Abstract
本発明は、良好な製剤安定性を有するベタメタゾン吉草酸エステル含有軟膏剤組成物を提供する。ベタメタゾン吉草酸エステル及び親油性基剤、又はベタメタゾン吉草酸エステル、親油性基剤及びクロタミトンに、HLB5.0以下の非イオン性界面活性剤を配合することで、上記軟膏剤組成物を調製することができる。
Description
本発明は、ベタメタゾン吉草酸エステルを含有する軟膏剤組成物に関する。また本発明は、ベタメタゾン吉草酸エステルを含有する軟膏剤組成物の製剤安定性を向上する方法に関する。
ステロイド骨格を有する化合物は、微量で高い生理活性を示すことが知られている。なかでも副腎皮質ホルモンは、炎症を伴う皮膚の痒み、赤み、及び腫れなどの症状の改善に有効であり、湿疹(アトピー性皮膚炎による湿疹を含む。以下、同じ。)、痒疹(汗疹、蕁麻疹、かぶれ、及び虫さされ等による痒疹を含む。以下、同じ。)、皮膚炎、および乾癬等の治療に広く用いられている。通常、副腎皮質ホルモンを含む外用製剤には、その他の有効成分として、非ステロイド系抗炎症剤、局所麻酔剤、血行促進剤、殺菌剤、皮膚保護剤、抗生物質、及び清涼剤などが、適宜に組み合わせて配合される。しかし、配合成分の組み合わせによっては、製剤的安定性を確保することが難しくなったり、皮膚刺激が惹起されたり、また使用感が悪いものになる等の製剤上の問題が発生する。
このため、製剤の設計には、薬理作用(効能・効果)に加えて、製剤の物理的安定性、皮膚等の生体への安全性、及び使用感等を総合的に考慮する必要がある。
前述する副腎皮質ホルモンのうち、17位または21位にヒドロキシル基を有するステロイドがアセチル化またはバレリル化してなるエステル化ステロイドは、併用する化合物によって、製剤中で分解されやすくなる等(特許文献1及び2参照)、安定性が低下することが知られている。
本発明は、良好な製剤安定性を有するベタメタゾン吉草酸エステル含有軟膏剤組成物を提供することを課題とする。また、本発明は、当該ベタメタゾン吉草酸エステル含有軟膏剤組成物の製剤安定性を改善又は向上する方法を提供することを課題とする。
本発明者らは、ベタメタゾン吉草酸エステルを含有する軟膏剤の設計に際して、親油性基剤をベタメタゾン吉草酸エステルと共存させることで、室温条件下で経時的に液状成分が滲み出してくること、つまりブリーディング現象が生じることを知見した。そこで、これを解消すべく、鋭意検討を重ねていたところ、ベタメタゾン吉草酸エステル、及び親油性基剤に、さらにHLB5.0以下の非イオン性界面活性剤を併用することで、前記ブリーディング現象が有意に改善され、製剤安定性が改善又は向上したベタメタゾン吉草酸エステル含有軟膏剤が得られることを見出した。
本発明は、これらの知見に基づいて、さらなる改良を重ねて完成したものであり、下記の実施形態を包含するものである。
(I)ベタメタゾン吉草酸エステル含有軟膏剤組成物
(I-1)ベタメタゾン吉草酸エステル、親油性基剤、及びHLB5.0以下、好ましくはHLB1.0~5.0の非イオン性界面活性剤を含有する軟膏剤組成物。

本発明によれば、良好な製剤安定性を有するベタメタゾン吉草酸エステル含有軟膏剤組成物を提供することができる。より詳細には、本発明によれば、親油性基剤を配合することで経時的に生じるブリーディングが、HLB5.0以下の非イオン性界面活性剤を配合することで有意に抑制されてなるベタメタゾン吉草酸エステル含有軟膏剤組成物を提供することができる。
また本発明によれば、ベタメタゾン吉草酸エステル及びクロタミトンを含有する、製剤安定性が良好な軟膏剤組成物を提供することができる。より詳細には、本発明によれば、ベタメタゾン吉草酸エステル及びクロタミトンに加えて親油性基剤を配合することで経時的に生じるクロタミトンの偏析が、HLB5.0以下の非イオン性界面活性剤を配合することで有意に抑制され、クロタミトンが経時的に分離することなく親油性基剤中に安定して分散してなる軟膏剤組成物を提供することができる。
(I)ベタメタゾン吉草酸エステル含有軟膏剤組成物
本発明のベタメタゾン吉草酸エステル含有軟膏剤組成物(以下、単に「本組成物」とも称する)は、下記の(A)、(B)及び(C)成分を含有する組成物である。
(A)ベタメタゾン吉草酸エステル
(B)親油性基剤、及び
