10質量%スキムミルク培地に乳酸菌の前培養菌液を0.01質量%接種し、37℃の恒温槽に静置して培養した場合に、 培養時間が4時間の時にpH6.0~6.5、 培養時間が8時間の時にpH5.0~6.0、 培養時間が12時間の時にpH4.5~5.5、 培養時間が24時間の時にpH4.0~4.5となる乳酸菌と、 ラクチカゼイバチルス・パラカゼイとを混合培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法またはスキムミルクを0.1~20質量%含有する培地に、ラクチカゼイバチルス・パラカゼイの前培養菌液を接種し、 培養時間が8時間の時にpH5.5~6.25、 培養時間が16時間の時にpH4.25~5.5、 培養時間が24時間の時にpH4.0~4.75となるようにpHを調整して培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法は、ラクチカゼイバチルス・パラカゼイの胃酸、胆汁酸の耐性を向上する新たな方法である。
Description
本発明は、ラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法に関する。
近年、プロバイオティクスとして、腸内細菌叢のバランスを改善し、宿主の健康に有益な作用をもたらす微生物が注目されている。このような微生物として乳酸菌やビフィズス菌が利用されており、これらを摂取することで、便秘・下痢症の改善効果、免疫機能改善による感染防御・アレルギー抑制効果、動脈硬化の予防効果など様々な生理効果を得られることが報告されている。
プロバイオティクスの定義に関しては、近年、生菌だけでなく死菌を含める考え方も提唱されている。しかし、プロバイオティクスの作用機序として、腸内細菌叢において有用な微生物が増殖し、様々な代謝産物を産生することによって、競合する有害な微生物を排除したり、増殖を抑制したりすることで、その機能がより効果的に発揮され得るといえることから、腸内において生きた状態で存在している方が有利と考えられる。このように、摂取された有用微生物は、生きたままの状態で腸内に到達することが望まれるが、腸内に到達するまでには、温度、pH、酸素など、微生物の生育を阻害する様々な因子が存在し、特に胃液や胆汁などの消化液は、胃酸や胆汁酸の他、様々な消化酵素を含み、これらの消化液に曝されると、死滅したり、生き残ったとしてもコロニー形成能(増殖能)を喪失したりするなど重大な傷害を受ける。
そのため、有用微生物を消化液から保護し、消化管内での生残性を向上させる技術が検討されている。例えば、微生物のhrcA遺伝子を破壊することにより、微生物自体の胃酸・胆汁酸耐性を増強する方法(特許文献1)や、乳酸菌を腸溶性カプセル内に封入し、胃液などとの直接的な接触を回避して生残性を高める方法(特許文献2)などが開示されている。しかし、前者は煩雑な遺伝子工学的手法を要するものであり、後者は剤型に制約が生じるなど、これらの技術は汎用性が高いものとは言い難かった。一方、より簡便な方法として、ラクトバチルス・ラクチスを、Tween 80を過剰添加した培地で培養することにより、その胆汁酸耐性が向上することが報告されている(非特許文献1)。しかし、本出願人が、ラクチカゼイバチルス・パラカゼイにこの技術を適用したところ、胆汁酸耐性の向上は認められず、むしろ低下してしまうことが確認された。
そこで、本出願人は、ラクチカゼイバチルス・パラカゼイを含む乳酸菌をオレイン酸又はその塩を含有する培地で培養することにより、胃酸胆汁酸耐性が向上することを見出し、特許出願している(特許文献3)。
しかしながら、ラクチカゼイバチルス・パラカゼイを含む乳酸菌の胃酸胆汁酸耐性の向上の技術は、生菌を取り扱う多種多様な飲食品に関わる技術であることから、多種多様な技術があった方がよいことも明らかである。

従って、本発明の課題は、ラクチカゼイバチルス・パラカゼイの胃酸、胆汁酸の耐性を向上する新たな方法を提供することである。
