Description
본 발명은 연교, 작약, 및 치자로 이루어진 군에서 선택되는 어느 하나 이상의 추출물 또는 이의 분획물을 포함하는 프리바이오틱스 조성물, 이를 포함하는 식품 조성물, 사료 조성물, 아토피 피부염 예방 또는 치료용 약학적 조성물, 및 아토피 피부염 예방 또는 개선용 의약외품 및 식품 조성물, 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 장 기능 개선 용도, 방법, 아토피 예방, 치료, 또는 개선 용도, 방법에 관한 것이다.
프로바이오틱스(probiotics)는 장내 균 총의 균형을 유지시킴으로써 유익하게 작용할 수 있는 살아있는 미생물로서, 다양한 기능을 갖는다는 것이 알려져 있다. 예를 들어, 프로바이오틱스는 장내 영양소와 장벽 부착 부위에 관한 경쟁을 통해 유해균의 생장을 저해하고, 면역활성화 작용, 유당불내증의 경감 등 인체에 유익한 기능을 하는 것으로 알려졌다.
이러한 유익한 기능을 증진시키기 위해, 프로바이오틱스를 보다 효과적으로 증식 및 유지시키기 위하여 사용되는 주요 성분인 프리바이오틱스에 대한 연구도 지속되어 왔다. 프리바이오틱스의 대표적인 예로서 프락토올리고당(fructo-oligosaccharides, FOS)이 알려져 있다. 프락토올리고당은 설탕에 과당분자가 β-1, 2로 결합한 형태의 올리고당으로서, 락토바실러스와 비피도박테리아에 의해 발효가 가능하기 때문에, FOS를 섭취하면 비피도박테리움과 락토바실러스 종들을 증가시킬 수 있다고 알려져 있다.
그러나, 장내 FOS의 대사가 가능한 유해균 및 프로바이오틱이 아닌 균주(non-probiotics)에 의해 역효과가 나타날 수도 있다는 문제점이 알려져 있어, 보다 효과적으로 프로바이오틱스를 증식 및 유지시킬 수 있는 프리바이오틱스 조성물의 개발이 요구되고 있다. 이에, 식물을 이용한 것으로서 포도씨 분말을 포함하는 프리바이오틱스가 연구되었으나(한국등록특허 제10-1973514호), 한방 추출물을 이용한 프리바이오틱스는 개발이 미미한 실정이다.
본 발명자들은 보다 효과적인 프리바이오틱스 조성물을 개발하고자 예의 노력한 결과, 프리바이오틱스 조성물의 유효성분으로 본 발명의 추출물을 발굴하고, 본 발명의 프리바이오틱스 조성물이 아토피 피부염도 치료할 수 있음을 확인함으로써, 본 발명을 완성하였다.
본 발명의 하나의 목적은 연교, 작약, 및 치자로 이루어진 군에서 선택되는 어느 하나 이상의 추출물 또는 이의 분획물을 포함하는 프리바이오틱스 조성물을 제공하는 것이다.
본 발명의 다른 하나의 목적은 상기 프리바이오틱스 조성물을 포함하는 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 하나의 목적은 상기 프리바이오틱스 조성물을 포함하는 사료 조성물을 제공하는 것이다.
본 발명의 또 다른 하나의 목적은 상기 프리바이오틱스 조성물을 포함하는 아토피 피부염 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
본 발명의 또 다른 하나의 목적은 상기 프리바이오틱스 조성물을 포함하는 아토피 피부염 예방 또는 개선용 의약외품 조성물을 제공하는 것이다.
본 발명의 또 다른 하나의 목적은 상기 프리바이오틱스 조성물을 포함하는 아토피 피부염 예방 또는 개선용 식품 조성물을 제공하는 것이다.

