Abstract
ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、血管内皮細胞を含む組織体の製造方法。
Description
本発明は、組織体の製造方法及びヒト由来の脂肪由来幹細胞の血管内皮細胞への分化促進方法に関する。
人工的に生体組織を模した構造体を作製する手法として、例えば、コラーゲンを含む被膜でコートされた細胞を三次元に配置して、三次元組織体を形成することを含む、三次元組織体を製造する方法(特許文献1)、細胞をカチオン性物質及び細胞外マトリックス成分と混合して混合物を得て、得られた混合物から細胞を集めて、基材上に細胞集合体を形成することを含む、立体的細胞組織の製造方法(特許文献2)等が知られている。また、本発明者らは、細胞と断片化された外因性コラーゲンを接触させることで、比較的少ない細胞数で、厚さが1mm以上である、サイズが大きい三次元組織体を製造する方法(特許文献3)を提案している。
上述の製造方法によれば、細胞培養によって人工的に作られる細胞の集合体である組織体を得ることができる。特に、血管内皮細胞を含む組織体は、実験動物の代替品、移植材料等としての利用が期待されている。ここで、人工的に組織体を製造する際には、ウシ等の他生物種を由来とする脂肪由来幹細胞、同じヒトであっても異なる個体から採取されたアロジェニックな細胞等が使用されることが多いものの、例えば、ヒトへの移植材料を製造する際には、当該ヒトの自家細胞を使用して移植材料を製造することが想定される。そのため、特にヒト由来の脂肪由来幹細胞を用いて組織体を製造する際に、当該脂肪由来幹細胞の血管内皮細胞への分化を促進する方法が望まれている。
本発明は上記事情に鑑みてなされたものであり、血管内皮細胞を含む組織体の製造において、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化を促進させる方法を提供することを目的とする。
本発明者らは鋭意研究を重ねた結果、ヒト由来の脂肪由来幹細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることにより、脂肪由来幹細胞の血管内皮細胞への分化が促進されることを見出し、本発明を完成させるに至った。
すなわち、本発明は、例えば、以下の発明を含む。

本発明によれば、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化が促進される。血管内皮細胞への分化を促進することで、短時間で血管内皮細胞を含む組織体を製造可能となる。
以下、本発明を実施するための形態について詳細に説明する。ただし、本発明は以下の実施形態に限定されるものではない。
本発明は、一実施形態として、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、血管内皮細胞を含む組織体の製造方法を提供する。
本明細書において、「組織体」とは、細胞培養によって人工的に作られる細胞の集合体を意味する。そのうち、「三次元組織体」とは、細胞培養によって人工的に作られ、細胞が三次元的に配置されている細胞の集合体(塊状の細胞集団)を意味する。三次元組織体が後述する細胞外マトリックス成分を含む場合には、細胞外マトリックス成分を介して細胞が三次元的に配置される。組織体の形状は、特に制限はなく、例えば、シート状、球体状、略球体状、楕円体状、略楕円体状、半球状、略半球状、半円状、略半円状、直方体状、略直方体状等が挙げられる。ここで、生体組織は、汗腺、リンパ管、脂腺等を含み、構成が組織体より複雑である。そのため、組織体と生体組織とは容易に区別可能である。
本明細書において「細胞」は、特に限定されないが、例えば、ヒト、サル、イヌ、ネコ、ウサギ、ブタ、ウシ、マウス、ラット等の哺乳類動物に由来する細胞であってよい。細胞の由来部位も特に限定されず、骨、筋肉、内臓、神経、脳、骨、皮膚、血液等に由来する体細胞であってもよく、生殖細胞であってもよい。さらに、細胞は、幹細胞であってもよく、また、初代培養細胞、継代培養細胞及び細胞株細胞等の培養細胞であってもよい。
本明細書において「幹細胞」とは、自己複製能及び多分化能を有する細胞を意味する。幹細胞には、任意の細胞種に分化する能力を持つ多能性幹細胞と、特定の細胞種に分化する能力を持つ組織幹細胞(体性幹細胞とも呼ばれる)が含まれる。多能性幹細胞としては、例えば、胚性幹細胞(ES細胞)、体細胞由来ES細胞(ntES細胞)及び人工多能性幹細胞(iPS細胞)が挙げられる。組織幹細胞としては、例えば、間葉系幹細胞(例えば、脂肪由来幹細胞、骨髄由来幹細胞)、造血幹細胞及び神経幹細胞が挙げられる。脂肪由来幹細胞としては、例えば、ヒト脂肪由来幹細胞(ADSC)が挙げられる。
細胞は、少なくともヒト由来の脂肪由来幹細胞を含む。ヒト由来の脂肪由来幹細胞は、ヒトの生体組織、例えば、ヒトの皮下脂肪組織及び心外膜由来脂肪組織等から採取した幹細胞を用いてもよい。本実施形態の方法により製造する血管内皮細胞を含む組織体を、最終的にヒトの生体の特定箇所の組織に見立てて利用する場合には、当該箇所の組織に対応する組織由来のものを用いることが好ましい。
本実施形態において細胞は、少なくともヒト由来の脂肪由来幹細胞を含むが、脂肪由来幹細胞以外の細胞を更に含んでいてもよい。脂肪由来幹細胞以外の細胞としては、例えば、成熟脂肪細胞、血管内皮細胞、線維芽細胞、軟骨細胞、骨芽細胞等の間葉系細胞、大腸がん細胞(例えば、ヒト大腸がん細胞(HT29))、肝がん細胞等のがん細胞、心筋細胞、上皮細胞(例えば、ヒト歯肉上皮細胞)、リンパ管内皮細胞、神経細胞、樹状細胞、肝細胞、接着性細胞(例えば、免疫細胞)、平滑筋細胞(例えば、大動脈平滑筋細胞(Aorta-SMC))、膵島細胞、角化細胞(例えば、ヒト表皮角化細胞)等が挙げられる。
生体組織からの脂肪由来幹細胞及び/又は成熟脂肪細胞の採取は、常法により行うことができるが、例えば、以下のように採取することができる。脂肪組織断片を洗浄及び切断し、コラゲナーゼで消化(例えば、37℃で1時間インキュベート)する。その後、消化された細胞をろ過等により回収し、遠心すると、成熟脂肪細胞を含む上層の黄色の油性層、脂肪幹細胞及び血球を含む下層のペレットに分離される。上層のみを回収することで成熟脂肪細胞を、ペレットのみを回収することでADSCをそれぞれ採取することができる。
本明細書において「血管内皮細胞」は、血管内腔の表面を構成する扁平状の細胞を意味する。血管内皮細胞としては、例えば、ヒト臍帯静脈由来血管内皮細胞(HUVEC)が挙げられる。脂肪幹細胞が血管内皮細胞に分化しているかは、例えば、血管内皮細胞のマーカーであるCD31、フォン・ウィルブランド因子(vWF)及びVE-カドヘリン等を用いた免疫染色やフローサイトメトリー解析等により確認することができる。
組織体における血管内皮細胞の含有率は、組織体における全細胞数に対して、例えば、5%以上、10%以上、15%以上、20%以上、25%以上、又は30%以上であってよく、95%以下、90%以下、80%以下又は75%以下であってよい。本発明による効果がより顕著になる観点から、脂肪由来幹細胞を少なくとも含む細胞、すなわちインキュベート前の細胞における血管内皮細胞の含有率は、インキュベート前の全細胞数に対して10%以下、5%以下、又は1以下であってよい。
組織体は、細胞間に血管網を有するものであってよい。細胞間に血管網が形成されていると、組織体を長期間維持できること、及び、組織体を哺乳類等に移植した際に生着しやすくなることが期待される。
「細胞間に血管網を有する」とは、生体組織と同様に、分岐した血管が細胞を取り囲むように細胞と細胞の間に延びた構造を有することを意味する。生体組織と同様の血管網が形成されているか否かについては、例えば、生体組織における血管の分岐数及び/又は血管の分岐間の長さ及び/又は血管の直径の多様性に基づき判断することができる。例えば、生体組織における血管の分岐数の平均値に対する、組織体における血管の分岐数の平均値が、80%以上150%以下、85%以上130%以下、又は90%以上120%以下である場合に、生体組織における血管の分岐数と類似していると判断してもよい。また、例えば、組織体における血管の分岐数の平均値が、2.5以上4.5以下、又は3.0以上4.2以下である場合に、生体組織における血管の分岐数と類似していると判断してもよい。例えば、生体組織における血管の分岐間の長さの平均値に対する、組織体における血管の分岐間の長さの平均値が、80%以上150%以下、85%以上130%以下、及び90%以上120%以下である場合に、生体組織における血管の分岐間の長さと類似していると判断してもよい。生体組織においては、太い血管及び細い血管の両方が観察される。そこで、例えば、生体組織と同様に直径の太いもの(例えば、10μm以上25μm未満)と細いもの(例えば、0μm超10μm未満)の両方が観察された場合には、生体組織における血管の直径と同様の多様性を有すると判断してもよい。また、例えば、0μm超25μm未満に血管直径全体の60%以上、70%以上又は80%以上が分布している場合に、生体組織における血管の直径と同様の多様性を有すると判断してもよい。血管内皮細胞を含む組織体は、血管内皮細胞間に血管網を有することが好ましい。その場合、血管網を有するだけでなく、血管に取り囲まれる血管内皮細胞も生体組織と近いことが好ましい。上記生体組織及び組織体の比較に際しては、同じ条件(例えば、一定体積当たり、画像解析であれば一定面積当たり、一定サンプル当たり等)にて生体組織と組織体を比較する。
