ここで、溶液中の標識コレステロール量を測定するためには、溶液中の磁気ビーズを分離する必要がある。コレステロールの固相化にゲルビーズ(ILG)を用いていた既報(非特許文献4、5)のCEC評価法では、ゲルビーズを分離するために遠心操作が必要であり、自動分析装置への搭載が困難であるという課題があった。一方、本実施形態の方法では、マグネットなどの磁力を用いて磁気ビーズを分離可能であるため、磁気ビーズを操作可能な分析装置を用いれば自動化が容易であり、臨床における日常検査への導入に適している。
また、磁気ビーズを磁気で分離する場合、ゲルビーズを遠心分離で分離する場合にくらべて、より短時間の工程で、より確実に分離できるとともに、遠心操作が必要ないので溶液中のHDLから安定に分離できるというメリットもある。
本実施形態のCEC測定方法では、まず、コレステロールを固相化した磁気ビーズを用意する。
本実施形態のCEC測定方法では、少なくとも一部が標識されたコレステロールをリポソームに複合化して、磁性ビーズに固相化することが好ましい。
リポソームは、標識コレステロールを含ませることができ、かつHDLがコレステロールを引き抜くことが可能であるため、CEC測定に好適な粒子である。また、リン脂質とコレステロールからなるリポソームは細胞膜の構造に近く、生体内の環境に類似しているため、生体内における引き抜き能をより正確に反映することができる。
また、一実施形態では、コレステロールをリポソームに含有させることで、後述するように、凍結融解などの簡便な方法でコレステロールを磁気ビーズに効率的に固相化することができるという利点もある。
なお、本明細書において、コレステロールを含むリポソームとは、コレステロール分子がリポソームの膜表面又は膜中に包埋されたリポソームを意味する。
磁気ビーズに固相化されたリポソームに包含されたコレステロールは、測定対象のHDLに引き抜かれると、HDLに取り込まれた状態で溶液中に移動することとなる。HDLのコレステロール引き抜き能は、磁気ビーズを分離後の溶液(本明細書では、磁気ビーズを分離後の溶液を「上清」と記載することもある。)中のコレステロール量(濃度)を測定することで評価することができる。したがって、本実施形態の測定方法では、上清中のコレステロール濃度の測定のために、少なくとも一部が標識されたコレステロールを用いる。
コレステロールの標識方法としては、特に限定されないが、蛍光物質、発色物質、発光物質、放射線物質、安定同位体などが挙げられるが、日常検査への導入の観点では、蛍光物質、発色物質、発光物質が好ましく、蛍光物質がより好ましい。
標識コレステロールとして、市販のものを適宜利用してもよい。
コレステロールを含有させたリポソームの調製方法の例としては、例えば、Yoshimoto et al., Biotechnol. Prog. 2006, 22, 459-464に記載の方法が挙げられる。
コレステロールとリン脂質との比率は適宜選択できるが、例えば、リン脂質:コレステロール=50:1~1:2、好ましくは20:1~1:1、より好ましくは10:1~2:1(重量比)とすることができる。
本実施形態のCEC測定方法で用いる磁気ビーズは、上述のコレステロールを含むリポソームを固相化し得るものであれば特に限定されず、例えば、磁性材料からなる粒子の他、磁性材料を含む粒子、例えば、磁性材料と、有機高分子化合物、無機化合物、生体高分子等との複合粒子等が挙げられる。
多孔質磁気粒子は、後述するように、凍結融解を行うことにより、簡便な方法で、効率よくコレステロールを固相化することができる。
コレステロールを含むリポソームを磁気ビーズに固相化する方法は、特に限定されず、磁気ビーズの種類などを考慮して適宜選択することができる。
このような工程によりコレステロールを含むリポソームを磁気ビーズに効率的に固相化できる理由としては、コレステロールを含むリポソームの懸濁液に、適切な孔径を有する多孔質磁気ビーズを混合・反応させることにより、リポソームが多孔質の磁気ビーズの孔内に入りこみ、この状態で凍結融解を繰り返すと、リポソームのサイズが大きくなり孔内に詰まった状態になるため、磁気ビーズにリポソームを効率的に固相化できるものと考えられる。
