Abstract
본 발명은 아토피성 피부염 예방, 개선 또는 치료 효과를 가지는 조성물에 관한 것으로, 천연물 추출물을 유효성분으로 포함하는 아토피성 피부염 예방, 개선 또는 치료용 조성물에 관한 것이다. 보다 구체적으로는 느타리 버섯 추출물 및 물푸레 나무 추출물을 유효성분으로 포함하는 아토피성 피부염 예방, 개선 또는 치료용 조성물에 관한 것이다.
Description
본 발명은 아토피성 피부염 예방 또는 개선 효과를 가지는 조성물에 관한 것으로, 보다 구체적으로는 천연물 추출물을 유효성분으로 포함하는 아토피성 피부염 예방 또는 개선용 조성물에 관한 것이다.
아토피는 최근 들어, 유병률이 급격히 증가함에 따라 그 위험성이 크게 부각되고 있는 과민증(hypersensitivity)의 일종으로, 면역학적 관점에서는 면역글로불린 E(immunoglobulin E; IgE)와 알레르겐(allergen)의 면역반응에 의한 주요증상을 동반하는 유전경향이 강한 알레르기성 질환군으로 정의된다. 특히, 아토피의 대표적 증상인 아토피 피부염(atopic dermatitis)은 전 인구의 0.5~1%, 어린이의 5~10%가 심각한 증상을 호소하고 있지만, 유전적인 요인과 면역계 결핍에 관련 있는 것으로 추정될 뿐 아직 정확한 원인은 밝혀진 바 없고, 환경 및 식생활 개선을 통해 다소 완화되는 것으로 기대될 뿐 근본적인 치료방법이 전무한 실정이다.
그러나 아토피 피부염은 개인차를 보이기는 하지만, 대부분 극심한 가려움증을 비롯한 피부 건조증, 발진, 진물, 부스럼딱지, 비늘 같은 껍질이 있는 피부(인비늘) 등을 수반하며, 이로 인한 정서적 불안, 스트레스, 긴장, 좌절, 분노의 감정과 함께 기타 알레르기 질환인 두드러기, 금속 알레르기, 천식이나 알레르기성 비염 등을 동반하는 경우가 빈번한 바, 오늘날 국민건강을 위협하는 심각한 사회문제로 떠오르고 있다. 한편, 현재까지 아토피 피부염에 효과가 있는 것으로 알려진 대표적 약물로는 스테로이드제(steroid), 항히스타민제(antihistamine), 항생제(antibiotic), 비스테로이드제를 비롯한 기타 진정제와 신경안정제 등을 들 수 있다.
한편, 알러지(allergy)는 면역 시스템의 오작동으로 보통 사람에게는 별 영향이 없는 물질이 어떤 사람에게는 두드러기, 가려움, 콧물 또는 기침 등의 이상 과민 반응을 일으키는 것을 말하며, WHO에서는 이미 알러지 질환을 21세기에 가장 문제가 되는 제7대 질병으로 포함하였다. 주요한 알러지 질환으로는 아토피 피부염, 천식, 비염 등이 있다. 알러지는 현재까지 뚜렷한 치료제가 없을 정도로 현대의학의 난치병에 가까운 질환이다. 현재 알러지 질환 치료에 있어서 근본적인 치료제가 아닌 항히스타민제, 효과는 우수하나 부작용이 많은 스테로이드의 사용으로 질환은 더욱 악화되고 환자의 경제적 부담은 계속 증가하고 있다.
대체로, 아토피 증상이나 알러지 질환은 산업의 발달, 식생활의 서구화 및 인스턴트화, 심각한 환경오염 발생, 새집 증후군과 같은 신종 질병 등과 함께 동반되면서, 21세기에 접어들어 더욱 꾸준히 증가하고 있는 추세이다.
상기와 같은 문제를 해결하기 위하여, 본 발명의 일측면은 아토피 피부염의 예방 또는 개선용 조성물을 제공하는 것이다.
