Abstract
본 발명은 글루타민 또는 이의 염을 유효성분으로 함유한 뇌손상 및 경도 인지장애의 예방, 개선 또는 치료용 조성물에 관한 것으로, 본 발명의 유효성분인 글루타민은 스트레스에 의해 감소한 기억력 및 인지기능을 회복시킬 뿐만 아니라, 신경세포 손상을 억제하며. 뇌조직에서 ROS/RNS의 함량을 감소시키고, 이와 관련된 단백질의 발현량도 감소시키며, 시냅스 가소성(Synaptic plasticity)을 증진시킬 수 있으며, 알츠하이머병 동물에게 글루타민을 식이한 경우, 인지능력이 증가하고, Aβ42의 축적이 억제되는 것을 확인한 것이다.
Description
본 발명은 글루타민을 유효성분으로 함유한 뇌손상 및 경도 인지장애의 예방, 개선 또는 치료용 조성물에 관한 것이다.
경도 인지장애는 기억력을 비롯한 인지 기능이 시간의 경과에 따라 점진적으로 저하되나, 일상생활 수행 능력은 보존되어 있는 상태를 말한다. 정상 노화와 치매의 중간 단계로, 알츠하이머병으로 이행할 위험이 크다. 노화 과정에 의해 자연스럽게 저하되는 객관적인 인지 기능의 범위를 벗어난 것으로, 이는 치매를 비롯한 주요 신경 인지 장애의 임상적인 전단계에 해당한다. 기억력 저하가 가장 주된 문제로 나타나나, 특정 자극에 대해 감정의 변화가 없거나 기분의 변화가 둔감하게 나타나는 경우도 경도 인지 장애로 인한 저하된 인지 기능을 시사한다. 그러나 주요 신경 인지 장애와는 달리 일상적인 생활이 비교적 정상적으로 가능한 단계이다.
기억력 손상을 중심으로 하는 인지 기능 저하에 대한 불편감을 주된 특징으로 하며, 이에 대해 객관적인 증거가 존재하나 일상생활의 기능은 비교적 경미한 수준의 손상을 입는 것이 특징이다. 이와 같은 진단 기준에 해당할 경우, 경도 인지 장애의 하위 유형 중 어느 것에 해당하는지를 파악해야 한다.
경도 인지 장애를 진단하기 위한 우선적인 도구로는 신경심리검사가 있으나, 하위 유형을 보다 구체적으로 살펴보기 위해서는 자기공명영상촬영(뇌 MRI) 또는 양전자방출단층촬영(PET)을 이용한 영상 검사가 필요하다. 검사를 통해 진단이 확증되면, 콜린에스터레이즈 억제제(cholinesterase inhibitor), 항산화제 및 NMDA 수용체 길항제 등을 이용한 약물요법을 시도한다. 경도 인지 장애가 알츠하이머병 및 치매로 이행할 가능성은 10~15% 정도이다.
한편, 글루타민은 몸 전체에 풍부하게 존재하며 많은 대사 과정에 관여한다. 글루탐산과 암모니아로부터 합성되고 인체 내 질소의 주된 운반체이며 많은 세포에서 중요한 에너지원이다. 글루타민은 경구투여를 통해 뇌의 striatal 조직에서 GABA를 증가시킬 수 있다는 것이 알려져 있고(FASEB J. 21, 1227-1232(2007)), 한국공개특허 제2011-0117591호에 글루타민을 포함하는 우울증 예방 또는 치료용 조성물이 개시되어 있으나, 아직까지 본 발명의 글루타민을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방, 개선 또는 치료용 조성물에 대해 개시된 바 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명은 글루타민을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방, 개선 또는 치료용 조성물을 제공하고, ORT(Objective Recognition Test) 및 OLT(Objective Location Test)에서 정상군 대비 스트레스에 의한 뇌손상과 경도 인지장애를 유도한 스트레스군(STR)의 물체 기억력 및 장소 기억력이 저하되었으나, 본 발명의 유효성분인 글루타민 식이군(STR+Gln)은 정상군에 버금가는 인지기능을 유지하고 있다는 것과, STR군에서는 신경세포 손상이 발생하였으나, 이에 대비 STR+Gln군에서는 신경세포 손상이 나타나지 않았으며, 뇌조직에서 ROS/RNS 함량 및 이와 관련된 단백질의 함량을 감소시키고, 시냅스 가소성(Synaptic plasticity)을 증진시킬 수 있으며, 알츠하이머병 동물에서 글루타민의 식이에 의해 인지능력이 증가했을 뿐만 아니라, Aβ42의 축적을 억제할 수 있다는 것을 확인함으로써, 본 발명을 완성하였다.
