Abstract
支持体層と粘着剤層とを備えるパップ剤であり、前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤。
Description
本発明は、パップ剤及びその製造方法に関する。
パップ剤は水溶性高分子及び水を含有する粘着剤層を備える貼付剤であり、薬物による薬効に加えて、冷却効果や温感刺激効果も期待できる製剤として汎用されている。
そのためパップ剤としては、薬物として、例えば、ジクロフェナク、ロキソプロフェン、インドメタシン等の抗炎症作用を有する薬物を含有するパップ剤が主に知られている。このようなパップ剤としては、例えば、特開平9-208462号公報(特許文献1)において、パップ剤基剤に、ジクロフェナクナトリウム、水、及び脂肪酸ジアルキロールアミドを配合したパップ剤に、ポリブテン及びゼラチンを添加した自己粘着型パップ剤が記載されている。
また、他にも様々なパップ剤が開発されており、例えば、特開平4-29927号公報(特許文献2)には、粘着剤層が、粘着基剤、粘着基剤に対して可塑化作用を有する化合物、無水ケイ酸、薬物、及び吸水性充填剤を含有する貼付剤が記載されている。さらに、例えば、特開平6-145049号公報(特許文献3)には、ポリアクリル酸及び/又はポリアクリル酸塩を主体とした架橋型ハップ(パップ)基剤において無水ケイ酸を配合するハップ剤が記載されている。
パップ剤は上記のように水を含有するため、一般に、非水系のテープ剤等に比べて皮膚への付着性に劣り、貼付中に剥がれやすいという欠点を有する。そのため、本発明者らがパップ剤の皮膚への付着性についてさらなる検討を行ったところ、皮膚への付着性を向上させようとすると、皮膚から剥離した際に皮膚に粘着剤層又は粘着剤層の一部若しくは粘着剤層に含有される成分の一部が残る現象、すなわち「べたつき」が生じる場合があることを見出した。
さらに、パップ剤の粘着剤層には、その保形性を向上させること等を目的として、無機充填剤を含有させることが知られている。このような無機充填剤としては、他の成分との混合時の均一性に優れるという観点から、従来、例えば、特許文献2に記載されているアエロジルA-200(日本アエロジル株式会社製)のように粒子径や嵩密度の小さい無水ケイ酸が好ましく用いられていた。しかしながら、このように粒子径や嵩密度の小さい無機充填剤では粉塵が発生しやすい等、製造時の取り扱いが困難であることから、本発明者らがパップ剤の無機充填剤についてもさらなる検討を行ったところ、その種類や粒子径によっては、製造時に凝集物が生成しやすくなって均一な外観の粘着剤層を得ることが困難になるという問題が生じることを見出した。さらには、かかる無機充填剤の種類や粒子径によっては、上記の皮膚への付着性が低下したり、べたつきが生じたりする場合があるという問題が生じることも見出した。
本発明は、上記従来技術の有する課題に鑑みてなされたものであり、皮膚への付着性に優れるとともに剥離後のべたつきの発生が十分に抑制され、かつ、粘着剤層の外観に優れたパップ剤、及び前記パップ剤を容易に得ることができるパップ剤の製造方法を提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意研究を重ねた結果、支持体層と粘着剤層とを備えるパップ剤において、前記粘着剤層に、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種と、無水ケイ酸とを組み合わせて含有させ、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50を特定の範囲内とすることによって、皮膚への付着性が十分に向上するとともに、剥離後のべたつきの発生も十分に抑制されることを見出した。
さらに、本発明者らは、このようなカルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種と、無水ケイ酸との組み合わせによれば、従来好ましく用いられていた粒子径や嵩密度の小さい無水ケイ酸を用いた場合に比べて製造時の無水ケイ酸の粉塵発生を抑制することが可能となることに加え、凝集物の生成も十分に抑制され、均一な外観の粘着剤層を容易に得ることが可能となることも見出し、本発明を完成するに至った。かかる知見により得られた本発明の態様は次のとおりである。

前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤。
前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤。
[2]前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩の含有量が、ナトリウム塩換算で、前記粘着剤層の全質量に対して1.0~4.0質量%である、[1]に記載のパップ剤。
[3]前記無水ケイ酸の含有量が、前記粘着剤層の全質量に対して0.1~6.0質量%である、[1]又は[2]に記載のパップ剤。
[4]前記粘着剤層における前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量と前記無水ケイ酸の含有量との質量比(カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量:無水ケイ酸の含有量)が、1:2~6:0.5である、[1]~[3]のうちのいずれか一項に記載のパップ剤。

本発明によれば、皮膚への付着性に優れるとともに剥離後のべたつきの発生が十分に抑制され、かつ、粘着剤層の外観に優れたパップ剤、及び前記パップ剤を容易に得ることができるパップ剤の製造方法を提供することが可能となる。
以下、本発明をその好適な実施形態に即して詳細に説明する。
<パップ剤>
本発明のパップ剤は、支持体層と粘着剤層とを備えるパップ剤であり、前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤である。
本発明のパップ剤は、支持体層と粘着剤層とを備えるパップ剤であり、前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤である。
本発明のパップ剤は、支持体層を備える。本発明に係る支持体層としては、後述の粘着剤層を支持し得るものであれば特に制限されず、パップ剤の支持体層として公知のものを適宜採用することができる。本発明に係る支持体層の材質としては、例えば、ポリエチレン、ポリプロピレン等のポリオレフィン;エチレン-酢酸ビニル共重合体、酢酸ビニル-塩化ビニル共重合体、ポリ塩化ビニル等;ナイロン等のポリアミド;ポリエチレンテレフタレート(PET)、ポリブチレンテレフタレート、ポリエチレンナフタレート等のポリエステル;セルロース誘導体;ポリウレタンなどの合成樹脂や、アルミニウムなどの金属が挙げられる。本発明に係る支持体層としては、フィルム、シート、布帛、箔、及びこれらの積層体が挙げられるが、中でも、前記材質の繊維からなる布帛を用いることが好ましく、前記布帛としては、前記繊維を編成、絡合、熱融着、圧着あるいはバインダー接着等の方法により加工した織布、不織布を用いることがより好ましい。
このような支持体層としては、例えば、ポリエステル繊維からなる不織布を用いることが好ましく、前記不織布の目付としては、50~200g/m2であることがより好ましい。前記不織布の目付が前記下限未満である場合には、パップ剤を剥がす際に破れやすいといった強度上の問題が発生したり、支持体層の裏面に粘着剤層に含有される成分が浸み出す現象、すなわち「染み出し」が発生することによりパップ剤の外観や使用感の悪化を招くといった問題が生じる傾向にある。他方、前記上限を超える場合には、支持体層の伸縮性や柔軟性が不足するため、パップ剤が剥がれやすくなる傾向にある。