(C)HLB5.0以下の非イオン性界面活性剤。
本発明のベタメタゾン吉草酸エステル含有軟膏剤組成物(以下、単に「本組成物」とも称する)は、下記の(A)、(B)及び(C)成分を含有する組成物である。
(A)ベタメタゾン吉草酸エステル
(B)親油性基剤、及び
(C)HLB5.0以下の非イオン性界面活性剤。
本組成物は、前記(A)及び(B)成分に加えて、下記の(D)成分を含有していてもよい。
(D)クロタミトン。
(D)クロタミトン。
以下、これらの成分について説明する。
(A)成分
ベタメタゾン吉草酸エステルは、17位のヒドロキシル基がバレリル化され、21位のヒドロキシル基がアセチル化されてなるエステル化ステロイドである。当該化合物は、抗炎症作用を有するストロングなステロイドであり、下記の症状を改善する効能・効果が知られている:
湿疹・皮膚炎群(進行性指掌角皮症、女子顔面黒皮症、ビダール苔癬、放射線皮膚炎、日光皮膚炎を含む)、皮膚そう痒症、痒疹群(じん麻疹様苔癬、ストロフルス、固定じん麻疹を含む)、虫さされ、乾癬、掌蹠膿疱症、扁平苔癬、光沢苔癬、毛孔性紅色粃糠疹、ジベルバラ色粃糠疹、紅斑症(多形滲出性紅斑、結節性紅斑、ダリエ遠心性環状紅斑)、紅皮症(悪性リンパ腫による紅皮症を含む)、慢性円板状エリテマトーデス、薬疹・中毒疹、円形脱毛症(悪性を含む)、熱傷(瘢痕、ケロイドを含む)、凍瘡、天疱瘡群、ジューリング疱疹状皮膚炎(類天疱瘡を含む)、痔核、鼓室形成手術・内耳開窓術・中耳根治手術の術創。
(A)成分

本組成物中に含まれる(A)成分の量は、本発明の効果を妨げることなく、所望の薬効を発揮する量であればよく、その範囲で適宜設定調整することができる。例えば、本組成物を、炎症を伴う皮膚の痒み、赤みまたは腫れなどの症状を有する皮膚疾患(例えば、湿疹、痒疹、皮膚炎または乾癬等)を改善する外用製剤(医薬製剤)として調製する場合、本組成物中の(A)成分の含有量としては、0.0002~10質量%の範囲を例示することができる。好ましくは0.002~2質量%であり、より好ましくは0.01~0.3質量%である。
(B)成分
(B)成分としては、軟膏剤の調製に一般的に使用される親油性基剤であればよい。その限りにおいて、特に制限されないものの、例えば、ワセリン、パラフィン、プラスチベース、ラノリン、及びミツロウなどが例示される。好ましくはワセリン、パラフィン、及びプラスチベース等の炭化水素系油からなる親油性基剤である。前記ワセリンには、白色ワセリン及び黄色ワセリンが含まれる。前記パラフィンには、流動パラフィンが含まれる。より好ましくは白色ワセリン、流動パラフィン、及びプラスチベースを挙げることができる。これらの親油性基剤は、1種を単独で使用することもできるし、また2種以上を任意に組み合わせて使用することもできる。
(B)成分としては、軟膏剤の調製に一般的に使用される親油性基剤であればよい。その限りにおいて、特に制限されないものの、例えば、ワセリン、パラフィン、プラスチベース、ラノリン、及びミツロウなどが例示される。好ましくはワセリン、パラフィン、及びプラスチベース等の炭化水素系油からなる親油性基剤である。前記ワセリンには、白色ワセリン及び黄色ワセリンが含まれる。前記パラフィンには、流動パラフィンが含まれる。より好ましくは白色ワセリン、流動パラフィン、及びプラスチベースを挙げることができる。これらの親油性基剤は、1種を単独で使用することもできるし、また2種以上を任意に組み合わせて使用することもできる。
本組成物中に含まれる(B)成分の量は、本発明の効果を妨げることなく、本組成物を軟膏剤の形状に成形することができる量であればよく、その範囲で適宜設定調整することができる。例えば、本組成物中の(B)成分の含有量としては、10~99.79質量%の範囲から適宜選択調製することができる。好ましくは80~99.7質量%であり、より好ましくは90~99質量%である。
(C)成分
(C)成分として配合するHLB5.0以下の非イオン性界面活性剤としては、外用医薬品、医薬部外品または化粧料に配合可能なものである。