상기 목적을 달성하기 위하여, 본 발명은 글루타민 또는 식품학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
또한, 본 발명은 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 의약외품을 제공한다.
본 발명은 글루타민을 유효성분으로 함유한 뇌손상 및 경도 인지장애 예방, 개선 또는 치료용 조성물에 관한 것으로, 스트레스를 받은 대조군에서는 기억력 및 인지기능이 감소하였으나, 글루타민을 보충하여 식이한 실험군은 정상군에 유사한 인지기능으로 회복될 뿐만 아니라, 신경세포 손상을 억제하며, 뇌조직에서 ROS/RNS의 함량을 감소시키고, 이와 관련된 단백질의 발현량도 감소시키는 것을 확인하였으며, 시냅스 가소성(Synaptic plasticity)을 증진시킬 수 있다는 것을 확인하였다. 또한, 알츠하이머병 동물에서 글루타민의 식이에 의해 인지능력이 증가했을 뿐만 아니라, Aβ42의 축적이 억제되는 것을 확인하였다. 따라서 본 발명의 유효성분인 글루타민은 만성 뇌손상 및 경도 인지장애에 대한 효과가 있는 것이다.

상기 경도 인지장애는 기억, 인지 및 학습능력 중에서 선택된 하나 이상의 기능의 저하인 것이 바람직하지만 이에 한정하지 않는다.
본 발명의 건강기능식품 조성물은 환, 정제(tablet), 캡슐(capsule), 산제, 분말, 과립, 캔디, 시럽 및 음료 중에서 선택된 어느 하나로 제조하거나, 식품의 성분으로 첨가하여 제조될 수 있으며, 통상적인 방법에 따라 적절하게 제조될 수 있다. 본 발명의 유효성분을 첨가할 수 있는 식품의 일례로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합제 중에서 선택된 어느 하나의 형태일 수 있으며, 통상적인 의미에서의 건강기능식품을 모두 포함한다. 상기 건강기능식품은 여러 가지 영양제, 비타민, 광물(전해질), 합성 및 천연 풍미제, 착색제 및 증진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 천연 과일 주스 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 본 발명의 건강기능식품 조성물은 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상기 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토스, 슈크로스와 같은 디사카라이드, 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다.
또한, 본 발명은 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 치료용 약학 조성물에 관한 것이다.
상기 유효성분 이외에 추가로 약제학적으로 허용가능한 담체, 부형제 또는 희석제를 더 포함할 수 있다. 본 발명의 약학 조성물은 경구 또는 비경구로 투여될 수 있으며, 비경구 투여 시 피부 외용 또는 복강 내, 직장, 정맥, 근육 또는 피하주사 방식을 선택하는 것이 바람직하지만 이에 한정하는 것은 아니다.
본 발명의 약학 조성물은 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조제제, 좌제가 포함된다. 비수성 용제 및 현탁 용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈 61, 카카오지, 라우린지, 글리세롤, 젤라틴 등이 사용될 수 있다.
본 발명에 따른 조성물은 약제학적으로 유효한 양으로 투여한다. 본 발명에 있어서, "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 조성물의 투여량은 환자의 체중, 연령, 성별, 건강상태, 식이, 투여시간, 투여방법, 배설률 및 질환의 중증도에 따라 그 범위가 다양하게 사용할 수 있다.
