Abstract
본 발명은 프테로신 화합물 및 이의 유도체를 유효성분으로 포함하는 PKA(protein kinase A, cAMP-dependent protein kinase) 관련 질환 예방 또는 치료용 조성물에 관한 것으로, 보다 상세하게는 화학식 1로 표시되는 프테로신 화합물 또는 이의 유도체를 유효성분으로 포함하는 신경정신성 질환 예방 또는 치료용 조성물에 관한 것이다.
Description
본 출원은 2020년 12월 14일에 출원된 대한민국 특허출원 10-2020-0174821호를 우선권으로 주장하고, 상기 명세서 전체는 본 출원의 참고문헌이다.
본 발명은 프테로신 화합물 및 이의 유도체를 유효성분으로 포함하는 PKA(protein kinase A, cAMP-dependent protein kinase) 관련 질환 예방 또는 치료용 조성물에 관한 것으로, 보다 상세하게는 화학식 1로 표시되는 프테로신 화합물 또는 이의 유도체를 유효성분으로 포함하는 신경정신성 질환 예방 또는 치료용 조성물에 관한 것이다.
우울증은 계속되는 슬픈 감정과 흥미 상실을 야기하는 기분 장애이다. 우울증이 있는 환자의 50% 이상은 공지된 투여 약물에 대하여 적절한 치료 반응을 경험하지 않은 것으로 추산된다. 우울증의 약물 치료에 대한 개방 연구(open-label study)에서 언급된 바와 같이, 대부분의 경우에는, 유의미한 개선이 관찰되기 전에 2주 이상의 약물 치료법이 필요하며(Rush et al, Am. J. Psychiatry 2006, 163: 1905), 아직까지 우울증 및 다른 관련된 질환에 대한 단일의 효과적인 치료가 없어 이에 관한 연구가 계속되고 있다.
한편, 마약성 진통계의 마약, 수면제, 마취제 등의 향정신성약물, 그중에서도 알코올, 헤로인 등과 같은 습관성 및 탐닉성이 강한 약물의 사용이 증가하며 약물중독이 심각한 사회적 문제로 인식되고 있으며, 이를 치료하기 위하여 약물중독 해독제에 대한 개발이 활발하게 이루어지고 있으나 고비용, 부작용 등의 여러 가지 문제점이 있으며 그 효과 또한 미미한 실정이다.
다른 한편, PKA(protein kinase A, cAMP-dependent protein kinase)는 단백질에 인산염을 공유결합시키는 인산화 효소로서 PKA의 특징은 세포 내 cAMP 수준의 변동에 따라서 활성이 조절된다는 것이다. 따라서 이 효소는 cAMP 신호전달경로를 통해 작동하는 다양한 호르몬의 최종 작동기로서 기능한다. 다시 말하면 PKA는 궁극적으로 cAMP 2차 전달 체계로 인해 모든 세포 반응에 필수적으로 관여한다. PKA는 효소활성을 갖는 두개의 촉매 소단위와 활성을 조절하는 기능을 하는 두개의 조절 소단위로 구성된 이종 4단위 중합체 (heterotetramer)로 작용을 한다. 촉매 소단위는 효소의 활성 기능 구역을 포함하고, 또한 ATP 결합 도메인과, 조절 소단위와 결합하는 도메인을 포함한다. 조절 소단위는 반대 방향으로 서로 결합하여 동종이량체 (homodimer)를 형성하는데, 이들은 cAMP가 결합하는 도메인 두 개와, 촉매 소단위와 결합하는 도메인, 그리고 촉매 도메인을 억제할 수 있는 '자동억제' 도메인을 갖고 있어, 촉매 소단위의 활성을 조절할 수 있다. 이들 조절 소단위들은 또한 촉매 소단위들을 조절하는 역할 이외의 다른 생물학적 활성도 가질 수 있다. PKA는 지방세포, 심근세포를 포함한 근세포, 간세포, 신장세포, 그리고 신경세포 등에서 역할을 갖고, 세포의 종류에 따라서 효소의 활성은 다른 기능을 갖는다.
CREB(cAMP response element-binding protein)는 세포내 주요 전사인자로서 cAMP 반응 요소(CRE; cAMP response elements)라고 불리는 특정 DNA 염기서열에 결합하여 하위 유전자들의 발현을 증가 혹은 감소시킨다. CREB은 1987년도에 소마토스타틴(somatostatin) 유전자를 조절하는 cAMP-반응성 전사인자로 처음 보고되었다. 세포의 화학에너지인 ATP는 막에 존재하는 아데닐산고리화효소(adenylyl cyclase)에 의해 cAMP (cyclic AMP)로 전환되어 세포내 주요한 단백질의 활성을 조절할 수 있다. CREB은 외부 자극으로 인한 cAMP 세포내 농도의 증가에 의해 활성화되는 PKA(Protein kinase A)로 인해 주로 활성화되며, 세포내 영양분 공급과 성장, 생리리듬, 공간 지각과 기억 등에 관련된 유전자들의 발현을 증가시키거나 억제시키는 기능을 한다.
PKA-CREB 신호전달은 신경 발달, 시냅스 및 신경가소성, 신경보호의 측면에서 중요한 역할을 하는 것으로 알려져 있으며 많은 연구들에서 PKA-CREB 신호전달이 우울증 등 다양한 신경정신성 질환의 치료에 밀접한 관련이 있다고 보고되었다.

이에 본 발명자들은 고사리 추출물에 있는 프테로신 화합물 및 이의 유도체가 생체에 미치는 영향을 확인한 결과, 프테로신 화합물 및 이의 유도체가 PKA-CREB 신호전달경로를 활성화함으로써 다양한 신경정신성 질환 등에 예방 또는 치료 효과를 나타낼 수 있다는 것을 확인하여 본 발명을 완성하였다.
따라서 본 발명의 목적은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
또한, 본 발명의 목적은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염으로 이루어진 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공하는 것이다.
또한, 본 발명의 목적은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염으로 필수적으로 이루어진 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공하는 것이다.

상기와 같은 목적을 달성하기 위하여, 본 발명은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염으로 이루어진 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공한다.

이하 본 발명을 상세히 설명한다.
본 발명은 하기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경정신성 질환 예방 또는 치료용 약학적 조성물을 제공한다.

본 발명에서 사용한 용어 "알킬"은 1 내지 4 탄소수를 함유하는 직쇄 또는 측쇄형 알킬기를 포함하는 기 또는 기의 일부분을 기술하기 위해 사용되며; 이러한 기의 예는 메틸, 에틸, 프로필, 이소프로필, n-부틸, 이소부틸, tert 부틸을 포함한다.
본 발명에서 사용한 용어 "알케닐"은 2-10개 탄소 원자 (예를 들어, C2-C10)를 가지며, 하나 이상의 이중 결합을 갖는 직쇄 또는 분지된 일가 또는 이가 탄화수소이다. 알케닐의 예로는 비제한적으로, 에테닐, 프로페닐, 프로페닐렌, 알릴 및 1,4-부타디에닐을 포함한다.
