Abstract
【課題】シモンコライトの新たな用途を提供すること。 【解決手段】本発明の育毛用化粧料は、シモンコライト結晶を有効成分として含有している。
Description
本発明は、化粧料用原料及び化粧料に関し、特に、育毛用又は頭皮ケア用として用いられる化粧料用原料及び化粧料に関する。
シモンコライトとは、亜鉛、水酸基及び塩素から構成される塩化水酸化亜鉛水和物であり、その化学式はZn5(OH)8Cl2・(H2O)nで表される化合物である。水及び有機溶媒に不溶性の白色結晶粉末であり、塩基性塩化亜鉛または塩化水酸化亜鉛とも称される。天然の鉱物でもあるシモンコライトは、1985年に新鉱物として発表され、シモンコライトの鉱物サンプルを採集したヴェルナー・シモン(Werner Simon)とクルト・コール(Kurt Kolle)にちなんで名付けられた。これまで、シモンコライトは、主に、動物用の飼料添加物である四塩基性塩化亜鉛(TBZC)を構成する主成分として利用され、動物にとって望ましい栄養補助食品とされている。
近年、このシモンコライトの新たな用途や工業的な製造方法に関する研究開発が進められている。例えば、特許文献1では、皮膚創傷または皮膚荒れ治療剤としてシモンコライトを用いる技術が報告されており、特許文献2では、医薬品の原体として用いられ得るシモンコライトの製造方法が報告されている。
他方、ヘアケア・頭皮ケアの分野において、亜鉛を含む化合物の効能が報告されている。例えば、特許文献3では、グルコン酸亜鉛、塩化亜鉛及び硫酸亜鉛などの亜鉛塩がトリプシン阻害活性を有することから、抜け毛の予防又は低減に有用であることが報告されている。また、特許文献4には、養毛・育毛成分であるオウゴンエキスと、グルコン酸亜鉛及びシクロヘキサンジカルボン酸ビスエトキシグリコールを含有する育毛用組成物が記載されており、このうち、グルコン酸亜鉛は毛髪にハリ・コシを付与する作用を有することが記載されている。さらに、非特許文献1では、亜鉛錯体化合物であるピリチオン亜鉛の毛髪密度・毛髪太さに対する効果の検討結果が報告されている。

特許文献3、4で報告されたグルコン酸亜鉛、塩化亜鉛及び硫酸亜鉛などの亜鉛塩は、水に対する溶解度が高く、水に溶けて亜鉛イオンを速やかに生成する物質であり、この亜鉛イオンが抜け毛の予防又は低減、毛髪にハリ・コシを付与する作用を奏している。それを裏付けるが如く、非特許文献1では、水に対する溶解度が非常に低いピリチオン亜鉛の毛髪密度・毛髪太さに対する効果が検討されたものの、ピリチオン亜鉛は、陽性対照であるミノキシジルの半分以下の効果しか観察されなかったことが報告されている。シモンコライトもピリチオン亜鉛同様に水中に溶出する亜鉛イオンの量が非常に微量であることから、シモンコライトには特許文献3、4で報告されたような効能は期待されていなかった。それゆえ、シモンコライトをヘアケア・頭皮ケアのために用いることについての検討はこれまでなされておらず、その有効性はまったく不明であった。
他方、上述した特許文献1において用いられ、特許文献2で紹介された製造方法にて製造されたシモンコライト結晶や、その他一般的に流通するシモンコライト結晶は、一次粒子径が約10μm(D50:メジアン径)程度と大きく、さらに凝集性も有することがわかった。本願発明者らが、これらのシモンコライト結晶を水やエタノール等の分散媒体に分散させ、他の一般的に使用される化粧料用原料と配合して化粧料の調製を試みたところ、回復不可能な、あるいは容易に再分散しない凝集沈殿を起こすという問題に直面した。
さらに、シモンコライト結晶は粒子径が大きい白色の粉体であるために、そのままでは勿論、水やエタノール等の分散媒体に分散させて肌に塗布した場合でも「ざらつき」が感じられ、白色の「肌残り」が生じた。このような物性は、ユーザーから嫌われる可能性が高く、特に化粧料用原料として用いるには大きな難点である。特に、頭皮ケア用化粧料にシモンコライトを配合した場合、頭皮に塗布すると、シモンコライト結晶による白色の「肌残り」は頭髪の色と極端に異なるために毛髪中で目立ち、「ふけ」と混同される可能性があり、美容上極めて好ましくない状況を呈した。
したがって、本発明は上述した点に鑑みてなされたもので、その目的は、シモンコライトの新たな用途を提供することにある。
また、本発明の他の目的は、水やエタノール等の分散媒体に分散させても凝集せずに容易に再分散され、取り扱いが容易な、シモンコライト結晶を含有する化粧料用原料及び化粧料を提供することにある。
また、本発明の他の目的は、使用時の「ざらつき」や白色の「肌残り」が低減され、滑らかな使用感と使用後においても美容上好ましい外観を呈する、シモンコライト結晶を含有する化粧料用原料及び化粧料を提供することにある。
本願発明者らは、シモンコライト結晶について様々な用途を検討したところ、シモンコライト結晶にミノキシジルを凌駕する毛の伸長促進作用を有することを見出した。この知見に基づき、本発明を完成するに至った。上記課題を解決するため、本発明の育毛用化粧料は、シモンコライト結晶を有効成分として含有する。シモンコライト結晶が毛包組織における毛の伸長を促進するため、毛髪の健やかな成長が維持され、健康な毛髪を得ることができる。
また、本発明の育毛用化粧料のシモンコライト結晶の配合量は、0.001重量%~10重量%であることも好ましい。これにより、育毛効果に優れるシモンコライト結晶の濃度が選択される。
また、本発明の育毛用化粧料のシモンコライト結晶が、レーザー回折散乱法による体積基準の粒子径分布における90%累積粒子径(D90)が800nm以下のナノ粒子であることも好ましい。シモンコライト結晶をD90が800nm以下のナノ粒子とするとすることにより、頭皮に塗布した際に、有効成分である不溶性のシモンコライト結晶によるざらつき感が解消すると共に、白色の肌残りが生じなくなるため、滑らかな使用感と使用後においても美容上好ましい外観を呈する育毛用化粧料が得られる。
また、本発明の育毛用化粧料は、さらに、分散媒体と分散剤を含有し、分散剤が、ポリエチレングリコール、ポリプロピレングリコール、エチレンオキサイド・プロピレンオキサイド共重合体、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース及びヒドロキシエチルセルロースからなる群から選択される少なくとも1種の化合物であることも好ましい。これにより、育毛用化粧料の剤形をシモンコライト結晶が分散媒体中に分散されてなるスラリー(懸濁液)とした際に、シモンコライト結晶が凝集し難く、保存安定性が高まると共に容易に再分散可能となるため、取り扱いも容易な育毛用化粧料が得られる。
本発明の化粧料用原料又は化粧料は、シモンコライト結晶のナノ粒子を含有する。これにより、化粧料として有用な新たな化粧料用原料及び化粧料が得られる。シモンコライト結晶をナノ粒子とすることにより、シモンコライト結晶によるざらつき感が低減すると共に、白色の肌残りが生じ難くなるため、滑らかな使用感と使用後においても美容上好ましい外観を呈する化粧料を得ることができる。なお、本明細書において、ナノ粒子とは、レーザー回折散乱法による、体積基準の粒子径分布における小粒子側からの90%累積粒子径(D90)が、1nm以上1000nm未満の粒子状物質のことをいう。
また、本発明の化粧料用原料又は化粧料のシモンコライト結晶のナノ粒子は、レーザー回折散乱法による体積基準の粒子径分布における90%累積粒子径(D90)が、800nm以下であることも好ましい。これにより、皮膚に塗布した際のシモンコライト結晶によるざらつき感と白色の肌残りが生じなくなるため、より滑らかな使用感と使用後においても美容上好ましい外観を呈する化粧料が得られる。
また、本発明の化粧料用原料又は化粧料のシモンコライト結晶のナノ粒子は、レーザー回折散乱法による体積基準の粒子径分布における50%累積粒子径(D50)が、250nm以下であることも好ましい。これにより、皮膚に塗布した際のシモンコライト結晶によるざらつき感と白色の肌残りが生じなくなるため、さらに滑らかな使用感と使用後においても美容上好ましい外観を呈する化粧料が得られる。
また、本発明の化粧料用原料又は化粧料は、さらに、分散媒体として、水、水混和性有機溶媒又はこれらの組み合わせを含有する。これにより、スラリー(懸濁液)状の化粧料用原料又は化粧料が得られる。
また、本発明の化粧料用原料又は化粧料は、さらに、分散剤を含有することも好ましい。これにより、スラリーに含まれるシモンコライト結晶が凝集し難くなり、保存安定性が高まると共に容易に再分散されるために、取り扱いも容易な化粧料用原料又は化粧料が得られる。
また、上述した分散剤は、ポリエチレングリコール、ポリプロピレングリコール、エチレンオキサイド・プロピレンオキサイド共重合体、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース及びヒドロキシエチルセルロースからなる群から選択される少なくとも1種の化合物であることも好ましい。これにより、分散剤として好適な化合物が選択される。
また、本発明の化粧料用原料又は化粧料のシモンコライト結晶の配合量は、0.001重量%~30重量%であることも好ましい。これにより、化粧料用原料又は化粧料として好適なシモンコライト結晶の濃度が選択される。
また、本発明の化粧料用原料又は化粧料は、育毛用又は頭皮ケア用であることも好ましい。シモンコライト結晶は毛包組織における毛の伸長を促進し、毛髪の健やかな成長を維持する効果を有することから、化粧料用原料又は化粧料として好適な用途が選択される。
本発明の化粧料用原料又は化粧料の製造方法は、シモンコライト結晶と分散媒体を混合して懸濁液を得る混合工程と、この懸濁液を1時間以上撹拌する撹拌工程と、撹拌工程後の懸濁液を微粒化装置に導入し、懸濁液に含まれるシモンコライト結晶をナノレベルのサイズに微細化する微細化処理工程と、を有する。シモンコライト結晶と分散媒体との懸濁液を1時間以上撹拌して、両者を馴染ませる撹拌工程を経ることによって、微粒化装置内の流路やノズル等の管路詰まりが生じ難くなり、シモンコライト結晶の微細化処理が容易となるため、シモンコライト結晶がナノレベルのサイズに微細化された化粧料用原料又は化粧料が得られる。
また、本発明の化粧料用原料又は化粧料の製造方法は、混合工程又は撹拌工程において、懸濁液にさらに分散剤を混合することを特徴とすることも好ましい。分散剤を混合工程又は撹拌工程に添加し、シモンコライト結晶と分散剤とを分散媒体に馴染ませることによって、微粒化装置によるシモンコライト結晶の微細化処理が容易となると共に、シモンコライト結晶が凝集し難く、分散性も向上した化粧料用原料又は化粧料が得られる。
本発明の化粧料用原料又は化粧料の製造方法は、シモンコライト結晶と、水混和性有機溶媒を有する分散媒体を混合して懸濁液を得る混合工程と、懸濁液を微粒化装置に導入し、懸濁液に含まれるシモンコライト結晶をナノレベルのサイズに微細化する微細化処理工程と、微細化処理後の懸濁液に含まれる水混和性有機溶媒を留去する工程と、を有する。分散媒体として水混和性有機溶媒を用いることにより、微粒化装置内の流路やノズル等の管路詰まりが生じ難くなるため、分散媒体とシモンコライト結晶とを馴染ませるための撹拌工程を行わずとも、両者の懸濁液を微粒化装置に導入して微細化処理を行うことができる。このとき、得られた懸濁液から水混和性有機溶媒を留去することにより、水混和性有機溶媒と他の溶媒、具体的には、水混和性有機溶媒を水に置換することができるため、微細化されたシモンコライト結晶の水懸濁液(スラリー)からなる化粧料用原料又は化粧料を得ることができる。
また、本発明の化粧料用原料又は化粧料の製造方法は、混合工程において、さらに分散剤を混合して懸濁液を得ることも好ましい。分散剤を添加することによって、シモンコライト結晶が凝集し難く、分散性も向上した化粧料用原料又は化粧料が得られる。
本発明によれば、以下のような優れた効果を有する育毛用化粧料を提供することができる。
(1)シモンコライト結晶が毛髪の伸長を促進するため、毛髪の健やかな成長が維持され、健康な毛髪が得られる。
(2)効果持続期間が長いため、使用頻度を少なくすることができる。
(3)有効成分であるシモンコライト結晶をナノ粒子とすることにより、シモンコライト結晶によるざらつき感や白色の肌残りが生じなくなるため、滑らかな使用感と使用後においても美容上好ましい外観を呈する。
(4)分散剤を配合することにより、シモンコライト結晶が凝集し難くなり、保存安定性が高まると共に容易に再分散可能となるため、取り扱いも容易である。
(1)シモンコライト結晶が毛髪の伸長を促進するため、毛髪の健やかな成長が維持され、健康な毛髪が得られる。
(2)効果持続期間が長いため、使用頻度を少なくすることができる。