또한, 본 발명은 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 의약외품에 관한 것이다.
본 발명의 유효성분인 글루타민 또는 약학적으로 허용가능한 이의 염을 의약외품 첨가물로 사용할 경우, 상기 유효성분을 그대로 첨가하거나 다른 의약외품 또는 의약외품 성분과 함께 사용할 수 있고, 통상적인 방법에 따라 적절하게 사용할 수 있다. 유효성분의 혼합량은 사용 목적에 따라 적합하게 결정될 수 있다.
이하, 실시예를 이용하여 본 발명을 더욱 상세하게 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로 본 발명의 범위가 이들에 의해 제한되지 않는다는 것은 당해 기술분야에서 통상의 지식을 가진 자에게 있어 자명한 것이다.
실시예 1. 만성 부동자세 스트레스(chronic immobilization stress, CIS) 및 글루타민 보충에 따른 체중, 사료 섭취량 및 코르티코스테론의 함량의 변화 확인
[동물 모델]
본 발명에서는 7주령 C57BL/6 수컷 마우스(한국 코아텍)를 표준 조건(온도 22~24℃, 습도 50~70%, 오후 6시에 점등되는 12시간 명암주기)으로 사육하였으며, 이때 식이요법과 물을 자유롭게 섭취할 수 있도록 하였다. 본 발명에서 사용한 동물은 국립보건원(NIH, Bechesda, MD, USA) 지침에 따르는 경상대학교 동물보호 및 사용 위원회(GNU IACUC)로부터 승인받은 프로토콜(GNU-161128-M0068)에 따라 실시하였다.
대조군(CTL)은 스트레스를 가하지 않으면서 일반식이(영향 균형이 맞는 일반적인 식이)를 실시한 군이고, CTL+Gln군은 스트레스를 가하지 않으면서 사료 1kg당 150mg의 글루타민을 보충한 일반식이군이며, STR은 스트레스를 가하면서 일반식이를 실시한 군이고, STR+Gln군은 스트레스를 가하면서 사료 1kg당 150mg의 글루타민을 보충한 일반식이 군이다. 스트레스군(STR)은 매일 2시간(오후 2~4시)동안 개별적으로 구속기에 강제동원하여 만성적 부동자세 스트레스를 15일 동안 주었다.
본 발명에서 실시한 동물 실험은 3개의 코호트를 구성하여, 제1 코호트는 군당 7마리씩 총 28마리로 구성하였고, 제2 코호트는 군당 12~13마리씩 총 51마리로 구성하였으며, 마지막 제3 코호트는 군당 8마리씩 총 32마리로 구성하였다.
상기 제1 코호트를 이용하여, 스트레스를 가하는 15일 동안 2일 간격으로, 각 동물모델군(각 군당 7마리씩 총 28마리)의 체중 변화와 사료 섭취량을 측정하였다.
그 결과 도 1에 개시한 바와 같이 체중은 스트레스를 가하지 않은 CTL 및 CTL+Gln은 시간 경과에 따라 점진적으로 증가하였으나, 스트레스를 가한 STR 및 STR+Gln은 시간 경과에 따라 체중이 증가하지 않고 오히려 감소하는 경향을 나타냈다. 이때 글루타민의 보충은 체중의 변화에 영향을 주지 않아, 글루타민 보충 여부에 따른 차이는 거의 없는 것으로 나타났다. 하지만, 사료 섭취량은 거의 차이가 없어 CTL, CTL+Gln, STR 및 STR+Gln은 모두 유사한 수준의 사료를 섭취하였다.
한편, 혈장 내 코르티코스테론의 함량 변화를 확인한 결과 도 1D에 개시한 바와 같이 대조군(CTL) 대비 스트레스군(STR)의 코르티코스테론의 함량이 증가하였고, 이에 대비하여 본 발명의 글루타민을 보충한 군(STR+Gln)의 코르티코스테론의 함량이 통계적으로 유의미하게 감소한 것을 확인하였다.