組織体は血管内皮細胞以外の細胞を更に含んでいてもよい。血管内皮細胞以外の細胞としては、例えば、成熟脂肪細胞、線維芽細胞、軟骨細胞、骨芽細胞等の間葉系細胞、大腸がん細胞(例えば、ヒト大腸がん細胞(HT29))、肝がん細胞等のがん細胞、心筋細胞、上皮細胞(例えば、ヒト歯肉上皮細胞)、リンパ管内皮細胞、神経細胞、樹状細胞、肝細胞、接着性細胞(例えば、免疫細胞)、平滑筋細胞(例えば、大動脈平滑筋細胞(Aorta-SMC))、膵島細胞、角化細胞(例えば、ヒト表皮角化細胞)等が挙げられる。ただし、本発明は、脂肪由来幹細胞を血管内皮細胞の分化を促進する効果を奏するため、本発明による効果がより顕著になることから、インキュベート後の組織体において、脂肪由来幹細胞が組織体における全細胞数に対して10%以下であることが好ましく、5%以下であることがより好ましく、脂肪由来幹細胞を含まないことがさらに好ましい。
脂肪細胞の成熟度を示す指標として、脂肪滴の大きさを用いることができる。脂肪滴とは、トリグリセリド(中性脂肪)、コレストロール等の脂質を貯蔵する細胞内小器官であり、上記脂質類がリン脂質の1重膜で覆われることで液滴様の形状を有している。また上記リン脂質の表面には脂肪組織特有のタンパク質(ペリリピン等)の発現が見られる。成熟した脂肪細胞の脂肪滴の大きさにはばらつきがあるが、例えば、脂肪滴の大きさの平均値が20μm以上である場合には、脂肪細胞がある程度成熟している、すなわち、成熟脂肪細胞であるとすることができる。なお、本明細書において「脂肪細胞」とは、脂肪由来幹細胞を除くすべての脂肪細胞を意味し、成熟脂肪細胞、及び脂肪由来幹細胞に含まれない脂肪細胞が含まれる。
上記組織体の厚さは10μm以上であることが好ましく、100μm以上であることがより好ましく、1000μm以上であることが更により好ましい。このような組織体は、生体組織により近い構造であり、実験動物の代替品、及び移植材料として好適なものとなる。組織体の厚さの上限は、特に制限されないが、例えば、10mm以下であってもよいし、3mm以下であってもよいし、2mm以下であってもよいし、1.5mm以下であってもよいし、1mm以下であってもよい。
ここで、「組織体の厚さ」とは、組織体がシート状、又は直方体状である場合、主面に垂直な方向における両端の距離を意味する。上記主面に凹凸がある場合、厚さは上記主面の最も薄い部分における距離を意味する。また、組織体が球体状又は略球体状である場合、その直径を意味する。さらにまた、組織体が楕円体状又は略楕円体状である場合、その短径を意味する。組織体が略球体状又は略楕円体状であって表面に凹凸がある場合、厚さは、組織体の重心を通る直線と上記表面とが交差する2点間の距離であって最短の距離を意味する。
本実施形態の方法は、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む。TGFβタイプI受容体阻害剤の存在下でインキュベートすることで、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化が促進される。本実施形態の方法においては、脂肪由来幹細胞が組織体に含まれた状態で分化を促進させること、又は脂肪由来幹細胞が組織体に含まれるように組織体が形成される状況において分化を促進することにより、最終的に血管内皮細胞を含む組織体を作製することができる。したがって、組織体の製造は、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含んでいればよく、三次元培養であっても、二次元培養であってもよい。組織体は、例えば、培地中でヒト由来の脂肪由来幹細胞を少なくとも含む細胞をインキュベートすることのみで得られたものであってもよく、足場がない状態で当該細胞を培地中に浮遊させながらインキュベートする浮遊培養により得られたものであってもよく、ゲル及びシート等の足場に当該細胞を付着させてインキュベートする付着型培養で得られたものであってもよく、後述するように、水性媒体中で当該細胞を細胞外マトリックスとともにインキュベートすることで得られたものであってもよい。
脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることは、細胞のみがTGFβタイプI受容体阻害剤存在下でインキュベートされてもよく、細胞及び細胞外マトリックス成分が混合された状態でTGFβタイプI受容体阻害剤存在下でインキュベートされてもよく、細胞及び細胞外マトリックス成分を含む組織体が形成された状態でTGFβタイプI受容体阻害剤存在下でインキュベートされてもよい。
TGFβタイプI受容体阻害剤としては、例えば、TGFβR1キナーゼである、ALK5、ALK2、ALK4及びALK7の阻害剤が挙げられる。TGFβタイプI受容体阻害剤は、TGFβタイプI受容体を阻害する作用を有するものであれば、特に制限されない。ALK5、ALK4及びALK7阻害剤としては、例えば、4-[4-(1,3-ベンゾジオキシオール-5-イル)-5-(2-ピリジニル)-1H-イミダゾール-2-イル]ベンズアミドが挙げられ、ALK5阻害剤としては、例えば、2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、2-(3-(6-メチルピリジン-2-イル)-1H-ピラゾール-4-イル)-1,5-ナフチリジン,塩酸塩、N-(2-フルオロフェニル)-4-([1,2,4]トリアゾロ[1,5-a]ピリジン-6-イル)-5-(6-メチル-2-ピリジル)-1H-イミダゾール-2-メタンアミン等が挙げられる。
TGFβタイプI受容体阻害剤の存在下でインキュベートするとは、例えば、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤が含まれる培地中でインキュベート(培養)することであってよい。
培地は特に制限はなく、培養する細胞の種類に応じて好適な培地を選択できる。培地は、固体培地であっても、液体培地であってもよいが、液体培地であることが好ましい。培地としては、例えば、Eagle’s MEM培地、DMEM、血管内皮細胞専用培地(EGM-2)、Modified Eagle培地(MEM)、Minimum Essential培地、RPMI、KBM培地、及びGlutaMax培地等が挙げられる。また、液体培地は、フィブリンゲル、ハイドロゲル、マトリゲル、コラーゲンゲル及びゼラチンゲルのようにゲル状のものを用いてもよい。ゲル状の液体培地に細胞を添加してもよく、細胞を含む液体培地をゲル化させて用いてもよい。培地は、血清を添加した培地であってもよいし、無血清培地であってもよい。培地は、二種類の培地を混合した混合培地であってもよい。
TGFβタイプI受容体阻害剤の量は、各脂肪由来幹細胞が当該阻害剤に接するのに十分な量であればよく、例えば、脂肪由来幹細胞1×106cellsに対して、2.0×10-11mol~2.0×10-8mol、又は1.0×10-9mol~2.0×10-8molであってよく、例えば、脂肪由来幹細胞5×106cellsに対して、1.0×10-10mol~1.0×10-7mol、又は5.0×10-9mol~1.0×10-7molであってよい。また、TGFβタイプI受容体阻害剤は、例えば、脂肪由来幹細胞1×106cellsに対して、6.0ng~6.0μg、又は300ng~6.0μgであってよく、例えば、脂肪由来幹細胞5×106cellsに対して、30ng~30μg、又は1500ng~30μgであってよい。
ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤が含まれる培地中でインキュベートする場合、培地におけるTGFβタイプI受容体阻害剤の含有量は、例えば、0.1μM以上100μM以下、0.5μM以上50μM以下、1μM以上10μM以下、又は2μM以上8μM以下であってもよく、0.5μM以上、0.7以上、1μM以上、2μM以上、3μM以上、4μM以上、5μM以上であってもよく、20μM以下、15μM以下、12μM以下、10μM以下、8μM以下又は7μM以下であってもよい。
インキュベート前の培地中の細胞密度は、目的とする組織体の形状、厚さ、培養器のサイズ等に応じて適宜決定できる。例えば、培地中の細胞密度は、1~108cells/mLであってよく、103~107cells/mLであってよい。
インキュベートは、特に制限はなく、培養する細胞の種類に応じて好適な条件で行うことができる。例えば、インキュベートの温度は20℃~40℃であってもよく、30℃~37℃であってもよい。培地のpHは、6~8であってもよく、7.2~7.4であってもよい。インキュベートの時間は、24時間以上336時間以下であってもよく、72時間以上336時間以下であってもよく、96時間以上288時間以下であってもよい。TGFβタイプI受容体阻害剤を添加してからのインキュベートの時間は、24時間以上336時間以下であってもよく、72時間以上336時間以下であってもよく、96時間以上288時間以下であってもよい。
細胞の培養に用いられる培養器(支持体)は、特に制限されず、例えば、ウェルインサート、低接着プレート、U字やV字等の底面形状を有するプレートであってよい。上記細胞を支持体と接着させたまま培養してもよく、上記細胞を支持体と接着させずに培養してもよく、培養の途中で支持体から引き離して培養してもよい。上記細胞を支持体と接着させずに培養する場合や、培養の途中で支持体から引き離して培養する場合には、細胞の支持体への接着を阻害するU字やV字等の底面形状を有するプレートや、低吸着プレートを用いることが好ましい。
本実施形態の方法は、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤が含まれる培地中でインキュベート(培養)することである場合、培地には血管内皮細胞増殖因子(VEGF)が含まれていてもよい。その際、例えば、予めVEGFとTGFβタイプI受容体阻害剤を含む培地を使用してもよく、VEGFを含む培地にTGFβタイプI受容体阻害剤を添加してもよく、TGFβタイプI受容体阻害剤を含む培地にVEGFを添加してもよい。
