【発明の詳細な説明】
ターゲッティングされた遺伝子輸送システム本発明の背景
細胞に外来遺伝子を導入するために種々の技術が使用されている。物理的な方
法には、リン酸カルシウムを用いる共沈殿、電気穿孔法、及び粒子衝撃法(par-
ticle bombardment)が含まれる。これらの直接移入技術は、in vitro において
は適切であるが、in vivo では非実用的である。in vivo での遺伝子治療を保証
するには、輸送のためのウイルス性ベクター又はリポソームのようなキャリアー
を頼りにする。ウイルス性ベクターに対しては長期の安定性についての懸念がま
だ残されている。他の制限はDNA配列のサイズであり、これは通常ウイルス性
ベクターに取り込ませうる7〜8Kbに制限されている。一方、リポソームは一

ポリマー系は、変更された薬物動力学及び生体内分布のようなリポソーム系の利

本発明の他の目的は、ポリマー粒子を使用して細胞にDNAを輸送する方法を
提供することである。
本発明のこれらの目的及び他の目的は、以下に説明する1以上の態様によって
提供される。一態様では、特異的な標的に遺伝子を輸送するためのミクロ粒子を
提供する。該ミクロ粒子は、ゼラチン及びDNAを具備し、結合分子又はターゲ
ッティングリガンドが前記ミクロ粒子の表面に結合されている。
本発明の他の態様では、特異的な標的に遺伝子を輸送するためのミクロ粒子を
形成する方法を提供する。該方法は、DNAとゼラチンのコアセルベーションに
よってミクロ粒子を形成すること、及び結合分子又はターゲッティングリガンド
を該ミクロ粒子の表面に接着することのステップを具備する。
本発明の更なる態様では、細胞に遺伝子を導入する方法を提供する。該方法は
、形トランスフェクションされる細胞をゼラチン及びDNAを含有する固体ミク
ロ粒子とインキュベートし、この場合にターゲッティングリガンドが前記ミクロ
粒子の表面に結合され、前記ターゲッティングリガンドが前記細胞の表面に結合
することを具備する。
従って、本発明は、製造が簡単であり、実用的なコストであり、放出を制御す
る能力を有し、保存に安定であり、生物適合性である魅力的なDNA輸送システ

種々の鎖長の核酸分子を水性条件下でポリマーカチオンと複合体形成させ、サ
ブミクロンからミクロンの範囲のサイズの固体ミクロ粒子を形成させうることが
本発明で明かとなった。適切にターゲッティングされた場合、これらの核酸を導
入したミクロ粒子は、食細胞活性を有する細胞をトランスフェクションすること
ができる。ミクロ粒子の分解及び核酸の放出速度は、架橋の割合を変化すること
で演繹的にデザインされうる。核酸の導入レベルは、>95%のカプセル化効率
で30%(w/w)程度の高さになりうる。
本発明に従えば、ゼラチン又はゼラチンと同様の電荷密度を有する他のポリマ
ーカチオンを使用し、核酸と複合体を形成させ、ミクロ粒子を形成する。ゼラチ
ン源は、厳密に考慮する必要はなく、ウシ、ブタ、ヒト、又は他の動物源から得
ることができる。典型的には、ポリマーカチオンは19,000〜30,000
の間の分子量を有しうる。ポリ−L−リジンは本発明のポリマーカチオンとして
特に有用である。望ましくは、硫酸ナトリウムを使用してポリマーカチオンと核
酸のコアセルベーションを誘導する。約40から60%の濃度のエタノールを使
用してコアセルベーションを誘導することもできる。コンドロイチン硫酸もミク
ロ粒子に導入されうる。これは、ミクロ粒子に医薬のような他の基質を含めるこ
とを望む場合に特に有益である。典型的にはコンドロイチン硫酸の濃度は、約0
.005%と0.1%の間である。
ターゲッティングリガンドは、ミクロ粒子の表面に直接に結合されうるか、又
は「架橋」若しくは「スペーサー」を使用して間接的に結合されうる。アミノ基
がゼラチンのリジン基に存在するのでミクロ粒子の表面を、ターゲッティング部
分の直接のカップリングのために容易に誘導体化することができる。他の方法と