상기 의약외품 조성물은 특별히 이에 제한되지 않으나, 피부외용제, 소독청결제, 샤워폼, 물티슈, 세제비누, 핸드워시, 마스크 또는 연고제를 포함한다. 상기 피부외용제는 특별히 이에 제한되지 않으나, 구체적으로 연고제, 로션제, 스프레이제, 패취제, 크림제, 산제, 현탁제, 겔제 또는 젤의 형태로 제조되어 사용될 수 있다. 상기 개인위생용품에는 특별히 이에 제한되지 않으나, 구체적으로 비누, 물티슈, 휴지, 샴푸, 치약, 모발 손질 제품, 에어프레쉬너 겔 또는 세정 겔일 수 있다.
본 발명의 또 다른 하나의 양태는 상기 프리바이오틱스 조성물을 포함하는 아토피 피부염 예방 또는 개선용 식품 조성물을 제공한다.
상기 프리바이오틱스, 아토피 피부염, 에방, 개선, 식품 조성물 등은 다른 양태에서 설명한 바와 같다.
본 발명의 또 다른 양태는 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 장 기능 개선 용도를 제공한다.
상기 추출물, 분획물, 장 기능 개선 등은 다른 양태에서 설명한 바와 같다.
상기 장 기능 개선은 장내 균총 개선을 포함하는 것일 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 하나의 양태는 상기 프리바이오틱스 조성물을 개체에 투여하는 단계를 포함하는 장 기능 개선 방법을 제공한다.

본 발명의 또 다른 양태는 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 아토피 예방 또는 개선 용도를 제공한다.
본 발명의 또 다른 양태는 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 아토피 예방 또는 치료 용도를 제공한다.
상기 추출물, 분획물, 아토피, 예방, 개선, 치료 등은 다른 양태에서 설명한 바와 같다.
이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나 이들 실시예 및 실험예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예 및 실험예에 한정되는 것은 아니다.
제조예: 연교, 작약, 치자, 및 이들의 조합의 제조 및 투여방법
연교, 작약, 및 치자는 한풍제약(Hanpoong Pharmaceutical Co., 전주, 한국)에서 구입하여, 연교, 작약, 치자, 및 이들의 조합을 각각 열수 추출 하였다. 이후, 감압 농축기를 이용하여 용매를 모두 제거하고 동결 건조하여 추출 분말을 제조하였다. 수득한 상기 분말은 200mg/kg로 음수에 녹인 후, 동물모델에 경구 투여하였다.
실시예 1: 장 내 미생물 억제 동물모델(ABX) 및 아토피 피부염을 동반하는 장 내 미생물 억제 동물 모델(ABX+DNCB) 구축
장 내 미생물 억제 동물모델(ABX 모델)을 구축하기 위해, 마우스를 대상으로 3% 수크로스 및 1% 글루코스를 용해시킨 식음수에 암피실린(ampicillin), 메트로니다졸(metronidazole), 반코마이신(vancomycin), 및 네오마이신 설페이트(neomycin sulfate)를 포함하는 항생제 4종을 혼합하여 7일씩 급여한 후 3일씩 일반 물로 바꾸어 급여하였다. 항생제 4종은 4주동안 점차적으로 농도를 높이면서 자율 급수 방법으로 장 내 미생물을 억제하도록 유도하였다.
아토피 피부염을 동반하는 장 내 미생물 억제 동물 모델(ABX+DNCB 모델)을 구축하기 위해, 전술한 ABX 모델 구축 방식과 마찬가지로 항생제 및 물을 급여함과 동시에, 마우스의 등 피부를 면도하고 1Х1 cm의 패취를 이용하여 100 ㎕의 2% DNCB(2,4-dinitrochlorobenzene)를 도포함으로써 아토피 피부염 (Atopic dermatitis; AD)을 유도하였다.
보다 구체적으로, 장 내 미생물 억제 동물모델(ABX 모델) 및 아토피 피부염을 동반하는 장 내 미생물 억제 동물 모델(ABX+DNCB 모델)을 구축하기 위한 급여 및 도포 방식을 도식화하여 도 1에 나타내었다.
실시예 2: 연교, 작약, 및 치자의 독성 평가
연교, 작약, 및 치자의 섭취로 인한 독성 유무를 확인하기 위해, 전술한 제조예의 연교, 작약, 및 치자를 섭취한 마우스의 체중 및 식이량을 확인하였다.
