Description
본 발명은 2'-FL을 함유하는 도파민 감소로 말미암아 발생하는 질환의 개선, 예방 또는 치료용 조성물에 관한 것이다.
도파민은 영국, 런던의 웰컴 연구소에서 조지 바거와 제임스 웬스에 의해 1910년 합성되었다. 모노아민이기 때문에 도파민이라 불리게 되었다. 도파민은 화학식 C8H11NO2로, 카테콜아민 계열의 유기 화합물이고, 다양한 동물의 중추신경계에서 발견되는 호르몬이나 신경전달물질이다. 뇌신경세포들 간에 신호를 전달하기 위해 분비되는 신경전달물질 중 하나로, 뇌의 도파민 수용체 D1, D2, D3, D4, D5 및 이의 변종을 활성화한다.

도파 반응성 근육긴장이상(dopa-responsive dystonia)은 도파민 생성 과정에 관련된 GCH-1 유전자의 돌연변이가 원인이다. 다만, 실제 환자의 FP-CIT 양전자방출단층촬영 결과는 정상 소견을 보인다. 이에 따라, 흑질-선조체의 시냅스 전 말단이 구조적으로 정상임을 확인할 수 있는데, 이를 통해 시냅스 전 신경 변화 외에 시냅스 후(postsynaptic) 신경 퇴행이 이상운동항진증 유발에 중요한 기전이 될 수도 있음을 유추할 수 있다.
실제 도파민 생성 경로에서 중요한 티록신 수산화효소(Tyrosine hydroxylase) 결핍 질환 환자에서도 도파 반응성 근육긴장이상이 빈번하게 관찰되는 것으로 알려져 있다. 이는 기본적으로 도파민 수용체의 민감도가 지나치게 항진되어 발생하는 것으로 해석되고 있다.
세가와병(segawa's syndrome)은 걷기가 불편하거나 비정상적인 걸음걸이와 디스토니아를 수반하는 희귀한 유전질환이다. 디스토니아(Dystonia)는 일반적으로 신체를 비정상적으로, 때로는 고통스럽게, 움직임과 위치(자세)를 강제하는 무의식적인 근육 수축으로 특징지어지는 근육 장애를 일컫는 총칭이다. 세가와병은 구아노신 삼인산 사이클로하이드롤라제 I(GCH-1) 유전자의 돌연변이에 의해 발생한다. GCH-1 유전자 돌연변이는 자가 우성 형질로 유전되거나 뚜렷한 이유 없이 산발적으로 발생하는 자발적 유전자 변화(즉, 새로운 돌연변이)로 발생한다. GCH-1 유전자의 돌연변이는 GTPCH1 효소의 결핍을 초래하여 궁극적으로 도파민 결핍을 초래한다.
다발성경화증(multiple sclerosis)은 뇌, 척수, 그리고 시신경을 포함하는 중추신경계에 발생하는 만성 신경면역계질환이다. 도파민 불균형이 다발성 경화증의 피로에 영향을 준다고 연구를 통해 밝혀졌다. 다발성 경화증 환자에게서 도파민은 말초 혈액 단핵세포(peripheral blood mononuclear cell, PBMC)를 통해 IL-17과 IFN-γ 생산을 억제한다.
파킨슨병(Parkinson's disease)은 노인성 환(age-related disorder)으로 신체 경직, 동작 둔화, 정지해 있을 때의 팔다리 떨림 등 운동장애(movement disorder)를 특징으로 한다. 병세가 진전되면 치매(dementia)로 이어지며 결국 사망에 이르게 된다. 주요 증상들은 중뇌의 흑색질(substantia nigra)에서의 도파민 분비세포의 사멸로 인하여 발생한다. 특히, 이러한 도파민 세포는 뇌염(encephalitis), 반복되는 운동성 뇌진탕(sports-related concussion)과 같은 외상이나 상해에 취약하며, MPTP와 같은 화학물 중독에 의해서도 야기될 수 있다.
