Le clofibrate de metformine selon l'invention est le mono-clofibrate de metformine et répond à la formule ~ ~H3 ~H ~H
Cl ~ / \ ~ O - ~ - COOH, NH2 ~ ~ - NH - ~ - N(CH3)2 Ce sel peut être préparé à partir de la metformine et de l'acide clofibrique selon une méthode connue en soi.
Comme Ia base libre n'est pas un produit commercial, on préconise, selon l'invention, te partir du chlorhydrate te metformine.
Le chlorhydrate de metformine, en solution dans un alcool inférieur, notamment le méthanol à 2% d'eau, est traité au moyen d'une résine anionique pour obtenir la metformine base. La metformine base est ensuite salifiee par l'acite clofibrique au sein t'acétone.
On a tonné ci-après un exemple te préparation nullement limitatif mais tonné à titre d'illustration. Cet exemple concerne la fabrication d'environ 5 kg de clofibrate de metformine.

L'agitatlon est poursuivie pentant 4 heures après le tébut te la crlstallisatlon.
. Stockage en chambre froide de la solutlon contenant le clofibrate de metformine formé.
. Filtration pcur recueillir le clofibrate de metformine formé, pUi8 rinçage par 4 fois 5 iitres d'acetone glacée et essorage maximal. Récuperation de l'acétone à partir du filtrat.
. Séchage en étuve ventilée régulée a 40C.
On obtient ainsi 5 kg,ou un peu pius, de clofibrate te metformine, sous la forme d'une poudre cristalline blanche, soit avec un rendement global dlenviron 80% par rapport a la quantit~ théorique calculée à partir des quantités stoechiometriques de chlorhydrate de metformine et d'acide clofibrique mises en Jeu. Si l'on prend soin de concentrer partiellement sous vide ( ~ température ambiante), la solution acétonlque de metformine base (par exemple en lui soustrayant environ 15 litres t'acétone) le rentement peut être accru ~usqu'a 90 - 95%.

Du point te vue physique, ce sel , qui ne peut être anhydre qu'initialement, a un point de fusion différent de celui de - l'acide clofibrique (120C en tube capilaire) et de la metformine base (110C en tube capillaire).
Le spectre infrarouge est différent de celui des spectres de l'acite clofibrique, de la metormine base et du m~lange équimoléculaire t'acide clofibrique et de metforn$ne base. Le spectre infrarouge met, , ~C~62280 en évidence la salification pulsque le~ piC9 ~ 2700, 2500, 1600 et 1300 cm 3 du COOH libre de l'acide clofibrique et le plc à 1320 cm 1 de la metformine base n'apparaissent pas pour le sel.
Compte tenu des résultats analytiques, il y a de très fortes présomptions pour que le sel obtenu selon l'invention soit le mono-clofibrate de metformine, c'est-~-dire le sel équimoléculaire.
Enfin le clofibrate de metformine est soluble dans l'eau à froid (1 g dans 5 ml d'eau) en donnant des solutions de pH proche de la neutralité. Ce sel est également très soluble dans le méthanol et l~éthanol. Il est peu soluble dans l'acétone (1 g dans 100 ml) à la température ambiante, et insoluble dans le benzène, le chloroforme et l'hexane.
Les résultats des essais pharmacologiques ont montré que le sel selon l'invention avait des propriétés avantageuses différentes de la somme de celles de l'acide clofibrique et de la metformine. En ce qui concerne la toxicité, on a obtenu, chez la souris de race Swiss,les valeurs suivantes :
DL-50 par voie orale : 1,60 g/kg DL-50 par voie intrapéritonéale : 0,85 g/kg.
Le sel selon l'invention possède une action hypoglycémiante sur le diabète alloxanique du lapin. En ce qui concerne l'action hypocholestérolémique, on a observé une amélioration des dép~ts lipidiques chez le lapin recevant un régime hypercholest~rolémique et chez la Aouris obèse dlabétique génétiquement, cette amélloration étant nette au niveau artériel.
Le sel selon l'invention permet donc la régulation glycé-mique sans risque d'accident hypoglycémique, le traitement et la prévention des complications vasculaires du diabète. Il permet également le traitement et la prévention des affections athéromateuses Selon l!invention, on préconise des compositions thérapeu-tiques renfermant, en association avec un véhicule physiologiquement acceptable, le clofibrate de metformine. Ces compositions seront de préfé-rqnce administrée~ par voie orale ou par voie parentérale, la dose quoti-dienne dépendant de l~état du patient selon que ce dernier est ou n'est pas diabétique.
