Abstract
본 발명은 침엽수 추출물을 함유하는 수면개선용 조성물에 관한 것으로서, 더욱 상세하게는 심신안정에 도움이 되는 알파 피넨을 다량으로 함유하는 침엽수 추출물을 함유하는 수면 개선 천연물 소재로서 숙면을 유도하여 장시간 편한 잠자리를 유도할 수 있도록 수면 개선효과를 발휘하는 수면 개선용 조성물이며, 특히 수면 개선과 수면장애의 예방 또는 치료가 가능하도록 하는 식품 조성물, 약학 조성물, 피부 외용제 조성물 등으로 사용할 수 있는 천연 소재 조성물에 관한 것이다.
Description
본 발명은 침엽수 추출물을 함유하는 수면개선용 조성물에 관한 것으로서, 더욱 상세하게는 심신안정에 도움이 되는 알파 피넨을 다량으로 함유하는 침엽수 추출물을 함유하는 수면 개선 천연물 소재로서 숙면을 유도하여 장시간 편한 잠자리를 유도할 수 있도록 수면 개선효과를 발휘하는 수면 개선용 조성물이며, 특히 수면 개선과 수면장애의 예방 또는 치료가 가능하도록 하는 식품 조성물, 약학 조성물 또는 피부 외용제 조성물로 사용할 수 있는 천연 소재 조성물에 관한 것이다.
수면이란 건강에 매우 중요한 영향을 주는 것으로 잘 알려져 있다. 수면은 중요한 생리기능이 뇌에 의해 뇌를 위하여 운영되는 능동적인 상태로 단순한 활동정지 시간이 아닌 고도의 생리기능으로 유지된 적응 행동이며 생체방어 기술이다. 특히, 발달한 대뇌를 가진 인간에게 있어서 수면의 질은 삶의 질을 좌우할 만큼 큰 영향을 미치기 때문에 숙면을 취한다는 것은 삶의 질 향상에 매우 중요한 것이다.
그동안 수면장애가 건강에 매우 부정적인 영향을 주는 것으로 알려지면서, 다양한 형태의 수면장애 치료용 의약품 등이 다양하게 제안되고 있다.
종래 수면장애 개선을 위한 기술로서, 병풍추출물, 옥촉서예 추출물, 시트러스 과피 추출물 등이 수면장애 개선이나 치료용 조성물로 개발된 바 있다.(한국특허등록 제10-2157248호, 한국특허공개 제10-2019-0106782호, 한국특허등록 제10-2139253호 등 참조)
그 외에도, 화학 물질을 이용한 수면장애 치료용 조성물(한국특허공개 제10-2019-0024593호)이나, 바실러스 섭틸리스 발효물로 얻어지는 폴리감마글루탐산을 이용한 수면장애 치료용 조성물도 제안되고 있다.(한국특허등록 제10-2149404호)
그러나 이러한 기존이 조성물들은 제조단가가 높거나 효과가 미미하여 수면장애 치료 용도로 적용하기 어려운 문제가 있다.
그러므로 최근 각종 스트레스 원인이 많아지면서 수면 장애를 호소하는 인구가 나날이 증가하고 있는 상황에서, 원료 수급이 용이하면서 수면장애 효과도 우수하여 실용화를 통해 다양한 계층에서 널리 활용 가능한 조성물의 개발이 절실한 실정이다.
상기와 같은 종래 기술의 문제점을 개선하기 위하여, 본 발명에서는 기존의 천연물과는 달리 자연에 풍부하게 존재하여 원료 수급과 제조가 용이하면서도 수면 개선 효과가 우수한 천연 소재를 발굴하는 것을 해결과제로 한다.
따라서 본 발명의 목적은 자연에 널리 존재하는 침엽수 추출물로부터 얻어지는 수면개선 또는 수면장애 개선용 조성물을 제공하는데 있다.
또한, 본 발명의 또 다른 목적은 잣나무 추출물을 유효성분으로 함유하는 수면의 질 또는 수면장애 개선용 조성물을 제공하는데 있다.