한편, 파킨슨병 환자의 80%가 장기능 장애로 변비를 앓고 있는 것으로 알려졌다. 본 발명의 일 실시예에 의하면, 2'-FL이 대조군에 비해 조눌린(zonulin)과 오클루딘(occludin)의 수치를 각각 조절하여 장내 미생물의 환경을 개선함과 동시에 소장에서의 ZO-1 유전자의 발현을 증가시켜 소장 상피세포의 밀착접합을 견고하게 하여, 장기능 개선에 효과가 있음을 확인한 바 있다. 따라서, 본 발명의 약학 조성물은 파킨슨병에 의해 유발되는 장기능 장애를 예방 또는 치료할 수 있다.
주의력결핍 과잉행동장애(attention deficit hyperactivity disorder)는 인지 조절(cognitive control) 장애와 연관된 증상으로, 주의력 조절(attentional control) 문제, 행동 억제 조절, 깜박 잊기 혹은 상세 내용 부주의(작업기억)를 야기한다. 도파민 수용체, 도파민 수송체, ADHD 간에 유전적 연결성이 있는 것으로 알려졌다.
투렛증후군(Tourette syndrome)은 틱 장애로도 일컬으며, 자신의 의사와는 상관없이 단순한 동작이나 소리를 반복하는 증상이다. 투렛증후군은 도파민 1 수용체의 과잉 활동이 원인인 것으로 알려져 있다. 다만, 현재 사용되고 있는 투렛증후군 치료제는 도파민 2 수용체를 표적으로 하는 약으로 환자에 따라 효과가 나타나지 않는 경우가 많은데다 체중 증가와 같은 부작용이 있다.
우울장애(depressive disorder)는 일상생활이 힘들 정도로 우울한 기분이 지속되는 질환을 말한다. 도파민의 분비 이상은 우울증과 관련이 있는 것으로 알려졌다.
조현병(schizophrenia)은 망상, 환청, 와해된 언어, 와해된 행동, 정서적 둔마 등의 증상이 주로 나타나고, 사회적 기능에 장애를 일으킬 수도 있는 질환이다. 많은 정신과의사와 신경과학자들은 조현병이 도파민체계 장애와 관련이 있다고 생각한다. 정신약학자(Psychopharmacologist) 스티븐 스탈(Stephen M. Stahl)은 2018년 리뷰에서, 조현병 등 많은 정신병 사례에서 도파민, 세로토닌(serotonin), 글루타메이트(glutamate)가 서로 다양하게 조합되어 선조체(ventral striatum) 내 도파민 D2 수용체를 과자극(overexcitation)하여 발생하는 것으로 제시하였다.
양극성 장애(bipolar disorder)는 조울증으로도 일컬으며, 우울증과 함께 정신건강의학과에서 기분장애에 속하는 질환이다. 한번 이상의 경조증삽화 또는 조증삽화를 경험하는 경우 양극성 장애(조울증)로 진단하며, 조증삽화를 경험한 경우 1형 양극성 장애, 경조증삽화를 경험한 경우 2형 양극성 장애로 구분한다. 그 중 1형 양극성장애 환자들에서 도파민을 포함하는 신경전달물질이 과도하게 생성되거나 부족한 경우들이 보고되고 있다.
도파민체계는 중독의 일부 측면에 있어 결정적인 역할을 수행한다. 각성제 복용은 수분에서 수시간 동안 지속되는 뇌 내 도파민 수치를 증가시킨다. 결국, 반복적으로 고용량의 각성제를 복용함으로써 발생하는 도파민 수치의 만성적 고양 상태는, 중독을 특징으로 하는 행동 이상(behavioral abnormality)을 담당하는 뇌 내 구조에 변화를 촉발한다. 이렇기 때문에 각성제 중독 치료는 매우 어렵다. 복용을 중단하더라도 금단으로 인한 갈망은 계속 나타나기 때문이다.
