Abstract

Además una construccion de ADN para su uso en el procedimiento para obtener insulina aspártica. Reivindicacion 1: Un procedimiento para obtener insulina aspártica caracterizado porque comprende: i) transformar una cepa de levaduras de Pichia pastoris con al menos dos vectores de expresion que posean marcadores de seleccion distintos entre sí y en donde cada vector comprende una construccion de ADN que codifica para an precursor de insulina aspártica de la formula B (1-30) -X1-Y-X2-A (1-21); ii) seleccionar entre las cepas transformadas de acuerdo con la etapa anterior, aquellas cepas recombinantes que sean MUT S y que contengan más de 5 copias de la construccion de ADN integradas al genoma de la levadura; iii) seleccionar entre las cepas recombinantes de la etapa anterior aquella que posea un mayor numero de copias y que tenga un mayor nivel de expresion del precursor de insulina aspártica; iv) fermentar dicha cepa recombinante; v) capturar el precursor de la insulina aspártica del medio de cultivo mediante técnicas cromatográficas, vi) digerir dicho precursor enzimáticamente en condiciones adecuadas corno para que al menos el 70% del precursor de insulina aspártica se convierta en insulina aspártica madura; y vii) purificar la insulina aspártica obtenida a partir de dicha digestion mediante técnicas cromatográficas convencionales.
