Description
본 발명은 플루오렌 유도체 화합물의 합성방법에 관한 것이다.
종래 9,9-비스(4-(2-히드록시에톡시)페닐)플루오렌 화합물을 합성하기 위한 하기 반응식 1의 반응에 대해, 반응 촉매로서 염산(HCl), 황산/티올계 화합물(thiophenol, thioacetic acid, mercaptopropionic acid 등), Ti4+-mont(Ti4+-exchanged montmorillonite) 등이 사용되었다.

반응 촉매로 염산을 사용하는 경우, 염산 사용으로 인한 유해성 및 다단계 공정에 따른 수율 저하의 문제점이 있었고, 반응 촉매로 황산과 티올계 화합물을 사용하는 경우, 티올계 화합물로 인한 공해 발생, 페녹시에탄올(phenoxyethanol)의 과량 사용으로 인한 폐기물 발생과 경제성 저하, 높은 반응온도 등의 문제점이 있었으며, 반응 촉매로 Ti4+-mont(Ti4+-exchanged montmorillonite)를 사용하는 경우에는 페녹시에탄올(phenoxyethanol)의 과량 사용으로 인한 폐기물 발생과 경제성 저하, 높은 반응온도 등의 문제점뿐만 아니라 촉매 제조 공정의 어려움 등의 문제점이 있었다.
본 발명의 목적은 플루오렌 유도체 제조에 있어 기존에 사용된 티올계 화합물 촉매의 사용을 배제할 수 있고, 페녹시에탄올 사용량을 감소시킬 수 있으며, 상대적으로 저온에서 반응을 진행할 수 있어서, 공해유발 물질을 배제하는 한편 폐기물 발생과 에너지 소비를 감소시켜 제조비용 절감이 가능한 플루오렌 유도체의 합성 방법을 제공하는 것이다.
본 발명의 일 실시예에 따른 플루오렌 유도체의 합성방법은 플루오레논과 그라파이트를 혼합한 후 반응온도까지 승온하는 제1 단계; 여기에 메탄설폰산 및 페녹시에탄올을 첨가하여 하기 반응식 1에 따라 플루오레논과 페녹시에탄올을 반응시켜 화합물 1의 플루오렌 유도체를 합성하는 제2 단계; 및 상기 화합물 1의 플루오렌 유도체를 수득하는 제3 단계를 포함하고, 상기 그라파이트 및 상기 메타설폰산은 상기 반응식 1의 반응의 반응 촉매로 작용할 수 있다.

일 실시예에 있어서, 상기 그라파이트 및 상기 메타설폰산은 반응 촉매로 작용할 수 있다.
일 실시예에 있어서, 상기 메타설폰산은 상기 플루오레논 1 mol을 기준으로 8.0 내지 13.0 mol로 사용될 수 있다. 이 경우, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 40 내지 70 g이 사용될 수 있다.
일 실시예에 있어서, 상기 반응식 1의 반응에서, 상기 페녹시에탄올은 상기 플루오레논 1 mol을 기준으로 2.0 내지 3.5 mol로 사용될 수 있다.

본 발명의 플루오렌 유도체의 합성방법에 따르면, 반응 촉매로서 그라파이트(Graphite)와 메탄설폰산(methanesulfonic acid)을 사용함으로써, 티올계 화합물 사용으로 인한 공해 유발 유해물질을 적용하지 않을 수 있고, 플루오레논 대비 페녹시에탄올 사용량을 감축시킬 수 있어서 원료비 감소 및 폐기물 발생 감소로 인한 경제성 향상을 기대할 수 있으며, 약 30 내지 50 ℃의 상대적인 저온 반응을 통해 부산물 생성 억제 및 정제 효율 증가, 그리고 에너지 소비 감소가 가능하다.
도 1a 및 도 1b는 실시예 1에 따라 합성된 FBPE의 1H NMR (in DMSO-d6) 분석 결과 및 HPLC 순도 분석 결과이다.