本発明者らは、上記課題を解決するために鋭意研究した結果、ラクチカゼイバチルス・パラカゼイと乳酸菌を混合培養するにあたり、乳酸菌として特定の経過時間におけるpHが特定の範囲になる乳酸菌を用いることにより、上記課題を解決することを見出し、本発明を完成させた。
また、本発明者らは、上記課題を解決するために鋭意研究した結果、ラクチカゼイバチルス・パラカゼイを培養するにあたり、特定の経過時間におけるpHを特定の範囲になるように調整することにより、上記課題を解決することを見出し、本発明を完成させた。
すなわち、本発明は、10質量%スキムミルク培地に107~109cfu/mlの乳酸菌の前培養菌液を0.01質量%接種し、37℃の恒温槽に静置して培養した場合に、
培養時間が4時間の時にpH6.0~6.5、
培養時間が8時間の時にpH5.0~6.0、
培養時間が12時間の時にpH4.5~5.5、
培養時間が24時間の時にpH4.0~4.5となる乳酸菌と、
ラクチカゼイバチルス・パラカゼイとを混合培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法である。
培養時間が4時間の時にpH6.0~6.5、
培養時間が8時間の時にpH5.0~6.0、
培養時間が12時間の時にpH4.5~5.5、
培養時間が24時間の時にpH4.0~4.5となる乳酸菌と、

また、本発明は、スキムミルクを0.1~20質量%含有する培地に、ラクチカゼイバチルス・パラカゼイの前培養菌液を接種し、
培養時間が8時間の時にpH5.5~6.25、
培養時間が16時間の時にpH4.25~5.5、
培養時間が24時間の時にpH4.0~4.75となるようにpHを調整して培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法である。
培養時間が8時間の時にpH5.5~6.25、
培養時間が16時間の時にpH4.25~5.5、
培養時間が24時間の時にpH4.0~4.75となるようにpHを調整して培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法である。
更に、本発明は、上記胃酸胆汁酸耐性向上方法により胃酸胆汁酸耐性が向上したラクチカゼイバチルス・パラカゼイである。
本発明によれば、ラクチカゼイバチルス・パラカゼイの胃酸、胆汁酸の耐性を向上することができる。
従って、本発明により得られる胃酸、胆汁酸の耐性が向上したラクチカゼイバチルス・パラカゼイを利用すれば、生菌を腸まで多く届けられるようになり、これまでラクチカゼイバチルス・パラカゼイが有している種々の効能についてもより強く得ることができる。
本発明のラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法(以下、「本発明向上方法」という)としては、以下の二つの方法が挙げられる。
<本発明向上方法1>
10質量%スキムミルク培地に107~109cfu/mlの乳酸菌の前培養菌液を0.01質量%接種し、37℃の恒温槽に静置して培養した場合に、

培養時間が12時間の時にpH4.5~5.5、
培養時間が24時間の時にpH4.0~4.5となる乳酸菌と、
ラクチカゼイバチルス・パラカゼイとを混合培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。

まず、本発明向上方法1について説明する。本発明向上方法1に用いられる、乳酸菌は、例えば、10質量%スキムミルク培地に乳酸菌の前培養菌液を0.01質量%接種し、37℃の恒温槽に静置して培養し、培養時間が4時間の時にpH6.0~6.5、培養時間が8時間の時にpH5.0~6.0、好ましくはpH5.4~5.9、培養時間が12時間の時にpH4.5~5.5、好ましくはpH4.6~5.2、培養時間が24時間の時にpH4.0~4.5となるものである。これら各培養時間におけるpHの範囲は適宜組み合わせることができる。
上記スキムミルク培地には、10質量%のスキムミルクを含む他、例えば、通常の乳酸菌培地に使用される成分を添加してもよく、ビタミンA、ビタミンB類、ビタミンC、ビタミンE等のビタミン類やカルシウム、マグネシウム等の塩類、肉エキス、ペプトン、酵母エキス、アミノ酸等の窒素源、グルコース、キシロース、フルクトース、マルトース等の炭素源等を含んでいてもよい。
