Abstract
Métodos y sistemas para mejorar la actividad de solubilización del fosfato en un microorganismo. También se proveen microorganismos identificados mediante el uso de los métodos y sistemas de la presente. Los métodos incluyen los pasos, y los sistemas incluyen las instrucciones para identificar una secuencia codificante asociada con la solubilización del fosfato, para modificar la secuencia codificante asociada con la solubilización del fosfato y la relación funcional entre la secuencia codificante modificada y un promotor, e introducir la secuencia codificante alterada asociada con la solubilización de fosfatos bajo el control del promotor en el microorganismo. Las modificaciones en la secuencia codificante puede incluir, por ejemplo, la asignación aleatoria de los codones nativos, la eliminación de secuencias codificantes y la inserción de secuencias reguladoras. Reivindicación 1: Un microorganismo modificado, caracterizado porque comprende una alteración en un gen asociado con la solubilización de fosfatos, en donde dicho gen asociado con la solubilización de fosfatos es nativo para dicho microorganismo, con lo cual dicho microorganismo modificado solubiliza fosfatos con una capacidad o índice mayor en comparación con un microorganismo no modificado de la misma especie. Reivindicación 83: Un método, caracterizado porque comprende: a) proveer una pluralidad de especies microbianas que están asociadas con una planta blanco de interés; b) evaluar la pluralidad de especies microbianas según un parámetro de colonización y la capacidad para solubilizar fosfatos; c) seleccionar una especie microbiana candidato entre la pluralidad de especies microbianas evaluadas; d) caracterizar, en la especie microbiana candidato, un gen seleccionado del grupo que consiste en: una fosfatasa ácida no específica, una fitasa, un gen de la biosíntesis del ácido glucónico, un transportador del ácido glucónico, una gluconato deshidrogenasa y una glucosa deshidrogenasa; e) introducir una o más variaciones genéticas buscadas en la especie microbiana candidato; f) confirmar la integración de dichas una o más variaciones genéticas buscadas en un locus genómico blanco; y g) repetir los pasos d) y e) una o más veces, hasta que la especie microbiana candidato haya adquirido una capacidad mejorada para solubilizar fosfatos.
