Abstract
본 발명의 공결정, 이를 포함하는 약학적 조성물 및 이의 제조 방법에서, 새로운 공결정은 니클로사마이드 및 나파모스타트 또는 이의 약학적으로 허용가능한 염을 포함한다. 본 발명의 새로운 공결정은 우수한 안정성 및 용해도를 가지고, 이의 제조 방법에 따라 우수한 순도 및 수율로 수득할 수 있다. 본 발명의 공결정을 이용하여, 암 또는 코로나바이러스 감염 질환을 효과적으로 예방 및/또는 치료할 수 있다.

【기술분야】 본 발명은 신규한 공결정, 이를 포함하는 약학 조성물 및 이의 제조 방법에 관한 것으로, 보다 구체적으로는 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정, 이를 포함하는 약학 조성물 및 이의 제조 방법에 관한 것이다.
【배경기술】 공결정 (cocrystal)은, 서로 다른 2종 이상의 약물들 또는 약물과 공결정 형성자 (co- former)가 수소 결합 (hydrogen bond) , 반데르발스 결합 (van der waals interaction) , 및 파이 상호작용 (冗-冗 stacking interact ion)과 같은 방법으로 결합된 결정성 배향을 갖는 고체를 말한다. 이러한 공결정은 2 분자 이상의 새로운 수소결합을 통해 결정 구조를 이루기 때문에, 약물의 용해도 및 용해 속도를 변화시키고, 나아가 인체 내의 약물의 흡수율을 변화시켜 생체 이용률을 변화시킬 수 있다 .
이러한 이유로 공결정화 기술은 난용성 물질 및 생체 흡수율이 낮은 물질에 대하여 용해도와 경구율을 개선할 목적으로 제약 또는 식품 분야에서 관심을 받고 있다. 니클로사마이드 (Niclosamide)는 하기 화학식 1 로 나타내는 약물이다.

니클로사마이드는 수용해도 및 장관투과도가 매우 낮고 생체이용율이 극도로 낮음에 따라 과량의 니클로사마이드를 경구 투여하는 방법으로 처방되고 있지만, 니클로사마이드 자체의 특성에 의해서 근본적으로 약물의 전신노출에 의한 약효를 기대하기 힘들다. 최근에는 니클로사마이드가 다양한 종류의 암에 대한 항암 요법에 사용될 수 있고 , 세포의 엔도좀 중성화를 통해 코로나 19 바이러스 Coronavirus 2019)의 세포 내입 및 전달과정을 억제하여 코로나를 치료할 수 있다고 보고되고 있으나 여전히 낮은 용해도에 의해 이들의 치료 또는 개선 효과를 얻는데 한계가 있는 실정이다.
나파모스타트 (Nafamostat)는 하기 화학식 2 로 나타내는 약물로서, 항응고제로서 췌장염 치료제로 사용될 뿐만 아니라, 잠재적인 항바이러스 및 항암 효과도 있는 것으로 알려져 있다. 최근에는 나파모스타트가 세포 표면의 TMPRSS2의 활동을 억제함으로써 , 예를 들면 TMPRSS2의 S 단백질 프라이밍이 필요한 코로나바이러스 감염증- 19(Coronavirus disease 2019, COVID- 19)의 치료 및 예방에 유용한 화합물인 것을 보고 되고 있다 (Ko, M. , Jeon, S., Ryu, W. 【발명의 상세한설명】
【기술적 과제】 본 발명자들은 니클로사마이드의 낮은 용해도 및 나파모스타트의 낮은 안정성을 향상시킬 수 있는 기술을 개발하고자 연구 및 노력하였다. 그 결과, 본 발명자들은 나파모스타트와 니클로사마이드를 포함하면서 안정성과 용해도가 모두 개선된 공결정을 확보할 수 있었으며 이를 통해 본 발명을 완성하게 되었다.
본 발명의 일 목적은 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정 , 이를 포함하는 약학적 조성물 및 이의 제조 방법을 제공하는 것이다. 본 발명의 다른 목적은 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정을 포함하는 , 암 또는 코로나바이러스 감염 질환의 예방 또는 치료를 위한 약학적 조성물을 제공하는 것이다. 본 발명의 또 다른 목적은 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정을 대상체에게 투여하는 단계를 포함하는 암 또는 코로나바이러스 감염 질환의 예방 또는 치료 방법을 제공한다. 본 발명의 또 다른 목적은 암 또는 코로나바이러스 감염 질환의 예방 또는 치료를 위한, 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정의 용도를 제공한다 . 본 발명의 또 다른 목적은 암 또는 코로나바이러스 감염 질환의 예방 또는 치료용 약제의 제조를 위한, 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정의 용도를 제공한다 .
【기술적 해결방법】 이를 구체적으로 설명하면 다음과 같다. 한편 , 본 발명에서 개시된 각각의 설명 및 실시형태는 각각에 대한 다른 설명 및 실시형태에도 적용될 수 있다. 즉 , 본 발명에 개시된 다양한 요소들의 모든 조합이 본 발명의 범주에 속한다. 또한, 하기에 기술된 구체적인 서술에 의하여 본 발명의 범주가 제한된다고 볼 수 없다.
공결정 및 이의 제조 방법 본 발명의 공결정은 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드를 포함한다 . 본 발명의 공결정은 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드를 포함하는 복합체일 수 있다 . 나파모스타트 또는 이의 약학적으로 허용가능한 염에서 , “약학적으로 허용가능한 염 ” 은 당업계의 통상의 기술자에게 공지된 통상적인 방법으로 제조되는 염이면 특별히 제한 되지 않는다. 예를 들어 , 나파모스타트의 약학적으로 허용가능한 염은, 메탄설폰산, 에탄설폰산, 벤젠설폰산, p- 톨루엔설폰산, 나프탈렌설폰산 등으로 제조된 설폰산염일 수 있다. 본 발명의 공결정은 한 분자의 니클로사마이드 또는 네 분자의 니클로사마이드가 한 분자의 나파모스타트와 결합된 구조를 가질 수 있다 . 일 실시 예에서 , 본 발명의 공결정은 나파모스타트와 니클로사마이드가 1:1 의 몰비로 포함된 공결정 A 일 수 있다. 일 실시 예에서 , 본 발명의 공결정은 나파모스타트와 니클로사마이드가 1:4 의 몰비로 포함된 공결정 B 일 수 있다. 일 실시 예에서 , 본 발명의 나파모스타트와 니클로사마이드가 1:1 의 몰비로 포함된 공결정 A는 92.96 내지 225.66 으仁에서 시차주사열량 (DSC) 흡열 피크를 나타낸다. 이때 , 시차주사열량 흡열 피크는 승온속도가 io °C /min 인 경우에 나타나는 것이다.

