本明細書において、アミノ酸配列の「同一性」とは、2つ以上の対比可能なアミノ酸配列の、お互いに対するアミノ酸配列の一致の程度をいう。従って、ある2つのアミノ酸配列の一致性が高いほど、それらの配列の同一性又は類似性は高い。アミノ酸配列の同一性のレベルは、例えば、配列分析用ツールであるFASTAを用い、デフォルトパラメータを用いて決定される。若しくは、Karlin及びAltschulによるアルゴリズムBLAST(Karlin S, Altschul SF.“Methods for assessing the statistical significance of molecular sequence features by using general scoringschemes”Proc Natl Acad Sci USA.87:2264-2268(1990)、KarlinS,Altschul SF.“Applications and statistics for multiple high-scoring segments in molecular sequences.”Proc Natl Acad Sci USA.90:5873-7(1993))を用いて決定できる。このようなBLASTのアルゴリズムに基づいたBLASTXと呼ばれるプログラムが開発されている。これらの解析方法の具体的な手法は公知であり、National Center of Biotechnology Information(NCBI)のウェブサイト(http://www.ncbi.nlm.nih.gov/)を参照すればよい。また、塩基配列の「同一性」も上記に準じて定義される。
本明細書中において、「保存的置換」とは、アミノ酸残基が類似の側鎖を有するアミノ酸残基に置換されることを意味する。例えば、リジン、アルギニン、ヒスチジンといった塩基性側鎖を有するアミノ酸残基同士で置換されることが、保存的な置換にあたる。その他、アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸残基;グリシン、アスパラギン、グルタミン、セリン、トレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸残基;アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸残基;トレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸残基;チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸残基同士での置換も同様に、保存的な置換にあたる。
本明細書において、DNA、RNAなどのヌクレオチドには、次に例示するように、公知の化学修飾が施されていてもよい。ヌクレアーゼなどの加水分解酵素による分解を防ぐために、各ヌクレオチドのリン酸残基(ホスフェート)を、例えば、ホスホロチオエート(PS)、メチルホスホネート、ホスホロジチオネート等の化学修飾リン酸残基に置換することができる。また、各リボヌクレオチドの糖(リボース)の2位の水酸基を、-OR(Rは、例えばCH3(2´-O-Me)、CH2CH2OCH3(2´-O-MOE)、CH2CH2NHC(NH)NH2、CH2CONHCH3、CH2CH2CN等を示す)に置換してもよい。さらに、塩基部分(ピリミジン、プリン)に化学修飾を施してもよく、例えば、ピリミジン塩基の5位へのメチル基やカチオン性官能基の導入、あるいは2位のカルボニル基のチオカルボニルへの置換などが挙げられる。さらには、リン酸部分やヒドロキシル部分が、例えば、ビオチン、アミノ基、低級アルキルアミン基、アセチル基等で修飾されたものなどを挙げることができるが、これに限定されない。また、ヌクレオチドの糖部の2´酸素と4´炭素を架橋することにより、糖部のコンフォーメーションをN型に固定したものであるBNA(LNA)等もまた、好ましく用いられ得る。
本明細書において、アミノ酸の変異は、具体的には、アミノ酸の欠失、置換、挿入、又は付加である。
本明細書において、アミノ酸配列中のアミノ酸の位置は、アミノ酸一文字表記+N末端アミノ酸から数えた場合のアミノ酸番号で示すことがある。例えば、「L29」は、N末端から29番目のアミノ酸であるロイシンを示す。また、アミノ酸置換変異は、変異前のアミノ酸一文字表記+N末端アミノ酸から数えた場合のアミノ酸番号+変異後のアミノ酸一文字表記で示すことがある。例えば、「L29V」は、N末端から29番目のアミノ酸であるロイシンのバリンへの置換変異を示す。
2.ポリペプチド
