- Poly(ester amide) is a polymer that contains both ester and amide bonds. These properties give poly(ester amide) the advantages of both polyester and polyamide. Although there have been no successful cases of poly(ester amide) production to date, many studies have reported the production of naturally occurring polyesters known as polyhydroxyalkanoates using metabolic engineering (Choi et al., Adv. Mat. 32: 1-37, 2020). Furthermore, there have been reports of the biosynthesis of poly(lactate-ran-glycolate) and poly(3-hydroxybutyrate-ran-phenyllactate), polyesters that did not exist in nature, using microorganisms through systems metabolic engineering (Choi et al., Nat. Biotech. 34: 435-440, 2016; Yang et al., 9:79, 2018).

Abstract
대사공학적으로 엔지니어링된 재조합 미생물효소 및 이를 이용한폴리(에스터 아마이드)의 제조방법에 관한 것으로, 본 발명의 재조합 미생물 및 이를 이용한 폴리(에스터 아마이드)의 제조방법은 포도당, 아미노산, 하이드록시산, 카르복실산 등과 같은 다양한 탄소원으로부터 다양한 폴리에스터 아마이드폴리(에스터 아마이드) 고분자를 생합성할 수 있으므로들을 제조할 수 있어, 폴리에스터 아마이드폴리(에스터 아마이드)의 산업적 생산에 유용하다.
Description
본 발명은 대사공학적으로 엔지니어링된 재조합 미생물효소 및 이를 이용한 폴리(에스터 아마이드)의 제조방법에 관한 것으로서, 더욱 상세하게는 아미노산을 포함하는 유기산을 기질로 하는 코엔자임 A 전이효소를 코딩하는 유전자 및 PHA 합성효소를 코딩하는 유전자가 도입 또는 증폭되어 있는 재조합 미생물 및 이를 이용한 폴리(에스터 아마이드)의 제조방법에 관한 것이다.
최근 석유 고갈문제와 환경 문제로 인해 미생물을 이용한 지속가능한 다양한 부가가치 화학제품 및 고분자 생산에 큰 이목이 집중되고 있다. 특히 석유화학기반의 플라스틱을 대체하기 위하여 여러 가지 종류의 바이오 고분자들이 생산 연구가 진행되고 있으나, 현재까지 재조합 미생물을 이용한 다양한 폴리(에스터 아마이드)들의 생산 성공 사례는 없다.
폴리(에스터 아마이드)는 에스터 결합과 아마이드 결합을 모두 갖고 있는 고분자이다. 이러한 특성으로 폴리(에스터 아마이드)는 폴리에스터의 장점과 폴리아마이드의 장점을 모두 갖고 있다. 비록 현재까지 폴리(에스터 아마이드) 생산 성공 사례는 없지만, 기존에 대사공학을 이용하여 폴리하이드록시알카노에이트 (Polyhydroxyalkanoate)라 알려진 자연계에 존재하는 폴리에스터를 생산하는 연구는 보고된 바가 많다 (Choi et al., Adv. Mat. 32: 1-37, 2020). 또한 시스템 대사공학을 통하여 미생물을 이용하여 자연계에 존재하지 않았던 폴리에스터인 poly(lactate-ran-glycolate), poly(3-hydroxybutyrate-ran-phenyllactate)의 생합성한 보고가 있다 (Choi et al., Nat. Biotech. 34: 435-440, 2016; Yang et al., 9:79, 2018).
이에, 본 발명자들은 미생물을 이용하여 다양한 탄소원으로부터 폴리(에스터 아마이드)를 효율적으로 생산하는 방법을 개발하고자 노력한 결과, 아미노산(amino acid)을 기질(substrate)로 하여 아미노아실-CoA로 전환하는 효소와 아미노아실-CoA를 고분자화하는 효소의 유전자가 도입된 재조합 미생물을 이용하여 폴리에스터아마이드를 생합성할 수 있다는 것을 확인하고 본 발명을 완성하였다.
발명의 요약
본 발명의 목적은 다양한 탄소원으로부터 폴리(에스터 아마이드)를 제조할 수 있는 재조합 미생물을 제공하는 데 있다.
본 발명의 다른 목적은 상기 재조합 미생물을 이용하여, 폴리(에스터 아마이드)를 제조하는 방법을 제공하는데 있다.
상기 목적을 달성하기 위하여, 본 발명은 코엔자임 A 전이효소(Coenzyme A transferase)를 코딩하는 유전자 및 PHA 합성효소(polyhydroxyalkanoate synthase)를 코딩하는 유전자가 도입 또는 증폭되어 있는, 탄소원으로부터 폴리(에스터 아마이드) 생성능을 가지는 재조합 미생물을 제공한다.
본 발명은 또한, (a) 본 발명의 재조합 미생물을 배양하여 폴리(에스터 아마이드)를 생성하는 단계 및; (b) 상기 생성된 폴리(에스터 아마이드)를 회수하는 단계를 포함하는 폴리(에스터 아마이드)의 제조방법을 제공한다.
본 발명은 또한, (a) 본 발명의 재조합 미생물을 코엔자임 A 전이효소의 기질이 되는 유기산이 첨가된 배지에서 배양하여 폴리(에스터 아마이드)를 생산하는 단계; 및
(b) 상기 생성된 폴리(에스터 아마이드)를 회수하는 단계를 포함하는 폴리(에스터 아마이드)의 제조방법을 제공한다.
본 발명은 또한, 상기 제조방법으로 제조된 폴리(에스터 아마이드)를 제공한다.
도 1은 다양한 폴리(에스터 아마이드)를 생산하는 대사경로와 이러한 폴리(에스터 아마이드)에 포함되어 있는 아미노산 단량체를 나타낸 것이다. 폴리(에스터 아마이드)를 생합성하기 위해 새롭게 디자인된 자연계에 존재하지 않던 대사 반응은 빨간 화살표로 표기하였다. 다른 식으로 정의되지 않는 한, 본 명세서에서 사용된 모든 기술적 및 과학적 용어들은 본 발명이 속하는 기술분야에서 숙련된 전문가에 의해서 통상적으로 이해되는 것과 동일한 의미를 갖는다. 일반적으로, 본 명세서에서 사용된 명명법은 본 기술분야에서 잘 알려져 있고 통상적으로 사용되는 것이다.
본 발명의 일 실시예에서, 다양한 아미노산을 기질로 하는 코엔자임 A 전이효소(coenzyme A transferase)를 코딩하는 유전자 및 PHA 합성효소(polyhydroxyalkanoate synthase)를 코딩하는 유전자가 도입된 재조합 미생물을 사용하여, 다양한 탄소원(예, 아미노산, 당, 유기산 등)으로부터 폴리(에스터 아마이드)를 합성할 수 있음을 확인하였다.
