<Desc/Clms Page number 4>
anti-inflammeatoire.
<Desc/Clms Page number 5>
EMI5.1
<Desc/Clms Page number 6>
<Desc/Clms Page number 7>

wherein R1 represents a free, esterified or etherified hydroxy group and Y represents a hydrogen atom or a free or esterified hydroxy group.
The esterified hydroxy groups indicated are above all those derived from organic carboxylic acids of the aliphatic, alicyclic, aromatic or heterocyclic series, in particular those comprising from one to 18 carbon atoms, for example formic, acetic, propionic acids, butyric acids, valerian acids, such as n-valerianic acid, or trimethylacetic, trifluoroacetic acids, caproic acids such as ss-trimethyl-propionic acid or diethylacetic acid, oenanthic, caprylic, pelargonic, capric acids , undecylic acids, for example undecylenic acid, lauric, myristic, palmitic or stearic acids, for example oleic, cyclopropane-carboxylic acids,

The ester groups can however also be derived from ortho-carboxylic acids, such as ortho-formic acid, ortho-acetic acid or ortho-propionic acid, these acids possibly, like the dicarboxylic acids indicated above, provide cyclic 17,21-esters.
As etherified hydroxy groups, mention should be made in particular of those derived from alcohols comprising from one to 8 carbon atoms, such as lower aliphatic alkanols, ethyl alcohol, methyl alcohol, propyl alcohol, etc. isopropyl alcohol, butyl or amyl alcohols, or araliphatic alcohols, in particular monocyclic lower aryl aliphatic alcohols such as benzyl alcohol, or heterocyclic alcohols.

The new compounds of formula (I) above possess valuable pharmacological properties. This is how to. Besides a thymolytic effect and an effect of inhibiting adrenals, they exhibit in particular an anti-inflammatory effect as shown in a test carried out on animals, for example on rats, in the granuloma test caused by foreign bodies, at doses of 0.3 to 10 mg / kg. The new compounds can therefore be used as analogues of corticosteroids. especially as anti-inflammatory agents. The new compounds are, however, also valuable intermediates for the preparation of other useful substances, in particular pharmacologically active compounds.
In particular, the

The substance is active as an anti-inflammatory agent in the same test at doses of about 0.1 mg per kg when administered locally. The substance shows after oral administration a dissociation between the anti-inflammatory effect and the thymolytic effect, the latter being about 10 times stronger than the effect.

In the corresponding 21-pivalate, doses of the same order of magnitude are observed with regard to the anti-inflammatory effect: 11 is a little greater than in the case of the aforementioned 21-acetate, while the thymolytic effect is greater. weak as the anti-inflammatory effect,
The compounds of the formula (1) above can be obtained in a manner known per se.
In particular They can be prepared when a) in a compound of the general formula

wherein R1 represents a free, esterified or etherified hydroxy group and Y represents a hydrogen atom or a free or esterified hydroxy group, chlorine is added to the 1,2-double bond, and the hydrochloric acid is removed from the 1,2-dichloro-compound obtained, or when b) in a compound of the general formula
EMI4.2
wherein R1 and Y have the meaning given in formula (I), the 11ss-hydroxy group is dehydrogenated to 11-oxo group, or when c) in compounds of the general formula
<Desc / Clms Page number 5>
EMI5.1
in which R1 and Y have the meaning indicated in the 3rd formula (I),
a double bond is introduced in a manner known per se in position 1,2, then, if desired, esterified in the compounds obtained free hydroxy groups and / or etherified the hydroxy group in position 21 and / or converted into free hydroxy groups esterified or etherified hydroxy groups.
The addition of chlorine to the 1,2-double bond according to method a) takes place in a manner known per se. The chlorination can, for example, be carried out in an inert solvent such as dioxane, in the presence of a carboxylic acid such as propionic, for example, at low temperature in the dark.
Removal of hydrochloric acid from 1,2-dichloro compounds takes place by treatment with a base, preferably with a nitrogenous tertiary organic base such as, for example, triethylamine, pyridine or enlumina.
The dehydrogenation of an 11-hydroxy group to an 11-oxo group according to method b) mentioned above is carried out in a manner known per se, for example with chromium trioxide and pyridine.
To introduce a double bond into compounds of formula (IV) according to method c), chemical or microbiological dehydrogenation methods are used which are known in themselves. Among those indicated in the first place, there may be mentioned, for example, dehydrogenation with the aid of selenium dioxide or selenious acid, preferably in a tertiary aliphatic alcohol such as tert-butanol or amyl alcohol