ヒト由来の脂肪由来幹細胞を少なくとも含む細胞を、TGFβタイプI受容体阻害剤及びVEGFを含む培地中でインキュベートすることで、より脂肪幹細胞から血管内皮細胞への分化が促進される。また、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞を、TGFβタイプI受容体阻害剤及びVEGFを含む培地中でインキュベートすることで、血管新生を促進することもできるため、血管網を有する組織体をより簡便に製造できる。
ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をVEGFが含まれる培地中でインキュベートする場合、培地におけるVEGFの含有量は、例えば、0.5ng/mL以上100ng/mL以下、1ng/mL以上85ng/mL以下、6ng/mL以上80ng/mL以下、10ng/mL以上70ng/mL以下、又は20ng/mL以上50ng/mL以下であってもよく、0.5ng/mL以上、1ng/mL以上、5ng/mL以上、6ng/mL以上、10ng/mL以上、15ng/mL以上、20ng/mL以上であってもよく、100ng/mL以下、90ng/mL以下、80ng/mL以下、70ng/mL以下、60ng/mL以下又は50ng/mL以下であってもよい。
本実施形態の方法は、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞を断片化細胞外マトリックス成分とともにTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含んでもよい。断片化細胞外マトリックス成分とともにインキュベートすることで、断片化細胞外マトリックス成分を介して細胞が三次元的に配置された三次元組織体を得ることができる。断片化細胞外マトリックス成分が上記細胞同士の隙間に配置されている三次元組織体であることが好ましい。細胞同士における細胞は、同種細胞であってもよく、異種細胞であってもよい。TGFβタイプI受容体阻害剤を含む培地を用いる場合において、断片化細胞外マトリックス成分は、TGFβタイプI受容体阻害剤と同時に培地に添加してもよく、TGFβタイプI受容体阻害剤より先に培地に添加してもよく、TGFβタイプI受容体阻害剤より後に培地に添加してもよい。
断片化細胞外マトリックス成分は、細胞外マトリックス成分を断片化して得ることができる。本明細書において「細胞外マトリックス成分」とは、複数の細胞外マトリックス分子によって形成されている細胞外マトリックス分子の集合体である。細胞外マトリックスとは、生物において細胞の外に存在する物質を意味する。細胞外マトリックスとしては、細胞の生育及び細胞集合体の形成に悪影響を及ぼさない限り、任意の物質を用いることができる。具体例としては、コラーゲン、エラスチン、プロテオグリカン、フィブロネクチン、ヒアルロン酸、ラミニン、ビトロネクチン、テネイシン、エンタクチン及びフィブリリン等が挙げられるが、これらに限定されない。細胞外マトリックス成分は、これらの1種単独で用いてもよく、組み合わせて用いてもよい。細胞外マトリックス成分は、例えば、コラーゲン成分を含んでいてよく、コラーゲン成分であってもよい。本実施形態における細胞外マトリックス成分は、動物細胞の外に存在する物質、すなわち動物の細胞外マトリックス成分であることが好ましい。
細胞外マトリックス分子は、細胞の生育及び細胞集合体の形成に悪影響を及ぼさない限り、上述の細胞外マトリックス分子の改変体及びバリアントであってもよく、化学合成ペプチド等のポリペプチドであってもよい。細胞外マトリックス分子は、コラーゲンに特徴的なGly-X-Yで表される配列の繰り返しを有するものであってよい。ここで、Glyはグリシン残基を表し、X及びYはそれぞれ独立に任意のアミノ酸残基を表す。複数のGly-X-Yは、それぞれ同一であっても異なっていてもよい。Gly-X-Yで示される配列の繰り返しを有することによって、分子鎖の配置への束縛が少なくなるため、例えば、細胞培養の際の足場材料としての機能がより一層優れたものとなる。Gly-X-Yで示される配列の繰り返しを有する細胞外マトリックス分子において、Gly-X-Yで示される配列の割合は、全アミノ酸配列のうち、80%以上であってよく、好ましくは95%以上である。また、細胞外マトリックス分子は、RGD配列を有するポリペプチドであってもよい。RGD配列とは、Arg-Gly-Asp(アルギニン残基-グリシン残基-アスパラギン酸残基)で表される配列をいう。RGD配列を有することによって、細胞接着がより一層促進されるため、例えば、細胞培養の際の足場材料としてより一層好適なものとなる。Gly-X-Yで表される配列と、RGD配列とを含む細胞外マトリックス分子としては、コラーゲン、フィブロネクチン、ビトロネクチン、ラミニン、カドヘリン等が挙げられる。
コラーゲンとしては、例えば、線維性コラーゲン及び非線維性コラーゲンが挙げられる。線維性コラーゲンとは、コラーゲン線維の主成分となるコラーゲンを意味し、具体的には、I型コラーゲン、II型コラーゲン、III型コラーゲン等が挙げられる。非線維性コラーゲンとしては、例えば、IV型コラーゲンが挙げられる。
プロテオグリカンとして、コンドロイチン硫酸プロテオグリカン、ヘパラン硫酸プロテオグリカン、ケラタン硫酸プロテオグリカン、デルマタン硫酸プロテオグリカンが挙げられるが、これらに限定されない。
細胞外マトリックス成分は、本発明による効果がより顕著になることから、コラーゲン、ラミニン及びフィブロネクチンからなる群より選択される少なくとも1種を含んでいてよく、コラーゲンを含むことが好ましい。コラーゲンは好ましくは繊維性コラーゲンであり、より好ましくはI型コラーゲンである。線維性コラーゲンは、市販されているコラーゲンを用いてもよく、その具体例としては、日本ハム株式会社製のブタ皮膚由来I型コラーゲンが挙げられる。
細胞外マトリックス成分は、動物由来の細胞外マトリックス成分であってよい。細胞外マトリックス成分の由来となる動物種として、例えば、ヒト、ブタ、ウシ等が挙げられるが、これらに限定されない。細胞外マトリックス成分は、一種類の動物に由来する成分を用いてもよいし、複数種の動物に由来する成分を併用して用いてもよい。
「断片化」とは、細胞外マトリックス分子の集合体をより小さなサイズにすることを意味する。断片化は、細胞外マトリックス分子内の結合を切断する条件で行われてもよいし、細胞外マトリックス分子内の結合を切断しない条件で行われてもよい。物理的な力の印加によって断片化された細胞外マトリックスは、酵素処理とは異なり、通常、分子構造は断片化する前とは変化しない(分子構造は維持されている)。断片化細胞外マトリックス成分は、上述の細胞外マトリックス成分を物理的な力の印加により解繊した成分である、解繊された細胞外マトリックス成分(解繊細胞外マトリックス成分)を含んでいてよい。解繊は、断片化の一態様であり、例えば、細胞外マトリックス分子内の結合を切断しない条件で行われるものである。
細胞外マトリックス成分を断片化する方法としては、特に制限されない。細胞外マトリックス成分を解繊する方法としては、例えば、超音波式ホモジナイザー、撹拌式ホモジナイザー、及び高圧式ホモジナイザー等の物理的な力の印加によって細胞外マトリックス成分を解繊してもよい。撹拌式ホモジナイザーを用いる場合、細胞外マトリックス成分をそのままホモジナイズしてもよいし、生理食塩水等の水性媒体中でホモジナイズしてもよい。また、ホモジナイズする時間、回数等を調整することでミリメートルサイズ、ナノメートルサイズの解繊細胞外マトリックス成分を得ることも可能である。解繊細胞外マトリックス成分は、凍結融解を繰り返すことで解繊することにより得ることもできる。
断片化細胞外マトリックス成分は、解繊された細胞外マトリックス成分を少なくとも一部に含んでいてよい。また、断片化細胞外マトリックス成分は、解繊された細胞外マトリックス成分のみからなっていてもよい。すなわち、断片化細胞外マトリックス成分は、解繊された細胞外マトリックス成分であってよい。解繊された細胞外マトリックス成分は、解繊されたコラーゲン成分(解繊コラーゲン成分)を含むことが好ましい。解繊コラーゲン成分は、コラーゲンに由来する三重らせん構造を維持していることが好ましい。解繊コラーゲン成分は、コラーゲンに由来する三重らせん構造を部分的に維持している成分であってよい。
断片化細胞外マトリックス成分の形状としては、例えば、繊維状が挙げられる。繊維状とは、糸状のコラーゲン成分で構成される形状、又は糸状の細胞外マトリックス成分が分子間で架橋して構成される形状を意味する。断片化細胞外マトリックス成分の少なくとも一部は、繊維状であってよい。線維状の細胞外マトリックス成分には、複数の糸状細胞外マトリックス分子が集合して形成された細い糸状物(細線維)、細線維が更に集合して形成される糸状物、これらの糸状物を解繊したもの等が含まれる。線維状の細胞外マトリックス成分ではRGD配列が破壊されることなく保存されている。
断片化細胞外マトリックス成分の平均長は、100nm以上400μm以下であってよく、100nm以上200μm以下であってよい。一実施形態において、断片化細胞外マトリックス成分の平均長は、5μm以上400μm以下であってよく、10μm以上400μm以下であってよく、22μm以上400μm以下であってよく、100μm以上400μm以下であってよい。他の実施形態において、断片化細胞外マトリックス成分の平均長は、再分散性がより一層優れたものとなる観点から、100μm以下であってよく、50μm以下であってよく、30μm以下であってよく、15μm以下であってよく、10μm以下であってよく、1μm以下であってよく、100nm以上であってよい。断片化細胞外マトリックス成分全体のうち、大部分の断片化細胞外マトリックス成分の平均長が上記数値範囲内であることが好ましい。具体的には、断片化細胞外マトリックス成分全体のうち95%の断片化細胞外マトリックス成分の平均長が上記数値範囲内であることが好ましい。断片化細胞外マトリックス成分は、平均長が上記範囲内である断片化コラーゲン成分であることが好ましく、平均長が上記範囲内である解繊コラーゲン成分であることがより好ましい。
