Abstract
本発明の課題は、色のくすみが抑制された組織化植物性タンパク質の製造技術を提供することである。グルタミナーゼ類及びプロテアーゼを用いて処理した非組織化状の植物性タンパク質材料を組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみを抑制できる。
Description
本発明は組織化植物性タンパク質の製造方法に関する。より具体的には、色のくすみが抑制された組織化植物性タンパク質の製造方法に関する。
近年、食品市場において、動物性肉の代替として組織化植物性タンパク質材料(以下において、「代替肉」とも記載する。)を用いた食品のシェアが伸びている。組織化植物性タンパク質含有食品は、かつてはベジタリアン又はヴィーガンといった一部の消費者を対象としていたが、ここ数年では需要の動向も変化し、健康志向、ダイエット、環境問題、動物愛護等の意識の高まりを受け、改めて注目されている。
動物性肉は、畜肉の赤色、サーモンのピンク色、イカの白色等のように、明色及び/又は鮮やかな色調を呈するのが一般的である。一方、代替肉は一般的に灰褐色を呈するのが一般的である。このような代替肉の外観特性のため、代替肉をビート液により着色することで生の畜肉に似た外観を付与した商品が販売されている(非特許文献1)。
一方、植物性タンパク質を脱色する技術も知られている。非特許文献2では、過酸化水素による脱脂ツバキ種子粕の脱色法について報告がある。また、特許文献1では、アルカリ性プロテアーゼと酸性プロテアーゼ及び/又は中性プロテアーゼとの相乗作用下で酵素加水分解反応を行い、所定量の過酸化水素を添加することで着色物質の生成を抑制することが記載されている。

近年の代替肉の普及に伴い、代替肉の特性を動物性肉の特性により一層近づけることが望まれている。そのような中で、代替肉において動物性肉の外観を再現する手法として、上記のような着色や脱色は一応の選択肢となり得る。しかしながら、着色については、元来の色味である灰褐色の上に着色することとなるため、生肉ほどの鮮やかさが再現できない、元来の灰褐色のくすみを隠蔽しようとすると過剰な色味を付ける必要がある、ピンク色等の薄い色の動物性肉の再現まではできない、等の問題がある。また、脱色については、過酸化水素の発ガン性を考慮すると、健康志向の高いユーザーの志向に応えることができない。従って、代替肉に動物性肉の外観を再現するためには、代替肉が呈する灰褐色のくすみを抑制できる新たな方法が望まれる。
本発明の課題は、色のくすみが抑制された組織化植物性タンパク質の製造技術を提供することである。
本発明者は、鋭意検討の結果、グルタミナーゼ類(glutaminases)及びプロテアーゼを用いて処理した非組織化状の植物性タンパク質材料を組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみが抑制されることを見出した。本発明は、この知見に基づいて、更に検討を重ねることにより完成したものである。
即ち、本発明は下記に掲げる態様の発明を提供する。
項1. 非組織化状の植物性タンパク質材料にグルタミナーゼ類(glutaminases)及びプロテアーゼを作用させ、酵素処理された植物性タンパク質材料を得る工程1と、前記酵素処理された植物性タンパク質材料を含む組織化用材料を組織化処理する工程2と、を含む、組織化植物性タンパク質の製造方法。
項2. 前記組織化用材料の乾燥重量100重量部に対する前記酵素処理された植物性タンパク質材料の乾燥重量が、3重量部以上である、項1に記載の製造方法。
項3. 前記グルタミナーゼ類がグルタミナーゼ(EC3.5.1.2)である、項1又は2に記載の製造方法。
項4. 前記グルタミナーゼ類がバチルス属由来グルタミナーゼ類である、項1~3のいずれかに記載の製造方法。

本発明によれば、色のくすみが抑制された組織化植物性タンパク質の製造技術が提供される。
1.組織化植物性タンパク質の製造方法
本発明の組織化植物性タンパク質の製造方法は、非組織化状の植物性タンパク質材料にグルタミナーゼ類(glutaminases)及びプロテアーゼを作用させ、酵素処理された植物性タンパク質材料を得る工程1と、前記酵素処理された植物性タンパク質材料を含む組織化用材料を組織化処理する工程2とを含むことを特徴とする。以下、本発明の組織化植物性タンパク質の製造方法について詳述する。
本発明の組織化植物性タンパク質の製造方法は、非組織化状の植物性タンパク質材料にグルタミナーゼ類(glutaminases)及びプロテアーゼを作用させ、酵素処理された植物性タンパク質材料を得る工程1と、前記酵素処理された植物性タンパク質材料を含む組織化用材料を組織化処理する工程2とを含むことを特徴とする。以下、本発明の組織化植物性タンパク質の製造方法について詳述する。
1-1.工程1
工程1では、非組織化状の植物性タンパク質材料にグルタミナーゼ類及びプロテアーゼを作用させ、酵素処理された植物性タンパク質材料を得る。

1-1-1.非組織化状の植物性タンパク質材料
非組織化状の植物性タンパク質材料(以下において、単に「植物性タンパク質材料」とも記載する。)は、植物を原料とし、当該植物の構成成分のうちタンパク質以外の少なくとも一部を除去する等の処理を行うことによって、当該植物に含まれるタンパク質含量の乾燥重量当たりの比率を高めたものであって、組織化されていないものをいう。
非組織化状の植物性タンパク質材料(以下において、単に「植物性タンパク質材料」とも記載する。)は、植物を原料とし、当該植物の構成成分のうちタンパク質以外の少なくとも一部を除去する等の処理を行うことによって、当該植物に含まれるタンパク質含量の乾燥重量当たりの比率を高めたものであって、組織化されていないものをいう。