실시예 2. 만성 부동자세 스트레스(CIS) 및 글루타민 보충에 따른 ORT(object recognition test) 및 OLT(object location memory test) 행동분석
상기 실시예 1의 코호트를 이용하여 행동분석을 실시하였고, 스트레스를 가한 15일 후, 즉시 행동분석을 실시하였다.
ORT는 습관화(habituation), 친숙화(familiarization) 및 테스트(test)의 세션으로 구분하였다.
습관화(habituation)하는 2일 동안 하루에 10분씩 빈 열린 공간에 두었고, 습관화(habituation) 후, 열린 공간에 두 개의 동일한 물체를 배치하여 친숙화(familiarization)하였다. 각 실험군의 쥐들은 자유롭게 10분 동안 물체를 탐색할 수 있도록 하였다. 다음날, 상기 물체 중 하나를 새로운 물체로 대체하고, 테스트(Test) 세션을 반복 실시하였다. 각 세션은 초기 5분을 분석에 사용하였다.
이후 ORT에 대한 차별지수(DI)를 하기 식 (1)에 따라 계산하였다.

DI가 0인 경우, 두 물체에 대한 선호도가 동일하다는 것을 의미하며, DI가 (-)인 경우는 새로운 물체를 혐오한다는 것으로 그 값이 커질수록 혐오 정도가 크다는 것을 의미한다.
ORT 이후, 동일한 열린 공간에서 OLT를 실시하였다. 상기 ORT에서의 새로운 물체를 다른 위치로 이동시킨 후, ORT와 동일한 방법으로 테스트하였고, OLT에 대한 변별도 지수(DI)를 하기 식 (2)에 따라 계산하였다.

그 결과, 도 2에 개시한 바와 같이 CTL 또는 CTL+Gln 대비 스트레스군(STR)의 ORT 및 OLT의 DI가 통계적으로 유의미하게 감소하였으나, 본 발명의 글루타민을 보충식이한 군(SRT+Gln)의 DI가 증가하였다. 한편, ORT 및 OLT에서의 이동 거리는 실험군 간에 차이가 거의 없는 것으로 나타나, 운동성이 감소한 것이 아니라, 인지정도의 차이라는 것을 알 수 있었다.
실시예 3. 만성 부동자세 스트레스 및 글루타민 보충에 따른 해마에서 형태학적 변화 확인
해마 부위의 뇌조직을 염색하여 만성 부동자세 스트레스 및 글루타민 보충에 따른 형태학적 변화를 확인하고자 하였다.
0.2% 아세트산에 용해시킨 0.1% 크레실 바이올렛(Cresyl violet)에 5분 동안 마운팅하고 에어드라이한 섹션(뇌조직 절편)을 염색하였다. 염색한 섹션은 세척하고 30초 동안 80%(v/v) 에탄올에 침지하고, 이어 30초 동안 90%(v/v) 에탄올에 침지하고, 이어 30초 동안 95%(v/v) 에탄올에 두 번 침지하고, 이후 30초 동안 100%(v/v) 에탄올에 두 번 침지하여 탈수하였다. 이후 1분 동안 두 번 자일렌 처리하여 투명화(clearing)하고, 자일렌 마운팅 용액(자일렌:Permount=1:1)으로 처리하고, 후드에서 건조하였다.
그 결과, 도 3에 개시한 바와 같이 CA1-SP(stratum pyramidale CA1)의 폭이 스트레스에 의해 감소한 것을 확인할 수 있었고, 본 발명의 글루타민을 보충하는 경우 CA1-SP의 폭이 회복되는 것을 확인할 수 있었다. 이와 같은 결과로부터 스트레스에 의해 유도되는 뇌손상을 글루타민 보충으로 개선/치료할 수 있는 것으로 판단하였다.