구체적으로, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에 식음수에 용해시킨 연교, 작약, 및 치자를 2주동안 동일한 시간에 매일 경구 투여하였다(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자).
그 결과, 체중 및 식이 변화는 적정 오차 범위 내에서 약간의 차이만 있었으므로, 연교, 작약, 및 치자의 유의미한 독성은 없음을 확인하였다(도 2 내지 도 4).
실시예 3: 연교, 작약, 및 치자 단독 추출물의 프리바이오틱스 효과 확인
실시예 3-1: 연교, 작약, 및 치자의 장 내 혈변 수치 및 상태 개선 확인
연교, 작약, 및 치자의 프리바이오틱스 효과를 평가하기 위해, 장 내 혈변 수치 및 상태 개선 여부를 확인하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)의 대변을 채취하여, 대변 샘플의 염증 또는 혈액 수치를 육안으로 확인할 수 있도록 Hemoccult kit를 사용하였다. 이 때, Hemoccult kit 용액을 상기 대변 샘플에 2방울 떨어뜨렸을 때에 파란색으로 변하는 정도가 증가할 수록 염증이나 혈변의 정도가 심한 것이며, 이를 객관화하기 위해 3인 이상 이를 점수화하여 나타내었다. 또한, 3인 이상이 마우스 대변의 점도 및 혈액 확인 정도를 점수화하는 stool consistency를 활용하였다.
그 결과, 혈액과 점액질이 확인된 장 내 미생물 억제군(ABX)의 대변보다 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)의 대변에서 혈액과 점액질 수준이 더 높음을 확인하였다. 또한, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)을 대상으로 연교, 작약, 또는 치자를 경구 투여한 결과(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 혈액 및 점액질 수준이 현저히 감소하여 장 내 미생물의 환경이 개선됨을 확인하였다(도 5 내지 도 10).
이에, 장 내 미생물 억제에 의해 증가된 대변 내 혈액 및 점액질 수준을 연교, 작약, 및 치자가 감소시킴을 확인하였다.
실시예 3-2: 연교, 작약, 및 치자의 장 길이 연장 감소 효과 확인
장 내 미생물이 억제될 경우 장 세포의 기능 저하로 장의 길이가 연장된다고 알려져 있으므로, 연교, 작약, 및 치자의 프리바이오틱스 효과를 평가하기 위해, 장 길이의 변화를 확인하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 연교, 작약, 또는 치자 추출물 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)으로부터 각각 분리된 장의 길이를 측정하였다.
그 결과, 정상군(Normal)에 비해 장 내 미생물 억제군(ABX) 및 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서 모
두 장의 길이가 연장된 반면, 연교, 작약, 및 치자 추출물(이하, '추출물' 생략할 수 있다)을 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 정상군과 유사한 수준으로 장의 길이가 감소됨을 확인하였다(도 11 및 도 16).
이에, 장 내 미생물 억제에 의해 연장된 장 길이를 연교, 작약, 및 치자가 감소시키는 것을 확인하였다.
실시예 3-3: 연교, 작약, 및 치자의 장 조직 개선 효과 확인
연교, 작약, 및 치자의 프리바이오틱스 효과를 평가하기 위해, 장 조직의 개선 여부를 확인하였다.
장의 형태 및 상태를 관찰하기 위해, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)으로부터 장 조직의 일부를 분리하여 4% 파라포름알데하이드(paraformaldehyde; PFA)로 고정한 후, 파라핀(Paraplast High Melt, Surgipath, Leica, Biosystem, The Netherlands)을 이용하여 파라핀 절편을 제작하였다.
제작한 장 샘플을 대상으로, 조직학적 염색인 H&E(Hematoxylin and Eosin) 염색과 함께 ZO-1 항체를 이용한 immunofluorescence 염색 및 Occludin 항체를 이용한 immunofluorescence 염색을 수행하였다. ZO-1 및 Occludin은 장 세포간 밀착 접합(tight junction)을 구성하는 단백질이므로, 밀착 접합 붕괴 여부를 평가하기 위해 이들을 대상으로 하였다.