断片化細胞外マトリックス成分の平均径は、10nm以上30μm以下であってよく、30nm以上30μm以下であってよく、50nm以上30μm以下であってよく、100nm以上30μm以下であってよく、1μm以上30μm以下であってよく、2μm以上30μm以下であってよく、3μm以上30μm以下であってよく、4μm以上30μm以下であってよく、5μm以上30μm以下であってよい。断片化細胞外マトリックス成分は、平均径が上記範囲内である断片化コラーゲン成分であることが好ましく、平均径が上記範囲内である解繊コラーゲン成分であることがより好ましい。
断片化細胞外マトリックス成分の平均長及び平均径は、光学顕微鏡によって個々の断片化細胞外マトリックス成分を測定し、画像解析することによって求めることが可能である。本明細書において、「平均長」は、測定した試料の長手方向の長さの平均値を意味し、「平均径」は、測定した試料の長手方向に直交する方向の長さの平均値を意味する。
断片化細胞外マトリックス成分は、例えば、断片化コラーゲン成分を含んでいてよく、断片化コラーゲン成分からなっていてよい。「断片化コラーゲン成分」とは、線維性コラーゲン成分等のコラーゲン成分を断片化したものであって、三重らせん構造を維持しているものを意味する。断片化コラーゲン成分の平均長は、100nm~200μmであることが好ましく、22μm~200μmであることがより好ましく、100μm~200μmであることがさらにより好ましい。断片化コラーゲン成分の平均径は、50nm~30μmであることが好ましく、4μm~30μmであることがより好ましく、20μm~30μmであることがさらにより好ましい。
断片化細胞外マトリックス成分の少なくとも一部は分子間又は分子内で架橋されていてよい。細胞外マトリックス成分は、細胞外マトリックス成分を構成する細胞外マトリックス分子の分子内又は分子間で架橋されていてよい。
架橋する方法としては、例えば、熱、紫外線、放射線等の印加による物理架橋、架橋剤、酵素反応等による化学架橋等による方法が挙げられるが、その方法は特に限定されない。細胞の生育を妨げない観点からは、物理架橋が好ましい。架橋(物理架橋及び化学架橋)は、共有結合を介した架橋であってよい。
細胞外マトリックス成分がコラーゲン成分を含む場合、架橋は、コラーゲン分子(三重らせん構造)の間で形成されていてもよく、コラーゲン分子によって形成されたコラーゲン細繊維の間で形成されていてもよい。架橋は、熱による架橋(熱架橋)であってよい。熱架橋は、例えば、真空ポンプを使って減圧下で、加熱処理を行うことにより実施することができる。コラーゲン成分の熱架橋を行う場合、細胞外マトリックス成分は、コラーゲン分子のアミノ基が、同一又は他のコラーゲン分子のカルボキシ基とペプチド結合(-NH-CO-)を形成することにより、架橋されていてよい。
細胞外マトリックス成分は架橋剤を使用することによっても、架橋させることができる。架橋剤は、例えば、カルボキシル基とアミノ基を架橋可能なもの、又はアミノ基同士を架橋可能なものであってよい。架橋剤としては、例えば、アルデヒド系、カルボジイミド系、エポキシド系及びイミダゾール系架橋剤が経済性、安全性及び操作性の観点から好ましく、具体的には、グルタルアルデヒド、1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミド・塩酸塩、1-シクロヘキシル-3-(2-モルホリニル-4-エチル)カルボジイミド・スルホン酸塩等の水溶性カルボジイミドを挙げることができる。
架橋度の定量は、細胞外マトリックス成分の種類、架橋する手段等に応じて、適宜選択することができる。架橋度は、1%以上、2%以上、4%以上、8%以上、又は12%以上であってよく、30%以下、20%以下、又は15%以下であってもよい。架橋度が上記範囲にあることにより、細胞外マトリックス分子が適度に分散することができ、また、乾燥保存後の再分散性が良好である。
細胞外マトリックス成分中のアミノ基が架橋に使用される場合、架橋度は、TNBS(トリニトロベンゼンスルホン酸)法に基づき定量することが可能である。TNBS法による架橋度が、上述の範囲内であってもよい。TNBS法による架橋度は、細胞外マトリックスが有するアミノ基のうち架橋に使われているアミノ基の割合である。細胞外マトリックス成分がコラーゲン成分を含む場合、TNBS法により測定される架橋度が上記範囲内であることが好ましい。
架橋度は、カルボキシル基を定量することにより、算出してもよい。例えば、水に不溶性の細胞外マトリックス成分の場合、TBO(トルイジンブルーO)法により定量してもよい。TBO法による架橋度が、上述した範囲内であってもよい。
三次元組織体における細胞外マトリックス成分含有率は、上記三次元組織体(乾燥重量)を基準として0.01~90質量%であってよく、10~90質量%であることが好ましく、10~80質量%であることが好ましく、10~70質量%であることが好ましく、10~60質量%であることが好ましく、1~50質量%であることが好ましく、10~50質量%であることが好ましく、10~30質量%であることがより好ましく、20~30質量%であることがより好ましい。
ここで、「三次元組織体における細胞外マトリックス成分」とは、三次元組織体を構成する細胞外マトリックス成分を意味し、内因性細胞外マトリックス成分に由来していてもよく、外因性細胞外マトリックス成分に由来していてもよい。
「内因性の細胞外マトリックス成分」とは、細胞外マトリックス産生細胞が産生する細胞外マトリックス成分を意味する。細胞外マトリックス産生細胞としては、例えば、上述した線維芽細胞、軟骨細胞、骨芽細胞等の間葉系細胞が挙げられる。内因性細胞外マトリックス成分は、線維性であってもよいし、非線維性であってもよい。
「外因性の細胞外マトリックス成分」とは、外部から供給される細胞外マトリックス成分を意味する。外因性の細胞外マトリックス成分は、由来となる動物種が内因性の細胞外マトリックス成分と同じであっても異なっていてもよい。由来となる動物種としては、例えば、ヒト、ブタ、ウシ等が挙げられる。また、外因性の細胞外マトリックス成分は、人工の細胞外マトリックス成分であってもよい。
細胞外マトリックス成分がコラーゲン成分である場合には、外因性の細胞外マトリックス成分は「外因性コラーゲン成分」とも称され、外部から供給されるコラーゲン成分を意味する「外因性コラーゲン成分」は、複数のコラーゲン分子によって形成されている、コラーゲン分子の集合体であり、具体的には、線維性コラーゲン、非線維性コラーゲン等が挙げられる。外因性コラーゲン成分は、線維性コラーゲンであることが好ましい。上記線維性コラーゲンは、コラーゲン線維の主成分となるコラーゲン成分を意味し、例えば、I型コラーゲン、II型コラーゲン、III型コラーゲンが挙げられる。上記線維性コラーゲンは、市販されているコラーゲンを用いてもよく、その具体例としては、日本ハム株式会社製のブタ皮膚由来I型コラーゲンが挙げられる。外因性の非線維性コラーゲンとしては、例えば、IV型コラーゲンが挙げられる。
外因性の細胞外マトリックス成分において、由来する動物種は、細胞とは異なっていてよい。また、細胞が、細胞外マトリックス産生細胞を含む場合、外因性の細胞外マトリックス成分において、由来する動物種は、細胞外マトリックス産生細胞とは異なっていてもよい。つまり、外因性の細胞外マトリックス成分は、異種細胞外マトリックス成分であってよい。
すなわち、三次元組織体が内因性細胞外マトリックス成分及び断片化細胞外マトリックス成分を含む場合、上記三次元組織体を構成する細胞外マトリックス成分含有率は、内因性細胞外マトリックス成分及び断片化細胞外マトリックス成分の合計量を意味する。上記細胞外マトリックス含有率は、得られた三次元組織体の体積、及び脱細胞化した三次元組織体の質量から算出することが可能である。
例えば、三次元組織体に含まれる細胞外マトリックス成分がコラーゲン成分である場合、三次元組織体におけるコラーゲン成分量を定量する方法としては、例えば、以下のようなヒドロキシプロリンを定量する方法が挙げられる。三次元組織体を溶解した溶解液に、塩酸(HCl)を混合し、高温で所定の時間インキュベートした後に室温に戻し、遠心分離した上澄みを所定の濃度に希釈することでサンプルを調製する。ヒドロキシプロリンスタンダード溶液をサンプルと同様に処理した後、段階的に希釈してスタンダードを調製する。サンプル及びスタンダードのそれぞれに対してヒドロキシプロリンアッセイバッファ及び検出試薬で所定の処理をし、570nmの吸光度を測定する。サンプルの吸光度をスタンダードと比較することでコラーゲン成分量を算出する。なお、三次元組織体を、高濃度の塩酸に直接懸濁して溶解した溶解液を遠心分離して上澄みを回収し、コラーゲン成分定量に用いてもよい。また、溶解させる三次元組織体は、培養液から回収したままの状態であってもよいし、回収後に乾燥処理を行い、液体成分を除去した状態で溶解させてもよい。但し、培養液から回収したままの状態の三次元組織体を溶解してコラーゲン成分定量を行う場合、三次元組織体が吸収している培地成分、及び実験手技の問題による培地の残りの影響で、三次元組織体重量の計測値がばらつくことが予想されるため、三次元組織体の重量及び単位重量あたりに占めるコラーゲン成分量を安定して計測する観点からは、乾燥後の重量を基準とすることが好ましい。
コラーゲン成分量を定量する方法として、より具体的には、例えば、以下のような方法が挙げられる。
(サンプルの調製)
凍結乾燥処理を行った三次元組織体の全量を6mol/L HClと混合し、ヒートブロックで95℃、20時間以上インキュベートした後、室温に戻す。13000gで10分遠心分離した後、サンプル溶液の上澄みを回収する。後述する測定において結果が検量線の範囲内に収まるように6mol/L HClで適宜希釈した後、200μLを100μLの超純水で希釈することでサンプルを調製する。サンプルは35μL用いる。