当該植物としては特に限定されず、大豆、えんどう、レンズ豆、ひよこ豆、黒豆、空豆、緑豆、ルピン豆、インゲン豆等の菽穀類;小麦、大麦、燕麦(オート)、ソルガム、米、ライムギ、そば、ひえ、あわ、テフ、キヌア、トウモロコシ、ポテト等の禾穀類;アーモンド、ココナッツ、ピーナッツ、カシューナッツ、ヘーゼルナッツ、ペカンナッツ、マカダミアナッツ、ピスタチオ、クルミ、ブラジルナッツ、ピリナッツ、栗、ゴマ、松の実、ヘンプシード(産業用ヘンプ)、チア種子、キア、アマランサス、カナリーシ―ド、アマニ等の種実類;藻類等が挙げられる。
植物性タンパク質材料は、これらの植物のうち1種から得られたものであってもよいし、複数種から得られたものであってもよい。これらの植物の中でも、好ましくは菽穀類が挙げられ、より好ましくは大豆、えんどうが挙げられ、さらに好ましくはえんどうが挙げられる。
植物性タンパク質材料(乾燥重量)中の植物性タンパク質の含有量としては特に限定されず、例えば40重量%以上、好ましくは50重量%以上、より好ましくは60重量%以上、さらに好ましくは70重量%以上、一層好ましくは75重量%以上が挙げられる。また、植物性タンパク質材料(乾燥重量)中の植物性タンパク質の含有量はその上限においても特に限定されるものではないが、例えば98重量%以下、好ましくは95重量%以下、より好ましくは90重量%以下、さらに好ましくは85重量%以下が挙げられる。
1-1-2.グルタミナーゼ類(glutaminases)
本発明において、「グルタミナーゼ類(glutaminases)」は、γ-グルタミル結合を加水分解する活性を有する酵素の総称であり、当該活性を有する限りにおいて、特に限定されない。従って、グルタミナーゼ類は、遊離グルタミンを基質とするもの、及びグルタミン残基を基質とするもののいずれであってもよく、転移活性を有しないもの、及び転移活性を有するもののいずれであってもよい。本発明で用いることができるグルタミナーゼ類としては、グルタミナーゼ(EC3.5.1.2;glutaminase;遊離グルタミンをグルタミン酸に加水分解する酵素)、プロテイングルタミナーゼ(EC3.5.1.XX;proteinglutaminase;タンパク質又はペプチド中のグルタミン残基を脱アミド化しグルタミン酸残基に変換する酵素)、トランスグルタミナーゼ(EC2.3.2.13;transglutaminase;タンパク質又はペプチド中のグルタミン残基を他のアミノ酸残基に転移し、磯ペプチド架橋を形成する酵素)が挙げられる。本発明において、グルタミナーゼとして、これらの酵素の中から1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。これらのグルタミナーゼ(glutaminases)の中でも、組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくはグルタミナーゼ(EC3.5.1.2;glutaminase;遊離グルタミンをグルタミン酸に加水分解する酵素)が挙げられる。

グルタミナーゼ類はその由来において特に限定されず、具体例としては、バチルス属等の微生物由来グルタミナーゼ類(好ましくはバチルス属等の微生物由来グルタミナーゼ(EC3.5.1.2))が挙げられる。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくはバチルス属由来グルタミナーゼ類(好ましくはバチルス属由来グルタミナーゼ(EC3.5.1.2))が挙げられ、より好ましくはバチルス・アミロリケファシエンス(Bacillus amyloliquefaciens)由来グルタミナーゼ類(好ましくはバチルス・アミロリケファシエンス由来グルタミナーゼ(EC3.5.1.2))が挙げられる。
微生物由来グルタミナーゼ類の使用において、上記のグルタミナーゼ類の由来元となる微生物の培養液、その破砕液及び抽出物を用いてもよいし、それらを任意のレベルで精製し、グルタミナーゼ類の含有量を高めた精製物を用いてもよい。また、グルタミナーゼ類の使用において、市販の酵素製剤(例えば、天野エンザイム株式会社製のバチルス・アミロリケファシエンス由来グルタミナーゼ等)を用いてもよい。
グルタミナーゼ類の使用量としては特に限定されないが、植物性タンパク質1g当たりの使用量として、例えば0.001U以上が挙げられる。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、グルタミナーゼ類のタンパク質1g当たりの使用量として、好ましくは0.005U以上、より好ましくは0.01U以上、0.05U以上、0.1U以上、0.3U以上、0.4U以上、0.5U以上、又は0.6U以上、さらに好ましくは0.7U以上、一層好ましくは0.8U以上、より一層好ましくは0.9U以上が挙げられる。グルタミナーゼ類の使用量は、その上限においても特に限定されないが、植物性タンパク質1g当たりの使用量として、例えば100U以下、又は50U以下、好ましくは10U以下、より好ましくは8U以下、6U以下、4U以下、3U以下、2U以下、又は1.5U以下が挙げられる。
