Abstract
La presente invención proporciona procesos y métodos mejorados y/o acortados para preparar TIL con el fin de preparar poblaciones terapéuticas de TIL modificados genéticamente con expresión reducida de CISH y opcionalmente de PD-1 como se describe en la presente descripción. Reivindicación 1: Un método para preparar linfocitos infiltrantes de tumores modificados genéticamente (TIL) que comprende la expresión reducida de CISH y opcionalmente de PD-1, el método que comprende: (a) introducir en los TIL ácido(s) nucleico(s) que codifique(n) una o más primeras nucleasas efectoras de tipo activador de la transcripción (nucleasa TALE) capaces de inactivar selectivamente mediante escisión de ADN un gen que codifica CISH, en donde la una o más primeras nucleasas TALE comprenden una nucleasa TALE que se dirige contra la secuencia de ácido nucleico de la SEQ ID Nº 175, y opcionalmente se introducen una o más segundas nucleasas TALE capaces de inactivar selectivamente por escisión de ADN un gen que codifica PD-1; y (b) expandir los TIL. Reivindicación 75: Un método para tratar el cáncer en un sujeto que necesite de este, el método comprende administrar al sujeto una población terapéutica de linfocitos infiltrantes de tumores modificados genéticamente (TIL) que comprende la expresión reducida de CISH y/o CISH y PD-1, en donde la población terapéutica de TIL modificados genéticamente se pueden obtener mediante el método de cualquiera de las reivindicaciones 1 a 20 y 32 a 73.
