Abstract
본 발명은 도축 혈액에서 분리된 아미노산을 포함하는 사료첨가제 제조방법 및 상기 방법에 의해 제조된 사료첨가제, 동물용 사료 조성물에 관한 것으로, 냄새가 나지 않으면서도 아미노산이 풍부하여 영양성분이 강화된 동물용 사료첨가제 및 이의 제조방법을 제공할 수 있다. 또한, 도축 부산물을 재활용하여 폐기물을 감소시킬 수 있다. {대표도} 도 1
Description
본 발명은 도축 혈액에서 전환된 아미노산을 포함하는 동물용 사료첨가제의 제조방법 및 상기 방법에 의해 제조된 동물용 사료첨가제 조성물에 관한 것으로, 구체적으로는 도축 혈액에서 전환된 아미노산을 포함하는 가축용 사료에 사용될 수 있는 다양한 형태의 사료첨가제 조성물의 제조방법에 관한 것이다.
가축의 도축 시 다량 발생하는 도축 부산물 중 혈액은 각종 단백질, 무기질과 같은 영양소가 풍부하게 함유되어 있어, 유용한 자원으로 활용될 수 있다. 미국, 스웨덴, 스위스, 덴마크, 네덜란드, 일본, 중국 등에서는 분말 형태로 1차 가공 후, 사료첨가제, 식품첨가제, 발색제, 향신료, 성장촉진제, 의약품 첨가제 등으로 이용되고 있다.
이에 비해 국내 도축장에서 발생하는 혈액은 선지나 순대로 소량 사용될 뿐 대부분이 폐기물로 폐기되고 있다. 이러한 폐기물은 해양 배출에 의존하고 있었으나, 해양투기방지 협약의 가입에 따라, 2006년부터 해양 배출 금지를 위한 정책을 단계적으로 시행하여, 2016년 1월부터는 육상 폐기물의 해양 배출이 전면 금지된다. 따라서 가급적 혈액과 같은 해양 배출 폐기물을 줄일 수 있는 방안이 고려되고 있으며, 도축 혈액의 재활용도 하나의 방안이 될 수 있다.
도축 부산물 중 혈액의 재활용 방안으로, 최근에는 동물의 혈액에 함유된 고단백질을 이용한 아미노산 함유 사료나 비료로 재활용하려는 기술이 많이 제안되고 있다. 특히 국내에서는 혈분 형태의 건조와 수거된 생혈을 그대로 산 분해하거나 효소로 발효하여 액상 비료를 제조하는 방법이 많이 제안되어 있다.
생혈을 이용하여 사료나 비료를 제조하는 방법은 혈액에 포함된 단백질을 최대한 손실 없이 아미노산 제제로 제조하기 위해 마련된 것이다. 그러나 동물의 혈액은 단백질 함량이 높아서, 비료나 사료의 제조 공정 중 및 제조 후에 부패 가능성, 세균, 박테리아 등의 번식에 따른 오염 가능성이 높은 문제점이 있고, 비료나 사료로 제조 후에도 좋지 않은 냄새가 나는 문제점이 있다.
이에 본 발명자는 제조 공정 중의 오염 가능성이 낮고, 제조 후의 부패 가능성 및 냄새 문제를 해결할 수 있는 방안에 대해 연구하였다.

상기와 같은 문제점을 해결하기 위해 본 발명은 도축혈액을 이용하여 냄새가 나지 않으면서도 아미노산이 풍부한 사료첨가제, 상기의 사료첨가제를 포함하는 사료 조성물 및 이의 제조방법을 제공하고자 한다.
상기와 같은 목적을 달성하기 위해 본 발명은 하기의 단계를 포함하는 도축 혈액 유래 아미노산을 포함하는 사료용 조성물의 제조방법을 제공한다.
(a) 가축의 도축된 혈액에 효소를 처리하여 단백질을 분해하는 전처리 단계;
(b) 상기 (a) 단계에서 전처리된 도축혈액을 내측과 외측이 반대로 결합된 더블리본 믹서를 사용하여 증숙하는 단계;
(c) 상기 (b) 단계에서 증숙된 도축 혈액을 냉각하는 단계;
(d) 상기 (c) 단계에서 냉각된 도축 혈액을 탈수하는 단계;
(e) 상기 (d) 단계에서 탈수되고 남은 아미노산 케이크를 조파쇄하여 압출 성형하여 건조시키는 단계.