また、本発明によれば、以下のような優れた効果を有する化粧料用原料又は化粧料及びその製造方法を提供することができる。
(1)シモンコライト結晶をナノ粒子としたため、シモンコライト結晶によるざらつき感が低減すると共に白色の肌残りも生じ難くなり、滑らかな使用感と使用後においても美容上好ましい外観を呈する化粧料を得ることができる。
(2)分散剤を配合したことにより、スラリー中のシモンコライト結晶が凝集し難くなり、保存安定性が高まると共に容易に再分散されるために、取り扱いも容易な化粧料用原料又は化粧料を得ることができる。
(3)シモンコライト結晶は毛髪の伸長を促進し、毛髪の健やかな成長を維持する効果を有することから、育毛用又は頭皮用の化粧料用原料又は化粧料として好適に使用することができる。
(4)シモンコライト結晶がナノレベルのサイズに微細化されたスラリー状の化粧料用原料又は化粧料を容易に得ることができる。
(1)シモンコライト結晶をナノ粒子としたため、シモンコライト結晶によるざらつき感が低減すると共に白色の肌残りも生じ難くなり、滑らかな使用感と使用後においても美容上好ましい外観を呈する化粧料を得ることができる。
(2)分散剤を配合したことにより、スラリー中のシモンコライト結晶が凝集し難くなり、保存安定性が高まると共に容易に再分散されるために、取り扱いも容易な化粧料用原料又は化粧料を得ることができる。
(3)シモンコライト結晶は毛髪の伸長を促進し、毛髪の健やかな成長を維持する効果を有することから、育毛用又は頭皮用の化粧料用原料又は化粧料として好適に使用することができる。
(4)シモンコライト結晶がナノレベルのサイズに微細化されたスラリー状の化粧料用原料又は化粧料を容易に得ることができる。
以下、本発明の育毛用化粧料、化粧料用原料及び化粧料並びにその製造方法について説明する。
本発明の育毛用化粧料、化粧料用原料又は化粧料において用いられるシモンコライト結晶とは、Zn5(OH)8Cl2・(H2O)nで表される、塩化水酸化亜鉛水和物の白色結晶粉末のことをいう。シモンコライト結晶は、上述した特許文献2に示される方法等によって工業的に製造されたものが一般的に流通しており、一例として、JFEミネラル株式会社製品のシモンコライトが好適に用いられるが、その他の製造方法にて得られたシモンコライトや、天然鉱物由来のシモンコライトを用いることも可能である。
本発明の育毛用化粧料に配合されるシモンコライト結晶は、頭皮に塗布した際のシモンコライト結晶に起因するざらつき感と、塗布後の白色の肌残りを低減するため、ナノ粒子であることが好ましい。本明細書において、ナノ粒子とは、レーザー回折・散乱法による、体積基準の粒子径分布における小粒子側からの90%累積粒子径(D90)が、1nm以上1000nm未満の粒子状物質のことをいう。本発明においては、シモンコライト結晶に起因するざらつき及び白色の肌残りがより解消される観点から、シモンコライト結晶を、90%累積粒子径(D90)が800nm以下のナノ粒子とすることが好ましく、50%累積粒子径(D50)が250nm以下のナノ粒子とすることがより好ましく、50%累積粒子径(D50)が150nm以下のナノ粒子とすることがさらに好ましく、90%累積粒子径(D90)が550nm以下かつ50%累積粒子径(D50)が、150nm以下のナノ粒子とすることが特に好ましい。
本発明における育毛用途、すなわち、育毛用化粧料又は育毛用の化粧料用原料とは、毛包組織における毛の伸長を促進する作用を有する化粧料又は化粧料用原料のことをいい、本発明の育毛用化粧料又は化粧料用原料を塗布又は添加されない状態のコントロールと比較して、毛包組織における毛の伸長を促進することのできる化粧料又は化粧料用原料のことを意味する。毛包組織における毛の伸長の促進レベルは、例えば、ラットの髭毛包の器官培養法等の公知の方法で測定され得る。
本発明の育毛用化粧料に含有されるシモンコライト結晶の配合量は、目標とする育毛効果、処方や剤形、使用方法・回数等によって適宜設定され得るが、その作用効果の観点から、0.001重量%~10重量%が好ましく、0.005重量%~5重量%がより好ましく、0.01重量%~3重量%がさらに好ましい。
他方、本発明の化粧料用原料又は化粧料に配合されるシモンコライト結晶はナノ粒子である。これにより、肌に塗布した際のシモンコライト結晶に起因するざらつき感と、塗布後の白色の肌残りが低減される。ナノ粒子の粒子径としては、具体的には、シモンコライト結晶に起因するざらつき及び白色の肌残りがより解消される観点から、シモンコライト結晶を、90%累積粒子径(D90)が800nm以下のナノ粒子とすることが好ましく、50%累積粒子径(D50)が、250nm以下のナノ粒子とすることがより好ましく、50%累積粒子径(D50)が150nm以下のナノ粒子とすることがさらに好ましく、90%累積粒子径(D90)が550nm以下かつ50%累積粒子径(D50)が、150nm以下のナノ粒子とすることが特に好ましい。
本発明の育毛用化粧料、化粧料用原料又は化粧料は、取り扱いのし易さの観点から、分散媒体中にシモンコライト結晶を分散してなる懸濁液(スラリー)であることが好ましい。分散媒体としては、水、水混和性有機溶媒又はこれらの組み合わせが用いられる。水混和性有機溶媒としては、エタノールやプロパノール等のアルコール類、テトラヒドロフラン等のエーテル類、酢酸メチル等のエステル類又はアセトン等のケトン類などの水に混和可能な有機溶媒が挙げられるが、安全性等の観点から、エタノールが好適に用いられる。
さらに、育毛用化粧料、化粧料用原料又は化粧料を懸濁液(スラリー)とした際には、懸濁液中でシモンコライト結晶が容易に再分散しない凝集沈殿を形成するのを防ぐため、分散剤を配合することが好ましい。分散剤としては、エチレンオキサイド重合体、エチレンオキサイド・プロピレンオキサイド共重合体、セルロース誘導体又はこれらの組み合わせが挙げられる。具体的には、特に限定されないが、ポリエチレングリコール、ポリプロピレングリコール、エチレンオキサイド・プロピレンオキサイド共重合体、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース又はヒドロキシエチルセルロースが好適に用いられる。このうち、懸濁液中のシモンコライト結晶の分散性をより向上させる観点から、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース、ポリエチレングリコール6000又はこれらの組み合わせがより好適に用いられる。懸濁液中における分散剤の配合量は、分散性を向上させる観点から、0.05重量%~10重量%が好ましく、0.1重量%~5重量%がより好ましい。
本発明の化粧料用原料に含有されるシモンコライト結晶の濃度は、化粧料用原料としての流通性向上及び製造時の取り扱いのし易さの観点から、1重量%~30重量%が好ましく、5重量%~20重量%がより好ましい。他方、本発明の化粧料に含有されるシモンコライト結晶の濃度は、目的とする用途(機能)・効果、処方や剤形、使用方法等によって適宜設定され得る。具体的には、特に限定されないが、一例として、0.001重量%~10重量%が好ましく、0.005重量%~5重量%がより好ましく、0.01重量%~3重量%がさらに好ましい。
本発明の化粧料用原料又は化粧料の用途としては、上述したように、シモンコライト結晶は育毛効果を有するため、育毛用又は頭皮ケア用として用いることができる。さらに、シモンコライト結晶が備える他の機能の発揮を目的とした化粧料用原料又は化粧料として用いることが可能である。
次に、本発明の育毛用化粧料、化粧料用原料又は化粧料の製造方法について説明する。これらに配合されるシモンコライト結晶はナノ粒子であるところ、一般的に流通しているシモンコライト結晶は、平均粒子径(D50)が約10μm程度と大きい。そのため、このシモンコライト結晶をナノレベルのサイズに微細化する必要がある。微細化処理にあたっては、ナノレベルのサイズへの微細化に優れることから湿式粉砕を行う。以下、詳細に説明する。
(混合工程)
まず、シモンコライト結晶と分散媒体とを混合し、懸濁液を得る。分散媒体としては、上述したように、水、水混和性有機溶媒又はこれらの組み合わせが用いられる。この混合工程において、上述した分散剤を添加し、懸濁液に分散剤を含有させておくことも好ましい。シモンコライト結晶が含まれる懸濁液中に分散剤を含有させ、その懸濁液を微粒化装置に導入してシモンコライト結晶を微細化処理することにより、より分散性が向上したスラリーを得ることができるためである。
まず、シモンコライト結晶と分散媒体とを混合し、懸濁液を得る。分散媒体としては、上述したように、水、水混和性有機溶媒又はこれらの組み合わせが用いられる。この混合工程において、上述した分散剤を添加し、懸濁液に分散剤を含有させておくことも好ましい。シモンコライト結晶が含まれる懸濁液中に分散剤を含有させ、その懸濁液を微粒化装置に導入してシモンコライト結晶を微細化処理することにより、より分散性が向上したスラリーを得ることができるためである。
(撹拌工程)
次に、シモンコライト結晶と分散媒体とを混合して得られた懸濁液を撹拌する。この撹拌工程によって、懸濁液中のシモンコライト結晶が分散媒体に馴化するため、微細化処理の際に、微粒化装置内の流路やノズル等の管路詰まりが生じ難くなり、シモンコライト結晶の微細化処理が容易となる。撹拌手段としては、懸濁液全体を撹拌できるものであればよく、特に限定されないが、例えば、プロペラ型撹拌機やマグネチックスターラ等を用いることができる。撹拌時間としては、分散媒体とシモンコライト結晶とを馴染ませるために、1時間以上が好ましく、2時間以上とすることがより好ましい。また、この撹拌工程において、上述した分散剤を添加し、懸濁液に分散剤を含有させておくことも好ましい。なお、分散媒体として、エタノール等の水混和性有機溶媒を選択した場合には、撹拌工程を行わずに微細化処理工程を行うことも可能である。
次に、シモンコライト結晶と分散媒体とを混合して得られた懸濁液を撹拌する。この撹拌工程によって、懸濁液中のシモンコライト結晶が分散媒体に馴化するため、微細化処理の際に、微粒化装置内の流路やノズル等の管路詰まりが生じ難くなり、シモンコライト結晶の微細化処理が容易となる。撹拌手段としては、懸濁液全体を撹拌できるものであればよく、特に限定されないが、例えば、プロペラ型撹拌機やマグネチックスターラ等を用いることができる。撹拌時間としては、分散媒体とシモンコライト結晶とを馴染ませるために、1時間以上が好ましく、2時間以上とすることがより好ましい。また、この撹拌工程において、上述した分散剤を添加し、懸濁液に分散剤を含有させておくことも好ましい。なお、分散媒体として、エタノール等の水混和性有機溶媒を選択した場合には、撹拌工程を行わずに微細化処理工程を行うことも可能である。
(微細化処理工程)