또한, 본 발명의 다른 실시예에서, 글루코스로부터 하이드록시 산 및 아미노산을 합성능을 갖도록 추가적으로 유전자 도입된 재조합 미생물에서 글루코스를 단일 탄소원으로 폴리(에스터 아마이드)를 합성할 수 있음을 확인하였다.
따라서, 본 발명은 일 관점에서, 코엔자임 A 전이효소(Coenzyme A transferase)를 코딩하는 유전자 및 PHA 합성효소(polyhydroxyalkanoate synthase)를 코딩하는 유전자가 도입 또는 증폭되어 있는 폴리(에스터 아마이드)의 합성능을 갖는 재조합 미생물에 관한 것이다.
본 발명에 있어서, 상기 재조합 미생물은 1종 이상의 코엔자임 A 전이효소(Coenzyme A transferase)를 코딩하는 유전자 및 1종 이상의 PHA 합성효소(polyhydroxyalkanoate synthase)를 코딩하는 유전자가 도입된 것을 특징으로 할 수 있다.
본 발명의 일 실시예에서, 상기 재조합 미생물은 아미노산을 포함하는 유기산을 전구체로 하여 이를 단량체로 포함하는 폴리(에스터 아마이드)를 합성할 수 있다.
본 발명의 다른 실시예에서, 상기 재조합 미생물은 아미노산 및 하이드록시산을 전구체로 하여 이들을 포함하는 폴리(에스터 아마이드)를 합성할 수 있다.
본 발명의 재조합 미생물에 도입 또는 증폭된 i) 코엔자임 A 전이효소(Coenzyme A transferase)는 아미노산을 포함하는 유기산을 전구체로 아실-CoA의 합성을 촉매하며, ii) PHA 합성효소는 합성된 아실-CoA의 중합을 통한 폴리(에스터 아마이드)의 합성을 촉매하는 것을 특징으로 할 수 있다.
본 발명의 용어, “코엔자임 A 전이효소(Coenzyme A transferase)”는 아실-CoA 공여체에서 카르복실산으로 코엔자임 A 그룹의 전이 반응을 촉매하는 전이효소를 의미한다.
본 발명에 있어서, 상기 코엔자임 A 전이효소(Coenzyme A transferase)는 아미노산을 기질로 하여, 아미노 아실-CoA의 합성을 촉매하는 것을 특징으로 할 수 있다. 본 발명에 있어서, 상기 아미노산은 단백질을 구성하는 단백질 구성 아미노산 및 비단백 아미노산을 모두 포함하는 의미로 사용된다.
본 발명에 있어서, 상기 단백질 구성 아미노산은 예를 들어 진핵 생물에서 발견되는 21종의 천연 아미노산 또는 이의 이성질체 일 수 있으나, 이에 제한되는 것은 아니다.

본 발명에 있어서, 앞서 재조합 미생물과 관련하여 설명한 것과 같이 숙주 미생물이 선천적으로, 또는 추가적인 유전자 엔지니어링을 통해 본 발명의 코엔자임 A 전이요소의 기질이되는 유기산 (예, 하이드록시산 또는 아미노산)의 생합성 경로를 갖는 경우, 단일 탄소원 첨가 또는 외부로부터 별도의 탄소원 첨가 없이도 본 발명의 재조합 미생물의 배양을 통해 폴리(에스터 아마이드)를 생산할 수 있다.
본 발명은 또 다른 관점에서. 다음의 단계를 포함하는 폴리(에스터 아마이드)의 제조방법에 관한 것이다:
(a) 본 발명의 재조합 미생물을 코엔자임 A 전이효소의 기질이 되는 유기산이 첨가된 배지에서 배양하여 폴리(에스터 아마이드)를 생산하는 단계; 및
(b) 상기 생성된 폴리(에스터 아마이드)를 회수하는 단계.
본 발명에 있어서, 재조합 미생물의 배양과정은 통상적으로 알려진 배양방법을 사용하여 수행될 수 있고, 본 발명의 실시예에서 사용된 특정 배지 및 특정 배양방법 이외에도 유청(whey), CSL(corn steep liquor) 등의 당화액과 다른 배지를 사용할 수 있고, 유가배양(fed-batch culture), 연속배양 등 다양한 방법을 사용할 수 있다(Lee et al., Bioprocess Biosyst. Eng., 26: 63, 2003; Lee et al., Appl. Microbiol. Biotechnol., 58: 663, 2002; Lee et al., Biotechnol. Lett., 25: 111, 2003; Lee et al., Appl. Microbiol. Biotechnol., 54: 23, 2000; Lee et al., Biotechnol. Bioeng., 72: 41, 2001).
본 발명의 용어 “배양”은 미생물을 배양하여, 목적하고자 하는 효능을 이끌어 내는 것을 의미하며, 본 발명에서 목적하고자 하는 효능은 소수성 물질의 생산을 의미한다. 상기 배양은 사용되는 미생물에 최적화되고 적어도 1종의 단순 에너지 공급원 및 필요한 경우에 공동-기질을 함유하는 적절한 배양 배지를 갖는 배양기에서 일반적으로 수행될 수 있다.
본 발명에 있어서, 상기 “배양”은 미생물 배양을 통한 소수성 물질의 생산의 의미에서, “발효”와 상호호환적으로 사용될 수 있다.
본 발명에 있어서, 상기 재조합 미생물을 배양하여 소수성 물질을 생산하는 단계는 유가식 발효를 통해 수행되는 것을 특징으로 할 수 있다.
본 발명의 용어 “유가식 배양” 또는 “유가식 발효”는 배지를 간헐적으로 추가 공급하여 배양액의 농도를 제어하여 배양/발효 시키는 것을 의미한다.
"적절한 배양 배지"는 탄소 공급원 또는 탄소 기질, 질소 공급원, 예를 들어 펩톤, 포도당, 글리세롤, 효모 추출물, 고기 추출물, 맥아 추출물, 우레아, 황산암모늄, 염화암모늄, 질산암모늄 및 인산암모늄; 인 공급원, 예를 들어 인산일칼륨 또는 인산이칼륨; 미량 원소 (예를 들어, 금속 염), 예를 들어 마그네슘 염, 코발트 염 및/또는 망가니즈 염; 뿐만 아니라 성장 인자, 예컨대 아미노산 및 비타민과 같이, 세포의 유지 및/또는 성장에 필수적이거나 유익한 영양소; 효모 추출물, 항생제 등을 포함할 수 있다.본 발명에 있어서, 상기 배양은 소수성 물질을 생산하기 위해 적절한 범위의 온도에서 배양되는 것을 특징으로 할 수 있으며, 예를 들어20 ℃ 내지 40 ℃, 바람직하게는 25 ℃ 내지 35 ℃에서 배양되는 것을 특징으로 할 수 있으나, 이에 제한되는 것은 아니며, 숙주 미생물의 종, 발현하고자 하는 물질 등에 따라 상이한 조건으로 배양될 수 있다.