本発明は、その一態様において、ヒトTNFR2アゴニストドメイン、及び多量体化ドメインを含み、前記ヒトTNFR2アゴニストドメインが前記多量体化ドメインに連結している、ポリペプチド(本明細書において、「本発明のポリペプチド」と示すこともある。)に関する。以下に、これについて説明する。
本発明は、その一態様において、ヒトTNFR2アゴニストドメイン、及び多量体化ドメインを含み、前記ヒトTNFR2アゴニストドメインが前記多量体化ドメインに連結している、ポリペプチド(本明細書において、「本発明のポリペプチド」と示すこともある。)に関する。以下に、これについて説明する。
ヒトTNFR2アゴニストドメインは、ヒトTNFR2に対するアゴニスト活性を有する特定のアミノ酸配列からなるポリペプチドであり、多量体化ドメインと連結して一本のポリペプチド鎖を形成可能なものである限り、特に制限されない。
アゴニスト活性の発揮対象であるヒトTNFR2は、公知のタンパク質である。具体的には、ヒトTNFR2のアミノ酸配列としては、配列番号12に示されるアミノ酸配列(NCBI Reference Sequence: NP_001057.1)が挙げられ、ヒトTNFR2のmRNA配列としては、配列番号13に示される塩基配列(NCBI Reference Sequence: NM_001066.3)が挙げられる。また、ヒトTNFR2としては、各種スプライシングバリアントも包含され得る。
ヒトTNFR2は、その本来の活性、すなわちリガンド(ヒトTNF)の結合に依拠してTregを増殖及び/又は活性化させる活性を有する限りにおいて、置換、欠失、付加、挿入などのアミノ酸変異を有していてもよい。変異としては、活性がより損なわれ難いという観点から、好ましくは置換、より好ましくは保存的置換が挙げられる。
ヒトTNFR2の好ましい具体例としては、下記(a)に記載するタンパク質及び下記(b)に記載するタンパク質:
(a)配列番号12に示されるアミノ酸配列からなるタンパク質、及び
(b)配列番号12に示されるアミノ酸配列と85%以上の同一性を有するアミノ酸配列からなり、且つTregを増殖及び/又は活性化させる活性を有するタンパク質
からなる群より選択される少なくとも1種が挙げられる。
(a)配列番号12に示されるアミノ酸配列からなるタンパク質、及び
(b)配列番号12に示されるアミノ酸配列と85%以上の同一性を有するアミノ酸配列からなり、且つTregを増殖及び/又は活性化させる活性を有するタンパク質
からなる群より選択される少なくとも1種が挙げられる。
上記(b)において、同一性は、より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは98%以上である。
上記(b)に記載するタンパク質の一例としては、例えば
(b’)配列番号12に示されるアミノ酸配列に対して1若しくは複数個のアミノ酸が置換、欠失、付加、又は挿入されたアミノ酸配列からなり、且つTregを増殖及び/又は活性化させる活性を有するタンパク質
上記(b’)において、複数個とは、例えば2~20個であり、好ましくは2~10個であり、より好ましくは2~5個であり、よりさらに好ましくは2又は3個である。
(b’)配列番号12に示されるアミノ酸配列に対して1若しくは複数個のアミノ酸が置換、欠失、付加、又は挿入されたアミノ酸配列からなり、且つTregを増殖及び/又は活性化させる活性を有するタンパク質
上記(b’)において、複数個とは、例えば2~20個であり、好ましくは2~10個であり、より好ましくは2~5個であり、よりさらに好ましくは2又は3個である。
ヒトTNFR2アゴニストドメインは、通常、ヒトTNF-αの変異体を含む。ヒトTNF-αは、分泌型であることが好ましく、例えば配列番号1に示されるアミノ酸配列Aからなる。ヒトTNF-αの変異体は、ヒトTNF-αにアミノ酸変異が導入されてなる変異体であって、ヒトTNFR2に対してアゴニスト活性を有するものである限り、特に制限されない。なお、ヒトTNF-αの変異体には、他の生物(例えばマウス)のTNF-α及びその変異体は包含されない。
ヒトTNF-αの変異体において導入されるアミノ酸変異については、例えば非特許文献1で報告されているような公知の変異を採用することができる。ヒトTNF-αの変異体は、好ましくは配列番号1に示されるアミノ酸配列Aが変異してなるアミノ酸配列Bを含み、且つ前記アミノ酸配列Bが、前記アミノ酸配列AにおけるL29、A145、E146、及びS147からなる群より選択される少なくとも1種のアミノ酸のアミノ酸置換変異を含むことができる。本発明の好ましい態様においては、A145及びS147のアミノ酸置換変異を含むことが好ましく、A145、E146、及びS147のアミノ酸置換変異を含むことがより好ましく、L29、A145、E146、及びS147のアミノ酸置換変異を含むことがさらに好ましい。