本発明のパップ剤としては、パップ剤を使用する時まで前記粘着剤層の表面を被覆して保護すること等を目的として、前記粘着剤層の前記支持体層とは反対の面上に剥離ライナーをさらに備えていてもよい。前記剥離ライナーの材質としては、特に制限されず、ポリエチレン、ポリプロピレン等のポリオレフィン;エチレン-酢酸ビニル共重合体、酢酸ビニル-塩化ビニル共重合体、ポリ塩化ビニル等;ナイロン等のポリアミド;ポリエチレンテレフタレート等のポリエステル;セルロース誘導体;ポリウレタンなどの合成樹脂や、アルミニウム、紙が挙げられる。前記剥離ライナーとしては、前記材質からなるフィルムやシート及びこれらの積層体が挙げられる。また、前記剥離ライナーとしては、前記粘着剤層から容易に剥離できるように該粘着剤層と接触する側の面に含シリコーン化合物コートや含フッ素化合物コート等の離型処理が施されたものであることが好ましい。これらの中でも、前記剥離ライナーとしては、ポリエチレンテレフタレート、ポリプロピレンからなるフィルムを用いることが好ましい。また、このような剥離ライナーの厚みとしては、特に制限されないが、例えば、20~150μmであることが好ましい。
本発明のパップ剤は、前記支持体層の面上(通常は一方の面上)に粘着剤層を備える。本発明に係る粘着剤層としては、特に制限されないが、単位面積(貼付面の面積)あたりの質量が430~1900g/m2であることが好ましく、665~1500g/m2であることがより好ましく、900~1100g/m2であることがさらに好ましい。また、本発明に係る粘着剤層の貼付面の面積としては、薬物の種類や含有量、治療の目的や適用対象等に応じて適宜調整されるものであるため特に限定されるものではないが、通常、70~280cm2の範囲である。
(カルボキシメチルセルロース及びその薬学的に許容される塩)
本発明に係る粘着剤層は、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種(本明細書中、場合により「カルボキシメチルセルロース及び/又はその薬学的に許容される塩」という)を含有する。
本発明に係る粘着剤層は、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種(本明細書中、場合により「カルボキシメチルセルロース及び/又はその薬学的に許容される塩」という)を含有する。
カルボキシメチルセルロースは、カルメロースとも称されるセルロースの誘導体であり、セルロースの一部のヒドロキシ基にカルボキシメチル基(-CH2-COOH)がエーテル結合した構造を有する。理論的にエーテル化度は0~3であるが、本発明に係るカルボキシメチルセルロースとしては、エーテル化度が0.5~1.5であることが好ましい。
また、本発明に係るカルボキシメチルセルロースとしては、特に制限されるものではないが、例えば、25℃において1.0%水溶液、回転60rpmで測定される粘度が、10~15,000mPa・sであることが好ましく、10~6,000mPa・sであることがより好ましく、10~3,000mPa・sであることがさらに好ましい。
本発明において前記粘着剤層中におけるカルボキシメチルセルロースの形態としては、遊離体(フリー体)であってもその薬学的に許容される塩であってもよく、製造中及び/又は製造された製剤中においてカルボキシメチルセルロースの薬学的に許容される塩が脱塩されて遊離体となったものであってもよく、これらのうちの1種であっても2種以上の組み合わせであってもよい。カルボキシメチルセルロースの薬学的に許容される塩としては、ナトリウム塩、カルシウム塩、カリウム塩、アンモニウム塩が挙げられ、これらの塩の1種であっても2種以上の組み合わせであってもよい。本発明に係る粘着剤層に含有されるカルボキシメチルセルロースとしては、これらの中でも、カルボキシメチルセルロースの塩であることが好ましく、ナトリウム塩であることがより好ましい。
また、前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩としては、例えば、CMCダイセルシリーズ(ダイセルミライズ株式会社製)、Aqualon CMCシリーズ(Ashland社製)、Blanose CMCシリーズ(Ashland社製)、セロゲンシリーズ(第一工業製薬株式会社製)、TPTシリーズ(五徳薬品株式会社製)等の市販のものを適宜用いることができる。
本発明において、前記粘着剤層におけるカルボキシメチルセルロース及び/又はその薬学的に許容される塩の含有量(カルボキシメチルセルロースの含有量若しくはカルボキシメチルセルロースの薬学的に許容される塩の含有量、又は、両者がいずれも含有されている場合にはそれらの合計含有量、以下同じ)としては、カルボキシメチルセルロースのナトリウム塩換算で、前記粘着剤層の全質量に対して1.0~4.0質量%であることが好ましく、1.5~3.5質量%であることがより好ましく、2.0~3.0質量%であることがさらに好ましい。カルボキシメチルセルロース及び/又はその薬学的に許容される塩の含有量が前記下限未満であると、皮膚から剥離した後のべたつきが発生しやすくなる傾向にあり、他方、前記上限を超えると、粘着剤層の皮膚への付着性が低下しやすくなる傾向にある。
(無水ケイ酸)
本発明に係る粘着剤層は、無水ケイ酸を含有する。無水ケイ酸は、シリカとも称され、SiO2で示されるケイ素の酸化物である。

本発明に係る無水ケイ酸の粒子径としては、粒度分布における累積体積が50%となる粒子径D50で0.2~9.0μmであることが必要であり、前記粒子径D50としては、0.5、1.0、2.0、3.0、4.0、4.5、又は4.8μm以上であることが好ましく、また、5.2、6.0、7.0、7.7、8.0、又は9.0μm以下であることが好ましい。前記粒子径D50の上限と下限とは任意に組み合わせてよく、特に制限されないが、例えば、2.0~8.0μm、3.0~7.0μm、4.0~8.0μm、4.0~6.0μm、4.8~7.7μm、4.8~5.2μmが挙げられ、2.0~8.0μmであることが好ましく、4.0~8.0μmであることがより好ましく、4.8~7.7μmであることがさらに好ましい。前記無水ケイ酸の粒子径D50が前記下限未満であると、粘着剤層を皮膚から剥離した後のべたつきの発生抑制効果が低下したり、製造時の取り扱いが困難になる傾向にあり、他方、前記上限を超えると、粘着剤層の皮膚への付着性が低下したり、粘着剤層に粒子が残存して外観不良となったりする傾向にある。
本発明において、前記無水ケイ酸の粒子径D50は、日本工業規格JIS.Z8825:2013のレーザ回折・散乱法により測定される粒子径を示す。より具体的には、測定サンプルとして、測定対象の無水ケイ酸の3質量%水分散液を準備し、次の測定条件:

で前記サンプル中の粒子径を測定し、粒度分布曲線を得る。得られた粒度分布曲線において累積体積が50%となる粒子径を、本発明における無水ケイ酸の粒子径D50[μm]とすることができる。なお、例えば、本発明者らがこの測定方法(より具体的には下記の実施例に記載の方法)によって特許文献2に記載されているアエロジルA-200(日本アエロジル株式会社製)の粒子径D50を測定したところ、0.055μm(n=3、1回目:0.055μm、2回目:0.055μm、3回目:0.055μm)であった。

屈折率:1.55