또한, 본 발명의 또 다른 목적은 침엽수 추출물을 유효성분으로 함유하는 수면개선 또는 수면장애 개선용 건강식품 조성물, 피부외용제 조성물, 또는 수면장애에 의한 질환의 예방 또는 치료용 약학 조성물 등 다양한 용도의 조성물을 제공하는데 있다.
위와 같은 과제 해결을 위하여, 본 발명은 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면 개선 또는 수면장애 관련 질환 또는 증상의 예방 및 개선용 조성물을 제공한다.
또한, 본 발명은 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면 개선 또는 수면장애 개선용 피부 외용제 조성물을 제공한다.
또한, 본 발명은 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면 개선 또는 수면장애 관련 질환 또는 증상의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
또한, 본 발명은 잣나무 추출물을 유효성분으로 포함하는 집중력 강화 및 수면 개선 또는 수면장애 관련 질환 또는 증상의 예방 및 개선용 건간기능식품 조성물을 제공한다.
또한, 본 발명은 잣나무 추출물을 유효성분으로 포함하는 집중력 저하 또는 수면장애 관련 질환 또는 증상의 예방 및 치료용 약학 조성물을 제공한다.
본 발명은 침엽수 추출물의 알파 피넨이 함유된 수면개선 효과를 가지는 천연물 소재로서, 본 발명에 따른 수면개선용 조성물은 수면개선 효과는 물론, 수면장애 관련 질환, 집중력 향상을 통한 집중력 개선 효과, 집중력 저하로 인해 발생될 수 있는 각종 질환의 치료에도 탁월한 효과가 있다.
또한, 본 발명의 천연물 소재로 이루어진 조성물은, 숙면 및 수면의 질 개선, 불면증 등 편안한 잠자리를 유도하는 효과가 있으면서, 또 피부 외용제 조성물로 적용이 가능하기 때문에, 미용 및 화장품으로 적용이 가능하고, 식품, 건강기능식품, 의약품 분야 등에서 다양하게 활용이 가능하다.
또한, 본 발명은 자연에 풍부하게 존재하는 침엽수, 특히 바람직하게는 잣나무로부터 추출물로 얻을 수 있어서, 원료 수급이 용이하며, 경제적으로 제조가 가능한 점에서 상업적으로 매우 유용한 장점이 있다.

이하, 본 발명을 하나의 구현예로서 더욱 상세하게 설명하면 다음과 같다.
본 발명은 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면개선을 위한 천연물 소재에 대한 것이다.
이러한 본 발명의 천연물 소재는 피부 외용제 조성물로 적용이 가능함은 물론 식품용 조성물이나 약학 조성물로 매우 유용한 바이오 소재로 제공될 수 있다.
본 발명의 바람직한 일 실시예에 의하면, 상기 침엽수 추출물에 사용되는 침엽수는 침엽수의 부산물이 사용될 수 있다.
본 발명의 바람직한 일 실시예에 의하면, 상기 침엽수 추출물을 얻기 위한 침엽수로서는 예컨대 소나무, 메타세콰이아, 푸른구상, 전나무, 잣나무, 개잎갈나무, 잎갈나무, 일본잎갈나무, 분비나무, 개비자나무, 눈측백, 편백나무 중에서 하나 이상이 사용될 수 있다. 더욱 바람직하게는 침엽수로 잣나무를 이용하는 경우 더욱 우수한 효과를 얻을 수 있다.
오늘날 현대인은 과중한 업무로 인하여 만성적 피로와 스트레스에 시달리고 있으며, 이러한 피로와 스트레스를 풀어주는 방법 중 하나로서 삼림욕을 즐기는 인구가 늘고 있다. 삼림욕이 피로와 스트레스에 좋은 이유 중 하나는 식품에서 병원균, 해충, 곰팡이에 저항하기 위해 발생하는 피톤치드(Phytoncide)때문인 것으로 알려져 있으며, 이러한 피톤치드는 테르펜을 비롯한 페놀 화합물, 알칼로이드 성분, 글리코시드 등으로 구성된 것이 알려졌다.