微細化処理工程では、懸濁液を微粒化装置に導入して、懸濁液中のシモンコライト結晶をナノレベルのサイズに微細化する。微粒化装置としては、湿式微粒化装置が使用され、例えば、高圧ホモジナイザー型の湿式微粒化装置やビーズミル型の湿式微粒化装置が用いられる。微細化処理は、シモンコライト結晶が、90%累積粒子径(D90)が800nm以下のナノ粒子となるように行うことが好ましく、50%累積粒子径(D50)が250nm以下のナノ粒子になるように行うことがより好ましく、50%累積粒子径(D50)が150nm以下のナノ粒子とすることがさらに好ましく、90%累積粒子径(D90)が550nm以下かつ50%累積粒子径(D50)が150nm以下のナノ粒子となるように行うことが特に好ましい。微細化処理にあたっては、高圧ホモジナイザー型装置の場合には、使用チャンバー、圧力、ノズル径、パス回数又は処理時間等の微細化処理条件があり、ビーズミル型装置の場合には、使用するビーズの材質、ビーズ径又は処理時間等の微細化処理条件があるが、これらの処理条件は、使用する装置の仕様と、所望の微細化レベルに基づき、適宜設定され得る。また、この微細化処理工程においては、前もって懸濁液中のシモンコライト結晶を大まかに粉砕する予備粉砕処理を行ったのち、結晶をさらに微細化する微細化処理を行ってもよい。予備粉砕処理としては、上述した微粒化装置と同じ装置を用いることができ、本処理としての微細化処理に係る条件よりも処理時間やパス回数、圧力等を少なくしたり、使用チャンバーのノズル径や使用するビーズ径等を大きくするなどして、懸濁液中のシモンコライト結晶を大まかに粉砕処理することが行われる。これにより、50%累積粒子径(D50)が小さく、ナノ粒子径が揃ったシモンコライト結晶を安定的に得ることができる。
微細化処理工程では、懸濁液を微粒化装置に導入して、懸濁液中のシモンコライト結晶をナノレベルのサイズに微細化する。微粒化装置としては、湿式微粒化装置が使用され、例えば、高圧ホモジナイザー型の湿式微粒化装置やビーズミル型の湿式微粒化装置が用いられる。微細化処理は、シモンコライト結晶が、90%累積粒子径(D90)が800nm以下のナノ粒子となるように行うことが好ましく、50%累積粒子径(D50)が250nm以下のナノ粒子になるように行うことがより好ましく、50%累積粒子径(D50)が150nm以下のナノ粒子とすることがさらに好ましく、90%累積粒子径(D90)が550nm以下かつ50%累積粒子径(D50)が150nm以下のナノ粒子となるように行うことが特に好ましい。微細化処理にあたっては、高圧ホモジナイザー型装置の場合には、使用チャンバー、圧力、ノズル径、パス回数又は処理時間等の微細化処理条件があり、ビーズミル型装置の場合には、使用するビーズの材質、ビーズ径又は処理時間等の微細化処理条件があるが、これらの処理条件は、使用する装置の仕様と、所望の微細化レベルに基づき、適宜設定され得る。また、この微細化処理工程においては、前もって懸濁液中のシモンコライト結晶を大まかに粉砕する予備粉砕処理を行ったのち、結晶をさらに微細化する微細化処理を行ってもよい。予備粉砕処理としては、上述した微粒化装置と同じ装置を用いることができ、本処理としての微細化処理に係る条件よりも処理時間やパス回数、圧力等を少なくしたり、使用チャンバーのノズル径や使用するビーズ径等を大きくするなどして、懸濁液中のシモンコライト結晶を大まかに粉砕処理することが行われる。これにより、50%累積粒子径(D50)が小さく、ナノ粒子径が揃ったシモンコライト結晶を安定的に得ることができる。
(溶媒留去工程)
また、分散媒体として、エタノール等の水混和性有機溶媒を選択した場合には、微細化処理後の水混和性有機溶媒スラリーに含まれる水混和性有機溶媒を留去することにより、分散媒体を他の分散媒体、具体的には水に置換できる。具体的には、一例として、微細化処理後のエタノールスラリーに水を加えて減圧環境とし、水蒸気蒸留する。これにより、エタノールが留去され、微細化処理されたシモンコライト結晶が分散された水スラリーを得ることができる。
また、分散媒体として、エタノール等の水混和性有機溶媒を選択した場合には、微細化処理後の水混和性有機溶媒スラリーに含まれる水混和性有機溶媒を留去することにより、分散媒体を他の分散媒体、具体的には水に置換できる。具体的には、一例として、微細化処理後のエタノールスラリーに水を加えて減圧環境とし、水蒸気蒸留する。これにより、エタノールが留去され、微細化処理されたシモンコライト結晶が分散された水スラリーを得ることができる。
上述した微細化処理工程又は留去工程によって得られたスラリーは、そのままスラリーとして育毛用化粧料、化粧料用原料又は化粧料に用いることができるほか、濃縮処理により濃縮液としたもの、乾燥処理を施して微粉末状としたものなども育毛用化粧料、化粧料用原料又は化粧料として用いることが可能である。
なお、育毛用化粧料として、配合成分中のシモンコライト結晶をナノ微粒子としない場合には、分散媒体にシモンコライト結晶と、必要に応じて分散剤を添加し、分散させることによって、育毛用化粧料を得ることも可能である。
本発明の化粧料用原料は、毛包組織の毛髪の伸長を促進し、毛髪の健やかな成長を維持し、健康な毛髪を得るための育毛用、頭皮ケア用及び皮膚外用剤のための原料として用いることができる。そして、本発明の化粧料は、毛包組織の毛髪の伸長を促進し、毛髪の健やかな成長を維持し、健康な毛髪を得るための育毛用、頭皮ケア用及び皮膚外用剤として用いることができる。
本発明の育毛用化粧料の使用量は、目標とする毛の伸長効果、使用方法、年齢や毛髪の状態などによって変化するので一概には規定できないが、シモンコライト結晶として、表皮単位面積(1cm2)当たり、0.01~1000μg/cm2・dayとすることが好ましく、0.1~500μg/cm2・dayとすることがより好ましい。
本発明の育毛用化粧料、化粧料用原料又は化粧料は、通常頭皮や皮膚に適用される剤形として、従来慣用されている方法により種々の外用剤の剤形に適用されることができ、例えば、スラリー、低粘度液体等の液剤、ジェル、乳液、ペースト、クリーム、フォーム、軟膏又は粉剤等が挙げられる。なお、本発明に係る育毛用化粧料、化粧料用原料又は化粧料は、化粧品、医薬部外品又は医薬品のいずれにも適用することができる。具体的な製品としては、特に限定されないが、ヘアトニック、頭皮用化粧水、シャンプー、コンディショナー、トリートメント、整髪料、石鹸、化粧水、化粧クリーム、化粧乳液、美容液、化粧パック、化粧洗浄料、浴用剤又はメーキャップ化粧料等が挙げられる。
また、本発明の育毛用化粧料、化粧料用原料又は化粧料には、本発明の作用効果を損なわない範囲において、通常化粧料や外用剤に用いられる種々の成分を配合することができる。例えば、血管拡張剤、血行促進剤、界面活性剤、抗ヒスタミン剤、抗炎症剤、増粘剤、ビタミン類、アミノ酸類、保湿剤、防腐剤、抗菌剤、香料及び色素などが挙げられる。また、シモンコライト結晶以外の育毛成分や発毛成分を配合することも可能である。他の育毛・発毛成分としては、例えば、ミノキシジル、アデノシン又はセンブリエキス等が挙げられる。
以下、本発明を実施例及び比較例によりさらに詳細に説明する。以下の実施例及び比較例で調製した各サンプル中のシモンコライト結晶の粒子径の測定方法及び各サンプルの評価方法は下記の通りである。
(1)粒子径
実施例及び比較例で得られたサンプルの粒子径分布の測定は、レーザー回折散乱式粒子径分布測定装置(型番:Partica LA-960、株式会社堀場製作所製品)を用い、シモンコライト結晶の屈折率パラメーターを1.700-0.000i、各サンプルにおいてシモンコライト結晶が分散されているイオン交換水又は無水エタノールを分散媒体として測定を行った。得られた粒子径分布から、体積基準の粒子径分布における90%累積粒子径(D90)と50%累積粒子径(D50)を求めた。
実施例及び比較例で得られたサンプルの粒子径分布の測定は、レーザー回折散乱式粒子径分布測定装置(型番:Partica LA-960、株式会社堀場製作所製品)を用い、シモンコライト結晶の屈折率パラメーターを1.700-0.000i、各サンプルにおいてシモンコライト結晶が分散されているイオン交換水又は無水エタノールを分散媒体として測定を行った。得られた粒子径分布から、体積基準の粒子径分布における90%累積粒子径(D90)と50%累積粒子径(D50)を求めた。
(2)ざらつき
実施例及び比較例で得られた各サンプルについて、サンプルに配合した分散媒体(イオン交換水又は無水エタノール)と同じ分散媒体を用いて、シモンコライト結晶の濃度が5wt%になるように希釈し、評価用サンプルをそれぞれ調製した。この評価用サンプルを50μLとり、前腕内側に滴下して指で直径4cmに静かに塗り広げた際のざらつきの感じ方を下記評価基準にて評価した。
◎:ざらつきを全く感じない。
〇:わずかにざらつきを感じる。
△:少しざらつきを感じる。
×:強くざらつきを感じる。
実施例及び比較例で得られた各サンプルについて、サンプルに配合した分散媒体(イオン交換水又は無水エタノール)と同じ分散媒体を用いて、シモンコライト結晶の濃度が5wt%になるように希釈し、評価用サンプルをそれぞれ調製した。この評価用サンプルを50μLとり、前腕内側に滴下して指で直径4cmに静かに塗り広げた際のざらつきの感じ方を下記評価基準にて評価した。
◎:ざらつきを全く感じない。
〇:わずかにざらつきを感じる。
△:少しざらつきを感じる。
×:強くざらつきを感じる。
(3)肌残り
上述の「(2)ざらつき」の評価において、前腕内側に塗り広げた範囲の評価用サンプル液が肌上で乾いてから、白色の肌残りの有無を下記評価基準にて評価した。
◎:白色の肌残りを全く認めない。
〇:白色の肌残りをほとんど認めない。
△:部分的に白色の肌残りをわずかに認める。
×:明確に白色の肌残りを全体に認める。
上述の「(2)ざらつき」の評価において、前腕内側に塗り広げた範囲の評価用サンプル液が肌上で乾いてから、白色の肌残りの有無を下記評価基準にて評価した。
◎:白色の肌残りを全く認めない。
〇:白色の肌残りをほとんど認めない。
△:部分的に白色の肌残りをわずかに認める。
×:明確に白色の肌残りを全体に認める。
(4)分散性
シモンコライト結晶の濃度が5wt%になるように希釈調製した各評価用サンプルを8mLずつ取り、それぞれ10mL容量のバイアル瓶(マイティバイアルNo.3、株式会社マルエム製品)に入れた。バイアル瓶を振とうして、評価用サンプル中のシモンコライト結晶を分散させた後、室温にて18時間静置した。18時間後、シモンコライト結晶の分散性を下記評価基準にて評価した。
◎:シモンコライト結晶の分散状態が保持されている、又は、バイアル瓶を軽く数回振とうするだけでシモンコライト結晶が分散する。
〇:バイアル瓶を振とうすることにより、シモンコライト結晶が分散する。
△:バイアル瓶を激しく振とうすることにより、シモンコライト結晶が分散する。
×:バイアル瓶を激しく振とうしてもシモンコライト結晶が瓶の底部に残存し、分散され難い部分がある。
シモンコライト結晶の濃度が5wt%になるように希釈調製した各評価用サンプルを8mLずつ取り、それぞれ10mL容量のバイアル瓶(マイティバイアルNo.3、株式会社マルエム製品)に入れた。バイアル瓶を振とうして、評価用サンプル中のシモンコライト結晶を分散させた後、室温にて18時間静置した。18時間後、シモンコライト結晶の分散性を下記評価基準にて評価した。