凍結乾燥処理を行った三次元組織体の全量を6mol/L HClと混合し、ヒートブロックで95℃、20時間以上インキュベートした後、室温に戻す。13000gで10分遠心分離した後、サンプル溶液の上澄みを回収する。後述する測定において結果が検量線の範囲内に収まるように6mol/L HClで適宜希釈した後、200μLを100μLの超純水で希釈することでサンプルを調製する。サンプルは35μL用いる。
(スタンダードの調製)
スクリューキャップチューブに125μLのスタンダード溶液(1200μg/mL in acetic acid)と、125μLの12mol/l HClを加え混合し、ヒートブロックで95℃、20時間インキュベートした後、室温に戻す。13000gで10分遠心分離した後、上澄みを超純水で希釈して300μg/mLのS1を作製し、S1を段階的に希釈してS2(200μg/mL)、S3(100μg/mL)、S4(50μg/mL)、S5(25μg/mL)、S6(12.5μg/mL)、S7(6.25μg/mL)を作製する。4mol/l HCl90μLのみのS8(0μg/mL)も準備する。

(アッセイ)
35μLのスタンダード及びサンプルをそれぞれプレート(QuickZyme Total Collagen Assayキット付属、QuickZyme Biosciences社)に加える。75μLのアッセイバッファ(上記キット付属)をそれぞれのウェルに加える。シールでプレートを閉じ、20分シェイキングしながら室温でインキュベートする。シールをはがし、75μLのdetection reagent (reagent A:B=30μL:45μL、上記キット付属)をそれぞれのウェルに加える。シールでプレートを閉じ、シェイキングで溶液を混合し、60℃で60分インキュベートする。氷上で十分に冷まし、シールをはがして570nmの吸光度を測定する。サンプルの吸光度をスタンダードと比較することでコラーゲン成分量を算出する。
35μLのスタンダード及びサンプルをそれぞれプレート(QuickZyme Total Collagen Assayキット付属、QuickZyme Biosciences社)に加える。75μLのアッセイバッファ(上記キット付属)をそれぞれのウェルに加える。シールでプレートを閉じ、20分シェイキングしながら室温でインキュベートする。シールをはがし、75μLのdetection reagent (reagent A:B=30μL:45μL、上記キット付属)をそれぞれのウェルに加える。シールでプレートを閉じ、シェイキングで溶液を混合し、60℃で60分インキュベートする。氷上で十分に冷まし、シールをはがして570nmの吸光度を測定する。サンプルの吸光度をスタンダードと比較することでコラーゲン成分量を算出する。
三次元組織体中に占めるコラーゲン成分を、その面積比又は体積比によって規定してもよい。「面積比又は体積比によって規定する」とは、例えば三次元組織体中のコラーゲン成分を既知の染色手法(例えば、抗コラーゲン抗体を用いた免疫染色、又はマッソントリクローム染色)等で他の組織構成物と区別可能な状態にした上で、肉眼観察、各種顕微鏡及び画像解析ソフト等を用いて、三次元組織体全体に占めるコラーゲン成分の存在領域の比率を算出することを意味する。面積比で規定する場合、三次元組織体中の如何なる断面もしくは表面によって面積比を規定するかは限定されないが、例えば三次元組織体が球状体等である場合には、その略中心部を通る断面図によって規定してもよい。
例えば、三次元組織体中のコラーゲン成分を面積比によって規定する場合、その面積の割合は、上記三次元組織体の全体の面積を基準として0.01~99%であり、1~99%であることが好ましく、5~90%であることが好ましく、7~90%であることが好ましく、20~90%であることが好ましく、50~90%であることがより好ましい。「三次元組織体におけるコラーゲン成分」については、上述したとおりである。三次元組織体を構成するコラーゲン成分の面積の割合は、内因性コラーゲン成分及び外因性コラーゲン成分を合わせた面積の割合を意味する。コラーゲン成分の面積の割合は、例えば、得られた三次元組織体をマッソントリクロームで染色し、三次元組織体の略中心部を通る断面の全体の面積に対する、青く染色したコラーゲン成分の面積の割合として算出することが可能である。
三次元組織体は、トリプシンの濃度0.25%、温度37℃、pH7.4、反応時間15分でトリプシン処理を行った後の残存率が70%以上であることが好ましく、80%以上であることがより好ましく、90%以上であることが更により好ましい。このような三次元組織体は、培養中又は培養後において酵素による分解が起きにくく、安定である。上記残存率は、例えば、トリプシン処理の前後における三次元組織体の質量から算出できる。
上記三次元組織体は、コラゲナーゼの濃度0.25%、温度37℃、pH7.4、反応時間15分でコラゲナーゼ処理を行った後の残存率が70%以上であってもよく、80%以上であることがより好ましく、90%以上であることが更により好ましい。このような三次元組織体は、培養中又は培養後における酵素による分解が起きにくく、安定である。
ヒト由来の脂肪由来幹細胞を少なくとも含む細胞を断片化細胞外マトリックス成分とともにTGFβタイプI受容体阻害剤の存在下でインキュベートするとは、例えば、細胞をTGFβタイプI受容体阻害剤及び断片化細胞外マトリックス成分が含まれる培地中でインキュベート(培養)することであってよい。その際、例えば、予め断片化細胞外マトリックス成分とTGFβタイプI受容体阻害剤を含む培地を使用してもよく、断片化細胞外マトリックス成分を含む培地にTGFβタイプI受容体阻害剤を添加してもよく、TGFβタイプI受容体阻害剤を含む培地に断片化細胞外マトリックス成分を添加してもよい。
インキュベート前に、水性媒体中において上記細胞と上記断片化細胞外マトリックス成分とを接触させる工程をさらに含んでいてもよい。この場合、TGFβタイプI受容体阻害剤は、水性媒体中に含まれていてもよく、インキュベート前に添加してもよく、インキュベート中に添加してもよい。水性媒体は、インキュベート時と同じであってもよく、異なっていてもよい。水性媒体中において上記細胞と上記断片化細胞外マトリックス成分とを接触させてインキュベートすることで、断片化細胞外マトリックス成分が上記細胞同士の隙間に配置されており、細胞と断片化細胞外マトリックス成分が三次元組織体全体に渡って三次元的に均一に分布した三次元組織体を製造することができる。例えば、水性媒体中において上記細胞と上記断片化細胞外マトリックス成分とを接触させてインキュベートした後に、TGFβタイプI受容体阻害剤の存在下でさらにインキュベートしてもよい。水性媒体中において上記細胞と上記断片化細胞外マトリックス成分とを接触させてインキュベートする時間は、例えば、12時間~72時間であってもよい。
「水性媒体」とは、水を必須構成成分とする液体を意味する。水性媒体としては、断片化細胞外マトリックス成分が安定に存在できるものであれば、特に制限はない。例えば、水性媒体として、リン酸緩衝生理食塩水(PBS)等の生理食塩水、Dulbecco’s Modified Eagle培地(DMEM)、血管内皮細胞専用培地(EGM-2)等の液体培地が挙げられるがこれに制限されない。
水性媒体のpHは細胞の生育及び細胞集合体の形成に悪影響を及ぼさない範囲が好ましい。水性媒体のpHは、細胞に投入した際の細胞への負荷を軽減する観点から、例えば、7.0以上であってよく、8.0以下であってよい。具体的には、水性媒体のpHは、7.0、7.1、7.2、7.3、7.4、7.5、7.6、7.7、7.8、7.9、又は8.0であってよい。水性媒体は、上記pHの範囲において緩衝能を有することが好ましく、より好ましくは液体培地である。液体培地は特に制限はなく、培養する細胞の種類に応じて好適な培地を選択できる。当該培地としては、例えば、Eagle’s MEM培地、DMEM、EGM-2、Modified Eagle培地(MEM)、Minimum Essential培地、RPMI、及びGlutaMax培地等が挙げられる。培地は、血清を添加した培地であってもよいし、無血清培地であってもよい。更に、液体培地は二種類以上の培地を混合した混合培地であってもよい。
断片化細胞外マトリックス成分は、水性媒体に分散させることにより、水性媒体中で細胞と接触しやすくなり、三次元組織体の形成を促進し得る。接触工程としては、断片化細胞外マトリックス成分を含有する水性媒体と細胞を含有する水性媒体とを混合する方法、断片化細胞外マトリックス成分を含有する水性媒体に細胞を添加する方法、細胞を含む培養液に細胞外マトリックス成分を含有する水性媒体を加える方法、細胞外マトリックス細胞外マトリックス成分を含有する水性媒体に細胞を加える方法、予め用意した水性媒体に、細胞外マトリックス成分及び細胞をそれぞれ加える方法等の方法が挙げられるが、これらに限られるものではない。
接触工程における断片化細胞外マトリックス成分の濃度は、目的とする三次元組織体の形状、厚さ、培養器のサイズ等に応じて適宜決定できる。例えば、接触工程における水性媒体中の断片化細胞外マトリックス成分の濃度は、0.1~90質量%であってもよいし、1~30質量%であってもよい。
接触工程における断片化細胞外マトリックス成分の量は、例えば、1.0×106cellsの細胞に対して、0.1~100mg、0.5~50mg、0.8~25mg、1.0~10mg、1.0~5.0mg、1.0~2.0mg、又は1.0~1.8mgであってよく、0.7mg以上、1.1mg以上、1.2mg以上、1.3mg以上又は1.4mg以上であってよく、7.0mg以下、3.0mg以下、2.3mg以下、1.8mg以下、1.7mg以下、1.6mg以下又は1.5mg以下であってもよい。
接触工程において、断片化細胞外マトリックス成分と細胞との質量比(断片化細胞外マトリックス成分/細胞)は、1/1~1000/1であることが好ましく、9/1~900/1であることがより好ましく、10/1~500/1であることがさらに好ましい。