그러나 이러한 자연의 피톤치드로 대표되는 유익한 성분 중에서 테르펜이 인체의 건강에 유익한 효과를 나타낼 수 있지만, 특히 침엽수에서 발생되는 테르펜류는 매우 우수한 효과를 나타낸다.
그러므로 본 발명의 바람직한 일 실시예에 의하면, 특별히 침엽수 추출물, 더욱 바람직하게는 잣나무 추출물이 더욱 우수한 효과를 나타내며, 특히 본 발명에 따르면 수면개선 효과, 수면장애 개선효과, 이들로부터 발생하는 질환, 그리고 집중력 개선 또는 집중력 저하로 인한 질환 등에 침엽수 추출물, 특히 잣나무 추출물이 매우 우수한 효과를 나타내는 것으로 확인되었다.
본 발명의 바람직한 일 실시예에 의하면, 상기 침엽수 추출물에 사용되는 침엽수의 부산물로서는, 줄기, 잎, 뿌리, 미성숙 구과, 성숙 구과 중 하나 이상을 이용하여 추출물로 사용할 수 있다. 더욱 바람직하게는 잣나무 부산물의 추출물이 사용될 수 있으며, 바람직하게는 미성숙 구과나 성숙 구과 추출물 또는 잣나무에서 잣을 제거하고 남은 구과를 사용할 수 있다.
본 발명의 일 구현예에 의하면, 잣나무는 일반 침엽수에 비해 피톤치드의 발생량이 적은 대신 인체에 대한 독성이 전혀 없는 것으로 밝혀졌다. 특히, 잣나무의 피톤치드에는 대뇌피질을 자극하여 집중력과 운동량을 증가시키는 알파 피넨의 함량이 가장 많고, 그 외 3-카렌, 보닐-아세테이트 등이 들어 있어 산림향 특유의 효능을 주는 것으로 알려져 있다.
특히, 본 발명의 바람직한 일 실시예에 의하면, 침엽수 추출물 중에서도 잣나무 추출물은 중추신경계에 진정작용을 하여 쾌적한 느낌을 가지게 하며, 수면시간을 연장하고 편안한 숙면을 취하는데 도움을 준다. 그 외에도, 잣나무 추출물은 인체 내에 내성이 생기지 않는 강력한 항균작용이 있으며, 천연물질인 잣나무 추출물은 거의 90 %까지 집먼지진드기 기피효과를 가져와 부작용이 없으며 알레르기 예방에도 매우 효과적이다.
본 발명에서는 다양한 물질들이 숙면 효과가 있는지에 대하여 검증하고자 지속적인 연구를 수행하던 중에 잣나무 추출물 속 알파 피넨(alpha-pinene)은 실험쥐에 투여한 결과 진정작용이 있다는 사실을 알아냈다. 그러나 특이하게도, 알파 피넨은 낮은 농도, 예컨대 25-80 mg/kg 정도의 농도에서는 진정작용을 보이지만, 높은 농도인 약 100 mg/kg 이상의 농도에서는 수면을 개선하는 효과가 월등하게 우수하다는 사실을 확인하였다.
특히, 본 발명의 바람직한 일 실시예에 의하면, 알파 피넨을 함유하는 침엽수 추출물에서 수면 개선 효과가 매우 우수하고, 더 좋기로는 잣나무 추출물에서 월등하게 우수함을 확인하였다.
본 발명의 바람직한 일 실시예에 의하면, 본 발명의 침엽수 추출물은 미성숙 구과, 성숙 구과, 잎, 가지, 뿌리, 껍질 등 중에서 어느 하나를 이용하여 추출하여 얻은 성분으로 사용될 수 있다. 보다 구체적으로는 구과를 사용하여 얻은 성분으로 예컨대 알파 피넨, 베타 피넨, 디-리모넨, 3-카렌 등 다양한 생리활성물질을 함유하고 활성 산소로부터 스트레스를 저하시키고, 불면증을 없애주거나 수면질을 개선시키는 기능이 있다.