본 발명에서 사용한 용어 "알카이닐"은 2-10개 탄소 원자 (예를 들어, C2-C10)를 가지며, 하나 이상의 삼중 결합을 갖는 직쇄 또는 분지된 일가 또는 이가 탄화수소이다. 알키닐의 예로는 비제한적으로, 에티닐, 에티닐렌, 1-프로피닐, 1- 및 2-부티닐 및 1-메틸-2-부티닐을 포함한다.
본 발명에서 사용한 용어 "알콜"은 1 내지 4 탄소 원자를 가지는 직쇄 또는 측쇄형 알콜 또는 알콕시기를 기술하기 위하여 사용되며; 이러한 기는 메탄올, 에탄올, 프로판올, 부탄올 및 그들의 알콕시기를 포함한다. 알콕시기의 예로는 메톡시, 에톡시, 이소프로폭시, tert-부톡시를 포함한다. 본 발명에서 사용한 용어 상기 "탄소수 1 내지 4의 알코올"은 "탄소수 1 내지 4의 히드록시 알킬"과 동일하게 해석될 수 있다.
본 발명의 일 양태에서, 상기 화학식 1의 R1 내지 R4 중 적어도 하나 이상은 OH 또는 탄소수 1 내지 4의 알콜인 것을 특징으로 할 수 있다.
본 발명의 다른 일 양태에서, 상기 화학식 1의 R1 내지 R4 중 어느 하나는 OH 또는 탄소수 1 내지 4의 알코올이며, 상기 R6는 수소인 것을 특징으로 할 수 있다.
본 발명의 바람직한 일 양태에서, 상기 화학식 1의 R3 및/또는 R4는 OH인 것을 특징으로 할 수 있다.
본 발명의 또 다른 바람직한 일 양태에서, 상기 R3는 OH, 상기 R4는 수소인 것을 특징으로 할 수 있다.
본 발명의 상기 ‘화학식 1로 표시되는 화합물’은 고사리에 존재하는 세스퀴터페노이드로, 고사리 추출물에서 정제된 프테로신 화합물일 수 있다. 또한 본 발명에서 상기 프테로신 화합물은 이에 한정되지는 않으나 프테로신 A(pterosin A), 프테로신 B(pterosin B), 프테로신 C(pterosin C), 프테로신 D(pterosin D), 프테로신 J(pterosin J), 프테로신 M(pterosin M), 프테로신 N(pterosin N), 프테로신 P(pterosin P), 프테로신 S(pterosin S), 프테로신 Z(pterosin Z), 프테로사이드 A(pteroside A), 프테로사이드 A2(pteroside A2), 프테로사이드 B(pteroside B), 프테로사이드 C(pteroside C), 프테로사이드 D(pteroside D), 프테로사이드 N(pteroside N), 프테로사이드 P(pteroside P), 프테로사이드 Z(pteroside Z), 또는 황산화 프테로신 C(sulfated Pterosin C)일 수 있으며, 바람직하게는 프테로신 A(pterosin A), 프테로신 C(pterosin C), 프테로신 D(pterosin D), 프테로신 N(pterosin D) 또는 이의 이성질체일 수 있다.
바람직하게는, 본 발명에서 상기 화학식 1로 표시되는 화합물은 상기 R3 및/또는 R4에 히드록시(-OH) 잔기가 포함되어 있는 프테로신 화합물로서, 구체적으로는 프테로신 A, C, D 또는 이들의 이성질체일 수 있고,
가장 바람직하게는, 본 발명에서 상기 화학식 1로 표시되는 화합물은 상기 R3 및/또는 R4가 히드록시(-OH)인 프테로신 C, D 또는 이들의 이성질체일 수 있다.
또한, 본 발명의 프테로신 화합물은 비방향족 이중 결합 및 하나 이상의 비대칭 중심을 가질 수 있으며, 이들은 라세미체 및 라세미체 혼합물, 단일 거울상 이성질체, 개별적인 부분입체 이성질체, 부분입체 이성질체 혼합물 및 시스- 또는 트랜스-이성질체로서 발생할 수 있고, 이와 같은 모든 이성질체 형태를 포함할 수 있다.
본 발명에 따른 상기 화학식 1로 표시되는 화합물 및 이의 유도체는 천연으로부터 분리되거나 당 업계에 공지된 신규한 화합물의 화학적 합성법으로 제조할 수 있다. 바람직하게는 본 발명의 상기 화학식 1로 표시되는 화합물 및 이의 유도체는 천연 식물로부터 분리 및 정제할 수 있다. 즉, 종래의 물질을 추출하고 분리하는 방법을 이용하여 식물 또는 식물의 일부로부터 수득될 수 있다. 줄기, 뿌리 또는 잎은 목적하는 추출물을 획득하기 위하여 적절히 건조하여 침연(macerated)하거나, 단지 건조시켜 적절한 유기용매로 추출하고, 목적하는 추출물은 본 발명이 속하는 기술분야의 당 업자에게 알려진 정제 방법을 이용하여 정제될 수 있다. 바람직하게는 본 발명의 상기 화학식 1로 표시되는 화합물 및 이의 유도체는 고사리로부터 분리 및 정제할 수 있다.
상기 고사리는 예컨대, 덴스태드티아새(Dennstaedtiaceae) 및 프테리다새 (Pteridaceae)일 수 있다. 이러한 식물의 특정 예로는 비제한적으로, 덴스태드티아 스캔덴스(Dennstaedtia scandens), 히스티옵테리스 인시사(Histiopteris incisa), 마이크롤피아 스펠운캐(Microlepia speluncae), 프테리듐 아퀼리늄 바르. 라티우스쿨룸(Pteridium aquilinum var. latiusculum), 프테리듐 레볼루툼(Pteridium revolutum), 하이폴피스 푼크타타(Hypolepis punctata), 세라토프테리스 탈릭트로이데스(Ceratopteris thalictroides), 프테리스 파우리에이(Pteris fauriei), 프테리스 디미디아타(Pteris dimidiata) 및 프테리스 엔시포르미(Pteris ensiformi)를 포함한다. 이들 식물은 전 세계적으로 분포하며 특히 우라이 타운쉽(Wulai Township), 타이페이 카운티(Taipei County) 및 마운틴 다툰(Mountain Datun), 타이페이 시티(Taipei City)에서 발견될 수 있다.
본 발명의 상기 화학식 1로 표시되는 화합물 및 이의 유도체는 하기와 같은 단계를 포함하는 방법에 의해 분리 및 정제될 수 있다:
(a) 고사리를 물 또는 탄소수 1 내지 4의 유기용매로 추출하는 단계;
(b) 상기 (a) 단계에서 얻은 추출물을 에틸-아세테이트로 분획화하는 단계; 및
(c) 상기 (b) 단계에서 얻은 에틸-아세테이트 분획을 농도구배 크로마토그래피로 분리 및 정제하는 단계.
