Abstract
本発明は、例えば、植物からプロテオグリカンを抽出する工程などを含む植物由来のプロテオグリカンの製造方法、に関する。 溶媒を水としてガディガムを溶解する工程、当該溶解する工程で得られる溶解液の分子量分画を行う工程を含む、分子量が1万より大きく500万以下の植物由来のプロテオグリカンの製造方法。
Description
本出願は、日本国において、2021年11月1日に出願された特願2021-178577号に基づく優先権を主張するものであり、当該出願に記載された内容はすべて参照によりそのまま本明細書に援用される。
本発明は、例えば、植物からプロテオグリカンを抽出する工程などを含む植物由来のプロテオグリカンの製造方法、に関する。
プロテオグリカン(以下「PG」と称する場合がある。)は、1本のコアタンパク質にコンドロイチン硫酸、ケラタン硫酸等のグリコサミノグリカンが数本から数十本、共有結合した糖タンパク質であり、細胞外マトリックスの一つとして皮膚や軟骨など体内に広く分布している。軟骨中のPGは、コラーゲンやヒアルロン酸と共に凝集体を形成しており、代表的な軟骨型PGは、アグリカンと称される。アグリカンはコアタンパク質に大量のグリコサミノグリカン糖鎖が結合すると共に、そのN末端側には、ヒアルロン酸およびリンクタンパク質の結合領域を有する。
グリコサミノグリカンは分岐をもたない長い直鎖構造を持ち、多数の硫酸基とカルボシキル基を持つため負に荷電しており、その電気的反発力のために伸びた形状をとる。また、糖の持つ水親和性により、多量の水を保持し、弾力や衝撃への耐性といった軟骨特有の機能を担っている。さらに、PGには抗炎症作用、ヒアルロン酸合成促進作用、上皮細胞増殖因子(EGF)様作用等多くの生理機能を有することが明らかとなり、食品や化粧品への応用に期待が寄せられている。動物由来のプロテオグリカンとしては、例えば、サケ鼻軟骨、イカ頭部軟骨を由来としたものが知られている。
しかし、動物由来のプロテオグリカンは軟骨に微量に存在する物質であるため、製造コストが高いという問題があった。また、化粧品市場や食品市場においては、動物由来成分よりも植物由来成分の方がイメージに優れるため、動物性プロテオグリカンも植物由来成分で代替することが求められていた(特許文献1)。

本発明は、このような状況下においてなされたものであってその目的とするところは、植物から優れた生理作用を有する植物由来のプロテオグリカンを製造する方法を提供することなど、である。
上記課題を解決するために、本発明者らは、ガディガムから、アラビノガラクタン-プロテインを所定量含む植物由来のプロテオグリカンを効率的及び所定の純度で製造できることを見出し、本発明を完成するに至った。
すなわち、本発明は以下の実施形態を含む。
(1)溶媒を水としてシクンシ科の植物の部位を溶解する工程、
当該溶解する工程で得られる溶解液の分子量分画を行う工程、を含む分子量が1万より大きく500万以下の植物由来のプロテオグリカンの製造方法。
(2)分子量が5万より大きく100万以下の植物由来のプロテオグリカンを製造する、(1)記載の方法。当該分子量の下限は、より好ましくは1000、更に好ましくは20万、更に好ましくは30万、である。当該分子量の上限は、より好ましくは50万、更に好ましくは40万、である。
(3)公称孔径0.1~250μm(より好ましくは0.1~10μm)の精密濾過膜を用いる、(1)又は(2)記載の方法。