tertiary or with 2,3-dichloro-5,6-dlcyano-1,4-benzoquinone in boiling benzene or dioxane.
<Desc / Clms Page number 6>
In the case of microbiological dehydrogenation, for example cultures of microorganisms of the type
Corynebacterium simplex, Septomyxa affinis or Didymella lycopersici, or enzymes isolated from said microorganisms and separated from the mycelium.
In accordance with the process, the free hydroxy groups at position 17 and 21 are esterified, if desired. It is possible to prepare both 17a-mono-esters or 21-mono-esters as well as 17a,
21-diesters. In order to prepare the 21-mono-esters, the 21-hydroxy-compounds are treated, in a manner known per se, with reactive functional derivatives of carboxylic acids, preferably with those of the acids indicated above, for example by an acid anhydride or with an acid halide, for example in a tertiary base such as pyridine.
A free hydroxy group at position 17a can be selectively esterified according to methods known per se.
For example, the 17a, 21-diesters are prepared by acylating with a carboxylic anhydride such as acetic anhydride, adding a strong acid, in particular an aromatic sulfonic acid such as, for example, p-toluene acid. sulfonic acid as a catalyst, then saponifies the 21-ester group under mild conditions. For example, solutions of an alkali metal carbonate or hydrogen carbonate in an aqueous solution of an aliphatic alcohol such as methanol or ethanol are used for this purpose.
The 17α-mono-esters can also be obtained in the following manner known per se: from compounds of the formula (I), which have free hydroxy groups in position 17O and in position 21, one obtains, by reaction on an ortho-ester of the type
R'-C (OR ") 3, where R 'represents a hydrogen atom or an alkyl residue and R" represents an alkyl residue, in the presence of a strong acid, such as p-toluenesulfonic acid, by example, as a catalyst, in an inert solvent such as benzene, for example, the cyclic 17Ó21-ortho-esters are obtained. By hydrolysis with a weak organic acid such as oxalic acid, for example, the ester linkage at 21 is then selectively hydrolyzed, resulting in a 17α-mono-ester.
<Desc / Clms Page number 7>
In accordance with the process, a free hydroxy group at position 21 is etherified if desired in a manner known per se, preferably with reactive derivatives of the alcohols mentioned above. With dihydropyran, for example, in a solvent which does not take part in the reaction, such as tetrahydrofuran, diethyl ether or chloroform, in the presence of phosphorus oxychloride, 21-tetrahydro-pyranyl ether is obtained. -
The compounds to be used as starting substances and corresponding to the formulas (II) to (IV) above are known, or can be prepared in a manner known per se.
The invention also relates to the embodiments of the process in which one starts with a compound obtained as an intermediate product at any stage of the process and carries out the phases still missing from said process, or interrupts the latter at one. any of its stages, or in which a starting substance is formed under the reaction conditions .. ¯ ¯
The present invention also relates to the production of pharmaceutical preparations intended for use in human or veterinary medicine and comprising the novel pharmacologically active substances which are described above as active substances, together with a pharmaceutical carrier material.
As carriers, organic or inorganic substances suitable for enterane administration, for example oral, parenteral or topical administration, are used. 'For the formation of these supports, substances which do not react with the new compounds, such as for example water, gelatin, lactose, starch, magnesium stearate, talc, vegetable oils, alcohols, are envisaged. benzyls, gums, polyalkylene glycols, petroleum jelly, cholesterin and other known excipients. The pharmaceutical preparations can be in solid form, for example in the form of tablets, dragees or capsules, or in liquid or semi-liquid form in the form of solutions, suspensions, emulsions, ointments. or creams.
Where appropriate, these pharmaceutical preparations are sterilized and / or contain auxiliary substances such as preservatives, stabilizers, wetting or emulsifying agents,: salts serving to vary the osmotic pressure, or buffers. They can also contain still other

therapeutically valuable substances. The novel compounds can also serve as starting materials for the preparation of other valuable compounds. The compounds of the present application can also be used as additives to animal feed.

The dark brown cloudy solution is stirred for a further 20 hours at room temperature and then poured into 2 liters of water. This mixture is extracted several times with methylene chloride, the extracts are washed several times with water,
dried over sodium sulphate and the solvent is completely evaporated off. The reaction product is purified by dissolving the slightly greenish residue which is partially crystallized (2.98 g) in a mixture (97: 5) of toluene and of ethyl acetate, and chromatography through 120 g of silica gel. The medium reaction is evaporated and the residue crystallized from ethyl acetate.