L29の変異後のアミノ酸は、側鎖の分子量がより低い脂肪族アミノ酸が好ましい。L29の変異後のアミノ酸としては、好ましくはバリン、アラニン等が挙げられ、特に好ましくはバリンが挙げられる。
A145の変異後のアミノ酸としては、例えばアスパラギン酸、グルタミン酸、ヒスチジン、アルギニン、トレオニン、グルタミン等が挙げられる。A145の変異後のアミノ酸としては、特に好ましくはアスパラギン酸が挙げられる。
E146の変異後のアミノ酸としては、例えばアスパラギン酸、セリン、グリシン等が挙げられる。E146の変異後のアミノ酸としては、好ましくはアスパラギン酸、グルタミン酸等が挙げられ、特に好ましくはアスパラギン酸が挙げられる。
S147の変異後のアミノ酸としては、例えばアスパラギン酸、グルタミン酸等が挙げられる。S147の変異後のアミノ酸としては、特に好ましくはアスパラギン酸が挙げられる。
アミノ酸配列Bにおいては、将来的な分子修飾時の障害にならないように(側鎖のアミノ基がランダムに修飾されることを防ぐために)、アミノ酸配列Aにおけるリシンが他のアミノ酸へ変異していることが好ましい。この観点から、アミノ酸配列Bは、アミノ酸配列AにおけるK11、K65、K90、K98、K112、及びK128からなる群より選択される少なくとも1種(好ましくは2種以上、より好ましくは3種以上、さらに好ましくは4種以上、よりさらに好ましくは5種以上、とりわけ好ましくは6種全て)のアミノ酸のアミノ酸置換変異を含むことが好ましい。変異後のアミノ酸は特に制限されない。本発明の一態様において、K11のアミノ酸置換変異はK11Mであることが好ましく、K65のアミノ酸置換変異はK65Sであることが好ましく、K90のアミノ酸置換変異はK90Pであることが好ましく、K98のアミノ酸置換変異はK98Rであることが好ましく、K112のアミノ酸置換変異はK112Nであることが好ましく、K128のアミノ酸置換変異はK128Pであることが好ましい。
アミノ酸配列Bは、ヒトTNFR2に対するアゴニスト活性が著しく損なわれない限りにおいて、上記以外にも他のアミノ酸変異(例えば置換、保存的置換)を含むことができる。他のアミノ酸変異の数は、例えば1~20個、1~10個、1~5個、又は1~2個である。
アミノ酸配列Bは、好ましくは配列番号2で示されるアミノ酸配列、又は配列番号2で示されるアミノ酸配列に対して85%以上(より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは98%以上)の同一性を有するアミノ酸配列であることができる。
ヒトTNFR2アゴニストドメインは、複数個のヒトTNF-αの変異体を含むことが好ましい。ヒトTNFR2アゴニストドメインは、例えば2~6、好ましくは2~5、より好ましくは2~4、さらに好ましくは3個のヒトTNF-αの変異体を含むことができる。この場合、ヒトTNF-αの変異体間は、リンカー(ペプチドリンカー)を介して連結していることが好ましい。リンカーは、αヘリックス構造やβシート構造などの2次構造を作らず、且つ自由に折り曲がるアミノ酸配列であることが好ましい。リンカーは、数アミノ酸残基からなる単位が連結してなるアミノ酸配列であることが好ましい。ペプチドリンカーはグリシン及びセリンから構成されるリンカー(GSリンカー)が好ましく、具体的に、その構成単位としては、例えばGGGGS(配列番号14)、GSG(配列番号15)、SAGG(配列番号16)、GGGS(配列番号17)、GGSG(配列番号18)、GGG(配列番号19)等が挙げられる。リンカーは、好ましくは配列番号14~18からなる群より選択される1種以上の構成単位1つ以上からなる。リンカーのアミノ酸残基数は、特に制限されるものではないが、例えば3以上、好ましくは3~20、より好ましくは4~10である。
ヒトTNFR2アゴニストドメインは、N末端側から、アミノ酸配列B-リンカー-アミノ酸配列B-リンカーアミノ酸配列B からなるドメイン構造を含むことが好ましい。当該ドメイン構造において、「-」はペプチド結合、又はその両側の配列とペプチド結合で連結された任意のアミノ酸配列(例えば1~10、1~5、又は1~2アミノ酸からなるアミノ酸配列)を示す。
多量体化ドメインは、互いに結合して多量体を形成可能なドメインであり、多ヒトTNFR2アゴニストドメインと連結して一本のポリペプチド鎖を形成可能なものである限り、特に制限されない。当該ドメインとしては、例えば抗体Fc領域、軟骨オリゴマーマトリックスタンパク質ドメイン、ロイシンジッパードメイン、コラーゲン様ドメイン、コレラトキシンBサブユニットドメイン、テトラブラキオンコイルドコアドメイン、レオウイルスσ1タンパク質ドメイン、ヘパチチスデルタ抗原ドメイン等が挙げられる。当該ドメインは、例えばLiprin-b2 Coil2、RP_P2ドメイン等、ヒトのダイマータンパク質の一部であってもよい。