かかる非イオン性界面活性剤としては、制限されないものの、ソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルを挙げることができる。これらの界面活性剤を構成する脂肪酸としては、炭素数5~25の脂肪酸を挙げることができる。例えば、ソルビタン脂肪酸エステルには、HLBが5.0以下であることを限度として、ソルビタンモノオレエート、ソルビタンモノイソステアレート、ソルビタンモノラウレート、ソルビタンモノパルミテート、ソルビタンモノステアレート、ソルビタンセスキオレエート、ソルビタントリオレエート、ペンタ-2-エチルヘキシル酸ジグリセロールソルビタン、及びテトラ-2-エチルヘキシル酸ジグリセロールソルビタンが含まれる。またグリセリン脂肪酸エステルには、HLBが5.0以下であることを限度として、カプリル酸グリセリン、カプリン酸グリセリン、ラウリン酸グリセリン、パルミチン酸グリセリン、オレイン酸グリセリン、ステアリン酸グリセリン、ジカプリル酸グリセリン、及びトリカプリル酸グリセリンが含まれる。これらのソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルは単独で使用されても、また2種以上を任意に組み合わせて用いてもよい。ソルビタン脂肪酸エステル及びグリセリン脂肪酸エステルのHLBとして、いずれも好ましくは1.0~5.0の範囲であり、より好ましくは2.0~5.0の範囲である。
(C)成分として配合するHLB5.0以下の非イオン性界面活性剤としては、外用医薬品、医薬部外品または化粧料に配合可能なものである。
かかる非イオン性界面活性剤としては、制限されないものの、ソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルを挙げることができる。これらの界面活性剤を構成する脂肪酸としては、炭素数5~25の脂肪酸を挙げることができる。例えば、ソルビタン脂肪酸エステルには、HLBが5.0以下であることを限度として、ソルビタンモノオレエート、ソルビタンモノイソステアレート、ソルビタンモノラウレート、ソルビタンモノパルミテート、ソルビタンモノステアレート、ソルビタンセスキオレエート、ソルビタントリオレエート、ペンタ-2-エチルヘキシル酸ジグリセロールソルビタン、及びテトラ-2-エチルヘキシル酸ジグリセロールソルビタンが含まれる。またグリセリン脂肪酸エステルには、HLBが5.0以下であることを限度として、カプリル酸グリセリン、カプリン酸グリセリン、ラウリン酸グリセリン、パルミチン酸グリセリン、オレイン酸グリセリン、ステアリン酸グリセリン、ジカプリル酸グリセリン、及びトリカプリル酸グリセリンが含まれる。これらのソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルは単独で使用されても、また2種以上を任意に組み合わせて用いてもよい。ソルビタン脂肪酸エステル及びグリセリン脂肪酸エステルのHLBとして、いずれも好ましくは1.0~5.0の範囲であり、より好ましくは2.0~5.0の範囲である。
本組成物中に含まれる(C)成分の量は、本発明の効果を奏する範囲であればよく、その範囲で適宜設定調整することができる。例えば、本組成物中の(C)成分の含有量としては、0.1~20質量%の範囲から適宜選択調製することができる。好ましくは1~10質量%であり、より好ましくは3~8質量%である。
(D)成分
本組成物は、前述する(A)~(C)成分に加えて、(D)成分としてクロタミトンを含有することができる。本組成物中に含まれる(D)成分の量は、本発明の効果を妨げない範囲であればよく、その範囲で適宜設定調整することができる。例えば、本組成物中の(D)成分の含有量としては、0.1~20質量%の範囲から適宜選択調製することができる。好ましくは1~15質量%であり、より好ましくは5~10質量%である。
本組成物は、前述する(A)~(C)成分に加えて、(D)成分としてクロタミトンを含有することができる。本組成物中に含まれる(D)成分の量は、本発明の効果を妨げない範囲であればよく、その範囲で適宜設定調整することができる。例えば、本組成物中の(D)成分の含有量としては、0.1~20質量%の範囲から適宜選択調製することができる。好ましくは1~15質量%であり、より好ましくは5~10質量%である。
(E)任意の薬理活性成分