이하, 첨부한 도면을 참조하여 본 발명의 실시예에 대해 상세히 설명한다. 본 발명은 다양한 변경을 가할 수 있고 여러 가지 형태를 가질 수 있는 바, 특정 실시예들을 도면에 예시하고 본문에 상세하게 설명하고자 한다. 그러나 이는 본 발명을 특정한 개시 형태에 대해 한정하려는 것이 아니며, 본 발명의 사상 및 기술 범위에 포함되는 모든 변경, 균등물 내지 대체물을 포함하는 것으로 이해되어야 한다. 각 도면을 설명하면서 유사한 참조부호를 유사한 구성요소에 대해 사용하였다.

본 발명의 일 실시예에 따른 플루오렌 유도체의 합성방법은, 플루오레논과 그라파이트를 혼합한 후 반응온도까지 승온하는 제1 단계; 여기에 메탄설폰산 및 페녹시에탄올을 첨가하여 하기 반응식 1에 따라 플루오레논과 페녹시에탄올을 반응시켜 화합물 1의 플루오렌 유도체를 합성하는 제2 단계; 및 상기 화합물 1의 플루오렌 유도체를 수득하는 제3 단계를 포함할 수 있다.
[반응식 1]
상기 제1 단계에 있어서, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 40 내지 70 g이 혼합될 수 있다. 예를 들면, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 45 내지 65 g 또는 약 50 내지 60 g이 사용될 수 있다. 그리고 상기 반응 온도는 약 30 내지 60℃일 수 있다. 예를 들면, 상기 반응 온도는 약 35 내지 50℃일 수 있다.
상기 제2 단계에 있어서, 상기 페녹시에탄올은 약 4.0 당량 이하로 사용될 수 있다. 일 실시예로, 상기 페녹시에탄올은 상기 플루오레논 1 mol을 기준으로 약 2.0 내지 3.5 mol 또는 약 2.5 내지 3.0 mol이 사용될 수 있다.
그리고 상기 메타설폰산은 상기 플루오레논 1 mol을 기준으로 약 8.0 내지 13.0 mol 또는 약 9.0 내지 11.5 mol로 사용될 수 있다. 한편, 상기 제2 단계의 반응은 약 8 내지 20 시간 또는 약 10 내지 15 시간 동안 교반하면서 수행될 수 있다.
상기 제2 단계의 반응에 있어서, 상기 그라파이트 및 상기 메타설폰산은 반응 촉매로 작용할 수 있다.
상기 제3 단계에 있어서, 상기 화합물 1의 플루오렌 유도체를 수득하기 위해 상기 제2 단계의 반응물로부터 여과 공정을 통해 그라파이트를 제거한 후 여과된 반응액을 정제수로 세정하여 수용액층을 폐기하고, 이어서 이의 pH를 중성으로 조절하여 얻어진 유기층을 감압하여 고체를 수득한 후 정제수를 추가하여 약 60 내지 80℃에서 슬러리 교반을 하고 상온에서 여과하여 흰색 고체 형태의 화합물 1의 플루오렌 유도체를 수득할 수 있다. 한편, 보다 고순도의 화합물 1의 플루오렌 유도체를 수득하기 위해, 상기 희색 고체를 추가적으로 정제할 수도 있다.
본 발명의 다른 실시예에 따른 플루오렌 유도체의 합성방법은, 플루오레논과 그라파이트를 혼합한 후 반응온도까지 승온하는 제1 단계; 여기에 메탄설폰산 및 나프톡시에탄올을 첨가하여 하기 반응식 2에 따라 플루오레논과 나프톡시에탄올을 반응시켜 화합물 2의 플루오렌 유도체를 합성하는 제2 단계; 및 상기 화합물 2의 플루오렌 유도체를 수득하는 제3 단계를 포함할 수 있다.
[반응식 2]
상기 제1 단계에 있어서, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 40 내지 70 g이 혼합될 수 있다. 예를 들면, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 45 내지 65g 또는 약 50 내지 60g이 사용될 수 있다. 그리고 상기 반응 온도는 약 30 내지 60℃일 수 있다. 예를 들면, 상기 반응 온도는 약 35 내지 50℃일 수 있다.