多量体化ドメインとして、特に好ましくは抗体Fc領域が挙げられる。抗体Fc領域は、抗体重鎖のCH2ドメイン及びCH3ドメインを含む領域であり、この限りにおいて特に制限されない。抗体Fc領域は、好ましくは配列番号7で示されるアミノ酸配列、又は配列番号7で示されるアミノ酸配列に対して85%以上(より好ましくは90%以上、さらに好ましくは95%以上、よりさらに好ましくは98%以上)の同一性を有するアミノ酸配列であることができる。
本発明の好ましい一態様においては、ヒトTNFR2アゴニストドメインが、抗体ヒンジ領域及び/又はリンカーを介して多量体化ドメインに連結している。抗体ヒンジ領域は、特に制限されず、例えば配列番号6で示されるアミノ酸配列、又は配列番号6で示されるアミノ酸配列に対して1又は複数個(好ましくは3個以下、より好ましくは2個以下)のアミノ酸が変異してなるアミノ酸配列であることができる。リンカーについては上記で説明したとおりである。
本発明のポリペプチドは、ヒトTNFR2に対するアゴニスト活性が著しく損なわれない限りにおいて、上記以外の他のアミノ酸配列、例えば分泌シグナル配列、タンパク質タグ、蛍光タンパク質、発光タンパク質、プロテアーゼ認識配列(TEVプロテアーゼ認識配列等)等のシグナル配列等のタンパク質又はペプチドが付加されたものであってもよい。タンパク質タグとしては、例えばビオチン、Hisタグ、FLAGタグ、Haloタグ、MBPタグ、HAタグ、Mycタグ、V5タグ、PAタグ等が挙げられる。
本発明のポリペプチドは、ヒトTNFR2に対するアゴニスト活性が著しく損なわれない限りにおいて、化学修飾されたものであってもよい。
本発明のポリペプチドは、C末端がカルボキシル基(-COOH)、カルボキシレート(-COO-)、アミド(-CONH2)またはエステル(-COOR)の何れであってもよい。
ここでエステルにおけるRとしては、例えば、メチル、エチル、n-プロピル、イソプロピル、n-ブチルなどのC1-6アルキル基;例えば、シクロペンチル、シクロヘキシルなどのC3-8シクロアルキル基;例えば、フェニル、α-ナフチルなどのC6-12アリール基;例えば、ベンジル、フェネチルなどのフェニル-C1-2アルキル基;α-ナフチルメチルなどのα-ナフチル-C1-2アルキル基などのC7-14アラルキル基;ピバロイルオキシメチル基などが用いられる。
本発明のポリペプチドは、C末端以外のカルボキシル基(またはカルボキシレート)が、アミド化またはエステル化されていてもよい。この場合のエステルとしては、例えば上記したC末端のエステルなどが用いられる。
さらに、本発明のポリペプチドには、N末端のアミノ酸残基のアミノ基が保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイルなどのC1-6アシル基など)で保護されているもの、生体内で切断されて生成し得るN末端のグルタミン残基がピログルタミン酸化したもの、分子内のアミノ酸の側鎖上の置換基(例えば-OH、-SH、アミノ基、イミダゾール基、インドール基、グアニジノ基など)が適当な保護基(例えば、ホルミル基、アセチル基などのC1-6アルカノイル基などのC1-6アシル基など)で保護されているもの、あるいは糖鎖が結合したいわゆる糖蛋白質などの複合蛋白質なども包含される。
本発明のポリペプチドは、酸または塩基との塩の形態であってもよい。塩は、特に限定されず、酸性塩、塩基性塩のいずれも採用することができる。例えば酸性塩の例としては、塩酸塩、臭化水素酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩; 酢酸塩、プロピオン酸塩、酒石酸塩、フマル酸塩、マレイン酸塩、リンゴ酸塩、クエン酸塩、メタンスルホン酸塩、パラトルエンスルホン酸塩等の有機酸塩; アスパラギン酸塩、グルタミン酸塩等のアミノ酸塩等が挙げられる。また、塩基性塩の例として、ナトリウム塩、カリウム塩等のアルカリ金属塩; カルシウム塩、マグネシウム塩等のアルカリ土類金属塩等が挙げられる。
本発明のポリペプチドは、溶媒和物の形態であってもよい。溶媒は、特に限定されず、例えば水、エタノール、グリセロール、酢酸等が挙げられる。
本発明のポリペプチドは、そのアミノ酸配列に応じて、公知の遺伝子工学的手法に従って容易に作製することができる。例えば、PCR、制限酵素切断、DNA連結技術、in vitro転写・翻訳技術、リコンビナントタンパク質作製技術等を利用して作製することができる。