本組成物には、本発明の効果を妨げないことを限度として、前述する成分に加えて、さらに他の薬理活性成分が含まれていてもよい。制限されないものの、非ステロイド系抗炎症剤、局所麻酔剤、鎮痒剤、血行促進剤、殺菌剤、皮膚保護剤、抗生物質、保湿剤、及び/又は清涼剤を例示することができる。これらは、外用剤、特に皮膚外用剤に配合できる成分であればよいが、本組成物を、炎症を伴う皮膚の痒み、赤みまたは腫れなどの症状を有する皮膚疾患(例えば、湿疹、痒疹、皮膚炎または乾癬等)を改善する外用製剤(医薬製剤)として調製する場合は、当該外用製剤に配合することができる成分であることが好ましい。
本組成物には、本発明の効果を妨げないことを限度として、前述する成分に加えて、さらに他の薬理活性成分が含まれていてもよい。制限されないものの、非ステロイド系抗炎症剤、局所麻酔剤、鎮痒剤、血行促進剤、殺菌剤、皮膚保護剤、抗生物質、保湿剤、及び/又は清涼剤を例示することができる。これらは、外用剤、特に皮膚外用剤に配合できる成分であればよいが、本組成物を、炎症を伴う皮膚の痒み、赤みまたは腫れなどの症状を有する皮膚疾患(例えば、湿疹、痒疹、皮膚炎または乾癬等)を改善する外用製剤(医薬製剤)として調製する場合は、当該外用製剤に配合することができる成分であることが好ましい。
(F)任意の担体・添加剤
本組成物は、前述する成分以外に、本発明の効果を妨げないことを限度として、適宜、軟膏剤の製造に使用される従来公知の担体や添加剤を任意に配合することができる。これらの担体や添加剤としては、例えば賦形剤、増粘剤、抗酸化剤、緩衝剤、乳化剤、pH調整剤、分散剤、溶解補助剤、流動化剤、着色剤、保存剤、及び/又は香料等を、制限なく、例示することができる。
本組成物は、前述する成分以外に、本発明の効果を妨げないことを限度として、適宜、軟膏剤の製造に使用される従来公知の担体や添加剤を任意に配合することができる。これらの担体や添加剤としては、例えば賦形剤、増粘剤、抗酸化剤、緩衝剤、乳化剤、pH調整剤、分散剤、溶解補助剤、流動化剤、着色剤、保存剤、及び/又は香料等を、制限なく、例示することができる。
本組成物は、前述する成分のうち、少なくとも(A)~(C)成分または(A)~(D)成分を、必要に応じて(E)成分及び/又は(F)成分と混合して、軟膏剤の慣用の製造方法に従って、軟膏剤の形態に製造することができる。また、製造された軟膏剤(膏体)は、慣用のチューブ容器に充填して軟膏剤製剤として提供することができる。
斯くして軟膏剤の形態に調製される本組成物は、炎症を伴う皮膚の痒み、赤みまたは腫れなどの症状を有する皮膚疾患(例えば、湿疹、痒疹、皮膚炎または乾癬等)を改善する外用製剤(医薬製剤)として好適に用いることができる。適用部は、制限されず、手、足、指、顔、頭、及び体など、皮膚全般に対して広く用いることができる。また肛門部や陰部周辺などの皮膚の角質が薄い部位にも適用することができる。これらの皮膚への適用量や用法も特に制限されず、患部の症状や有効成分の含有量に応じて、一日1回~数回、適量を皮膚などの外皮に塗布することなどにより用いることができる。制限されないものの、本組成物1回塗布量(単回投与量)に含まれる(A)成分の量として10~1000mgの範囲を例示することができる。この範囲で皮膚疾患の種類や患部の症状に応じて、適宜選択することができる。
(II)ベタメタゾン吉草酸エステル含有軟膏剤組成物の安定性改善方法
本発明は、前述する(A)成分を含有する軟膏剤組成物(ベタメタゾン吉草酸エステル含有軟膏剤組成物)について、その安定性を改善する方法を提供する。
後述する実験例に示すように、(A)成分に(B)成分を配合することで、25℃条件下で保存した場合でも、安定性が低下し、経時的にブリーディング現象が生じる。なお、ブリーディング現象とは液状成分が滲み出てくる現象(離漿現象)を意味する。この現象は、(A)成分及び(B)成分に、低融点物質である(D)クロタミトン(融点<25℃)を配合することで、さらに顕著に生じる。またこの場合、膏体中で(D)成分が偏析するという問題も生じる。こうしたブリーディング現象や偏析という問題は、これらの組成物((A)及び(B)成分含有組成物、並びに(A)、(B)及び(D)成分含有組成物)中に、前述する(C)成分を配合し、当該成分を共存させることで顕著に改善することができる。
本発明は、前述する(A)成分を含有する軟膏剤組成物(ベタメタゾン吉草酸エステル含有軟膏剤組成物)について、その安定性を改善する方法を提供する。
