EMI1.2
Un degré inconvenable de cette suite réactionnelle est justement la préparation des 2-dialcoyle-amino-éthyle-mercaptanes libres (III), dont le rende- ment n'est pas du tout satisfaisant. La cause probable en est la considérable al- térabilité de ces substances, étant donné que ces substances posèdent à la fois une fonction acide et une fonction basique, dues à la présence simultanée d'un groupement acide et d'un groupement basique dans la molécule;
<Desc/Clms Page number 2>

<Desc / Clms Page number 1>
The object of the present invention lies in a new process for the preparation of bis-quaternary ammonium salts of aliphatic sulphides, having as general formula
EMI1.1
R1 and R2 mean in this general formula the low molecular weight alkyl residues; A y signifies a monovalent anion, for example halide anion.
The compounds of this group possess ganglioplegic activity as well as remarkable hypotensive activity, while being at the same time very little toxic (J. Fakstorp, J.G.A. Pedersen, Acta Pharmacolo Toxicol. 10, 7, 1954); it can be used to treat certain forms of hypertension (hypertension diseases).
@ The already known preparation processes of these compounds (I) (see KJM Andrews. Fo Bergel, AoL. Morrisson, Jo Chemo Soc. 1953, 2998; A. Fakstorp, J. Christiansen, Acta Chem. Scande 8, 346, 1954 ; M. Borovicka, M. Protiva, Chem.
Listy 48, 1374, 1954) have very serious technical drawbacks. In all these preparation processes, the same starting materials are used - namely hydrochlorides of 2-dialkyl-amino-ethyl-isothiuronium-chlorides (II); these hydrochlorides are decomposed by the action of dilute sodium hydrate, and they are converted into 2-dialkyl-amino-ethylemercaptanes (III); their sodium salts are condensed with a 2-dialkyl-amino-ethyl-chloride. Finally, the bis- / 2-dialkoyie-amino-ethyl / -sulfides (IV) thus created are converted into the corresponding bis-quaternary ammonium salts (see general formula I) by addition of alkyl halide.
Here is the reaction scheme:
EMI1.2
An inconvenient stage of this reaction sequence is precisely the preparation of the free 2-dialkyl-amino-ethyl-mercaptans (III), the yield of which is not at all satisfactory. The probable cause is the considerable alterability of these substances, given that these substances have both an acid function and a basic function, due to the simultaneous presence of an acid group and a basic group in the molecule. ;
on the other hand, this alterability is given by the presence of a very sensitive sulfhydryl group -SE, which is easily oxidizable by atmospheric oxygen to a disulfide group -SoS-o
It is for the same reason that the subsequent transformation of these mercaptans into sulphides (IV) proceeds with an unsatisfactory yield. Thus, for example, we obtain a most important mercaptane, namely 2-
<Desc / Clms Page number 2>
dimethyl-amino-ethyl-mercaptane, (see formula III, where R = CH3à in yield of 30-40 percent of theory (Chem. listy 48, 1374, 1954); subsequent condensation to obtain a sulfide (see Formula IV, where R1 = CE) runs with a yield of 60 percent of theory.
The other Danes get the same mercaptane with a yield of 31 percent of theory and the condensation product (IV) with a yield of 36.5 percent. (Acta Chemo Scando 8, 346, 1954). The English authors obtain the same mercaptane according to
EMI2.1
RO Clinton, U. Jo Salvador, SoCo Laskowski and CoMo Suter (JoAmoChemo Soc. 70, 950, 1948) with a yield of 22 percent of theory and the condensation product with a yield of 54 percent of theory (Jo Chem.
Soc. 1953, 2998) o It follows that the sulphide (see formula IV or R1 = CH) can only be obtained with a total yield of 11-21 per cent, starting from the iso- salt.
EMI2.2
tRiuronium (see formula II, where H 1 = CH3) a
It has now been found that one can work with a considerably better yield if the isolation of 2-dialkyl-amino-ethyl-mercaptans (see formula III) is eliminated. For hydrolysis of the isothiuronium salt, an aqueous - alcoholic alkaline hydrate solution is advantageously used in a quantity of four equivalents o This hydrolysis (consuming two equivalents of hydrate) already passes through at normal temperature and the mercaptane (III) is directly neutralized by the third equivalent of alkali forming its alkali salt.
A corresponding alcoholic solution of 2-dial-coyl-amino-ethyl-chloride hydrochloride is then added to this solution thus created.
EMI2.3
The β-dialkyl-amino-ethyl-chloride base is liberated by the action of the fourth equivalent of alkali hydrate so far. The condensation required to obtain the sulphide (IV) takes place. The reaction mixture is heated to the boiling point in order to terminate this condensation. The reaction mixture is then worked up in a manner known per se and the product is obtained by distillation of the sulphide (IV) in pure form with a yield exceeding 50%. cent of theory This yield exceeds 2-4 times the yields described so far in the literature.
The synthesis is then carried out by adding an alkyl halide to the sulphide (IV) under usual conditions, while acetone is advantageously used as a solvent.
This new process for the preparation of bis-quaternary ammonium salts of aliphatic sulphides, having the general formula (I), is precisely the subject of the present invention.
The procedure is described in more detail in the following example:
Example of realization
Is added with stirring to a solution of 60 g of potassium hydrate in 150 com of methyl alcohol and in 15 ccm of water 57 g of 2- hydrochloride.
EMI2.4
dimethyl-amino-ethyl-isothiuronichloride at normal temperature. The mixture is stirred for three hours, still at normal temperature. Then the mixture is left to stand overnight.
The next day we add to it while shaking
EMI2.5
a solution of 38 g of 2-dimethyl-amino-ethyl-chloride hydrochloride in 100 tsp of methyl alcohol and the mixture is stirred first for one hour at normal temperature, then stirring is continued at reflux by heating to boiling point in a water bath, after which heating is continued for 5 hours with constant stirring. After standing, overnight, the potassium chloride removed is filtered off and washed several times in methyl alcohol. The combined filtrates are evaporated under reduced pressure to a volume of 200 ccm and 200 ccm are added. of water. The extraction of an oleaginous product is done by ether. The ethereal solution is dried with solid sodium hydrate and evaporated in a water bath after filtration.
By distillation of the residue under reduced pressure, 24 g (53 percent) of bis- / 2-dimethyl-amino- are obtained.

The following day, the removed product is filtered off and washed in 40 cc of acetone. The crude product is obtained, the yield of which corresponds approximately to theory, and it is purified by recrystallization from boiling aqueous ethyl alcohol. Recrystallization of 33g of the product is carried out in a mixture of 90 ccm of ethyl alcohol and 12 ccm of water.