본 발명에서, 용어 "벡터 (vector)"는 적합한 숙주 내에서 DNA를 발현시킬 수 있는 적합한 조절 서열에 작동가능하게 연결된 DNA 서열을 함유하는 DNA 제조물을 의미한다. 벡터는 플라스미드, 파지 입자, 또는 간단하게 잠재적 게놈 삽입물일 수 있다. 적당한 숙주로 형질전환 되면, 벡터는 숙주 게놈과 무관하게 복제하고 기능할 수 있거나, 또는 일부 경우에 게놈 그 자체에 통합될 수 있다. 플라스미드가 현재 벡터의 가장 통상적으로 사용되는 형태이므로, 본 발명의 명세서에서 "플라스미드 (plasmid)" 및 "벡터 (vector)"는 때로 상호 교환적으로 사용된다. 그러나, 본 발명은 당업계에 알려진 또는 알려지게 되는 바와 동등한 기능을 갖는 벡터의 다른 형태를 포함한다. 포유동물 세포 배양물 발현을 위한 전형적인 발현 벡터는 예를 들면 pRK5 (EP 307,247호), pSV16B (WO 91/08291호) 및 pVL1392 (Pharmingen)을 기초로 한다.
"발현 조절 서열 (expression control sequence)"이라는 표현은 특정한 숙주 생물에서 작동가능하게 연결된 코딩 서열의 발현에 필수적인 DNA 서열을 의미한다. 그러한 조절 서열은 전사를 실시하기 위한 프로모터, 그러한 전사를 조절하기 위한 임의의 오퍼레이터 서열, 적합한 mRNA 리보좀 결합 부위를 코딩하는 서열 및 전사 및 해독의 종결을 조절하는 서열을 포함한다. 예를 들면, 원핵생물에 적합한 조절 서열은 프로모터, 임의로 오퍼레이터 서열 및 리보좀 결합 부위를 포함한다. 진핵세포는 프로모터, 폴리아데닐화 시그날 및 인핸서가 이에 포함된다. 플라스미드에서 유전자의 발현 양에 가장 영향을 미치는 인자는 프로모터이다. 고 발현용의 프로모터로서 SRα 프로모터와 사이토메가로바이러스 (cytomegalovirus) 유래 프로모터 등이 바람직하게 사용된다.
본 발명의 DNA 서열을 발현시키기 위하여, 매우 다양한 발현 조절 서열 중 어느 것이라도 벡터에 사용될 수 있다. 유용한 발현 조절서열의 예에는, 예를 들어, SV40 또는 아데노바이러스의 초기 및 후기 프로모터들, lac 시스템, trp 시스템, TAC 또는 TRC 시스템, T3 및 T7 프로모터들, 파지 람다의 주요 오퍼레이터 및 프로모터 영역, fd 코드 단백질의 조절 영역, 3-포스포글리세레이트 키나제 또는 다른 글리콜분해 효소에 대한 프로모터, 상기 포스파타제의 프로모터들, 예를 들어 Pho5, 효모 알파-교배 시스템의 프로모터 및 원핵세포 또는 진핵 세포 또는 이들의 바이러스의 유전자의 발현을 조절하는 것으로 알려진 구성과 유도의 기타 다른 서열 및 이들의 여러 조합이 포함된다. T7 RNA 폴리메라아제 프로모터 Φ10은 이. 콜라이에서 단백질 NSP를 발현시키는데 유용하게 사용될 수 있다.
핵산은 다른 핵산 서열과 기능적 관계로 배치될 때 "작동가능하게 연결 (operably linked)"된다. 이것은 적절한 분자 (예를 들면, 전사 활성화 단백질)은 조절 서열(들)에 결합될 때 유전자 발현을 가능하게 하는 방식으로 연결된 유전자 및 조절 서열(들)일 수 있다. 예를 들면, 전서열 (pre-sequence) 또는 분비 리더 (leader)에 대한 DNA는 폴리펩타이드의 분비에 참여하는 전단백질로서 발현되는 경우 폴리펩타이드에 대한 DNA에 작동가능하게 연결되고; 프로모터 또는 인핸서는 서열의 전사에 영향을 끼치는 경우 코딩서열에 작동가능하게 연결되거나; 또는 리보좀 결합 부위는 서열의 전사에 영향을 끼치는 경우 코딩 서열에 작동가능하게 연결되거나; 또는 리보좀 결합 부위는 번역을 용이하게 하도록 배치되는 경우 코딩 서열에 작동가능하게 연결된다. 일반적으로, "작동가능하게 연결된"은 연결된 DNA 서열이 접촉하고, 또한 분비 리더의 경우 접촉하고 리딩 프레임 내에 존재하는것을 의미한다. 그러나, 인핸서 (enhancer)는 접촉할 필요가 없다. 이들 서열의 연결은 편리한 제한 효소 부위에서 라이게이션(연결)에 의해 수행된다. 그러한 부위가 존재하지 않는 경우, 통상의 방법에 따른 합성 올리고뉴클레오티드 어댑터 (oligonucleotide adaptor) 또는 링커(linker)를 사용한다.
본원 명세서에 사용된 용어 "발현 벡터"는 통상 이종의 DNA의 단편이 삽입된 재조합 캐리어 (recombinant carrier)로서 일반적으로 이중 가닥의 DNA의 단편을 의미한다. 여기서, 이종 DNA는 숙주 세포에서 천연적으로 발견되지 않는 DNA인 이형 DNA를 의미한다. 발현 벡터는 일단 숙주 세포내에 있으면 숙주 염색체 DNA와 무관하게 복제할 수 있으며 벡터의 수 개의 카피 및 그의 삽입된 (이종) DNA가 생성될 수 있다.
당업계에 주지된 바와 같이, 숙주세포에서 형질감염 유전자의 발현 수준을 높이기 위해서는, 해당 유전자가, 선택된 발현 숙주 내에서 기능을 발휘하는 전사 및 해독 발현 조절 서열에 작동가능 하도록 연결되어야만 한다. 바람직하게는 발현 조절서열 및 해당 유전자는 세균 선택 마커 및 복제 개시점 (replication origin)을 같이 포함하고 있는 하나의 발현 벡터 내에 포함되게 된다. 발현 숙주가 진핵세포인 경우에는, 발현 벡터는 진핵 발현 숙주 내에서 유용한 발현 마커를 더 포함하여야만 한다.
상술한 발현 벡터에 의해 형질전환 또는 형질 감염된 숙주 세포는 본 발명의 또 다른 측면을 구성한다. 본원 명세서에 사용된 용어 "형질전환"은 DNA를 숙주로 도입하여 DNA가 염색체외 인자로서 또는 염색체 통합완성에 의해 복제가능하게 되는 것을 의미한다. 본원 명세서에 사용된 용어 "형질감염"은 임의의 코딩 서열이 실제로 발현되든 아니든 발현 벡터가 숙주 세포에 의해 수용되는 것을 의미한다.