本発明において、前記粘着剤層における無水ケイ酸の含有量としては、前記粘着剤層の全質量に対して0.1~6.0質量%であることが好ましく、0.2~5.0質量%又は0.1~4.0質量%であることがより好ましく、0.7~4.5質量%であることがさらに好ましく、1.0~4.0質量%であることが特に好ましい。無水ケイ酸の含有量が前記範囲を外れると、粘着剤層の皮膚への付着性が低下したり、粘着剤層を皮膚から剥離した後のべたつきの発生抑制効果が低下したりする傾向にある。
また、本発明において、前記粘着剤層における前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量と前記無水ケイ酸の含有量との質量比(カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量:無水ケイ酸の含有量)としては、1:2~6:0.5であることが好ましく、1:1.5~6:0.9であることがより好ましく、1:1.5~6:1であることがさらに好ましく、1:1.4~3:1であることが特に好ましい。カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量に対する無水ケイ酸の含有量が前記下限未満であると、粘着剤層の皮膚への付着性が低下したり、皮膚から剥離した後のべたつきが発生しやすくなったりする傾向にあり、他方、前記上限を超えると、粘着剤層を皮膚から剥離した後のべたつきの発生抑制効果が低下する傾向にある。
(薬物)
本発明に係る粘着剤層に含有される薬物としては、特に制限されず、貼付剤の粘着剤層に含有可能な薬物を適宜採用することができる。このような薬物としては、例えば、ジクロフェナク、フェルビナク、フルルビプロフェン、サリチル酸エステル、インドメタシン、ケトプロフェン、イブプロフェン等の非ステロイド系抗炎症剤;ジフェンヒドラミン、クロルフェニラミン等の抗ヒスタミン剤;アスピリン、アセトアミノフェン、ロキソプロフェン等の鎮痛剤;リドカイン、ジブカイン等の局所麻酔剤;塩化スキサメトニウム等の筋弛緩剤;クロトリマゾール等の抗真菌剤;クロニジン等の降圧剤;ニトログリセリン、硝酸イソソルビド等の血管拡張剤;ビタミンA、ビタミンE、ビタミンK、オクトチアミン、酪酸リボフラビン等のビタミン類;プロスタグランジン類;グリチルレチン酸、スコポラミン、フェンタニール、トウガラシエキス、カプサイシン、ノニル酸ワニリルアミド、l-メントール、dl-カンフル;及びこれらが塩を形成可能な場合にはこれらの薬学的に許容される塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。前記薬学的に許容される塩としては、例えば、ナトリウム塩、カリウム塩、カルシウム塩、アンモニウム塩等の無機塩基塩;エポラミン塩(1-(2-ヒドロキシエチル)ピロリジン塩)等の有機塩基塩;塩酸塩、硝酸塩、リン酸塩等の無機酸塩;酢酸塩、乳酸塩、ベンゼンスルホン酸塩等の有機酸塩が挙げられる。
本発明に係る粘着剤層に含有される薬物としては、特に制限されず、貼付剤の粘着剤層に含有可能な薬物を適宜採用することができる。このような薬物としては、例えば、ジクロフェナク、フェルビナク、フルルビプロフェン、サリチル酸エステル、インドメタシン、ケトプロフェン、イブプロフェン等の非ステロイド系抗炎症剤;ジフェンヒドラミン、クロルフェニラミン等の抗ヒスタミン剤;アスピリン、アセトアミノフェン、ロキソプロフェン等の鎮痛剤;リドカイン、ジブカイン等の局所麻酔剤;塩化スキサメトニウム等の筋弛緩剤;クロトリマゾール等の抗真菌剤;クロニジン等の降圧剤;ニトログリセリン、硝酸イソソルビド等の血管拡張剤;ビタミンA、ビタミンE、ビタミンK、オクトチアミン、酪酸リボフラビン等のビタミン類;プロスタグランジン類;グリチルレチン酸、スコポラミン、フェンタニール、トウガラシエキス、カプサイシン、ノニル酸ワニリルアミド、l-メントール、dl-カンフル;及びこれらが塩を形成可能な場合にはこれらの薬学的に許容される塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。前記薬学的に許容される塩としては、例えば、ナトリウム塩、カリウム塩、カルシウム塩、アンモニウム塩等の無機塩基塩;エポラミン塩(1-(2-ヒドロキシエチル)ピロリジン塩)等の有機塩基塩;塩酸塩、硝酸塩、リン酸塩等の無機酸塩;酢酸塩、乳酸塩、ベンゼンスルホン酸塩等の有機酸塩が挙げられる。
これらの中でも、本発明に係る薬物としては、パップ剤に好適であるという観点から、ジクロフェナク、フェルビナク、フルルビプロフェン、サリチル酸エステル、インドメタシン、ケトプロフェン、ジフェンヒドラミン、ロキソプロフェン、リドカイン、ビタミンE、グリチルレチン酸、トウガラシエキス、ノニル酸ワニリルアミド、l-メントール、dl-カンフル、及びこれらの薬学的に許容される塩のうちの少なくとも1種であることが好ましい。
例えば、ジクロフェナクは、非ステロイド系の鎮痛、抗炎症薬であり、日本薬局方の他、世界各国の薬局方に収載されている。本発明において、ジクロフェナクが前記粘着剤層に含有される場合、その形態としては、遊離体(フリー体)であってもその薬学的に許容される塩であってもよく、製造中及び/又は製造された製剤中においてジクロフェナクの薬学的に許容される塩が脱塩されて遊離体となったものであってもよく、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、本発明に係る粘着剤層に含有されるジクロフェナクの形態としては、薬物の安定性が向上し、酸によりひきおこされる皮膚への刺激や粘着剤層の物性(強度、弾性、耐久性、保水性等)の低下をより抑制できる傾向にあるという観点から、ジクロフェナクの薬学的に許容される塩であることが好ましく、ナトリウム塩であることがより好ましい。
また例えば、サリチル酸エステルも非ステロイド系の鎮痛、抗炎症薬であり、本発明に係るサリチル酸エステルとしては、サリチル酸メチル、サリチル酸グリコール等が挙げられる。これらの中でも、本発明に係る粘着剤層に含有されるサリチル酸エステルとしては、臭いが少ない観点からはサリチル酸グリコールが好ましい。
本発明において、前記粘着剤層における薬物の含有量(薬物が2種以上の場合にはそれらの合計含有量、以下同じ)としては、その効果や目的等に応じて適宜調整されるものであるため、一概にいえるものではないが、前記粘着剤層の全質量に対して0.2~10.0質量%であることが好ましい。
例えば、本発明に係る粘着剤層がジクロフェナク及びその薬学的に許容される塩からなる群から選択される少なくとも1種(以下、場合により「ジクロフェナク及び/又はその薬学的に許容される塩」という)を含有する場合、ジクロフェナク及び/又はその薬学的に許容される塩の含有量(ジクロフェナクの含有量若しくはジクロフェナクの薬学的に許容される塩の含有量、又は、両者がいずれも含有されている場合にはそれらの合計含有量、以下同じ)としては、ジクロフェナクのナトリウム塩換算で、前記粘着剤層の全質量に対して0.5~3.0質量%であることが好ましく、0.75~2.5質量%であることがより好ましく、1.0~2.0質量%であることがさらに好ましい。
また例えば、本発明に係る粘着剤層がサリチル酸エステルを含有する場合、サリチル酸エステルの含有量(2種以上のサリチル酸エステルが含有されている場合にはそれらの合計含有量、以下同じ)としては、サリチル酸グリコール換算で、前記粘着剤層の全質量に対して0.5~3.0質量%であることが好ましく、0.75~2.5質量%であることがより好ましく、1.0~2.0質量%であることがさらに好ましい。