상기 제2 단계에 있어서, 상기 나프톡시에탄올은 4.0 당량 이하로 사용될 수 있다. 일 실시예로, 상기 나프톡시에탄올은 상기 플루오레논 1mol을 기준으로 약 2.0 내지 3.5 mol 또는 약 2.5 내지 3.0 mol이 사용될 수 있다. 그리고 상기 메타설폰산은 상기 플루오레논 1mol을 기준으로 약 8.0 내지 13.0 mol 또는 약 9.0 내지 11.5 mol로 사용될 수 있다. 한편, 상기 제2 단계의 반응은 약 8 내지 20 시간 또는 약 10 내지 15 시간 동안 교반하면서 수행될 수 있다.
상기 제2 단계의 반응에 있어서, 상기 그라파이트 및 상기 메타설폰산은 반응 촉매로 작용할 수 있다.
상기 제3 단계에 있어서, 상기 화합물 2의 플루오렌 유도체를 수득하기 위해 상기 제2 단계의 반응물로부터 여과 공정을 통해 그라파이트를 제거한 후 여과된 반응액을 정재수로 린싱하여 수용액층을 폐기하고, 이어서 이의 pH를 중성으로 조절하여 얻어진 유기층을 감압하여 고체를 수득한 후 정제수를 추가하여 약 60 내지 80℃에서 슬러리 교반을 하고 상온에서 여과하여 흰색 고체 형태의 화합물 2의 플루오렌 유도체를 수득할 수 있다. 한편, 보다 고순도의 화합물 2의 플루오렌 유도체를 수득하기 위해, 상기 희색 고체를 추가적으로 정제할 수도 있다.
본 발명의 또다른 실시예에 따른 플루오렌 유도체의 합성방법은, 플루오레논과 그라파이트를 혼합한 후 반응온도까지 승온하는 제1 단계; 여기에 메탄설폰산 및 비페닐옥시에탄올을 첨가하여 하기 반응식 3에 따라 플루오레논과 바이페닐옥시에탄올을 반응시켜 화합물 3의 플루오렌 유도체를 합성하는 제2 단계; 및 상기 화합물 3의 플루오렌 유도체를 수득하는 제3 단계를 포함할 수 있다.
[반응식 3]
상기 제1 단계에 있어서, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 40 내지 70 g이 혼합될 수 있다. 예를 들면, 상기 그라파이트는 상기 플루오레논 100 g을 기준으로 약 45 내지 65 g 또는 약 50 내지 60 g이 사용될 수 있다. 그리고 상기 반응 온도는 약 30 내지 60℃일 수 있다. 예를 들면, 상기 반응 온도는 약 35 내지 50℃일 수 있다.

그리고 상기 메타설폰산은 상기 플루오레논 1 mol을 기준으로 약 8.0 내지 13.0 mol 또는 약 9.0 내지 11.5 mol로 사용될 수 있다. 한편, 상기 제2 단계의 반응은 약 8 내지 20 시간 또는 약 10 내지 15 시간 동안 교반하면서 수행될 수 있다.
상기 제2 단계의 반응에 있어서, 상기 그라파이트 및 상기 메타설폰산은 반응 촉매로 작용할 수 있다.
상기 제3 단계에 있어서, 상기 화합물 3의 플루오렌 유도체를 수득하기 위해 상기 제2 단계의 반응물로부터 여과 공정을 통해 그라파이트를 제거한 후 여과된 반응액을 정제수로 세정하여 수용액층을 폐기하고, 이어서 이의 pH를 중성으로 조절하여 얻어진 유기층을 감압하여 고체를 수득한 후 정제수를 추가하여 약 60 내지 80℃에서 슬러리 교반을 하고 상온에서 여과하여 흰색 고체 형태의 화합물 3의 플루오렌 유도체를 수득할 수 있다. 한편, 보다 고순도의 화합물 3의 플루오렌 유도체를 수득하기 위해, 상기 희색 고체를 추가적으로 정제할 수도 있다.