발명의 숙주 세포는 원핵 또는 진핵생물 세포일 수 있다. 또한, DNA의 도입효율이 높고, 도입된 DNA의 발현효율이 높은 숙주가 통상 사용된다. 이. 콜라이, 슈도모나스, 바실러스, 스트렙토마이세스, 진균, 효모와 같은 주지의 진핵 및 원핵 숙주들, 스포도프테라 프루기페르다(SF9)와 같은 곤충 세포, CHO 및 생쥐 세포같은 동물 세포, COS 1, COS 7, BSC 1, BSC 40 및 BMT 10과 같은 아프리카 그린 원숭이 세포, 및 조직배양된 인간 세포는 사용될 수 있는 숙주 세포의 예이다. 본 발명의 NSP 단백질을 코딩하는 cDNA를 클로닝할 때에는 동물세포를 숙주로 하는 것이 바람직하다. COS 세포를 이용하는 경우에는 COS 세포에서 SV40 라지 T안티겐(large T antigen)이 발현하고 있으므로 SV40의 복제개시점을 갖는 플라스미드는 세포중에서 다수 카피(copy)의 에피솜(episome)으로 존재하도록 되고 통상보다 고 발현이 기대될 수 있다. 도입된 DNA 서열은 숙주 세포와 동일한 종으로부터 얻을 수 있거나, 숙주 세포와 다른 종의 것일 수 있거나, 또는 그것은 어떠한 이종 또는 상동성 DNA를 포함하는 하이브리드 DNA 서열일 수 있다.
물론 모든 벡터와 발현 조절 서열이 본 발명의 DNA 서열을 발현하는데 모두 동등하게 기능을 발휘하지는 않는다는 것을 이해하여야만 한다. 마찬가지로 모든 숙주가 동일한 발현 시스템에 대해 동일하게 기능을 발휘하지는 않는다. 그러나 당업자라면 과도한 실험적 부담 없이 본 발명의 범위를 벗어나지 않는 채로 여러 벡터, 발현 조절 서열 및 숙주 중에서 적절한 선택을 할 수 있다. 예를 들어, 벡터를 선택함에 있어서는 숙주를 고려하여야 하는데, 이는 벡터가 그 안에서 복제되어야만 하기 때문이다. 벡터의 복제 수, 복제 수를 조절할 수 있는 능력 및 당해 벡터에 의해 코딩되는 다른 단백질, 예를 들어 항생제 마커의 발현도 또한 고려되어야만 한다. 발현 조절 서열을 선정함에 있어서도, 여러 가지 인자들을 고려하여야만 한다. 예를 들어, 서열의 상대적 강도, 조절가능성 및 본 발명의 DNA 서열과의 상용성 등, 특히 가능성있는 이차 구조와 관련하여 고려하여야 한다. 단세포 숙주는 선정된 벡터, 본 발명의 DNA 서열에 의해 코딩되는 산물의 독성, 분비 특성, 단백질을 정확하게 폴딩시킬 수 있는 능력, 배양 및 발효 요건들, 본 발명 DNA 서열에 의해 코딩되는 산물을 숙주로부터 정제하는 것의 용이성 등의 인자를 고려하여 선정되어야만 한다. 이들 변수의 범위 내에서, 당업자는 본 발명의 DNA 서열을 발효 또는 대규모 동물 배양에서 발현시킬 수 있는 각종 벡터/발현 조절 서열/숙주 조합을 선정할 수 있다. 발현 클로닝에 의해 NSP 단백질의 cDNA를 클로닝 하려고 할 때의 스크리닝법으로서 바인딩법(binding법), 페닝법(panning법), 필름에멀션법(film emulsion 법)등이 적용될 수 있다.
본 발명의 정의상 "실질적으로 순수한"이란 본 발명에 따른 폴리펩타이드 및 폴리펩타이드를 코딩하는 DNA 서열이 박테리아로부터 유래된 다른 단백질을 실질적으로 포함하지 않는 것을 의미한다.
재조합 단백질을 발현하기 위한 숙주세포는 짧은 시간 내 고농도 균체 배양이 가능하고 유전자 조작이 용이하고 유전학적, 생리적 특징이 잘 밝혀져 있는 대장균 (Escherichia coli), 고초균 (Bacillus subtillis) 등과 같은 원핵세포가 널리 이용되어 왔다. 그러나, 단백질의 번역 후 수식 (post-translational modification), 분비과정 및 활성형의 3차원 구조, 단백질의 활성상태의 문제점들을 해결하기 위해 최근 단세포 진핵세포인 효모 계열 (Pichia pastoris, Saccharomyces cerevisiae, Hansenula polymorpha 등), 사상성 진균류 (filamentous fungi), 곤충세포 (insect cells), 식물세포, 포유동물세포 (mammalian cells) 등 고등생물에 이르기까지 재조합 단백질 생산의 숙주세포로 활용하고 있으므로, 실시예에서 예시된 대장균뿐만 아니라 다른 숙주세포를 이용하는 것은 당업계의 통상의 지식을 가진 자에게 있어서 용이하게 적용 가능하다.
본 발명은 또 다른 관점에서, 상기 방법으로 제조된 폴리(에스터 아마이드)에 관한 것이다.
[실시예]
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: 유전자 엔지니어링을 위한 백터의 제작
실시예 1-1: pTac15k_davAB 백터
pKE112_davAB 백터(Park et al., Metab. Eng. 16:42-47 2013)를 DNA를 주형으로 하고, 서열번호 14 및 15의 프라이머로 PCR을 수행하여, 5-aminovaleramide aminohydrolase와 L-lysine monooxygenase를 코딩하는 davAB 유전자 절편을 제작하였다.

실시예 1-2: pTac15k_act_davAB 백터
pKE112_davAB 백터(Park et al., Metab. Eng. 16:42-47 2013)를 DNA를 주형으로 하고, 서열번호 16 및 17의 프라이머로 PCR을 수행하여, 5-aminovaleramide aminohydrolase와 L-lysine monooxygenase를 코딩하는 davAB 유전자 절편을 제작하였다.

실시예 1-3: pTac15k_act_panD 백터
코리네박테리움 글루타미쿰 (Corynebacterium glutamicum) 균주의 염색체 DNA를 주형으로 하고, 서열번호 18과 19의 프라이머로 PCR을 수행하여, aspartate decarboxylase를 코딩하는 panD 유전자 절편을 제작하였다.

실시예 1-4: pPs619C1437PhaAB 백터
큐프리아비더스 네케터 (Cupriavidus necator) 균주의 염색체 DNA를 주형으로 하고, 서열번호 20과 21의 프라이머로 PCR을 수행하여, acetyl-CoA acetyltransferase와 acetoacetyl-CoA reductase를 코딩하는 phaAB 유전자 절편을 제작하였다.