また、グルタミナーゼ類の植物性タンパク質材料1g当たりの使用量としては、例えば0.0008U以上が挙げられる。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、グルタミナーゼ類のタンパク質材料1g当たりの使用量として、好ましくは0.004U以上、より好ましくは0.008U以上、0.04U以上、0.08U以上、0.24U以上、0.32U以上、0.4U以上、又は0.48U以上、さらに好ましくは0.56U以上、一層好ましくは0.64U以上、より一層好ましくは0.72U以上が挙げられる。グルタミナーゼ類の使用量は、その上限においても特に限定されないが、植物性タンパク質材料1g当たりの使用量として、例えば80U以下、又は40U以下、好ましくは8U以下、より好ましくは6.4U以下、4.8U以下、3.2U以下、2.4U以下、1.6U以下、又は1.2U以下が挙げられる。
なお、グルタミナーゼ類の活性については、L-グルタミンを基質とし、1分間に1μmolのL-グルタミン酸を生成する酵素量を1単位(1U)とする。
1-1-3.プロテアーゼ
プロテアーゼは、エンドペプチダーゼであることを限度として特に限定されない。プロテアーゼの例としては、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼが挙げられる。プロテアーゼとして、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼのうち、いずれか一方を用いてもよいし、両方を組み合わせて用いてもよい。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼの両方を組み合わせて用いることが好ましい。
プロテアーゼは、エンドペプチダーゼであることを限度として特に限定されない。プロテアーゼの例としては、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼが挙げられる。プロテアーゼとして、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼのうち、いずれか一方を用いてもよいし、両方を組み合わせて用いてもよい。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼの両方を組み合わせて用いることが好ましい。
糸状菌由来プロテアーゼとしては特に限定されないが、アスペルギルス(Aspergillus)属、リゾプス(Rhizopus)属、ムコール(Mucor)属、ニューロスポーラ(Neurospora)属、ペニシリウム(Penicillium)属、リゾムコール(Rhizomucor)属、及びスクレロティニア(Sclerotinia)属等に由来するプロテアーゼが挙げられる。アスペルギルス属由来プロテアーゼとしては、アスペルギルス・オリゼ(Aspergillus oryzae)由来プロテアーゼ、アスペルギルス・メレウス(Aspergillus melleus)由来プロテアーゼ等が挙げられる。また、糸状菌由来プロテアーゼは、酸性プロテアーゼであってもよいし、中性プロテアーゼであってもよい。より具体的な糸状菌由来プロテアーゼの例として、アスペルギルス・オリゼ由来酸性プロテアーゼ、アスペルギルス・オリゼ由来中性プロテアーゼ、アスペルギルス・メレウス由来中性プロテアーゼが挙げられる。
これらの糸状菌由来プロテアーゼは、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。これらの糸状菌由来プロテアーゼの中でも、組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくはアスペルギルス属由来プロテアーゼ、より好ましくはアスペルギルス・オリゼ由来プロテアーゼ、さらに好ましくはアスペルギルス・オリゼ由来酸性プロテアーゼが挙げられる。
糸状菌由来プロテアーゼの使用において、上記の糸状菌の培養液、その破砕液及び抽出物を用いてもよいし、それらを任意のレベルで精製しプロテアーゼの含有量を高めた精製物を用いてもよい。また、糸状菌由来プロテアーゼの使用において、市販の酵素製剤(例えば、天野エンザイム株式会社製のアスペルギルス・オリゼ由来酸性プロテアーゼ、アスペルギルス・オリゼ由来中性プロテアーゼ及びアスペルギルス・メレウス由来中性プロテアーゼ等)を用いてもよい。
細菌プロテアーゼとしては特に限定されないが、バチルス(Bacillus)属及びジオバチルス(Geobacillus)属等に由来するプロテアーゼが挙げられる。
バチルス属(又はジオバチルス属)由来プロテアーゼの具体例としては、バチルス・アミロリケファシエンス(Bacillus amyloliquefaciens)、バチルス・セレウス(Bacillus cereus)、バチルス・クラウジー(Bacillus clausii)、バチルス・インターミディウス(Bacillus intermedius)、バチルス・レンタス(Bacillus lentus)、バチルス・リケニフォルミス(Bacillus licheniformis)、バチルス・ステアロサーモフィラス(Bacillus stearothermophilus)、バチルス・サブティリス(Bacillus subtilis)、及びバチルス・サーモプロテオリティカス(Bacillus thermoproteolyticus)、及びこれらのジオバチルス属由来のプロテアーゼが挙げられる。
これらのバチルス属(又はジオバチルス属)由来プロテアーゼは、1種を単独で用いてもよいし、複数種を組み合わせて用いてもよい。これらのバチルス属(又はジオバチルス属)由来プロテアーゼの中でも、組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくはバチルス・ステアロサーモフィラス、ジオバチルス・ステアロサーモフィラスが挙げられる。