コレステロールを含むリポソームが固相化された磁気ビーズの保存安定性は、ILM懸濁液の懸濁媒体中(例えば上清中)の標識物質強度の変化で評価することができる。例えば、ILMの保存安定性が良好であれば、保存中に磁気ビーズから懸濁媒体中に遊離する標識物質(例えば、蛍光標識コレステロール)が少ない。一実施形態では、所望の保存条件(例えば4℃)において、少なくとも10日間、好ましくは少なくとも20日間、より好ましくは少なくとも30日間、さらに好ましくは少なくとも40日間、ILM懸濁液から採取した懸濁媒体の標識強度(例えば蛍光強度)が有意差をもって増加しない(実質的に変化しない)。
なお、本明細書において、コレステロールを含むリポソームを固相化した磁気ビーズを単に「コレステロールを固相化した磁気ビーズ」と記載することもある。
上記のとおり調製したコレステロールを含むリポソームを固相化した磁気ビーズを、測定対象のHDL含有試料と溶液中で接触させることにより、磁気ビーズに固定されたリポソームからコレステロールを引き抜く。
本実施形態の測定方法において測定対象となるHDL含有試料は、哺乳動物(例えば、ヒト、ウシ、ブタ、ウマ、イヌ、ネコ、ヒツジ、ヤギ、ウサギ、ハムスター、モルモット、マウス、ラット等)由来、好ましくはヒト由来の単離されたHDL含有試料である。
HDL含有試料としては、全血、血清、血漿等の血液試料、apoB除去血清(apoB deleted serum:BDS)、精製HDL等を用いることができる。
また一実施形態では、後述の実施例に示すように、洗浄操作に代えて、ブランク補正を行ってもよい。ブランク補正を行うことにより、洗浄操作を行わなくても、洗浄操作を行った場合と同等の精度でCEC測定を行うことができうる。ブランク補正は、例えば、CEC測定において、測定対象のHDL含有試料に代えて、HDL含有試料の希釈媒体(例えば上記の水性媒体)をブランク試料として用いてCECを測定し、その値を、測定対象のCEC測定値から、ブランクとして差し引くことで行うことができる。
コレステロールを固相化した磁気ビーズと測定対象のHDL含有試料とを上記のとおり反応させると、溶液中のHDLの働きにより、磁気ビーズに固相化されたリポソームからコレステロールが引き抜かれて、HDLに取り込まれた状態で溶液中に移動することになる。
この引き抜かれたコレステロールを測定するために、まず、磁気ビーズを分離する。磁気ビーズの分離は、マグネットなどの磁力を用いて行えばよい。このように、本実施形態のCEC測定方法は、遠心分離操作を必要としないため、自動分析装置への適用が容易である。
また、磁気ビーズは、迅速な分離が可能なので測定時間の短縮が可能である。また、ゲルビーズは、遠心操作後に上清を分取する際、ゲルが浮遊して上清に混入する場合があるが、磁気ビーズであれば、磁気を用いて固定することにより、溶液から磁気ビーズを確実に分離することができる。
さらに、遠心操作が必要でないため、安定に分離できるというメリットもある。
なお、マグネットなどの磁力による分離に追加して、又は場合によっては磁力による分離に代えて、遠心分離操作を行ってもよい。
このように、ILG法において用いるゲルビーズは、遠心後、上清を回収する際に軽いゲルビーズが舞いやすく、正誤差の影響を受けるという課題があるが、本実施形態の磁気ビーズ(ILM)は、磁石を用いて容易に分離が可能であり、また、重く、かつ磁石で強く引き寄せることができるため、ゲルビーズのように舞うことがなく、より容易なプロセスで、より正確な測定が可能であるという利点がある。
コレステロール引き抜き能に関連する状態としては、脂質異常症、粥状動脈硬化症、家族性コレステロール血症、炎症性疾患(特には、コレステロール引き抜き能の低下が関与する炎症性疾患等)、糖尿病、心血管疾患イベント発症リスク等が挙げられる。