また、以下の実施例及び比較例で使用した配合成分の仕様を表1に示す。
[実施例1]
1.シモンコライトの水スラリーの調製
シモンコライト(JFEミネラル株式会社、ロット番号:S1C231)を32メッシュの篩にて篩過し、シモンコライト篩過品5gを得た。これに分散媒体としてイオン交換水を加え100mLとした後、2分間の超音波照射を行い、シモンコライトの水スラリー(水懸濁液)100mLを得た。このスラリー中のシモンコライト結晶の粒子径は、90%累積粒子径(D90)が9458nm、50%累積粒子径(D50)が5600nmであった。
1.シモンコライトの水スラリーの調製
シモンコライト(JFEミネラル株式会社、ロット番号:S1C231)を32メッシュの篩にて篩過し、シモンコライト篩過品5gを得た。これに分散媒体としてイオン交換水を加え100mLとした後、2分間の超音波照射を行い、シモンコライトの水スラリー(水懸濁液)100mLを得た。このスラリー中のシモンコライト結晶の粒子径は、90%累積粒子径(D90)が9458nm、50%累積粒子径(D50)が5600nmであった。
[実施例2]
2.シモンコライト・ナノ結晶の水スラリーの調製(1)
イオン交換水840gにシモンコライト(JFEミネラル株式会社、ロット番号:TOL81)160gを投入して混合し、プロペラ型撹拌機を用いて撹拌速度300~500rpmで4時間30分間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を高圧ホモジナイザー型の湿式微粒化装置(スターバーストラボ、株式会社スギノマシン製品)に導入し、使用チャンバー:ボール衝突チャンバー、チャンバーノズル径:0.14mm、圧力:245MPa、パス回数:30パスの条件にて微細化処理を施した。その結果、シモンコライト結晶がナノレベルのサイズに微細化された水スラリーを得た。このスラリー中のシモンコライト結晶の粒子径は、90%累積粒子径(D90)が401nm、50%累積粒子径(D50)が133nmであった。
2.シモンコライト・ナノ結晶の水スラリーの調製(1)