これら薬物の含有量が前記下限未満であると、鎮痛消炎効果が低下する傾向にあり、他方、前記上限を超えると、副作用が発現しやすくなる傾向にある。
(水溶性高分子)
本発明に係る粘着剤層は、カルボキシメチルセルロース及び/又はその薬学的に許容される塩以外の水溶性高分子(本明細書中場合により単に「水溶性高分子」という)をさらに含有する。このような水溶性高分子としては、特に制限されず、従来パップ剤の粘着剤層に含有される水溶性高分子として公知の水溶性高分子を適宜採用することができる。
本発明に係る粘着剤層は、カルボキシメチルセルロース及び/又はその薬学的に許容される塩以外の水溶性高分子(本明細書中場合により単に「水溶性高分子」という)をさらに含有する。このような水溶性高分子としては、特に制限されず、従来パップ剤の粘着剤層に含有される水溶性高分子として公知の水溶性高分子を適宜採用することができる。
本発明に係る水溶性高分子(カルボキシメチルセルロース及び/又はその薬学的に許容される塩以外の水溶性高分子)としては、例えば、ポリアクリル酸中和物、ポリアクリル酸、カルボキシビニルポリマー、メチルセルロース、エチルセルロース、ヒドロキシエチルセルロース、ヒドロキシプロピルセルロース、結晶セルロース、ゼラチン、カゼイン、プルラン、寒天、デキストラン、デキストリン、アルギン酸ナトリウム、可溶性デンプン、カルボキシル化デンプン、ポリビニルアルコール、ポリエチレンオキサイド、ポリアクリルアミド、ポリビニルピロリドン、ポリビニルエーテル-無水マレイン酸共重合体、メトキシエチレン-無水マレイン酸共重合体、イソブチレン-無水マレイン酸共重合体、ポリエチレンイミン、ヒアルロン酸ナトリウム、グアーガム、キサンタンガム、ジェランガム、カラギーナン、ローカストビーンガム、ペクチンが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
これらの中でも、本発明に係る水溶性高分子としては、粘着剤層の保形性をより向上させる観点から、ポリアクリル酸中和物、ポリアクリル酸、ゼラチン、寒天、アルギン酸ナトリウム、ポリビニルアルコール、ポリビニルピロリドン、ヒアルロン酸ナトリウム、グアーガム、キサンタンガム、ジェランガム、カラギーナン、ローカストビーンガム、及びペクチンからなる群から選択される少なくとも1種であることが好ましく、少なくともポリアクリル酸中和物を含むことがより好ましい。
本発明において、「ポリアクリル酸中和物」は、ポリアクリル酸完全中和物及びポリアクリル酸部分中和物を包含し、ポリアクリル酸の全部又は一部が、ナトリウム、カリウム等のアルカリ金属やアンモニウムイオン等によって中和されたものを示す。前記ポリアクリル酸中和物としては、例えば、ポリアクリル酸ナトリウム、ポリアクリル酸カリウム、ポリアクリル酸アンモニウム等が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよいが、得られる粘着剤層の保形性がさらに高くなる傾向にある観点から、ポリアクリル酸部分中和物が好ましく、また、ポリアクリル酸ナトリウムが好ましい。前記ポリアクリル酸中和物の中和率としては、特に制限されるものではないが、例えば、30~100%であることが好ましく、35~65%であることがより好ましく、40~60%であることがさらに好ましい。
本発明において、前記粘着剤層における前記水溶性高分子の含有量(水溶性高分子が2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して5.0~15.0質量%であることが好ましく、8.5~9.0質量%であることがより好ましい。前記水溶性高分子の含有量が前記下限未満であると、粘着剤層の保形性が低下する傾向にあり、他方、前記上限を超えると、粘着剤層の皮膚への付着性が低下しやすくなる傾向にある。また、本発明に係る粘着剤層が前記ポリアクリル酸中和物を含有する場合、その含有量(ポリアクリル酸中和物が2種以上の場合にはそれらの合計含有量、以下同じ)としては、同様の観点から、前記粘着剤層の全質量に対して1.0~10.0質量%であることが好ましく、2.0~7.0質量%であることがより好ましく、3.0~4.0質量%であることがさらに好ましい。
(水)
本発明に係る粘着剤層は、水も含有する。前記水としては、イオン交換、蒸留、濾過等の精製を施された水であることが好ましく、例えば、日本薬局方(第十八改正日本薬局方)に記載の「精製水」を好適に用いることができる。前記粘着剤層における水の含有量としては、前記粘着剤層の全質量に対して20~70質量%であることが好ましい。前記水の含有量が前記範囲を外れる場合には、粘着剤層の保形性が低下したり、皮膚への付着性が低下しやすくなったりする傾向にある。
本発明に係る粘着剤層は、水も含有する。前記水としては、イオン交換、蒸留、濾過等の精製を施された水であることが好ましく、例えば、日本薬局方(第十八改正日本薬局方)に記載の「精製水」を好適に用いることができる。前記粘着剤層における水の含有量としては、前記粘着剤層の全質量に対して20~70質量%であることが好ましい。前記水の含有量が前記範囲を外れる場合には、粘着剤層の保形性が低下したり、皮膚への付着性が低下しやすくなったりする傾向にある。
(多価アルコール)
本発明に係る粘着剤層としては、湿潤剤として機能し、また、皮膚への付着性がより向上する傾向にある観点から、多価アルコールをさらに含有することが好ましい。このような多価アルコールとしては、特に制限されないが、例えば、グリセリン;メチレングリコール、エチレングリコール、プロピレングリコール、ブチレングリコール等のアルキレングリコール;ポリエチレングリコール等のポリアルキレングリコール;1,3-プロパンジオール、1,4-ブタンジオール等のアルカンジオール;ソルビトールが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
本発明に係る粘着剤層としては、湿潤剤として機能し、また、皮膚への付着性がより向上する傾向にある観点から、多価アルコールをさらに含有することが好ましい。このような多価アルコールとしては、特に制限されないが、例えば、グリセリン;メチレングリコール、エチレングリコール、プロピレングリコール、ブチレングリコール等のアルキレングリコール;ポリエチレングリコール等のポリアルキレングリコール;1,3-プロパンジオール、1,4-ブタンジオール等のアルカンジオール;ソルビトールが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
これらの中でも、前記多価アルコールとしては、粘着剤層の皮膚への付着性がさらに向上する傾向にあるという観点から、グリセリン及びアルキレングリコールからなる群から選択される少なくとも1種であることが好ましく、少なくともグリセリンを含むことがより好ましい。
本発明に係る粘着剤層が前記多価アルコールを含有する場合、その含有量(多価アルコールが2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して10~50質量%であることが好ましく、35~45質量%であることがより好ましい。前記多価アルコールの含有量が前記下限未満であると、前記付着性の向上効果が十分に奏されなくなる傾向にあり、他方、前記上限を超えると、粘着剤層の保形性が低下しやすくなる傾向にある。また、本発明に係る粘着剤層がグリセリンを含有する場合、その含有量としては、同様の観点から、前記粘着剤層の全質量に対して20~50質量%であることが好ましく、25~43質量%であることがより好ましく、30~35質量%であることがさらに好ましい。