프로판올 등을 들 수 있다. 일례로, 상기 용매로서는 물과 함께 에탄올을 이용하거나, 물과 함께 아세톤을 이용할수 있다. 일 실시예에서, 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드는 1:1 또는 1:4의 몰 비로혼합될 수 있다. 상기 용매를 제거하여 공결정을 얻는 단계는, 상기 혼합 용액을 여과를 통해 용매를 제거한후 감압조건에서 건조 시키는 단계를 포함할수 있다. 건조 공정을 통해서 최종적으로고체 상의 공결정 분말을 얻을수 있다. 일 실시예에서, 나파모스타트의 염으로서 나파모스타트 메실산염을 이용할 수 있다. 약학적 조성물 및 용도 본 발명의 공결정을 유효 성분으로서 포함하는 약학적 조성물은, 통상적으로 사용되고 있는 약학적으로 허용 가능한 제제용의 담체나 부형제, 그 밖의 첨가제를 이용하여 정제, 산제, 세립제, 과립제, 캡슐제, 환제, 액제, 주사제, 좌제, 연고 및 첩부제 등으로 제조되어 경구적 또는 비경구적으로 투여될 수 있다. 본 발명에 따른 공결정을 대상체에 대한 투여량 (치료 유효량)은 적응증, 질환의 중증도, 체중, 연령, 성별 등을 고려하여 적절하게 결정될 수 있으며, 투여 용량 및 용법은 여러 가지 조건으로 변동될 수 있다. 약학적으로 허용 가능한 담체는, 구체적으로 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알지네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로오스, 폴리비닐피롤리딘, 셀룰로오스, 물,
시럽, 메틸 셀룰로오스, 메틸 히드록시벤조네이트, 프로필 히드록시벤조네이트, 활석, 스테아르산 마그네슘, 미네랄, 또는 오일일 수 있으나, 이에 제한되는 것은 아니다. 제제는 당업계에서 제제화에 사용되는 통상의 방법으로 제조될 수 있으며 , 각 질환또는 성분에 따라다양한 제제로 제제화될 수 있다. 경구 투여를 위한 액체 조성물은, 약제적으로 허용되는 유탁제, 용액제, 현탁제, 시럽제, 엘릭실제 등을 포함하며, 일반적으로사용되는 불활성인 희석제, 예를 들면 정제수, DMSO, Tween 20, Tween 80 또는 에틸알콜을 포함할 수 있다. 당해 조성물은 불활성인 희 석제 이외에 가용화제 , 용해보조제 , 습윤제 , 현탁제와 같은 보조제, 감미제, 풍미제, 방향제, 방부제를 함유할수 있다. 비경구 투여를 위한 주사제로서는, 무균의 수성 또는 비수성의 용액제, 현탁제, 유탁제 등을 포함할 수 있다. 수성의 용액제, 현탁제의 희석제로서는, 예를 들면 주사제용 증류수, 생리식염수 등을 들 수 있다. 비수용성의 용액제, 현탁제의 희석제로서는 예를 들면 프로 필렌글리콜, 폴리에틸렌글리콜, 올리브유, 콘 오일 등과 같은 식물유, 에틸알콜과 같은 알코올류, 폴리솔베이트 80과 같은 계면활성제 등이 있다. 본 발명의 약학적 조성물은 등장화제, 방부제, 습윤제, 유화제, 분산제, 안정화제, 가용화제 또는 용해보조제와 같은 첨가제를 포함할 수 있다. 이들은 무균화 하기 위해 박테리아 보류 필터를 통과시키는 여과, 살균제의 배합 또는 자외선 조사에 의해서 무균화 될 수 있다. 또한, 본 발명에서는 무균의 고체 조성물을 제조하여 사용 전에 무균화된 주사용 용매에 용해시켜 사용할수 있다.