실시예 1-5: pCneCPct540 백터
큐프리아비더스 네케터 (Cupriavidus necator) 균주의 염색체 DNA를 주형으로 하고, 서열번호 22와 23의 프라이머로 PCR을 수행하여, polyhydroxyalkanoate synthase를 코딩하는 phaCCne 유전자 절편을 제작하였다.
다음으로, 상기 phaCCne 절편과 pPs619C1437Pct540 플라스미드(Jung et al., Biotechnol. Bioeng., 105: 161- 171, 2010)에 제한효소(BstBI 및 SbfI)를 처리한 후, T4 DNA 라이게이즈를 처리하여, 제한효소로 절단된 phaCCne절편 및 pPs619C1437Pct540 플라스미드를 접합시킴으로써, 재조합 플라스미드인 pCneCPct540를 제작하였다.
실시예 1-6: pAcaCPct540, pAhyCPct540, pAsaCPct540 백터
에어로모나스 케비에 (Aeromonas caviae), 에어로모나스 하이드로필리아 (Aeromonas hydrophila), 에어로모나스 살모니시다 (Aeromonas salmonicida) 균주의 염색체 DNA를 주형으로 하고, 서열번호 24과 25의 프라이머로 PCR을 수행하여, polyhydroxyalkanoate synthase를 코딩하는 phaCAca, phaCAhy, phaCAsa 유전자 절편을 제작하였다.
다음으로, 상기 phaCAca, phaCAhy, phaCAsa 유전자 절편과 pPs619C1437Pct540 플라스미드(Jung et al., Biotechnol. Bioeng., 105: 161- 171, 2010)에 제한효소(BstBI 및 SbfI)를 처리한 후, T4 DNA 라이게이즈를 처리하여, 제한효소로 절단된 phaCAca, phaCAhy, phaCAsa 유전자 절편 및 pPs619C1437Pct540 플라스미드를 접합시킴으로써, 재조합 플라스미드인 pAcaCPct540, pAhyCPct540, pAsaCPct540를 제작하였다.
실시예 1-1 내지 1-6에서 제작된 벡터의 정보는 아래 표 1과 같다.
실시예 2: 재조합 미생물 제작
실시예 1에서 제조된 벡터를 사용하여 아래 표 2의 재조합 미생물들을 제작하였다.
미생물 내에서 beta-alanine coenzyme A transferase를 코딩하는 act 유전자가 발현이 되도록 제작한 pTac15k_act 플라스미드(Chae et al., Metab. Eng. 41: 82-91, 2017)와 엔지니어링 된 polyhydroxyalkanoate synthase를 코딩하는 phaC1437Ps6-19 유전자와 엔지니어링된 propionate coenzyme A transferase를 코딩하는 pct540 유전자가 발현이 되로록 제작한 pPs619C1437Pct540 플라스미드 (Jung et al., Biotechnol. Bioeng., 105: 161- 171, 2010)를 대장균 XL1-Blue (Stratagene)에 도입하여 재조합 미생물을 제조하였으며(PEA01), 공백터인 pTac15k가 도입된 대장균 (PEA00)을 대조군 균주로 사용하였다.
또한, 다양한 탄소원에서의 생산 가능성을 확인하기 위해, 미생물 내에서 beta-alanine coenzyme A transferase를 코딩하는 act 유전자, 5-aminovaleramide aminohydrolase를 코딩하는 davA 유전자, L-lysine monooxygenase를 코딩하는 davB 유전자가 발현이 되도록 실시예 1-2에서 제작된 pTac15k_act_davAB 백터를 대장균 XL1-Blue/pPs619C1437Pct540에 도입하여 재조합 미생물을 제조하였으며(PEA03), 실시예 1-1에서 제작된 pTac15k_davAB가 도입된 대장균 (PEA02)를 대조군 균주로 사용하였다.
또한, 포도당을 유일한 탄소원으로 이용한 생산 가능성 및 유가식 배양 발효를 통한 고효율 생산을 확인하기 위해, 엔지니어링 된 polyhydroxyalkanoate synthase를 코딩하는 phaC1437Ps6-19 유전자, acetyl-CoA acetyltransferase를 코딩하는 phaA 유전자와 acetoacetyl-CoA reductase를 코딩하는 phaB 유전자가 발현이 되도록 실시예 1에서 제작한 pPs619C1437PhaAB 플라스미드를 대장균 XL1-Blue ΔldhA에 도입하여 재조합 미생물을 제조하였으며(XL1-Blue ΔldhA/pPs619C1437PhaAB), 해당 균주에 beta-alanine coenzyme A transferase를 코딩하는 act 유전자와 aspartate decarboxylase를 코딩하는 panD 유전자가 발현이 되도록 실시예 1에서 제작된 pTac15k_act_panD 백터 혹은 beta-alanine coenzyme A transferase를 코딩하는 act 유전자와 엔지니어링 된 glutamate decarboxylase를 코딩하는 gadB_mut 유전자가 발현이 되도록 제작된 pTac15k_act_gadB_mut(Chae et al., Metab. Eng. 41: 82-91, 2017)가 도입된 대장균 (PEA06와 PEA07) 균주를 사용하였다. 해당 균주들의 대조군으로 XL1-Blue ΔldhA/pPs619C1437PhaAB 균주에 panD 혹은 gadB_mut 유전자를 발현하지 않는 pTac15k_act (Chae et al., Metab. Eng. 41: 82-91, 2017)가 도입된 대장균 (PEA05) 균주를 대조군으로 사용하였다.
또한, 다양한 미생물 유래의 polyhydroxyalkanoate synthase의 활용 가능성을 확인하기 위하여, 큐프리아비더스 네케터 (Cupriavidus necator), 에어로모나스 케비에 (Aeromonas caviae), 에어로모나스 하이드로필리아 (Aeromonas hydrophila), 에어로모나스 살모니시다 (Aeromonas salmonicida), 슈도모나스 MBEL 6-19 (Pseudomonas sp. MBEL 6-19), 슈도모나스 61-3 (Pseudomonas sp. 61-3), 슈도모나스 푸티다 KT2440 (Pseudomonas putida KT2440), 슈도모나스 아르지노사 PAO1 (Pseudomonas aeruginosa PAO1), 슈도모나스 클로로라피스 (Pseudomonas chlororaphis), 슈도모나스 레시노보렌스 (Pseudomonas resinovorans)유래의 polyhydroxyalkanoate synthase를 코딩하는 phaC 유전자를 발현이되도록 실시예 1에서 구축한 pCneCPct540, pAcaCPct540, pAhyCPct540, pAsaCPct540 백터들과 앞서 보고된 pPs619C1437Pct540, pPs613C1437Pct540, pPpuC1437Pct540, pPaeC1437Pct540, pPchC1437Pct540, pPreC1437Pct540 백터들(Yang et al., Appl. Microbiol. Biotechnol. 90: 603-614, 2011)을 각각 대장균 XL1-Blue ΔldhA/pTac15k_act에 도입하여 재조합 미생물을 제조하였다 (PEA08, PEA09, PEA10, PEA11, PEA04, PEA12, PEA13, PEA14, PEA15, PEA16)
실시예 3: 재조합 미생물을 이용한 3-아미노프로피오네이트, 4-아미노뷰티레이트를 단량체로 포함한 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA00, PEA01)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 MR배지에 접종하여 배양하였다.
