Description
本発明は、細胞培養用成分、細胞培養用培地、血清の製造方法、及び、細胞の製造方法に関する。特に、成牛血液成分の血清を利用した、新たな細胞培養用成分に関する。
従前より、細胞の培養には、栄養分として血清が用いられるが、なかでもウシ胎児血清(FBS;Fetal Bovine Serum)が最も広く用いられている。ウシ胎児血清は、成長因子や低レベルでの抗体を含むため、さまざまな細胞培養に使用可能であり、汎用性が高い(特許文献1、2)。
しかしながら、ウシ胎児血清を用いる場合、ウシ胎児からの採血後に即座に遠心分離をする必要があり、取り扱いが困難であるという課題があった。また、胎児血液は成牛と比較し、血液量が限られるため、大量にウシ胎児血清を得ることも困難であるという課題があった。このような特徴から、特に、工業用途の細胞培養に、ウシ胎児血清を利用しようとすると、量を確保するためにコストがかかり、また、ウシ胎児血清の取り扱いの難しさから工業プロセスでの利用が困難であるという課題があった。さらに、倫理的観点からも、胎児血清を使用しない血清の開発が望まれていた。
本発明は、ウシ胎児血清ではなく、成牛の血液を利用した細胞培養用成分を提供することを課題とする。また、成牛の血液を利用した細胞培養用成分を細胞培養の栄養分として用い、細胞を製造することを課題とする。
本発明者らは、上記事情に鑑みて鋭意検討した結果、成牛の血液を利用した細胞培養用成分を見出した。また、該細胞培養用成分を用いた培地で細胞培養し細胞を得る方法を見出した。
すなわち、本発明の細胞培養用成分は、成牛血液に由来する血清を含むことを特徴とする。

本発明の細胞培養用培地は、上記細胞培養用成分を含む。
本発明の血清の製造方法は、抗凝固剤を含む成牛血液から血清を得る、血清の製造方法であって、前記抗凝固剤を含む成牛血液に、凝固剤を添加するステップと、前記凝固剤を添加した成牛血液を遠心分離することで血清を得るステップと、を含む。

本発明の細胞培養用成分は、ウシ胎児血清を含まず、成牛の血液を利用することで提供されるため、大量に安価な栄養分として細胞培養に利用できる。また、成牛血液の血清は、ウシ胎児血清に比べ、取り扱いの困難性が低く、安定した材料であるため、工業プロセスでの利用が可能となる。また、成牛は食用にもなり得るため、成牛の血液を利用した細胞培養用成分で筋細胞を培養すれば、より安全性の高い人工食肉を作成することも期待される。さらに、胎児血清を使用しなくて済むため、倫理的観点からも望ましい細胞培養用成分となり得る。
本発明の実施形態について、以下に具体的に説明する。
(細胞培養用成分、血清及び血清の製造方法)
本発明の細胞培養用成分は、成牛血液に由来する血清、又は、成牛血漿に由来する血清を含むものである。
本発明において、成牛とは、胎児ではない牛を指す。牛の年齢による名称や分類は専門分野により異なるものの、本発明の成牛は、雌牛から分娩された後の、幼い牛、若い牛、成熟した牛、老いた牛のいずれの年齢の範囲にあってもよい。なお、市場からの入手のしやすさや屠畜解体される量の観点から、成熟した牛がより好ましい。
本発明の細胞培養用成分は、成牛血液に由来する血清、又は、成牛血漿に由来する血清を含むものである。
本発明において、成牛とは、胎児ではない牛を指す。牛の年齢による名称や分類は専門分野により異なるものの、本発明の成牛は、雌牛から分娩された後の、幼い牛、若い牛、成熟した牛、老いた牛のいずれの年齢の範囲にあってもよい。なお、市場からの入手のしやすさや屠畜解体される量の観点から、成熟した牛がより好ましい。
成牛の血液は、食肉市場や卸売市場において屠畜解体される牛から検体として採取する方法や、その他の方法により得られる。
成牛から採取された直後の成牛血液は、自然放置あるいは遠心分離により血清と血餅に分離することができる。血餅は、血液が凝固するときにできる暗赤色の塊であり、血漿中のフィブリノゲンが線維状のフィブリンに変わり、赤血球、白血球、血小板からなる血球と絡み合って沈殿したものである。血清は、血漿からフィブリノゲンその他の凝固因子が除かれた淡黄色の液体である。すなわち、血清には凝固因子が含まれないか、もしくは凝固因子がほとんど含まれない。
成牛血液から遠心分離にて上清を回収して得た血清は、細胞培養時の栄養分たる細胞培養用成分として使用することができる。