본 발명의 바람직한 일 실시예에 의하면, 이러한 침엽수 추출물의 수면개선 효과는 예컨대 수면 지속시간을 연장시킴으로써, 수면 강도를 손상시키지 않으면서 수면의 양을 증가시키는 효과를 가지고 있으며, 수면의 질 등을 개선하고, 수면장애를 예방 또는 치료할 수 있다.
이러한 본 발명의 침엽수 추출물은 수면 개선이나 수면장애 치료를 위해 널리 알려진 졸피뎀보다 더 우수한 것으로 확인되었으며, 기존의 약물과는 달리 천연 소재이므로 부작용 없이 수면 개선, 수면 장애 또는 불안정한 생활, 집중력 저하 등과 관련된 질환의 예방과 치료에 매우 바람직하게 적용할 수 있는 것으로 확인된다.
본 발명의 바람직한 일 실시예에 의하면, 질 좋은 수면을 유도하고 수면 장애 또는 불안을 개선할 수 있는 조성물로서 알파 피넨이 함유된 침엽수 추출물이 함유된 수면개선 천연물 소재를 제공할 수 있다.
또한, 본 발명의 바람직한 일 실시예에 의하면, 알파 피넨이 함유된 침엽수 추출물을 함유하는 수면 개선 천연물 소재를 포함하는 수면장애 예방 및 개선용 식품 조성물 또는 건강기능식품 조성물을 제공할 수 있다.
또한, 본 발명의 바람직한 일 실시예에 의하면, 알파 피넨이 함유된 침엽수 추출물을 함유하는 수면 개선 천연물 소재를 포함하는 수면장애 예방 및 치료용 약학 조성물을 제공할 수 있다.
한편, 본 발명의 바람직한 일 실시예에 의하면, 알파 피넨이 함유된 침엽수 추출물을 함유하는 수면 개선 천연물 소재를 포함하는 피부 외용제 조성물을 제공할 수 있다.
본 발명의 바람직한 일 실시예에 의하면, 상기 피부 외용제 조성물은 상기 천연 소재를 함유하는 조성물 총 중량에 대하여 80~100 중량%의 양으로 포함하는 피부 외용제 조성물이 바람직하다.
본 발명의 바람직한 일 실시예에 의하면, 본 발명의 식품 조성물은 식품, 기능성 식품, 영양보조제, 건강식품 및 식품 첨가제 등의 모든 천연소재의 가공 형태를 포함한다. 상기 유형의 식품 조성물은 당 업계에 공지된 통상적인 방법에 따라 다양한 형태로 제조할 수 있다.
예를 들면, 건강식품으로는 잣나무 추출물 자제를 차, 주스 및 드링크의 형태로 제조하여 음용하도록 하거나, 과립화, 캡슐화 및 분말화하여 섭취할 수 있다. 또한, 본 발명의 잣나무 추출물과 집중력 강화, 수면장애 증상의 예방 또는 개선의 효과가 있다고 알려진 공지의 활성 성분과 함께 혼합하여 조성물의 형태로 제조할 수 있다.
또한, 식품으로는 비알콜성 음료, 알코올성 음료, 과실 및 그의 가공식품(예: 과일통조림, 병조림, 잼, 마아말레이드 등), 어류, 육류 및 그 가공식품(예: 햄, 소시지 콘비이프 등), 빵류 및 면류(예: 우동, 메밀국수, 라면, 스파게티, 마카로니 등), 과즙, 각종 드링크, 쿠키, 엿, 유제품(예: 버터, 치즈 등), 식용식물유지, 마아가린, 식물성 단백질, 레토르트 식품, 냉동식품, 각종 조미료(예: 된장, 간장, 소스 등) 등에 잣나무 추출물을 첨가하여 제조할 수 있다. 또한 본 발명의 잣나무 추출물을 식품 첨가제의 형태로 사용하기 위해서는 분말 또는 농축액 형태로 제조하여 사용할 수 있다.
본 발명의 식품 조성물 중 잣나무 추출물의 바람직한 함량은 식품 조성물 총 중량에 대하여 0.01 내지 100 중량%일 수 있다.