MR배지 (pH 7.0)의 조성은 증류수 1리터당 20g 포도당, 6.67g KH2PO4, 4.0g (NH4)2HPO4, 0.8g citric acid, 0.8g MgSO4ㆍ7H2O, 0.01g CaCl2ㆍ2H20, 5mL trace metal solution (증류수 1리터 당 10 g FeSO4ㆍ7H2O, 2.2g ZnSO4ㆍ4H2O, 0.58g MnSO4ㆍ4H2O, 1g CuSO4ㆍ5H2O, 0.1g (NH4)6Mo7O24ㆍ4H2O, 0.02g Na2B4O7ㆍ10H2O)의 성분으로 구성된 배지이다. 상기 조성에서 기존 beta-alanine coenzyme A transferase의 기질로 알려진 아미노산들인 3-아미노프로피오네이트, 4-아미노뷰티레이트, 5-아미노발러레이트, 6-아미노카프로에이트, 7-아미노헵타노에이트를 2g/L 농도로 추가 탄소원으로 공급하였다. 또한 2 g/L의 sodium (RS)-3-하이드록이뷰티레이트 역시 추가 공급하였다. 배양은 96시간동안 30℃와 200rpm으로 작동하는 shaking incubator(jSR, 한국)에서 진행하였다. 배양이 끝난, 배양액은 13,200 rpm에서 10분간 원심분리하여, 세포를 채취하여 분석을 수행함으로써, 폴리에스터 아미이드 생산을 확인하였다.
그 결과, 도 2에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 3-아미노프로피오네이트와 4-아미노뷰티레이트를 단량체로 갖고있는 폴리(에스터 아마이드)를 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 3-아미노프로피오네이트와 4-아미노뷰티레이트를 단량체로 갖고있는 폴리(에스터 아마이드)를 생성함을 확인하였다.
실시예 4: 재조합 미생물을 이용한 5-아미노발러레이트를 단량체로 포함한 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA02, PEA03)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 MR배지에 접종하여 배양하였다.
MR배지 (pH 7.0)의 조성은 증류수 1리터당 20g 포도당, 6.67g KH2PO4, 4.0g (NH4)2HPO4, 0.8g citric acid, 0.8g MgSO4ㆍ7H2O, 0.01g CaCl2ㆍ2H20, 5mL trace metal solution (증류수 1리터 당 10 g FeSO4ㆍ7H2O, 2.2g ZnSO4ㆍ4H2O, 0.58g MnSO4ㆍ4H2O, 1g CuSO4ㆍ5H2O, 0.1g (NH4)6Mo7O24ㆍ4H2O, 0.02g Na2B4O7ㆍ10H2O)의 성분으로 구성된 배지이다. 상기 조성에서 davAB 유전자로 인해 5-아미노발러레이트로 전환될 수 있는 L-라이신을 2g/L 농도로 추가 탄소원으로 공급하였다. 또한 2 g/L의 sodium (RS)-3-하이드록이뷰티레이트 역시 추가 공급하였다. 배양은 96시간동안 30℃와 200rpm으로 작동하는 shaking incubator(jSR, 한국)에서 진행하였다. 배양이 끝난, 배양액은 13,200 rpm에서 10분간 원심분리하여, 세포를 채취하여 분석을 수행함으로써, 폴리 에스터 아미이드 생산을 확인하였다.
그 결과, 도 2에 개시된 바와 같이, 본 발명에 따른 재조합 미생물은 L-라이신을 탄소원으로 사용하여 5-아미노발러레이트를 단량체로 가지는 폴리(에스터 아마이드)를 생산하는 것을 확인하였다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 5-아미노발러레이트를 단량체로 가지는 폴리(에스터 아마이드)를 성공적으로 생성함을 확인하였다.
실시예 5: beta-alanine coenzyme A transferase의 추가 기질 확인을 위한 in vitro assay
폴리(에스터 아마이드)의 아미노산 단량체 종류들 더 넓히기 위하여 기존 알려진 기질 외에 beta-alanine coenzyme A transferase가 활성을 갖고 있는 기질을 찾기 위해 in vitro assay를 진행하였다. beta-alanine coenzyme A transferase의 정제를 위해서 pETa_his_act 백터(Chae et al., Metab. Eng. 41: 82-91, 2017)를 대장균 BL21(DE3)(F- ompT hsdSB(rB- mB-) gal dcm (DE3) a prophage carrying the T7 RNA polymerase gene) (New England Biolabs, 미국)에 도입하였다.
Enzyme assay는 50mM potassium phosphate buffer (pH 7.5)에서 진행하였다. Enzyme assay에 필요한 기질 및 효소는 아래와 같이 양으로 넣어주었다. 10mM의 타겟 기질, 1mM의 acetyl-CoA 그리고 2.5μg의 정제된 beta-alanine coenzyme A transferase를 첨가하여 2시간동안 30℃에서 반응을 진행하였다.
그 결과, 도 4에 개시된 바와 같이, 기존 알려진 3-아미노프로피오네이트, 4-아미노뷰티레이트, 5-아미노발러레이트, 6-아미노카프로에이트, 7-아미노헵타노에니트 이외에 아미노산 기질인 (RS)-3-아미노뷰티레이트, 3-아미노아이소뷰티레이트, (RS)-3-아미노발러레이트에도 활성을 갖고 있음을 확인하였다. 또한 아미노산이 아닌 다섯가지의 하이드록시산 (글라이콜레이트, (RS)-락테이트, (RS)-2-하이드록시뷰티레이트, 3-하이드록시프로피오네이트, (RS)-3-하이드록시뷰티레이트) 및 일곱가지의 카르복실산 (프로피오네이트, 뷰티레이트, 아이소뷰티레이트, 발러레이트, 아이소발러레이트, 2-메틸뷰티레이트, 카프로에이트)에도 활성이 있음을 확인하였다.
이 결과로부터, 본 발명에 따라 beta-alanine coenzyme A transferase가 성공적으로 기존 알려진 기질 이외에 여러 가지 아미노산, 하이드록시산 및 카르복실산을 coenzyme A 유도체로 전환시키는 것을 확인하였다.
실시예 6: 재조합 미생물을 이용한 (R)-3-아미노뷰티레이트, 3-아미노아이소뷰티레이트, (R)-3-아미노발러레이트를 단량체로 포함한 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA00, PEA01)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 MR배지에 접종하여 배양하였다.