상기 (a) 단계에서 고사리는 식물 전체를 그대로 또는 그 건체를 사용할 수 있으며, 추출효율을 증대시키기 위해 고사리의 건체를 분쇄기로 분쇄한 것을 사용할 수 있다. 건조방법으로는 양건, 음건, 열풍건조, 동결건조 및 자연건조 방법을 모두 사용할 수 있다. 바람직하게는 열풍건조 및 동결건조 방법을 사용할 수 있다.
상기 고사리 추출에 사용되는 탄소수 1 내지 4의 유기용매는 메탄올, 에탄올, 프로판올, 이소프로판올, 부탄올, 아세톤, 에테르, 클로로포름, 에틸아세테이트, 메틸렌클로라이드, 헥산, 시클로헥산, 디클로로메탄 또는 석유에테르일 수 있다. 본 발명의 일실시예에서는 고사리를 물로 추출하였다.
상기 (b) 단계에서는 상기 (a) 단계에서 수득한 추출물을 분획화하는 단계로 에틸-아세테이트를 이용하여 분배 추출한다. (b) 단계에서 수득한 에틸-아세테이트 분획을 농도구배 크로마토그래피로 분리한다. 상기 크로마토그래피로는 실리카겔(silica gel)이나 활성 알루미나(alumina)등의 각종 합성수지를 충진한 컬럼 크로마토그래피(column chromatography) 및 고속액체크로마토그래피(HPLC)등을 단독으로 혹은 병행하여 사용할 수 있다. 바람직하게는 상기 (b) 단계에서 수득한 분획 들 중에서 에틸-아세테이트 분획을 실리카겔 컬럼에 적용하고, 용출용매의 조성을 조절하여 극성을 점차 높여가면서 다양한 분획을 얻을 수 있다. 상기와 같은 과정에서 얻은 분획들 중에서 활성을 가진 분획을 다시 용출용매의 조성을 조절하여 극성을 점차 높여가는 농도구배 실리카겔 크로마토그래피를 수행할 수 있다. 그러나 화합물의 추출 및 분리정제 방법은 반드시 상기한 방법에 한정되는 것은 아니다.
상기 (c) 단계에서는 상기 (b) 단계에서 얻은 에틸-아세테이트 분획을 농도구배 크로마토그래피로 분리 및 정제하는 단계이다.
상기 크로마토그래피로는 실리카겔(silica gel)이나 활성 알루미나(alumina)등의 각종 합성수지를 충진한 컬럼 크로마토그래피 (column chromatography) 및 고속액체크로마토그래피(HPLC)등을 단독으로 혹은 병행하여 사용할 수 있다. 바람직하게는 고속액체크로마토그래피를 이용하여 신규한 화합물을 분리 및 정제할 수 있다.
이들 화합물을 분리하는데 이용되는 방법은 당해 분야에 널리 공지되어 있다. 예를 들어, 문헌 [Takahashi et al, Phytother. Res, 2004, 18, 573, Sheridan et al, Planta Med., 1999, 65, 271, Nagao et al., Mutation Research, 1989, 215, 173, Murakami et al., Chem. Pharm. Bull, 1976, 24, 2241, and Kuraishi et al.,Chem. Pharm. 본 발명의 일실시예에서 고사리 200~500g으로 열수 추출한 다음, 에틸아세테이트(ethyl acetate, EA)를 첨가하고, 잘 혼합하여 건조시켰다. 그 다음 에틸아세테이트 추출물을 DMSO에 녹인 후 물로 희석하여 컬럼 크로마토그래피를 이용하여 7개 분획으로 나누었다. 나눈 분획은 HPLC로 분석하여 본 발명의 화합물들을 추출하였다(실시예 1 참조).
본 발명에서 상기 신경정신성 질환은 PKA 및 CREB의 신호전달과 관련된 질환으로서, 본 발명의 질환의 구체적인 예는 이에 한정되지 아니하나, 헌팅턴병, 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD), 하지마비, 우울증, 약물 중독, 루빈스타인 테이비 증후군(Rubinstein Taybi syndrome), 불면증, 수면위상지연증후군(Delayed sleep syndrome) 및 수면위상전진증후군(Advanced sleep phase syndrome)으로 이루어진 군에서 선택되는 어느 하나 이상일 수 있다.
하기에 기재한 바와 같이, PKA 및 CREB 활성화가 상기에 기재된 신경정신성 질환의 예방 또는 치료 효과가 있음은 여러 문헌을 통해 알려져 있다.
우울증, 약물중독, 루빈스타인-테이비 증후군, 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD), 하지마비
CREB 결합 단백질 (CBP) 인 CREB의 KID 도메인과 상호 작용하는 단백질의 이상은 Rubinstein-Taybi 증후군과 관련이 있으며 CREB의 기능 부족이 주요 우울 장애와 관련이 있음을 시사하는 몇 가지 증거가 있다. [Belmaker, R. H.; Agam, Galila (2008). "Major depressive disorder". 불면증, 수면위상지연증후군 및 수면위상전진증후군
포유류의 일주기 시계의 동반은 PER의 광 유도를 통해 설정된다. 빛은 망막시상하부 (RHT)를 통해 교차상핵 (SCN)에 신호를 보내는 멜라놉신 함유 감광성 망막 신경절 세포를 흥분시킨다. 본 발명의 상기 화학식 1로 표시되는 화합물에는 이의 유도체들도 포함되는데, 이는 고사리 유래의 모든 프테로신 유도체들을 포함한다. 야생형(wild type)의 프테로신 유도체들을 포함하여, PKA 활성화 기능을 하는 한, 화학기의 결실, 삽입, 비보전적 또는 보전적 치환 또는 이들의 조합에 의한 프테로신의 유도체를 모두 포함한다.
프테로신의 유도체란 야생형의 구조식과 하나 이상의 화학기가 상이한 화학적 유도체를 의미한다. 삽입은 통상적으로 약 1개 내지 20개 원자의 연속 서열로 이루어지나, 보다 큰 삽입도 가능할 수 있다. 결실은 통상적으로 약 1개 내지 30개의 원자로 이루어지나, 일부의 경우에는 곁사슬(side chain) 중 하나가 결실될 수 있는 것과 같이 보다 큰 결실 또한 가능할 수 있다. 이런 유도체는 당해 분야에 공지된 화학적 합성방법에 의해 제조될 수 있다. 분자의 활성을 전체적으로 변경시키지 않는 화학적 유도체에서의 화학기 교환은 당해 분야에 공지되어 있다. 가장 통상적으로 일어나는 교환은 화학기 간의 교환이다.
또한, 상기 화학식 1로 표시되는 화합물은 경우에 따라서는 인산화(phosphorylation), 황화(sulfation), 아크릴화(acrylation), 당화(glycosylation), 메틸화(methylation), 파네실화(farnesylation) 등으로 수식(modification) 될 수도 있다.