본 발명에 따른 약학적 조성물은 약학적으로 유효한 양의 잣나무 추출물을 단독으로 포함하거나 하나 이상의 약학적으로 허용되는 담체, 부형제 또는 희석제를 추가로 포함할 수 있다. 상기에서 “약학적으로 허용되는”이란 생리학적으로 허용되고 인간에게 투여될 때, 활성성분의 작용을 저해하지 않으며 통상적으로 위장 장애, 현기증과 같은 알레르기 반응 또는 이와 유사한 반응을 일으키지 않는 비독성의 조성물을 말한다.
상기 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말리톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하리드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다. 또한 상기 약학 조성물은 충진제, 항응집제, 윤활제, 습윤제, 항료, 유화제 및 방부제 등을 추가로 포함할 수 있다.
상기에서 “약학적으로 유효한 양”이란 음성 대조군에 비해 그 이상의 반응을 나타내는 양을 말하며 바람직하게는 집중력 향상 및 수면장애로 유발되는 증상 예방 및 치료의 효과를 나타내기에 충분한 양을 말한다.
본 발명에 따른 잣나무 추출물의 약학적으로 유효한 양으로는 0.01 내지 200 mg/day/kg체중이다. 그러나 상기 약학적으로 유효한 양은 질환의 종류 및 이의 중증정도, 환자의 연령, 체중, 건강상태, 성별, 투여 경로 및 치료기간 등과 같은 여러 인자에 따라 적절히 변화될 수 있다.
또한, 본 발명의 약학적 조성물은 포유동물에 투여된 후 활성 성분의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 당업계에 공지된 방법을 사용하여 제형화 할 수 있다, 제형은 분말, 과립, 정제, 에멀젼, 시럽, 에어로졸, 연질 또는 경질 젤라틴 캡슐, 멸균 주사용액, 멸균 분말의 형태일 수 있다.
본 발명 조성물의 투여 경로로는 이에 한정되지는 않으나, 경구적 또는 비경구적으로 투여될 수 있다. 비경구적 투여 경로로는 예를 들면, 경피, 비강, 복강, 근육, 피하 또는 정맥 등의 여러 경로가 포함될 수 있다.
본 발명의 약학적 조성물은 집중력 강화 및/또는 수면장애 관련 질환 또는 증상의 예방 또는 치료의 효과를 가지는 공지의 화합물과 병행하여 투여할 수 있다.
이와 같이, 본 발명의 침엽수 추출물, 더욱 바람직하게는 잣나무 추출물은 남녀 모두 연령과 상관없이 수면장애 관련 질환 또는 증상의 예방 및 치료를 위한 건강식품 및 의약품, 피부 외용제 등에 유용하게 적용될 수 있다.
본 발명의 바람직한 일 실시예에 의하면, 본 발명에 의한 침엽수 추출물을 함유하는 수면개선 조성물은 향수, 화장품 등의 피부 외용제 조성물에 배합할 수 있으며, 식품 조성물로도 배합할 수 있다. 배합량은 당업계에 통상적인 기술에 따라 목적하는 효과를 이루기 위해 적절하게 선택하여 배합할 수 있으며, 바람직하게는 조성물 총 중량에 대하여 0.01~40 중량%의 양으로 혼합될 수 있다.
본 발명의 바람직한 일 실시예에 의하면, 상기 피부 외용제는 그 제형에 있어서 한정적이지 않고 당업계에 주지된 어떠한 제형에도 배합할 수 있다. 제형의 예로는 팩, 연고, 로션, 가용화상, 현탁액, 에멀젼, 크림, 젤, 스프레이, 파스계, 경고제, 패치제 또는 물파스 등을 포함한다.
또한, 멤브레인 등 용기에 액상, 고체, 겔상 등을 충진하여 사용할 수도 있다.
본 발명의 바람직한 일 실시예에 의하면, 상기 식품 조성물은 본 발명에서는 잣나무 추출물 수면 개선 효과를 구명하기 위하여, 수면을 취하는 중 뇌파, 안전도, 심전도를 측정하여 수면다원 검사 및 수면에 대한 평가를 실시하였다.