[実施例3]
3.シモンコライト・ナノ結晶の水スラリーの調製(2)
実施例2で得られた水スラリー62.5gにイオン交換水を加え200mLとし、よく振り混ぜて、スラリー中のシモンコライト結晶濃度を5w/v%とした希釈スラリーを調製した。

[比較例1]
4.シモンコライト・ナノ結晶の水スラリーの上澄液の調製
シモンコライトは水に不溶であるが、非常に微量の亜鉛イオンが水中に溶出するとされている。そこで、シモンコライト・ナノ結晶の水スラリー中に溶出する亜鉛イオンの影響を調べるため、水スラリーの上澄液を調製した。具体的には、実施例3で得た、シモンコライト結晶濃度が5w/v%の水スラリーを1830×gで30分間遠心分離(型番:AX-521、株式会社トミー精工製品)処理した。上澄み部分を回収し、孔径0.22μmのメンブランフィルター(Millex(登録商標)-GVフィルター、メルク社製品)でろ過し、無色澄明なろ液を水スラリー上澄液として得た。
4.シモンコライト・ナノ結晶の水スラリーの上澄液の調製
シモンコライトは水に不溶であるが、非常に微量の亜鉛イオンが水中に溶出するとされている。そこで、シモンコライト・ナノ結晶の水スラリー中に溶出する亜鉛イオンの影響を調べるため、水スラリーの上澄液を調製した。具体的には、実施例3で得た、シモンコライト結晶濃度が5w/v%の水スラリーを1830×gで30分間遠心分離(型番:AX-521、株式会社トミー精工製品)処理した。上澄み部分を回収し、孔径0.22μmのメンブランフィルター(Millex(登録商標)-GVフィルター、メルク社製品)でろ過し、無色澄明なろ液を水スラリー上澄液として得た。
以下表2に、実施例1~3及び比較例1で調製したサンプルの配合、製造条件、粒子径及び各種評価について示す。
[実施例4]
5.シモンコライト結晶による育毛効果の検討(1)
実施例1、3及び比較例1で調製したサンプルを用いて、ラットの髭毛包の器官培養を行い、シモンコライトの育毛促進効果を調べた。試験は次のようにして行った。7週齢、体重210-230gの雄性SDラット(日本エスエルシー株式会社)を麻酔し、無菌的にヒゲ部皮膚を摘出した。摘出皮膚の髭が生えている1列目及び2列目の中で、1コの毛包から2本生えている毛包(成長期毛包)を選び、実体顕微鏡下で単離した。単離した毛包は、DMEM培地(サーモフィッシャーサイエンティフィック株式会社)に、ペニシリンG(100IU/mL)、ストレプトマイシン(100μg/mL)及びアンフォテリシンB(0.25μg/mL)を添加した培養液で洗浄した。次に、10%FBS含有DMEM培地に対し、実施例1、3及び比較例1で調製したサンプル並びに陽性対照であるミノキシジルをそれぞれ添加した培養液を準備した。下記表3に示すように、10%FBS含有DMEM培地(陰性対照)に、下記表3に示すサンプル及びミノキシジルをそれぞれ所定の濃度となるように添加し、各試験群(試験4a~4e)用の培養液を調製した。

