Abstract
a presente invenção refere-se a uma cadeia pesada de imunoglobulina recombinante que compreende um domínio vh, um domínio ch1, uma região de articulação que vai até a cisteína mais terminal c da região de articulação intermediária, um circuito de conjugação de sortase, um domínio ch2 e um domínio ch3, em que o circuito de conjugação de sortase (a) consiste de pelo menos 16 aminoácidos localizados entre a cisteína mais terminal c da região de articulação intermediária e a sequência de consenso vfx1fpp (seq id nº 2, em que x1 é l ou i) no terminal n de um domínio ch2 de cadeia pesada de imunoglobulina; (b) compreende um motivo de reconhecimento de sortase que consiste da sequência de aminoácidos lpx1tx2 (seq id nº 1), em que x1 pode ser qualquer resíduo de aminoácido e x2 é g ou a; (c) compreende, terminal n para a sequência de (b), pelo menos dois aminoácidos; (d) compreende, terminal c para a sequência de (b), pelo menos seis aminoácidos; e (e) em que os terminais c e n de aminoácidos para a sequência de (b) não são cisteína e seu uso em conjugação dirigida a local com base na enzima sortase.