본 발명의 바람직한 일 실시예에 의하면, 하기의 시험예의 결과에서 볼 수 있듯이 본 발명의 잣나무 추출물 수면개선 조성물을 사용함으로써, 수면 다원검사 결과로부터 수면 효율이 증가하고 입면 지연시간이 단축됨을 확인할 수 있다. 또한, 수면 후에 실시한 주관적인 평가 결과로부터 긴장감의 감소, 뒤척임 정도의 감소, 전체적 수면의 만족감이 높은 것을 확인할 수 있다.
이하. 본 발명을 실시예에 의하여 본 발명을 보다 상세히 설명하겠는바, 본 발명이 이들 예로만 한정되는 것은 아니다.
시험예 1 : 잣나무 추출물과 소나무 추출물을 이용한 수면개선효과실험
잣나무 추출물의 수면개선효과를 확인하기 위하여, 같은 침엽수 종인 소나무 추출물과의 비교실험을 통해 잣나무 추출물의 수면개선효과 유의성을 확인하고자, 펜토바르비탈로 유도한 동물모델 마우스의 수면유도시간 및 수면시간을 측정하였다. 구체적으로, 펜토바르비탈 45 mg/kg을 복강투여하였으며, 잣나무 추출물과 소나무 추출물은 경구투여하여 실험하였다.
수면은 정향반사가 소실되는 시점으로 하였으며, 깨는 시간은 동물이 정향으로 돌아오는 시간으로 정하였다.
잣나무 추출물과 소나무 추출물을 이용하여 수면개선효과를 확인하고자, 마우스에 각각 100 mg/kg의 추출물을 경구투여 후 펜토바르비탈 수면 테스트를 진행한 결과, 다음 표 1과 같다.

상기 표 1에 나타난 바와 같이, 마우스에 펜토바르비탈 복강투여 후 잣나무 추출물을 경구 투여한 마우스의 수면유도시간은 129.55±18.2904초로서, 소나무 추출물을 경구 투여한 마우스의 수면유도시간인 154.22±24.3090초에 비해 다소 감소하는 것을 확인할 수 있었다. 또한, 상기 동물모델에 잣나무 추출물을 투여한 마우스의 수면시간은 5406.00±455.8588초로서, 소나무 추출물을 경구 투여한 마우스의 수면시간인 2461.37±246.9644초에 비해, 유의하게 증가하는 것을 확인할 수 있었다.
이러한 결과는 잣나무 추출물이 수면유도시간을 단축시키고, 특히 수면시간을 유의성 있게 증가시키는 효과가 있음을 보여주는 것이다.
이러한 결과를 통하여, 수면장애에 대하여 잣나무 추출물이 수면유도시간을 단축시키고, 수면시간을 연장시킬 수 있음을 확인함으로써, 잣나무 추출물이 수면개선효과가 있음을 확인할 수 있었다.
시험예 2 : 잣나무 추출물 수면개선 조성물 소재의 숙면 효과 측정
본 발명에 따른 잣나무 추출물(잣을 제거한 구과 추출물) 수면개선 조성물 소재의 숙면 효과를 객관적으로 평가하기 위하여 상기 표 1에서 제조된 잣나무 추출물 수면개선 조성물 소재를 이용하여, 수면을 취하는 중 뇌파, 안전도, 심전도를 측정하여 수면다원 검사 및 수면에 대한 주관적 평가를 실시하였다. 구체적인 방법은 아래와 같다.
1. 피험자
조사대상자는 건강하고, 현재 약물 복용을 하지 않으며, 취맹이 아닌 10~40대 여성 200명을 대상으로 하였다.
피험자는 실험 전일에 무리한 운동이나 과다한 음주 섭취를 자제하도록 하였고, 실험 당일에는 중추신경계와 후각감각에 영향을 줄 수 있는 흡연, 음료 및 약물, 껌 섭취를 삼가도록 하였다. 성인 여성을 대상으로 하는 실험으로 여성의 성주기에 따른 향에 대한 감각의 변화, 여성의 생리주기와 기분이 상관관계가 있다고 보고되어 조사대상자의 생리주기를 고려하여 실시하였다.