SARS-CoV-2 감염 질환의 증상은 발열, 권태감, 기침, 호흡곤란, 가래, 인후통, 두통, 객혈, 오심, 위장관 증상, 신장질환, 호흡기 질환, 설사 등일 수 있다. 본 발명은, 나파모스타트 또는 이의 약학적으로 허용가능한 염; 및 니클로사마이드의 공결정을 대상체에 투여하는 단계를 포함하는 암 또는 코로나바이러스 감염 질환의 예방또는 치료방법을 제공한다. 본 발명의 암 또는 코로나바이러스 감염 질환의 예방 또는 치료 방법은 나파모스타트 또는 이의 약학적으로 허용가능한 염; 및 니클로사마이드의 공결정을 치료학적으로유효한 양으로투여하는 것일 수 있다. 본 발명의 용어, "치료학적으로 유효한 양"이란 의학적 치료에 적용가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분하며 부작용을 일으키지 않을 정도의 양을 의미하며, 이는 환자의 성별, 연령, 체중, 건강상태, 질병의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 방법, 투여 시간, 투여 경로, 배출 비율, 치료 기간, 배합 또는 동시에 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 결정될 수 있다. 특정 환자에 대한 구체적인 치료적 유효량은 달성하고자 하는 반응의 종류와 정도, 경우에 따라 다른 제제가 사용되는지의 여부를 비롯한 구체적 조성물, 환자의 연령, 체중, 일반 건강 상태, 성별 및 식이, 투여 시간, 투여 경로 및 조성물의 분비율, 치료기간, 구체적 조성물과 함께 사용되거나 동시 사용되는 약물을 비롯한 다양한 인자와 의약 분야에 잘 알려진 유사 인자에 따라 다르게 적용하는 것이 바람직하다.
본 발명은 , 암 또는 코로나바이러스 감염 질환의 예방 또는 치료를 위한, 나파모스타트 또는 이의 약학적으로 허용가능한 염 ; 및 니클로사마이드의 공결정의 용도를 제공한다. 본 발명은 , 암 또는 코로나바이러스 감염 질환의 예방 또는 치료용 약제의 제조를 위한, 나파모스타트 또는 이의 약학적으로 허용가능한 염 ; 및 니클로사마이드의 공결정의 용도를 제공한다 . 본 발명의 약학적 조성물 , 치료 방법 및 용도에서 언급된 사항은 서로 모순되지 않는 한 동일하게 적용된다.
【발명의 효과】 본 발명에 따르면 , 안정성과 수용해도가 개선된 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정을 확보함으로써 복합체 원료 의약품으로 유용하게 사용할 수 있다. 본 발명에 따른 공결정은 특정 용매환경에 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 당량, 교반 속도, 교반 시간, 교반 용매의 종류 , 교반 용매의 양, 교반 용매 간의 비율을 조절하여 공결정화 시킴으로써 최적 비율의 신규 공결정을 우수한 순도 및 수율로 수득할 수 있다. 안정성과 수용해도가 개선된 나파모스타트 또는 이의 약학적으로 허용가능한 염과 니클로사마이드의 공결정을 이용하여 , 암 또는 코로나바이러스 감염 질환을 효과적으로 예방 및/또는 치료할 수 있다.