도파민은 척수(spinal cord), 중심회색질(periaqueductal gray), 시상(thalamus), 기저핵(basal ganglia), 대상피질(cingulate cortex) 등의 중추신경계(central nervous system)의 복합적 수준에서 통증 과정에 중요한 역할을 한다. 도파민 수치 저하는 파킨슨병에서 자주 발생하는 통증과 관련이 있다. 도파민 신경전달작용(dopaminergic neurotransmission)에서 발생하는 기형은 또한 구강작열감증후군(burning mouth syndrome), 섬유근육통(fibromyalgia), 하지불안증후군(restless legs syndrome) 등의 질환에서도 발생한다.
구역질(nausea)과 구토(vomiting)는 화학수용체자극대(chemoreceptor trigger zone)로 알려진, 뇌간(brainstem)의 숨뇌(medulla oblongata)에 있는 구토중추(area postrema)의 활동에 의해 결정된다. 이 영역은 D2형 도파민 수용체(type D2 dopamine receptor)가 분포되어 있다. 그 결과, D2 수용체를 활성화하는 약물은 구역질 유발 가능성이 높다.
보다 상세하게는, 본 발명은 2'-FL (2'-fucosyllactose)을 유효성분으로 함유하며, 도파민 신경세포의 감소를 억제하며, 운동활성증진효과를 가지는 것을 특징으로 하는 도파민 감소로 말미암은 파킨슨병의 예방 또는 치료용 약학 조성물을 제공한다.
파킨슨병은 도파민 신경세포의 소실로 인해 발생하는 신경계의 만성 진행성 퇴행성 질환으로, 파킨슨 환자들에게서는 서동증(운동 느림), 안정 시 떨림, 근육 강직, 자세 불안정 등의 증상이 발생하는데, 본 발명의 일 실시예에 의하면, 2'-FL은 신경독성물질인 6-OHDA가 유발하는 도파민 신경세포의 감소를 효과적으로 억제함을 확인하였다 (도 2). 또한, 본 발명의 일 실시예에서는 2'-FL의 투여로 인해 뇌의 흑색질에서의 TH-면역반응성이 대조군에 비해 현저히 증가되어 뇌의 흑색질에서 도파민 감소 억제능이 있음을 확인하였다 (도 6).
특히, 본 발명의 일실시예에서는 파킨슨이 유발된 동물모델에서 2'-FL의 투여가 마우스의 움직임을 증가시켜 운동활성증진 효과가 있으며 (도 4), 2-FL의 투여는 6-OHDA 투여 후 메스암페타민(Meth)에 의해 유발되는 로테이션 행동을 전체적으로 완화 및 억제시켰음을 확인하였다 (도 5).
한편, 본 발명에서 사용되는 용어, "예방"이란 본 발명에 따른 약학적 조성물의 투여에 의해 도파민 감소로 말미암아 발생하는 질환을 억제하거나 발병을 지연시키는 모든 행위를 의미한다.
본 발명에서 사용되는 용어, "치료"란 본 발명에 따른 약학적 조성물의 투여에 의해 도파민 감소로 말미암아 발생하는 질환에 의한 증세가 호전되거나 이롭게 변경되는 모든 행위를 의미한다.
본 발명의 조성물은 2'-FL에 추가로 동일 또는 유사한 기능을 나타내는 유효성분을 1종 이상 함유할 수 있다.
본 발명의 약학적 조성물은 2'-FL 이외에 약학적으로 허용 가능한 담체를 추가로 포함할 수 있다.
본 발명에서 사용될 수 있는 담체의 종류는 특별히 제한되지 아니하며 당해 기술 분야에서 통상적으로 사용되는 담체라면 어느 것이든 사용할 수 있다. 상기 담체의 비제한적인 예로는, 락토오스, 덱스트로오스, 수크로오스, 솔비톨, 만니톨, 식염수, 멸균수, 링거액, 완충 식염수, 알부민 주사 용액, 자일리톨, 에리스리톨, 말티톨, 말토덱스트린, 글리세롤, 에탄올 등을 들 수 있다. 이들은 단독으로 사용되거나 2종 이상을 혼합하여 사용될 수 있다.