2. 실험 시료
실험 시료는 비교예로서 향을 제시하지 않은 무향 조건과 잣나무 추출물 수면개선 조성물 소재를 사용하였다. 수면 실험실에 일정하게 향을 제시하였고, 수면을 취하는 중의 뇌파(EEG), 안전도(EOG), 심전도(ECG)를 측정하여 수면다원 검사를 실시하였다.
3. 실험장소
실험은 표준과학 연구원에서 구축한 수면 실험실(4.8 m x 3 m x 2.4 m)에서 수행되었고, 내부에는 실험 시 외부 환경의 영향을 배제하기 위하여 방음장치가 설치되어 있다. 수면 실험실은 항온·항습이 유지되며 온도는 24 ℃, 습도는 40 %였다.
4. 측정방법
수면 중 생리신호는 수면다원검사장치를 이용하여 뇌파(EEG), 안전도(EOG), 심전도(ECG)를 측정하였다. 수면에 대한 주관적 평가는 피실험자가 느끼는 수면에 대해서 실험이 끝난 후 작성하도록 하였다.
5. 실험순서
수면 다원검사는 대조예와 실시예의 조건에서 3회 실험을 실시하였다.
수면에 대한 효과를 배제하기 위하여 피험자마다 시료를 무작위로 실시하였으며, 본 실험을 실시하기 전에 수면 실험실에 대한 적응을 위해서 1~2회 수면 실험실에서 수면을 취하도록 하였다. 수면 시간은 주간에 2시간 동안 취하도록 하였으며, 각 피실험자 마다 같은 시간에 수면을 취하도록 하였다. 또한 수면 이후에 수면에 대한 주관적인 평가를 설문지를 통하여 수집하였다.
6. 분석방법
피실험자가 수면을 취하는 동안 수면다원검사장치를 이용하여 생리신호를 측정하였으며, 측정된 신호를 이용하여 수면단계를 분석하였다. 수면단계의 판정은 국제표준인 Rechtschaffen and Kales(1968)의 방법을 사용하였다.
구분되어진 수면단계를 이용하여, 입면 지연시간(sleep latency) 및 수면효율(sleep efficiency)을 비교하였다. 또한, 주어진 조건에서 수면을 실시한 이후 주관적인 느낌을 설문을 통하여 수집하고 SPSS프로그램을 이용하여 t-test를 사용하여 통계적인 분석을 실시하였다.
7. 실험결과
가. 입면 지연시간(sleep latency)

상기 표 2 및 도 1 에서 확인할 수 있는 바와 같이, 입면 지연시간의 평균은 잣나무 추출물이 함유된 실험군이 잣나무 추출물이 함유되지 않은 대조군에서 보다 짧았으며 이는 잣나무 추출물이 포함된 실험군이 빠른 시간 안에 수면에 드는 것을 의미한다.
나. 수면효율(sleep efficiency)
수면효율은 침상에 있는 시간에 대한 총 수면시간의 비율이며, 실험 기간 동안 수면을 취한 시간의 비율을 비교하였다.
상기 수면효율에 대한 결과는 하기 표 3 및 도 2에 나타내었다.

상기 표 3 및 도 2에서 확인할 수 있는 바와 같이, 수면효율의 평균을 보면, 잣나무 추출물을 함유한 실험군이 대조군에서보다 수면효율이 높았으며, 이는 잣나무 추출물이 함유된 실험군이 향이 없을 때 보다 오랜 시간 동안 수면에 드는 것을 의미한다.
다. 주관적 평가
① 긴장 정도
하기 표 4 및 도 3는 평소 수면과 비교하여 수면 시 긴장 정도를 보여주는 도표이다.