本実施形態のCEC測定用試薬キットは、コレステロールを固相化した磁気ビーズ、又はコレステロールを固相化した磁気ビーズの製造に用いるための磁気ビーズを少なくとも含む。本実施形態のCEC測定用試薬キットは、さらに、コレステロールを固相化した磁気ビーズの製造に用いるためのコレステロール、HDL含有試料の調製に用いるための各種試薬、コレステロールを固相化した磁気ビーズとHDL含有試料との反応に用いるための媒体、標識コレステロール量の測定に用いるための各種試薬等の本実施形態のCEC測定方法に用いられる任意の試薬、並びにそれらの試薬の保管及び反応のための容器等を含み得る。
したがって、本発明に係るCEC測定方法、CECの評価のための検査方法、CECの評価のための試験方法、及びCEC測定用試薬キットは、自動分析装置への適用が容易であり、臨床における日常検査への適用に適している。
(1) 固相化リポソーム結合ビーズの作製法
(調製例1)
固相化リポソーム結合磁気ビーズの作製
卵レシチン(10.6 mg)とコレステロール(2.3 mg)を6 mLのクロロホルムに溶解し、30 μLの0.5 mM蛍光標識コレステロール(4,4-difluoro-4-bora-3a,4a-s-indacene-labeled cholesterol,Avanti Polar Lipids)を溶液に加えた。窒素ガス下で形成された脂質膜をエーテルに溶解し、溶媒を蒸発により除去した。この工程を2回行った後、脂質膜を窒素ガス下で完全に乾燥させ、7 mLの10 mM Tris-HCl(pH 7.4)containing 150 mM NaCl and 1 mM EDTA-2Na(Buffer A)に懸濁した(リポソーム懸濁液)。その後、2.45 gの磁気ビーズ(コアシェル形状フェライト粉・MTFH-35,POWDERTECH、粒子サイズ:40.2 μm、孔径:0.8 μm)をリポソーム懸濁液に加え、室温で30分間反応させた。混合物に対して3, 5, 7, 9サイクルの凍結(-80℃)及び解凍(室温、水中)を繰り返し、ビーズ内へのリポソームの固相化を誘導した。最後に、Buffer Aで洗浄を5回行い、Buffer A 5 mLに再懸濁した。ビーズ混濁液は4℃の暗所で保存した。
ILG(固相化リポソーム結合ゲルビーズ)の作製
2.45 gの磁気ビーズに代えて0.35 gの乾燥したゲルビーズ(Sephacryl S-300 gel beads,GE Healthcare)をリポソーム懸濁液に加えたこと、及び、凍結・解凍を7サイクル行ったこと以外は、調製例1と同様にして、ILG(固相化リポソーム結合ゲルビーズ)を調製した。
血清に20%ポリエチレングリコール溶液(200 mM glycine buffer; pH 7.4)を血清に対し100:40の割合で添加し、インキュベーション後(20 min, 室温)、遠心(10,000 rpm, 30 min, 4℃)し、上清をBDSとして回収した。
蛍光標識コレステロールを含む固相化リポソーム結合磁気ビーズ懸濁液100 μL(調製例1)又は固相化リポソーム結合ゲルビーズ懸濁液100 μL(比較調製例1)に対し、希釈したHDL(測定試料)又はBDS(参照試料)を150 μL添加し、それぞれ最終総コレステロール濃度 4~15 mg/dL(HDLの場合)、最終血清濃度2%(BDSの場合)になるようにした。遮光条件下でインキュベーション(16h、30℃)し、固相化リポソーム結合磁気ビーズはマグネットを用いて、固相化リポソーム結合ゲルビーズは遠心分離を用いて、反応溶液から分離し、得られた上清 75 μL の蛍光強度(Ex: 485 nm, Em: 538 nm)を測定した。
なお、(4)以降の実施例においても、特記しない限り、CEC測定は本実施例と同様の方法で行った。
(1) 凍結融解によるリポソームの固相化
調製例1において、磁気ビーズに蛍光標識コレステロール含有リポソームが固相化されていることを確認するため、凍結融解後、洗浄前の上清を回収した。反応前のリポソーム懸濁液の蛍光強度と、磁気ビーズと反応後の上清の蛍光強度を比較することで、ビーズ内の蛍光強度を評価した。