また、107~109cfu/mlの乳酸菌の前培養菌液は、凍結保存された乳酸菌をスキムミルク培地に接種して恒温槽で乳酸菌の生育に適した温度(好ましくは30~37℃)で24時間静置培養するようにして調製すればよい。ここで用いられる恒温槽は特に限定されず、例えば、インキュベーターや恒温水槽等を用いればよい。ここで静置とは、撹拌を行わず静止した状態で置くことをいう。培養時におけるpHの測定は、例えば、pHメーター等で規定の時間に測定すればよい。
このような乳酸菌としては、例えば、ストレプトコッカス属、ラクトコッカス属、ラクチカゼイバチルス属、ラクチプランチバチルス属、ラチラクトバチルス属、リギラクトバチルス属、リモシラクトバチルス属、レンチラクトバチルス属、レビラクトバチルス属等の乳酸菌が挙げられる。これらの乳酸菌の中でもストレプトコッカス属、ラクトコッカス属の乳酸菌が好ましい。これらのストレプトコッカス属、ラクトコッカス属の乳酸菌の中でもストレプトコッカス・サーモフィルス、ラクトコッカス・ラクチスが好ましく、ストレプトコッカス・サーモフィルス YIT 2001、ストレプトコッカス・サーモフィルス YIT 2021、ラクトコッカス・ラクチスYIT 2027がより好ましい。これら乳酸菌は1種または2種以上を用いることができる。
上記した乳酸菌のうち、ストレプトコッカス・サーモフィルス YIT 2001は、ストレプトコッカス・サーモフィルス YIT 2001(FERM BP-7538、受託日:2001年4月5日)としてブダペスト条約の国際寄託当局である独立行政法人製品評価技術基盤機構特許生物寄託センター(〒292-0818日本国千葉県木更津市かずさ鎌足2-5-8 120号室)に国際寄託されている。
また、ストレプトコッカス・サーモフィルス YIT 2021は、ストレプトコッカス・サーモフィルス YIT 2021(FERM BP-7537、受託日:2001年4月5日)として上記した独立行政法人製品評価技術基盤機構特許生物寄託センターに国際寄託されている。
更に、ラクトコッカス・ラクチスYIT 2027は、ラクトコッカス・ラクチスYIT 2027(FERM BP-6224、受託日:1998年1月16日)として上記した独立行政法人製品評価技術基盤機構特許生物寄託センターに国際寄託されている。
本発明向上方法1において、上記した乳酸菌と混合培養されるラクチカゼイバチルス・パラカゼイは、特に限定されないが、例えば、ラクチカゼイバチルス・パラカゼイ YIT 0209(基準株)、ラクチカゼイバチルス・パラカゼイ YIT 9029株、ラクチカゼイバチルス・パラカゼイ YIT 0180等が挙げられる。これらラクチカゼイバチルス・パラカゼイは1種または2種以上を用いることができる。これらラクチカゼイバチルス・パラカゼイの中でもラクチカゼイバチルス・パラカゼイYIT 9029が好ましい。
上記したラクチカゼイバチルス・パラカゼイYIT 9029は、ラクトバチルス・カゼイYIT 9029(FERM BP-1366、受託日:昭和56年5月1日)としてブダペスト条約の国際寄託当局である独立行政法人製品評価技術基盤機構特許生物寄託センター(〒292-0818日本国千葉県木更津市かずさ鎌足2-5-8 120号室)に国際寄託されている。なお、ラクチカゼイバチルス・パラカゼイ YIT 9029は以前の分類ではラクトバチルス・カゼイ(Lactobacillus casei)とされていたが、近年ラクトバチルス属の再分類がなされ、ラクチカゼイバチルス・パラカゼイ(Lacticaseibacillus paracasei)に分類された(Zheng et al., A taxonomic note on the genus Lactobacillus : Description of 23 novel genera, emended description of the genus Lactobacillus Beijerinck 1901, and union of Lactobacillaceae and Leuconostocaceae. Int. J. Syst. Evol. Microbiol. 2020 Apr; 70(4):2782-2858 DOI 10.1099/ijsem.0.004107)。本明細書では、乳酸菌の分類について、上記文献に基づいて分類された新分類で表記している。