상기 표 4 및 도 3에서 확인할 수 있는 바와 같이, 수면효율이 평균은 잣나무 추출물 수면개선 조성물 소재를 포함한 실험군이 대조군에서보다 수면효율이 높았으며, 이는 잣나무 추출물 수면개선 조성물 소재가 함유된 실험예가 향이 없을 때 보다 오랜 시간 동안 수면에 드는 것을 의미한다.
② 뒤척임 정도
하기 표 5 및 도 4는 평소와 비교해 수면시 뒤척거린 정도를 보여주는 도표이다.

상기 표 5 및 도 4에서 확인할 수 있는 바와 같이, 대조군 조건에서는 평소보다 뒤척인 것으로 답하였으며, 실험군 조건에서는 평소보다 적게 뒤척인 것으로 답하여 실험군을 사용한 경우 뒤척임 정도가 현저하게 차이가 있는 것으로 나타났다.
③ 전체적인 수면 정도
하기 표 6 및 도 5는 평소와 비교해 전체적인 수면에 대한 느낌 정도를 보여주는 도표이다.

상기 표 6 및 도 5에서 확인할 수 있는 바와 같이, 대조군 조건에서는 평소와 비교해 비슷한 수준의 수면을 취하였으나 실험군 조건에서는 평소와 좋은 수면을 취하였다고 답하여, 잣나무 추출물 수면개선 소재가 함유된 실험군을 사용한 경우 전체적인 수면 만족도가 매우 우수한 것으로 나타났다.
하기 조성에 따라 다양한 제형의 건강기능식품 및 약학조성물, 피부 외용제를 제조하였다.
아래에 본 발명의 약학 조성물 또는 건강기능식품 또는 피부 외용제 조성물의 제제예를 설명하나, 본 발명은 이를 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.
[제제예 1] 정제의 제조
침엽수 추출물 120 mg
옥수수 전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
상기의 성분들을 혼합한 후 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조한다.
[제제예 2] 캅셀제의 제조
침엽수 추출물 120 mg
결정성 셀룰로오스 3 mg
옥수수 전분 100 mg
스테아린산 마그네슘 2 mg
통상의 캡슐제 제조방법에 따라 상기의 성분을 혼합하고 젤라틴 캡슐에 충전하여 캡슐제를 제조한다.
[제제예 3] 주사제의 제조
침엽수 추출물 120 mg
만니톨 180 mg
주사용 멸균 증류수 3000 mg
통상의 주사제의 제조방법에 따라 1앰플 당 상기의 성분 함량으로 제조한다.
[제제예 4] 피부 외용제
침엽수 추출물 100 중량%
상기 제조된 침엽수 추출물을 그대로 피부 외용제로 사용할 수 있다.
Claims (9)
- 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면 개선 또는 수면장애 관련 질환 또는 증상의 예방 또는 개선용 조성물.
- 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면 개선 또는 수면장애 개선용 피부 외용제 조성물.
- 청구항 2에 있어서, 침엽수는 잣나무인 피부 외용제 조성물.
- 청구항 2에 있어서, 상기 피부 외용제 조성물은 상기 침엽수 추출물을 전체 중량의 80~100 중량%의 양으로 포함하는 피부 외용제 조성물.
- 상기 청구항 2 내지 청구항 4 중 어느 하나의 항에 따른 피부 외용제 조성물을 함유하는 화장료 조성물.
- 상기 청구항 2 내지 청구항 4 중 어느 하나의 항에 따른 피부 외용제 조성물을 함유하는 수면장애 질환 피료용의 외피용 약학 조성물.
- 알파 피넨이 함유된 침엽수 추출물을 유효성분으로 함유하는 수면개선 또는 수면장애 관련 질환 또는 증상의 예방 또는 개선용 건강기능식품 조성물.
- 잣나무 추출물을 유효성분으로 포함하는 집중력 강화, 수면 개선 또는 수면장애 관련 질환 또는 증상의 예방 또는 개선용 건강기능식품 조성물.
- 잣나무 추출물을 유효성분으로 포함하는 집중력 저하 또는 수면장애 관련 질환 또는 증상의 예방 또는 치료용 약학 조성물.
