Abstract
Predmetni pronalazak obezbedjuje postupak za prečišćavanje u kome se deferoksamin B, proizveden od strane mikroorganizama i koji je pomešan sa drugim polihidroksamatima stvorenih od strane mikroorganizama, može prevesti u svoju metilsulfonil so, koja je oslobodjena prisustva drugih polihidroksamata, kao i jona hlora. Postupak uključuje adsorbciju i desorbciju deferoksamina B na adsorbcionoj smoli, direktno taloženje deforksamina u vidu slobodne baze iz eluenta iz adsorpcione smole, reakciju deferoksamina u obliku slobodne baze sa metansulfonsom kiselinom i izolovanje deferoksamin B metilsulfonila putem taloženja. Ovaj postupak minimizuje razlaganje deferoksamina B.[The present invention provides a purification process whereby deferoxamine B produced by a microorganism and in mixture with other polyhydroxamates produced by the microorganism may be converted into its mesylate salt substantially free of the other polyhydroxamates and substantially free of chloride ion. The process includes adsorption and desorption of the deferoxamine B on an adsorption resin, direct precipitation of the deferoxamine free base out of the eluent from the adsorption resin, contacting of the deferoxamine B free base with methanesulfonic acid and isolation of the deferoxamine B mesylate sait by precipitation. This process minimizes decomposition of deferoxamine B.