실시예 4. 만성 부동자세 스트레스 및 글루타민 보충에 따른 혈장, PFC 및 해마에서 ROS/RNS 함량 변화 확인
제2코호트는 군당 12~13마리로 구성된 51마리의 동물모델은 행동분석하지 않고, 마지막 구속 스트레스를 끝내고 바로 희생시켜, 혈장, PFC 및 해마에서의 ROS/RNS를 확인하였다. ROS/RNS는 ROS/RNS 어세이 키트(STA-347; Cell Biolabs, San Diego, CA, USA)를 이용하여 제조사의 방법에 따라 실시하였다.
PFC 및 해마 조직은 Bullert 블랜더를 이용하여 RIPA(radioimmunoprecipitation assay) 완충용액에 글라스 비드를 첨가하여 조직파쇄하였다. 이후 2분 동안 초음파 처리하고, 4℃에서 10분 동안 12,000×g의 조건으로 원심분리하였다. 이후 Tecan Infinite F200 PRO 마이크로 리더를 이용하여 형광강도를 측정하기를 3번 반복하였다.
그 결과 도 4에 개시한 바와 같이, 혈장, PFC 및 해마조직 모두에서 스트레스에 의해 활성 산소/질소종(ROS/RNS)의 함량이 증가하였으나, 본 발명의 글루타민을 보충한 군에서는 활성 산소/질소종의 함량이 감소한 것을 확인할 수 있었다.
실시예 5. 만성 부동자세 스트레스 및 글루타민 보충에 따른 단백질 발현량 변화 분석
글루타민 보충에 따른 구체적인 뇌 조직의 세포 위치를 확인하기 위하여, 세포 분포 위치 마커와 함께 이중염색하여 ROS/RNS 함량 변화에 관여하는 단백질(iNOS; nNOS; NOX의 서브유닛 p47phox와 p67phox)을 IHC(immunohistochemistry) 분석하였다.
마지막 스트레스를 끝내고 24시간 후 마우스를 아베르틴으로 마취하고, PBS(pH7.4) 및 4% 파라포름알데히드를 관류하였다. 이후, 뇌를 적출하여 고정하고 40㎛의 두께로 자르고, 항체 nNOS(sc-5302, 200㎍/㎖, Santa Cruz, Dallas, TX, USA, 1:50), iNOS(sc-7271, 200㎍/㎖, Santa Cruz, 1:20), p47phox(sc-17845, 200㎍/㎖, Santa Cruz, 1:20), p67phox(15551-1-AP, 43㎍/150㎕, Proteintech, Rosemont, IL, USA, 1:100), PSD-95(postsynaptic density-95)(ab12093, 1mg/㎖, Abcam, Cambridge, UK, 1:200) 및 시냅토파이신(synaptophysin, ab14692, 0.65mg/㎖, Abcam, 1:200))을 각각 첨가하여 밤새 인큐베이션하였다. 세포 분포 마커로, Iba1(Ionized calcium-binding adapter molecule 1, ab5076, 0.5mg/㎖, Abcam, 1:200), NeuN(neuronal nuclei, MAB377, 30㎍/㎖, Merck Millipore, St. Louis, MO, USA, 1:200), 및 GAD2(glutamate decarboxylase, #3988, Cell Signaling, Danvers, MA, USA, 1:100)를 사용하였다. 뇌조직 절편들은 AlexaFluor 594 및 488-결합된 2차 항체(2mg/㎖, Invitrogen, Carlsbad, CA, USA, 1:1000)들과 함께 인큐베이션 하였다. 이후, 올림푸스 디스크 스피닝 유닛이 장착된 공초점 현미경 및 이미지J 프로그램을 이용하여 분석하였다.
그 결과, 도 5 및 도 6에 개시한 바와 같이 infralimbic cortex의 미아교세포 및 해마 CA1 지역에서 iNOS 및 NOX 서브유닛인 p47phox 및 p67phox의 발현량이 스트레스 군에서 통계적으로 유의미하게 증가하였고, 이와는 대조적으로 글루타민을 보충식이한 군에서 통계적으로 유의미하게 감소하였다.