그 결과, 정상군(Normal)에 비해 장 내 미생물 억제군(ABX) 및 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서 모두 장 내 미생물의 억제가 장 조직 내 융모(vilus) 및 샘(crypt)의 형태를 붕괴시키고 장 세포간 밀착 접합(tight junction)이 억제된 반면, 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)에 정상군과 유사한 수준으로 융모 및 샘의 형태가 회복됨을 확인하였다(도 17).
이에, 장 내 미생물 억제에 의해 저하되었던 장 조직의 기능을 연교, 작약, 및 치자가 회복시키는 것을 확인하였다.
실시예 3-4: 연교, 작약, 및 치자 의 장 내의 유익균 비율 증가 및 유해균 비율 감소 효과 확인
연교, 작약, 및 치자의 프리바이오틱스 효과를 평가하기 위해, 장 내 미생물 변화를 확인하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 및 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)에서 채취한 대변을 대상으로 16s rRNA metagenome sequencing을 통해 미생물 수의 변화를 관찰하였다.
미생물 수는 유익균(Lactobaciilacea, Lachnospiraceae, Rikenellaceae, Prevotellaceae, Acetobacteraceae) 및 유해균(Clostridiaceae 1, Ruminococcaceae, Erysipelotrichaceae, Porphyromonadaceae, Bacteroidaceae, Desulfovibrionaceae, Pasteurellaceae, Enterobacteriaceae)을 모두 평가하였다.
그 결과, 정상군(Normal)에 비해 장 내 미생물 억제군(ABX) 및 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서 모두 전체적인 미생물의 수치가 낮음을 확인하였다. 특히, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서 염증과 관련된 미생물인 유해균의 수가 더 많은 것으로 관찰되었다.
반면, 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)미생물 균총이 정상군과 유사한 수준으로 나타나는 것을 확인하였다(도 18).
이에, 연교, 작약, 및 치자가 장 내 미생물 억제에 의해 증가되었던 장 내 유해균의 수를 감소시키고, 감소되었던 장 내 유익균의 수를 증가시키며, 정상과 유사한 수준의 균총으로 회복시키는 것을 확인하였다.
실시예 4: 연교, 작약, 및 치자 조합의 프리바이오틱스 시너지 효과 확인
연교, 작약, 및 치자 조합의 프리바이오틱스 효과를 평가하기 위해, 장 내 미생물 변화를 보다 면밀하게 확인하였다. 구체적으로, 연교, 작약, 치자, 및 이들의 조합을 유익균(Lactobacillus pentosus 및 Lactobacillus plantarum)에 처리하여 상기 유익균의 수를 관찰하였고, 이는 600nm에서 OD 값을 측정하여 표시하였다. 유산균의 종 배양은 Lactobacilli MRS broth (Difco, Franklin Lakes, NJ, USA) 배지를 사용하였으며, 30℃, 200rpm 조건에서 정지기까지 배양 후 본배양을 위한 종배양액으로 사용하였다. 연교, 작약 및 치자를 최소배지 (peptone 1%, sodium chloride 1%)에 첨가하여 Autoclave로 멸균한 후 Lactobacillus pentosus 와 Lactobacillus plantarum의 종배양액이 생육곡선의 정지기에 도달한 유산균 배양액을 1% 접종하여 30℃, 200rpm의 조건에서 배양하며 시간 경과에 따른 OD값을 측정하여 생육도를 확인하였다.
Lactobacillus plantarum에 대한 OD 값은, 연교, 작약, 및 치자 혼합 추출물(90 ㎍/ml) 및 대비 연교, 작약, 및 치자 단독 추출물(45 ㎍/ml)에 대한 값으로 도 19 내지 도 21에 각각 나타내었다(FPG=연교:작약:치자=1:1:1, FPG1=연교:작약:치자=2:1:1, FPG2=연교:작약:치자=1:2:1, 및 FPG3=연교:작약:치자=1:1:2 비율).