細菌由来プロテアーゼの使用において、上記の細菌の培養液、その破砕液及び抽出物を用いてもよいし、それらを任意のレベルで精製しプロテアーゼの含有量を高めた精製物を用いてもよい。また、細菌由来プロテアーゼの使用において、市販の酵素製剤(例えば、天野エンザイム株式会社製のジオバチルス・ステアロサーモフィラス由来プロテアーゼ等)を用いてもよい。
プロテアーゼの使用量(総量)としては特に限定されないが、植物性タンパク質1g当たりの使用量として、例えば4U以上が挙げられる。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、プロテアーゼのタンパク質1g当たりの使用量として、好ましくは40U以上、より好ましくは400U以上、さらに好ましくは1,000U以上、一層好ましくは2,000U以上、より一層好ましくは3,000U以上が挙げられる。プロテアーゼの使用量(総量)は、その上限においても特に限定されないが、植物性タンパク質1g当たりの使用量として、例えば100,000U以下、又は50,000U以下、好ましくは10,000U以下、より好ましくは8,000U以下、又は6,000U以下が挙げられる。
また、プロテアーゼの植物性タンパク質材料1g当たりの使用量(総量)としては、例えば3.2U以上が挙げられる。組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、プロテアーゼのタンパク質材料1g当たりの使用量として、好ましくは32U以上、より好ましくは320U以上、さらに好ましくは800U以上、一層好ましくは1,600U以上、より一層好ましくは2,400U以上が挙げられる。プロテアーゼの使用量(総量)は、その上限においても特に限定されないが、植物性タンパク質材料1g当たりの使用量として、例えば80,000U以下、又は40,000U以下、好ましくは8,000U以下、より好ましくは6,400U以下、又は4,800U以下が挙げられる。
また、糸状菌由来プロテアーゼ及び細菌由来プロテアーゼの両方を組み合わせて用いる場合、その比率については特に限定されないが、糸状菌由来プロテアーゼ1U当たりの細菌由来プロテアーゼの使用量として、例えば0.01~2U、好ましくは0.05~1U、より好ましくは0.1~0.7U、さらに好ましくは0.2~0.5U、一層好ましくは0.3~0.4Uが挙げられる。
プロテアーゼの活性については、カゼインを基質とし、1分間にチロシン1μgに相当するフォリン試液呈色物質の増加をもたらす酵素量を1単位(1U)とする。
1-1-4.酵素処理反応
非組織化状の植物性タンパク質材料にグルタミナーゼ類及びプロテアーゼを作用させる酵素処理反応は、通常、非組織化状の植物性タンパク質材料とグルタミナーゼ類とプロテアーゼとを水中に含む酵素処理用混合液を、当該酵素の反応条件に供することにより行うことができる。
非組織化状の植物性タンパク質材料にグルタミナーゼ類及びプロテアーゼを作用させる酵素処理反応は、通常、非組織化状の植物性タンパク質材料とグルタミナーゼ類とプロテアーゼとを水中に含む酵素処理用混合液を、当該酵素の反応条件に供することにより行うことができる。
酵素処理用混合液中の植物性タンパク質材料の乾燥重量としては特に限定されないが、例えば1~30重量%、好ましくは3~20重量%、より好ましくは5~15重量%、さらに好ましくは8~12重量%が挙げられる。
なお、グルタミナーゼ類による酵素処理とプロテアーゼによる酵素処理は、同時に行ってもよいし、段階的に行ってもよい。これら酵素処理を同時に行う場合、非組織化状の植物性タンパク質材料とグルタミナーゼ類とプロテアーゼとを水中に含む酵素処理用混合液を調製し、その後、当該混合液を当該酵素の反応条件に供する。これら酵素処理を段階的に行う場合は、非組織化状の植物性タンパク質材料と、グルタミナーゼ類及びプロテアーゼのいずれか一方とを水中に含む第1の酵素処理用混合液を調製し、その後、当該第1の混合液を当該一方の酵素の反応条件に供し、その後、第1の酵素処理用混合液に、グルタミナーゼ類及びプロテアーゼのいずれか他方を加えて第2の酵素処理用混合液を調製し、その後、当該第2の混合液を当該他方の酵素の反応条件に供することができる。
反応条件(温度及びpH)については、用いる酵素の熱特性及びpH特性に応じて適宜設定することができる。反応温度の具体例としては、例えば15℃~70℃、好ましくは30℃~65℃、更に好ましくは40℃~60℃が挙げられる。酵素処理用混合液のpH(25℃)の具体例としては、例えばpH3~11、好ましくはpH4~10、より好ましくはpH5~9、更に好ましくはpH6~8が挙げられる。また、反応時間については、例えば分~48時間、好ましくは10分~24時間、更に好ましくは15分~12時間が挙げられる。
1-1-5.酵素処理された植物性タンパク質材料(酵素処理物)
工程1で得られる酵素処理された植物性タンパク質材料(以下において、「酵素処理物」とも記載する。)の形態については、工程2に適用可能な形態であれば特に限定されない。酵素処理物の具体的な形態としては、液状、ペースト状、固体状(例えば、粉末状、顆粒状等)等が挙げられ、好ましくは固体状が挙げられ、より好ましくは粉末状が挙げられる。
工程1で得られる酵素処理された植物性タンパク質材料(以下において、「酵素処理物」とも記載する。)の形態については、工程2に適用可能な形態であれば特に限定されない。酵素処理物の具体的な形態としては、液状、ペースト状、固体状(例えば、粉末状、顆粒状等)等が挙げられ、好ましくは固体状が挙げられ、より好ましくは粉末状が挙げられる。