本発明の植物由来のプロテオグリカンの製造方法は、植物から優れた生理作用を有する植物由来のプロテオグリカンを製造することができる。

本発明における「植物由来のプロテオグリカン」は、シクンシ科の植物の部位(ガディガムなどの樹液も含む)から抽出等の工程を経て得られるものである。本発明者は、当該植物の部位を用いることで、高純度及び/又は高効率でアラビノガラクタン-プロテイン(AGP)が含有された組成物を製造できると考え、本発明を発明した。
本発明における「植物由来のプロテオグリカン」は、シクンシ科の植物の部位(ガディガムなどの樹液も含む)から抽出等の工程を経て得られるものである。本発明者は、当該植物の部位を用いることで、高純度及び/又は高効率でアラビノガラクタン-プロテイン(AGP)が含有された組成物を製造できると考え、本発明を発明した。
シクンシ科(Quisqualis indica L.)の植物は、中国南部や東南アジアなどに分布し、薬用や観賞用に栽培されている常緑のつる性木本植物である。生育初期には低木状だが、その後、他物に絡みつきながら10mくらい伸長する。
ガティノキは、主にインド中部の森林に自生し、滑らかな白色の樹皮が特徴的な落葉喬木である。
ガティガムは、シクンシ科ガティノキ(Anogeissus latifolia Wallich)の幹の裂傷から分泌される樹液が乾燥固化したもので、別名Indian Gumとも呼ばれる。ガティガムの主成分は、アラビノース、ガラクトース、マンノース、キシロース、及びグルクロン酸を構成糖とする水溶性多糖類で、天然には主にCa、Mg、K塩として存在する。ガディガムは、通常、室温、又はそれ以上の温度条件下で、30質量%程度まで水に溶解する。ガディガムは、約3%のタンパク質を含み、アラビアガムと同様に「多糖タンパク質複合体」を形成する(特許文献2)。
本発明における「植物由来のプロテオグリカン」は、アラビノガラクタン-プロテイン(AGP)も含む。アラビノガラクタン-プロテイン(AGP)は植物組織に普遍的に分布しているプロテオグリカンで、主に細胞壁(細胞外マトリックス)に局在している。AGPは一般的にはヒドロキシプロリン(Hyp)に富むコアタンパク質に、ガラクトース(Gal)とL-アラビノース(L-Ara)に富むアラビノガラクタン(AG)糖鎖が結合している。コアタンパク質の種類も多く、糖鎖の構造も複雑で多様性に富んでいる(非特許文献1)。
本発明で使用する溶媒は、例えば、酢酸、水、クエン酸、アルカリ溶液、界面活性剤溶液などを適宜選択できる。
プロテオグリカン抽出液を粉末セルロース及び/又は吸油マットなどを用いることにより、混入する脂質成分などを簡便に吸着除去することも可能である。例えば、粉末セルロースとしてはセルロースファイバー(日本製紙(株);KCフロックW-50(S)、KCフロックW-50、KCフロックW-100、KCフロックW-100G、KCフロックW-200、KCフロックW-200G、KCフロックW-250、KCフロックW-300G、KCフロックW-400G、NPファイバーW-100F、NPファイバーW-300F、NPファイバーW-10MG2、NPファイバーW-06MG、KCフロックW-50GK、KCフロックW-100GK、レッテンマイヤー社製;VITACEL-L10、VITACEL-L20、VITACEL-L600-30、VITACEL-L90、ARBOCEL-BMW40、VITACEL-L500)などが挙げられる。吸油マットとしては、主にポリプロピレンを原料とする繊維不織布が好ましい。その例として、例えば前田工繊(株)製;油吸着シートSP-1300N(DX)、油吸着シートSP-1100N、日本製紙クレシア(株)製;オイル吸着マットPP-100シリーズ、田中産業(株);ルックリンA-50、ルックリンB-50、その他、ポリオレフィンとポリエステルを原料とする繊維不織布として、日本製紙クレシア(株)製;パワフルECOシリーズ、主にコットンを原料とする吸油マットとして、大王製紙(株);プロワイプ・コットンオイルマットなどが挙げられる。
次に実施例を挙げ、本発明を更に詳しく説明するが、本発明はこれら実施例に何ら制約されるものではない。なお、以下の実施例において、各種成分の添加量を示す数値の単位%は、質量%を意味する。
(製造例1:ガディガム由来のプロテオグリカンを含有する組成物の製造)
以下のように、当該組成物を製造した。