一方、成牛血液は、流通や保管時の凝固を防ぐため、抗凝固剤を添加してから配送されることが多い。抗凝固剤が添加された成牛血液は、赤血球と、血漿と、白血球及び血小板を含む層に遠心分離はできるものの、血漿中の凝固因子が取り除かれていない。
このため、抗凝固剤を含む成牛血液の場合、さらに凝固剤を添加し、血漿中の凝固因子を凝固させ、遠心分離することにより、血清を得ることが可能となる。このように回収して得た血清は、細胞培養時の栄養分たる細胞培養用成分として使用することができる。

クエン酸ナトリウムの場合、成牛血液及び抗凝固剤全量に対し、2体積%~4体積%、好ましくは、3体積%前後となるように添加することがより望ましい。

DMEMには、L-アルギニン、L-シスチン、L-ヒスチジン、L-イソロイシン、L-ロイシン、L-リシン、L-フェニルアラニン、L-トレオニン、L-トリプトファン、L-チロシン、L-バリン、塩化カルシウム、塩化カリウム、硫酸マグネシウム、ナトリウム塩化物、リン酸二水素ナトリウム、D-グルコース、葉酸、ニコチンアミド、リボフラビン、ビタミンB12、コリン、イノシトール、パントテン酸、ピリドキサールリン酸、チアミン、鉄等が含まれる。DMEMを凝固剤として用いる場合、DMEMの量に対し、成牛血液及び抗凝固剤の合計が、5体積%以上、好ましくは10体積%以上、より好ましくは10体積%~30体積%となるように添加することができる。DMEMに含まれるカルシウム量に対し、成牛血液及び抗凝固剤の合計が多くなると、硬化しにくくなる。
(細胞培養用培地)
本発明の細胞培養用培地は、上記細胞培養用成分を含む。

細胞培養用培地には、上記成牛血液の血清を含む細胞培養用成分の他、増殖培養用培地や、培地に使用することができる添加剤成分を適宜配合することができる。
増殖培養用培地としては、DMEM(GIBCO等)や、EMEM(GIBCO)、MEM ALPHA(GIBCO)、RPMI-1640(Roswell Park Memorial Institute 1640培地;GIBCO)等が挙げられる。
添加剤成分としては、抗生物質の他、ビタミン類、核酸、アミノ酸、無機塩、糖、ポリアミン、炭水化物、タンパク質、脂肪酸、脂質、pH調整剤、亜鉛、銅、セレン等が例示される。
増殖培養用培地としては、DMEM(GIBCO等)や、EMEM(GIBCO)、MEM ALPHA(GIBCO)、RPMI-1640(Roswell Park Memorial Institute 1640培地;GIBCO)等が挙げられる。
添加剤成分としては、抗生物質の他、ビタミン類、核酸、アミノ酸、無機塩、糖、ポリアミン、炭水化物、タンパク質、脂肪酸、脂質、pH調整剤、亜鉛、銅、セレン等が例示される。
培地中の細胞培養用成分の濃度は、細胞や培養条件により適宜設定される。すなわち、培地中の細胞培養用成分の下限濃度は細胞が生存維持できる濃度であり、好適濃度は細胞培養用成分が添加されていない培地に比べて細胞増殖の量が最大となる濃度であり、上限濃度は培地の組成として有害とならない最大濃度である。細胞培養用成分の濃度の一例は、培地全体に対し0.5体積%~20体積%、好ましくは2体積%~15体積%、より好ましくは2体積%~13体積%である。
(細胞の製造方法)
本発明の細胞の製造方法は、上記成牛血液に由来する血清、あるいは、上記成牛血漿に由来する血清を含む細胞培養用成分を用いて細胞を培養し、所望の増殖・分化細胞を得るものである。

培養する細胞は、成牛血液に由来する血清又は成牛血漿に由来する血清を含む細胞培養用成分を栄養分として利用可能な細胞であれば、特に種類の限定はない。
ウシ筋芽細胞等の初代培養細胞、C2C12、CHO、HEK293、HL-60、HeLa、MDCK、NIH3T3、PC12、S2、Vero等の細胞が例示される。