【발명의 실시를 위한 형태】 이하, 본 발명을 실시하기 위한 형태를 구체적으로 설명한다.
본 발명의 발명자들은 나파모스타트와 니클로사마이드의 몰 비가 1:1인 공결정 A 와 나파모스타트와 니클로사마이드의 몰 비가 1:4 인 공결정 으 을 개발하였으며, 공결정 형성을 확인하기 위해 아래와 같이 얻어진 결과물을 다양한 분석 방법으로 분석하였다. 본 발명의 공결정에 있어서는, 상기한 바와 같이 일정한 품질을 갖는 단일의 공결정이 재현성이 양호하게 수득된다. 또한, 본 발명의 공결정은 의약의 제조에 사용되는 원약(의약원료 물질)의 결정으로서 안정적으로 공급할 수 있고 보존 안정성이 우수하다. 단순 혼합물과 공결정의 결정형의 차이는 시차주사열량계 분석 (DSC 분석) , 열중량 분석 (TGA 분석) 및 분말 X선 회절 (PXRD분석)의 결과로부터 명확하다고 할 수 있다. 니클로사마이드 및 나파모스타트 메실산염은 표 1, 도 1 및 도 2 에 기재되는 PXRD 회절각 및 상대 강도에 의해서 특정된다. 이와 대조적으로, 본 발명의 공결정으로서 , 공결정 A 및 공결정 B는 표 2, 표 3 및 표 4 각각에 기재되는 회절각, 열흐름 및 열중량 측정결과 의하여 특정 지어진다. 또한 XRD 회절은 데이터 특성상, 결정의 동일성 인정에 있어서는 결정 격자 간 격이나 전체적인 패턴이 중요하고, 열흐름 및 열중량 측정결과는 결정 성장의 방향, 입자의 크기, 측정 조건에 따라 다소 변할 수 있기 때문에 엄밀하게 해석되어서는 안 된다. 본 발명에 따른 공결정의 제조 방법은, 나파모스타트 또는 이의 약학적으로 허용가능한 염 및 니클로사마이드를 혼합하여 공결정화시키는 단계를 포함한고, 이때의 공결정화시키는 단계는, 공결정의 제조 방법으로 알려진 다양한 방법으로 수행될 수 있다. 예를 들어 , 본 발명에 따른 공결정 A및 으는
각각 독립적으로 액체 보조 연삭법 (liquid-assisted grinding), 슬러리법
(slurry method) , 용매 증발법 (solvent evaporation method) , 용매 냉각법 (solvent cooling method) 등의 결정화 방법을 활용해 제조할 수 있다. 액체 보조 연삭법에서는, 나파모스타트 또는 이의 약학적으로 허용 가능한 염과 알칼리화제를 미량의 증류수 등의 용매와 함께 막자와 사발과 같은 기구를 사용해 갈아서 반응시킨 뒤 미량의 아세톤 혹은 알코올류의 유기용매와 니클로사미드를 첨가하여 지속적으로 갈아서 본 발명에 따른 공결정을 합성할 수 있다. 슬러리 법에서는, 나파모스타트 또는 이의 약학적으로 허용 가능한 염과 알칼리화제를 증류수 등의 용매에서 교반하여 반응시킨 뒤 아세톤 혹은 알코올류의 유기용매와 니클로사미드를 첨가하여 과포화 용액으로 만든 뒤 지속적으로 교반하여 본 발명에 따른 공결정을 합성할 수 있다. 용매 증발법에서는, 나파모스타트 또는 이의 약학적으로 허용 가능한 염과 알칼리화제를 증류수 등의 제 1 용매에서 교반하여 반응시킨 뒤 아세톤 혹은 알코올류의 제 2 용매에서 가열하여 깨끗하게 녹인 니클로사미드 용액을 첨가하고 용매를 증발시키면서 교반하여 본 발명에 따른 공결정을 합성할 수 있다. 상기 제 1 용매와 제 2 용매의 부피비는 10:1 내지 1:10일 수 있다. 일례로, 상기 제 1 용매와 제 2 용매의 부피비는 4:1 내지 5:1일 수 있다. 용매 냉각법에서는, 나파모스타트 또는 이의 약학적으로 허용 가능한 염과 알칼리화제를 증류수 등의 제 1 용매에서 교반하여 반응시킨 뒤 아세톤 혹은 알코올류의 제 2 용매에서 가열하여 깨끗하게 녹인 니클로사미드 용액을
첨가하고 용매를 냉각시키면서 교반하여 본 발명에 따른 공결정을 합성할 수 있다. 상기 방법들에서 , 알칼리화제로는 탄산수소나트륨, 탄산나트륨, 수산화나트륨, 수산화칼륨, 수산화칼슘, 수산화마그네슘 등의 염기성 물질을 이용할 수 있고, 특별히 이에 제한되지 않는다. 이하에서는 본 발명을 실시예에 의해 구체적으로 설명하지만, 본 발명은 이러한 실시예에 의해 특별히 제한되는 것은 아니다. 일 실시예에서 , 본 발명에 따른 공결정 A를 슬러리 법을 통해서 합성하는 경우, 니클로사미드 무수물 및 나파모스타트 메실산염 각각의 함량, 알칼리화제의 종류 및 함량, 제 1 용매 (용매 (2)) 및 제 2 용매 (용매 (1))의 종류 및 함량은 하기 표 1에 따라 이용될 수 있다.

일 실시예에서, 본 발명에 따른 공결정 B 를 용매 냉각법을 통해서 합성하는 경우, 니클로사미드 무수물 및 나파모스타트 메실산염 각각의 함량, 알칼리화제의 종류 및 함량, 제 1 용매 (용매 (2)) 및 제 2 용매 (용매 (1))의 종류 및 함량은 하기 표 2에 따라 이용될 수 있다.