製造例1の組成物に、アラビノガラクタン―プロテイン(AGP)が含有されているかを確認した。AGPに特異的に結合するヤリブ試薬(Biosupplies Australia Pty Ltd、100-2)を用いて、ヤリブ沈殿により当該確認を行った。塩化ナトリウム(NaCl)水溶液ではAGP-ヤリブ試薬複合体として沈殿する性質を利用した。製造例1の組成物を0.15M塩化ナトリウム(NaCl)水溶液に溶解させた。ヤリブ試薬も0.15M塩化ナトリウム(NaCl)水溶液に溶解させた。調整した溶液を等量混合し、4℃で一晩静置した。当該静置後、当該溶液の沈殿を見たところ、赤色の沈殿を確認した。当該確認より、製造例1の組成物に、アラビノガラクタン―プロテイン(AGP)が含有されていることを確認した。この確認により、製造例1の組成物が植物由来のプロテオグリカンを含有していることを確認した。

アミノ酸の分析にて、当該組成物中のタンパク質において、約0.5%のヒドロキシプロリン(Hyp)が含まれことを確認した。ヒドロキシプロリンは、植物性プロテオグリカンの糖鎖の結合点である。

(5)HPLC分析
製造例1の組成物のHPLC分析を行った。製造例1の組成物(乾燥物)で得た品約1gを精密に量り、リン酸緩衝液(pH6.8)を加えて正確に10mLとしたものを試料溶液とした。各試料を0.45μmメンブレンフィルターに通した後、以下の操作条件でHPLCを行い以下検量線から分子量を求めた。
製造例1の組成物のHPLC分析を行った。製造例1の組成物(乾燥物)で得た品約1gを精密に量り、リン酸緩衝液(pH6.8)を加えて正確に10mLとしたものを試料溶液とした。各試料を0.45μmメンブレンフィルターに通した後、以下の操作条件でHPLCを行い以下検量線から分子量を求めた。
また、分子量マーカーとして、Shodex STANDARD P-82(昭和電工社製)を用いて作成した検量線からピークトップの分子量を求めた。なお、分子量5000~800000の範囲の検量線(標準プルラン)を作成した。

図1に、HPLCの結果を示す。図1に示す結果において、カラムの保持時間から計算したピークトップの分子量は、30万から40万であった。
[試験例1]ヒト表皮角化細胞増殖試験
ヒト表皮角化細胞NHEK(成人表皮角化細胞、クラボウ、KK-4109)を、3×103個/wellの細胞数となるように、当該準備した96ウェルプレートに播種し、増殖添加剤を添加した表皮角化細胞増殖基本培地(KBM培地、クラボウ)にて、37℃、5%CO2の環境下で、24時間、当該ヒト表皮角化細胞を培養した。その後、増殖添加剤としてFGF及びBPE(ウシ脳下垂体抽出物)を除いた表皮角化細胞増殖基本培地へ置換し、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、3日間培養した。3日間の培養後、Cell Counting Kit-8を用いて、各群(試料1から試料5の群)のヒト表皮角化細胞数を測定した。当該測定は、各群(試料1から試料5の群)において5サンプルずつ行った。以下表1では、各群において5サンプルの平均値を算出した結果を用いて、試料1の群の平均値の値を100としたときの相対値を示す。表1の「**」は、試料1の群の値と比べてのDunnett’s testによる有意差(p<0.05)、を示す。