して、アビジン−ビオチンのようなスペーサー(結合分子及びターゲッティング
リガンドの誘導体化した部分)使用し、ミクロ粒子にターゲッティングリガンド
を間接的に結合することができる。ビオチニル化された抗体及び/又は他のビオ
チニル化されたリガンドは、アビジンに対してビオチンが高親和性(Ka〜101 5
M-1)であるのでアビジンでコートされたミクロ粒子表面に効果的にカップリ
ングされる(Hazuda,et al.,1990,Processing of precursor interleukinl b

ology,184:5-13)。IgGの定位選択的な結合は、Fc部分に見出されるオリ
ゴ糖基で抗体をビオチニル化することによって達成されうる(O'Shannessy,et
al.,1984,A novel procedure for labeling immunoglobulins by conjuga-tio
n to oligosaccharides moieties,Immunol.Lett.,8:273-277)。このデザイ
ンは、利用しうる結合部位の総数を保護し、マクロファージのようなFc受容体
−結合細胞に対する結合した抗体の免疫原生を低くする手助けとなる。当分野で
公知のように、アビジン−ビオチン架橋以外のスペーサーも使用しうる。例えば
、スタフィロコッカル(Staphhylococcal)タンパクAを、ミクロ粒子に免疫グ
ロブリン分子のFc部分を結合するためにミクロ粒子上にコートしうる。
ミクロ分子への結合分子又はターゲッティングリガンドの架橋を使用し、ミク
ロ粒子の安定性を増し、更にミクロ粒子に結合分子又はターゲッティングリガン
ドを共有結合的に付加する。架橋の割合はミクロ分子からの核酸の放出の速度に
直接に影響する。架橋はグルタルアルデヒド、EDC(1−エチル−3−(3−
ジメチルアミノプロピル)−カルボジイミド、DCC(N,N’−ジシクロヘキ
シルカルボジイミド)のようなカルボジイミド、カルボキシルズ(ペプチド結合
)結合、ビス(スルホスクシンイミジル)スベレート(bis(sulfosccinimid-yl)
suberate)、ジメチルスベリイミデート(demethylsuberimidate)等を用いて達
成しうる。
本発明のターゲッティングリガンドは、体内の特異的なタイプの細胞に結合す
る何れかの細胞である。これらは、細胞受容体が存在する何れのタイプの分子で
あってもよい。好ましくは、細胞受容体は特異的な細胞タイプにのみ発現される
。使用されうるターゲッティングリガンドの例は、ホルモン、抗体、細胞接着分
子、糖、医薬、及び神経伝達物質である。
本発明のミクロ粒子はよい導入特性を有する。典型的には、本発明の方法に従
えば、少なくとも5%(w/w)の核酸を有するミクロ粒子が得られる。導入は
核酸が10又は15%よりも多くなることが好ましい。しばしば、核酸が20又
は30%よりも多いミクロ粒子が達成されうる。典型的には、95%以上のミク
ロ粒子への核酸の導入効率が達成されうる。
本発明の方法は、ポリマーカチオンと核酸のコアセルベーションを含む。この
過程は正に荷電したポリマーカチオンと負に荷電した核酸の相互作用に依存する
ので、複雑なコアセルベーション過程を考えることができる。しかし、硫酸ナト
リウム(又はエタノール)は相変化を誘導することによってコアセルベーション
反応を誘導する。従って、簡単なコアセルベーション反応も考慮されうる。核酸
は、1 ng/ml から500μg/ml の間の濃度でコアセルベーション混合物中に
存在する。望ましくは、核酸は少なくとも約2〜3kbの長さである。硫酸ナト
リウムは、7から43 mM の間の濃度で存在する。ゼラチン又は他のポリマーカ
チオンは、約2から7%の間でコアセルベーション混合物中に存在する。
注目すべきミクロ粒子輸送システムは、調製の容易さ、実用的なコスト、核酸
の導入レベル、制御された放出能力、保存安定性、及び成分の免疫原生のような
