La présente invention a pour objet la préparation de 1-déhydro-testostérones renfermant en position 17a un reste alcoylique ou alcénylique comportant de 2 à 4 atomes ,de carbone et de leurs esters, par exemple de la 1-déhydro- 17a-propyl-, -17a-butyl-, -17a-allyl- ou métallyl-, -17a- vinyl-, -17a-isopropényl-testostérone et de ses esters, tout particulièrement toutefois de la 1-déhydro-17a-éthyltestostérone et de ses esters. anabolique, ces composés possèdent un Intérêt thérapeutique et peuvent être utilisés comme médicaments dans les maladies correspondantes. L'effet anabolique n'en est pas, ce qui est surprenant, associé à des effets sexuels marqués.
Ces propriétés rendent ces composés particulièrement intéressants pour le traitement des affections exigeant une édification protéinique accrue et dans lesquelles l'effet sexuel est Indésirable.,comme le manque de poids, des états post-opératoires ou post-infectieux, l'asthénie après la grossesse et dans la gériatrie.

La réaction conforme au procédé a lieu dans la culture de champignon décrite ou à l'aide des enzymes éventuellement enrichis ou séparés qu'elle renferme, ainsi, dans le cas le plus.simple, dans une suspension du mycélium de champignon séparé, du mycélium de champignon homogénéisé ou dans des filtrats ou dans des extraits aqueux dudit mycélium. La séparation des produits du procédé peut avoir lieu, par exemple, en extrayant le mélange réactionnel avec un solvant organique, par exemple avec du chlorure de méthylène ou de l'acétate d'éthyle.
Pour purifier davantage l'extrait ainsi obtenu, il est approprié de faire appel notamment à la chromatographie, par exemple sur de l'oxyde d'aluminium ou du gel de silice, d'utiliser des méthodes de répartition, par exemple le procédé à contre-courant ou la séparation à l'aide des réactifs de Girard comme l'acéthydrazide de triméthyl-ammonium ou de pyridinium. Si l'on part, lors de la préparation des 1-déhydro- testostérones de la présente invention ou de leurs esters, du ¯1,4-3,17-dioxo-androstadiene, on introduit alors en position 17a le reste hydrocarboné désiré à l'aide du com- posé organo-métallique correspondant comme le composé de Grignard,ou à l'aide du composé de métal alcalin, par exemple à l'aide de chlorure, de bromure ou d'iodure d'éthyl- magnésium ou à l'aide de lithium-éthyle. Avant la réaction sur le composé organo-métallique, on protège le groupe 3-oxo, par exemple en le transformant en un éther 3-énolique comme l'éther 3-énoléthylique, en un 3-cétal, tel qu'un ,

Exemple 1.
Dans 1 litre d'eau de conduite, on dissout 2 g de nitrate de sodium, 1 g d'orto-phosphate de potassium, 0,5 g de sulfate de magnésium-heptahydrate, 0,5 g de chlorure de potassium, 50 g de glucose et 1 g d'extrait de levure Difco, ajuste à un pH de 5 par addition d'une solution d'hydroxyde de sodium, puis stérilise. On ensemence la solution nutritive obtenue avec 50 cm3 d'une culture 6'agitation de Didymella lycopersici âgée de 4 jours et l'agite 48 heures à 27 , ce qui fait que la culture se développe bien.
A 2 litres de cette culture, on ajoute, dans des conditions stériles, une solution de 500 mg de 17a-éthyl-testostérone dans 15 car d'acétone. On secoue pendant 3 jours à 27 , sépare ensuite le mycélium par essorage, lave à l'eau et à l'acétate d'é- thyle, puis après avoir réuni les filtrats, extrait ces derniera à l'acétate d'éthyle. On lave à l'eau les solutions d'acétate d'éthyle, les sèche et les évapore soua vide. On recristallise le résidu d'extraction dans un mélange d'acé- tone et d'éther de pétrole, et lton obtient la 1-déhydro- 17a-éthyl-testostérone fondant à 133 - 134 .
D'une manière analogue,, on peut transformer la 17c-vinyl-testostérone en 1-déhydro-17a-v@nyl-testostérone.
Après recristallisation dans un mélange d'acétone et d'éther isopropylique, ce dernier composé fond à 165 - 166 .

Exemple 3
Dans 250 cm3 d'ocide acétique glacial, on dissout 31,8 g de puis ajoute 20 gouttes d'une solution saturée d'acide bromhydrique dans l'acide acétique glacial.
On ajoute ensuite d'abord lentement, puis plus rapidement une solution de 32 g de brome dans 200 cm3 d'acide acétique glacial, de manière à empêcher un excès de brome dans la solution réactionnelle* La tempé- rature réactionnelle monta alors peu à peu de 15 à 40 .
Lorsque la bromuration est terminée, on refroidit et préci- pite le produit réactionnel en le versant dans 3-litres d'eau.
On essore, lave à l'eau à neutralité et finalement au métha- nol pour éliminer les impuretés plus facilement solubles.
On sèche le produit dans une ermoire à vide. On obtient le 2,4-dibrome-3-ox-17ss-hydroxy-17a-éthyl-androstane.
Dans 50 cm3 de collidine, on chauffe pendant 25 minutes à l'ébullition en agitant 24 g du dibromure ci-dessus.
Après refroidissement, on essore le bromhydrate de collidine et le lave à la collidine et à l'éther. Après avoir rassemblé les filtrats, on les lave avec de l'acide sulfurique et à l'eau, puis les sèche sur du sulfate de sodium. On concentre la solution éthérée jusque oristallisation commençante.
Pour compléter la cristallisation, on laisse reposer pendant 10 à 12 heures à 0 . On rassemble sur l'entonnoir filtrant la 1-déhydro-17a-éthyl-testostérone ainsi obtenue, la lave

avec un peu d'éther froid et la sèche dans l'armoire à vide.
La matière de départ utilisée ci-dessus peut, par exemple, être obtenue comme suit :
Dans une solution d'acide acétique glacial et avec de l'oxyde de platine comme catalyseur, on hydrogène à la température ambiante du ¯5-3ss,17ss, dihydroxy-17a-éthylandrostène. Après absorption de la quantité d'hydrogène calculée pour une double liaison, on sépare le catalyseur, précipite le produit d'hydrogénation par addition d'eau, essore et sèche.
Dans 500 cm3 de toluène et 150 cm3 de cyclohexanone,

on dissout environ 30 g du 30, 17fi-dihydroxy-17a-éthyl- androstane ainsi obtenu. On chauffe à l'ébullition et élimine par distillation 100 à 200 cm3 de toluène jusqu'à ce qu'il ne reste absolument plus d'humidité. On ajoute dans la solution encore chaude 5 g d'isopropylate d'aluminium et fait ensuite bouillir pendant 30 minutes à reflux.