酵素処理物は、組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみを抑制することができる。
1-2.工程2
工程2では、工程1で得られた酵素処理された植物性タンパク質材料(酵素処理物)を含む組織化用材料を組織化処理する。

1-2-1.組織化用材料
組織化用材料は、工程1で得られた酵素処理された植物性タンパク質材料(酵素処理物)を含み、組織化可能な混錬物として調製されることを限度として、その組成について特に限定されるものではない。

組織化用材料に配合できる酵素処理物以外の成分としては、植物性タンパク質材料(工程1の酵素処理を経ておらず、且つ組織化されていないもの)、多糖類、水、油等が挙げられる。
組織化用材料に配合できる植物性タンパク質材料の具体例としては、上記工程1における非組織化状の植物性タンパク質材料の具体例から1種又は複数種を選択することができ、好ましくは菽穀類、より好ましくは大豆、えんどう、さらに好ましくはえんどうに由来する植物性タンパク質材料が挙げられる。多糖類としては、デキストリン、セルロース、デンプン、食物繊維等から1種又は複数種を選択することができ、好ましくはデキストリンが挙げられる。油としては、好ましくは植物油が挙げられる。
組織化用材料の100重量部(乾燥重量)に対する酵素処理物の含有量(乾燥重量)としては、組織化植物性タンパク質の色のくすみを抑制する効果の求める程度に応じて適宜設定することができる。組織化用材料の100重量部(乾燥重量)に対する酵素処理物の含有量(乾燥重量)の具体例としては、例えば3重量部以上が挙げられ、組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくは5重量部以上、より好ましくは10重量部以上、さらに好ましくは13重量部以上、一層好ましくは15重量部以上が挙げられる。組織化用材料の100重量部(乾燥重量)に対する酵素処理物の含有量(乾燥重量)は、その上限においても特に限定されず、例えば100重量部以下、80重量部以下、60重量部以下、40重量部以下、又は30重量部以下が挙げられる。
組織化用材料に植物性タンパク質材料を含む場合、組織化用材料の100重量部(乾燥重量)に対する植物性タンパク質材料の含有量(乾燥重量)としては特に限定されないが、例えば40重量部以上、50重量部以上、60重量部以上、65重量部以上、70重量部以上、又は75重量部以上が挙げられる。組織化用材料の100重量部(乾燥重量)に対する植物性タンパク質材料の含有量(乾燥重量)は、その上限においても特に限定されないが、例えば90重量部以下、好ましくは85重量部以下、より好ましくは80重量部以下、さらに好ましくは75重量部以下が挙げられる。
組織化用材料に植物性タンパク質材料を含む場合、植物性タンパク質材料100重量部(乾燥重量)に対する酵素処理物の含有量(乾燥重量)としては、例えば5重量部以上が挙げられ、組織化植物性タンパク質の色のくすみ抑制効果をより一層高める観点から、好ましくは10重量部以上、より好ましくは15重量部以上、さらに好ましくは18重量部以上、一層好ましくは20重量部以上が挙げられる。植物性タンパク質材料100重量部(乾燥重量)に対する酵素処理物の含有量(乾燥重量)は、その上限においても特に限定されず、例えば80重量部以下、60重量部以下、40重量部以下、30重量部以下、又は25重量部以下が挙げられる。
組織化用材料に植物性タンパク質材料を含む場合、組織化用材料100重量部(乾燥重量)に対する植物性タンパク質材料及び酵素処理物の総量(乾燥重量)としては、例えば50重量部以上、好ましくは60重量部以上、より好ましくは70重量部以上、さらに好ましくは80重量部以上、一層好ましくは83重量部以上が挙げられる。組織化用材料100重量部(乾燥重量)に対する植物性タンパク質材料及び酵素処理物の総量(乾燥重量)は、その上限においても特に限定されず、例えば95重量部以下、90重量部以下、又は87重量部以下が挙げられる。
組織化用材料に多糖類を含む場合、組織化用材料の100重量部(乾燥重量)に対する多糖類の含有量(乾燥重量)としては特に限定されないが、例えば5重量部以上、好ましくは10重量部以上、より好ましくは13重量部以上が挙げられる。組織化用材料の100重量部(乾燥重量)に対する多糖類の含有量(乾燥重量)は、その上限においても特に限定されないが、例えば30重量部以下、好ましくは25重量部以下、より好ましくは20重量部以下、さらに好ましくは18重量部以下が挙げられる。
1-2-2.組織化処理
組織化処理の具体的な手法については、組織化植物性タンパク質が得られる方法であれば特に限定されず、具体的例として、エクストルーダーを用いた高温・高圧での押出が挙げられる。押出された材料は、更に、乾燥及び/又は冷凍、好ましくは凍結乾燥の処理に供することもできる。
組織化処理の具体的な手法については、組織化植物性タンパク質が得られる方法であれば特に限定されず、具体的例として、エクストルーダーを用いた高温・高圧での押出が挙げられる。押出された材料は、更に、乾燥及び/又は冷凍、好ましくは凍結乾燥の処理に供することもできる。
1-2-3.組織化植物性タンパク質
本発明により得られる組織化植物性タンパク質は、一般的に代替肉(擬似肉)として利用できるものである。本発明により得られる組織化植物性タンパク質が模する「肉」とは、食用とされる動物の筋肉を意味し、「肉」と記載する場合、哺乳類及び鳥類の筋肉だけでなく、魚介の身も包含する意味で用いる。