本発明のポリペプチドは、合成後、精製してもよい。例えば、培養物から遠心、濾過等により集菌し採取した菌体から本発明のタンパク質分子を抽出する。先ず、抽出第一段階としての菌体の破砕には、酵素による消化、浸透圧による破壊、急激な加圧減圧、超音波、種々のホモジナイザー等々を用いることができる。次いで、菌体破砕物を分別するための手段として、物理的な低速遠心、超遠心、濾過、分子ふるい、膜濃縮等、化学的な沈殿剤、可溶化剤、吸脱着剤、分散剤等、物理化学的な電気泳動、カラムクロマトグラフィー、支持体、透析、塩析等を、それぞれ組合せて用いることができる。また、これ等の手段の適用においては、温度、圧力、pH、イオン強度等の物理化学的条件を適宜、設定できる。
3.ポリヌクレオチド、細胞
本発明は、その一態様において、本発明のポリペプチドのコード配列を含む、ポリヌクレオチド(本明細書において、「本発明のポリヌクレオチド」と示すこともある。)、本発明のポリヌクレオチドを含む、細胞(本明細書において、「本発明の細胞」と示すこともある。)に関する。以下に、これらについて説明する。
本発明は、その一態様において、本発明のポリペプチドのコード配列を含む、ポリヌクレオチド(本明細書において、「本発明のポリヌクレオチド」と示すこともある。)、本発明のポリヌクレオチドを含む、細胞(本明細書において、「本発明の細胞」と示すこともある。)に関する。以下に、これらについて説明する。
本発明のポリペプチドのコード配列は、本発明のポリペプチドをコードする塩基配列からなるポリヌクレオチドである限り、特に制限されない。
本発明のポリヌクレオチドは、その一態様において、本発明のポリペプチドの発現カセットを含む。
本発明のポリペプチドの発現カセットは、細胞内で本発明のポリペプチドを発現可能なポリヌクレオチドである限り特に制限されない。本発明のポリペプチドの発現カセットの典型例としては、プロモーター、及びそのプロモーターの制御下に配置された本発明のポリペプチドのコード配列を含むポリヌクレオチドが挙げられる。
本発明のポリペプチドの発現カセットに含まれるプロモーターとしては、特に制限されず、対象細胞に応じて適宜選択することができる。プロモーターとしては、例えばpol II系プロモーターを各種使用することができる。pol II系プロモーターとしては、特に制限されないが、例えばCMVプロモーター、EF1プロモーター、SV40プロモーター、MSCVプロモーター等が挙げられる。その他にも、プロモーターとして、例えばtrcやtac等のトリプトファンプロモーター、lacプロモーター、T7プロモーター、T5プロモーター、T3プロモーター、SP6プロモーター、アラビノース誘導プロモーター、コールドショックプロモーター、テトラサイクリン誘導性プロモーター等が挙げられる。
本発明のポリヌクレオチドは、必要に応じて、他のエレメント(例えば、マルチクローニングサイト(MCS)、薬剤耐性遺伝子、複製起点、エンハンサー配列、リプレッサー配列、インスレーター配列、レポータータンパク質(例えば、蛍光タンパク質等)コード配列、薬剤耐性遺伝子コード配列、ガイドRNA発現カセットなどが挙げられる。)を含んでいてもよい。
本発明のポリヌクレオチドは、ベクターの形態であることができる。使用目的(クローニング、タンパク質の発現)に応じて、また宿主細胞の種類を考慮して適当なベクターが選択される。大腸菌を宿主とするベクターとしてはM13ファージ又はその改変体、λファージ又はその改変体、pBR322又はその改変体(pB325、pAT153、pUC8など)等、酵母を宿主とするベクターとしてはpYepSec1、pMFa、pYES2、pPIC3.5K等、昆虫細胞を宿主とするベクターとしてはpAc、pVL等、哺乳類細胞を宿主とするベクターとしてはpcDNA、pCDM8、pMT2PC等を例示することができる。
本発明の細胞は、本発明のポリヌクレオチドを含む限り特に制限されない。細胞としては、例えば、Escherichia coli K12等の大腸菌、Bacillus subtilis MI114等のバチルス属細菌、Saccharomyces cerevisiae AH22等の酵母、Spodoptera frugiperda 由来の Sf細胞系もしくはTrichoplusia ni由来のHighFive細胞系、嗅神経細胞等の昆虫細胞、COS7細胞等の動物細胞等を挙げることができる。動物細胞としては、好ましくは、哺乳動物由来の培養細胞、具体的には、COS7細胞、CHO細胞、HEK293細胞、HEK293FT細胞、Hela細胞、PC12細胞、N1E-115細胞、SH-SY5Y細胞等が挙げられる。
本発明の細胞は、その一態様において、本発明のポリペプチドを発現している。
4.複合体