後述する実験例に示すように、(A)成分に(B)成分を配合することで、25℃条件下で保存した場合でも、安定性が低下し、経時的にブリーディング現象が生じる。なお、ブリーディング現象とは液状成分が滲み出てくる現象(離漿現象)を意味する。この現象は、(A)成分及び(B)成分に、低融点物質である(D)クロタミトン(融点<25℃)を配合することで、さらに顕著に生じる。またこの場合、膏体中で(D)成分が偏析するという問題も生じる。こうしたブリーディング現象や偏析という問題は、これらの組成物((A)及び(B)成分含有組成物、並びに(A)、(B)及び(D)成分含有組成物)中に、前述する(C)成分を配合し、当該成分を共存させることで顕著に改善することができる。
本発明が対象とする「ベタメタゾン吉草酸エステル含有軟膏剤組成物の安定性改善」には、好ましくは、ベタメタゾン吉草酸エステル含有組成物に(B)成分を配合することによって生じる経時的なブリーディング現象が抑制されることが含まれる。少なくとも(A)~(C)成分または(A)~(D)成分を含む組成物(測定対象)が、(C)成分を含まない以外は前記と同じ成分からなる組成物(比較対象)と比較して、経時的に生じる離漿量が少ない場合、前者の組成物は、(C)成分が配合されていることによりブリーディング現象が抑制されており、安定性が改善されていると判断することができる。
経時的ブリーディング現象の有無は、前記測定対象の組成物と比較対象の組成物とを、それぞれ25℃、60%RHの恒温恒湿条件の暗所条件下に1~4週間程度保存し、その期間中、経時的に離漿量を測定し、その離漿量を比較することで評価することができる。詳細は、後述する実験例1の記載を参照することができる。
経時的ブリーディング現象の有無は、前記測定対象の組成物と比較対象の組成物とを、それぞれ25℃、60%RHの恒温恒湿条件の暗所条件下に1~4週間程度保存し、その期間中、経時的に離漿量を測定し、その離漿量を比較することで評価することができる。詳細は、後述する実験例1の記載を参照することができる。
また本発明が対象とする「ベタメタゾン吉草酸エステル含有軟膏剤組成物の安定性改善」には、好ましくは、ベタメタゾン吉草酸エステル含有組成物に(B)及び(D)成分を配合することによって生じる、(D)成分の経時的な偏析が抑制されることが含まれる。少なくとも(A)~(D)成分を含む組成物(測定対象)が、(C)成分を含まない以外は前記と同じ成分からなる組成物(比較対象)と比較して、経時的に生じる(D)成分の偏析が少ない場合、前者の組成物は、(C)成分が配合されていることにより(D)成分の偏析が抑制されており、安定性が改善されていると判断することができる。
(D)成分の経時的な偏析の有無は、前記測定対象の組成物と比較対象の組成物とを、それぞれ40℃、75%RHの恒温恒湿条件の暗所条件下に3ヶ月程度保存し、その後、膏体中の(D)成分含量の偏析を測定し、その偏析の程度を比較することで評価することができる。詳細は、後述する実験例2の記載を参照することができる。
(D)成分の経時的な偏析の有無は、前記測定対象の組成物と比較対象の組成物とを、それぞれ40℃、75%RHの恒温恒湿条件の暗所条件下に3ヶ月程度保存し、その後、膏体中の(D)成分含量の偏析を測定し、その偏析の程度を比較することで評価することができる。詳細は、後述する実験例2の記載を参照することができる。
(A)及び(B)成分(または(A)、(B)及び(D)成分)を含有する組成物に、(C)成分を共存させる方法としては、本発明の効果が得られる方法であればよく、制限されない。制限されないものの、例えば、(B)成分と(C)成分との共存状態を形成し、これに(A)成分または(A)成分と(D)成分を順番に分散させる方法であってもよい。また、本発明の効果を妨げないことを限度として、他の成分として、前述する(E)成分及び/又は(F)成分を配合することもできる。これらの各成分を混合し、少なくとも(A)~(C)成分又は(A)~(D)成分との共存状態を形成した後、慣用の方法により、軟膏剤の形態に調製する。組成物中の各成分の割合、及び配合比などは、前記(I)に記載した通りであり、当該記載はここに援用することができる。
以上、本明細書において、「含む」及び「含有する」の用語には、「からなる」及び「から実質的になる」という意味が含まれる。