또한, 시냅스 가소성(synaptic plasticity)을 확인할 수 있는 PSD-95 및 시냅토파이신(synaptophysin)의 co-localization을 확인한 결과, 글루타민은 synaptic puncta를 유의미하게 증진시키는 것을 확인하였다(도 5E 및 도 6F).
실시예 6. 알츠하이머병 모델의 글루타민 식이에 의한 경도 인지장애 예방 효과 확인
(1) 글루타민 식이 알츠하이머병 모델 구축
인지기능에 대한 평가를 위한 동물 모델로서, 7주령 3xTG-AD 암컷 마우스(알츠하이머병 모델) 8마리와 C57BL/6 암컷 마우스(대조군) 10마리를 채택하였으며, 각각 두 그룹으로 분류하고, 각각의 그룹에 정상사료(normal diet, ND)와 글루타민 사료(Gln)를 제공하였다. 글루타민 식이 그룹은 150mg/kg의 글루타민이 포함된 사료를 2개월령부터 실험 종료 시까지(6~7개월령) 섭취하도록 하였으며, 주 1회 체중과 먹이 섭취량을 측정하여 건강 상태를 확인하였다(도 7A).
(2) 인지능력시험
상기 구축한 동물모델인 마우스의 인지능력은 ORT(object recognition test)를 통해 측정하였다. 이틀 동안 박스 적응시킨 후, 3일 차에 두 개의 동일한 물체를 10분 동안 탐색하게 하였다. 4일차에 사물 한 개를 새로운 것으로 바꾸어 5분 동안 탐색하게 하였다(도 7B). 양쪽 물체의 주변 1cm 이내에 접근하여 탐색한 시간을 측정하고, 식 (3)을 이용하여 계산하여 DI(discrimination index) 값을 계산하여 비교하였다. 본 발명에서는 시험을 수행할 때마다 새로운 사물을 사용하였다.

(3) 글루타민 식이에 따른 알츠하이머병 모델의 경도 인지장애 발병 지연 효과 확인
2, 4, 6개월령에 인지기능을 시험하였다. 정상군(NonTg, n=5), 글루타민을 식이한 정상군(NonTg+Gln, n=5), 알츠하이머병 유도군(3xTG, n=4) 및 글루타민을 식이한 알츠하이머병 유도군(3xTG+Gln, n=4)으로 분류하여 분석하였다.
그 결과, 정상군인 NonTg 마우스 군은 6개월령까지 인지기능 저하가 나타나지 않았고, 글루타민 식이에 따른 차이도 없었다. 3xTG-AD 마우스군은 2개월령에 비해 6개월령의 사물인지 능력이 현저히 저하된 반면, 글루타민 식이군의 인지기능은 유의한 저하를 보이지 않았다.
3xTG-AD군은 2개월령에 비해 6개월령에 개체별 ORT의 DI값이 평균 0.4 감소하여, 상기 시험군 중에서 가장 큰 변화를 보였으나, 글루타민 식이 3xTG-AD군은 0.1 정도만 감소하여 NonTg군과 유의한 차이가 나타나지 않았다. 따라서 알츠하이머병 모델에서의 글루타민 식이는 경도 인지장애의 발병을 지연시키는 것으로 판단하였다(도 8).
(4) 글루타민 식이에 따른 아밀로이드 베타 42(Aβ42
)의 축적 지연
6개월령의 인지기능 시험 후, 이산화탄소로 마우스를 안락사시키고, 혈액 내 신경분비 엑소좀(neuronally derived blood exosome, NDE) 단백질 변화를 조사하였다. 혈액 세럼에서 총 엑소좀을 분리하고, 항-CD171 항체와 1시간 반응시킨 후, 스트렙타비딘이 융합된 자성비드와 1시간 동안 반응시켰다. 이후, 0.05M 글리신-HCl(pH3.0)로 용출하고, 1M Tris-HCl(pH8.0)을 첨가하였다. 아밀로이드 베타 42(Aβ42) 마우스 ELISA 키트를 이용하여 분석하였다.