본 발명에서 상기 화학식 1로 표시되는 화합물의 유도체는 천연 화합물과 동일한 생물학적 활성을 나타내는 기능적 등가물이나, 필요에 따라서는 약효의 극대화 및 부작용의 최소화를 위해 이의 화합물의 특성을 변형시킨 유도체일 수 있다. 바람직하게는 곁사슬의 변이와 수식에 의해서 화합물의 열, pH 등에 대한 구조적 안정성이 증가하거나 체내 흡수율이 증가하거나 생리 활성이 증가한 화합물이다.
상기 화학식 1로 표시되는 화합물의 유도체들은 당해 기술분야에 널리 공지된 방법으로 고사리를 비롯한 천연에서 추출 및 정제하여 얻을 수 있다. 달리는, 프테로신의 구조식이 밝혀져 있으므로 화학적으로 합성하여 얻을 수 있다. 화학적으로 합성하여 제조하는 경우, 당해 기술분야에 널리 공지된 화합물 합성법을 이용하여 얻을 수 있다.
본 발명의 또 다른 일실시예에서 고사리 추출물에서 정제한 화합물들을 핵자기공명(NMR)을 이용하여 각 화합물의 종류 및 화학구조를 규명하였다(실시예 2 참조).
본 명세서에서, "약학적으로 허용가능한 염"이란, 상기 조성물에 노출되는 세포나 인간에게 독성이 없어, 인간에게 투여하기에 적합한 안전성 및 효능 프로파일을 갖는 염을 의미한다.
상기 염은 약학적으로 허용가능한 염기성 염 또는 산성염 중 어느 하나의 형태로 사용할 수 있다. 염기성염은 유기 염기염, 무기 염기염 중 어느 하나의 형태로 사용할 수 있으며, 나트륨염, 칼륨염, 칼슘염, 리튬염, 마그네슘염, 세슘염, 아미늄염, 암모늄염, 트리에칠아미늄염 및 피리디늄염으로 이루어진 군에서 선택될 수 있다.
산성염은 유리산(free acid)에 의해 형성된 산부가염이 유용하다. 유리산으로는 무기산과 유기산을 사용할 수 있으며, 무기산으로는 염산, 브롬산, 황산, 아황산, 인산, 이중 인산, 질산 등을 사용할 수 있고, 유기산으로는 구연산, 초산, 말레산, 말산, 퓨마르산, 글루코산, 메탄설폰산, 벤젠설폰산, 캠퍼설폰산, 옥살산, 말론산, 글루타릭산, 아세트산, 글리콘산, 석신산, 타타르산, 4-톨루엔설폰산, 갈락투론산, 엠본산, 글루탐산, 시트르산, 아스파르탄산, 스테아르산 등을 사용할 수 있으나, 이에 제한되지 않고 당업계에서 통상적으로 사용되는 다양한 무기산 및 유기산을 이용하여 형성되는 염이 모두 포함될 수 있다.
또한, 상기 화학식 1로 표시되는 화합물은 상기의 염뿐만 아니라, 통상의 방법에 의해 제조될 수 있는 모든 염, 수화물, 용매화물, 유도체 등을 모두 포함할 수 있다. 부가염은 통상의 방법으로 제조할 수 있고, 수혼화성 유기용매, 예를 들면 아세톤, 메탄올, 에탄올, 또는 아세토니트릴 등에 녹여 과량의 유기염기를 가하거나 무기염기의 염기 수용액을 가한 후 침전시키거나 결정화시켜서 제조할 수 있다. 또는 이 혼합물에서 용매나 과량의 염기를 증발시킨 후 건조시켜서 부가염을 얻거나 또는 석출된 염을 흡인 여과시켜 제조할 수 있다.
본 발명에 따른 약학적 조성물은 상기 화학식 1로 표시되는 화합물, 이의 이성질체, 이의 유도체 또는 이의 약학적으로 허용가능한 염을 단독으로 함유하거나 약학적으로 허용되는 담체와 함께 적합한 형태로 제형화될 수 있으며, 부형제 또는 희석제를 추가로 함유할 수 있다. 상기에서 '약학적으로 허용되는'이란 생리학적으로 허용되고 인간에게 투여될 때, 통상적으로 위장 장애, 현기 증 등과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 비독성의 조성물을 말한다.
약학적으로 허용되는 담체로는 예컨대, 경구 투여용 담체 또는 비경구 투여 용 담체를 추가로 포함할 수 있다. 경구 투여용 담체는 락토스, 전분, 셀룰로스 유도체, 마그네슘 스테아레이트, 스테아르산 등을 포함할 수 있다. 아울러, 펩티드 제제에 대한 경구투여용으로 사용되는 다양한 약물전달물질을 포함할 수 있다. 또한, 비경구 투여용 담체는 물, 적합한 오일, 식염수, 수성 글루코오스 및 글리콜 등을 포함할 수 있으며, 안정화제 및 보존제를 추가로 포함할 수 있다. 적합한 안정화제로는 아황산수소나트륨, 아황산나트륨 또는 아스코르브산과 같은 항산화제가 있다. 적합한 보존제로는 벤즈알코늄 클로라이드, 메틸-또는 프로필-파라벤 및 클로로부탄올이 있다. 본 발명의 약학적 조성물은 상기 성분들 이외에 윤활제, 습윤 제, 감미제, 향미제, 유화제, 현택제 등을 추가로 포함할 수 있다. 그 밖의 약학적으로 허용되는 담체 및 제제는 당업계에 공지되어 있는 것을 참고로 할 수 있다.
본 발명의 조성물은 인간을 비롯한 포유동물에 어떠한 방법으로도 투여할 수 있다. 예를 들면, 경구 또는 비경구적으로 투여할 수 있다. 비경구적인 투여방법으 로는 이에 한정되지는 않으나, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장내 투여일 수 있다.
본 발명의 약학적 조성물은 상술한 바와 같은 투여 경로에 따라 경구 투여용 또는 비경구 투여용 제제로 제형화 할 수 있다.