なお、市販のアラビアゴム含有組成物として、アラビアゴム(富士フイルム和光純薬、016-00025、CAS RNTM:9000-01-5)を用いた。
表1に示す結果より、製造例1で得られた組成物を添加した群(試料2及び3の群)では、市販のアラビアゴム含有組成物の添加群(試料4及び5の群)と比較して、ヒト表皮角化細胞増殖能が高いことが確認できた。
[試験例2]ヒト線維芽細胞増殖試験
製造例1で得られた組成物についてのヒト線維芽細胞増殖能の有無を評価した。5%FBS(Thermo Trace製)を含むDMEM培地が各ウェルに存在した96ウェルプレートを準備した。正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに4×103個/wellの細胞数となるように、当該準備した96ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、当該正常ヒト皮膚線維芽細胞を培養した。その後、5%FBS(Thermo Trace製)を含むDMEM培地から0.25%FBSを含むDMEM培地へ置換して、置換後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、3日間、当該正常ヒト皮膚線維芽細胞を培養した。3日間の培養後、Cell Counting Kit-8(DOJINDO)を用いて、各群(試料1から試料5の群)の正常ヒト皮膚線維芽細胞数を測定した。当該測定は、各群(試料1から試料7の群)において5サンプルずつ行った。以下表2では、各群において5サンプルの平均値を算出した結果を用いて、試料1の群の平均値の値を100としたときの相対値を示す。表2の「**」は、試料1の群の値と比べての Tukey-kramer testによる有意差(p<0.05)、を示す。表2の「(††)」は、試料7の値と比べてのTukey-kramer testによる有意差(p<0.05)、を示す。
製造例1で得られた組成物についてのヒト線維芽細胞増殖能の有無を評価した。5%FBS(Thermo Trace製)を含むDMEM培地が各ウェルに存在した96ウェルプレートを準備した。正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに4×103個/wellの細胞数となるように、当該準備した96ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、当該正常ヒト皮膚線維芽細胞を培養した。その後、5%FBS(Thermo Trace製)を含むDMEM培地から0.25%FBSを含むDMEM培地へ置換して、置換後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、3日間、当該正常ヒト皮膚線維芽細胞を培養した。3日間の培養後、Cell Counting Kit-8(DOJINDO)を用いて、各群(試料1から試料5の群)の正常ヒト皮膚線維芽細胞数を測定した。当該測定は、各群(試料1から試料7の群)において5サンプルずつ行った。以下表2では、各群において5サンプルの平均値を算出した結果を用いて、試料1の群の平均値の値を100としたときの相対値を示す。表2の「**」は、試料1の群の値と比べての Tukey-kramer testによる有意差(p<0.05)、を示す。表2の「(††)」は、試料7の値と比べてのTukey-kramer testによる有意差(p<0.05)、を示す。

なお、市販のアラビアゴム含有組成物として、アラビアゴム(富士フイルム和光純薬、016-00025、CAS RNTM:9000-01-5)を用いた。
表2に示す結果より、製造例1で得られた組成物を添加した群(試料2及び3の群)では、市販のアラビアゴム含有組成物の添加群(試料4及び5の群)と比較して、ヒト線維芽細胞増殖能が高いことが確認できた。また、試料7の群に比べ、試料4の群では、ヒト線維芽細胞増殖能が有意に高いことが確認できた。
[試験例3]1型コラーゲン産生促進活性(ELISAによる検出)
製造例1で得られた組成物についての1型コラーゲン産生促進活性の有無をELISAにより評価した。
5%FBS(Thermo Trace製)を含むDMEM培地が各ウェルに存在した24ウェルプレートを準備した。正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに2×104個/wellの細胞数となるように、当該準備した24ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、コンフルエントになるまで当該正常ヒト皮膚線維芽細胞を培養した。その後、5%FBS(Thermo Trace製)を含むDMEM培地から0.25%FBSを含むDMEM培地へ置換して、置換後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、1日間、当該正常ヒト皮膚線維芽細胞を培養した。当該培養後の細胞培養上清に対して、PIP EIA KIT(タカラバイオ)を用いて、各群の1型コラーゲンの産生量(ng/mL)を評価した。当該測定は、各群(試料1から試料3の群)において3サンプルずつ行った。以下表4では、各群において3サンプルの平均値を算出した結果を示す。表3の「**」は、試料1の群の平均値と比べてのDunnett’s testによる有意差(p<0.05)、を示す。
製造例1で得られた組成物についての1型コラーゲン産生促進活性の有無をELISAにより評価した。
5%FBS(Thermo Trace製)を含むDMEM培地が各ウェルに存在した24ウェルプレートを準備した。正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに2×104個/wellの細胞数となるように、当該準備した24ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、コンフルエントになるまで当該正常ヒト皮膚線維芽細胞を培養した。その後、5%FBS(Thermo Trace製)を含むDMEM培地から0.25%FBSを含むDMEM培地へ置換して、置換後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、1日間、当該正常ヒト皮膚線維芽細胞を培養した。当該培養後の細胞培養上清に対して、PIP EIA KIT(タカラバイオ)を用いて、各群の1型コラーゲンの産生量(ng/mL)を評価した。当該測定は、各群(試料1から試料3の群)において3サンプルずつ行った。以下表4では、各群において3サンプルの平均値を算出した結果を示す。表3の「**」は、試料1の群の平均値と比べてのDunnett’s testによる有意差(p<0.05)、を示す。
・試料1の群:コントロールとして、所定量の精製水を添加。
・試料2の群:最終濃度10μg/mLとなるように、製造例1で得られた組成物を添加。
・試料3の群:最終濃度100μg/mLとなるように、製造例1で得られた組成物を添加。