(キレート剤)
本発明に係る粘着剤層としては、粘着剤層の展延ムラをより抑制できる傾向にある観点から、キレート剤をさらに含有することが好ましい。このようなキレート剤としては、特に制限されないが、例えば、エデト酸塩(EDTA)、ピロリン酸塩、ヘキサメタリン酸塩、グルコン酸塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記キレート剤としては、エデト酸ナトリウムが好ましい。
本発明に係る粘着剤層としては、粘着剤層の展延ムラをより抑制できる傾向にある観点から、キレート剤をさらに含有することが好ましい。このようなキレート剤としては、特に制限されないが、例えば、エデト酸塩(EDTA)、ピロリン酸塩、ヘキサメタリン酸塩、グルコン酸塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記キレート剤としては、エデト酸ナトリウムが好ましい。
本発明に係る粘着剤層が前記キレート剤を含有する場合、その含有量(キレート剤が2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して0.01~1質量%であることが好ましく、0.01~0.1質量%であることがより好ましい。前記キレート剤の含有量が前記下限未満であると、粘着剤層の展延ムラ抑制効果が十分に発揮されずに外観が損なわれやすくなる傾向にあり、他方、前記上限を超えると、粘着剤層の保形性が低下しやすくなる傾向にある。
(架橋剤)
本発明に係る粘着剤層としては、粘着剤層の保形性がより向上する傾向にある観点及び展延ムラをより抑制できる傾向にある観点から、架橋剤をさらに含有することが好ましい。このような架橋剤としては、特に制限されないが、例えば、硫酸アルミニウムカリウム(ミョウバン)、塩化カルシウム、塩化マグネシウム、水酸化アルミニウム、ジヒドロキシアルミニウムアミノアセテートが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記架橋剤としては、硫酸アルミニウムカリウムが好ましい。
本発明に係る粘着剤層としては、粘着剤層の保形性がより向上する傾向にある観点及び展延ムラをより抑制できる傾向にある観点から、架橋剤をさらに含有することが好ましい。このような架橋剤としては、特に制限されないが、例えば、硫酸アルミニウムカリウム(ミョウバン)、塩化カルシウム、塩化マグネシウム、水酸化アルミニウム、ジヒドロキシアルミニウムアミノアセテートが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記架橋剤としては、硫酸アルミニウムカリウムが好ましい。
本発明に係る粘着剤層が前記架橋剤を含有する場合、その含有量(架橋剤が2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して0.1~1.0質量%であることが好ましく、0.2~0.5質量%であることがより好ましい。前記架橋剤の含有量が前記下限未満であると、粘着剤層の保形性の向上効果が十分に奏されなくなる傾向にあり、他方、前記上限を超えると、粘着剤層の展延ムラ抑制効果が十分に発揮されずに外観が損なわれやすくなる傾向にある。
また、本発明に係る粘着剤層が前記キレート剤と前記架橋剤との両方を含有する場合、前記キレート剤の含有量と前記架橋剤の含有量との質量比(キレート剤の含有量:架橋剤の含有量)としては、1:0.7~1:10であることが好ましく、1:4~1:9であることがより好ましい。前記キレート剤の含有量に対する前記架橋剤の含有量が前記下限未満であると、染み出しが発生しやすくなったり、粘着剤層の保形性の向上効果が十分に奏されなくなる傾向にあり、他方、前記上限を超えると、粘着剤層の展延ムラ抑制効果が十分に発揮されずに外観が損なわれやすくなったり、粘着剤層の粘着力が低下しやすくなったりする傾向にある。
(pH調整剤)
本発明に係る粘着剤層としては、そのpHを調整するために、pH調整剤をさらに含有することも好ましい。このようなpH調整剤としては、例えば、酢酸、乳酸、シュウ酸、クエン酸、酒石酸等の有機酸;塩酸、硫酸、硝酸、リン酸等の無機酸;前記有機酸及び前記無機酸の薬学的に許容される塩;モノエタノールアミン、ジエタノールアミン、トリエタノールアミン、イソプロパノールアミン、ジイソプロパノールアミン等のアミン類;水酸化ナトリウム、水酸化カリウム、水酸化マグネシウム等の水酸化物の塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。また、本発明に係る粘着剤層のpHとしては、7.0~9.0であることが好ましく、7.0~8.5であることがより好ましい。
本発明に係る粘着剤層としては、そのpHを調整するために、pH調整剤をさらに含有することも好ましい。このようなpH調整剤としては、例えば、酢酸、乳酸、シュウ酸、クエン酸、酒石酸等の有機酸;塩酸、硫酸、硝酸、リン酸等の無機酸;前記有機酸及び前記無機酸の薬学的に許容される塩;モノエタノールアミン、ジエタノールアミン、トリエタノールアミン、イソプロパノールアミン、ジイソプロパノールアミン等のアミン類;水酸化ナトリウム、水酸化カリウム、水酸化マグネシウム等の水酸化物の塩が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。また、本発明に係る粘着剤層のpHとしては、7.0~9.0であることが好ましく、7.0~8.5であることがより好ましい。
(粘着増強剤)
本発明に係る粘着剤層としては、粘着剤層の皮膚への付着性がより向上する傾向にある観点から、粘着増強剤をさらに含有することが好ましい。このような粘着増強剤としては、特に制限されないが、例えば、アミノアルキルメタクリレートコポリマーE等のアクリル酸エステル共重合体;アクリル酸メチル・アクリル酸2-エチルヘキシル共重合体が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記粘着増強剤としては、少量の添加で効果が奏される傾向にある観点からは、アミノアルキルメタクリレートコポリマーEが好ましい。
本発明に係る粘着剤層としては、粘着剤層の皮膚への付着性がより向上する傾向にある観点から、粘着増強剤をさらに含有することが好ましい。このような粘着増強剤としては、特に制限されないが、例えば、アミノアルキルメタクリレートコポリマーE等のアクリル酸エステル共重合体;アクリル酸メチル・アクリル酸2-エチルヘキシル共重合体が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。これらの中でも、前記粘着増強剤としては、少量の添加で効果が奏される傾向にある観点からは、アミノアルキルメタクリレートコポリマーEが好ましい。
本発明に係る粘着剤層が前記粘着増強剤を含有する場合、その含有量(粘着増強剤が2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して0.1~1.0質量%であることが好ましく、0.4~0.6質量%であることがより好ましい。前記粘着増強剤の含有量が前記下限未満であると、粘着剤層の皮膚への付着性向上効果が十分に奏されなくなる傾向にあり、他方、前記上限を超えると、粘着剤層を皮膚から剥離する際に痛みが生じやすくなる傾向にある。
(その他添加剤)
