Abstract

L'invention concerne un essai à base de cellules destiné à cribler des substances pour détecter leur capacité à inhiber l'activité de COX-2. Cet essai utilise, en tant que réactif de criblage, une lignée cellulaire qui a été modifiée afin de parvenir à une expression constitutive ou inductible de COX-2. Dans un mode de réalisation, les cellules expriment une forme mutante à activation constitutive et déficient en GTPase de la sous-unité .alpha. de la protéine G12 qui induit la production d'acide arachidonique et de COX-2. Une diminution de la prolifération cellulaire en présence de la substance test indique que la substance présente une activité inhibitrice de COX-2.