ファクターの中で、微妙なバランスを必要とする。本明細書で開示される遺伝子
輸送システムは、リポソームシステムを含めた他の粒子の輸送システムに比べて
有利である。不安定性、低い導入レベル、及び制御された放出能力の問題は、ポ
リマーミクロ粒子システムでよりよく解決される。ゼラチンは、in vivo でのそ
の生物適合性及び酵素分解性により、外科手術の組織接着剤(Weinschelbaum,e

酸/ポリグリコール酸コポリマー(polylactic/polyglycolic co-polymers)の
ような他の合成ポリマーシステムに比べて、穏和な条件でのミクロ粒子の形成を
アピールすることができる。溶媒の蒸発及びホットメルト技術は合成ポリマーミ
クロ粒子を形成するのに使用されるが、複合コアセルベーションは
有機溶媒との接触も熱も必要としない。これは、不活性溶媒捕捉によるばかりで
なく、直接の電荷−電荷相互作用によっても、核酸のような生物マクロ分子をカ
プセル化するのに特に適している。
10kb以上の遺伝子を輸送することができないウイルス性ベクターと異なっ
て、本発明のミクロ粒子輸送システムではこのような大きさの制限はない。約2
kb以上の核酸分子を使用し得、10kbよりも大きい核酸分子さえも使用する
ことができる。
一般に、可能なターゲットの範囲は、注入の経路、例えば静脈内若しくは動脈
内、皮下、腹腔内、髄腔内等に依存する。全身性の注入に対しては、この輸送シ
ステムの特異性は血液に結合したミクロ粒子に対するターゲットの接近性に影響
され、これはまた粒子のサイズの範囲に影響される。粒子のサイズは、温度、成
分の濃度、及びコアセルベーション混合物のpHに影響される。粒子はまた、例
えば蔗糖勾配超遠心分離法によって大きさで分別されうる。150ナノメータ以
下のサイズの粒子は、ほとんどの血管壁に一列に並んだ開窓を介して浸透するこ
とによって間隙スペースに近づくことができる。このような状況下では、ターゲ
ッティングされうる細胞の範囲は広がる。ターゲッティングされうる細胞の省略
したリストには、肝シヌソイドの実質細胞、結合組織の繊維芽細胞、膵臓のラン
ゲルハンス島内の細胞、心筋細胞、腸のチーフ(Chief)及び壁細胞、骨の骨細
胞及びクロンドサイト(chrondocytes)、ケラチン生成細胞、末梢神経系の神経
細胞、腎臓及び肺の上皮細胞、精巣のセルトリ細胞等が含まれる。0.2ミクロ
ン以上の大きさのサイズを有する粒子に対するターゲットは、血管区画に大きく
制限されるであろう。ここで、ターゲットとなりうる細胞のタイプには、赤血球
、白血球(即ち、単核細胞、マクロファージ、B及びTリンパ球、好中球、ナチ
ュラルキラー細胞、始原細胞、マスト細胞、エオシン好性細胞)、血小板、及び
内皮細胞が含まれる。
皮下注入に対して、ターゲッティングしうる細胞には、結合組織(例えば、繊
維芽細胞、マスト細胞等)、ランゲルハンス細胞、ケラチン生成細胞、及び筋細
胞に存する全ての細胞が含まれる。髄腔内注入に対して、ターゲッティングしう
る細胞には、ニューロン、グリア細胞、星状細胞、及び血液脳関門内皮細胞が含
まれる。腹腔内注入に対して、ターゲッティングしうる細胞には、マクロファー
ジ及び好中球が含まれる。
例
マトリックス材料:ゼラチン(60ブルーム、ブタ皮膚より得たタイプA)、
コンドロイチン4−硫酸、グルタルアルデヒド(25%、グレード1)、1−エ
チル−3−(3−ジメチルアミノプロピル)−カルボジイミド塩酸塩(EDC塩
酸塩)、及び超高純度蔗糖をシグマケミカルCo.(セントルイス、MO)より
購入した。ビオチンLCヒドラジド、及びニュートロアビジン(NeutrAvidin)
、及びクーマッシー(Coomassie)タンパクアッセイ試薬をピアース(Pierce)
(ロックフォード、IL)より得た。セントリコンミクロコンセントレーターを
アミコン(Amicon)(ベヴァリー、MA)より得た。
モノクローナル掟体:mAbPLM−2、BALB/cマウス抗−ブタLFA