또한, 하기 표 1과 같이, 연교, 작약, 및 치자 각각은 45 ㎍/ml와 90 ㎍/ml에서 유사한 수준의 OD 값을 나타내는 것을 확인하였다.

실시예 5: 연교, 작약, 및 치자 프리바이오틱스의 아토피 개선 효과 확인
실시예 5-1: 연교, 작약, 및 치자 프리바이오틱스의 조직 두께 감소 확인
아토피가 있다면 등 조직의 두께도 증가되므로, 연교, 작약, 및 치자를 포함하는 프리바이오틱스의 아토피 개선 효과를 확인하기 위하여, 등 조직 두께를 측정하고, 관능평가 법(SCORAD)을 수행하였다. 관능평가 법은 피부 건선/홍반/부종/출혈 등 육안으로 확인 가능한 아토피 피부염 증상을 관찰해 점수화한 것으로서, 객관성을 위해 3인 이상의 점수를 평균화 하여 점수화 하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염군(DNCB), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 및 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)의 등을 면도하여 육안으로 관찰하였고, 이들의 등 조직을 일부 분리하여 4% 포름알데하이드에 보관한 후 제작한 파라핀 조직 절편 샘플을 이용하여 등 조직의 두께를 측정하였다.
그 결과, 정상군(Normal) 및 장 내 미생물 억제군(ABX)에 비해 아토피 피부염군(DNCB)의 등 조직이 두꺼웠으며, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서의 등 조직이 더 두꺼운 것을 확인하였다. 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 정상군과 유사한 수준으로 등 조직의 두께가 감소함을 확인하였다(도 25 내지 도 30).
이에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스는 아토피로 인해 증가된 조직 두께를 감소시키는 효과가 있음을 확인하였다.
실시예 5-2: 연교, 작약, 및 치자 프바이오틱스의 염증 매개 면역세포 감소 효과 확인
아토피 피부염에 의해 면역 세포가 과 발현되기 때문에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스의 아토피 개선 효과를 평가하기 위하여, 면역세포 감소 여부를 확인하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염군(DNCB), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 및 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)에서 마우스의 전체 혈액 샘플을 심장에서 채혈하였고, EDTA(BD science, NJ, USA)가 포함된 Vacutainer TM 튜브에 넣어 보관하였다. 채혈 및 보관한 혈액 샘플을 대상으로, 다음 면역 세포 6종 각각의 개수를 HEMAVET 950 혈구계산기(Drew Scientific, Inc., Oxford, USA)를 이용하여 분석하였다: WBC, 림프구(lymphocytes), 단핵백혈구(monocytes), 호산구(eosinophils), 호염구(basophils) 또는 호중구(neutrophils).
그 결과, 정상군(Normal) 및 장 내 미생물 억제군(ABX)에 비해 아토피 피부염군(DNCB)의 면역세포 6종의 수가 현저히 증가하였으며, 이보다도 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서의 면역세포 6종의 수가 더 증가되는 것을 확인하였다. 또한, 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 정상군과 유사한 수준으로 면역세포 수가 감소함을 확인하였다 (도 31 내지 도 33).
이에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스는 아토피로 인해 증가된 면역세포 수를 감소시키는 효과가 있음을 확인하였다.
실시예 5-3: 연교, 작약, 및 치자 프리바이오틱스의 염증 사이토카인 감소 효과 확인
아토피의 특징 중 하나는 혈중 IgE 및 염증 사이토카인이 높은 수준으로 발현되는 것이다. 이에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스의 아토피 개선 효과를 확인하기 위하여, 염증 매개 사이토카인 (IgE, IL-12, TNF-α, IL-6) 수준을 분석하였다.