以下表3で示すように、製造例1で得られた組成物を投与することにより、1型コラーゲン産生促進活性が向上されることが確認できた。
[試験例4]1型コラーゲン遺伝子発現(qRT-PCRによる検出)
製造例1で得られた組成物についての1型コラーゲン遺伝子発現をqRT-PCRにより評価した。
正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに2×104個/wellの細胞数となるように、当該準備した24ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、コンフルエントになるまで当該正常ヒト皮膚線維芽細胞を培養した。その後、DMEM培地から試験用DMEM培地(試験例2と異なり血清を含有しない培地)へ置換してさらに24時間培養した。培養後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、24時間、当該正常ヒト皮膚線維芽細胞を培養した。RNeasy Mini Kit (QIAGEN)のプロトコルに従って、当該培養した細胞を回収し、mRNAを精製した。精製したmRNAはPrime Script RT Master Mix (TaKaRa Bio)のプロトコルに従って逆転写反応を行い、反応生成物に対してTB Green Premix EX Taq II (TaKaRa Bio)を使用してRT-PCR反応を行った。RT-PCRはLightCycler96システム(Roche)を用いて行い、付属のソフトウェアを用いて目的遺伝子の発現量を定量化した。当該測定は、各群(試料1から試料3の群)において3サンプルずつ行った。以下表4では、各群において3サンプルの平均値を算出した結果を用いて、試料1の群の平均値の値を100としたときの相対値を示す。表4の「**」は、試料1の群の値と比べてのDunnett’s testによる有意差(p<0.05)、を示す。

正常ヒト皮膚線維芽細胞(クラボウ)を、各ウェルに2×104個/wellの細胞数となるように、当該準備した24ウェルプレートに播種した。この播種後、37℃、5%CO2の環境下で、24時間、コンフルエントになるまで当該正常ヒト皮膚線維芽細胞を培養した。その後、DMEM培地から試験用DMEM培地(試験例2と異なり血清を含有しない培地)へ置換してさらに24時間培養した。培養後、以下試料を各ウェルに投与し、37℃、5%CO2の環境下で、24時間、当該正常ヒト皮膚線維芽細胞を培養した。RNeasy Mini Kit (QIAGEN)のプロトコルに従って、当該培養した細胞を回収し、mRNAを精製した。精製したmRNAはPrime Script RT Master Mix (TaKaRa Bio)のプロトコルに従って逆転写反応を行い、反応生成物に対してTB Green Premix EX Taq II (TaKaRa Bio)を使用してRT-PCR反応を行った。RT-PCRはLightCycler96システム(Roche)を用いて行い、付属のソフトウェアを用いて目的遺伝子の発現量を定量化した。当該測定は、各群(試料1から試料3の群)において3サンプルずつ行った。以下表4では、各群において3サンプルの平均値を算出した結果を用いて、試料1の群の平均値の値を100としたときの相対値を示す。表4の「**」は、試料1の群の値と比べてのDunnett’s testによる有意差(p<0.05)、を示す。