경구 투여용 제제의 경우에 본 발명의 조성물은 분말, 과립, 정제, 환제, 당의정제, 캡슐제, 액제, 겔제, 시럽제, 슬러리제, 현탁액 등으로 당업계에 공지된 방법을 이용하여 제형화될 수 있다. 예를 들어, 경구용 제제는 활성성분을 고체 부형제와 배합한 다음 이를 분쇄하고 적합한 보조제를 첨가한 후 과립 혼합물로 가공함으로써 정제 또는 당의정제를 수득할 수 있다. 적합한 부형제의 예로는 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨 및 말티톨 등을 포 함하는 당류와 옥수수 전분, 밀 전분, 쌀 전분 및 감자 전분 등을 포함하는 전분 류, 셀룰로즈,메틸 셀룰로즈, 나트륨 카르복시메틸셀룰로오즈 및 하이드록시프로필 메틸-셀룰로즈 등을 포함하는 셀룰로즈류, 젤라틴, 폴리비닐피롤리돈 등과 같은 충 전제가 포함될 수 있다. 또한, 경우에 따라 가교결합 폴리비닐피롤리돈, 한천, 알 긴산 또는 나트륨 알기네이트 등을 붕해제로 첨가할 수 있다. 나아가, 본 발명의 약학적 조성물은 항응집제, 윤활제, 습윤제, 향료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
비경구 투여용 제제의 경우에는 주사제, 크림제, 로션제, 외용연고제, 오일 제, 보습제, 겔제, 에어로졸 및 비강 흡입제의 형태로 당업계에 공지된 방법으로 제형화할 수 있다. 이들 제형은 모든 제약 화학에 일반적으로 공지된 처방서에 기재되어 있다.
본 발명의 조성물의 총 유효량은 단일 투여량(single dose)으로 환자에게 투여될 수 있으며, 다중 투여량(multiple dose)으로 장기간 투여되는 분할 치료 방법(fractionated treatment protocol)에 의해 투여될 수 있다. 본 발명의 약학적 조성물은 질환의 정도에 따 라 유효성분의 함량을 달리할 수 있다. 바람직하게 본 발명의 약학적 조성물의 바람직한 전체 용량은 1일당 환자 체중 1㎏ 당 약 0.01㎍ 내지 10,000mg, 가장 바람직하게는 0.1㎍ 내지 500mg일 수 있다. 그러나 상기 약학적 조성물의 용량은 제제화 방법, 투여 경로 및 치료 횟수뿐만 아니라 환자의 연령, 체중, 건강 상태, 성별, 질환의 중증도, 식이 및 배설율 등 다양한 요인들을 고려하여 환자에 대한 유효 투여량이 결정되는 것이므로, 이러한 점을 고려할 때 당 분야의 통상적인 지식을 가진 자라면 본 발명의 조성물의 적절한 유효 투여량을 결정할 수 있을 것이다. 본 발명에 따른 약학적 조성물은 본 발명의 효과를 보이는한 그 제형, 투여 경로 및 투여 방법에 특별히 제한되지 아니한다.
본 발명은 하기 화학식 1로 표시되는 화합물, 이의 이성질체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경정신성 질환 예방 또는 개선용 식품 조성물을 제공한다.

본 발명에 따른 상기 화학식 1로 표시되는 화합물을 포함하는 식품용 조성물은 기능성 식품(functional food), 영양보조제(nutritional supplement), 건강식품(health food) 및 식품첨가제(food additives) 등의 모든 형태를 포함한다. 상기 유형들은 당업계에 공지된 통상적인 방법에 따라 다양한 형태로 제조할 수 있다.
예를 들면, 건강식품으로는 본 발명의 식품용 조성물 자체를 차, 쥬스 및 드링크의 형태로 제조하여 음용하도록 하거나, 과립화, 캡슐화 및 분말화하여 섭취할 수 있다. 또한 본 발명의 식품용 조성물은 체지방 감소, 콜레스테롤 개선, 혈압강하의 효과가 있다고 알려진 공지의 물질 또는 활성 성분과 함께 혼합하여 조성물의 형태로 제조할 수 있다.
또한 기능성 식품으로는 음료(알콜성 음료 포함), 과실 및 그의 가공식품(예를 들어 과일 통조림, 병조림, 잼, 마멀레이드 등), 어류, 육류 및 그 가공식품(예를 들어 햄, 소시지콘비프 등), 빵류 및 면류(예를 들어 우동, 메밀국수, 라면, 스파게티, 마카로니 등), 과즙, 각종 드링크, 쿠키, 엿, 유제품(예를 들어 버터, 치즈 등), 식용식물유지, 마가린, 식물성 단백질, 레토르트 식품, 냉동식품, 각종 조미료(예를 들어 된장, 간장, 소스 등) 등에 본 발명의 식품용 조성물을 첨가하여 제조할 수 있다.
본 발명에 따른 식품용 조성물의 바람직한 함유량으로는 이에 한정되지 않지만 바람직하게는 최종적으로 제조된 식품 총 중량 중 0.01 내지 50 중량% 이다. 본 발명의 식품용 조성물을 식품첨가제의 형태로 사용하기 위해서는 분말 또는 농축액 형태로 제조하여 사용할 수 있다.
또한, 본 발명의 식품 조성물은 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상기 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 조성물 100mL당 일반적으로 약 0.01~0.04g, 바람직하게는 약 0.02~0.03g 일 수 있으나 이에 제한되지 않는다.
본 발명의 상기 부형제, 결합제, 붕해제, 활택제, 교미제, 착향제 등에 대한 용어 정의는 당업계에 공지된 문헌에 기재된 것으로 그 기능 등이 동일 내지 유사한 것들을 포함한다.
본 발명의 다른 일실시예에서 정상 세포인 NIH3T3 세포와 암 세포인 B16F10 세포를 각각 96 웰 플레이트에 배양한 뒤 화합물 2 내지 11을 각각 세포 배지에 첨가하여 배양한 후, MTT를 처리하고, 마이크로 플레이트 리더기로 550nm에서 흡광도를 측정하였다. 측정한 결과에 근거하여 LD50값을 계산하였다. 그 결과, 암 세포와 정상세포에서 각 화합물의 LD50값이 높은 것으로 나타났고, 이에 각 화합물은 세포 독성이 낮은 것을 확인할 수 있었다(실시예 3 참조).
또한 본 발명은 신경정신성 질환 치료용 제제를 제조하기 위한 하기 화학식 1로 표시되는 화합물, 이의 이성질체 또는 이의 약학적으로 허용가능한 염의 용도를 제공한다.

본 발명의 상기 '유효량'이란 개체에게 투여하였을 때, 신경정신성 질환의 개선, 치료, 예방, 검출, 진단 또는 상기 질환의 억제 또는 감소 효과를 나타내는 양을 말하며, 상기 '개체'란 동물, 바람직하게는 포유동물, 특히 인간을 포함하는 동물일 수 있으며, 동물에서 유래한 세포, 조직, 기관 등일 수도 있다. 상기 개체는 상기 효과가 필요한 환자(patient) 일 수 있다.
본 발명의 상기 '치료'는 신경정신성 질환 또는 신경정신성 질환의 증상을 개선시키는 것을 포괄적으로 지칭하고, 이는 신경정신성 질환을 치유하거나, 실질적으로 예방하거나, 또는 상태를 개선시키는 것을 포함할 수 있으며, 상기 질환으로부터 비롯된 한 가지 증상 또는 대부분의 증상을 완화시키거나, 치유하거나 예방하는 것을 포함하나, 이에 제한되는 것은 아니다.