구체적으로, 정상군(Normal), 장 내 미생물 억제군(ABX), 아토피 피부염군(DNCB), 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB), 및 연교, 작약, 또는 치자 투여군(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자)에서 채취한 혈액을 대상으로, BD PharMingen mouse 또는 human ELISA set(Pharmingen, San Diego, CA, USA)를 이용한 샌드위치 ELISA를 통해 발현 수준을 분석하였다. ELISA 코팅 버퍼(sigma, Louis, MO, USA)에 포함된 포획 항체(capture antibody)로 플레이트를 코팅하였고, 4℃에서 하루 동안 보관하였다. 플레이트를 0.05%의 PBS-Tween 20로 헹구었고, 이어서 1시간 동안 20℃에서 10% FBS가 포함된 PBS로 블로킹하였다. 희석 버퍼(10% FBS가 포함된 PBS)에 단계 희석한 표준 항원 또는 샘플을 플레이트에 넣고, 20℃에서 1시간 동안 인큐베이션하였다. 이후, 비오틴-접합 항-마우스 IgE 및 SAv-HRP(streptavidin-horseradish peroxidase conjugate)를 플레이트에 넣고, 20℃에서 1시간 동안 인큐베이션하였다. 마지막으로, TMB(tetramethylbenzidine) 기질 용액을 플레이트에 넣고, 암조건에서 15분 동안 배양하였으며, 2N H2SO4 용액으로 반응을 종결하였다. ELISA reader(Versa Max, Molecular Devices, CA, USA)를 이용하여 450nm에서 광학밀도를 측정하였다.
그 결과, 정상군(Normal) 및 장 내 미생물 억제군(ABX)에 비해 아토피 피부염군(DNCB)에서의 염증 매개 사이토카인(IgE, IL-6, IL-12, 및 TNF-α) 수준이 증가하였으며, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서 아토피 피부염군(DNCB)과 유사한 수준 또는 더 높은 수준으로 증가함을 확인하였다. 반면, 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 염증 매개 사이토카인(IgE, IL-6, IL-12, 및 TNF-α) 수준이 현저하게 감소함을 확인하였다 (도 34 내지 도 36).
이에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스는 아토피로 인해 증가된 염증 매개 사이토카인의 수준은 감소시키는 효과가 있음을 확인하였다.
실시예 5-4: 연교, 작약, 및 치자 프리바이오틱스의 아토피 피부염 완화 효과 확인
연교, 작약, 및 치자를 포함하는 프리바이오틱스의 아토피 개선 효과를 확인하기 위하여, 염증 매개 지표인 염증 세포, 비만 세포(mast cell), TSLP, 및 피부 표피 두께를 측정하였다.
구체적으로, 파라핀 조직 절편 등 조직 내 H&E 염색을 통해 염색된 염증세포를 관찰하였고, Toluidun blue 염색을 통해 염색된 비만세포를 관찰했으며, TSLP 염색을 통해 염색된 TSLP를 관찰하였다. 피부 표피 두께는 조직 염색사진에서 측정하였다.
그 결과, 정상군(Normal) 및 장 내 미생물 억제군(ABX)에 비해 아토피 피부염군(DNCB)에서의 염증 세포, 비만 세포(mast cell), 및 TSLP이 더욱 높은 수준으로 관찰되며, 피부 표피의 두께도 증가함을 확인하였다. 피부의 두께가 증가되는 것은 피부 재생을 위한 세포 증식이 이루어지는 것으로 관찰되었다. 또한, 아토피 피부염을 동반하는 장 내 미생물 억제군(ABX+DNCB)에서는 염증 세포, 비만 세포, TSLP, 피부 두께 모두 아토피 피부염군(DNCB)에서 더욱 증가함을 확인하였다.
반면, 연교, 작약, 및 치자를 투여하였을 때(ABX+DNCB+연교, ABX+DNCB+작약, ABX+DNCB+치자), 염증 세포, 비만 세포, TSLP, 피부 두께 모두 현저하게 감소함을 확인하였다(도 37).
이에, 연교, 작약, 및 치자를 포함하는 프리바이오틱스는 아토피를 개선하는 효과가 있음을 확인하였다.
전술한 결과로부터, 연교, 작약, 및 치자는 장 내 미생물 환경을 개선시키는 프리바이오틱스로의 효과가 있으며, 본 발명의 연교, 작약, 및 치자를 포함하는 프리바이오틱스는 아토피 피부염도 개선 및 치료할 수 있음을 확인하였다.