본 발명의 플루오렌 유도체의 합성방법에 따르면, 반응 촉매로서 그라파이트(Graphite)와 메탄설폰산(methanesulfonic acid)을 사용함으로써, 티올계 화합물 사용으로 인한 공해 유발 유해물질을 적용하지 않을 수 있고, 플루오레논 대비 페녹시에탄올 사용량을 감축시킬 수 있어서 원료비 감소 및 폐기물 발생 감소로 인한 경제성 향상을 기대할 수 있으며, 약 30 내지 50 ℃의 상대적인 저온 반응을 통해 부산물 생성 억제 및 정제 효율 증가, 그리고 에너지 소비 감소를 기대할 수 있다.
한편, 상기 그라파이트는 회수하여 약 10회까지 재사용이 가능하고, 이에 따라 본 발명에 따라 플루오렌 유도체를 합성하는 경우 폐기물을 저감시킬 수 있을 뿐만 아니라 경제성을 향상시킬 수 있다.
이하 본 발명의 실시예에 대해 상술한다. 다만, 하기 실시예는 본 발명의 실시형태 중 하나에 불과한 것으로서, 본 발명의 범위가 하기 실시예에 한정되는 것은 아니다.
[실시예1]
온도계, 교반기가 구비된 1구 플라스크에 9-플루오레논 5 g (0.028 mol), 그라파이트 2.8 g (0.231 mol), 메탄설폰산 27.4 g (0.285 mol) 및 2-페녹시에탄올 11.6 g (0.084 mol)을 첨가하였다. 반응 온도를 40℃로 승온한 뒤 12시간 교반하면서 9-플루오레논과 2-페녹시에탄올을 반응시켰다.
반응 종료 후, 반응물에 클로로폼(80 g)을 첨가하고 10분간 교반한 뒤 여과를 통해 그라파이트를 제거하였다.
여과된 반응액에 정제수(250 g)으로 씻어준 뒤 수용액층은 폐기하고 포화 NaHCO3 수용액 (150 g)으로 씻어 pH를 중성으로 조정하였고, 이로부터 얻어진 유기층을 감압하여 고체를 얻어낸 뒤 정제수(80 g)를 추가하여 70 ℃에서 2시간 동안 슬러리(slurry) 교반한 뒤 상온에서 여과하여 흰색 고체 (10.1 g 수율 83%, 순도 96%)를 얻었다.
얻어진 고체에 메탄올 (70 g)을 첨가하고 80 ℃에서 교반하여 균일 용액으로 만든 뒤 상온까지 천천히 냉각시켜 흰색 고체 형태의 9,9-비스(4-(2-히드록시에톡시)페닐)플루오렌을 얻었다 (수율 = 72%, 순도 = 99.7%).
[비교예]
종래 기술 1(Chemistry Letters 1998, 1055)에 의하면 머캅도프로피온산 촉매와 메탄설폰산 용매 하에 페녹시엔탄올을 4.0 당량 사용한 경우 65 ℃ 고온에서도 반응이 진행되지 않는다고 보고하였다. 또한 종래 기술 2(PCT/JP2013/055177) 역시 메탄설폰산을 이용하지만 머캅토프로피온산 사용은 물론 페녹시에탄올을 4.0 당량 사용하고 있다.
이에 제안된 기술이 메탄설폰산을 사용하고 있기 때문에 그라파이트 사용량, 반응 온도 및 시간에 따른 반응 최적화 실험을 진행하였다. 이때 9-플루오레논을 5 g 기준으로 진행하였다.
[실시예 2]
온도계, 교반기가 구비된 1구 플라스크에 9-플루오레논 5 g (0.028 mol), 그라파이트 2.77 g (0.231 mol), 메탄설폰산 27.38 g (0.285 mol) 및 2-(2-나프톡시)엔탄올 18.0 g (0.084 mol)을 첨가하였다. 반응 온도를 40℃로 승온한 뒤 16시간 교반하였다.
반응 종료 후, 반응물에 클로로폼 (80 g)과 메탄올 (10 g)을 첨가하고 10분간 교반한 뒤 여과를 통해 그라파이트를 제거하였다.