그 결과, 도 2에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 (R)-3-아미노뷰티레이트, 3-아미노아이소뷰티레이트, (R)-3-아미노발러레이트를 단량체로 갖고있는 폴리(에스터 아마이드)를 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 (R)-3-아미노뷰티레이트, 3-아미노아이소뷰티레이트, (R)-3-아미노발러레이트를 단량체로 갖고있는 폴리(에스터 아마이드)를 생성함을 확인하였다.
실시예 7: 재조합 미생물을 이용한 여러가지 아미노산을 단량체로 포함한 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA00, PEA01)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 MR배지에 접종하여 배양하였다.

그 결과, 도 3에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 두가지 (3-아미노프로피오네이트와 (R)-3-아미노뷰티레이트) 및 세가지 (3-아미노프로피오네이트, (R)-3-아미노뷰티레이트와 (R)-3-아미노아이소뷰티레이트)를 통시에 단량체로 갖고있는 폴리(에스터 아마이드)를 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 여러가지 아미노산을 단량체로 갖고 있는 폴리(에스터 아마이드)를 생성함을 확인하였다.
실시예 8: 재조합 미생물을 이용한 포도당을 유일한 탄소원으로 사용한 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA05, PEA06 혹은 PEA07)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 MR배지에 접종하여 배양하였다. PEA06 균주와 PEA07균주는 각각 3-아미노프로피오네트와 4-아미노뷰티레이트를 생합성하는 대사흐름을 증가시키기 위해 panD와 gadB_mut 유전자가 과발현된 균주이다. 또한 두 균주는 포도당으로 부터 3-하이드록시뷰티릴 코엔자임 A의 생합성이 가능하게 만들어 외부 하이드록시산의 도입 없이도 하이드록시산 단량체를 제공하는 것을 가능하게 하기 위해 phaAB 유전자가 도입되어 있다. 또한 목표로한 3-하이드록시뷰티레이트 이외에 락테이트가 하이드록시산 단량체로 고분자화 되는 것을 방지하기 위하여 락테이트 생합성에 필요한 ldhA유전자가 결실된 균주를 사용하였다.
MR배지 (pH 7.0)의 조성은 증류수 1리터당 20g 포도당, 6.67g KH2PO4, 4.0g (NH4)2HPO4, 0.8g citric acid, 0.8g MgSO4ㆍ7H2O, 0.01g CaCl2ㆍ2H20, 5mL trace metal solution (증류수 1리터 당 10 g FeSO4ㆍ7H2O, 2.2g ZnSO4ㆍ4H2O, 0.58g MnSO4ㆍ4H2O, 1g CuSO4ㆍ5H2O, 0.1g (NH4)6Mo7O24ㆍ4H2O, 0.02g Na2B4O7ㆍ10H2O)의 성분으로 구성된 배지이다. 상기 조성에서 앞 실시예들과 다르게 어떠한 아미노산 및 sodium (RS)-3-하이드록시뷰티레이트를 추가로 공급하지 않아 포도당이 유일한 탄소원으로 이용될 수 있도록 하였다. 배양은 96시간동안 30℃와 200rpm으로 작동하는 shaking incubator(jSR, 한국)에서 진행하였다. 배양이 끝난, 배양액은 13,200 rpm에서 10분간 원심분리하여, 세포를 채취하여 분석을 수행함으로써, 폴리에스터 아미이드 생산을 확인하였다.
그 결과, 도 5에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 3-아미노프로피오네티트(도 5a,b) 혹은 4-아미노뷰티레이트(도 5c,d)를 단량체로 갖고있는 폴리(에스터 아마이드)를 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 포도당을 유일한 탄소원으로 이용하여 폴리(에스터 아마이드)를 생성함을 확인하였다.
실시예 9: 재조합 미생물의 유가식 배양 발효를 이용한 3-아미노프르피오네이트를 단량체로 갖는 폴리(에스터 아마이드)의 고효율 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA06)을 200mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 200mL을 6.6L 발효기에에 2L의 MR배지에 접종하여 유가식 배양 발효를 하였다.
MR배지 (pH 7.0)의 조성은 증류수 1리터당 20g 포도당, 6.67g KH2PO4, 4.0g (NH4)2HPO4, 0.8g citric acid, 0.8g MgSO4ㆍ7H2O, 0.01g CaCl2ㆍ2H20, 5mL trace metal solution (증류수 1리터 당 10 g FeSO4ㆍ7H2O, 2.2g ZnSO4ㆍ4H2O, 0.58g MnSO4ㆍ4H2O, 1g CuSO4ㆍ5H2O, 0.1g (NH4)6Mo7O24ㆍ4H2O, 0.02g Na2B4O7ㆍ10H2O)의 성분으로 구성된 배지이다. 유일 탄소원인 포도당이 모두 소모될때 마다 1리터당 700g 포도당, 8g MgSO4ㆍ7H2O로 구성된 feeding solution을 추가로 공급하였다. 배양은 96시간동안 30℃와 dissolved oxygen level을 40%로 유지하도록 작동하는 bioreactor(New Brunswick Scientific Co, NJ)에서 진행하였다. 배양이 끝난, 배양액은 13,200 rpm에서 10분간 원심분리하여, 세포를 채취하여 분석을 수행함으로써, 폴리에스터 아미이드 생산을 확인하였다.
그 결과, 도 5e에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 3-아미노프로피오네티트를 단량체로 갖고있는 폴리(에스터 아마이드)를 높은 생산 농도, 생산성 및 수율로 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물이 유가식 배양 발효를 활용하여 높은 효율로 폴리(에스터 아마이드)를 생성함을 확인하였다.
실시예 10: 다양한 미생물 유래의 polyhydroxyalkanoate를 사용하여 폴리(에스터 아마이드) 생산 확인
실시예 2에서 제조한 재조합 미생물(PEA08, PEA09, PEA10, PEA11, PEA04, PEA12, PEA13, PEA14, PEA15, PEA16)을 10mL LB 배지에 접종하여 37℃에서 8시간 동안 전배양을 수행하고, 전배양한 배양액 1.0mL을 350mL 플라스크에 100mL의 LB배지에 접종하여 배양하였다.
LB배지의 조성은 증류수 1리터당 10g tryptone, 5g yeast extract, 10g NaCl의 성분으로 구성된 배지이다. 상기 조성에서 beta-alanine coenzyme A transferase의 기질로 알려진 아미노산들인 3-아미노프로피오네이트를 2 g/L 농도로 추가 탄소원으로 공급하였다. 또한 50 g/L의 sodium (RS)-3-하이드록이뷰티레이트 역시 추가 공급하였다. 배양은 96시간동안 30℃와 200rpm으로 작동하는 shaking incubator(jSR, 한국)에서 진행하였다. 배양이 끝난, 배양액은 13,200 rpm에서 10분간 원심분리하여, 세포를 채취하여 분석을 수행함으로써, 폴리에스터 아미이드 생산을 확인하였다.
