Abstract

"ISOLAMENTO DE PROTEìNAS ENVOLVIDAS NO SILENCIAMENTO GêNICO PóS-TRANSCRICIONAL E MéTODOS DE USO". A presente invenção inclui um método para a detecção e isolamento de proteínas da cana-de-açúcar que interagem com as proteínas HC-Pro e P1 da SRMV e com outras proteìnas envolvidas no silenciamento de genes, particularmente na cana-de-açúcar. O método utiliza um ensaio duplo híbrido como uma HC-Pro, P1, ou outra proteína relacionada ao silenciamento, contendo uma proteína isca e uma proteína presa contendo um polipeptídio codificado por uma molécula de DNA em uma biblioteca CDNA. O método também inclui a identificação de falsos positivos através de ensaios duplo híbrido reversos e utilizando técnicas e in vitro tais como o farwestern blot ou prestes destrutivos nos quais as condições fisiológicas da planta podem ser replicadas. Finalmente, as interações podem ser confirmadas na planta. Algumas novas proteínas utilizadas aqui e descobertas utilizando esses métodos também são identificadas. Também são discutidos métodos para regular o silenciamento em plantas tais como a cana-de-açúcar utilizando proteínas virais e de planta.