このうち、筋芽細胞は、増殖して多核化すると、筋管(筋管細胞)又は筋繊維の形態である骨格筋芽細胞となるものである。筋芽細胞は、サル、ウシ、ウマ、ブタ、ヒツジ、ヤギ、イヌ、ネコ、モルモット、ラット、マウス等の哺乳類、ダチョウ、ニワトリ、カモ、スズメ等の鳥類、ヘビ、ワニ、トカゲ、カメ等の爬虫類、カエル、イモリ、サンショウウオ等の両生類、サケ、マグロ、サメ、タイ、コイ等の魚類等の脊椎動物に由来するものが例示される。なかでも、増殖して多核化した骨格筋芽細胞を食用とする可能性を検討する場合は、ウシ、ブタ、ヒツジ、ヤギ、ウマ等の畜産のために飼育される哺乳類に由来する筋芽細胞がより望ましい。
細胞の製造方法の一例として、成牛血液に由来する血清を含む細胞培養用成分を用いて、細胞を培養する。
細胞の製造方法の他の一例として、成牛血漿に由来する血清を含む細胞培養用成分を用いて、細胞を培養する。
細胞の製造方法の他の一例として、上記細胞培養用成分を含む細胞培養用培地を用いて、細胞を培養する。
細胞の製造方法の他の一例として、抗凝固剤を含む成牛血液に凝固剤を添加して得られた血清、又は、成牛血漿に凝固剤を添加して得られた血清を用いて、細胞を培養する。
細胞の製造方法の他の一例として、成牛血漿に由来する血清を含む細胞培養用成分を用いて、細胞を培養する。
細胞の製造方法の他の一例として、上記細胞培養用成分を含む細胞培養用培地を用いて、細胞を培養する。
細胞の製造方法の他の一例として、抗凝固剤を含む成牛血液に凝固剤を添加して得られた血清、又は、成牛血漿に凝固剤を添加して得られた血清を用いて、細胞を培養する。
具体例として、成牛血液の血清を、DMEMに対し10体積%となるように調製する。これにC2C12マウス筋芽細胞を播種し、継代方法を0.25%トリプシンによる細胞剥離と、継代・培地交換頻度を2-3日に1回、培養温度を37℃、二酸化炭素濃度を5%として培養する方法が挙げられるが、これに限定されない。
細胞培養に供する細胞の好適濃度は、成牛血液の血清を含む細胞培養用成分の濃度や培地の組成、播種する面積等により変わるが、播種面積1cm2あたり1,000個~50,000個の割合となるような濃度で播種することが望ましい。
次に実施例により本発明をさらに具体的に説明する。なお、下記実施例は本発明の範囲を限定するものではない。
(成牛血液硬化評価試験1)
DMEM(GIBCO社製)100体積%に対し、抗凝固剤となるクエン酸ナトリウムが3.13%配合された成牛の血液(東京芝浦臓器株式会社製)の配合量を変えて配合し、血液硬化の有無を確認した。
評価結果を表1に示す。

表1の結果より、DMEM100体積%に対し、クエン酸ナトリウム入り成牛血液を10体積%、30体積%配合すると、成牛血液の硬化が見られることが分かった。
DMEMに含まれる塩化カルシウムや他の成分が血液と反応し、クエン酸ナトリウムとカルシウムイオンとの平衡状態が変わることにより、血液凝固に不可欠なカルシウムイオンが血液を硬化させるものと推察される。

以上より、DMEMはクエン酸ナトリウム入り成牛血液を凝固させることができ、かかる試料を遠心分離することにより成牛血液由来の血清を得ることができることが分かった。
また、成牛血漿をDMEM100体積%に対し10体積%、30体積%配合すると、同様に成牛血漿の硬化が見られることを確認した。
(成牛血液硬化評価試験2)
抗凝固剤となるクエン酸ナトリウムが3.13%配合された成牛の血液(東京芝浦臓器株式会社製)に、塩化カルシウムの濃度を変えて配合し、血液硬化の有無を確認した。
評価結果を表2に示す。

表2の結果より、クエン酸ナトリウム入り成牛血液に、塩化カルシウムをCa濃度にて12mM、24mM、60mM配合すると、成牛の血液の硬化が見られることが分かった。成牛血液硬化評価試験2で評価した範囲においては、特にCa濃度が24mMのときに、血液の硬化がより進むことが分かった。
塩化カルシウムが血液に配合され、クエン酸ナトリウムとカルシウムイオンとの平衡状態が変わることにより、血液凝固に不可欠なカルシウムイオンが血液を硬化させるものと推察される。