−1(IgG1k)(これは、ネズミLFA−1とも交差反応する。)を、先に開
示されたように単離し、精製した(Hildreth,et al.,1989,Monoclonal an-ti
bodies against porcine LFA-1: species cross-reactivity and functionaleff
ects of b-subunit-specific antibodies,Molec.Immunol.,26:883-895)。I
B−4B、ラット抗−マウスLAMP−1腹水液及びマウス抗−ヒトCD44m
Abを先に開示されたように単離した(de Wet,et al.,1987,Firefly lu-cif
erase gene: structure and expression in mammalian cells,Mol.& Cell Bio
l.,7:725-37)。CHAは、何れの公知の in vivo マウスエピトープ(ハイブ
リテック社(hybritech Inc.)サンディエゴ、CA)も認識しないIgG1であ
る。親和性により精製したFITC及びテキサスレッドラベルポリクローナル(
Texas Red-labeled polyclonal)抗−ラットIgGをシグマより得た。
遺伝子:2種類の遺伝子を、本発明の輸送システムの実行可能性を示すために
使用した。LAMP−acDNAは、CMVプロモータを有するインビトロゲン
プラスミド(Invitrogen plasmid)cDNAに挿入されたマウスLAMP−1遺
伝子(2.4kb)を有する6.4kbの環状スーパーコイルプラスミドcDN
Aである(Guaaranieri,et al.,1993,J.Biol.Chem.,268:1941)。LAM
P−1の発現の検出は、抗−LAMP−1mAb及びテキサスレッドと複合体形
成された第二の抗IgGmAbで細胞を染色することによって行った。ルシフェ
ラーゼ酵素をコードする遺伝子は、高感度のアッセイ、その簡便性、及び低コス
ト故に遺伝子発現の研究の細胞生物学で広く使用されている。加えて、該酵素は
、これが酵素活性のための翻訳後のプロセッシングを必要としないサイトソルタ
ンパクであるから、遺伝子発現のよいレポーターである(de Wet,et al.,1987
,Firefly luciferase gene: structure and expression inmammmalian cells,

01)。ルシフェラーゼの存在は、フォトンの生成(化学ルミネッセンスの形態)
に伴う甲虫ルシフェリンの酸化を含む酵素反応によって容易に検出されうる。ア
ッセイは、プロメガ杜(Promega Corp.)(マジソン、WI)から購入したアッ
セイキットを用いて行った。
ミクロスフェアみ今成:ゼラチン−DNAコアセルベートの合成の詳細な系統
図を図1に示した。示された濃度は全て、特に示さない限り67℃にセットした
反応混合物の最終濃度である。アビジンでコートされたゼラチン/プラスミドD
NAミクロ粒子は、まず、42 mM 硫酸ナトリウム(Na2SO4)中のリソソー
ム関連膜タンパク−1(LAMP−1)(6.7Kb、環状スーパーコイル)を
コードするプラスミドDNAの3.5mg/ml 溶液を調製することによって合成し
た。コアセルベーションを、高スピードで1分間ボルテックスしながらDNA/
Na2SO4溶液にゼラチン(5%)を等容量で加えることによって開始した。ゼ
ラチンとのコアセルベーションを開始する前に、DNA/Na2SO4溶液へ直接
に添加することによって医薬と他の薬剤を共にカプセル化した。アビジン(5mg
/ml)を、ミクロスフェア懸濁液に75μgアビジン/ml ミクロスフェア溶液の
最終濃度で加えた。ミクロスフェア混合物を、70%スクロース(w/v)の層
上に層上に重ね、4分間6,000×gで遠心した(ブリンクマンイン
スツルメンツ社(Brinkman Instruments Inc.)、ウエストベリー、NY、モデ