以下、本発明の構成及び効果について、その理解を助けるために、実験例を用いて本発明を説明する。但し、本発明はこれらの実験例によって何ら制限を受けるものではない。以下の実験は、特に言及しない限り、室温(25±5℃)、及び大気圧条件下で実施した。なお、特に言及しない限り、以下に記載する「%」は「質量%」、「部」は「質量部」を意味する。
以下の実験例で使用した化合物は下記の通りである。
ベタメタゾン吉草酸エステル(仮比重(密度)0.16g/mL):Sicorより入手
クロタミトン(比重0.98、融点25℃以下):住友化学(株)より入手
流動パラフィン(比重0.88):SONNEBORNより入手
白色ワセリン(比重0.85):CALUMET(商品名:スノーホワイトV)より入手ソルビタン脂肪酸エステル:NIKKOL SS-15V(HLB5.0:日光ケミカルズ(株)より入手)
グリセリン脂肪酸エステル:SYグリスターTS-3S(HLB4.0:阪本薬品工業(株)より入手)
ベタメタゾン吉草酸エステル(仮比重(密度)0.16g/mL):Sicorより入手
クロタミトン(比重0.98、融点25℃以下):住友化学(株)より入手
流動パラフィン(比重0.88):SONNEBORNより入手
白色ワセリン(比重0.85):CALUMET(商品名:スノーホワイトV)より入手ソルビタン脂肪酸エステル:NIKKOL SS-15V(HLB5.0:日光ケミカルズ(株)より入手)

実験例1 ベタメタゾン吉草酸エステル含有軟膏剤組成物の調製、並びにその評価
ベタメタゾン吉草酸エステル含有軟膏組成物を調製し、ブリーディング(離漿性、滲み出し)、及び展延性を評価した。
ベタメタゾン吉草酸エステル含有軟膏組成物を調製し、ブリーディング(離漿性、滲み出し)、及び展延性を評価した。
(1)軟膏剤の調製
表1に記載する成分を混合して、半固形状のベタメタゾン吉草酸エステル含有軟膏組成物(比較例1~2、実施例1~4)を調製した。実施例1及び2並びに比較例1は、(B)成分に(C)成分を溶解し、この中に(A)成分を添加混合して調製した。また実施例3及び4並びに比較例2は、(B)成分に(C)成分を溶解し、この中に(A)成分、及び(D)成分を順番に分散して調製した。斯くして調製した軟膏剤(膏体)を、調製後すみやかに後述する実験に供した。
表1に記載する成分を混合して、半固形状のベタメタゾン吉草酸エステル含有軟膏組成物(比較例1~2、実施例1~4)を調製した。実施例1及び2並びに比較例1は、(B)成分に(C)成分を溶解し、この中に(A)成分を添加混合して調製した。また実施例3及び4並びに比較例2は、(B)成分に(C)成分を溶解し、この中に(A)成分、及び(D)成分を順番に分散して調製した。斯くして調製した軟膏剤(膏体)を、調製後すみやかに後述する実験に供した。
(2)ブリーディング評価
(a)試験方法
コーンメッシュ(メッシュ孔径:45μm、φ50mm×H60mm)の円錐先端部の中に、前記で調製した各軟膏剤(膏体)10gを充填し、これをガラスビーカーで受けた状態で、25℃/60%RHの環境下で4週間静置保存して、コーンメッシュの金網先端部から滴下する液体(低融点物質)の量を経時的に測定した。1週間後、及び4週間後に測定した滴下量(g)から、液状部分の滲み出し易さの指標であるブリーディング率(%)を算出した。
(a)試験方法
コーンメッシュ(メッシュ孔径:45μm、φ50mm×H60mm)の円錐先端部の中に、前記で調製した各軟膏剤(膏体)10gを充填し、これをガラスビーカーで受けた状態で、25℃/60%RHの環境下で4週間静置保存して、コーンメッシュの金網先端部から滴下する液体(低融点物質)の量を経時的に測定した。1週間後、及び4週間後に測定した滴下量(g)から、液状部分の滲み出し易さの指標であるブリーディング率(%)を算出した。
なお、1週間後及び4週間後のブリーディング率(%)は、具体的には、静置保存から1週間後及び4週間後に、コーンメッシュからの液体滴下を受けたガラスビーカーの質量を測定し、下記式に基づいて、算出した。

(b)試験結果
結果を図1に示す。図1Aは比較例1及び実施例1~2の結果を、図1Bは比較例2及び実施例3~4の結果を示す。