本発明により得られる組織化植物性タンパク質は、一般的に代替肉(擬似肉)として利用できるものである。本発明により得られる組織化植物性タンパク質が模する「肉」とは、食用とされる動物の筋肉を意味し、「肉」と記載する場合、哺乳類及び鳥類の筋肉だけでなく、魚介の身も包含する意味で用いる。
組織化植物性タンパク質の形状としては、粒状及び繊維状が挙げられる。粒状の形状には、小粒型(ミンチ)、大粒型、ブロック型等の様々な大きさ(小粒型、大粒型、ブロック型の順にサイズが大きくなる)の塊状形状;フレーク型、フィレ型、スライス型等の様々な大きさ(フレーク型、フィレ型、スライス型の順にサイズが大きくなる)の扁平状形状が挙げられる。
2.組織化植物性タンパク質を含む食品
本発明は、上記「1.組織化植物性タンパク質の製造方法」によって得られる組織化植物性タンパク質を含む食品も提供する。

本発明の食品の具体的な形態については、食品形態から選択でき、例えば、畜肉、鳥肉、及び/又は魚すり身加工食品に準拠することができる。つまり、本発明の食品としては、肉様加工食品(畜肉、鳥肉、及び/又は魚すり身加工食品を模した食品を指す。)が挙げられ、より好ましくは、畜肉及び/又は鳥肉様加工食品(畜肉及び/又は鳥肉加工食品を模した食品を指す。)が挙げられる。このような畜肉及び/又は鳥肉加工食品は、畜肉及び/又は鳥肉を用いた肉種を成形し加熱することで調理される食品であればよく、その具体例としては、ハンバーグ、ミートボール、パティ、ミートローフ、ミンチカツ、点心等が挙げられる。
本発明の食品は、上記の製造方法によって得られる組織化植物性タンパク質を任意の調理工程に供することによって得ることができる。調理工程においては、膨潤、着色、調味、形態調整、成形、加熱調理、発酵、及び/又は冷凍等が行われ得る。調理工程における具体的な方法としては、必要に応じて水で膨潤し、並びに/若しくは、着色料その他の添加物及び/又は調味料を加えて所望の形状に成型し、さらに必要に応じて加熱調理する方法が挙げられる。加熱調理の具体的な方法については、食品の種類に応じて当業者が適宜決定することができ、具体的には、煮沸、焼成(ロースト、トースト、ベイク、グリル、ブロイル)、蒸し、及び/又は揚げ等が行われる。
3.組織化植物性タンパク質の色のくすみ抑制剤
上述の通り、上記「1.組織化植物性タンパク質の製造方法」の工程1で得られる酵素処理された植物性タンパク質材料(酵素処理物)は、組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみを抑制することができる。従って、本発明は、非組織化状の植物性タンパク質材料の、グルタミナーゼ類及びプロテアーゼによる酵素処理物を含む、組織化植物性タンパク質の色のくすみ抑制剤も提供する。
上述の通り、上記「1.組織化植物性タンパク質の製造方法」の工程1で得られる酵素処理された植物性タンパク質材料(酵素処理物)は、組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみを抑制することができる。従って、本発明は、非組織化状の植物性タンパク質材料の、グルタミナーゼ類及びプロテアーゼによる酵素処理物を含む、組織化植物性タンパク質の色のくすみ抑制剤も提供する。
組織化植物性タンパク質の色のくすみ抑制とは、組織化植物性タンパク質(上記「1.組織化植物性タンパク質の製造方法」の工程1で得られる酵素処理物を用いずに製造されるもの)の外観が元来呈する灰褐色の色味の明度及び黄味が増すことをいう。具体的には、JIS(JIS Z 8781-4)に準拠するL*値及びb*値が増大することによって確認することができる。
組織化植物性タンパク質の色のくすみ抑制剤において、使用する成分、使用量、具体的な使用方法等については、前記「1.組織化植物性タンパク質の製造方法」の欄に示す通りである。
4.組織化植物性タンパク質の色の着色抑制剤
組織化植物性タンパク質が呈する灰褐色の色味は、組織化形態に成形されるまでの少なくともいずれかの過程で、植物材料中に含まれる糖とアミノ酸とのメイラード反応により生じた褐色物質による着色の結果と考えられる。従って、上記「1.組織化植物性タンパク質の製造方法」の工程1で得られる酵素処理された植物性タンパク質材料(酵素処理物)を組織化用材料に配合することで、得られる組織化植物性タンパク質の色のくすみを抑制するのは、当該酵素処理で用いられたグルタミナーゼ類及びプロテアーゼの組み合わせが、植物材料中に含まれる糖とアミノ酸とのメイラード反応を抑制することにより、組織化植物性タンパク質の着色を抑制したものと考えられる。従って、本発明は、グルタミナーゼ類及びプロテアーゼを含む、組織化植物性タンパク質の着色抑制剤も提供する。

組織化植物性タンパク質の着色抑制剤において、使用する成分、使用量、具体的な使用方法等については、前記「1.組織化植物性タンパク質の製造方法」の欄に示す通りである。
以下の実施例により本発明を具体的に説明するが、本発明は実施例によって限定されるものではない。
(1)材料
(1-1)非組織化状植物性タンパク質材料
・エンドウタンパク質:PURIS Pea870(PURIS製、タンパク質含量80重量%)
(1-2)酵素
・グルタミナーゼ類:バチルス・アミロリケファシエンス由来グルタミナーゼ(EC3.5.1.2)(天野エンザイム株式会社)
・糸状菌由来プロテアーゼ:アスペルギルス・オリゼ由来酸性プロテアーゼ(天野エンザイム株式会社)
・細菌由来プロテアーゼ:ジオバチルス・ステアロサーモフィラス由来プロテアーゼ(天野エンザイム株式会社)