上記したラクチカゼイバチルス・パラカゼイYIT 0209は基準株なので、ATCC等から入手できる。
本発明向上方法1において、上記した乳酸菌と、ラクチカゼイバチルス・パラカゼイとを混合培養する方法は、特に限定されないが、例えば、培地に上記した乳酸菌を0.005~0.5質量%、好ましくは0.01~0.1質量%、ラクチカゼイバチルス・パラカゼイを0.05~5質量%、好ましくは0.1~1質量%で接種し、これを25~40℃、好ましくは30~37℃で、24~72時間、好ましくは40~50時間培養する方法が挙げられる。培養は、静置、振盪であってもよい。
上記培地としては、特に限定されないが、例えば、スキムミルク培地である。また、培地には通常の乳酸菌培地に使用される成分を添加してもよく、ビタミンA、ビタミンB類、ビタミンC、ビタミンE等のビタミン類やカルシウム、マグネシウム等の塩類、肉エキス、ペプトン、酵母エキス、アミノ酸等の窒素源、グルコース、キシロース、フルクトース、マルトース等の炭素源等の成分を含んでいてもよい。
上記のようにして乳酸菌と、ラクチカゼイバチルス・パラカゼイとを混合培養することにより、ラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性が向上する。ここでラクチカゼイバチルス・パラカゼイの胃酸、胆汁酸の耐性を確認する方法は、特に限定されないが、例えば、人工胃酸や人工胆汁酸を用いる胃酸胆汁酸連続試験方法等が挙げられる。胃酸胆汁酸連続試験方法は、例えば、実施例に記載の方法で行うことができる。
また、上記した胃酸胆汁酸耐性が向上したラクチカゼイバチルス・パラカゼイは、例えば、本発明向上方法1を行わない場合と比較して、胃酸胆汁酸連続試験後の生残率が3~100倍となる。
次に、本発明向上方法2について説明する。本発明向上方法2においては、スキムミルクを0.1~20質量%含有する培地にラクチカゼイバチルス・パラカゼイの前培養液を接種し、下記培養時間で下記範囲になるようにpHを調整しながら培養する。培養条件は特に限定されないが、例えば、37℃の恒温槽に静置して24~72時間、好ましくは40~50時間培養すればよい。
上記培養において、培養時間が8時間の時にpH5.5~6.25、好ましくはpH5.6~6.2、培養時間が16時間の時にpH4.25~5.5、好ましくはpH4.4~5.4、培養時間が24時間の時にpH4.0~4.75となるようにpHを調整する方法は、特に限定されず、例えば、規定の時間に培地のpHを乳酸、塩酸等の酸や、炭酸水素ナトリウム、水酸化ナトリウム等のアルカリで調整すればよい。また、特定の微生物等を同時に培養することや、培養温度を調整することでpHを調整しても良い。これら各培養時間におけるpHの範囲は適宜組み合わせることができる。
本発明向上方法2において用いられるラクチカゼイバチルス・パラカゼイは本発明向上方法1で用いられるものと同様でよい。
本発明向上方法2に用いられるスキムミルク培地は、本発明向上方法1で用いられるものと同様でよい。また、恒温槽、前培養菌液の調製方法、静置、pHの測定についても本発明向上方法1で用いられるものと同様でよい。
上記のようにして、ラクチカゼイバチルス・パラカゼイの培養をpHの調整をしながら行うことにより、ラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性が向上する。ここでラクチカゼイバチルス・パラカゼイの胃酸、胆汁酸の耐性を確認する方法は、本発明向上方法1で用いられる方法と同様でよい。