以上より、塩化カルシウムはクエン酸ナトリウム入り成牛血液を凝固させることができ、遠心分離により成牛血液の血清を得ることができることが分かった。
また、抗凝固剤となるクエン酸ナトリウムが3.13%配合された成牛血漿に対しても成牛血液硬化評価試験1、2と同様の試験を行った。その結果、塩化カルシウムをCa濃度にて12mM、24mM、60mM配合すると、成牛血漿の硬化が見られることが分かった。
(細胞培養試験1)
抗凝固剤となるクエン酸ナトリウムが3.13%配合された成牛の血液(東京芝浦臓器株式会社製)に、塩化カルシウムをCa濃度で20mM配合し、遠心分離にて上清の血清を回収した。
次に、DMEMに、成牛血液の血清を含む細胞培養用成分とペニシリン-ストレプトマイシンを配合し、全培地体積に対して成牛血液の血清を含む細胞培養用成分が10体積%、ペニシリン-ストレプトマイシンが1体積%となるように培養用培地を調製した。この培養用培地中にC2C12マウス筋芽細胞を播種し、培地交換頻度を1-3日に1回、培養温度を37℃、二酸化炭素濃度を5%として培養した。
1日目、3日目、6日目、8日目、10日目、12日目に、培養された細胞を顕微鏡にて観察した。

比較対照として、成牛血液の血清を含む細胞培養用成分に代えて、ウシ胎児血清を用いて同様に細胞培養を行った。
具体的には、DMEMと、ウシ胎児血清とペニシリン-ストレプトマイシンを配合し、全培地体積に対してウシ胎児血清が10体積%、ペニシリン-ストレプトマイシンが1体積%となるように培養用培地を調製した。この培養用培地中にC2C12マウス筋芽細胞を播種し、培地交換頻度を1-3日に1回、培養温度を37℃、二酸化炭素濃度を5%として培養した。
1日目、3日目、6日目、8日目、10日目、12日目に、培養された細胞を顕微鏡にて観察した。
具体的には、DMEMと、ウシ胎児血清とペニシリン-ストレプトマイシンを配合し、全培地体積に対してウシ胎児血清が10体積%、ペニシリン-ストレプトマイシンが1体積%となるように培養用培地を調製した。この培養用培地中にC2C12マウス筋芽細胞を播種し、培地交換頻度を1-3日に1回、培養温度を37℃、二酸化炭素濃度を5%として培養した。
1日目、3日目、6日目、8日目、10日目、12日目に、培養された細胞を顕微鏡にて観察した。
細胞培養試験1の結果を、図1A及び図1Bに示す。
図1A及び図1Bより、成牛血液の血清を含む細胞培養用成分でも、ウシ胎児血清と同様にC2C12マウス筋芽細胞が増殖されることが確認された。増殖速度もほぼ同様であった。また、成牛血液の血清を含む細胞培養用成分でも、ウシ胎児血清と同様にC2C12マウス筋芽細胞が分化されることが10日目以降に確認された。
以上より、成牛血液の血清を含む細胞培養用成分は、ウシ胎児血清と同等にC2C12マウス筋芽細胞を増殖し、分化させることができることが分かった。
(細胞培養試験2)
抗凝固剤となるクエン酸ナトリウムが3.13%配合された成牛の血液(東京芝浦臓器株式会社製)に、塩化カルシウムをCa濃度で20mM配合し、遠心分離にて上清の血清を回収した。

細胞培養試験2の結果を、図2に示す。
図2より、成牛血液の血清を含む細胞培養用成分の濃度が2体積%と低くても、C2C12マウス筋芽細胞が分化されることが確認された。
また、成牛血漿に対しても、塩化カルシウムをCa濃度にて20mM配合し、遠心分離にて上清の血清を回収し、細胞培養試験1及び2と同様の条件で細胞培養を行った。その結果、成牛血漿に由来する血清を用いた場合にも、成牛血液の血清の場合と同様にC2C12マウス筋芽細胞が増殖および分化されることが確認された。

- 成牛血液に由来する血清を含む、細胞培養用成分。
- 成牛血漿に由来する血清を含む、細胞培養用成分。
- 請求項1又は2記載の細胞培養用成分を含む、細胞培養用培地。
- 抗凝固剤を含む成牛血液から血清を得る、血清の製造方法であって、
前記抗凝固剤を含む成牛血液に、凝固剤を添加するステップと、
前記凝固剤を添加した成牛血液を遠心分離することで血清を得るステップと、
を含む、血清の製造方法。 - 成牛血漿に凝固剤を添加するステップと、
前記凝固剤を添加した成牛血漿を遠心分離することで血清を得るステップと、