그 결과, 3xTG-AD 암컷의 NDE에서 Aβ42가 증가하였고, 글루타민 식이군에서는 Aβ42의 증가가 억제된 것을 확인하였다. 이와 같은 결과로부터, 글루타민 식이에 의해 체내 Aβ42의 축적이 억제되어 인지기능 저하에 대한 예방 효과를 보이는 것으로 판단하였다(도 9).
(5) 글루타민 식이의 인지기능 개선 효과 확인
인지기능을 개선시키는 효과도 있는지를 확인하고자, 글루타민을 정상 마우스에 경구투여하여 2시간 후에 인지기능이 개선되는지 확인하였다. 인지기능은 단기작업기억능력을 시험하는 Y-maze, 장기기억능력을 시험하는 ORT 및 OLT(object location recognition test)로 시험하였다.
Y-maze: A, B, C로 지정된 120°간격으로 이루어진 3개의 팔로 구성된 Y자형 미로에 마우스를 넣고, 3회 연속으로 서로 다른 팔에 들어간 자발적 변경률(spontaneous alternation)을 식 4에 적용하여 계산하였다(도 10A).

ORT/OLT: 상기한 바와 같이 ORT를 10분 동안 수행하고, 24시간 후 물체 하나의 위치를 이동시켜 5분 동안 탐색하게 하였다(도 10B).
그 결과, Y-maze에서는 자발적 변경률은 차이가 없었으나, 장기기억능력시험인 ORT와 OLT에서는 정상 마우스에 비해 인지기능이 향상된 것으로 나타났다(도 11).
(5) 글루타민 식이에 따른 경도 인지장애 개선 효과 확인
만성 신체 구금 스트레스(chronic immobilization stress, CIS)에 의해 인지장애가 발생한 후에 글루타민 경구투여한 후, 인지장애 치료 효과를 확인하였다.
CIS 2주 수행 후 ORT-OLT로 인지능력을 시험하였다. 이후 1주 동안 글루타민을 경구투여하면서(45mg 글루타민/kg 체중/day) 4일차부터 인지기능 변화를 조사하였다(4~7일차에 ORT-OLT를 실시하였다).
2주 동안 가한 만성 스트레스 군은 ORT와 OLT 모두에서 인지기능 저하가 확인되었으나, 이후 1주일 동안 글루타민을 경구투여한 그룹은 인지기능이 비스트레스 대조군 수준으로 회복되는 것을 확인하였다. 이와 같은 결과로부터 글루타민 식이는 경도 인지장애의 예방 및 치료 효과가 있는 것으로 판단하였다(도 12).

- 제1항에 있어서, 상기 뇌손상 및 경도 인지장애는 스트레스에 의한 것임을 특징으로 하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 건강기능식품 조성물.

- 제1항에 있어서, 상기 경도 인지장애는 기억, 인지 및 학습능력 중에서 선택된 하나 이상의 기능의 저하인 것을 특징으로 하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 건강기능식품 조성물.
- 제1항에 있어서, 상기 조성물은 환, 정제(tablet), 캡슐(capsule), 산제, 분말, 과립, 캔디, 시럽 및 음료 중에서 선택된 어느 하나의 제형으로 제조되는 것을 특징으로 하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 건강기능식품 조성물.
- 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 치료용 약학 조성물.
- 제6항에 있어서, 상기 유효성분 이외에 추가로 약제학적으로 허용가능한 담체, 부형제 또는 희석제를 더 포함하는 것을 특징으로 하는 뇌손상 및 경도 인지장애의 예방 또는 치료용 약학 조성물.
- 글루타민 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 뇌손상 및 경도 인지장애의 예방 또는 개선용 의약외품.