図1に示すのは、凍結回数を変え、固相化されたリポソームの量を比較したものである。結果は、凍結融解7回に対するビーズ内の蛍光強度の比で示す。凍結融解の繰り返しにより、リポソームが固相化された。
調製例1において、蛍光標識コレステロール含有リポソームを磁気ビーズに固相化、洗浄した後、固相化磁気ビーズを蛍光顕微鏡で観察した(図2)。磁気ビーズ内に蛍光が観察され、リポソームが固相化されていることが確認できた。
本発明の磁性ビーズと従来法のゲルビーズのリポソーム固相化効率を比較した結果を図3に示す。
図3中、「M」は、調製例1(凍結融解5回)と同様にして調製した固相化リポソーム結合磁性ビーズの固相化量(調製2回(各回3チューブ、計6チューブ)の平均)を示し、「ILG」は、比較調製例1(凍結融解7回)と同様にして調製した固相化リポソーム結合ゲルビーズの固相化量(調製2回(各回2チューブ、計4チューブ)の平均)を示す。ゲルビーズと同等又はそれ以上の効率で、磁気ビーズにリポソームを固相化できることが確認された。
調製例1の凍結回数5回の磁気ビーズを用いて、濃度の異なるHDLのコレステロール引き抜き能を評価した。蛍光コレステロールの引き抜き能はHDL濃度依存的に増加した(図4)。
様々なHDL濃度(24系列、1~15 mg/mL)を用いて、ILGを用いた従来法と磁気ビーズを用いた本発明の方法のCECの相関性を調べた(図5)。各試料のCECは、同一の参照血清を用いて測定したCECで規格化した。本発明の方法は従来法と良好な相関性を示した(r = 0.987)。なお、本実施例に示したILGを用いた従来法は、培養細胞を用いた従来のCEC評価法との相関が高いことが報告されている(非特許文献4、r =0.932)。
調製例1の凍結回数5回の磁気ビーズと同様にして蛍光コレステロール含有リポソームを磁気ビーズに固相化させた。このILMを4℃、暗所にて0~60日間まで保管し、それぞれの保存日数における上清の蛍光強度を測定した。上清の蛍光強度(保存中に遊離した蛍光コレステロール含有リポソーム量を反映)は60日間にかけて微増するものの、0日と比較して45日までは有意な差は認められなかった(図6)。それぞれの日数につき3つのチューブで保管し、さらにそれぞれの蛍光強度は3重測定した。* p < 0.05。
調製例1の凍結回数5回の磁気ビーズと同様にして蛍光コレステロール含有リポソームを磁気ビーズに固相化させた。このILMを用いて、緩衝液のみ(blank)、参照血清、血清サンプルA, B, CのそれぞれのCECを測定した。CEC測定の際に、磁気ビーズを緩衝液で洗浄操作あり(白い棒グラフ)と洗浄なし(黒い棒グラフ)のCECを比較した(図7上図)。洗浄操作は、ILMをマグネットを用いて回収し、同容量のBuffer Aに再懸濁する操作を2回繰り返すことで行った。磁気ビーズを洗浄操作の有無によってほとんどのサンプル(血清B以外)でCECに有意な差が認められた(*p < 0.05, **p < 0.01)。これは、洗浄しない場合、磁気ビーズから遊離した蛍光強度を測り込むからである。一方、参照血清、血清A, B, CのCECをブランクの値で差し引くと(ブランク補正)(図7下図)、洗浄操作の有無で有意差は認められなかった。つまり、洗浄操作をしなくても、ブランク補正を行うことにより、CECの測定が可能である。
超遠心で得られたHDLを0.6 - 9.0 mg cholesterol/dLの範囲で24系列希釈した後、それぞれの希釈したHDLのCECを測定したところ、1.2 - 7.8 mg/dLまで直線性が認められた(図8上図、TC:トータルコレステロール)。同様にBDSを0.2 - 5.0%の範囲に希釈し、測定すると、1.0 - 5.0%の範囲まで直線性が認められた(図8下図)。これは血清HDLコレステロール濃度に換算すると、およそ34 - 223 mg/dLまで測定できることを意味し、健常者および患者検体を十分に測定することが可能である。
上記(8)において直線性の認められた濃度範囲のサンプルを用いて、ILG法とILM法の相関性試験を行った(図9上図:HDL、図9下図:BDS)。どちらも良好な相関性が認められた。