본 명세서에서 용어 "을 포함하는(comprising)"이란 "함유하는(including)" 또는 "특징으로 하는(characterized by)"과 동일한 의미로 사용되며, 본 발명에 따른 조성물 또는 방법에 있어서, 구체적으로 언급되지 않은 추가적인 구성 성분 또는 방법의 단계 등을 배제하지 않는다. 또한 용어 "로 이루어지는(consisting of)"이란 별도로 기재되지 않은 추가적인 요소, 단계 또는 성분 등을 제외하는 것을 의미한다. 용어 "필수적으로 이루어지는(essentially consisting of)"이란 조성물 또는 방법의 범위에 있어서, 기재된 물질 또는 단계와 더불어 이의 기본적인 특성에 실질적으로 영향을 미치지 않는 물질 또는 단계 등을 포함할 수 있는 것을 의미한다.
이상 살펴본 바와 같이 본 발명이 제공하는 화합물, 이의 이성질체 또는이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 조성물은 PKA 및 CREB를 활성화하는 효과를 나타내어 헌팅턴병, 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD), 하지마비, 우울증, 약물 중독, 루빈스타인 테이비 증후군(Rubinstein Taybi syndrome), 불면증, 수면위상지연증후군(Delayed sleep syndrome) 및 수면위상전진증후군(Advanced sleep phase syndrome)으로 이루어진 군에서 선택되는 질환의 치료제 또는 상기 질환 개선용 식품 개발에 유용하게 활용이 될 수 있다.

이하 본 발명을 상세히 설명한다.
단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 한정되는 것은 아니다.
실험방법
실험 기구
1 H NMR 및 13 C NMR 스펙트럼은 중수소 클로로포름 (CDCl3), 디메틸 술폭시드 (DMSO-d 6)로 1H NMR로 400 MHz에서, 13C NMR로 100MHz에서 JEOL JUM ECP-400 분광계(Jeol, Tokyo, Japan)를 이용하여 측정하였다.
컬럼 크로마토그래피는 실리카겔(70-230 메쉬; Merk, Darmstadt, Germany), RP-18 (40-63mm; Merk) 및 Sephadex LH-20 (20-100mm; Sigma, St. Louis, MO, USA)로 실시되었다. 얇은 층 크로마토그래피 (TLC)는 Kiesel gel 60 F254 plates (0.25 mm; Merck) 및 25 RP-18 F 254s plate (5 -10 cm, Merk)로 미리 코팅 된 상태로 실시하였다. 분무 시약은 50 % H2SO4였다. 컬럼 크로마토그래피에 사용된 모든 화학 물질 및 용매는 시약 등급 내이며 구입하여 수령한 그대로 사용하였다.
MTT assay
마우스 섬유아세포주인 NIH3T3 세포 및 마우스 멜라노마세포주인 B16F10 세포를 2×103cells/well의 96 well plate에 분주하였고, 10% FBS(fetal bovine serum)가 첨가된 DMEM를 이용하여 5% CO2, 37℃ 조건에서 배양하였다. 화합물(pterosin A, pterosin B, pterosin C, pterosin D, pterosin P, pterosin Z, pteroside A, pteroside B, pteroside Z)을 각각 농도별로 처리한 후, 48시간 동안 배양하였다. 그 후 각 웰에 MTT 용액(5 mg/ml)을 10μg/well의 농도로 첨가하고, 4시간 동안 37℃에서 배양하였다. 각 well에 100μl의 DMSO를 가하고 상층액을 제거하였다. 10분간 배양한 후, Microplate Reader (Molecular Devices, Spectra Max, USA)를 이용하여 550nm에서 흡광도(Optical density)를 측정하였다. 흡광도는 생존한 세포의 수를 나타내는 지표로서 하단의 식으로 계산되며, 3회의 실험으로 재현성을 확인하였다.

웨스턴 블롯 분석법
웨스턴 블롯 분석은 다음과 같은 방법을 사용하였다. 세포를 RIPA 완충액 (20mM Tris-Cl pH 7.4, 1mM EDTA, 150mM NaCl, 1 % Triton X-100, 1mM EGTA, 1μM Na3VO4, 2.5mM 나트륨 파이로 포스페이트, 1μM 페닐 메틸 설포닐 플루오 라이드, 1 μg/ml 류 펩틴,)에 용해시켰다. 단백질 농도는 BCA 단백질 분석 시약 (BioRad, Hercules, CA, USA)을 사용하여 계산되었다. 단백질 샘플을 SDS-PAGE로 분리하고 폴리 비닐 리덴 디 플루오 라이드 (PVDF) 막 (Millipore, Bedford, MA, USA)으로 옮겼다. 막을 1 차 항체와 함께 4℃에서 밤새 배양 한 다음 HRP-접합 염소 항-토끼 IgG (1 : 5,000; Pierce; Carlsbad, CA, USA)를 사용했습니다. 사용 된 1 차 항체는 actin (sigma; 1:1000), p-CREB (Cell signaling; 1:1000), tubulin (Cell signaling; 1:1000), p-PKA (Cell signaling; 1:1000), p-trk (Cell signaling; 1:1000) 그리고 BDNF (Abcam; 1:1000) 이다. 모든 막은 UVP Autochemi Darkroom Imaging System (Utra-Violet Products Ltd. UK) 및 Immobilon Crescendo Western HRP Substrate 또는 Luminata Forte Western HRP Substrate (Millipore, USA)를 사용하여 시각화되었다.
전기생리학 실험
1. 4개월 된 생쥐의 뇌를 차가운 해부버퍼 용액(ice-cold dissection buffer) 위에 놓고, 바이브라톰을 사용해서 300μm 두께의 시상면의 해마 슬라이스(sagittal hippocampal slice)를 준비했다 (dissection buffer: contents in mM: sucrose, 25 NaHCO3, 5 KCl, 1.25 NaH2PO4, 0.5 CaCl2, 3.5 MgSO4, 10 D-glucose, 1.25 L-ascorbic acid and 2 Na-pyruvate bubbled with 95% O2/5% CO2)
2. 슬라이스는 정상 ACSF에서 1시간 동안 32℃에서 회복되었다(ACSF: mM: 125 NaCl, 2.5 KCl, 1.25 NaH2PO4, 25 NaHCO3, 10 포도당, 2.5 CaCl2 및 1.3 MgCl2 산소가 95% O2/5% CO2).
3. 레코딩 시에는, 슬라이스를 28℃의 ACSF에 담긴 챔버(submerged chamger)로 이동한 뒤, 95% O2/5% CO2에 포화된 ACSF(2ml/min)를 지속적으로 관류했다. 자극과 기록을 위한 마이크로피펫은 전극 풀러(micropipette electrode puller)를 사용하여, 붕소실산 유리 모세혈관(Harvard Instruments)을 이용해서 제작했다.