(1-1)非組織化状植物性タンパク質材料
・エンドウタンパク質:PURIS Pea870(PURIS製、タンパク質含量80重量%)
(1-2)酵素
・グルタミナーゼ類:バチルス・アミロリケファシエンス由来グルタミナーゼ(EC3.5.1.2)(天野エンザイム株式会社)
・糸状菌由来プロテアーゼ:アスペルギルス・オリゼ由来酸性プロテアーゼ(天野エンザイム株式会社)
・細菌由来プロテアーゼ:ジオバチルス・ステアロサーモフィラス由来プロテアーゼ(天野エンザイム株式会社)
(2)酵素活性値測定法
(2-1)プロテアーゼ活性測定法
0.6%(w/v)カゼイン溶液(0.05mol/Lリン酸水素ナトリウム、pH8.0[細菌由来プロテアーゼの場合])若しくは0.6%(w/v)カゼイン溶液(0.7%(w/v)乳酸、pH3.0[糸状菌由来プロテアーゼの場合])の5mLを、37℃で10分間加温した後、プロテアーゼを含む試料溶液1mLを加え、直ちに振り混ぜた。この液を37℃で10分間放置した後、トリクロロ酢酸試液(1.8%(w/v)トリクロロ酢酸、1.8%(w/v)酢酸ナトリウム及び0.33mol/L酢酸を含む水溶液)5mLを加えて振り混ぜ、再び37℃で30分間放置し、ろ過した。初めのろ液3mLを除き、次のろ液2mLを量り、0.55mol/L炭酸ナトリウム試液5mL及びフォリン試液(1→3)1mLを加え、よく振り混ぜ、37℃で30分間放置した。この液(酵素反応液)につき、水を対照とし、波長660nmにおける吸光度ATを測定した。
(2-1)プロテアーゼ活性測定法
0.6%(w/v)カゼイン溶液(0.05mol/Lリン酸水素ナトリウム、pH8.0[細菌由来プロテアーゼの場合])若しくは0.6%(w/v)カゼイン溶液(0.7%(w/v)乳酸、pH3.0[糸状菌由来プロテアーゼの場合])の5mLを、37℃で10分間加温した後、プロテアーゼを含む試料溶液1mLを加え、直ちに振り混ぜた。この液を37℃で10分間放置した後、トリクロロ酢酸試液(1.8%(w/v)トリクロロ酢酸、1.8%(w/v)酢酸ナトリウム及び0.33mol/L酢酸を含む水溶液)5mLを加えて振り混ぜ、再び37℃で30分間放置し、ろ過した。初めのろ液3mLを除き、次のろ液2mLを量り、0.55mol/L炭酸ナトリウム試液5mL及びフォリン試液(1→3)1mLを加え、よく振り混ぜ、37℃で30分間放置した。この液(酵素反応液)につき、水を対照とし、波長660nmにおける吸光度ATを測定した。
別に、プロテアーゼを含む試料溶液1mLを量り、トリクロロ酢酸試液5mLを加えて振り混ぜた後、試料ごとに設定された測定pHのカゼイン溶液5mLを加え、直ちに振り混ぜ、37℃で30分間放置したことを除いて上述の酵素反応液と同様に操作した液(ブランク)について、吸光度ABを測定した。
1分間にチロシン1μgに相当するフォリン試液呈色物質の増加をもたらす酵素量を1単位(1U)とした。
1mg/mLチロシン標準原液(0.2mol/L塩酸)1mL,2mL,3mL及び4mLを量り、それぞれに0.2mol/L塩酸試液を加え、100mLとした。それぞれの液2mLを量り、0.55mol/L炭酸ナトリウム試液5mL及びフォリン試液(1→3)1mLを加え、直ちに振り混ぜ、37℃で30分間放置した。これらの液につき、0.2mol/L塩酸試液2mLを量り上記と同様に操作して得た液を対照とし、波長660nmにおける吸光度A1,A2,A3及びA4を測定した。縦軸に吸光度A1,A2,A3及びA4を、横軸にそれぞれの液2mL中のチロシン量(μg)をとり、検量線を作成し、吸光度差1に対するチロシン量(μg)を求めた。
(2-2)グルタミナーゼ類活性測定法
酵素溶液1mLを試験管に量りとり、37℃の恒温水槽に5分間放置した。これにあらかじめ37℃に予熱した2%(w/v)L-グルタミン溶液(0.1mol/L酢酸緩衝液(pH6.0))1mLを加えて混ぜ、正確に10分間放置した。放置後、5%(v/v)過塩素酸試液1mLを加えて混ぜ、直ちに氷水中に入れた。1分間以上放置した後、水酸化ナトリウム試液(0.75mol/L)1mLを加えて混ぜ、反応液とした。反応液のL-グルタミン酸を、L-グルタミン酸測定キット「ヤマサ」NEO(ヤマサ醤油製)を用いて定量した。本条件下、1分間に1μmolのL-グルタミン酸を生成する酵素量を1単位とした。
酵素溶液1mLを試験管に量りとり、37℃の恒温水槽に5分間放置した。これにあらかじめ37℃に予熱した2%(w/v)L-グルタミン溶液(0.1mol/L酢酸緩衝液(pH6.0))1mLを加えて混ぜ、正確に10分間放置した。放置後、5%(v/v)過塩素酸試液1mLを加えて混ぜ、直ちに氷水中に入れた。1分間以上放置した後、水酸化ナトリウム試液(0.75mol/L)1mLを加えて混ぜ、反応液とした。反応液のL-グルタミン酸を、L-グルタミン酸測定キット「ヤマサ」NEO(ヤマサ醤油製)を用いて定量した。本条件下、1分間に1μmolのL-グルタミン酸を生成する酵素量を1単位とした。
試験例
1.酵素処理された植物性タンパク質材料(酵素処理物粉末)の調製
エンドウタンパク質を約10重量%になるように水に懸濁し、撹拌しながら50℃に調整した。そして酵素(バチルス・アミロリケファシエンス由来グルタミナーゼ 1U/gタンパク質、アスペルギルス・オリゼ由来酸性プロテアーゼ 3,000U/gタンパク質、ジオバチルス・ステアロサーモフィラス由来プロテアーゼ 1,000U/gタンパク質)をエンドウタンパク質原料に添加し、50℃で1時間反応させた。その後、85℃に昇温してから10分間保持して酵素を失活させた。得られた酵素処理タンパク質液をスプレードライヤー(モデル:Henningsen Pilot Plant Tower Spray Dryer Model T-20、入口温度:140~150℃、出口温度:90~100℃)で粉末化することで、酵素処理された植物性タンパク質材料(酵素処理物粉末)を得た。
