Abstract
Patente de Invenção: <B>"RESISTêNCIA A DOENçAS POR PLANTAS MEDIADA POR PATOGENICIDAS MOLECULARES"<D>. A invenção proporciona construtos de genes que compreendem seq³ência(s) de nucleotídeos que codifica(m) um anticorpo, anticorpos ou parte(s) dos mesmos que se ligam especificamente a proteínas estruturais ou não-estruturais de agente(s) patogênico(s). A invenção também proporciona seq³ência(s) de nucleotídeos que codifica(m) um peptídeo, ou proteína(s), ou enzimas que são danosas ou tóxicas a esse(s) agente(s) patogênico(s). A invenção proporciona os meios e métodos para a montagem in vivo de duas ou mais proteínas, codificadas por seq³ências de nucleotídeos separadas, em um complexo de proteínas. Esses anticorpos, toxinas ou complexos poderão ser expressos e bombardeados em membranas celulares ou compartimentos de células de plantas em diferentes orientações e também podem ser clivados in vivo por diferentes proteases para tornar-se ativos. Esses agentes são denominados "patogenicidas moleculares". Os meios e métodos são proporcionados para imobilizar uma proteína recombinante capaz de ligar-se a um agente patogênico, através de técnicas de DNA recombinante. Os meios e métodos são também proporcionados para expressar patogenicidas moleculares em membranas de plantas monocotiledóneas ou dicotiledóneas. Se o agente patogênico é um vírus, por conseguinte o anticorpo poderá ser específico a proteínas virais estruturais ou não-estruturais envolvidas em entrada, desmontagem, replicação, movimento e/ou transmissão virais. Também proporcionados são seq³ências de ácidos nucléidos recombinantes que codificam patogenicidas moleculares, vetores, suspensão de plantas transformadas e células de calo, bem como plantas transgênicas regeneradas que expressam "patogenicidas moleculares" imobilizados em sua(s) membrana(s), seja diretamente ou após montagem in vivo de duas ou mais cadeias de polipeptídeos independentes. Também proporcionados são sistemas de expressão e plantas resistentes a agentes patogênicos geneticamente engenheiradas estavelmente transformadas com esses sistemas, bem como patogenicidas moleculares codificados pelos construtos de genes e organismos que expressam os patogenicidas moleculares. Além disso, são proporcionados kits que compreendem as proteínas de fusão acima mencionadas, patogenicidas, polinucleotídeos e vetores.