実施例1、3及び比較例1で調製したサンプルを用いて、ラットの髭毛包の器官培養を行い、シモンコライトの育毛促進効果を調べた。試験は次のようにして行った。7週齢、体重210-230gの雄性SDラット(日本エスエルシー株式会社)を麻酔し、無菌的にヒゲ部皮膚を摘出した。摘出皮膚の髭が生えている1列目及び2列目の中で、1コの毛包から2本生えている毛包(成長期毛包)を選び、実体顕微鏡下で単離した。単離した毛包は、DMEM培地(サーモフィッシャーサイエンティフィック株式会社)に、ペニシリンG(100IU/mL)、ストレプトマイシン(100μg/mL)及びアンフォテリシンB(0.25μg/mL)を添加した培養液で洗浄した。次に、10%FBS含有DMEM培地に対し、実施例1、3及び比較例1で調製したサンプル並びに陽性対照であるミノキシジルをそれぞれ添加した培養液を準備した。下記表3に示すように、10%FBS含有DMEM培地(陰性対照)に、下記表3に示すサンプル及びミノキシジルをそれぞれ所定の濃度となるように添加し、各試験群(試験4a~4e)用の培養液を調製した。
6ウェル細胞培養プレートにセルカルチャーインサート(Millicell(登録商標)、メルク社製品)を載せ、各試験群(試験4a~4e)の培養液2mLでメンブレンを下面からそれぞれ湿潤させた。PBSに浸した毛包を、各試験群20本を無作為に選び、ピンセットを用いてメンブレン上に移し、CO2インキュベーター(Napco 7100)に入れ、37℃、5%CO2条件下で培養した。培養は7日間行い、毛の伸長を実体顕微鏡下で毛根部黒色組織の始点から毛切断部までの全体の長さ及び皮膚表面から毛切断部までの毛露出部の長さの変化を観察、記録した。結果を図1に示す。
図1に示すように、試験開始から4日目までは、いずれの試験群も似たような毛の伸長を示したが、試験開始から4日目以降は、実施例1のシモンコライトの試験群(4b)及び実施例3のシモンコライトのナノ結晶の試験群(4c)のみが顕著な毛の伸長を示した。この伸長効果は、陰性対照の試験群(4a)のみならず、陽性対照であるミノキシジルの試験群(4e)と比べても際立っていた。なお、比較例1のシモンコライトの上澄液の試験群(4d)にはこのような際立った効果は認められなかった。これらのことから、実施例1のシモンコライトの試験群(4b)及び実施例3のシモンコライトのナノ結晶の試験群(4c)が示す顕著な毛の伸長効果は、単にシモンコライトと平衡状態になる水相に溶出した亜鉛イオンの効果によるものでなく、水に不溶性の粉末であるシモンコライト結晶に特有の効果であることを示している。
[実施例5]
6.シモンコライト・ナノ結晶スラリーの分散性の検討(1)
微細化されていないシモンコライト(実施例1)の粒子径分布はD50が5.6μm、D90が9.5μmと大きい。そのため、化粧料として皮膚に適用した時、結晶によるざらつきが感じられると共に、塗布後には塗布した箇所が白く残るため、美容上好ましくない。また、実施例2、3で得たシモンコライト・ナノ結晶スラリーはナノ粒子であるため、ざらつき感と白色の肌残りは解消されているが、シモンコライト結晶の凝集沈殿が生じており、使用する際には、シモンコライト結晶が再分散されるまで容器を激しく振とうしなければならず、手間がかかる。そこで、微細化されていないシモンコライトから、分散性が向上したシモンコライト・ナノ結晶スラリーを得るため、下記表4及び表5に示す配合及び製造条件にて各種サンプルを調整し、分散剤の検討を行った。

(試験5-8)
日本薬局方 無水エタノールに、ポリエチレングリコール6000(PEG6000、キシダ化学株式会社製品)が0.8重量%、シモンコライト(JFEミネラル株式会社、ロット番号:T0I71-A5)が16重量%となるように混合し、懸濁液を得た。この懸濁液をビーズミル型の湿式微粒化装置(ラボスターミニLMZ015、アシザワ・ファインテック株式会社製品)に導入し、0.2mm径のPSZビーズを用いて150分間粉砕した後、ポリエチレングリコール6000を最終濃度1.6重量%となるように追加し、0.2mm径のPSZビーズを0.05mm径のPSZビーズに変更して、さらに120分間粉砕することにより、微細化処理を施した。得られたエタノールスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表5に示す。