1.酵素処理された植物性タンパク質材料(酵素処理物粉末)の調製
エンドウタンパク質を約10重量%になるように水に懸濁し、撹拌しながら50℃に調整した。そして酵素(バチルス・アミロリケファシエンス由来グルタミナーゼ 1U/gタンパク質、アスペルギルス・オリゼ由来酸性プロテアーゼ 3,000U/gタンパク質、ジオバチルス・ステアロサーモフィラス由来プロテアーゼ 1,000U/gタンパク質)をエンドウタンパク質原料に添加し、50℃で1時間反応させた。その後、85℃に昇温してから10分間保持して酵素を失活させた。得られた酵素処理タンパク質液をスプレードライヤー(モデル:Henningsen Pilot Plant Tower Spray Dryer Model T-20、入口温度:140~150℃、出口温度:90~100℃)で粉末化することで、酵素処理された植物性タンパク質材料(酵素処理物粉末)を得た。
2.組織化植物性タンパク質の調製
酵素処理物粉末を用いて下表に示す組成物に水を適量加えて混錬し、エクストルーダー(Wenger Manufacturing TX-57 model)で組織化(圧力:150-175psi、ステージ1温度:60℃、ステージ2温度:80℃、ステージ3温度:90℃、ステージ4温度:100℃)し、組織化タンパク質(粒状)を得た。
酵素処理物粉末を用いて下表に示す組成物に水を適量加えて混錬し、エクストルーダー(Wenger Manufacturing TX-57 model)で組織化(圧力:150-175psi、ステージ1温度:60℃、ステージ2温度:80℃、ステージ3温度:90℃、ステージ4温度:100℃)し、組織化タンパク質(粒状)を得た。
3.組織化植物性タンパク質の色調評価
上記方法で得られた組織化植物タンパク質(乾燥状態)について、分光測色計(CM-700d:コニカミノルタセンシング(株))を用いてL*a*b*値を測定し、色調を数値化した。L*値は明度を表し、大きいほど白に近く、小さいほど黒に近い。a*値は色度を表し、大きいほど赤味が強く、小さいほど緑味が強い。b*値は色度を表し、大きいほど黄味が強く、小さいほど青味が強い。結果を表2に示す。また、上記方法で得られた組織化植物タンパク質(乾燥状態)の外観写真を図1に示す。
上記方法で得られた組織化植物タンパク質(乾燥状態)について、分光測色計(CM-700d:コニカミノルタセンシング(株))を用いてL*a*b*値を測定し、色調を数値化した。L*値は明度を表し、大きいほど白に近く、小さいほど黒に近い。a*値は色度を表し、大きいほど赤味が強く、小さいほど緑味が強い。b*値は色度を表し、大きいほど黄味が強く、小さいほど青味が強い。結果を表2に示す。また、上記方法で得られた組織化植物タンパク質(乾燥状態)の外観写真を図1に示す。
表2に示されるように、酵素処理タンパク質粉末を用いずに製造した比較例1の組織化植物性タンパク質に比べ、酵素処理物粉末を組織化用材料に配合して製造した実施例1及び2の組織化植物性タンパク質では、L*値およびb*値が高くなり、その傾向は、酵素処理タンパク質粉末の配合比率が大きいほど高いことが判った。すなわち、酵素処理タンパク質粉末を用いずに製造した比較例1の組織化植物性タンパク質が灰褐色であることに対し、酵素処理物粉末を組織化用材料に配合して製造した実施例1及び2の組織化植物性タンパク質では、明度と黄味が増し、灰褐色のくすみが抑制されていた。明度と黄味が増すことで組織化植物性タンパク質の色のくすみが抑制されていることは、図1に示すように、組織化植物性タンパク質の外観からも目視で確認できた。
Claims (13)
- 非組織化状の植物性タンパク質材料にグルタミナーゼ類及びプロテアーゼを作用させ、酵素処理された植物性タンパク質材料を得る工程1と、前記酵素処理された植物性タンパク質材料を含む組織化用材料を組織化処理する工程2と、を含む、組織化植物性タンパク質の製造方法。
- 前記組織化用材料の乾燥重量100重量部に対する前記酵素処理された植物性タンパク質材料の乾燥重量が、3重量部以上である、請求項1に記載の製造方法。
- 前記グルタミナーゼ類がグルタミナーゼである、請求項1に記載の製造方法。
- 前記グルタミナーゼ類がバチルス属由来グルタミナーゼ類である、請求項1に記載の製造方法。
- 前記プロテアーゼが糸状菌由来プロテアーゼ及び/又は細菌由来プロテアーゼである、請求項1に記載の製造方法。
- 前記糸状菌プロテアーゼが酸性プロテアーゼである、請求項5に記載の製造方法。
- 前記糸状菌プロテアーゼがアスペルギルス属由来プロテアーゼである、請求項5に記載の製造方法。
- 前記細菌プロテアーゼがバチルス属又はジオバチルス属由来プロテアーゼである、請求項5に記載の製造方法。
- 前記植物性タンパク質材料が菽穀類由来である、請求項1に記載の製造方法。
- 前記タンパク質材料がエンドウ由来である、請求項1に記載の製造方法。
- 請求項1に記載の製造方法により製造される組織化植物性タンパク質を含む食品。
- 非組織化状の植物性タンパク質材料の、グルタミナーゼ類及びプロテアーゼによる酵素処理物を含む、組織化植物性タンパク質の色のくすみ抑制剤。
- グルタミナーゼ類及びプロテアーゼを含む、組織化植物性タンパク質の着色抑制剤。