상기 (a) 단백질 분리 단계에서 효소는 제한되는 것은 아니지만, 바람직하게는 알칼라아제일 수 있고, 구체적인 알칼라아제의 예로는 노보자임 알칼라아제일 수 있다.
상기 효소는 도축혈액 중량을 기준으로 0.1 내지 5 중량%, 바람직하게는 0.1 내지 2 중량% 첨가될 수 있다. 효소를 상기 범위 미만으로 투입하는 경우, 아미노산으로 제대로 전환되지 않을 수 있고, 상기 범위를 초과하여 투입하는 경우, 아미노산 외에 다른 불순물까지 분리될 우려가 있다.
또한, 상기 (a) 단계에서의 단백질 분해 시간은 4 내지 16 시간, 바람직하게는 6 내지 12 시간 동안 진행할 수 있다. 상기 시간 미만인 경우 아미노산이 제대로 전환되지 않을 수 있고, 상기 시간을 초과하는 경우 불순물까지 분해되어 오히려 아미노산이 제대로 분리되지 않을 수 있다.
또한, 상기 (a) 단계는 단백질 분해 효율을 고려할 때 pH 8 내지 8.5가 바람직하다.
상기 (a) 단계 이후 (b) 단계를 진행하기 전에 추가적으로, (a)’ 맷돌 방식의 그라인드밀을 사용하여 액화시키는 단계를 더 포함할 수 있다. 상기 액화 단계는 3000 내지 7000, 바람직하게는 4000 내지 5000rpm으로 3 내지 5회 반복하여 수행할 수 있다. 이 단계를 통해 혈액 내에 공존하는 혈관, 털, 응고된 혈구 등을 액화시켜 단백질의 아미노산전환 효율이 증가되며, 부패 및 강한 암모니아 악취 발생을 낮출 수 있다.
상기 (b) 증숙 단계의 온도는 제한되는 것은 아니지만 바람직하게는 80 내지 95℃, 증숙 시간은 0.5 내지 4시간, 바람직하게는 1 내지 2시간 동안 진행될 수 있다.
상기 온도 미만인 경우, 증숙이 제대로 이루어지지 않고, 상기 온도를 초과하는 경우 단백질이 겔화되므로, 이후의 탈수 단계가 원활하게 진행되지 않을 수 있다.
또한, 상기 (b) 증숙 단계에서 더블리본 믹서를 사용하는 이유는 증숙되는 과정에서 고형화가 이루어지면 아미노산 덩어리가 상층에 부유하게 되어 믹싱이 어렵기 때문이다. 증숙 효율을 높이기 위해 바람직하게는 교반기의 안쪽(샤프트쪽) 스크류 날개는 하향식이 되도록 설정하고, 바깥쪽(탱크벽쪽) 스크류 날개는 상향식이 되도록 설정할 수 있으나 이에 제한되는 것은 아니다.
상기 (c) 단계는 (b) 단계에서 증숙된 도축 혈액을 냉각하는 단계로, 냉각 온도는 제한되는 것은 아니지만 바람직하게는 65 내지 75℃일 수 있다.
상기 온도 미만이면 온도가 너무 낮아 탈수가 어렵고, 상기 온도보다 높으면 여과포가 수축되어 또한 탈수가 어렵다. 또한, 상기 냉각 단계는 축을 중심으로 U형 날개 교반기를 사용하여 10 내지 30rpm, 바람직하게는 10 내지 20rpm하에서 진행될 수 있다.
상기 (d) 단계는 (c) 단계에서 냉각된 아미노산을 탈수하는 단계로, 탈수 시 압력은 1 내지 5kg/cm2, 바람직하게는 2 내지 3kg/cm2의 압력 하에서 진행한다. 탈수 압력이 상기 압력보다 낮은 경우 탈수가 원활히 되지 않는다.
탈수된 아미노산 케이크의 함수율은 80% 미만, 바람직하게는 70% 미만이 되도록 탈수한다. 함수율이 상기 범위를 초과하는 경우 수분이 너무 많이 포함되어 있어 제형화가 어려워진다.
또한, 상기 탈수 단계는 유압프레스 또는 여과 필터프레스를 사용할 수 있고, 바람직하게는 여과 필터프레스를 사용할 수 있으나 이에 제한되는 것은 아니다.
상기 (d) 탈수 단계 이후에 (d)’ 제형화 단계를 추가적으로 더 포함할 수 있다. 상기 (d)’ 제형화 단계는 탈수된 아미노산 또는 아미노산 케이크를 정제(tablet), 펠렛(pellet), 입상(granule) 등의 형태로 압출 성형시키는 단계이다.