또한, 본 발명의 약학적 조성물은 필요한 경우, 항산화제, 부형제, 희석제, 완충액 또는 정균제 등 기타 약학적으로 허용 가능한 첨가제들을 첨가하여 사용할 수 있으며, 계면활성제, 결합제, 충진제, 증량제, 습윤제, 붕해제, 분산제 또는 윤활제 등을 부가적으로 첨가하여 사용할 수 있다.
본 발명의 약학적 조성물에 있어서, 2'-FL은 약학적 조성물의 전체의 중량을 기준으로 0.00001 중량% 내지 99.99 중량%로 포함될 수 있으며, 바람직하게는 0.1 중량% 내지 90 중량%, 보다 바람직하게는 0.1 중량% 내지 70 중량%, 더욱 바람직하게는 0.1 중량% 내지 50 중량%로 포함될 수 있으나, 이에 한정되지 않으며 투여 대상의 상태, 구체적인 병증의 종류, 진행 정도 등에 따라 다양하게 변경될 수 있다. 필요한 경우, 약학적 조성물의 전체 함량으로도 포함될 수 있다.
즉, 본 발명의 약학적 조성물의 약학적 유효량, 유효 투여량은 약학적 조성물의 제제화 방법, 투여 방식, 투여 시간 및/또는 투여 경로 등에 의해 다양해질 수 있으며, 약학적 조성물의 투여로 달성하고자 하는 반응의 종류와 정도, 투여 대상이 되는 개체의 종류, 연령, 체중, 일반적인 건강 상태, 질병의 증세나 정도, 성별, 식이, 배설, 해당 개체에 동시 또는 이시에 함께 사용되는 약물 기타 조성물의 성분 등을 비롯한 여러 인자 및 의약 분야에서 잘 알려진 유사 인자에 따라 다양해질 수 있으며, 당해 기술 분야에서 통상의 지식을 가진 자는 목적하는 치료에 효과적인 투여량을 용이하게 결정하고 처방할 수 있다. 예를 들어, 본 발명의 약학적 조성물의 일일 투여량은 약 0.01 내지 1,000 mg/kg이고, 바람직하게는 0.1 내지 100 mg/kg이며, 하루 일회 내지 수회에 나누어 투여할 수 있다.
본 발명의 약학적 조성물의 투여는 하루에 1회 투여될 수 있고, 수 회에 나누어 투여될 수도 있다. 본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양으로 투여할 수 있으며, 이는 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물은 도파민 감소로 말미암아 발생하는 질환을 예방 또는 치료하기 위하여 추가적으로 호르몬 치료, 약물치료 등의 다양한 방법들과 병용하여 사용될 수 있다.
본 발명에서 사용된 용어, "투여"는 어떠한 적절한 방법으로 환자에게 본 발명의 약학적 조성물을 도입하는 것을 의미하며, 본 발명의 약학적 조성물의 투여 경로 및 투여 방식은 각각 독립적일 수 있으며, 목적하는 해당 부위에 상기 약학적 조성물이 도달할 수 있는 한, 특별한 제한 없이 임의의 투여 경로 및 투여 방식에 따를 수 있다.
상기 약학적 조성물은 경구 투여 또는 비경구 투여 방식으로 투여할 수 있으며, 경구 투여 또는 비경구 투여를 위한 적합하고 다양한 제형으로 제제화되어 사용될 수 있다.
본 발명의 약학적 조성물을 이용한 경구 투여용 제제의 비제한적인 예로는, 유성 현탁액, 트로키제(troches), 로젠지(lozenge), 정제, 수용성 현탁액, 조제 분말, 과립, 에멀젼, 하드 캡슐, 소프트 캡슐, 시럽 또는 엘릭시르제 등을 들 수 있다.