4. 입력/출력 반응(input/output), 쌍체-펄스 비율(paired-pulse ratio), 장기간 강화(Long-term potentiation, LTP) 실험의 경우, ACSF로 채워진 피펫(저항: 1MΩ)을 사용하여 해마 CA1 영역에서 fEPSP(field excitatory post-synaptic potential)을 측정하였다. fEPSP 신호는 Multiclamp 700B(Molecular Devices)를 통해 증폭하고, Digidata 1440A(Molecular Devices)를 이용해 디지털화하였다. 섀퍼 측방향 경로(Schaffer collateral pathway)는 20초(LTP) 혹은 15초(입력/출력, 쌍체-펄스 비율)마다 ACSF(0.3-0.5MΩ)로 채워진 피펫으로 자극되었다.
5. LTP를 유도하기 위해서, 안정적인 베이스라인 신호를 획득한 후 theta-burst stimulation(40 trains of pulse, each train is composed with 4 stimulus in 100 Hz; 40 trains are divided by 4 burst, each containing 10 trains with 1 sec inter-burst-interval; Within the 10 trains, there were 170 ms time inter-train-interval)의 전기자극을 적용하였다. 쌍체-펄스 비율은 25, 50, 75, 100, 200, 300 ms의 간격으로 쌍자극을 준 뒤 측정하였다.
6. 시냅스 성질에 대한 약물 A (pterosin A)/C (pterosin C)/X (Drug X, phosphodiesterase(PDE) inhibitor인 amx)의 약리학적 영향을 분석하기 위해, 각 약물을 DMSO에 1000배의 재고농도로 용해한 후, ACSF에서의 농도가 각각 10μM(X)와 1μM(A/C)가 되도록 약물을 적용하였다. 컨트롤 약물로는 실험 약물과 같은 volume의 DMSO를 적용하였다.
실시예 1: 프테로신(pterosin) 화합물 및 이의 유도체의 정제
고사리 열수 추출물을 에틸아세테이트(ethyl acetate)를 이용하여 추출한 뒤, 7개의 분획으로 분리하였고, 각 분획에서 HPLC 방법을 이용하여 프테로신 화합물을 분리하였다.
구체적으로, 고사리 추출물은 경기도 가평군에서 채취한 고사리 200~250g을 깨끗이 씻어 증숙용기(OSK-2002, 홍삼박사, Well sosanaTM, 대웅제약)에 넣고 물 1.5L를 첨가하여 24시간 증숙한 후, 물 3.5L를 추가로 첨가하여 72시간 동안 숙성시키고 냉장보관한 것을 열수 추출물로 사용하였다. 열수 추출물에 동일한 부피의 에틸 아세테이트(ethyl acetate, EA)를 첨가하고 잘 혼합한 후, EA 층을 rotary evaporator로 건조하여 EA 추출물로 사용하였다. EA 추출물을 최소량의 DMSO에 녹인 후 물로 희석하였는데, 이 때 희석 배수는 수율을 대략 70%로 예상하여 열수 추출물 원래 부피의 70%까지 희석하였다.
EA 추출물을 실리카겔(silica gel) 컬럼 크로마토그래피(column chromatography)를 이용하여 클로로포름(CHCl3)과 메탄올 조건으로 7개의 분획으로 나누었다. 그 다음 각각의 분획물을 Prep-HPLC로 분석한 결과, 분획물 2, 4, 5, 6, 7에서 화합물 2 내지 11을 추출할 수 있었다. 각 화합물을 추출하는 과정은 도 1에 나타내었다.
실시예 2: 화합물 2-11의 프테로신(pterosin) 화합물 및 이의 유도체의 동정
상기 실시예 1에서 정제한 각각의 화합물을 동정하기 위하여 EA 분획물에서 단일 화합물을 분리한 뒤, 상기 실험방법에 기재되어 있는 핵자기공명(NMR) 방법으로 화합물의 종류와 화학구조를 규명하였다. 고사리 추출물에서 동정된 각 화합물의 구조 및 화합물의 명칭은 하기 표 1에 기재되어 있으며, 구체적인 NMR 분석 결과는 하기 표 2에 기재되어 있다.
실시예 3: 프테로신 화합물의 세포독성 평가(MTT assay)
상기 실시예 2의 화합물 2 내지 11이 세포에 독성 효과를 나타내는지 확인하기 위하여 다음과 같이 실험을 실시하였다.
상기 실험방법에 기재된 바에 따라 정상 세포인 NIH3T3(mouse embryo fibroblast cell-line)와 암 세포인 B16F10(Mouse melanoma cell-line)을 96웰 플레이트에 배양한 뒤 각 화합물을 농도별로 세포 배지에 첨가하고 48시간 동안 배양한 뒤 MTT를 투여한 뒤, 마이크로플레이트 리더(microplate reader)를 이용하여 550nm에서 흡광도를 측정하였다. 흡광도는 생존한 세포의 수를 나타내는 지표로서 하단의 식으로 계산되며, 3회의 실험으로 재현성을 확인하였다.
세포 증식율(%)=OD550(sample)/OD550(control)×100
그 다음 세포 증식율의 변화에 근거하여 LD50(Lethal Dose 50)값을 산출하였다.
그 결과 표 3에 나타난 바와 같이, 암세포에서 화합물 6의 LD50값이 522±25μM로 가장 낮은 것으로 나타났으며, 정상 세포에서 화합물 2의 LD50값은 3,110±130μM, 정상 세포에서 화합물 3의 LD50값은 553±37μM, 암세포에서 화합물 3의 LD50값은 618±71μM인 것으로 나타났다. 이를 제외하고 정상세포 및 암세포에서 다른 화합물의 LD50값은 5000μM 초과인 것으로 나타났다.
이를 통해, 화합물 3은 약한 세포 독성이 있으며, 화합물 6은 암세포에서만 약한 세포 독성이 있고, 화합물 2, 4, 5, 7 내지 11은 정상 세포와 암세포에서 세포 독성이 거의 없는 것을 확인할 수 있었다.
실시예 4: PKA의 cAMP 결합 부위에 대한 Rigid Docking 실험
프테로신 화합물 또는 이의 유도체들의 PKA 결합모델 예측을 위한 실험을 수행하였다.
실험은 Schrodingr 사의 MAESTRO 소프트웨어를 사용하였으며, Ligand docking과 induced fit docking 방법을 이용하여 단백질과 약물(프테로신 A) 사이의 결합을 측정하였다.
PKA 조절소단위의 CBD(cAMP binding domain)는 2곳 (CBD1, CBD2)으로 각각에 대한 결합을 측정하였고(도 3 참조), 그 결과 Docking score가 CBD1 및 CDB2 각각에서 -8.307과 -9.808로 측정되어 단백질-약물 사이의 결합이 매우 안정한 것을 확인할 수 있었다(표 4 참조).