상기 성형은 1 내지 5mm, 바람직하게는 2 내지 3mm 크기로 제형화시킨다. 제형화 단계에서 상기 크기 범위를 벗어나는 경우, 건조가 잘되지 않거나 부패될 우려가 있다.
상기 (e) 단계는 (d) 단계 또는 (d’) 단계에서 제조된 아미노산을 감압 원적외선 파이프 히터를 사용하여 건조시키는 단계로, 건조 온도는 제한되는 것은 아니지만 바람직하게는 60 내지 90℃에서 건조시킨다.
상기 온도보다 낮은 경우, 건조시간이 오래걸리고 열효율이 떨어지며, 상기 온도보다 높은 경우, 아미노산이 탄화되어 펩신 소화율이 낮아질 뿐만 아니라 상품성도 저하될 수 있다.
상기 원적외선은 제한되는 것은 아니지만 바람직하게는 8~16um의 파장을 사용할 수 있다. 원적외선 파이프 히터를 사용하여 건조시키는 것은 아미노산이 파괴되지 않으면서도 도축 혈액 특유의 잡내를 잡아줄 수 있기 때문이다. 또한, 상기 건조단계는 배치식 감압 건조장치를 추가적으로 더 포함하여 진행할 수 있다.
상기 (e) 단계 이후에는 건조된 아미노산 또는 아미노산 케이크를 메쉬망을 이용하여 분류하는 단계; 및 분류된 아미노산 또는 아미노산 케이크를 포장 및 출하하는 단계를 추가적으로 더 포함하여 진행할 수 있다.
본 발명에서 아미노산은 하기와 같이 기재된다;
알라닌(Ala(A)), 라이신(Lys(K)), 아스파라긴(Asn(N)), 아스파르트 산Asp(D), 시스테인(Cys(C)), 히스티딘(His(H)), 이소류신(Ile(I)), 메티오닌(Met(M)), 세린(Ser(S)), 발린(Val(V)), 글리신(Gly(G)), 류신(Leu(L)), 프롤린(Pro(P)), 트레오닌(Thr(T)), 페닐알라닌(Phe(F)), 알기닌(Arg(R)), 티로신(Tyr(Y)), 트립토판(Trp(W)), 글루탐산(Glu(E)), 글루타민(Gln(Q)).
이중 필수아미노산은 히스티딘, 이소류신, 류신, 라이신, 메티오닌, 페닐알라닌, 트레오닌, 트립토판, 발린이다.
본 발명에 따른 아미노산 사료첨가제는 상기 필수 아미노산을 적어도 7종 이상 포함할 수 있다.
또한, 상기 필수 아미노산은 사료첨가제에 포함된 전체 아미노산 함량 대비 40 내지 60 중량%, 바람직하게는 45 내지 55중량% 포함될 수 있다.
또한, 본 발명에 따른 사료첨가제를 포함하는 사료 조성물은 상기 사료첨가제를 사료 조성물 총 중량의 60 내지 80 중량%, 바람직하게는 65 내지 75 중량% 포함할 수 있다.
상기 방법에 의해 도축 혈액에서 분리된 아미노산은 도축된 혈액에서 나는 특유의 악취가 나지 않고, 영양 성분이 높기 때문에 동물 사료용, 비료용 등의 용도로 사용될 수 있다.
상기 동물은 예를 들어 소, 돼지와 같은 가축용 동물이거나 개, 고양이 등과 같은 애완용 동물일 수 있으나, 이에 제한되는 것은 아니다.
또한 상기 방법에 의해 분리된 아미노산을 동물용 사료용으로 제형화시킬 경우, 당 업계에서 동물용 사료를 제조할 때 통상적으로 사용하는 첨가제, 안정제 등을 추가적으로 더 포함할 수 있다.
상기 첨가제, 안정제는 솔비톨, 아스코르빈산, 말티톨, 카제인산나트륨, 소디움클로라이드, 잔탄검, 폴록사머, 아황산나트륨 등일 수 있으나 이에 제한되는 것은 아니다.
본 발명은 가축용 사료로 사용될 수 있는 도축 혈액에서 분리된 아미노산을 포함하는 사료첨가제 및 이의 제조방법에 관한 것으로, 도축 혈액 특유의 냄새가 나지 않으면서도 아미노산이 풍부하여 영양성분이 강화된 동물용 사료첨가제 및 이의 제조방법을 제공할 수 있다. 또한, 도축 부산물을 재활용하여 폐기물을 감소시킬 수 있다.
