método para gerar uma molécula de rna com atividade de silenciamento, célula geneticamente modificada, planta geneticamente modificada, célula vegetal, planta, semente, métodos de produção de uma planta, de tratamento de uma doença e de introdução da atividade de silenciamento. um fornecido um método de gerar uma molécula de rna que possui uma atividade de silenciamento em uma célula, compreendendo: (a) a identificação de sequências de ácidos nucleicos que codificam moléculas de rna que exibem faixa de homologia de sequência predeterminada, não incluindo identidade completa, com relação as sequências de ácidos nucleicos que codificam moléculas de rna envolvidas com risc; (b) a determinação da transcrição de sequências de ácidos nucleicos que codificam moléculas de rna, de modo a selecionar sequências de ácidos nucleicos que codificam moléculas de rna transcritíveis; (c) a determinação da processabilidade em transcritos de rnas pequenos das sequências de ácidos nucleicos transcritíveis que codificam moléculas de rna que exibem uma faixa de homologia de sequência predeterminada de modo a selecionar sequências de ácidos nucleicos transcritíveis que codificam moléculas de rna processadas de forma aberrante, exibindo a faixa de homologia de sequência predeterminada; (d) a modificação de uma sequência de ácido nucleico das sequências de ácidos nucleicos processadas de forma aberrante, transcritíveis, de modo a conferir capacidade de processamento em rnas pequenos que estão envolvidos com risc e são complementares a um primeiro rna alvo ou a um rna alvo de interesse.