本発明に係る粘着剤層としては、他にも、乳化剤、清涼化剤、安定化剤(酸化防止剤)、防腐剤、本発明に係る無水ケイ酸以外の充填剤等の添加剤をさらに含有していてもよい。
本発明に係る粘着剤層としては、他にも、乳化剤、清涼化剤、安定化剤(酸化防止剤)、防腐剤、本発明に係る無水ケイ酸以外の充填剤等の添加剤をさらに含有していてもよい。
前記乳化剤としては、例えば、モノオレイン酸ポリアルキレングリコール(モノオレイン酸ポリエチレングリコール、モノオレイン酸ポリプロピレングリコール、モノオレイン酸ポリエチレンポリプロピレングリコール等)、モノステアリン酸ポリエチレングリコール、モノオレイン酸ポリオキシエチレンソルビタンが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
前記清涼化剤としては、例えば、チモール、dl-メントール、l-イソプレゴール、ハッカ油が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
前記安定化剤としては、例えば、亜硫酸ナトリウム、ブチルヒドロキシアニソール、ジブチルヒドロキシトルエン、ノルジヒドログアヤレチン酸、クエン酸、アスコルビン酸、没食子酸プロピルが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
前記防腐剤としては、例えば、パラオキシ安息香酸メチル、パラオキシ安息香酸エチル、パラオキシ安息香酸プロピル、パラオキシ安息香酸ブチル、1,2-ペンタンジオール、安息香酸及びその塩、サリチル酸及びその塩、ソルビン酸及びその塩、デヒドロ酢酸及びその塩、4-イソプロピル-3-メチルフェノール、2-イソプロピル-5-メチルフェノール、フェノール、ヒノキチオール、クレゾール、2,4,4’-トリクロロ-2’-ヒドロキシジフェニルエーテル、3,4,4’-トリクロロカルバニリド、クロロブタノール、塩化ベンザルコニウム、塩化ベンゼトニウムが挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。
前記本発明に係る無水ケイ酸以外の充填剤としては、本発明に係る無水ケイ酸の粒子径を外れる無水ケイ酸の他、例えば、酸化チタン、ケイ酸アルミニウム、カオリン、タルク、ベントナイト、メタケイ酸アルミン酸マグネシウム等の無機充填剤が挙げられ、これらのうちの1種であっても2種以上の組み合わせであってもよい。ただし、本発明に係る粘着剤層としては、本発明に係る無水ケイ酸以外の充填剤を実質的に含有しないことが好ましく、含有する場合であってもその含有量(充填剤が2種以上の場合にはそれらの合計含有量、以下同じ)としては、前記粘着剤層の全質量に対して3.0質量%以下であることが好ましく、1.0質量%以下であることがより好ましく、0.1質量%以下であることがさらに好ましく、0.05質量%以下であることが特に好ましい。
<パップ剤の製造方法>
本発明のパップ剤は、特に制限されず、公知のパップ剤の製造方法を適宜採用することによって製造することができるが、好ましくは、次の製造方法:
支持体層と粘着剤層とを備えるパップ剤の製造方法であり、
前記カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、前記無水ケイ酸、前記薬物、及び前記水溶性高分子を混合して粘着剤層組成物を得る工程と、前記粘着剤層組成物を展延して前記粘着剤層を得る工程とを含む、製造方法で製造することができる。
本発明のパップ剤は、特に制限されず、公知のパップ剤の製造方法を適宜採用することによって製造することができるが、好ましくは、次の製造方法:
支持体層と粘着剤層とを備えるパップ剤の製造方法であり、
前記カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、前記無水ケイ酸、前記薬物、及び前記水溶性高分子を混合して粘着剤層組成物を得る工程と、前記粘着剤層組成物を展延して前記粘着剤層を得る工程とを含む、製造方法で製造することができる。
前記粘着剤層組成物を得る工程においては、前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩、前記無水ケイ酸、前記薬物、前記水溶性高分子、水、並びに、必要に応じて前記多価アルコール、前記キレート剤、前記架橋剤、前記pH調整剤、前記粘着増強剤、及び前記添加剤等を混合し、粘着剤層組成物とする。このとき、混合する各成分の配合量としては、その好ましい態様も含めて、それぞれ、上記の本発明のパップ剤の粘着剤層における各成分の含有量として述べたとおりである。
前記混合方法としては、特に制限されず、従来パップ剤の製造方法において公知の方法又はそれに準じた方法を適宜採用することができ、例えば、プロペラミキサー、パドルミキサー、アンカーミキサー、プラネタリーミキサー、V型混合機、ヘンシェルミキサー等の混合機を用いて混合する方法が挙げられる。混合条件についても特に制限されず、従来公知の条件又はそれに準じた条件に適宜調整することができる。
次いで、前記粘着剤層を得る工程においては、上記で得られた粘着剤層組成物を展延して前記粘着剤層とする。前記展延方法としては、特に制限されず、従来パップ剤の製造方法において公知の方法又はそれに準じた方法を適宜採用することができ、例えば、前記粘着剤層組成物を前記支持体層の面上(通常は一方の面上)に所定の厚みで展延することで前記支持体層上に前記粘着剤層を備えた本発明のパップ剤を得ることができる。この場合、必要に応じて前記粘着剤層の前記支持体層と反対の面上に前記剥離ライナーを貼り合わせて、前記支持体層、前記粘着剤層、及び前記剥離ライナーがこの順で積層されたパップ剤としてもよい。
また、前記展延方法の別の態様としては、例えば、前記粘着剤層組成物を先ず前記剥離ライナーの一方の面上に所定の厚みで展延して前記粘着剤層とした後に、前記粘着剤層の前記剥離ライナーと反対の面上に前記支持体層を貼り合わせることにより、前記支持体層、前記粘着剤層、及び前記剥離ライナーがこの順で積層されたパップ剤を得ることができる。
さらに、本発明のパップ剤の製造方法としては、必要に応じてさらにパップ剤を所定の形状に裁断する工程を含んでいてもよい。
また、本発明のパップ剤としては、必要に応じて、保存用包装容器(例えば、アルミニウム包装袋)に封入して包装体としてもよい。
以下、実施例及び比較例に基づいて本発明をより具体的に説明するが、本発明は以下の実施例に限定されるものではない。なお、各実施例及び比較例において用いた無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径測定、並びに、各実施例及び比較例において得られたパップ剤の付着性評価、べたつき評価、及び粘着剤層外観評価は、それぞれ、以下に示す方法により行った。
(粒子径測定)
無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径は、日本工業規格JIS.Z8825:2013のレーザ回折・散乱法により、次の測定条件:
測定機器:株式会社島津製作所製レーザ回折式粒子径分布測定装置SALD-2300
測定モード:高濃度サンプル測定モード
サンプル濃度:3質量%
分散媒:精製水
屈折率:1.55
でサンプル中の粒子径を測定し、粒度分布曲線を得た。得られた粒度分布曲線から、累積体積が50%となる粒子径D50(μm)を求めた。測定は各無水ケイ酸サンプル又はメタケイ酸アルミン酸マグネシウムサンプルについて3回ずつ行い、その平均を当該無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径(粒子径D50[μm])とした。