도 1은 본 발명에 따른 도축 혈액을 이용한 동물용 아미노산 사료첨가제 제조방법의 전체 공정을 도시한 것이다.
이하, 본 발명을 실시예 및 실험예에 의해 상세히 설명한다.
단, 하기 실시예 및 실험예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예 및 실험예에 한정되는 것은 아니다.
<실시예> 본 발명에 따른 아미노산 제조
도축되고 남은 동물의 혈액을 준비하였다. 상기 혈액량 대비 0.5~1.5중량%의 노보자임 알칼라아제를 투입하여 56~60도, 6~12시간 동안 20~40rpm에서 교반하여 단백질을 분해시켰다 이때 pH는 8~8.5로 조정하였다. 이후 맷돌방식으로 특수 제작된 그라인드밀로 4,000~5,000rmp 3~5회 리사이클링하면서 액화시켰다. 이후 80~95도 사이에서 1시간~2시간 더블리본 믹서로 20~30rpm으로 교반시키고, 65~75℃로 냉각시켰다. 냉각 이후 증숙 입자가 깨지지 않게 에어구동식 다이아프람 펌프를 이용하여 탈수기로 압력이송하고, 유압프레스로 2~3kg/cm2 압력하에 탈수시켰다. 이때 탈수된 아미노산 케이크의 함수율은 70%미만이었다. 탈수케이크를 패들 타입 형식으로 제작된 타력파쇄기로 조파쇄하여 펠렛 타입 제형기로 압출하여 2~3mm굵기로 제형화하였다. 압출된 아미노산 펠렛을 배치식 감압건조 장치를 활용하여, 감압건조 장치의 하부에는 스팀 열 교환 발열장치를 설치하고, 건조물 트레이 상단에는 원적외선 파이프 히터를 부착하여 60~90℃로 건조시켜 최종 산물을 얻었다.
<비교예1>
실시예에서의 단백질 분해 효소 중량을 5 중량%로 투여한 것을 제외하고 동일한 방법으로 아미노산을 전환하였다.
<비교예2>
실시예에서의 건조 단계에서 원적외선을 사용한 것 대신 연속식 로터리킬른로에서 60~90℃로 건조시킨 것을 제외하고는 동일한 방법으로 아미노산을 전환하였다.
<비교예3>
실시예에서의 그라인드밀로 혈액 분쇄 후 단백질을 발효분해 과정을 거쳐 액화시키는 단계를 진행하지 않는 것을 빼고 동일한 방법으로 아미노산을 전환하였다.
<실험예1> 아미노산 분리 및 제형화 여부 평가
상기 실시예 및 비교예 1 내지 3에서 전환된 아미노산의 특성을 평가하였다.
실시예에 의해 제조된 아미노산은 도축혈액에서 나는 특유의 악취가 나지 않았을 뿐만 아니라, 펠렛 형태가 잘 유지되었다.
비교예 1은 실시예에 의해 제조된 아미노산 전환구성비는 유사하였으나, 아미노산 고형물이 형성되지 않았다.
비교예 2는 분리된 아미노산이 제대로 건조되지 않아 제형화가 제대로 되지 않아 상품화가 어렵다고 판단되었다. 부스러져서 펠렛 형태가 아니고 분말에 가까웠다. 동물 급이 시 씹어 먹게 되는데 분말은 비산될 뿐 아니라 핥아먹게 되어 불편하고 보관시 굳어지는 경향이 있다.
비교예 3은 제형화는 제대로 이루어졌으나 도축 혈액에서 나는 특유의 악취가 잡히지 않았고, 단백질이 아미노산으로 미전환되어 사료로 사용하기에는 적합지 않다.

<실험예 2> 섭취시 소화율 평가
본 발명에 따른 사료의 섭취시 소화율을 평가하기 위해 위 환경과 유사하게 pH 2로 용액을 조성하여 실시예 및 비교예 1 내지 3에 의해 제조된 사료의 소화율을 평가하였다.
그 결과, 실시예에 따른 사료의 소화율은 98%이상으로 나타났다. 반면 비교예 1에 의한 아미노산은 약 85%, 비교예 2는 약 70%, 비교예 3은 약 90%로 나타났다.
비교예 1은 불순물이 일부 섞여 있어 소화율이 낮아진 것으로 판단되고, 비교예 2는 건조가 제대로 이루어지지 않았기 때문에 소화율이 낮아진 것으로 판단된다. 비교예 3은 소화율은 실시예와 유사하였으나, 악취가 심해 사료용으로 적합하지 않은 것으로 판단되었다.