본 발명의 일 실시예에 따른 아토피 피부염의 예방 또는 개선용 화장료 조성물은, 느타리버섯 추출물 및 물푸레나무 추출물을 유효성분으로 포함할 수 있다.
하나의 바람직한 예에서, 상기 느타리버섯 추출물은 큰느타리버섯 추출물일 수 있다.
하나의 바람직한 예에서, 상기 물푸레나무 추출물은 물푸레나무 진피 추출물일 수 있다.
본 발명의 일 실시예에 따른 아토피 피부염의 예방 또는 치료용 약학적 조성물은, 느타리버섯 추출물 및 물푸레나무 추출물을 유효성분으로 포함할 수 있다.
하나의 바람직한 예에서, 상기 느타리버섯 추출물은 큰느타리버섯 추출물일 수 있다.

다르게 정의되지 않는 한, 기술적이거나 과학적인 용어를 포함해서 여기서 사용되는 모든 용어들은 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자에 의해 일반적으로 이해되는 것과 동일한 의미이다. 일반적으로 사용되는 사전에 정의되어 있는 것과 같은 용어들은 관련 기술의 문맥상 가지는 의미와 일치하는 의미인 것으로 해석되어야 하며, 본 출원에서 명백하게 정의하지 않는 한, 이상적이거나 과도하게 형식적인 의미로 해석되지 않는다.
본 발명은 느타리버섯 추출물 및 물푸레나무 추출물을 유효성분으로 포함하는 아토피 피부염 예방, 개선 또는 치료용 조성물에 관한 것이다.
느타리버섯은 담자균류 주름버섯목 느타리과의 버섯으로, 활엽수의 고목에 군생하며, 특히 늦가을에 많이 발생한다. 갓은 너비 5~15 cm로 반원형 또는 약간 부채꼴이며 가로로 짧은 줄기가 달린다. 표면은 어릴 때는 푸른빛을 띤 검은색이지만 차차 퇴색하여 잿빛에서 흰빛으로 되며 매끄럽고 습기가 있다. 살은 두텁고 탄력이 있으며 흰색이다.
느타리버섯의 한 종류로 큰느타리버섯이 있는데, 이는 송이버섯과 비슷한 식감을 가진다 해서 일반적으로 새송이버섯으로 많이 알려져 있다.
물푸레나무는 쌍떡잎식물 용담목 물푸레나무과의 낙엽교목으로, 산기슭이나 골짜기 물가에서 자란다. 높이는 10 m이고, 나무 껍질은 회색을 띤 갈색이며 잿빛을 띤 흰 빛깔의 불규칙한 무늬가 있다. 잎은 마주나고 홀수 1회 깃꼴겹잎이고, 작은 잎은 5~7개이며 길이 6~15 cm의 넓은 바소 모양 또는 바소 모양이고 가장자리에 물결 모양의 톱니가 있으며 앞면에 털이 없고 뒷면 맥 위에 털이 있다.
추출물은 용매를 사용하여 성분을 추출해낸 것으로, 추출용매는 물, C1~C4의 저급 알코올 또는 이들의 혼합물일 수 있다. 바람직하게는, 물 또는 에탄올일 수 있지만 이들로 한정되는 것은 아니다.
본 발명의 화장료 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 계면활성제 함유 클렌징, 오일, 분말 파운데이션, 유탁액, 파운데이션, 왁스 파운데이션 및 스프레이 중에서 선택되는 어느 하나의 제형으로 화장품으로 제조될 수 있지만, 이것으로 한정되는 것은 아니고 화장품으로 사용하기에 적합한 제형이면 어느 것이나 가능하다.
상기 화장품의 제형이 용액 또는 유탁액일 경우에는 담체 성분으로 용매, 용매화제 또는 유탁화제가 이용될 수 있다. 예를 들어, 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있지만 이것만으로 한정되는 것은 아니다.