無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径は、日本工業規格JIS.Z8825:2013のレーザ回折・散乱法により、次の測定条件:
測定機器:株式会社島津製作所製レーザ回折式粒子径分布測定装置SALD-2300
測定モード:高濃度サンプル測定モード
サンプル濃度:3質量%
分散媒:精製水
屈折率:1.55
でサンプル中の粒子径を測定し、粒度分布曲線を得た。得られた粒度分布曲線から、累積体積が50%となる粒子径D50(μm)を求めた。測定は各無水ケイ酸サンプル又はメタケイ酸アルミン酸マグネシウムサンプルについて3回ずつ行い、その平均を当該無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径(粒子径D50[μm])とした。
(付着性評価)
先ず、同一の実施例又は比較例によりパップ剤を6枚ずつ調製し、各パップ剤から剥離ライナーを剥離除去して、6名の被験者の肘にそれぞれ貼付した。次いで、被験者の腕を屈伸(屈曲及び伸展)させ、40回屈伸後の、粘着剤層の皮膚からの剥離状況を観察した。下記の表1の基準に基づいて剥離状況をスコア付けし、被験者6名のスコアの平均値(平均スコア)から付着性の評価を判定した。パップ剤の付着性評価は、「B」以上で、皮膚への付着性が製剤として十分許容可能に優れていると判断できる。
先ず、同一の実施例又は比較例によりパップ剤を6枚ずつ調製し、各パップ剤から剥離ライナーを剥離除去して、6名の被験者の肘にそれぞれ貼付した。次いで、被験者の腕を屈伸(屈曲及び伸展)させ、40回屈伸後の、粘着剤層の皮膚からの剥離状況を観察した。下記の表1の基準に基づいて剥離状況をスコア付けし、被験者6名のスコアの平均値(平均スコア)から付着性の評価を判定した。パップ剤の付着性評価は、「B」以上で、皮膚への付着性が製剤として十分許容可能に優れていると判断できる。
(べたつき評価)
上記付着性評価の後、パップ剤をそれぞれ皮膚から剥離し、パップ剤を貼付していた部位を指先で触ったときのべたつき状況を観察した。下記の表2の基準に基づいてべたつき状況をスコア付けし、被験者6名のスコアの平均値(平均スコア)からべたつきの評価を判定した。パップ剤のべたつき評価は、「B」以上で、皮膚へのべたつきの発生が製剤として十分許容可能な程度に抑制されていると判断できる。
上記付着性評価の後、パップ剤をそれぞれ皮膚から剥離し、パップ剤を貼付していた部位を指先で触ったときのべたつき状況を観察した。下記の表2の基準に基づいてべたつき状況をスコア付けし、被験者6名のスコアの平均値(平均スコア)からべたつきの評価を判定した。パップ剤のべたつき評価は、「B」以上で、皮膚へのべたつきの発生が製剤として十分許容可能な程度に抑制されていると判断できる。
(粘着剤層外観評価)
実施例又は比較例で得られた直後のパップ剤から剥離ライナーを剥離除去し、粘着剤層の表面を目視で観察した。粘着剤層表面に凝集物又は凹凸が確認され、製剤として用いることが困難なものは「N」、粘着剤層表面に凝集物や凹凸が確認されず均一なものは「Y」と評価した。
実施例又は比較例で得られた直後のパップ剤から剥離ライナーを剥離除去し、粘着剤層の表面を目視で観察した。粘着剤層表面に凝集物又は凹凸が確認され、製剤として用いることが困難なものは「N」、粘着剤層表面に凝集物や凹凸が確認されず均一なものは「Y」と評価した。
(実施例1)
先ず、カルボキシメチルセルロースナトリウム(エーテル化度:0.8~1.0、粘度(25℃、1.0%水溶液、60rpm):13mPa・s)2.0質量部、無水ケイ酸1.5質量部、ジクロフェナクナトリウム1.0質量部、水溶性高分子(ゼラチン:ポリアクリル酸部分中和物(ポリアクリル酸ナトリウム):ポリビニルアルコール=1.8:3.5:3.5(質量比))8.8質量部、多価アルコール(グリセリン:アルキレングリコール=31.6:7.6(質量比))39.2質量部、キレート剤(エデト酸ナトリウム水和物)0.05質量部、架橋剤(乾燥硫酸アルミニウムカリウム)0.41質量部、pH調整剤(ジエタノールアミン)1.9質量部、粘着増強剤(アミノアルキルメタクリレートコポリマーE)0.5質量部、その他添加剤(乳化剤、清涼化剤、及び安定化剤)2.72質量部、及び精製水41.92質量部を混合し、粘着剤層組成物を調製した。上記粒子径測定により測定した前記無水ケイ酸の粒子径は、粒子径D50:4.8μmであった。
先ず、カルボキシメチルセルロースナトリウム(エーテル化度:0.8~1.0、粘度(25℃、1.0%水溶液、60rpm):13mPa・s)2.0質量部、無水ケイ酸1.5質量部、ジクロフェナクナトリウム1.0質量部、水溶性高分子(ゼラチン:ポリアクリル酸部分中和物(ポリアクリル酸ナトリウム):ポリビニルアルコール=1.8:3.5:3.5(質量比))8.8質量部、多価アルコール(グリセリン:アルキレングリコール=31.6:7.6(質量比))39.2質量部、キレート剤(エデト酸ナトリウム水和物)0.05質量部、架橋剤(乾燥硫酸アルミニウムカリウム)0.41質量部、pH調整剤(ジエタノールアミン)1.9質量部、粘着増強剤(アミノアルキルメタクリレートコポリマーE)0.5質量部、その他添加剤(乳化剤、清涼化剤、及び安定化剤)2.72質量部、及び精製水41.92質量部を混合し、粘着剤層組成物を調製した。上記粒子径測定により測定した前記無水ケイ酸の粒子径は、粒子径D50:4.8μmであった。
次いで、得られた粘着剤層組成物を、パップ剤1枚(140mm×100mm)当たり14gとなるように剥離ライナー(ポリプロピレン製フィルム)上に均一に展延し、その上に支持体層(ポリエステル製不織布、目付:100g/m2)を積層して、パップ剤を得た。得られた粘着剤層の組成を下記の表3に示す。また、得られたパップ剤について付着性評価、べたつき評価、及び粘着剤層外観評価を行なった結果を下記の表3に合わせて示す。
(実施例2、比較例1~6)
粘着剤層の組成をそれぞれ下記の表3に示す組成としたこと以外は実施例1と同様にして各パップ剤をそれぞれ得た。得られたパップ剤についてそれぞれ付着性評価、べたつき評価、及び粘着剤層外観評価を行なった結果を、各粘着剤層の組成と共に下記の表3に示す。
粘着剤層の組成をそれぞれ下記の表3に示す組成としたこと以外は実施例1と同様にして各パップ剤をそれぞれ得た。得られたパップ剤についてそれぞれ付着性評価、べたつき評価、及び粘着剤層外観評価を行なった結果を、各粘着剤層の組成と共に下記の表3に示す。
表3に示した結果から明らかなように、本発明のパップ剤(例えば、実施例1~2)では、無水ケイ酸とカルボキシメチルセルロースナトリウムとをいずれも含有することにより、付着性評価、べたつき評価、及び粘着剤層外観評価が全てパップ剤として十分に優れるものであることが確認された。他方、無水ケイ酸、並びに、カルボキシメチルセルロース及び/又はその薬学的に許容される塩のうちの少なくともいずれかを含有しないパップ剤(例えば、比較例1~3)や、カルボキシメチルセルロース及び/又はその薬学的に許容される塩に代えてヒドロキシプロピルセルロース、カルボキシビニルポリマー、又は結晶セルロースを含有するパップ剤(例えば、比較例4~6)では、付着性評価、べたつき評価、及び粘着剤層外観評価のうちの少なくともいずれかの評価が劣り、パップ剤として用いることが困難であることが確認された。
(実施例3~4)
粘着剤層の組成をそれぞれ下記の表4に示す組成としたこと以外は実施例1と同様にして各パップ剤をそれぞれ得た。

(比較例7~9)