15名の健常者BDS中のCECをILM法で測定し(3重測定)、それぞれのHDL-C濃度との相関性を確認した。CECとHDL-Cには正の相関性が認められるが、中には同程度のHDL-C濃度であってもCECが異なる検体があり、HDLの能力の違いを反映するものと考えられる(図10上図)。また、ILM法とILG法は良好な相関性を示した(図10下図)。
3種類の濃度の異なるBDSのCECをILM法において、同日中にそれぞれ20重測定したところ,CV%はいずれも7%以内であった(表1:Sample 1-3)。また、同様に3種類のBDSを20日間測定したところ、CV%は10%以内であった(表1:Sample 4-6)。従来の細胞法を用いた場合、これらのCV%は10%を超え(データ図示せず)、本発明のILM法の再現性が従来法に比べて良好であることが示された。
Claims (15)
- HDLのコレステロール引き抜き能を測定する方法であって、
コレステロールを固相化した磁気ビーズを用意する工程、
前記コレステロールを固相化した磁気ビーズと測定対象のHDL含有試料とを溶液中で接触させる工程、及び
前記溶液から磁気ビーズを分離し、磁気ビーズ分離後の溶液中のコレステロール量を測定する工程
を含む、方法。 - 磁気ビーズが多孔質磁気粒子である、請求項1に記載の方法。
- 前記コレステロールを固相化した磁気ビーズを用意する工程が、
コレステロールを含むリポソームを調製する工程、及び
前記リポソームと磁気ビーズとを含む溶液を凍結融解させる工程
を含む、請求項1又は2に記載の方法。 - 磁気ビーズに固相化されたコレステロールの少なくとも一部が蛍光標識されている、請求項1~3のいずれか1項に記載の方法。
- HDLのコレステロール引き抜き能を測定する方法であって、
少なくとも一部が標識されたコレステロールを含むリポソームを調製する工程、及び
前記リポソームと多孔質磁気粒子とを含む溶液を凍結融解させる工程
を含む工程により、コレステロールを固相化した磁気ビーズを用意する工程と、
前記コレステロールを固相化した磁気ビーズと測定対象のHDL含有試料とを溶液中で接触させる工程と、
前記溶液から磁気ビーズを分離し、磁気ビーズ分離後の溶液中のコレステロール量を、溶液の標識強度に基づき測定する工程と
を含む、請求項1~4のいずれか1項に記載の方法。 - コレステロールを固相化した磁気ビーズが、該磁気ビーズの懸濁液から採取した懸濁媒体の標識強度が少なくとも10日間実質的に変化しないことによって表される保存安定性を有する、請求項5に記載の方法。
- 磁気ビーズが磁性セラミックス粒子である、請求項1~6のいずれか1項に記載の方法。
- 溶液から磁気ビーズを分離する工程が、遠心分離操作を含まない、請求項1~7のいずれか1項に記載の方法。
- 測定対象のHDL含有試料のコレステロール量測定値から、ブランク試料のコレステロール量測定値を差し引くことで、ブランク補正を行う工程をさらに含む、請求項1~8のいずれか1項に記載の方法。
- コレステロールを固相化した磁気ビーズと測定対象のHDL含有試料とを溶液中で接触させる工程の前に、コレステロールを固相化した磁気ビーズを洗浄する工程を含まない、請求項9に記載の方法。
- 測定対象のHDL含有試料が、全血、血清、血漿、apoB除去血清、又は精製HDLを含む溶液である、請求項1~10のいずれか1項に記載の方法。
- 無細胞系で実施される、請求項1~11のいずれか1項に記載の方法。
- 同一サンプルの少なくとも10日間にわたるコレステロール量測定値の変動係数が10%以下であることによって表される再現性を有する、請求項1~12のいずれか1項に記載の方法。
- コレステロールを固相化した磁気ビーズを含む、HDLのコレステロール引き抜き能測定用試薬キット。
- コレステロールを固相化した磁気ビーズを用いて、被験者由来のHDL含有試料のコレステロール引き抜き能を測定する工程を含む、被験者のコレステロール引き抜き能に関連する状態又はそのリスクに関する情報を取得する方法。