図1に示すように、ベタメタゾン吉草酸エステルを親油性基剤と共存させることで、経時的に液体が滲み出てくること(ブリーディング現象)が確認された(比較例1)。なお、拘束されるものではないが、滲み出てきた液体は、流動パラフィン及び白色ワセリン中の低融点物質に由来するものと考えられる。また当該ブリーディング現象は、ベタメタゾン吉草酸エステルに低融点物質であるクロタミトンを共存させることでより増大することが確認された(比較例2)。比較例1及び2のベタメタゾン吉草酸エステル含有軟膏剤でみられたブリーディング現象は、HLBが5以下の非イオン性界面活性剤を共存させておくことで、いずれも顕著に改善され、軟膏剤の保存安定性(親油性基剤中でのベタメタゾン吉草酸エステル、及びクロタミトンの保存安定性)が向上することが確認された。
結果を図1に示す。図1Aは比較例1及び実施例1~2の結果を、図1Bは比較例2及び実施例3~4の結果を示す。
図1に示すように、ベタメタゾン吉草酸エステルを親油性基剤と共存させることで、経時的に液体が滲み出てくること(ブリーディング現象)が確認された(比較例1)。なお、拘束されるものではないが、滲み出てきた液体は、流動パラフィン及び白色ワセリン中の低融点物質に由来するものと考えられる。また当該ブリーディング現象は、ベタメタゾン吉草酸エステルに低融点物質であるクロタミトンを共存させることでより増大することが確認された(比較例2)。比較例1及び2のベタメタゾン吉草酸エステル含有軟膏剤でみられたブリーディング現象は、HLBが5以下の非イオン性界面活性剤を共存させておくことで、いずれも顕著に改善され、軟膏剤の保存安定性(親油性基剤中でのベタメタゾン吉草酸エステル、及びクロタミトンの保存安定性)が向上することが確認された。
(3)展延性の評価
(a)試験方法
前記で調製した軟膏剤(膏体)(比較例1及び2、実施例3及び4)を25℃/60%RHの環境下で8週間保存した後、展延性(伸びやすさ)を、スプレッドメーター(井元製作所製)を用いて、23~25℃条件下での流動性を測定することで評価した。
(b)試験結果
結果を表2に記載する。なお、表2には、同じく8週間保存した後に測定したブリーディング率(%)も合わせて示す。

前記で調製した軟膏剤(膏体)(比較例1及び2、実施例3及び4)を25℃/60%RHの環境下で8週間保存した後、展延性(伸びやすさ)を、スプレッドメーター(井元製作所製)を用いて、23~25℃条件下での流動性を測定することで評価した。
(b)試験結果
結果を表2に記載する。なお、表2には、同じく8週間保存した後に測定したブリーディング率(%)も合わせて示す。
表2に示すように、ベタメタゾン吉草酸エステル、クロタミトン及び親油性基剤に加えて、HLBが5以下の非イオン性界面活性剤を配合することで(実施例3及び4)、これを配合しない場合(比較例1及び2)と比較して、展延性(軟膏の伸びやすさ)が若干低下すること、言い換えると膏体の粘度がやや上昇することが確認された。HLBが5以下の非イオン性界面活性剤の特徴として、親油性基剤に溶解し、その後、冷却することで結晶化することなく、当該基剤中においてゲル構造を形成すると考えられる。このため、理論に拘束されないものの、本発明の軟膏剤組成物は、ゲル化した非イオン性界面活性剤が存在する親油性基剤中に、クロタミトンを安定して分散させたことで、ブリーディング現象が大幅に減少したと考えられる。
実験例2 ベタメタゾン吉草酸エステル含有軟膏剤組成物中成分の偏析評価
ベタメタゾン吉草酸エステル含有軟膏組成物の保存安定性を評価するために、前記実験例1で調製したベタメタゾン吉草酸エステル含有軟膏組成物(比較例2、実施例3及び4)を用いて、成分(クロタミトン)の含量偏析を測定した。
具体的には、実験例1に記載する方法でベタメタゾン吉草酸エステル含有軟膏組成物(比較例2、実施例3及び4)を調製し、これを5g、チューブ形状のアルミニウム製容器(φ:13.8mm、長さ:75mm)に充填し、密封した状態で、40℃/75%RHの環境下で3ヶ月間静置保存した。保存後、チューブ容器の内部に充填した膏体の形状が崩れないように容器を開封し、略円柱状の膏体を3等分(画分1~3)に分け、各画分に含まれるクロタミトンの含量を、液体クロマトグラフィーにより測定した。