이하, 표 1의 실시예 1.1와 표 2의 실시예 2.1 및 실시예 2.3에 따라 공결정의 제조 방법 및 이를 통해 얻은 공결정의 분석 실험과 그 결과에 대해서 보다 구체적으로 설명한다. 실시예 1.1 - 공결정 A (나파모스타트와 니클로사마이드의 몰 비 1)의
제조
나파모스타트 메실산염 86,332.8 mg을 상온에서 증류수 1,950 mL에 500 rpm으로 교반 하며 용해시켰다. 또한, 수산화나트륨 12,799.04 mg을 상온에서 증류수 50 mL에 500 rpm으로교반 하며 용해시켰다. 얻어진 수산화나트륨 용액을 나파모스타트 메실산염 용액에 첨가하여 혼합하고 , 500 rpm으로 교반 하였다. 혼합된 용액이 충분히 불투명해졌을 때, 여기에 에탄올 8므를 첨가하였다. 이어서 , 인산염 16,679.8 uL를 첨가하였고, 이어서 니클로사마이드 무수물 52,339.04 mg을 첨가하였다. 3시간 동안 4,000 rpm으로 강하게 교반한 후 진공 펌프와 종이 필터를 사용하여 여과하여 용매를 제거하였다. 용매 제거 후 진공 건조기를 사용하여 감압 조건에서 하룻동안 건조하여 건조 분말 형태의 공결정 원료(수율 94.94%, 함량 75.47%)를 얻었다. 실시예 2.1 - 공결정 B (나파모스타트와 니클로사마이드의 몰 비 =1:4)의 제조 나파모스타트 메실산염 40,468.5 mg을 상온에서 증류수 l,000mL에 500 rpm으로 교반 하며 용해시켰다. 또한, 탄산수소나트륨 12,601 mg을 상온에서 증류수 500 mL에 500 rpm으로 교반 하며 용해시켰다. 얻어진 탄산수소나트륨 용액을 나파모스타트 메실산염 용액에 첨가하여 혼합하고 500 rpm 으로 교반하였다. 혼합된 용액이 불투명해졌을 때 , 에탄올 6,500 mL를 첨가하였다. 에탄올 첨가후, 니클로사마이드 무수물 98,135.7 mg을 첨가하였고, 3시간 동안 4,000 rpm으로 강하게 교반 하였고, 진공 펌프와 종이 필터를 사용하여 여과하여 용매를 제거하였다. 용매 제거 후 진공 건조기를 사용하여 감압
조건에서 하룻동안 건조하여 건조 분말 형태의 공결정 원료(수율 88.27%, 함량 100.4%)를 얻었다. 실시예 2.3 - 공결정 B(나파모스타트와 니클로사마이드의 몰 비 :4)의
제조 나파모스타트 메실산염 40,468.5mg 을 상온에서 증류수 l,000mL 에 500 rpm으로 교반 하며 용해시켰다. 깨끗하게 녹은 나파모스타트 메실산염 용액을 진공 펌프와 0.45 iim pore size의 나일론 필터를 이용하여 여과하였다. 또한, 탄산수소나트륨 12,601 mg을 상온에서 증류수 500mL에 500 rpm으로 교반 하며 용해시켰다. 깨끗하게 녹은 탄산수소나트륨 용액을 진공 펌프와 0.45 iim pore size 의 나일론 필터를 이용하여 여과하였다. 여과된 탄산수소나트륨 용액을 여과된 나파모스타트 메실산염 용액과혼합하고 500 rpm으로 10분간 교반하였다. 니클로사미드 무수물 98,135.7 mg 을 60 °C에서 아세톤 6,500mL 에 500 rpm으로 1시간 동안교반 하며 용해시켰다. 깨끗하게 녹은 니클로사미드 무수물 용액을 진공 펌프와 0.45 ym pore size의 나일론 필터를 이용하여 여과하였다. 여과된 니클로사미드 무수물 용액을 나파모스타트 메실산염 /탄산수소나트륨 혼합물에 첨가하였고, 이어서 4,000 rpm 의 속도로 상온에서 1 시간 동안 교반하였다. 교반 후 진공 펌프와 종이 필터를 사용하여 여과하여 용매를 제거하였고, 용매 제거 후 진공 건조기를 사용하여 감압 조건에서 하룻동안 건조하여 건조분말 형태의 공결정 원료를 얻었다. 실험 1 - PXRD분석 및 결과
분말 오선 회절 (PXRD) 분석 장치로 D8 ADVANCE with Davinci (상품명, Bruker AXS Inc, GmbH, 독일)를 사용하여 하기 조건에서 본 발명의 실시예 1.1 및 실시예 2.1에 따른공결정 샘플들과 비교 샘플들 각각에 대해서 측정하였다. 검출기 : 고속 LynxEye 검출기 , 관구: Cu, 관 전류: 40mA, 관 전압: 40 kV, 샘플링 폭: 0.020 ° , 주사 속도: 0.1 sec/step, 파장: 1.54056 A, 측정 회절각 범위 (20): 2.5 에서 40 ° 본 발명의 공결정 샘플들은 각각 실시예 1.1 및 실시예 2.1에 따라 얻은 건조 분말 형태의 공결정이고, 비교샘플들은 공결정을 형성하기 위해 이용한 원료물질인 니클로사마이드 및 나파모스타트 메실산염이다. 본 발명의 실시예 1.1 및 실시예 2.1에 따른 공결정 샘플들 및 비교샘플들의 PXRD를 분석된 오선 회절 패턴의 회절각 (20)은 하기 표 3 및 표 4에 나타낸다. 표 3]