本発明は、その一態様において、本発明のポリペプチド複数個が、前記多量体化ドメインを介して連結してなる、複合体(本明細書において、「本発明の複合体」と示すこともある。)、に関する。以下に、これについて説明する。

本発明の複合体においては、連結した本発明のポリペプチドの数に応じて、複数のヒトTNFR2アゴニストドメインを含む。
本発明の複合体は、ヒトTNFR2アゴニストとして好適に使用することができる。本発明の複合体は、エフェクターTregを選択的に増加させることができる。
5.医薬
本発明は、その一態様において、本発明のポリペプチド、本発明のポリヌクレオチド、本発明の細胞、及び本発明の複合体からなる群より選択される少なくとも1種を含有する、医薬(本明細書において、「本発明の医薬」と示すこともある。)、に関する。以下に、これについて説明する。

本発明の医薬は、本発明のポリペプチド、本発明の複合体のヒトTNFR2に対するアゴニスト活性に基づいて、或いは本発明のポリヌクレオチド、本発明の細胞から発現する本発明のポリペプチド、本発明の複合体のヒトTNFR2に対するアゴニスト活性に基づいて、各種用途、例えば免疫抑制剤、自己免疫疾患の予防又は治療剤、臓器移植の拒絶反応の抑制剤、又はアレルギー疾患の予防又は治療剤として用いることができる。
自己免疫疾患としては、特に制限されないが、例えば全身性エリテマトーデス(SLE)、CREST症候群(石灰沈着症、レイノー症候群、食道運動障害、強指症及び毛細血管拡張症)、オプソクローヌス、炎症性ミオパチー(例えば、多発性筋炎、皮膚筋炎及び封入体筋炎)、全身性強皮症、原発性胆汁性肝硬変、セリアック病(例えば、グルテン過敏性腸疾患)、疱疹状皮膚炎、ミラー・フィッシャー症候群、急性運動性軸索型ニューロパチー(AMAN)、伝導ブロックを伴う多巣性運動ニューロパチー、自己免疫性肝炎、抗リン脂質抗体症候群、ウェゲナー肉芽腫症、顕微鏡的多発血管炎、チャーグ・ストラウス症候群、関節リウマチ、慢性自己免疫性肝炎、硬化性筋炎、重症筋無力症、ランバート・イートン筋無力症候群、橋本甲状腺炎、グレーブス病、傍腫瘍性小脳変性症、スティッフパーソン症候群、辺縁系脳炎、アイザックス症候群、シデナム舞踏病、小児自己免疫性溶連菌関連性精神神経疾患(PANDAS)、脳炎、1型真性糖尿病並びに視神経脊髄炎が挙げられる。他の自己免疫性障害としては、悪性貧血、アジソン病、乾癬、炎症性腸疾患、乾癬性関節炎、シェーグレン症候群、紅斑性狼瘡(例えば、円板状紅斑性狼瘡、薬物誘発性紅斑性狼瘡及び新生児紅斑性狼瘡)、多発性硬化症反応性関節炎等が挙げられる。
アレルギー疾患としては、特に制限されないが、例えば喘息、呼吸器アレルギー(例えば花粉アレルギー)、虫刺されによるアレルギー、アレルギー性鼻炎(花粉症)、食物アレルギー、薬物アレルギー、ラテックスアレルギー、アトピー性皮膚炎(湿疹)、接触性皮膚炎等が挙げられる。
本発明の医薬中の有効成分の含有量は、対象とする疾患の種類、目的とする治療効果、投与方法、治療期間、患者の年齢、及び患者の体重などを考慮して適宜設定することができる。例えば、本発明の医薬中の有効成分の含量は、本発明の医薬全体を100重量部として0.0001重量部~100重量部程度をすることができる。
本発明の医薬の投与形態は、所望の効果が得られる限り特に制限されず、経口投与、及び非経口投与(例えば静脈注射、筋肉注射、皮下投与、直腸投与、経皮投与、局所投与)のいずれかの投与経路でヒトを含む哺乳類に投与することができる。好ましい投与形態は非経口投与である。経口投与および非経口投与のための剤形およびその製造方法は当業者に周知であり、有効成分を、薬学的に許容される坦体などと混合などすることにより、常法に従って製造することができる。
非経口投与のための剤形は、注射用製剤(例えば、点滴注射剤、静脈注射剤、筋肉注射剤、皮下注射剤、皮内注射剤)、外用剤(例えば、軟膏剤、パップ剤、ローション剤、クリーム、ゲル剤)、坐剤、吸入剤、眼剤、眼軟膏剤、点鼻剤、点耳剤、リポソーム剤などが挙げられる。例えば、注射用製剤は、本発明のオリゴペプチドを注射用蒸留水に溶解して調製し、必要に応じて溶解補助剤、緩衝剤、pH調整剤、等張化剤、無痛化剤、保存剤、及び安定化剤などを添加することができる。本発明の医薬は、用事調製用の凍結乾燥製剤とすることもできる。
本発明の医薬は、疾患の治療又は予防に有効な他の薬剤を更に含有していてもよい。本発明の医薬は、必要に応じて殺菌剤、消炎剤、細胞賦活剤、ビタミン類、及びアミノ酸などの成分を配合することもできる。
本発明の医薬の製剤化に用いる担体には、当該技術分野において通常用いられる賦形剤、結合剤、崩壊剤、滑沢剤、着色剤、矯味矯臭剤や、必要により安定化剤、乳化剤、吸収促進剤、界面活性剤、pH調整剤、防腐剤、抗酸化剤、増量剤、湿潤化剤、表面活性化剤、分散剤、緩衝剤、保存剤、溶解補助剤、無痛化剤などを用いることができる。
