Abstract

a invenção inclui materiais e métodos para gerar numerosos pequenos rnas a partir de uma construção polinucleotídica (gene sintético) para facilitar a reação multiplex guiada por rna na edição de genoma, modificação, inibição de expressão e outras tecnologias com base no rna. a construção do gene/polinucleotídeo sintético codifica componentes de rna policistrônicos separados pelo rnat e, preferencialmente, também inclui componentes regulatórios, tais como um promotor ou terminador para formar um cassete de expressão. uma vez transcrito em uma célula, o transcrito é processado pela célula a múltiplas moléculas de rna pelo sistema de processamento endógeno de rnat. o sistema pode ser aplicado para qualquer método de manipulação de genes baseado no rna, incluindo a edição de genoma mediada por rna, silenciamento de genes mediado por microrna artificial, manipulação genética por rna mediada por pequenos rnas, silenciamento de genes de rna dupla fita, mecanismos antisense e semelhantes.