また、接触工程における断片化細胞外マトリックス成分の量は、培地全体の重量に対して、0.1重量%~10重量%、0.5重量%~8重量%、又は1~5重量%であってもよい。
三次元組織体の製造方法の一実施形態は、接触工程、又は接触工程後かつインキュベート前に、フィブリノゲン及びトロンビンを同時に又は別々に混合することを含んでいてよい。フィブリノゲン及びトロンビンを混合することによって、これらが反応してフィブリンが形成される。フィブリノゲンの含有量は、例えば、培地に対して、1~10mg/mLであってよく、2~8mg/mLであってよく、5~6mg/mLであってもよい。
接触工程後かつインキュベート前に、水性媒体中における断片化細胞外マトリックス成分と細胞とを共に沈降させる工程を更に含んでもよい。このような工程を行うことで、三次元組織体における断片化細胞外マトリックス成分及び細胞の分布が、より均一になる。具体的な方法としては、特に制限はないが、例えば、断片化細胞外マトリックス成分と細胞とを含む培養液を遠心操作する方法が挙げられる。
上述したインキュベート前の培地中の細胞密度は、接触工程における水性媒体中の細胞密度と同じであってもよい。
本発明は、一実施形態として、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、脂肪由来幹細胞の血管内皮細胞への分化促進方法も提供する。上述したように、TGFβタイプI受容体阻害剤の存在下で脂肪由来幹細胞をインキュベートすることにより、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化を促進することができる。本実施形態の方法は、ヒト由来の脂肪由来幹細胞が組織体に含まれた状態で分化を促進させること、又はヒト由来の脂肪由来幹細胞が組織体に含まれるように組織体が形成される状況において分化を促進することを含み、それにより最終的に血管内皮細胞を含む組織体を作製することができる。
本実施形態の方法は、少なくとも一部のヒト由来の脂肪由来幹細胞がTGFβタイプI受容体阻害剤と接触することで分化が促進されればよいため、上述した三次元組織体の製造における三次元培養だけでなく、他の三次元培養、二次元培養においても適用することができる。
また、本実施形態の方法は、上述した三次元組織体の製造方法におけるヒト由来の脂肪由来幹細胞の血管内皮細胞への分化促進方法であってもよい。
上述のとおり、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞を、TGFβタイプI受容体阻害剤及びVEGFを含む培地中でインキュベートすることで、脂肪幹細胞から血管内皮細胞への分化、及び血管新生を促進することができる。すなわち、本実施形態の方法は、ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤及びVFGFが含まれる培地中でインキュベート(培養)することを含む、血管新生促進方法とすることもできる。血管新生促進とは、血管網形成促進であってもよい。
組織体(三次元組織体を含む)、細胞、インキュベート、断片化細胞外マトリックス成分及び各工程等、本実施形態の方法における具体的な態様等は、上述した具体的な態様等を制限なく適用できる。例えば、本実施形態の方法においても、インキュベート前に、水性媒体中において上記細胞と上記断片化細胞外マトリックス成分とを接触させる工程を含んでもよい。
以下、本発明を実施例に基づいてより具体的に説明する。ただし、本発明は、以下の実施例に限定されるものではない。
組織体の作製において用いた細胞、試薬及び作成方法等は以下のとおりである。
(細胞及びコラーゲン)

・50mg/mL フィブリノゲンストック溶液:フィブリノゲン 50mgをエッペンドルフチューブに秤とり、DMEM(0%FBS、1%抗生物質)1mLをすぐに加える。手動でチューブを振って混合した後、37℃のウォーターバスに3~5分間置き、孔径0.2μmのフィルターでろ過し、エッペンドルフチューブに等量分注したものを使用。
・202U/mL トロンビンストック溶液:トロンビン 202Uをエッペンドルフチューブに秤とり、DMEM(0%FBS、1%抗生物質)1mLをすぐに加え、37℃のウォーターバスに3~5分間置いて溶解させる。その後、孔径0.2μmのフィルターでろ過し、エッペンドルフチューブに等量分注したものを使用。

・50mg/mL フィブリノゲンストック溶液:フィブリノゲン 50mgをエッペンドルフチューブに秤とり、DMEM(0%FBS、1%抗生物質)1mLをすぐに加える。手動でチューブを振って混合した後、37℃のウォーターバスに3~5分間置き、孔径0.2μmのフィルターでろ過し、エッペンドルフチューブに等量分注したものを使用。
・202U/mL トロンビンストック溶液:トロンビン 202Uをエッペンドルフチューブに秤とり、DMEM(0%FBS、1%抗生物質)1mLをすぐに加え、37℃のウォーターバスに3~5分間置いて溶解させる。その後、孔径0.2μmのフィルターでろ過し、エッペンドルフチューブに等量分注したものを使用。
(解繊されたコラーゲン成分)
ブタ皮膚由来コラーゲンI型スポンジ断片(日本ハム株式会社製)100mgを200℃で24時間加熱を行うことにより、少なくとも一部が架橋されているコラーゲン成分(架橋コラーゲン成分)を得た。なお、200℃の加熱前後において、コラーゲンに外見上の大きな変化は確認されなかった。架橋コラーゲン成分50mgを15mLチューブに入れ、5mLの超純粋を加え、ホモジナイザー(アズワン社 VH-10)を用いて6分間ホモジナイズすることで架橋コラーゲン成分を解繊した。
ブタ皮膚由来コラーゲンI型スポンジ断片(日本ハム株式会社製)100mgを200℃で24時間加熱を行うことにより、少なくとも一部が架橋されているコラーゲン成分(架橋コラーゲン成分)を得た。なお、200℃の加熱前後において、コラーゲンに外見上の大きな変化は確認されなかった。架橋コラーゲン成分50mgを15mLチューブに入れ、5mLの超純粋を加え、ホモジナイザー(アズワン社 VH-10)を用いて6分間ホモジナイズすることで架橋コラーゲン成分を解繊した。
21℃の条件下、10000rpmで10分間遠心した。上清を吸引し、コラーゲンペレットを5mLの新しい超純水と混合し、コラーゲン溶液を作製した。コラーゲン溶液の入ったチューブを氷上に維持したまま、ソニケーター(Sonics and Materials社 VC50)を用いて100Vで20秒間超音波処理し、ソニケーターを取り出した後コラーゲン溶液の入ったチューブを氷上で10秒間冷却することを100回繰り返した。超音波処理を100回行った後、コラーゲン溶液を孔径40μmのフィルターでろ過し、解繊されたコラーゲン成分(CMF)を含む分散液を得た。分散液を常法によって凍結乾燥させることにより、乾燥体として、解繊コラーゲン成分(CMF)を得た。CMFの平均長(長さ)は、14.8±8.2μm(N=20)であった。
(成熟脂肪細胞及びADSCの採取方法)
ヒト脂肪組織断片を5%の抗生物質を含むPBSで洗浄した。4~6gの組織を6ウェルプレートの6ウェル分に分けた。2mg/mL コラゲナーゼ溶液2mL中ではさみ及びピンセットを使用しておよそ1~3mmのサイズに細かく切り刻んだ。37℃及び230rpmで1時間インキュベートした後、10mLピペットで30分間混合した。溶解物を孔径500μmの鉄メッシュフィルターでろ過し、1ウェル当たり2mLのDMEMを添加して消化された細胞すべてを回収した後、室温(15~25℃)下にて200gで3分間遠心した。成熟脂肪細胞は上層の黄色の油性層に、脂肪幹細胞及び血球はペレットに含まれる。長い針及び10mLシリンジを用いて、上層と下層の間の媒体を吸引及び廃棄し、上層に含まれる成熟脂肪細胞並びに下層に含まれる脂肪幹細胞及び血球を25mLのPBS(5%BSA、1%抗生物質)で二回洗浄した。洗浄を行う際には、上述したのと同様に遠心することで、上層及び下層及び上層と下層の間の媒体の3層に分離し、上層と下層の間の媒体を吸引及び廃棄した。二回の洗浄をした後、25mLのDMEMで洗浄した。
ヒト脂肪組織断片を5%の抗生物質を含むPBSで洗浄した。4~6gの組織を6ウェルプレートの6ウェル分に分けた。2mg/mL コラゲナーゼ溶液2mL中ではさみ及びピンセットを使用しておよそ1~3mmのサイズに細かく切り刻んだ。37℃及び230rpmで1時間インキュベートした後、10mLピペットで30分間混合した。溶解物を孔径500μmの鉄メッシュフィルターでろ過し、1ウェル当たり2mLのDMEMを添加して消化された細胞すべてを回収した後、室温(15~25℃)下にて200gで3分間遠心した。成熟脂肪細胞は上層の黄色の油性層に、脂肪幹細胞及び血球はペレットに含まれる。長い針及び10mLシリンジを用いて、上層と下層の間の媒体を吸引及び廃棄し、上層に含まれる成熟脂肪細胞並びに下層に含まれる脂肪幹細胞及び血球を25mLのPBS(5%BSA、1%抗生物質)で二回洗浄した。洗浄を行う際には、上述したのと同様に遠心することで、上層及び下層及び上層と下層の間の媒体の3層に分離し、上層と下層の間の媒体を吸引及び廃棄した。二回の洗浄をした後、25mLのDMEMで洗浄した。
成熟脂肪細胞を含む上層のみを採取し、エッペンドルフチューブに分注した。ヘキスト染色により核を染色し(1000倍希釈したヘキスト、15分間染色)、蛍光顕微鏡を用いて、Turker Burk血球計数器上で細胞数をカウントした。
ADSCを含むペレットをDMEM 10mLに懸濁し、10cmディッシュ内に播種し、継代培養した。トリプシン/EDTAを使用してADSCをディッシュから分離し、DMEM 1mLに懸濁し、細胞数をカウントした。
<組織体の製造>
製造例1
