Abstract
Un método para detectar una interaccion proteína-proteína, que consiste en: (a) presentar una célula huésped que contiene (i) un primer gen reporterooperativamente ligado a una secuencia de ADN que incluye un primer sitio de union de proteína; (ii) un segundo gen reportero operativamente ligado a unasecuencia de ADN que incluye un segundo sitio de union de proteína; (iii) un primer gen de fusion que expresa una primera proteína de fusion, primera proteínade fusion que incluye una primera proteína ligada en forma covalente a una porcion de ligamiento, que es apta para ligarse específicamente al primer sitio deunion de proteínas; (iv) un segundo gen de fusion que expresa una segunda proteína de fusion, segunda proteína de fusion que incluye una segunda proteínaligada en forma covalente a una porcion de ligamiento,que es apta para ligarse específicamente a dicho segundo sitio de union de proteínas; y (v) un tercer gende fusion que expresa una tercera proteína de fusion, tercera proteína de fusion que incluye una tercera proteína ligada en forma covalente a una porcion deactivacion génica; (b) medir la informacion de salida de la expresion de dicho primer gen reportero como medida de dicha interaccion entre dichas primera ytercera proteínas; (c) medir la informacion de salida de la expresion de dicho segundo gen reportero como medida de dicha interaccion entre dichas segunda ytercera proteínas; y (d) interpretar los resultados de la informacion de salida de la expresion del paso(b) y el paso (c), por lo cual (i) una mayorinformacion de salida tanto de dicho primero como de dicho segundo gen reportero indica que dicha tercera proteína de fusion interactua tanto con dichaprimera como con dicha segunda proteína de fusion; (ii)una mayor informacion de salida de dicho primer gen reportero, pero no así de dicho segundo genreportero indica que dicha tercera proteína de fusion interactua con dicha primera proteína de fusion pero no con dicha segunda proteína de fusion; (iii)unamayor informacion de salida de dicho segundo gen reportero pero no de dicho primer gen reportero indica que dicha tercera proteína de fusion interactua con