상기 화장품의 제형이 현탁액인 경우에는 담체 성분으로서, 물, 에탄올 똔느 프로필렌 글리콜과 같은 액상 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
상기 화장품의 제형이 페이스트, 크림, 또는 겔인 경우에는 담체 성분으로서 동물섬유, 식물섬유, 왁스, 파라핀, 전분, 트라가칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다. 본 발명의 화장료 조성물의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판-부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
상기 화장품의 제형이 계면-활성제 함유 클렌징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 아세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 라놀린 유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
상기 화장품은 본 발명의 화장료 조성물 이외에 추가로 기능성을 나타내는 성분을 1종 이상 함유할 수 있다.
본 발명의 약학적 조성물은 상기 유효성분 이외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 더 포함할 수 있으며, 본 발명의 약학 조성물은 경구 또는 비경구로 투여될 수 있으며, 비경구 투여 시 피부 외용 또는 복강 내, 직장, 정맥, 근육 또는 피하주사 방식을 선택하는 것이 바람직하지만 이에 한정하는 것은 아니다.
본 발명의 약학 조성물은 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성 용제 및 현탁 용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
본 발명에 따른 유효성분은 약학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물의 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설률 및 질환의 중증도에 따라 그 범위가 다양하게 사용할 수 있다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하기로 한다. 이들 실시예는 본 발명을 보다 구체적으로 설명하기 위한 것으로서, 본 발명의 범위가 이들 실시예로 한정되는 것은 아니다.
스크리닝 원리
사람의 생체 내에서 알러지 반응을 일으키는 주된 면역 항체인 IgE와 비만세포(Dendritic cell)의 표면에 존재하는 IgE 수용체(FcεR1)가 결합하면서 세포 내 신호전달 과정을 통해 히스타민이 방출되고 이로 인해 여러 자극들이 나타나게 된다. 즉, IgE와 FcεR1 사이의 상호작용(interaction)을 방해할 수 있는 소재를 발굴할 경우, 아토피 피부염의 예방, 개선 또는 치료용 유효성분으로 사용이 가능하다.
특히, 특정 알러젠(allergen)이 들어와서 IgE에 결합되더라도 FcεR1에 결합하는 것을 방해할 수 있다면 아토피 피부염 뿐만 아니라 알러지 반응이 나타나지 않을 것이다.

각 well 내부가 단백질이 고정될 수 있도록 표면이 Linker 물질로 코팅된 단백질칩(자체 제작)에 1X PBS(10% PEG-200, pH 7.4) 완충용액에 적당한 농도로 FcεR1을 희석한 용액을 가하여 4 ℃에서 8시간 이상 고정시키고, 1X PBST(0.5% Tween-20, pH 7.4) 용액으로 10분간 2회 세척하고 비특이적 결합을 방지하기 위하여 1% BSA(1X PBS, pH 7.4) 용액에 담가 상온에서 Shaking 하면서 1시간 동안 Blocking 한다. 이후 1X PBST(0.5% Tween-20, pH 7.4) 용액으로 10분간 2회 세척하고 증류수로 rinsing 한 다음 질소 가스를 이용하여 웰에 남아있는 물기를 건조한다. 느타리버섯 추출물을 각 well에 0.5 ㎕씩 spotting 한 다음, 같은 위치에 biotin이 결합된 IgE 용액을 0.5 ㎕씩 spotting 하고 건조되지 않도록 wet box에 넣어 상온에서 1시간 동안 배양한다. 앞서 단계와 동일한 방법으로 wash & dry 한 다음 상기 biotin과 강하게 결합하는 Streptavidin(Cy5-labelled) 용액을 1 ㎕씩 spotting 하고 wet box에 넣어 상온에서 1시간 동안 배양하여 단백질칩을 처리하였다.
실시예 2(B)
느타리버섯 추출물 0.5 ㎕ 대신 물푸레나무 추출물 0.5 ㎕를 사용한 것 이외에 실시예 1과 동일한 방법으로 단백질칩을 처리하였다.