이상의 설명으로부터, 본 발명이 속하는 기술분야의 당업자는 본 발명이 그 기술적 사상이나 필수적 특징을 변경하지 않고서 다른 구체적인 형태로 실시될 수 있다는 것을 이해할 수 있을 것이다. 이와 관련하여, 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적인 것이 아닌 것으로 이해해야만 한다. 본 발명의 범위는 상기 상세한 설명보다는 후술하는 특허 청구범위의 의미 및 범위 그리고 그 등가 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
Claims (19)
- 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 프리바이오틱스 조성물.
- 제1항에 있어서, 상기 추출물은 연교, 작약, 및 치자의 혼합 추출물인 것인, 프리바이오틱스 조성물.
- 제1항에 있어서, 상기 추출물은 물, 탄소수 1 내지 4의 알코올 및 이들의 혼합 용매로 이루어진 군에서 선택된 1종 이상의 용매로 추출한 것인, 프리바이오틱스 조성물.
- 제2항에 있어서, 상기 혼합 추출물은 연교, 작약, 및 치자로 구성되는 군에서 선택되는 어느 하나 이상의 중량비가 더 높은 것인, 프리바이오틱스 조성물.
- 제2항에 있어서, 상기 혼합 추출물은 연교, 작약, 및 치자를 0.5 내지 1.5 : 0.5 내지 1.5 : 0.5 내지 1.5 의 중량비로 포함하는 것인, 프리바이오틱스 조성물.
- 제1항에 있어서, 상기 조성물은 연교, 작약, 및 치자로 이루어진 군에서 선택되는 하나 이상의 추출물을 총 조성물의 중량 대비 0.001 내지 80 중량%로 포함하는 것인, 프리바이오틱스 조성물.
- 제1항에 있어서, 상기 프리바이오틱스 조성물은 장 내 유해균의 증식을 억제하고, 장내 유익균의 증식을 촉진하는 것인, 프리바이오틱스 조성물.
- 제7항에 있어서, 상기 장내 유해균은 클로스트리디움과 1(Clostridiaceae 1), 루미노코카세과(Ruminococcaceae), 에리시펠로트릭스과(Erysipelotrichaceae), 포르피로모나스과(Porphyromonadaceae), 박테로이데스과(Bacteroidaceae), 디설포비브리오과(Desulfovibrionaceae), 파스테우렐라과(Pasteurellaceae), 및 엔테로박테리아과(Enterobacteriaceae)로 구성되는 군에서 선택되는 어느 하나 이상이고, 상기 장내 유익균은 락토바실러스과(Lactobaciilacea), 라크노스피라과(Lachnospiraceae), 리케넬라과(Rikenellaceae), 프로보텔라과(Prevotellaceae), 및 아세토박터과(Acetobacteraceae)로 구성되는 군에서 선택되는 어느 하나 이상인 것인, 프리바이오틱스 조성물.
- 제1항 내지 제8항 중 어느 한 항의 조성물을 포함하는 식품 조성물.
- 제1항 내지 제8항 중 어느 한 항의 조성물을 포함하는 사료 조성물.
- 제1항 내지 제8항 중 어느 한 항의 조성물을 포함하는 아토피 피부염 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제8항 중 어느 한 항의 조성물을 포함하는 아토피 피부염 예방 또는 개선용 의약외품 조성물.
- 제1항 내지 제8항 중 어느 한 항의 조성물을 포함하는 아토피 피부염 예방 또는 개선용 식품 조성물.
- 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 장 기능 개선 용도.
- 제14항에 있어서, 상기 장 기능 개선은 장내 균총 개선을 포함하는 것인, 장 기능 개선 용도.
- 제1항의 조성물을 개체에 투여하는 단계를 포함하는 장 기능 개선 방법.
- 연교, 작약, 및 치자로 구성되는 군에서 선택되는 하나 이상의 추출물 또는 이의 분획물을 포함하는 조성물의 아토피 예방 또는 개선 용도.