ベタメタゾン吉草酸エステル含有軟膏組成物の保存安定性を評価するために、前記実験例1で調製したベタメタゾン吉草酸エステル含有軟膏組成物(比較例2、実施例3及び4)を用いて、成分(クロタミトン)の含量偏析を測定した。
具体的には、実験例1に記載する方法でベタメタゾン吉草酸エステル含有軟膏組成物(比較例2、実施例3及び4)を調製し、これを5g、チューブ形状のアルミニウム製容器(φ:13.8mm、長さ:75mm)に充填し、密封した状態で、40℃/75%RHの環境下で3ヶ月間静置保存した。保存後、チューブ容器の内部に充填した膏体の形状が崩れないように容器を開封し、略円柱状の膏体を3等分(画分1~3)に分け、各画分に含まれるクロタミトンの含量を、液体クロマトグラフィーにより測定した。
表3に示すように、非イオン性界面活性剤を配合していない軟膏剤(比較例2)は、保存により、クロタミトンが偏析することが認められたのに対して、HLBが5以下の非イオン性界面活性剤を配合することで(実施例3及び4)、これが顕著に改善することが確認された。このことから、ベタメタゾン吉草酸エステル、クロタミトン及び親油性基剤を含有する軟膏剤について、これに加えてHLBが5以下の非イオン性界面活性剤を配合することで、製剤が安定化し、低融点物質であるクロタミトンの偏析が抑制できることが確認された。
Claims (15)
- ベタメタゾン吉草酸エステル、親油性基剤、及びHLB5.0以下の非イオン性界面活性剤を含有する軟膏剤組成物。
- さらにクロタミトンを含有する、請求項1に記載する軟膏剤組成物。
- 前記親油性基剤が、ワセリン、パラフィン、プラスチベース、ラノリン、及びミツロウからなる群より選択される少なくとも1種である、請求項1または2に記載する軟膏剤組成物。
- 前記非イオン性界面活性剤がソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルからからなる群より選択される少なくとも1種である、請求項1~3のいずれか一項に記載する軟膏剤組成物。
- ベタメタゾン吉草酸エステル、親油性基剤、及びHLB5.0以下の非イオン性界面活性剤を下記の割合で含有する、請求項1~4のいずれか一項に記載する軟膏剤組成物:
ベタメタゾン吉草酸エステル:0.0002~10質量%、
親油性基剤:10~99.79質量%、
HLB5.0以下の非イオン性界面活性剤:0.1~20質量%。 - クロタミトンの含有割合が0.1~20質量%である、請求項2~5のいずれか一項に記載する軟膏剤組成物。
- ブリーディングが抑制されてなることを特徴とする、請求項1~6のいずれか一項に記載する軟膏剤組成物。
- クロタミトンを含有する軟膏剤であって、軟膏剤組成物中のクロタミトンの偏析が抑制されてなることを特徴とする、請求項2~7のいずれか一項に記載する軟膏剤組成物。
- 請求項1~8のいずれか一項に記載する軟膏剤組成物がチューブ容器に充填されてなる軟膏剤製剤。
- 親油性基剤、並びに有効成分としてベタメタゾン吉草酸エステル、またはベタメタゾン吉草酸エステルとクロタミトンを含有する軟膏剤組成物の製剤安定性を向上する方法であって、前記軟膏剤組成物中にHLB5.0以下の非イオン性界面活性剤を共存させることを特徴とする、前記製剤安定性向上方法。
- 前記製剤安定性向上が、軟膏剤組成物のブリーディングの抑制、及び/又は、軟膏剤組成物の有効成分の偏析抑制である、請求項10に記載する製剤安定性向上方法。
- 前記親油性基剤が、ワセリン、パラフィン、プラスチベース、ラノリン、及びミツロウからなる群より選択される少なくとも1種である、請求項10または11に記載する製剤安定性向上方法。
- 前記非イオン性界面活性剤がソルビタン脂肪酸エステル、及びグリセリン脂肪酸エステルからからなる群より選択される少なくとも1種である、請求項10~12のいずれか一項に記載する製剤安定性向上方法。
- ベタメタゾン吉草酸エステル、親油性基剤、及びHLB5.0以下の非イオン性界面活性剤を下記の割合で含有する、請求項10~13のいずれか一項に記載する製剤安定性向上方法:
ベタメタゾン吉草酸エステル:0.0002~10質量%、
親油性基剤:10~99.79質量%、
HLB5.0以下の非イオン性界面活性剤:0.1~20質量%。 - クロタミトンの含有割合が0.1~20質量%である、請求項10~14のいずれか一項に記載する製剤安定性向上方法。