なお、本発明向上方法2において、ラクチカゼイバチルス・パラカゼイに影響を及ぼさない範囲で、培養の際に、本発明向上方法1で用いる乳酸菌と同様の乳酸菌をラクチカゼイバチルス・パラカゼイと共に用いてもよい。
また、上記した胃酸胆汁酸耐性が向上したラクチカゼイバチルス・パラカゼイは、例えば、本発明向上方法2を行わない場合と比較して、胃酸胆汁酸連続試験後の生残菌数が3~19倍、生残率が10~70倍高くなる。ここで、胃酸胆汁酸耐性が向上したとは、胃酸胆汁酸連続試験後の生残率が3倍以上となることを指す。
以上説明した本発明向上方法によれば、胃酸、胆汁酸の耐性が向上したラクチカゼイバチルス・パラカゼイが得られる。
このラクチカゼイバチルス・パラカゼイは、乳酸菌飲料、発酵乳等の飲食品に利用することができる。また、このラクチカゼイバチルス・パラカゼイは医薬品、健康食品等の製剤にも利用することができる。
具体的に、ラクチカゼイバチルス・パラカゼイを飲食品に利用する場合、本発明向上方法において培養後のラクチカゼイバチルス・パラカゼイ(および必要により混合培養した乳酸菌)を含む培地を、従来公知の乳酸菌を含む培地を利用した飲食品と同様に製造すればよい。
また、ラクチカゼイバチルス・パラカゼイを製剤に利用する場合、本発明向上方法において培養後のラクチカゼイバチルス・パラカゼイ(および必要により混合培養した乳酸菌)を含む培地から、必要により分離・精製・乾燥を行って、従来公知の乳酸菌を含む培地を利用した製剤と同様に製造すればよい。
このような飲食品や製剤は、これに含まれる胃酸胆汁酸耐性が向上したラクチカゼイバチルス・パラカゼイを含むため、従来のラクチカゼイバチルス・パラカゼイを含む飲食品や製剤よりも効果が高いものとなる。効果としては、便通改善、NK細胞の活性化、睡眠の質の向上などがある。
以下、本発明を本発明の実施例を挙げて詳細に説明するが、本発明はこれら実施例に何ら限定されるものではない。

以下の(A)の単独培養、(B)~(E)の混合培養を行い、培養0、8、12、16、24、48時間後のpHを均一になるまで撹拌した菌液にポータブルpHメーターを浸漬することで測定した。それらの結果を図1に示した。

(E)(1)および(5)で調製した前培養菌液を用いて10質量%スキムミルク培地にラクチカゼイバチルス・パラカゼイYIT 9029およびラクトコッカス・ラクチスYIT 2027をそれぞれ0.5質量%、0.01質量%接種し、37℃の恒温槽に静置して培養した。
<胃酸胆汁酸連続耐性試験>
(A)~(E)の単独培養および混合培養について48時間の培養後、胃酸胆汁酸連続耐性試験を行った。胃酸の条件はpH4.0、1時間、37℃であり、その後の胆汁酸の条件はpH4.4、オックスゴール(牛の胆汁酸)0.3質量%、5分、37℃である。胃酸暴露前および胆汁酸暴露後にサンプリングした菌液を適宜PBSで希釈し、LLV寒天培地に塗抹した。37℃で好気的に2~3日培養してコロニーを計測し、菌液中の生菌数(log10cfu/ml)および生残率(%)を測定した。生残率(%)は胃酸曝露後および胆汁酸曝露後の生菌数を胃酸暴露前の生菌数で除することで算出した。同様の試験を4回繰り返し、それらの結果を図2および図3に示した。

<結果>
これらの結果から、ラクチカゼイバチルス・パラカゼイYIT 9029と、ストレプトコッカス・サーモフィルスYIT 2001、ストレプトコッカス・サーモフィルスYIT 2021またはラクトコッカス・ラクチスYIT 2027と混合培養することにより胃酸胆汁酸耐性が顕著に向上することが分かった。
これらの結果から、ラクチカゼイバチルス・パラカゼイYIT 9029と、ストレプトコッカス・サーモフィルスYIT 2001、ストレプトコッカス・サーモフィルスYIT 2021またはラクトコッカス・ラクチスYIT 2027と混合培養することにより胃酸胆汁酸耐性が顕著に向上することが分かった。
実 施 例 2
単独培養試験:
どのような性質を有する菌株がラクチカゼイバチルス・パラカゼイYIT 9029と混合培養した時に胃酸胆汁酸耐性が顕著に向上するかを判断するために、混合培養に用いた菌株を単独培養し、培養0、4、8、12、16、24時間後のpHをpHメーターで測定した。なお、培養は菌株を全て単独で用いる以外は全て実施例1と同様である。それらの結果を図4に示した。