실시예 5: CREB 인산화 수준 측정을 위한 웨스턴 블롯
도 2에 나타낸 바와 같이, 신경세포에서 프테로신 A, C, D 등 프테로신의 오각형 ring의 아래쪽에 곁사슬로 -OH기가 있는 프테로신 유도체들은 10μM에서 CREB 인산화를 양성대조군으로서 phosphodiesterase(PDE) inhibitor인 amx 이상 수준으로 크게 증가시켰다.
반면 프테로신 B, P, Z 등 프테로신의 오각형 ring의 아래쪽에 곁사슬로 -OH기가 없는 프테로신 유도체들은 10μM에서 CREB 인산화를 전혀 증가시키지 못하였다.
뇌의 결체조직 세포인 성상 세포(astrocyte)에서는 프테로신 A, C, D 등 프테로신의 오각형 ring의 아래쪽에 곁사슬로 -OH기가 있는 프테로신 유도체뿐만 아니라 프테로신 B, P, Z 등 프테로신의 오각형 ring의 아래쪽에 곁사슬로 -OH기가 없는 프테로신 유도체 모두 CREB 인산화에 전혀 영향이 없었다.
그 다음으로, 신경세포에서 프테로신 A, C, D, B가 CREB, BDNF, trk 인산화에 미치는 영향을 웨스턴블롯을 통해 확인하였다.
그 결과, 도 8에 나타낸 바와 같이 프테로신 중 오각형 ring의 아래쪽에 곁사슬로 -OH기가 있는 유도체들이 곁사슬 -OH가 없는 프테로신 B와 비교했을 때, 프테로신 A, C 및 D는 PKA 활성화를 통해 CREB의 인산화를 통계적으로 유의하게 크게 증가시켰다. 이에 따라 인지장애에서 가장 중요한 표적 중 하나로 작용하는 뇌 유래 신경 영양 인자인 BDNF를 증가시킴으로서 프테로신이 PKA-CREB-BDNF 신호전달경로를 활성화함으로써 학습 및 기억 장애의 완화 효능을 가질 수 있다는 것을 확인하였다.
실시예 6: 전기생리학 실험
Hippocampal slice에서 전기생리학적 실험한 결과를 참고하면, 정상적인 synaptic transmission에는 실험물질의 처리에 의해 특별한 변화가 없으나(도 9), Long-term memory를 담당하는 뇌 조직 hippocampus mossy fiber-CA1 LTP가 pterosin 화합물 처리에 의해 강화되며, 이러한 효과는 특히 pterosin C 및 D에서 더 강하게 나타나는 것으로 확인되었다(도 10).
실시예 7: 신경세포(Neuron)의 증식, 분화 및 재생 촉진능
프테로신 화합물의 신경세포 증식, 분화 및 재생 촉진능을 평가하였다.
도 11에 나타낸 바와 같이, 프테로신 D를 처리한 신경세포의 길이가 대조군 대비 현저하게 길어진 것으로 나타나, 신경세포 증식 및 분화 촉진 효과가 나타났음을 확인할 수 있었다.
실시예 8: 혈액-뇌 장벽(Blood-brain barrier) 투과성 평가
상기 실시예에서 가장 우수한 활성을 나타낸 화합물 중 하나인 Pterosin D의 혈액-뇌 장벽 투과성을 아래 방법에 따라 평가하였다. 양성대조군으로는 progesterone, 음성대조군으로는 ranitidine을 사용하였다:
1) Deep well plate에 pH 7.4의 donor buffer를 분주한 후 시험 물질을 섞어서 준비했다.
2) Donor buffer를 U.V plate에 well당 150ul씩 분주한 후 흡광도를 측정하였다: Blank값
3) Deep well plate에 있는 mixture를 well당 150ul씩 U.V plate로 분주한 후 흡광도를 측정했다: Blank값
4) Stirwell PAMPA sandwich의 donor plate에 상기 1) 단계의 mixture를 200ul씩 분주했다.
5) Stirwell PAMPA sandwich의 acceptor plate의 membrane을 5ul의 BBB-1-lipid로 코팅했다.
6) Acceptor plate에 brain sink buffer를 200ul씩 각 well에 분주했다.
7) Acceptor plate와 donor plate를 합치고 25도씨에서 4시간 동안 incubation 했다.
8) 4시간 incubation 후, acceptor plate와 donor plate를 분리한 후 각각 150ul씩 U.V plate로 옮긴 후 흡광도를 측정했다.
투과성 평가 기준은 아래 표 5와 같다:

이상 살펴본 바와 같이 본 발명이 제공하는 프테로신 화합물 및 이의 유도체를 유효성분으로 포함하는 질환 예방 또는 치료용 조성물은 PKA 및 CREB를 활성화하는 효과를 나타내어 우울증, 약물 중독, 루빈스타인 테이비 증후군(Rubinstein Taybi syndrome), 불면증, 수면위상지연증후군(Delayed sleep syndrome), 수면위상전진증후군(Advanced sleep phase syndrome), 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD) 및 하지마비 치료제 또는 상기 질환 개선용 식품 개발에 유용하게 활용이 될 수 있어 산업상 이용가능성이 매우 우수하다.
Claims (15)
- 제1항에 있어서, 상기 R1 내지 R4 중 적어도 하나 이상은 OH 또는 탄소수 1 내지 4의 알콜인 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 R1 내지 R4 중 어느 하나는 OH 또는 탄소수 1 내지 4의 알코올이며, 상기 R6는 수소인 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 화학식 1로 표시되는 화합물은 프테로신 A(pterosin A), 프테로신 C(pterosin C), 프테로신 D(pterosin D) 및 프테로신 N(pterosin N)으로 이루어진 군에서 선택되는 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 화학식 1로 표시되는 화합물은 PKA(Protein kinase A)를 활성화하는 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 화학식 1로 표시되는 화합물은 CREB 인산화를 증가시키는 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 화학식 1로 표시되는 화합물은 고사리로부터 추출된 것을 특징으로 하는 약학적 조성물.
- 제1항에 있어서, 상기 신경정신성 질환은 헌팅턴병, 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD), 하지마비, 우울증, 약물 중독, 루빈스타인 테이비 증후군(Rubinstein Taybi syndrome), 불면증, 수면위상지연증후군(Delayed sleep syndrome) 및 수면위상전진증후군(Advanced sleep phase syndrome)으로 이루어진 군에서 선택되는 것을 특징으로 하는 약학적 조성물.
- 제10항에 있어서, 상기 화학식 1로 표시되는 화합물은 고사리로부터 추출된 것을 특징으로 하는 용도.
- 제10항에 있어서, 상기 신경정신성 질환은 헌팅턴병, 뇌성마비, 자폐증, 주의력결핍 과잉행동장애(ADHD), 하지마비, 우울증, 약물 중독, 루빈스타인 테이비 증후군(Rubinstein Taybi syndrome), 불면증, 수면위상지연증후군(Delayed sleep syndrome) 및 수면위상전진증후군(Advanced sleep phase syndrome)으로 이루어진 군에서 선택되는 것을 특징으로 하는 용도.