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が下記の表4に示す粒子径であった無水ケイ酸又はメタケイ酸アルミン酸マグネシウムを用い、かつ、粘着剤層の組成をそれぞれ下記の表4に示す組成としたこと以外は実施例1と同様にして、各パップ剤をそれぞれ得た。無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径D50は、それぞれ上記粒子径測定により測定した。
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が下記の表4に示す粒子径であった無水ケイ酸又はメタケイ酸アルミン酸マグネシウムを用い、かつ、粘着剤層の組成をそれぞれ下記の表4に示す組成としたこと以外は実施例1と同様にして、各パップ剤をそれぞれ得た。無水ケイ酸又はメタケイ酸アルミン酸マグネシウムの粒子径D50は、それぞれ上記粒子径測定により測定した。
(実施例5)
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が2.2μmであった無水ケイ酸を用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が2.2μmであった無水ケイ酸を用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
(実施例6)
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が3.1μmであった無水ケイ酸を用い、ジクロフェナクナトリウムに代えてサリチル酸グリコールを用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が3.1μmであった無水ケイ酸を用い、ジクロフェナクナトリウムに代えてサリチル酸グリコールを用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
(実施例7)
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が7.7μmであった無水ケイ酸を用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
粒子径D50:4.8μmの無水ケイ酸に代えて粒子径D50が7.7μmであった無水ケイ酸を用い、かつ、粘着剤層の組成を下記の表4に示す組成としたこと以外は実施例1と同様にしてパップ剤を得た。無水ケイ酸の粒子径D50は上記粒子径測定により測定した。
実施例3~7及び比較例7~9で得られたパップ剤について、それぞれ付着性評価、べたつき評価、及び粘着剤層外観評価を行なった結果を、各粘着剤層の組成と共に下記の表4に示す。なお、比較例9は、粘着剤層外観評価結果がNとなって製剤として使用困難であったため、付着性評価及びべたつき評価は実施しなかった。また、表4には、実施例2の結果及び粘着剤層の組成も合わせて示す。
表4に示した結果から明らかなように、本発明のパップ剤では、無水ケイ酸とカルボキシメチルセルロース及び/又はその薬学的に許容される塩とをいずれも含有することに加えて、無水ケイ酸の粒子径が粒子径D50で特定の範囲内(例えば、2.0~8.0μm)にあることにより、付着性評価、べたつき評価、及び粘着剤層外観評価が全てパップ剤として十分に優れるものであることが確認された。他方、無水ケイ酸の粒子径が上記範囲を外れるパップ剤(例えば、比較例7~8)や無水ケイ酸に代えてメタケイ酸アルミン酸マグネシウムを用いた場合(例えば、比較例9)では、付着性評価、べたつき評価、及び粘着剤層外観評価のうちの少なくともいずれかの評価が劣り、パップ剤として用いることが困難であることが確認された。
以上説明したように、本発明によれば、皮膚への付着性に優れるとともに剥離後のべたつきの発生が十分に抑制され、かつ、粘着剤層の外観に優れたパップ剤、及び前記パップ剤を容易に得ることができるパップ剤の製造方法を提供することが可能となる。
Claims (10)
- 支持体層と粘着剤層とを備えるパップ剤であり、
前記粘着剤層が、カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を含有し、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤。 - 前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩の含有量が、ナトリウム塩換算で、前記粘着剤層の全質量に対して1.0~4.0質量%である、請求項1に記載のパップ剤。
- 前記無水ケイ酸の含有量が、前記粘着剤層の全質量に対して0.1~6.0質量%である、請求項1又は2に記載のパップ剤。
- 前記粘着剤層における前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量と前記無水ケイ酸の含有量との質量比(カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量:無水ケイ酸の含有量)が、1:2~6:0.5である、請求項1又は2に記載のパップ剤。
- 前記薬物が、ジクロフェナク、フェルビナク、フルルビプロフェン、サリチル酸エステル、インドメタシン、ケトプロフェン、ジフェンヒドラミン、ロキソプロフェン、リドカイン、ビタミンE、グリチルレチン酸、トウガラシエキス、ノニル酸ワニリルアミド、l-メントール、dl-カンフル、及びこれらの薬学的に許容される塩からなる群から選択される少なくとも1種である、請求項1又は2に記載のパップ剤。
- 支持体層と粘着剤層とを備えるパップ剤の製造方法であり、
カルボキシメチルセルロース及びその薬学的に許容される塩からなる群から選択される少なくとも1種、無水ケイ酸、薬物、及び水溶性高分子を混合して粘着剤層組成物を得る工程と、前記粘着剤層組成物を展延して前記粘着剤層を得る工程とを含み、かつ、前記無水ケイ酸の粒度分布における累積体積が50%となる粒子径D50が0.2~9.0μmである、パップ剤の製造方法。 - 前記粘着剤層組成物及び前記粘着剤層における前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩の含有量が、ナトリウム塩換算で、前記粘着剤層の全質量に対して1.0~4.0質量%である、請求項6に記載のパップ剤の製造方法。
- 前記粘着剤層組成物及び前記粘着剤層における前記無水ケイ酸の含有量が、前記粘着剤層の全質量に対して0.1~6.0質量%である、請求項6又は7に記載のパップ剤の製造方法。
- 前記粘着剤層組成物及び前記粘着剤層における前記カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量と前記無水ケイ酸の含有量との質量比(カルボキシメチルセルロース及び/又はその薬学的に許容される塩のナトリウム塩換算での含有量:無水ケイ酸の含有量)が、1:2~6:0.5である、請求項6又は7に記載のパップ剤の製造方法。
- 前記薬物が、ジクロフェナク、フェルビナク、フルルビプロフェン、サリチル酸エステル、インドメタシン、ケトプロフェン、ジフェンヒドラミン、ロキソプロフェン、リドカイン、ビタミンE、グリチルレチン酸、トウガラシエキス、ノニル酸ワニリルアミド、l-メントール、dl-カンフル、及びこれらの薬学的に許容される塩からなる群から選択される少なくとも1種である、請求項6又は7に記載のパップ剤の製造方法。
