TECHNICAL FIELD [0001] The present invention relates to the field of microbial technology, in particular to a
Bacillus pumilus strain, and an application thereof beneficial for a weaned piglet.
BACKGROUND [0002] A nursery piglet is referred to as a weaned piglet, the pig at this stage is very fragile, and various diseases, even massive deaths easily occur, since the maternal antibody disappears, the new immune system has not yet been established, besides, there are other reasons, such as a rapid growing speed and many stress factors. Although antibiotics and chemicals can solve the emerging problem in a relatively short period of time, drug-resistance and drug residue problems caused therefrom may have an impact on the late growth of the piglet. How to improve the health condition, survival rate, and uniformity of the weaned piglet as well as how to fully play aftereffects of growth are problems commonly concerned by every farmer.
[0003] The microbial ecological preparation has roles of regulating and controlling animal intestinal microbial balance, proliferating beneficial bacteria, inhibiting harmful bacteria, reducing intestinal PH values, improving the enzyme activity in the digestive tract, nutrient digestibility and the organism immunity, and etc. Reports on the application of the microbial ecological preparation in piglet breeding are increasing. CN 101703136B reports an application of the Lactobacillus plantarum viable bacteria preparation in preventing and treating piglet diarrhea, the efficiency rate for the piglet diarrhea is more than 90% and the cure rate is more than 80%. CN 103255080B reports an application of the Bacillus coagulans (Bacillus coagulans HM-09) in the preparation of feedstuff for improving the productive performance and diarrhea of piglets, the occurrence rate of diarrhea of a weaned piglet can be reduced, thus the occurrence of enterogenous diseases can be reduced, the feedstuff digestibility is improved, and the utilization rate of the piglet feedstuff is improved. CN 103205373B reports a strain of Clostridium butyricum for breeding a piglet, which can antagonize animal pathogenic bacteria in the intestinal tract of the piglet, so as to maintain and regulate the microbial ecological balance, enhance the immune function of the organism, produce amylase and vitamins, and promote digestion and absorption and nutrition effects of the animals on the feedstuff.
[0004] In general, the microbial ecological preparation is slow in effect and capable of receiving a good effect after being used consistently in the livestock breeding process. Therefore, there is a need to develop a microbial ecological preparation capable of a long-term health care, but also quickly solving problems of piglet diarrhea, loose bowels, etc., and it is particularly necessary for realizing stable production of the weaned piglets.
SUMMARY [0005] In view of the above problems in the prior art, the applicant provides a Bacillus pumilus strain and a microbial ecological combined agent for a weaned piglet. Through the present invention, not only problems of piglet diarrhea, loose bowels, etc can be solved quickly, but also the health level of a weaned piglet can be improved during a long-term use. [0006] Technical solutions of the present invention are as follows:
[0007] A Bacillus pumilus strain ZS008 is deposited in the China Center for Type Culture Collection (CCTCC) on July 13, 2015 with a deposit number of CCTCC No. M2015451.
[0008] A microbial ecological Bacillus combined agent for a weaned piglet is combined and formulated by the following bacterial species: a Bacillus pumilus (CCTCC No. M2015451), a Bacillus coagulans (CCTCC No. M2015273), a Bacillus subtilis (CCTCC No. M2011443) and a. Bacillus licheniformis (CCTCC No. M2012458).
[0009] The microbial ecological Bacillus combined agent has an effective viable count of

1.4x 10 -7.0x 10 cfu/g, and the bacterial species composition is as follows:

Bacillus licheniformis CCTCC No. M 2012458 are commercially available from Taizhou
Lvshengyuan Biotechnology Co., Ltd. The product specifications thereof are both 5.OxlOlocfu/g.
[0013] The beneficial technical effects of the prevent invention are as follows:
[0014] 1) Problems of piglet diarrhea, loose bowels, etc can be solved quickly;
[0015] 2) After a long-term use, the health level of a weaned piglet can be improved, and thus the effect of improving production capacity of the weaned piglet can be achieved.
DETAILED DESCRIPTION [0016] 1. Screening of Bacillus pumilus [0017] 2g of soil sample near Jiangsu Wuxi Fangdingding Pig Farm is weighed, and added with 98 ml of sterile physiological saline, after heat treatment for 10 minutes at the temperature of 80°C, oscillated in a rotary shaker(120 rpm) at 37°C for 20 min, 1 ml of the supernatant is aspirated and added to LB culture medium for dilution and coating, so as to isolate bacterial strains. Genuses and species of bacterial species are determined effectively and accurately taking means, such as morphological observation, physiological biochemical experiment, 16S rDNA analysis on bacterial strains.
[0018] Finally, a strain with a serial number of ZS008 is obtained, and identified as a Bacillus pumilus. The strain is deposited in the China Center for Type Culture Collection on July 13, 2015 with a deposit number of CCTCC No. M2015451.

Screening of Bacillus coagulans [0020] 2g of soil sample near a certain dairy farm in Mashan, Wuxi, Jiangsu is weighed, and added with 98 ml of sterile physiological saline, after heat treatment for 10 minutes at the temperature of 80°C, oscillated in a rotary shaker(120 rpm) at 37°C for 20 min, 1 ml of the supernatant is aspirated and added to LB culture medium for dilution and coating, so as to isolate bacterial strains. Genuses and species of bacterial species are determined effectively and accurately taking means, such as morphological observation, physiological biochemical experiment, 16S rDNA analysis on bacterial strains.
[0021] Finally, a strain with a serial number of ZS007 is obtained, and identified as a Bacillus coagulans.

Preparation of a microbial ecological combined agent [0024] The microbial ecological combined agent is combined and formulated by a Bacillus pumilus (CCTCC No. M2015451), a Bacillus coagulans (CCTCC No. M2015273), a Bacillus subtilis (CCTCC No. M2011443) and a Bacillus licheniformis (CCTCC No. M2012458).
[0025] In particular, single bacterium powders of Bacillus subtilis CCTCC No. M 2011443 and Bacillus licheniformis CCTCC No. M 2012458 are commercially available from Taizhou Lvshengyuan Biotechnology Co., Ltd. The product specifications thereof are both 5.OxlOlocfir/g.

B, and Group C is the same with Group B, except that there is no Bacillus pumilus', the dosage of the microbial ecological preparation is 1.0-5.Og/head/day. The piglets are weaned at 21 days old under an average weight of 6.5kg, the pre-test is 7 days and feeding test is 30 days. The breeding site is a certain breeding cooperative in Nanjing.

[0036] After piglet diarrhea occurs, 5g of microbial ecological preparation product is used to each piglet for treatment. After being stirred with 10 mL of cold boiled water, the product is drawn into a syringe, and drenched in the corner of the piglet’s mouth with the syringe. When 15 the microbial ecological preparation product contains Bacillus pumilus, the mental state of the piglet gets better after 4 to 6 hours, the piglet diarrhea disappears within 24 hours, and the appetite returns to normal. When the microbial ecological preparation product doesn’t contain Bacillus pumilus, the mental state of the piglet gets better after 12 hours; consolidation treatment is carried out once, the piglet diarrhea disappears after about 2 days, and the 5 appetite returns to normal.

- THE CLAIMS DEFINING THE INVENTION ARE AS FOLLOWS:1. A Bacillus pumilus strain ZS008, deposited in the China Center for Type Culture Collection on July 13, 2015 with a deposit number of CCTCC No. M2015451, in an isolated form.
- 2. A microbial ecological Bacillus combined agent for a weaned piglet, characterized in that, being combined and formulated by the following bacterial species: a Bacillus pumilus strain ZS008, deposited in the China Center for Type Culture Collection on July 13, 2015 with a deposit number of CCTCC No. M2015451, a Bacillus coagulans (CCTCC No. M2015273), a Bacillus subtilis (CCTCC No. M2011443) and a Bacillus licheniformis (CCTCC No. M2012458).
- 3.