[実験5]ヒトモニター試験による肌状態改善作用の評価
2021年12月から2022年1月の期間で、30歳以上の健康な男女12名(平均年齢49.8歳)を被験者として、このモニター試験を行った。なお、以下表5で示すローションでは、製造例1で得られた組成物を最終濃度1質量%となるように、30%1,3-ブチレングリコールに溶解した溶液(表5では、製造例1で得られた組成物の溶液と記載)を用いた。

(試験方法)
表5で示す組成のローション(表5ローション)を被験者の顔の左半分に所定間隔にて塗布し、表6に示すローション(表6ローション)を被験者の顔の右半分に所定間隔にて塗布した。この所定間隔は、1日2回ずつ(朝晩)4週間である。4週間後には、被験者1人あたり、表5ローションが24g及び表6ローションが24gの量が塗布された。そして、これらのローションの塗布前の0日と、ローションを塗布してから4週間目に、測定(シワの有無の測定など)を行った。
表5で示す組成のローション(表5ローション)を被験者の顔の左半分に所定間隔にて塗布し、表6に示すローション(表6ローション)を被験者の顔の右半分に所定間隔にて塗布した。この所定間隔は、1日2回ずつ(朝晩)4週間である。4週間後には、被験者1人あたり、表5ローションが24g及び表6ローションが24gの量が塗布された。そして、これらのローションの塗布前の0日と、ローションを塗布してから4週間目に、測定(シワの有無の測定など)を行った。
(シワの有無の測定)
ANTERA 3DTM(ミラベックス)を用いて、被験者12名の目尻のシワの有無の測定及びほうれい線の有無の測定を行った。以下、「目尻のシワの測定」及び「ほうれい線のシワの測定」において、当該12名の平均値(当該水分分布の測定の平均値)を記載する。

表5ローション塗布群及び表6ローション塗布群の0日(当該塗布前)の値を100とした場合、当該4週間後において、表6ローション塗布群は103.99であったが、表5ローション塗布群は96.59(0日の表5ローション塗布群の値と比べp<0.05(Wilcoxon検定))であった。

(ほうれい線のシワの測定)
表5ローション塗布群及び表6ローション塗布群の0日(当該塗布前)の値を100とした場合、当該4週間後において、表6ローション塗布群は99.69であったが、表5ローション塗布群は98.10(0日の表5ローション塗布群の値と比べp<0.05(Wilcoxon検定))であった。

半顔(下眼瞼部から頬全体の部位)のシミの個数を測定することによってシミの有無の測定を行った。当該測定は、VISIATMEvolution(VISIA-Evo、Canfield Scientific)を用いて行った。表5ローション塗布群及び表6ローション塗布群の0日(当該塗布前)の値を100とした場合、当該4週間後において、表6ローション塗布群は103.78であったが、表5ローション塗布群は97.79であった。

鼻翼側部においてのDermaLab(登録商標)Comboを用いて、真皮コラーゲン密度の測定を行った。表5ローション塗布群及び表6ローション塗布群の0日(当該塗布前)の値を100とした場合、当該4週間後において、表6ローション塗布群は94.07であったが、表5ローション塗布群は97.22(0日の表5ローション塗布群の値と比べp<0.01(Wilcoxon検定))であった。
鼻翼側部においてのDermaLab(登録商標)Comboを用いて、真皮コラーゲン密度の測定を行った。表5ローション塗布群及び表6ローション塗布群の0日(当該塗布前)の値を100とした場合、当該4週間後において、表6ローション塗布群は94.07であったが、表5ローション塗布群は97.22(0日の表5ローション塗布群の値と比べp<0.01(Wilcoxon検定))であった。
Claims (3)
- 溶媒を水としてシクンシ科の植物の部位を溶解する工程、
当該溶解する工程で得られる溶解液の分子量分画を行う工程、
を含む、分子量が1万より大きく500万以下の植物由来のプロテオグリカンの製造方法。 - 分子量が5万より大きく100万以下の植物由来のプロテオグリカンを製造する、請求項1記載の方法。
- 分子量が20万から50万の植物由来のプロテオグリカンを製造する、請求項2記載の方法。