표 3 및 표 4를 도 1, 도 2, 도 7 및 도 8과 함께 참조하면, 공결정 형성자들의 공결정을 형성하는 과정은 비화학량론적인 수화물을 가짐으로써, 먼 간격에 어긋남이 생기며, 회절각 (20)이 시프트하는 현상이 나타났음을 확인할 수 있다. 즉, 도 1 및 도 2의 기존의 니클로사마이드 및 나파모스타트 각각과 비교하였을 때, 도 7 및 도 8 의 결과를 통해서 본 발명의 공결정들은 원료물질과는 다른 회절 패턴을 나타내는새로운 결정형임을 확인할수 있다. 실험 2 - DSC분석 및 결과 온도 시차주사열량법에 따른 분석을 위해, DSC 분석 장치로서 DSC Q2000 System (상품명 , TA Instrument , 미국)을 사용하여 , 0°C 에서부터 녹는점까지 10 °C/min의 승온 속도로 온도를 높이며 측정하였다. 측정 시 , N2 가스를 50 mL/min 의 속도로 공급하였고, 알루미늄 샘플 팬에서 즉정하였다. Universal Analysis 2000 software (상품명, TA instruments , 미국)를 사용하여 자료를 분석하였다. DSC를 통해 얻은 융해열 값은 하기 표 5에 나타낸다.

도 3, 도 4, 도 9 및 도 10에서 , x축은 온도 (단위 : °C)를 나타내고, y축은 열 흐름 (heat flow, 단위: W/g)을 나타낸다. 표 5를 도 9 및 도 10과 함께 참조하면, 승온 속도가 10 °C /min인 경우, 공결정 A에서는 92.96 내지 225.66 QC에서 시차주사열량 (DSC) 흡열 피크를 나타나고, 공결정 B에서는 116.49 내지 210.19으仁에서 시차주사열량 (DSC) 흡열 피크를 나타내는 것을 확인할 수 있다. 즉, 도 3 및 도 4 의 기존의 니클로사마이드 및 나파모스타트 각각과 비교하였을 때, 도 9 및 도 10 의 결과를 통해서 본 발명의 공결정들은 원료물질과는 다른새로운 열역학적 특성을 나타내는 결정임을 확인할수 있다. 실험 3 - TGA분석 및 결과
TGA Q50 System (상품명 , TA Instrument , 미국)을사용하여 , 약 2 - 5mg의 샘플을 백금 샘플 팬에 넣어, 실온에서부터 300 °C (10 °C /min.)까지 무게 변화를 측정하였다. 측정 시 , balance에는 40 mL/min. 그리고, 샘플에는 60 mL/min 의 고순도 此 가스를 공급하였다. Universal Analysis 2000 software (상품명 , TA instruments, 미국)를 사용하여 자료를 분석하였다. TGA를 통해 얻은 열중량손실값은 하기 표 6에 나타낸다.

도 5 , 도 6 , 도 11 및 도 12 에서 , x 축은 온도(단위 : °C)를 나타내고 , y죽은 중량(단위 : %)을 나타낸다 . 표 6 을 도 5 , 도 6 , 도 11 및 도 12 와 함께 참조하면 , 본 발명의 공결정 A는 약 4%의 용매를 결정 내 포함하여 공결정 B 는 약 2%의 용매를 결정 내 포함함을 알 수 있다. 실험 4 - 용해도
DMSO와 polysorbate 80 (Tween 80)을 정제수에 각 0.5% 농도가 되도록 용해시켰다. 이 용액을 바이알에 10 mL 가량 담고 니클로사미드와 나파모스타트의 공결정을 10 mg씩 넣은 뒤 10 초간 초음파 처리하여 균질하게 퍼지도록 하였다. 이후 24 시간동안 실온에서 교반한 뒤 용해된 액만을 얻기 위해 검체를 여과하였다. 여과된 검체를 메탄올로 2 배 희석하여 검 액으로 제조하였다. 용해도 측정은 HPLC를 통하여 확인되었다. 측정은 주입량 20 ul 일 때 332 nm 에서 0.7 ml/min 으로 30 분간 진행되었으며 이동상으로는 암모늄수용액과 메탄올을 혼합한 액을 사용하였다. 분석 조건은 하기 표 7 과 같고 , 그 결과를 하기 표 8 에 나타낸다.