本発明の医薬の投与量は、例えば、投与経路、疾患の種類、症状の程度、患者の年齢、性別、体重、疾患の重篤度、薬物動態および毒物学的特徴などの薬理学的知見、薬物送達系の利用の有無、並びに他の薬物の組合せの一部として投与されるか、など様々な因子を元に、決定することができる。本発明の医薬の投与量は、例えば、一日当たりで、1μg/kg(体重)~10g/kg(体重)程度とすることができる。本発明の医薬の投与スケジュールも、その投与量と同様の要因を勘案して決定することができる。例えば、上記の1日当たりの投与量で、1日~1月に1~5回投与することができる。
以下に、実施例に基づいて本発明を詳細に説明するが、本発明はこれらの実施例によって限定されるものではない。
実施例1.scR2-7-Fcタンパク質の作製
N末端側から、
(1)マウスIgG重鎖由来分泌シグナル配列(配列番号3)、
(2)ヒトTNF-αの変異体配列(配列番号2:ヒトTNF-α野生型アミノ酸配列(配列番号1)に対して、ヒトTNFR2結合領域の変異(L29V、A145D、E146D、及びS147D)を導入し、さらに将来的な分子修飾時の障害にならないように(側鎖のアミノ基がランダムに修飾されることを防ぐために)リシンの他のアミノ酸への変異(K11M、K65S、K90P、K98R、K112N、及びK128P)を導入してなるアミノ酸配列)、
(3)リンカー配列(配列番号4)、
(4)ヒトTNF-αの変異体配列(配列番号2)、
(5)リンカー配列(配列番号4)、
(6)ヒトTNF-αの変異体配列(配列番号2)、
(7)リンカー・スペーサー配列(配列番号5)、
(8)抗体ヒンジ配列(配列番号6)、
(9)抗体Fc配列(配列番号7:ヒトIgG CH2配列(配列番号8)+ヒトIgG CH3配列(配列番号9))、
の順に配置されてなるアミノ酸配列(配列番号10:図1)からなるscR2-7-Fcタンパク質のコード配列(配列番号11)が哺乳類発現用ヒトIgG1-Fc 融合プラスミドベクター(pCAG-Hyg hIgG-Fc)のマルチクローニングサイト内に配置されてなるscR2-7-Fc発現用プラスミド(pCAG-scR2-7-Fc)を作製した。
N末端側から、

(7)リンカー・スペーサー配列(配列番号5)、
(8)抗体ヒンジ配列(配列番号6)、
(9)抗体Fc配列(配列番号7:ヒトIgG CH2配列(配列番号8)+ヒトIgG CH3配列(配列番号9))、
の順に配置されてなるアミノ酸配列(配列番号10:図1)からなるscR2-7-Fcタンパク質のコード配列(配列番号11)が哺乳類発現用ヒトIgG1-Fc 融合プラスミドベクター(pCAG-Hyg hIgG-Fc)のマルチクローニングサイト内に配置されてなるscR2-7-Fc発現用プラスミド(pCAG-scR2-7-Fc)を作製した。
Expi293F細胞(Thermo)を用いてscR2-7-Fcタンパク質を発現させた。Expi293F 細胞は、無血清培地 Expi293 Expression Medium(Thermo scientific社)を用いて培養した。リポフェクション試薬(ExpiFectamine 293 reagent)を用いて、pCAG-scR2-7-FcプラスミドをExpi293F細胞に遺伝子導入した後、37℃, 8% CO2下で7日間振とう培養した。培養上清を回収した後、プロテインAアフィニティクロマトグラフィー(Wako)及びゲルろ過クロマトグラフィー(Hiload 16/600 Superdex 200 pg column, GE Healthcare)の2段階精製を行い、目的物(scR2-7-Fc複合体:2つのscR2-7-Fcタンパク質が抗体Fc領域を介して連結してなる複合体)を回収した。
実施例2.TNFR2 発現細胞を用いたアゴニスト活性の測定
TNFR2(細胞外)と細胞死誘導受容体Fas(細胞内)を融合したTNFR2/Fas強制発現細胞(mouse TNFR2/Fas-preadipocyte)を用いて、細胞死を指標にしてTNFR2を介したアゴニスト活性を調べた。具体的には以下のようにして調べた。
TNFR2(細胞外)と細胞死誘導受容体Fas(細胞内)を融合したTNFR2/Fas強制発現細胞(mouse TNFR2/Fas-preadipocyte)を用いて、細胞死を指標にしてTNFR2を介したアゴニスト活性を調べた。具体的には以下のようにして調べた。