表4の結果から、分散剤として、セルロース誘導体であるヒドロキシプロピルセルロース及びヒドロキシプロピルメチルセルロースを用いることにより、分散性が向上すると共に、ざらつき感及び白色の肌残りが解消されたシモンコライト・ナノ結晶スラリーが得られることがわかった。また、表5の結果から、分散剤として、ポリエチレングリコールも好適に用いられること、特にポリエチレングリコール6000(PEG6000)が、分散性の向上及びざらつき感・白色の肌残りの解消に好適であることが明らかとなった。他方、分散剤としてポリエチレングリコール1000、プロピレングリコール 1-メトキシ-2-アセテート、エトキシエチルアセテートを用いた場合には、分散性の改善がみられないばかりか、シモンコライト結晶の微細化も不十分であり、ナノレベルのサイズまでの微細化は困難であった。
また、表4及び表5の結果から、シモンコライト結晶を90%累積粒子径(D90)が800nm以下のナノ粒子に微細化することにより、ざらつき感及び白色の肌残りが低減され、使用感に優れるスラリーが得られることが明らかとなった。さらに、90%累積粒子径(D90)が550nm以下かつ50%累積粒子径(D50)が、150nm以下のナノ粒子にまで微細化することにより、ざらつき感及び白色の肌残りが解消され、化粧品としての使用感に非常に優れるスラリーが得られることが示された。
また、微細化処理条件として、高圧ホモジナイザー型の微粒化装置を使用した場合、チャンバー型の違いによる影響はあまり見られず、ボール衝突型チャンバーとスリット型チャンバーのいずれにおいても、同様のレベルまでの微細化処理が行われた。また、パス回数が増えるほど、より小さなサイズに微細化されることが確認された。さらに、微粒化装置として、高圧ホモジナイザー型だけでなく、ビーズミル型の微粒化装置も好適に用いることができることが示された(試験5-8)。
[実施例6]
7.シモンコライト・ナノ結晶スラリーの分散性の検討(2)
分散剤が配合されたサンプルと、分散剤が配合されていないサンプルについて、サンプル調整直後と、同サンプルを室温で1週間保存した後のシモンコライト結晶の粒子径分布を測定した。分散剤が配合されたサンプルとしては、実施例5の試験5-3(分散剤:ヒドロキシプロピルセルロース)で得たエタノールスラリーを用い、分散剤が配合されていないサンプルとしては、実施例2で得た水スラリーを用いた。結果を図2に示す。
7.シモンコライト・ナノ結晶スラリーの分散性の検討(2)
分散剤が配合されたサンプルと、分散剤が配合されていないサンプルについて、サンプル調整直後と、同サンプルを室温で1週間保存した後のシモンコライト結晶の粒子径分布を測定した。分散剤が配合されたサンプルとしては、実施例5の試験5-3(分散剤:ヒドロキシプロピルセルロース)で得たエタノールスラリーを用い、分散剤が配合されていないサンプルとしては、実施例2で得た水スラリーを用いた。結果を図2に示す。
図2から明らかなように、分散剤が配合されていない実施例2のサンプル中のシモンコライト粒子は、1週間後には、200nm以下の粒子がほとんど見られなくなり、10μmの粒子に凝集してしまっていた。他方、分散剤としてヒドロキシプロピルセルロースを配合した実施例5の試験5-3のスラリーサンプルでは、シモンコライトのナノ粒子の粒子径分布の変化がほとんど見られず、サンプル調整直後と同じ粒子径分布を維持していた。このことから、分散剤はスラリーの分散性の向上のみならず、シモンコライト粒子のナノレベルサイズの維持、保存安定性に顕著な効果を示していることが明らかとなった。
[実施例7]
8.シモンコライト・ナノ結晶の水スラリーの製造方法の検討(1)
本発明において、シモンコライト結晶の分散媒体として水を選択した場合、下記試験7-1等に示すように、シモンコライト粒子の微細化処理が困難となる事象が生じやすいことがわかった。そこで、分散媒体として水を選択した際における、シモンコライト・ナノ結晶の水スラリーの製造方法について種々検討を行った。

本発明において、シモンコライト結晶の分散媒体として水を選択した場合、下記試験7-1等に示すように、シモンコライト粒子の微細化処理が困難となる事象が生じやすいことがわかった。そこで、分散媒体として水を選択した際における、シモンコライト・ナノ結晶の水スラリーの製造方法について種々検討を行った。
(試験7-1)
イオン交換水840gにシモンコライト(JFEミネラル株式会社、ロット番号:TOL81)160gを投入して混合し、プロペラ型撹拌機を用いて撹拌速度300~400rpmで3分間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を高圧ホモジナイザー型の湿式微粒化装置(スターバーストラボ、株式会社スギノマシン製品)に導入し、使用チャンバー:ボール衝突チャンバー、チャンバーノズル径:0.14mm、圧力:245MPaの条件にて微細化処理を施そうと試みたが、管路に固着等の詰まりが生じ、微細化処理することが出来なかった。結果を表6に示す。

イオン交換水121.2gに分散剤として、ヒドロキシプロピルセルロース4.8gを溶解し、この溶液にシモンコライト(JFEミネラル株式会社、ロット番号:TOL81)24gを添加し、懸濁液を得た。この懸濁液をプロペラ型撹拌機を用いて撹拌速度500~1000rpmで2時間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を高圧ホモジナイザー型の湿式微粒化装置(スターバーストミニ、株式会社スギノマシン製品)に導入し、使用チャンバー:ボール衝突チャンバー、チャンバーノズル径:0.1mm、圧力:245MPa、パス回数:30パスの条件にて微細化処理を施し、水スラリーを得た。このスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表6に示す。

(試験7-7)
イオン交換水1760gに分散剤として、ヒドロキシプロピルセルロース40gを溶解し、この溶液にシモンコライト(JFEミネラル株式会社、ロット番号:S1J201)200gを添加し、懸濁液を得た。この懸濁液をプロペラ型撹拌機を用いて撹拌速度500~1000rpmで3時間攪拌後一夜静置した。このシモンコライト結晶の水スラリー(水懸濁液)を高圧ホモジナイザー型の湿式微粒化装置(スターバーストラボ、株式会社スギノマシン製品)に導入し、使用チャンバー:スリット型チャンバー、チャンバーノズル径:0.16mm、圧力:150MPa、パス回数:5パスの条件にて予備粉砕処理を行った。予備粉砕処理後、湿式微粒化装置のチャンバーを別のスリット型チャンバー(型番:ESC-101、ノズル径は予備粉砕処理で用いたチャンバーよりも小さいと推測される)に変更し、圧力:150MPa、パス回数:50パスの条件にて微細化処理を施し、水スラリーを得た。このスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表7に示す。

(試験7-8)
イオン交換水765.6gに分散剤として、ポリエチレングリコール6000(PEG-6000P、三洋化成工業株式会社製品)17.4gを溶解し、シモンコライト(JFEミネラル株式会社、ロット番号:S1J201)87gを添加し、懸濁液を得た。この懸濁液をプロペラ型撹拌機を用いて撹拌速度500~1000rpmで2時間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を試験7-7と同様の条件・方法にて予備粉砕処理及び微細化処理を施し、水スラリーを得た。このスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表7に示す。