単離したADSCを、ALK5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)を0μM又は5μM含有するEGM-2 2mL、又は10%FBS含有DMEM2mLを培地に懸濁し、24ウェルプレート(IWAKI製)に10000cells/wellの濃度で播種して、37℃でインキュベートした。
製造例1
単離したADSCを、ALK5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)を0μM又は5μM含有するEGM-2 2mL、又は10%FBS含有DMEM2mLを培地に懸濁し、24ウェルプレート(IWAKI製)に10000cells/wellの濃度で播種して、37℃でインキュベートした。
2日ごとに培地を交換し、播種後7日間培養し、組織体を得た。
組織体の蛍光イメージングを次の手順で行った。得られた組織体を含むトランスウェルを24ウェルプレートに移動させた。PBS 2mLの洗浄の後、4%PFA 2mLを使用して4℃の条件下で一晩組織を固定した。PBS 2mLを用いて3回洗浄した。0.05% Triton/PBSを用いて、室温条件下で7分間、細胞を透過処理し(トランスウェル内に500μL、トランスウェル外に500μL)、PBS 2mLを用いて3回洗浄した。
1% BSA/PBS溶液を用いて、室温条件下で1時間組織をブロック処理した(トランスウェル内に500μL、トランスウェル外に500μL)。BSA溶液を吸引し、一次抗体溶液(1% BSA/PBS溶液で100倍に希釈した抗CD31抗体)100μLを添加した(トランスウェル内に50μL、トランスウェル外に50μL)。プレート下にウェットティッシュを敷き、アルミホイルで覆って、4℃の条件下で一晩インキュベートした。PBS 2mLを用いて3回洗浄した後、二次抗体溶液(1% BSA/PBS溶液で200倍に希釈したAlexaFluor647標識抗マウスIgG抗体及び1000倍希釈したヘキスト)100μLを添加した(トランスウェル内に50μL、トランスウェル外に50μL)。プレート下にウェットティッシュを敷き、アルミホイルで覆って、室温条件下で2時間インキュベートした後、PBS 2mLを用いて4回洗浄した。
トランスウェルのメンブレンを切断し、少量のPBSを含むガラスボトムディッシュの底に直接ゲルを配置し、染色した組織体を共焦点レーザー走査型顕微鏡(FV3000、オリンパス株式会社製)を用い、レーザー励起光を640nm(AlexaFluor647)として観察した。
図1は、血管網を有する組織体の蛍光観察結果(20倍拡大)を示す。(a)はALK5阻害剤(ALK5i)を含有しないEGM-2培地において培養した組織体、(b)はALK5阻害剤を5μM含有するEGM-2培地において培養した組織体、(c)は10%FBSを含有するDMEMにおいて培養した組織体である。(c)において、血管網の形成は確認できなかった。(a)と比較し、(b)の組織体では、生体組織と同様に直径の太い血管(例えば、10μm以上25μm未満)と細い血管(例えば、0μm超10μm未満)を有し、より血管の分岐数が多い血管網が形成されていることが確認された。
ALK5阻害剤を含む培地での培養により、脂肪由来幹細胞の血管内皮細胞への分化が促進されていることが示された。
製造例2
単離したADSCを、ALK5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)を0μM又は5μM含有するEGM-2培地に懸濁し、10cmディッシュに播種して、37℃でインキュベートした。
単離したADSCを、ALK5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)を0μM又は5μM含有するEGM-2培地に懸濁し、10cmディッシュに播種して、37℃でインキュベートした。
2日ごとに培地を交換し、7日間培養した。その後、トリプシン/EDTAを使用してADSCをディッシュから分離し、DMEM 1mLに懸濁し、細胞数をカウントした。更に、回収されたADSCを予めマトリゲルでコーティングした24ウェルプレート上に、1.0×105cells/dish、1.5×105cells/dish、2.0×105cells/dishの濃度で播種し、37℃で24時間インキュベートした。その後、組織体の形態を位相差顕微鏡で評価した。図2は、製造した組織体の位相差観察画像を示す。ADSCの細胞数にかかわらず、ALK5阻害剤(ALK5i)を含有するEGM-2において培養した組織体の方が、ALK5阻害剤を含有しないEGM-2において培養した組織体と比較して、より管腔形成が進んでいることが観察された。特に、ADSCの細胞数が1.5×105cells/mL以上の場合は、より血管の分岐数が多い血管網が形成されていることが確認されたことから、ALK5阻害剤を含む培地での培養により、脂肪由来幹細胞の血管内皮細胞への分化が促進されていることが示唆された。
製造例3
単離したADSCを、ALK5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)を5μM含有するEGM-2培地に懸濁し、10cmディッシュに播種して、37℃でインキュベートした。2日ごとに培地を交換し、7日間培養して、組織体を得た。その後、0.25%Trypsinを用いた5分間のインキュベーションにより、ADSCを細胞表面のタンパク質を残したまま穏やかに剥離してディッシュから回収し、DMEM 1mLに懸濁し、細胞数をカウントした。

BD Facsmelody(登録商標)cellsorter(Beckton Dickinson社製)を用いたフローサイトメトリー解析により、組織体中の血管内皮細胞の比率を解析した。上記の通り回収された組織体を、遠心分離(100g、5分間、4℃)した。その後、alexa fluor647で標識した抗CD31抗体(100×希釈)を加え、細胞懸濁液を4℃でさらに30分間インキュベートした。その後、遠心分離(100g、5分間、4℃)して細胞を洗浄し、ペレット化した細胞をバッファーに懸濁して10,000イベントを記録した。さらに、コンジュゲート抗体のアイソタイプコントロールも同時に測定した。その後、Flowjo(登録商標)Software(Beckton Dickinson社)を用いて解析し、CD31+細胞の比率を求めた。
結果を図3に示す。(a)は製造した組織体を用いた結果、(b)は組織消化後に単離した細胞をそのまま用いた結果を示す。(a)においては、組織体から回収された全細胞数を基準として29.29%が血管内皮細胞であることが分かった。一方、(b)では、わずか1.20%が血管内皮細胞であった。
製造例4
成熟脂肪細胞、ADSC及びADSC由来血管内皮細胞を、2.5:1.5:1.5の比率(細胞数)で用いて製造した組織体(細胞ボール)、並びに成熟脂肪細胞及びADSC由来血管内皮細胞を2.5:3の比率(細胞数)で用いて製造した組織体(細胞ボール)を、成熟脂肪細胞、ADSC及びHUVECを2.5:2:1の比率(細胞数)で用いて同様に製造した組織体(細胞ボール)と比較した。いずれの組織体も用いた細胞の総細胞数は4.0×104cells/ballである。成熟脂肪細胞及びADSCは、上記(成熟脂肪細胞及びADSCの採取)に記載のように単離したヒト由来の細胞を用いた。ADSC由来血管内皮細胞は、製造例3で調整した組織体から得られたものを用いた。
成熟脂肪細胞、ADSC及びADSC由来血管内皮細胞を、2.5:1.5:1.5の比率(細胞数)で用いて製造した組織体(細胞ボール)、並びに成熟脂肪細胞及びADSC由来血管内皮細胞を2.5:3の比率(細胞数)で用いて製造した組織体(細胞ボール)を、成熟脂肪細胞、ADSC及びHUVECを2.5:2:1の比率(細胞数)で用いて同様に製造した組織体(細胞ボール)と比較した。いずれの組織体も用いた細胞の総細胞数は4.0×104cells/ballである。成熟脂肪細胞及びADSCは、上記(成熟脂肪細胞及びADSCの採取)に記載のように単離したヒト由来の細胞を用いた。ADSC由来血管内皮細胞は、製造例3で調整した組織体から得られたものを用いた。
sCMFを2.4mg秤取り、DMEM 1mLを添加し、sCMFの小さな粒子のみが観察されるようになるまで穏やかに混合した。室温下にて10000rpmで1分間遠心し、上清を吸引し、sCMFペレットを得た。ADSC及び/又はADSC由来血管内皮細胞(比較例ではさらにHUVEC)をsCMFペレット上に穏やかに添加し、室温下にて3500rpmで1分間遠心し、上清を吸引した。フィブリノゲン(50mg/mL フィブリノゲンストック溶液32μL)を添加し、細胞及びsCMFと穏やかに混合した。さらに成熟脂肪細胞を添加して穏やかに混合した。少量のDMEMを添加し、総量を200μLに調整した。すぐにトロンビン(202U/mL トロンビンストック溶液3.9μL)を添加し、少し混合した後、低接着96ウェルプレート(IWAKI #4860-800LP)上に混合物(細胞ボール40個分)を各ウェル5μL(細胞ボール1個分)ずつ播種した。
37℃のインキュベータ内で15分間インキュベートしてゲル化させ、最終濃度10μg/mLのインスリンを含むEGM-2 300μLを添加した。
24時間培養した後、低接着24ウェルプレート(IWAKI #4820-800LP)に移し、2mLの上記EGM-2を添加した。培地を2~3日毎に培養7日目まで交換した。
組織体の蛍光イメージングを次の手順で行った。得られた組織体を含むトランスウェルを24ウェルプレート(IWAKI #4820-800LP)に移動させた。PBS 200μLの洗浄の後、4%PFA 200μLを使用して4℃の条件下で一晩組織を固定した。PBS 200μLを用いて3回洗浄した。免疫染色のため、0.05% Triton/PBS 200μLを用いて、室温条件下で7分間、細胞を透過処理し、PBS 200μLを用いて3回洗浄した。