실시예 3(A+B)
느타리버섯 추출물 0.5 ㎕ 대신 느타리버섯 추출물과 물푸레나무 추출물의 1:1 혼합물(A+B) 0.5 ㎕를 사용한 것 이외에 실시예 1과 동일한 방법으로 단백질칩을 처리하였다.
비교예 1(PC1)
느타리버섯 추출물 0.5 ㎕ 대신 DMSO buffer 0.5 ㎕를 사용한 것 이외에 실시예 1과 동일한 방법으로 단백질칩을 처리하였다.
비교예 2(NC1)
느타리버섯 추출물 0.5 ㎕ 및 IgE 0.5 ㎕ 대신 DMSO buffer 1.0 ㎕를 사용한 것 이외에 실시예 1과 동일한 방법으로 단백질칩을 처리하였다.
실험예 1
비교예 1, 비교예 2, 실시예 1, 실시예 2 및 실시예 3 각각의 단백질칩을 마이크로어레이 스캐너(microarray scanner)로 관찰하여 결과를 분석하였다. 스캔 결과는 각각의 well에서 나타나는 형광값을 0 부터 216값인 65535 사이의 수치로 표현하였다.
도 1은 실험예 1에 따른 스캔 결과를 나타내는 그래프이다. 도 1을 참조하면, 비교예 1(PC1)은 IgE와 FcεR1 사이에 인위적인 방해요소가 없는 경우의 형광 발현 수준이고, 비교예 2(NC1)는 IgE가 없는 경우의 형광 발현 수준이다. 이들 비교예 1 및 비교예 2가 스크리닝 하고자 하는 실시예 1 내지 3의 결과값들의 기준값이 되고 이들에 대한 상대적인 비교를 통해 IgE와 FcεR1의 결합 방해 효과를 확인할 수 있다.
실시예 1(A) 및 실시예 2(B)의 경우, 비교예 1(PC1)과 비교하여 IgE와 FcεR1의 결합 방해 효과가 일부 있기는 하지만, 유의미한 수준은 아닌 것으로 판단된다. 다만, 실시예 3(A+B)의 경우에는 비교예 1(PC1) 대비 약 절반에 가까운 IgE와 FcεR1의 결합 방해 효과를 나타내고 있다.
따라서 느타리버섯 추출물과 물푸레나무 추출물의 혼합물의 경우 각각의 단독 사용에 비해 현저한 시너지 효과를 나타내는 것으로 판단된다.
상술한 바와 같이, 본 발명의 바람직한 실시예를 참조하여 설명하였지만 해당 기술 분야에서 통상의 지식을 가진 자라면 하기의 특허청구범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.

- 느타리버섯 추출물 및 물푸레나무 추출물을 유효성분으로 포함하는 것을 특징으로 하는 아토피 피부염의 예방 또는 개선용 화장료 조성물.
- 제 1 항에 있어서,상기 느타리버섯 추출물은 큰느타리버섯 추출물인 것을 특징으로 하는 아토피 피부염의 예방 또는 개선용 화장료 조성물.
- 제 1 항에 있어서,물푸레나무 추출물은 물푸레나무 진피 추출물인 것을 특징으로 하는 아토피 피부염의 예방 또는 개선용 화장료 조성물.
- 느타리버섯 추출물 및 물푸레나무 추출물을 유효성분으로 포함하는 것을 특징으로 하는 아토피 피부염의 예방 또는 치료용 약학적 조성물.
- 제 4 항에 있어서,상기 느타리버섯 추출물은 큰느타리버섯 추출물인 것을 특징으로 하는 아토피 피부염의 예방 또는 치료용 약학적 조성물.
- 제 4 항에 있어서,물푸레나무 추출물은 물푸레나무 진피 추출물인 것을 특징으로 하는 아토피 피부염의 예방 또는 치료용 약학적 조성물.