여과된 반응액에 정제수 (250 g)로 씻어준 뒤 수용액층은 폐기하고 포화 NaHCO3 수용액 (150 g)으로 씻어 pH를 중성으로 조정하였고, 이로부터 얻어진 유기층을 감압하여 고체를 얻어낸 뒤 메탄올 (70 g)과 정제수 (140 g)를 추가하여 70 ℃에서 2시간 동안 슬러리(slurry) 교반한 뒤 상온에서 여과하여 흰색 고체 (13.8 g 수율 84%, 순도 94%)를 얻었다.
얻어진 고체에 클로로폼 (35 g)과 메탄올 (100 g)을 첨가하고 80 ℃에서 교반하여 균일 용액으로 만든 뒤 상온까지 천천히 냉각시켜 흰색 고체 형태의 화합물 2의 플루오렌 유도체를 얻었다 (수율 = 78%, 순도 = 99.5%).
[실시예3]
온도계, 교반기가 구비된 1구 플라스크에 9-플루오레논 5 g (0.028 mol), 그라파이트 2.77 g (0.231 mol), 메탄설폰산 27.38 g (0.285 mol) 및 바이페닐옥시에탄올 18.0 g (0.084 mol)을 첨가하였다. 반응 온도를 40℃로 승온한 뒤 16시간 교반하였다.
반응 종료 후, 반응물에 클로로폼 (80 g)과 메탄올 (10 g)을 첨가하고 10분간 교반한 뒤 여과를 통해 그라파이트를 제거하였다.
여과된 반응액에 정제수 (250 g)으로 씻어준 뒤 수용액층은 폐기하고 포화 NaHCO3 수용액 (150 g)으로 씻어 pH를 중성으로 조정하였고, 이로부터 얻어진 유기층을 감압하여 고체를 얻어낸 뒤 메탄올 (70 g)과 정제수 (140 g)를 추가하여 70 ℃에서 2시간 동안 슬러리(slurry) 교반한 뒤 상온에서 여과하여 흰색 고체를 얻었다.
얻어진 고체에 클로로폼 (35 g)과 메탄올 (100 g)을 첨가하고 80 ℃에서 교반하여 균일 용액으로 만든 뒤 상온까지 천천히 냉각시켜 흰색 고체 형태의 화합물 3의 플루오렌 유도체를 얻었다 (수율 = 80%, 순도 = 99.8%).
[실험예]
도 1a 및 도 1b는 실시예 1에 따라 합성된 FBPE의 1H NMR (in DMSO-d6) 분석 결과 및 HPLC 순도 분석 결과이다.
도 1a 및 도 1b를 참조하면, 실시예 1에 따를 경우에 화합물 1의 플루오렌 유도체인 FBPE가 고순도로 높은 수율로 합성됨을 확인할 수 있다.
하기 표 1은 실시예 1에 따른 반응에서 그라파이트 및 페녹시 에탄올의 사용량을 변경하면서 측정된 반응 온도, 반응 시간, 수율을 나타낸다.

도 3a 및 도 3b는 실시예 3에 따라 합성된 화합물 3의 1H NMR (in DMSO-d6) 분석 결과 및 HPLC 순도 분석 결과이다.
도 3a 및 도 3b를 참조하면, 실시예 3에 따를 경우에 화합물 3의 플루오렌 유도체가 고순도로 높은 수율로 합성됨을 확인할 수 있다.
상기에서는 본 발명의 바람직한 실시예를 참조하여 설명하였지만, 해당 기술 분야의 숙련된 당업자는 하기의 특허 청구 범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.

- 제1항 내지 제3항 중 선택된 어느 한 항에 있어서,상기 메타설폰산은 상기 플루오레논 1mol을 기준으로 8.0 내지 13.0 mol로 사용되는 것을 특징으로 하는, 플루오렌 유도체의 합성방법.
- 제5항에 있어서,상기 그라파이트는 상기 플루오레논 100 g을 기준으로 40 내지 70 g이 사용되는 것을 특징으로 하는, 플루오렌 유도체의 합성방법.
- 제6항에 있어서,상기 반응식 1의 반응에서, 상기 페녹시에탄올은 상기 플루오레논 1mol을 기준으로 1.7 내지 2.3 mol로 사용되는 것을 특징으로 하는, 플루오렌 유도체의 합성방법.