상기 표 8 을 참조하면, 니클로사마이드의 용해도는 13.1 mg/mL 이고, 공결정 A의 용해도는 14.6 mg/mL이며 , 공결정 으의 용해도는 19.7 mg/mL으로 측정되었다. 이 결과를 통해 , 공결정화 하지 않은 니클로사마이드의 용해도 대비 본 발명에 따른 공결정 A 및 공결정 B가 각각 111.5 % 및 150.4 % 상승됨을 알 수 있다. 상기 실험 1 내지 실험 4의 결과인 도 3, 도 4, 도 9 및 도 10에 나타낸 DSC 곡선과, 도 1, 도 2, 도 7 및 도 8에 나타난 PXRD 프로파일을 참조하면, 본 발명에 따른 공결정은 니클로사마이드와 나파모스타트의 단순한 혼합물이 아니라 결합된 몰 비율에 따른 공결정 A와 공결정 B가 얻어진 것으로 확인된다. 또한, 이들 본 발명에 따른 공결정은 Vehicle 로 사용하기 위한 충분한 용해도를 확보하였다. 실험 5 - 다양한 암세포주에서 공결정의 항암효능 검증을 위한 세포 증식 억제를 평가 CCK- 8을 이용한 cell viability측정
췌장암, 유방암, 비소세포폐암 세포주에서 공결정에 의한 증식 억제 평가를 위한 항암 in vitro 약효 시험의 조건은 하기 표 9와 같다. 이하, 공결정 A는 실시예 1.1에 따라 얻어진 공결정이고, 공결정 B는 실시예 2.1에 따라 얻어진 공결정이다. 췌장암세포주 2종 (PANC- 1, MIAPACA- 2)과 유방암 세포주 2종 (MCF- 7, MDA- MB- 231)과 비소세포폐암 세포주 2종 (A- 549, H-1299), 간암 세포주 2종 (Hep- 3B, Huh-7) 총 8종의 세포주를 이용하여 96 -조직배양플레이트에 웰당 2〜 4 x 1어개의 세포를 접종하였다. 24시간세포배양기에 배양한후, 공결정 약물을 0.3, 3, 30, 300, 3000, 30000 ng/mL 총 6 개 농도로 세포에 처리하였다. 공결정 외에 단일제제인 Niclosamide 와 Nafamostat는 공결정에서 각 성분 비율에 해당하는 양으로 설정하여 약물 처리하였다. 약물 처리 48시간 뒤에 CCK-8 assay kit를 이용하여 1〜 4시간 시약을 반응시켜 색의 변화를 관찰하고 마이크로리더 장비로 450nm파장에서 흡광도를 측정하여 세포 생존능을 측정하였다.

다양한 종류의 암세포에 대한 공결정의 항암 효능을 검증하기 위해 췌장암, 유방암, 비소세포폐암, 간암 세포에서 세포의 생존력 (Cell viability)를 측정하여 IC50를 도출하였다. 그 결과를 도 13 및 하기 표 10에 나타낸다.

도 13 내지 도 16 및 표 10을 참조하면 , 본 발명에 따른 공결정 A의 IC50는 췌장암 세포주인 PANC- 1세포에서 355 nM, MIAPACA-2 세포에서 287 nM, 유방암 세포주인 MCF-7 세포에서 589 nM, MDA-MB-231 세포에서 38 nM, 비소세포폐암 세포주인 A- 549세포에서 720 nM, H- 1299세포에서 254 nM, 간암 세포주인 Hep-3B 세포에서 125 nM, Hu7- 7 세포에서 672 nM로 확인되었다. 공결정 으의 IC50는 PANC- 1세포에서 794 nM, MIAPACA-2 세포에서 190 nM, MCF- 7세포에서 96 nM, MDA-MB-231 세포에서 33 nM, A- 549세포에서 44 nM, H- 1299 세포에서 1 nM, Hep-3B 세포에서 82 nM, Hu7-7 세포에서 272 nM 로 확인되었다.
이를 통해 공결정 2종을 비교하였을 때에 , 공결정 A보다 공결정 으에 의한 항암 효과가 더 크게 나타난 것을 확인하였다. 실험 6 - 유형별 유방암 세포주에서 공결정의 항암효능 검증을 위한 세포 증식 억제를평/} Cel ITiter— Gio (Promega G9632) 3D cell viability측정 유형별 유방암 세포주에서 공결정에 의한 증식억제 평가를 위한 항암 in vitro 약효평가 시험의 조건은 하기 표 11과 같다. 총 10종의 유방암 세포주 (T47D, MCF7, MCF7/ADR, ZR-75-1 , BT474, BT20, HCC70, BT549, HCC38, MDA-MB-231, Hs578T)를 대상으로 공결정 약물에 대한 검증을 하였다. 각 약물을 세포주별로 10배수씩 농도 6개 범위로 설정하여 Cel ITiter gio를 이용하여 Luminescent Cell viabi 1 ity assay를 수행하였다. 약물 처리 용량은 ng/ml 단위로 질량을 측정하여 처리하였고, 각 공결정 비율에 해당하는 용량으로 단일제제의 용량을 설정하였다. 각 약물의 조건별 발광도를 이용하여 IC50값을 산출하여 단위를 몰농도로 환산했고 계산시 필요한 분자량의 값은 공결정의 경우 단일제제 함량 비율에 따른분자량의 합으로설정하였다.