mouse TNFR2/Fas-preadipocyteは、10% ウシ胎児血清、1% 抗生物質カクテル(Wako)及び5 μg/mLブラストサイジンS(Wako)を含むD-MEM(Wako)を用いて、37℃で培養した。96ウェルプレートに 3×104 cells/well の細胞を播種し、段階希釈した scR2-7-Fc複合体を添加した。また、コントロールとして、配列番号2で示されるアミノ酸配列からなるタンパク質が3つ会合してなる3量体(TNFR2アゴニスト(R2-7))、及び配列番号2に代えて野生型配列(配列番号1)を採用する以外はR2-7と同じ構成のタンパク質(ヒト野生型TNF(humanwtTNF))についても、段階希釈して添加した。37℃で48 時間培養した後、WST-8アッセイ(Dojindo)を行い、吸光度(450 nm)を測定した。scR2-7-Fc添加群の吸光度値を非添加群の吸光度値で除して、細胞生存率を算出し、アゴニスト活性を比較した。
結果を図2に示す。ヒト野生型TNFやR2-7(TNFR2アゴニスト)に比べて、scR2-7-Fc複合体は低濃度で効果を示し、約100倍活性が高いことが分かった。
実施例3.In vitroヒトPBMCアッセイ
ヒト末梢血単核球(Precision for Medicine社)を96ウェルプレートに播種(5*10E4 cells/well)した後、scR2-7-Fc複合体(100 ng/mL)を加えて、37℃で72時間培養した。また、コントロールとしてIL-2(10 U/mL)を添加した。細胞を回収した後、抗ヒトCD4抗体-APC、抗ヒトCD25抗体-PE、抗ヒトCD45RA抗体-FITCを加えて、蛍光免疫染色を行った。フローサイトメトリーを用いて、分画I(CD4+CD45RA+CD25low細胞)、分画II(CD4+CD45RA-CD25high細胞)、分画III(CD4+CD45RA-CD25low細胞)におけるTregの割合(%)を測定した(N=5)。
ヒト末梢血単核球(Precision for Medicine社)を96ウェルプレートに播種(5*10E4 cells/well)した後、scR2-7-Fc複合体(100 ng/mL)を加えて、37℃で72時間培養した。また、コントロールとしてIL-2(10 U/mL)を添加した。細胞を回収した後、抗ヒトCD4抗体-APC、抗ヒトCD25抗体-PE、抗ヒトCD45RA抗体-FITCを加えて、蛍光免疫染色を行った。フローサイトメトリーを用いて、分画I(CD4+CD45RA+CD25low細胞)、分画II(CD4+CD45RA-CD25high細胞)、分画III(CD4+CD45RA-CD25low細胞)におけるTregの割合(%)を測定した(N=5)。
結果を図3及び図4に示す。scR2-7-Fc複合体により、エフェクターTregが選択的に増殖したことが分かった。
Claims (13)
- ヒトTNFR2アゴニストドメイン、及び多量体化ドメインを含み、前記ヒトTNFR2アゴニストドメインが前記多量体化ドメインに連結している、ポリペプチド。
- 前記ヒトTNFR2アゴニストドメインがヒトTNF-αの変異体を含む、請求項1に記載のポリペプチド。
- 前記ヒトTNF-αの変異体が、配列番号1に示されるアミノ酸配列Aが変異してなるアミノ酸配列Bを含み、且つ前記アミノ酸配列Bが、前記アミノ酸配列AにおけるL29、A145、E146、及びS147からなる群より選択される少なくとも1種のアミノ酸のアミノ酸置換変異を含む、請求項2に記載のポリペプチド。
- L29の変異後のアミノ酸がバリン又はアラニンであり、且つA145、E146、及びS147の変異後のアミノ酸がアスパラギン酸又はグルタミン酸である、請求項3に記載のポリペプチド。
- 前記ヒトTNFR2アゴニストドメインが、N末端側から、アミノ酸配列B-リンカー-アミノ酸配列B-リンカーアミノ酸配列B からなるドメイン構造を含む、請求項3又は4に記載のポリペプチド。
- 前記多量体化ドメインが抗体Fc領域を含む、請求項1~5のいずれかに記載のポリペプチド。
- 前記ヒトTNFR2アゴニストドメインが、抗体ヒンジ領域及び/又はリンカーを介して前記多量体化ドメインに連結している、請求項1~6のいずれかに記載のポリペプチド。
- 請求項1~7のいずれかに記載のポリペプチドのコード配列を含む、ポリヌクレオチド。
- 請求項8に記載のポリヌクレオチドを含む、細胞。
- 請求項1~7のいずれかに記載のポリペプチド複数個が、前記多量体化ドメインを介して連結してなる、複合体。
- 請求項10に記載の複合体を含む、ヒトTNFR2アゴニスト。
- 請求項1~7のいずれかに記載のポリペプチド、請求項8に記載のポリヌクレオチド、請求項9に記載の細胞、及び請求項10に記載の複合体からなる群より選択される少なくとも1種を含有する、医薬。
- 免疫抑制剤、自己免疫疾患の予防又は治療剤、臓器移植の拒絶反応の抑制剤、又はアレルギー疾患の予防又は治療剤である、請求項12に記載の医薬。