(試験7-10)
イオン交換水1043gに、分散剤としてヒドロキシプロピルセルロース56gを、防腐剤としてフェノキシエタノール21gを溶解し、シモンコライト(JFEミネラル株式会社、ロット番号:S1J201)280gを添加し、懸濁液を得た。この懸濁液をプロペラ型撹拌機を用いて撹拌速度500~1000rpmで3時間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を試験7-9と同様の条件・方法にて予備粉砕処理及び微細化処理を施し、水スラリーを得た。このスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表7に示す。
イオン交換水1043gに、分散剤としてヒドロキシプロピルセルロース56gを、防腐剤としてフェノキシエタノール21gを溶解し、シモンコライト(JFEミネラル株式会社、ロット番号:S1J201)280gを添加し、懸濁液を得た。この懸濁液をプロペラ型撹拌機を用いて撹拌速度500~1000rpmで3時間攪拌した。撹拌後のシモンコライト結晶の水スラリー(水懸濁液)を試験7-9と同様の条件・方法にて予備粉砕処理及び微細化処理を施し、水スラリーを得た。このスラリーサンプルについて、シモンコライト結晶の粒子径の測定及びサンプルの各種評価を行った。結果を表7に示す。
表6に実施例2の結果も再掲して示す。表6に示すように、撹拌工程を設けていない試験7-1、試験7-4及び7-5は高圧ホモジナイザー内にシモンコライト結晶によると思われる固着が生じ、微細化処理ができない状態を生じた。他方、表6及び表7に示すように、微細化処理工程前に、シモンコライト結晶と水を含む懸濁液を所定時間撹拌する撹拌工程を設けることにより、分散媒体として水を選択した場合においても、スムーズにシモンコライト結晶がナノレベルのサイズに微細化処理できることが明らかとなった(実施例2、試験7-2及び試験7-3、試験7-6~7-10)。なお、分散剤を配合させた場合においても、撹拌工程を経ることにより、微粒化装置が問題なく作動し、シモンコライト結晶がナノレベルのサイズに微細化された(試験7-2及び試験7-3、試験7-6~7-10)。また、分散媒体として水を選択した場合においても、高圧ホモジナイザー型の微粒化装置では、ボール衝突型チャンバー及びスリット型チャンバーのいずれのタイプのチャンバーを用いても良好なレベルまで微細化されることが確認された。さらに、予備粉砕処理として、予め少ないパス回数で大まかな粉砕処理を施した後に、微細化処理を行うことによって、50%累積粒子径(D50)が安定的により小さくなり(150nm以下)、ざらつき、肌残り及び分散性に至るまで、優れた評価のスラリーが得られることが明らかとなった(試験7-7~7-10)。また、スラリー中に防腐剤であるフェノキシエタノールを配合した場合においても、シモンコライト結晶の微細化のレベルには影響を及ぼさず、各種評価も優れたままであった(試験7-10)。
[実施例8]
9.シモンコライト・ナノ結晶の水スラリーの製造方法の検討(2)
シモンコライト・ナノ結晶のエタノールスラリーの分散媒体を水に変換することにより、シモンコライト・ナノ結晶の水スラリーを製造できるかどうか検討を行った。まず、実施例5の試験5-3で得られたエタノールスラリー20gにイオン交換水32gを加えた。これをロータリーエバポレーターにて減圧下、エタノールを水蒸気蒸留により留去し、27.11gの水スラリーを得た。これにイオン交換水を加えて32gとし、シモンコライト・ナノ結晶を10w/w%含有する水スラリーを得た。本品25gをイオン交換水で希釈して50mLとし、シモンコライト・ナノ結晶を5w/v%、ヒドロキシプロピルセルロースを1w/v%それぞれ含有する水スラリーを得た(表8)。
9.シモンコライト・ナノ結晶の水スラリーの製造方法の検討(2)
シモンコライト・ナノ結晶のエタノールスラリーの分散媒体を水に変換することにより、シモンコライト・ナノ結晶の水スラリーを製造できるかどうか検討を行った。まず、実施例5の試験5-3で得られたエタノールスラリー20gにイオン交換水32gを加えた。これをロータリーエバポレーターにて減圧下、エタノールを水蒸気蒸留により留去し、27.11gの水スラリーを得た。これにイオン交換水を加えて32gとし、シモンコライト・ナノ結晶を10w/w%含有する水スラリーを得た。本品25gをイオン交換水で希釈して50mLとし、シモンコライト・ナノ結晶を5w/v%、ヒドロキシプロピルセルロースを1w/v%それぞれ含有する水スラリーを得た(表8)。
表8に示すように、シモンコライト・ナノ結晶のエタノールスラリーから、エタノールを水蒸気蒸留して留去することにより、分散媒体をエタノールから水に変換できることが示された。エタノールを分散媒体として選択する場合には撹拌工程が不要であるので、エタノールスラリーとしてシモンコライト・ナノ結晶スラリーを調製しておき、必要に応じて、水スラリーをエタノール留去によって調製する、というような製造方法が提案され得る。
[実施例9]
10.シモンコライト結晶による育毛効果の検討(2)
実施例8で調製した分散剤が配合されたサンプルを用いて、ラットの髭毛包の器官培養を行い、シモンコライトの育毛促進効果を調べた。試験は上述した実施例4と同様の方法にて行った。各試験群(試験1~3)で用いた培養液を下記表9に示す。培養は7日間行い、毛の伸長を実体顕微鏡下で毛根部黒色組織の始点から毛切断部までの全体の長さ及び皮膚表面から毛切断部までの毛露出部の長さの変化を観察、記録した。結果を図3に示す。
10.シモンコライト結晶による育毛効果の検討(2)
実施例8で調製した分散剤が配合されたサンプルを用いて、ラットの髭毛包の器官培養を行い、シモンコライトの育毛促進効果を調べた。試験は上述した実施例4と同様の方法にて行った。各試験群(試験1~3)で用いた培養液を下記表9に示す。培養は7日間行い、毛の伸長を実体顕微鏡下で毛根部黒色組織の始点から毛切断部までの全体の長さ及び皮膚表面から毛切断部までの毛露出部の長さの変化を観察、記録した。結果を図3に示す。
実施例4の図1の結果同様、図3に示すように、試験開始から5日目以降は、実施例8のシモンコライトのナノ結晶と分散剤を含むサンプルが添加された試験群(試験2)のみが顕著な毛の伸長を示した。この伸長効果は、陰性対照の試験群(試験1)のみならず、陽性対照であるミノキシジルの試験群(試験3)と比べても際立っていた。これらのことから、実施例4同様に、シモンコライトのナノ結晶にも毛の伸長効果を有することが示された。

本発明は、上記の実施形態又は実施例に限定されるものでなく、特許請求の範囲に記載された発明の要旨を逸脱しない範囲内での種々、設計変更した形態も技術的範囲に含むものである。
本発明の育毛用化粧料、化粧料用原料及び化粧料は、優れた育毛効果を有し、使用感及び取り扱いのし易さ、安定性にも優れるため、美容・ヘアケアの分野において幅広く利用されるものである。

- シモンコライト結晶を有効成分として含有することを特徴とする育毛用化粧料。
- 前記シモンコライト結晶の配合量は、0.001重量%~10重量%であることを特徴とする請求項1に記載の育毛用化粧料。
- 前記シモンコライト結晶が、レーザー回折散乱法による体積基準の粒子径分布における90%累積粒子径(D90)が800nm以下のナノ粒子であることを特徴とする請求項1又は2に記載の育毛用化粧料。
- さらに、分散媒体と分散剤を含有し、
前記分散剤が、ポリエチレングリコール、ポリプロピレングリコール、エチレンオキサイド・プロピレンオキサイド共重合体、ヒドロキシプロピルセルロース、ヒドロキシプロピルメチルセルロース及びヒドロキシエチルセルロースからなる群から選択される少なくとも1種の化合物であることを特徴とする請求項1~3のいずれか1項に記載の育毛用化粧料。 - シモンコライト結晶のナノ粒子を含有することを特徴とする化粧料用原料又は化粧料。

- シモンコライト結晶と、水混和性有機溶媒を有する分散媒体を混合して懸濁液を得る混合工程と、
前記懸濁液を微粒化装置に導入し、前記懸濁液に含まれる前記シモンコライト結晶をナノレベルのサイズに微細化する微細化処理工程と、
前記微細化処理後の懸濁液に含まれる前記水混和性有機溶媒を留去する工程と、を有することを特徴とする化粧料用原料又は化粧料の製造方法。 - 前記混合工程において、さらに分散剤を混合して懸濁液を得ることを特徴とする請求項15に記載の化粧料用原料又は化粧料の製造方法。