単独培養試験:
どのような性質を有する菌株がラクチカゼイバチルス・パラカゼイYIT 9029と混合培養した時に胃酸胆汁酸耐性が顕著に向上するかを判断するために、混合培養に用いた菌株を単独培養し、培養0、4、8、12、16、24時間後のpHをpHメーターで測定した。なお、培養は菌株を全て単独で用いる以外は全て実施例1と同様である。それらの結果を図4に示した。
この結果から、ラクチカゼイバチルス・パラカゼイYIT 9029と混合培養する乳酸菌としては、ストレプトコッカス・サーモフィルスYIT 2001、ストレプトコッカス・サーモフィルスYIT 2021またはラクトコッカス・ラクチスYIT 2027のように培養時間が4時間の時にpH6.0~6.5、培養時間が8時間の時にpH5.0~6.0、培養時間が12時間の時にpH4.5~5.5、培養時間が24時間の時にpH4.0~4.5となるものがよいことが分かった。一方、実施例1でラクチカゼイバチルス・パラカゼイYIT 9029と混合培養してもラクチカゼイバチルス・パラカゼイYIT 9029の胃酸胆汁酸耐性が向上しなかったストレプトコッカス・サーモフィルスYIT 2037は、このような性質ではなかった。

凍結保存されたラクチカゼイバチルス・パラカゼイYIT 0180をスキムミルク培地に0.1%(v/v)接種して37℃で好気的に48時間培養した。

(I)(9)および(11)で調製した前培養菌液を用いて10質量%スキムミルク培地にラクチカゼイバチルス・パラカゼイ YIT 0209(基準株)およびラクトコッカス・ラクチスYIT 2027をそれぞれ0.5質量%、0.01質量%接種し、37℃の恒温槽に静置して培養した。
(J)(10)および(11)で調製した前培養菌液を用いて10質量%スキムミルク培地にラクチカゼイバチルス・パラカゼイYIT 9029およびラクトコッカス・ラクチスYIT 2027をそれぞれ0.5質量%、0.01質量%接種し、37℃の恒温槽に静置して培養した。
<胃酸胆汁酸連続耐性試験>
(F)~(J)の混合培養および単独培養について48時間の培養後、選択培地以外は実施例1と同様にして胃酸胆汁酸連続耐性試験を行い、生残率(%)を算出した。生残率(%)は胃酸曝露後および胆汁酸曝露後の生菌数を胃酸暴露前の生菌数で除することで算出した。(F)と(G)では選択培地としてテトラサイクリンを0.5 μg/ml含有するMRS寒天培地を使用し、(H)~(J)ではバンコマイシンを10μg/ml含有するMRS寒天培地を使用した。同様の試験を4~5回繰り返し、それらの結果を図5に示した。なお、ラクトバチルス・ヘルベティカスYIT 0083(基準株)の生残率を評価する際には胆汁酸の条件をpH4.4、オックスゴール0.15質量%、5分、37℃とした。

<結果>
これらの結果から、ラクトコッカス・ラクチスYIT 2027と、ラクチカゼイバチルス・パラカゼイYIT 0180、ラクチカゼイバチルス・パラカゼイYIT 0209(基準株)またはラクチカゼイバチルス・パラカゼイYIT 9029と混合培養することにより胃酸胆汁酸耐性が顕著に向上することが分かった。一方、ラクチカゼイバチルス・パラカゼイに属さないラクトバチルス・ヘルベティカスやラクトバチルス・ガセリはラクトコッカス・ラクチスYIT 2027と混合培養しても胃酸胆汁酸耐性が向上することはなかった。
これらの結果から、ラクトコッカス・ラクチスYIT 2027と、ラクチカゼイバチルス・パラカゼイYIT 0180、ラクチカゼイバチルス・パラカゼイYIT 0209(基準株)またはラクチカゼイバチルス・パラカゼイYIT 9029と混合培養することにより胃酸胆汁酸耐性が顕著に向上することが分かった。一方、ラクチカゼイバチルス・パラカゼイに属さないラクトバチルス・ヘルベティカスやラクトバチルス・ガセリはラクトコッカス・ラクチスYIT 2027と混合培養しても胃酸胆汁酸耐性が向上することはなかった。
実 施 例 4
pH制御培養試験:
pH制御装置を備えた培養装置(Bio Jr.8、(株)バイオット製)を用いて実施例1と同様にし、ラクチカゼイバチルス・パラカゼイYIT 9029とラクトコッカス・ラクチスYIT 2027とを混合培養した際のpHの挙動にあわせて、ラクチカゼイバチルス・パラカゼイYIT 9029の単独培養の時のpHを培養時間が8時間の時にpH5.5~6.25、培養時間が16時間の時にpH4.25~5.5、培養時間が24時間の時にpH4.0~4.75となるように乳酸で調整した。その結果を図6に示した。
pH制御培養試験:
pH制御装置を備えた培養装置(Bio Jr.8、(株)バイオット製)を用いて実施例1と同様にし、ラクチカゼイバチルス・パラカゼイYIT 9029とラクトコッカス・ラクチスYIT 2027とを混合培養した際のpHの挙動にあわせて、ラクチカゼイバチルス・パラカゼイYIT 9029の単独培養の時のpHを培養時間が8時間の時にpH5.5~6.25、培養時間が16時間の時にpH4.25~5.5、培養時間が24時間の時にpH4.0~4.75となるように乳酸で調整した。その結果を図6に示した。
48時間の培養後、実施例1と同様にして、胃酸胆汁酸連続耐性試験を行い、胃酸反応前、胃酸反応後、胆汁酸反応後の生残菌数および生残率を算出した。同様の試験を3回繰り返し、それらの結果を図7および図8に示した。なお、胆汁酸の条件をpH4.4、オックスゴール1.0質量%、5分、37℃とした。
この結果から、ラクチカゼイバチルス・パラカゼイYIT 9029を単独で培養する時に、培養時間が8時間の時にpH5.5~6.25、培養時間が16時間の時にpH4.25~5.5、培養時間が24時間の時にpH4.0~4.75となるように調整することでも、実施例1のような混合培養と同様に、ラクチカゼイバチルス・パラカゼイYIT 9029の胃酸胆汁酸耐性が顕著に向上することが分かった。
本発明により得られる胃酸、胆汁酸の耐性が向上したラクチカゼイバチルス・パラカゼイを利用すれば、生菌を腸まで多く届けられるようになり、健康の増進に利用することができる。
Claims (6)
- 10質量%スキムミルク培地に107~109cfu/mlの乳酸菌の前培養菌液を0.01質量%接種し、37℃の恒温槽に静置して培養した場合に、
培養時間が4時間の時にpH6.0~6.5、
培養時間が8時間の時にpH5.0~6.0、
培養時間が12時間の時にpH4.5~5.5、
培養時間が24時間の時にpH4.0~4.5となる乳酸菌と、
ラクチカゼイバチルス・パラカゼイとを混合培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。 - 乳酸菌が、ストレプトコッカス・サーモフィルス、ラクトコッカス・ラクチスからなる群から選ばれる1種または2種である請求項1記載のラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。
- ラクチカゼイバチルス・パラカゼイが、ラクチカゼイバチルス・パラカゼイ YIT 9029(FERM BP-1366)である請求項1記載のラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。
- スキムミルクを0.1~20質量%含有する培地に、ラクチカゼイバチルス・パラカゼイの前培養菌液を接種し、
培養時間が8時間の時にpH5.5~6.25、
培養時間が16時間の時にpH4.25~5.5、
培養時間が24時間の時にpH4.0~4.75となるようにpHを調整して培養することを特徴とするラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。 - ラクチカゼイバチルス・パラカゼイが、ラクチカゼイバチルス・パラカゼイ YIT 9029(FERM BP-1366)である請求項4記載のラクチカゼイバチルス・パラカゼイの胃酸胆汁酸耐性向上方法。
- 請求項1~5の何れか1項に記載の胃酸胆汁酸耐性向上方法により胃酸胆汁酸耐性が向上したラクチカゼイバチルス・パラカゼイ。
