Abstract
"PROTEìNAS DE ESTRUTURAçãO DE MEMBRANAS". A presente invenção refere-se a proteínas de membranas são difíceis de expressar em forma recombinante, purificar e caracterizar, pelo menos em parte devido às suas propriedades hidrofóbicas ou parcialmente hidrofóbicas. Proteínas de estruturação de membranas (MSP) agrupam-se com proteínas de membranas-alvo ou outras proteínas hidrofóbicas ou parcialmente hidrofóbicas ou fragmentos de membranas para formar partículas de nanoescala solúveis que preservam sua estrutura e função nativas; elas são aperfeiçoadas em relação a lipossomos e micelas de detergente. Na presença de fosfolipídeo, MSPs formam discos nanoscópicos de bicamada de fosfolipídeo, com a MSP estabilizando a partícula no perímetro do domínio da bicamada. A estrutura em bicamada de partículas permite manipulação de proteínas incorporadas em solução ou em suportes sólidos, incluindo uso com técnicas sensíveis a superfície tais como microscopia de sonda de varredura ou ressonância em plasmónios superficiais superficiais.