그 결과, 도 6에 개시된 바와 같이, 본 발명에 따른 재조합 미생물이 모두 3-아미노프로피오네이트를 단량체로 갖고 있고 다양한 분자량을 갖고있는 폴리(에스터 아마이드)를 생산함을 확인할 수 있었다.
이 결과로부터, 본 발명에 따른 재조합 미생물들이 3-아미노프로피오네이트를 단량체로 갖고 있는 폴리(에스터 아마이드)를 생성함을 확인하였다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시태양일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
본 발명의 재조합 미생물은 포도당, 아미노산, 하이드록시산, 카르복실산 등과 같은 다양한 탄소원으로부터 폴리(에스터 아마이드) 고분자를 생합성할 수 있으므로, 폴리(에스터 아마이드)의 산업적 생산에 유용하다.

- 숙주 미생물에 코엔자임 A 전이효소(Coenzyme A transferase)를 코딩하는 유전자 및 PHA 합성효소(polyhydroxyalkanoate synthase)를 코딩하는 유전자가 도입 또는 증폭되어 있는, 폴리(에스터 아마이드)의 생성능을 갖는 재조합 미생물.
- 제1항에 있어서, 탄소원으로부터 상기 코엔자임 A 전이효소의 기질이 되는 유기산의 생합성 경로를 갖는 것을 보유하는 것을 특징으로 하는 재조합 미생물.
- 제2항에 있어서, 상기 탄소원은 당 또는 유기산인 것을 특징으로 하는 재조합 미생물.
- 제2항에 있어서, 상기 코엔자임 A 전이효소의 기질이 되는 유기산은 아미노산 및 하이드록시산 중 어느 하나 이상인 것을 특징으로 하는 재조합 미생물.

- 제5항에 있어서, 상기 아미노산은 글리신, 알라닌, 2-아미노뷰티레이트(2-aminobutyrate), 3-아미노프로피오네이트(3-aminopropionate), 아이소세린(Isoserine), 3-아미노뷰티레이트(3-aminobutyrate), 3-아미노발러레이트(3-aminovalerate), 베타-페닐알라닌 (beta-phenylalanine; BPA), 4-아미노뷰티레이트(4-aminobutyrate), 4-아미노-3-메틸뷰티레이트(4-amino-3-methylbutyrate), 5-아미노발러레이트(5-aminovalerate), 6-아미노카프로에이트(6-aminocaproate), 7-아미노헵타노에이트(7-aminoheptanoate), 3-아미노아이소뷰티레이트(3-aminoisobutyrate), 파라-아미노벤조에이트(para-aminobenzoate; PAB) 및 이의 이성질체로 구성된 군에서 선택되는 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 코엔자임 A 전이효소는 베타-알라닌 코엔자임 A 전이효소인 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 코엔자임 A 전이효소를 코딩하는 유전자는 클로스트리듐 프로피오니쿰(Clostridium propionicum) 유래의 act 유전자인 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 코엔자임 A 전이효소는 서열번호1 내지 3에서 선택되는 서열을 포함하는 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 PHA 합성효소는 PhaC인 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 PHA 합성효소를 코딩하는 유전자는 큐프리아비더스 네케터 (Cupriavidus necator), 에어로모나스 케비에 (Aeromonas caviae), 에어로모나스 하이드로필리아 (Aeromonas hydrophila), 에어로모나스 살모니시다 (Aeromonas salmonicida), 슈도모나스 MBEL 6-19 (Pseudomonas sp. MBEL 6-19), 슈도모나스 61-3 (Pseudomonas sp. 61-3), 슈도모나스 푸티다 KT2440 (Pseudomonas putida KT2440), 슈도모나스 아르지노사 PAO1 (Pseudomonas aeruginosa PAO1), 슈도모나스 클로로라피스 (Pseudomonas chlororaphis), 및 슈도모나스 레시노보렌스 (Pseudomonas resinovorans)로 구성된 군에서 선택되는 균주로부터 유래한 phaC 유전자인 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 PHA 합성효소는 서열번호 4 내지 13으로 구성된 군에서 선택되는 서열을 포함하는 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 폴리(에스터 아마이드)는 상기 코엔자임 A 전이효소의 기질이 되는 유기산을 단량체로 포함하는 것을 특징으로 하는 재조합 미생물.
- 제13항에 있어서, 상기 코엔자임 A 전이효소의 기질이 되는 유기산은 글리신, 알라닌, 2-아미노뷰티레이트(2-aminobutyrate), 3-아미노프로피오네이트(3-aminopropionate), 아이소세린(Isoserine), 3-아미노뷰티레이트(3-aminobutyrate), 3-아미노아이소뷰티레이트(3-aminoisobutyrate), 3-아미노발러레이트(3-aminovalerate), 베타-페닐알라닌 (beta-phenylalanine; BPA), 4-아미노뷰티레이트(4-aminobutyrate), 4-아미노-3-메틸뷰티레이트(4-amino-3-methylbutyrate), 파라-아미노벤조에이트(para-aminobenzoate; PAB), 5-아미노발러레이트(5-aminovalerate), 6-아미노카프로에이트(6-aminocaproate), 7-아미노헵타노에이트(7-aminoheptanoate), 글리콜레이트, 락테이트, 2-하이드록시뷰티레이트(2-hydroxybutyrate), 3-하이드록시프로피오네이트(3-hydroxypropionate), 3-하이드록시뷰티레이트(3-hydroxybutyrate), 프로피오네이트(propionate), 뷰티레이트(butyrate), 이소뷰티레이트(isobutyrate), 발러레이트(valerate), 이소발러레이트(isovalerate), 2-메틸뷰티레이트(2-methylbutyrate), 카프로에이트(caproate), 말로네이트(malonate), 숙시네이트(succinate) 및 이의 이성질체로 구성된 군에서 선택되는 것을 특징으로 하는 재조합 미생물.
- 제1항에 있어서, 상기 숙주 미생물은 상기 재조합 미생물은 박테리아, 효모 및 곰팡이로 구성된 군에서 선택되는 것을 특징으로 하는 재조합 미생물.
- 다음의 단계를 포함하는 폴리(에스터 아마이드)의 제조방법:(a) 제1항 내지 제15항 중 어느 한 항의 재조합 미생물을 배양하여 폴리(에스터 아마이드)를 생산하는 단계; 및(b) 상기 생산된 폴리(에스터 아마이드)를 회수하는 단계.
- 다음의 단계를 포함하는 폴리(에스터 아마이드)의 제조방법:(a) 제1항 내지 제15항 중 어느 한 항의 재조합 미생물을 코엔자임 A 전이효소의 기질이 되는 유기산이 첨가된 배지에서 배양하여 폴리(에스터 아마이드)를 생산하는 단계; 및(b) 상기 생산된 폴리(에스터 아마이드)를 회수하는 단계.
- 제16항의 방법으로 제조된 폴리(에스터 아마이드).