<실험예 3> 본 발명에 따른 사료용 조성물의 아미노산 배합비 분석
본 발명에 따라 제조된 사료용 조성물에 포함된 아미노산의 배합비를 분석하여 표 1에 나타내었다. 분석 결과, 본 발명에 따른 사료첨가제에 포함된 아미노산은 전체 사료용 조성물의 69.2중량%로 나타났다.
상기 사료용 조성물에 포함된 아미노산 중 필수아미노산은 히스티딘, 이소류신, 류신, 라이신, 메티오닌, 페닐알라닌, 트레오닌, 발린 등 총 7종이 포함되어 있었고, 이는 전체 아미노산 함량 대비 약 46.7%에 해당하는 중량이었다.
따라서 본 발명에 따라 제조된 사료첨가제 또는 사료용 조성물은 아미노산 함량이 매우 높을 뿐만 아니라, 필수 아미노산 함량이 높아 사료로 사용할 경우, 동물의 성장 및 면역력 증진 효과에 뛰어날 것으로 판단된다.

본 발명은 가축용 사료로 사용될 수 있는 도축 혈액에서 분리된 아미노산을 포함하는 사료첨가제 및 이의 제조방법에 관한 것으로, 도축 혈액 특유의 냄새가 나지 않으면서도 아미노산이 풍부하여 영양성분이 강화된 동물용 사료첨가제 및 이의 제조방법을 제공할 수 있다. 또한, 도축 부산물을 재활용하여 폐기물을 감소시킬 수 있기 때문에 산업상 이용가능성이 매우 높다.
Claims (9)
- (a) 가축의 도축된 혈액에 알칼라아제 0.1 내지 2 중량%를 처리하여 6 내지 12시간 동안 단백질을 분해하는 전처리 단계;(b) 상기 (a) 단계에서 전처리된 도축혈액을 80 내지 95℃에서 1 내지 2시간 동안 내측과 외측이 반대로 결합된 더블리본 믹서를 사용하여 증숙하는 단계;(c) 상기 (b) 단계에서 증숙된 도축 혈액을 65 내지 75℃에서 냉각하는 단계;(d) 상기 (c) 단계에서 냉각된 도축 혈액을 2 내지 3kg/cm2의 압력 하에서 함수율 70% 미만으로 탈수하는 단계;(e) 상기 (d) 단계에서 탈수되고 남은 아미노산을 60 내지 90℃에서 감압 원적외선 파이프 히터를 사용하여 건조시키는 단계;를 포함하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항에 있어서, 상기 (a) 단계의 알칼라아제는 노보자임 알칼라아제인 것을 특징으로 하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항에 있어서, 상기 (d) 단계는 함수율 70% 미만으로 탈수하는 것을 특징으로 하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항에 있어서, 상기 (e) 단계의 원적외선은 8~16um의 파장을 갖는 것을 특징으로 하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항에 있어서, 상기 (a) 단계와 (b) 단계 사이에 (a)’ 맷돌방식의 그라인드밀로 4000 내지 5000rpm을 3 내지 5회 반복 수행하는 액화 단계; 를 추가적으로 더 포함하는 것을 특징으로 하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항에 있어서, 상기 (d) 단계 이후에 탈수된 아미노산 또는 아미노산 케이크를 2 내지 3mm의 크기로 정제, 펠렛 또는 입상 형태로 압출 성형시키는 (d)’ 제형화 단계를 추가적으로 더 포함하는 것을 특징으로 하는 도축 혈액 유래 아미노산을 포함하는 사료첨가제 조성물의 제조방법.
- 제1항 내지 제6항 중 어느 한 항의 방법에 따라 제조된 사료첨가제 조성물.
- 제7항에 있어서, 상기 사료첨가제 조성물은 히스티딘, 이소류신, 류신, 라이신, 메티오닌, 페닐알라닌, 트레오닌, 트립토판, 발린으로 이루어진 군에서 선택되는 7종 이상의 필수 아미노산을 포함하고,상기 필수 아미노산은 사료첨가제 조성물에 포함된 전체 아미노산 함량의 45 내지 55 중량%로 포함되는 것을 특징으로 하는 사료첨가제 조성물.
- 제7항 또는 제8항의 사료첨가제 조성물을 전체 중량의 65 내지 75 중량% 포함하는 동물용 사료 조성물.