ルL8−75)。蔗糖層から回収されたミクロスフェア画分を水で5倍に希釈し
、次いで、室温で10分間グルタルアルデヒド(12.5 mM 最終濃度)で架橋
した。未反応のグルタルアルデヒドをエタノールアミン(1M)を加え、10分
間クエンチした。ミクロスフェアを上述のように蔗糖遠心によって透析した。
アビジンでコートされカミクロスフェアへのビオチニル化されたmAbの結合
:30 ug の抗体(刊行物に記載された手順に従ってビオチニル化した(O'Shar
messy,et al.,1984,A novel procedure for labeling immunoglobu-lins by
conjugation to oligosaccharides moietys,Immunol.Lett.,8:273-277))を
1ml のアビジンでコートされたミクロスフェア懸濁液(11 mg/ml)に穏やか
に撹拌しながら1時間で加えた。未結合のmAbを透析によってミクロスフェア
から除去した。
ミクロスフェアの特徴づけ及び結合能:DNAを導入したミクロスフェアは、
多形コロイド型を示した。これは、光学顕微鏡による測定で3ミクロン以下の多
分散系の粒子サイズを有していた。精製されたミクロスフェアは、評価し得るほ
どの分解をすることなく少なくとも1カ月間安定であった。LAMP−1プラス
ミドDNAの導入レベルは、20%(w/w)であった。カプセル化効率は、典
型的には>95%であった。1%アガロースゲル電気泳動での遊離のLAMP−
1及び(ミクロスフェアから)遊離されたDNAの可動性を示した(図2)。こ
れはカプセル化したDNAがそのもとの形態で放出されることを示唆している。
ミクロスフェアからのcDNAの放出速度は、架橋密度及び酵素レベルに依存す
る(図4)。数週間までの継続的な放出が容易に得られた。
我々は、組織培養中でのヒト組織球リンパ腫細胞(U937)に結合し、引き
続いてこれにトランスフェクションするようにミクロスフェアを導入したLAM
P−1DNAの能力を試験した。抗−LFA又は抗−CD44モノクローナル抗
体(両タンパクターゲットは、U937細胞表面に多量に発現される。)の何れ
かでコートした場合、LAMP−1を認識する抗体で染色すると、発現LAMP
−1タンパクは3日目まで検出された(図4A、蛍光性の顆粒)。LAMP−1
ミクロスフェアでインキュベートしたU937細胞の染色パターンは、リン酸カ
ルシウム法のトランスフェクションと同様であった(図4B)。アビジン又は非
特異的CHAmAbの何れかでコートされたミクロスフェアは、顆粒状染色パタ
ーンを示さず、未処理細胞と同様であった(図4C)。
フローサイトメトリーを用いて、我々は、LAMP−1の発現が培養物中の5
%のU937細胞まで検出されることを示した。対照--ブランクのミクロスフェ
ア、cDNAを用いるが抗体を全く用いないミクロスフェア、cDNAを用い、
ミスマッチ抗−P−糖タンパク抗体(MRK−Msp.)でコートされたミクロ
スフェア、及びミクロスフェアにトラップされているよりも6倍高い濃度の遊離
のcDNA--のどれも、トランスフェクションの何れの証拠も示さなかった。ト
ランスフェクションの効率は、投与量−応答的であると思われる。一般に、この
個々の遺伝子及び細胞タイプのトランスフェクション効率は、提案されたミクロ
スフェア輸送システム(1〜10%)及びリン酸カルシウム沈殿法(2〜15%
)の間に位置する。図6は、異なったモノクローナル抗体を使用した異なった細
胞タイプにおける概念を示す。再度言うが、cDNAは細胞をトランスフェクシ
ョンしない。結局、LAMP−1発現は幾つかの継代の後に消失する。陽性の結
果はルシフェラーゼレポーター遺伝子システムに対しても得られた。トランスフ
ェクションは、ルシフェラーゼ遺伝子を導入したミクロスフェアとインキュベー
トされたU293細胞でルシフェラーゼ酵素活性の測定によって明確に検出され
た。

Claims (1)