1% BSA/PBS溶液 200μLを用いて、室温条件下で1時間組織をブロック処理した。BSA溶液を吸引し、一次抗体溶液(1% BSA/PBS溶液で100倍に希釈した抗CD31抗体)100μLを添加した。プレート下にウェットティッシュを敷き、アルミホイルで覆って、4℃の条件下で一晩インキュベートした。PBS 200μLを用いて3回洗浄した後、二次抗体溶液(1% BSA/PBS溶液で200倍に希釈したAlexaFluor647標識抗マウスIgG抗体)を添加した。プレート下にウェットティッシュを敷き、アルミホイルで覆って、室温条件下で2時間インキュベートした後、PBS 200μLを用いて4回洗浄した。
共焦点定量イメージサイトメーターCQ1(横河電機製)のコンプリートプレート上に直接細胞ボールを配置し、染色した組織体を上記共焦点定量イメージサイトメーターCQ1を用い、レーザー励起光を640nm(AlexaFluor647)として観察した。
図4は、CD31染色による、血管網を有する細胞ボールの蛍光観察結果を示す。ALK5阻害剤により脂肪幹細胞の血管内皮細胞への分化が促進された組織体から得られた血管内皮細胞を使用した細胞ボールでは、ADSCの使用の有無及び各種の細胞の比率にかかわらず、HUVECを使用して作成した組織体と同様に血管網が形成されていた。これらの血管網は細胞ボールの内部の方まで形成されていた。
製造例5

結果を図5に示す。5μMのALK5阻害剤含有EGM2培地下で培養したサンプルでは、回収された全細胞数を基準として32.4%が血管内皮細胞であることが分かった。一方、ALK5阻害剤を含まないEGM2培地下で培養したサンプルでは、14.1%が血管内皮細胞であり、ALK5阻害剤がADSCからの血管内皮細胞への分化を顕著に誘導したことが示された。
単離したADSCを、EGM-2培地に懸濁し、10cmディッシュに播種して、37℃でインキュベートした。2日ごとに培地を交換し、7日間培養して、組織体を得た。この組織体から、ADSC、ADSC由来血管内皮細胞、及び血管内皮前駆細胞を含む細胞を回収した。
上記回収した細胞、sCMF、フィブリン、成熟脂肪細胞、及びトロンビンを製造例4と同様の方法で混合し、低接着96ウェルプレート(IWAKI #4860-800LP)上に混合物(細胞ボール40個分)を各ウェル5μL(細胞ボール1個分)ずつ播種した。用いた組織体(細胞ボール)の総細胞数は、4.0×104cells/ballであり、成熟脂肪細胞、及び上記回収した細胞は、3:2.5の比率で用いて製造した。
低接着96ウェルプレートに播種した細胞ボールを、37℃のインキュベータ内で15分間インキュベートしてゲル化させ、EGM-2又はKBM 300μLを添加した。なお、上記EGM-2又はKBMは、最終濃度10μg/mLのインスリン、5μMのALK-5阻害剤(2-[3-(6-メチル-2-ピリジニル)-1H-ピラゾール-4-イル]-1,5-ナフチリジン、Cayman Chemical社 No. 14794)及び、0.5ng/mL、20ng/mL、50ng/mL、100ng/mLのVEGF、又は1μg/mLのレプチンを含むように調整したものを用いた。また、コントロールとして最終濃度10μg/mLのインスリン、及び5μMのALK-5阻害剤のみを含むEGM-2又はKBMを用いた。
24時間培養した後、低接着24ウェルプレート(IWAKI #4820-800LP)に移し、2mLの上記EGM-2又はKBMを添加した。培地を2~3日毎に培養7日目まで交換した。
組織体の蛍光イメージングを、一次抗体溶液として1%BSA/PBS溶液で100倍に希釈した抗フォン・ウィルブランド因子(vWF)抗体を用いたこと、及び二次抗体溶液として1% BSA/PBS溶液で200倍に希釈したAlexaFluor647標識抗ウサギIgG抗体溶液を用いたこと以外は製造例4と同様にして行った。vWFは、血管内皮細胞のマーカーとして用いた。また、観察は共焦点定量イメージサイトメーターCQ1(横河電機製)のコンプリートプレート上に直接細胞ボールを配置し、染色した組織体を、上記共焦点定量イメージサイトメーターCQ1を用い、レーザー励起光を640nmとした。
vWF染色による血管網を有する細胞ボールの蛍光観察結果を図6に示す。VEGFを含む培地中で培養した細胞ボールでは、VEGFを含まない培地中で培養したコントロールの細胞ボールと比較して、より血管の分岐数が多い血管網が形成されていた。また、レプチンを含む培地で培養した細胞ボールと比較しても、VEGFを含む培地中で培養した細胞ボールは、より血管の分岐数が多い血管網が形成されていた。VEGFを添加した培地中で培養した細胞ボールの中でも、培地に20ng/mL以上のVEGFを含む培地で培養することで、さらに血管の多い血管網が形成されていた。
この結果から、培地にALK-5阻害剤に加えてVEGFを添加することにより、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化及び血管新生がより促進されていることが示された。
製造例7
製造例6と同様の方法で、組織体(細胞ボール)を製造した。製造した組織体を低接着96ウェルプレート(IWAKI #4860-800LP)上に混合物(細胞ボール40個分)を各ウェル5μL(細胞ボール1個分)ずつ播種した。

低接着96ウェルプレートに播種した細胞ボールを、37℃のインキュベータ内で15分間インキュベートしてゲル化させ、最終濃度10μg/mLのインスリン、5μMのAKL-5阻害剤、及び50ng/mLのVEGFを含むEGM-2 300μLを添加した。
24時間培養した後、低接着24ウェルプレート(IWAKI #4820-800LP)に移し、2mLの上記EGM-2又はKBMを添加した。培地を2~3日毎に培養7日目まで交換した。
組織体の蛍光イメージングを、様々な一次抗体溶液及びそれらに対応する二次抗体溶液を用いたこと以外は製造例4と同様にして行った。一次抗体溶液は、1%BSA/PBS溶液で100倍に希釈したウサギ抗CD34抗体、1%BSA/PBS溶液で100倍に希釈したマウス抗CD31、又は1%BSA/PBS溶液で100倍に希釈したウサギ抗フォン・ウィルブランド因子(vWF)抗体を用いた。二次抗体溶液は、1%BSA/PBS溶液で200倍に希釈したAlexaFluor546標識抗マウスIgG抗体、1%BSA/PBS溶液で200倍に希釈したAlexaFluor647標識抗ウサギIgG抗体を用いた。CD34は、血管内皮前駆細胞のマーカーとして用いた。CD31は、血管内皮前駆細胞及び血管内皮細胞のマーカーとして用いた。vWFは、血管内皮細胞のマーカーとして用いた。また、ヘキスト染色による核の染色も行った。
観察は、共焦点定量イメージサイトメーターCQ1(横河電機製)のコンプリートプレート上に直接細胞ボールを配置し、染色した組織体を、上記共焦点定量イメージサイトメーターCQ1を用いた。AlexaFluor546に対するレーザー励起光を556nmとし、AlexaFluor647に対するレーザー励起光を640nmとした。
その結果を図7に示す。VEGFの培地への添加により、全CD31陽性細胞に対するvWF陽性細胞の割合が、CD34陽性細胞の割合よりも高くなっていた。つまり、上記のとおり製造した細胞ボールにおいて、血管内皮前駆細胞数よりも血管内皮細胞数の方が多かった。また、血管網も形成されていた。
この結果から、培地にALK-5阻害剤に加えてVEGFを添加することにより、ヒト由来の脂肪由来幹細胞の血管内皮細胞への分化及び血管新生がより促進されていることが示された。
Claims (14)
- ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、血管内皮細胞を含む組織体の製造方法。
- ヒト由来の脂肪由来幹細胞を少なくとも含む細胞が、血管内皮細胞を含まない、請求項1に記載の方法。
- 前記TGFβタイプI受容体阻害剤の存在下でインキュベートすることが、前記TGFΒタイプI受容体阻害剤が含まれる培地中でインキュベートすることであり、培地における前記TGFβタイプI受容体阻害剤の含有量が1μM以上10μM以下である、請求項1に記載の製造方法。
- インキュベートが、96時間以上288時間以下で行われる、請求項1に記載の製造方法。
- 前記細胞を断片化細胞外マトリックス成分とともにTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、請求項1に記載の製造方法。
- 前記組織体において、前記断片化細胞外マトリックス成分が前記細胞同士の隙間に配置されている、請求項5に記載の製造方法。
- 前記断片化細胞外マトリックス成分が断片化コラーゲン成分である、請求項5に記載の製造方法。
- インキュベート前に、水性媒体中において前記細胞と前記断片化細胞外マトリックス成分とを接触させる工程をさらに含む、請求項5~7のいずれか一項に記載の製造方法。
- ヒト由来の脂肪由来幹細胞を少なくとも含む細胞をTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、脂肪由来幹細胞の血管内皮細胞への分化促進方法。
- 前記TGFβタイプI受容体阻害剤の存在下でインキュベートすることが、前記TGFβタイプI受容体阻害剤が含まれる培地中でインキュベートすることであり、培地における前記TGFβタイプI受容体阻害剤の含有量が1μM以上10μM以下である、請求項9に記載の方法。
- インキュベートが、96時間以上288時間以下で行われる、請求項9に記載の方法。
- 脂肪由来幹細胞を少なくとも含む細胞を断片化細胞外マトリックス成分とともにTGFβタイプI受容体阻害剤の存在下でインキュベートすることを含む、請求項9に記載の方法。
- 前記断片化細胞外マトリックス成分が断片化コラーゲン成分である、請求項12に記載の方法。
- インキュベート前に、水性媒体中において前記細胞と前記断片化細胞外マトリックス成分とを接触させる工程をさらに含む、請求項12又は13に記載の方法。