유형별 유방암세포주에 대한 공결정의 항암 효능을 검증하기 위해 총 10 종의 유방암 세포에서 용량-반응 곡선 시험을 진행하였고, 세포의 생존력 (Cell viability)를 측정하여 IC50를 도출하였다. 그 결과를 도 17, 및 하기 표 12에 나타낸다. 표 12]
도 17 및 표 12를 참조하면 , 약물반응은 각 세포마다 다르게 나타났으나 모든 세포주에서 공결정 으가 공결정 A 보다 낮은 IC50값이 확인되었다. 이를 통해 공결정 으가 유방암 세포주에서 항암 효과가 더 높음을 알 수 있었다.
실험 7 - 췌장암 환자 세포를 이용하여 3D 조건에서 organoid 형태로 세포를 증식시킨 뒤 공결정에 의한 증식 억제를 평가 (CellTiter— Gio (Promega G9632) 3D cell viability측정) 췌장암 환자 세포를 이용하여 공결정에 의한 증식 억제를 평가하기 위한 항암 in vitro 약효 시험을 위한조건은 하기 표 13과 같고, 시험 방법의 개요는 도 18에 나타낸다. 췌장암 환자의 췌장암 세포주를 이용하여 96 -조직배양 플레이트에 웰당 2xl03개의 환자 췌장암 세포가 들어있는 배지를 Matrigel과 1:1비율로 섞은 혼합배지로 접종하였다. 7일간세포배양기에 배양한후, 공결정 약물을 0, 2, 5, 10, 50 uM로 세포에 처리하였다. 화합물 처리 24시간, 48시간 뒤에 현미경을 이용하여 배양중인 세포를 촬영하였다. 48시간 현미경 촬영 후 공결정에 의한 세포 증식 억제를 평가하기 위해 CellTiter-Glo(Promega G9632)를 이용하여 3D 세포 생존능을 측정하였다. 표 13]
췌장암에 대한 공결정의 항암 효능을 검증하기 위해 췌장암 organoid model에서 세포의 생존력 (Cell viability)를 측정하였다. 그 결과를 도 19 및 도 20에 나타낸다. 도 19 및 도 20을 참조하면, 본 발명에 따른공결정 A를 2 uM처리시 cell viability 68.0%, 공결정 B를 2 yM 처리시 cell viability 36.7%로 나타났고, 이를 통해 공결정 A보다는 상대적으로 공결정 으에 의한 항암 효과가 더 크게 나타난 것을 확인할수 있다.
10 yM에서 공결정 A와 공결정 B에 의한 세포 생존율은 10% 이하인 것을 확인할 수 있다. 본 발명의 공결정은 젬시타빈 대비 암세포 생존 억제능이 우수함을 확인하였다. 실험 8 - SARS- CoV- 2에 대한공결정의 항바이러스효능 검증을위한 SARS- CoV- 2세포감염모델에서의 용량-반응곡선 실험 진행
SARS-CoV-2 세포 감염모델을 이용해 공결정의 항바이러스 효능 검증을 위한 실험 조건은 하기 표 14와 같고, 시험 방법의 개요는 도 21에 나타낸다.
SARS-CoV-2 에 대한 공결정 물질의 항바이러스 효능을 검증하고자 384 - 조직배양 플레이트에 웰당 1.2xl04개의 Vero 세포를 접종하였다 . 24 시간 후 , DMS0에 2배 연속 희석하여 10 포인트로 준비된 공결정의 최고농도를 50 uM로 하여 세포에 처리하였다. 공결정 처리 약 1시간후, BSL3 시설에서 세포에 SARS- CoV-2 (0.125 M0I)를 감염시키고, 37°C에서 24시간 동안 배양하였다. 이후 4% paraforma 1 dehyde (PFA)로 세포를 고정한 뒤, permeabi 1 izat ion 하였다. 그 후 ant i -SARS- CoV- 2 nucleocapsid (N) 1차 항체를 처리하고, Alexa Fluor 488 -
conjugated goat ant i -rabbi t IgG 2차 항체와 Hoechst 33342 를 처리하여 세포를 염색하였다. 감염된 세포의 형광 이미지는 대용량 이미지 분석 기기인 Operetta(Perkin Elmer )를 이용하여 획득했다.
SARS_CoV- 2 에 대한 공결정의 항바이러스 효능을 검증하기 위해 SARS-CoV-2 세포 감염 모델에서 용량-반응 곡선 시험을 진행하였고 , 그 결과를 도 22 , 도 23 및 하기 표 15 에 나타낸다.

도 22 및 도 23 과 표 15 를 참조하면 , 공결정 A 의 IC50는 0.17 yM 로 나타났고 공결정 B 의 IC50는 0.17 uM 이었으며 공결정 A와 B 모두 0.4
농도에서 바이러스 감염 억제 효과가 100%에 가깝게 나타났다. 도 24 를 참조하면 , 본 발명에 따른 공결정 A와 B는 적은 농도로 바이러스 감염 억제 효과가 뚜렷하게 관찰되었고 기존 치료제로 사용 중인 렘데시비르보다 IC5o 값이 현저히 낮아, 매우 우수한 바이러스 감염 억제 효과가 있는 것을 확인하였다. 상기에서는 본 발명의 바람직한 실시 예를 참조하여 설명하였지만, 해당 기술 분야의 숙련된 당업자는 하기의 특허 청구 범위에 기재된 본 발명의 사상 및 영 역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경시킬 수 있음을 이해할 수 있을 것이다.