본 발명의 약학적 조성물을 경구 투여용으로 제제화하기 위하여, 솔비톨, 만니톨, 전분, 아밀로펙틴, 셀룰로오스 락토오스, 사카로오스 또는 젤라틴 등과 같은 결합제; 스테아르산 마그네슘, 스테아르산 칼슘, 스테아릴 푸마르산 나트륨 또는 폴리에틸렌 글리콜 왁스 등과 같은 윤활유; 디칼슘 포스페이트 등과 같은 부형제; 옥수수 전분 또는 고구마 전분 등과 같은 붕해제 등을 사용할 수 있으며, 방향제, 시럽제, 감미제 등도 사용할 수 있다. 나아가 캡슐제의 경우에는 상기 언급한 물질 외에도 지방유와 같은 액체 담체 등을 추가로 사용할 수 있다.
본 발명의 약학적 조성물의 비경구 투여 방법으로는, 근육 내 투여, 경피 투여, 정맥 내 투여, 복강 내 투여 또는 피하 투여 등을 이용할 수 있으며, 상기 조성물을 질환 부위에 도포하거나 분무, 흡입하는 방법 또한 이용할 수 있으나 이에 제한되지 않는다.
본 발명의 약학적 조성물을 이용한 비경구용 제제의 비제한적인 예로는, 주사액, 좌제, 연고, 도포용 파우더, 오일, 호흡기 흡입용 분말, 스프레이용 에어로졸제, 크림 등을 들 수 있다.
본 발명의 약학적 조성물을 비경구 투여용으로 제제화하기 위하여, 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결 건조 제제, 외용제 등을 사용할 수 있으며, 상기 비수성용제, 현탁제로는 올리브 오일과 같은 식물성 기름, 프로필렌글리콜, 폴리에틸렌글리콜, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
본 발명의 약학적 조성물을 주사액으로 제제화하는 경우, 본 발명의 약학적 조성물을 안정제 또는 완충제와 함께 물에서 혼합하여 용액 또는 현탁액으로 제조하고 이를 앰플(ampoule) 또는 바이알(vial)의 단위 투여용으로 제제화할 수 있다.
본 발명의 약학적 조성물을 에어로졸제로 제제화하는 경우, 수분산된 농축물 또는 습윤 분말이 분산되도록 추진제 등이 첨가제와 함께 배합할 수 있다.
본 발명의 약학적 조성물을 연고, 오일, 크림, 도포용 파우더, 피부 외용제 등으로 제제화하는 경우에는, 동물성 유, 식물성 유, 왁스, 파라핀, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크, 전분, 트라칸트, 셀룰로오스 유도체, 산화 아연 등을 담체로 사용하여 제제화할 수 있다.
한편, 본 발명은 2'-FL (2'-fucosyllactose)을 유효성분으로 함유하는 도파민 분비 감소로 말미암은 질환의 개선용 식품 조성물을 제공한다.
이때, 상기 도파민 감소로 말미암아 발생하는 질환은, 일 예로 도파 반응성 근육긴장이상(dopa-responsive dystonia), 세가와병(segawa's syndrome), 다발성경화증(multiple sclerosis), 파킨슨병(Parkinson's disease), 주의력결핍 과잉행동장애(attention deficit hyperactivity disorder), 투렛증후군(Tourette syndrome), 우울장애(depressive disorder), 조현병(schizophrenia), 양극성 장애(bipolar disorder), 중독(addiction), 통증 및 구역질 중 선택되는 어느 하나인 것일 수 있다. 이에 대한 구체적 기재는 앞서 상술한 내용과 동일한바 그 기재를 생략한다.
보다 상세하게는, 본 발명은 2'-FL (2'-fucosyllactose)을 유효성분으로 함유하며, 도파민 신경세포의 감소를 억제하며, 운동활성증진효과를 가지는 것을 특징으로 하는 도파민 감소로 말미암은 파킨슨병의 개선용 식품 조성물을 제공한다.
본 발명의 일 실시예에 의하면, 2'-FL은 신경독성물질인 6-OHDA가 유발하는 도파민 신경세포의 감소를 효과적으로 억제함을 확인하였다 (도 2). 또한, 본 발명의 일 실시예에서는 2'-FL의 투여로 인해 뇌의 흑색질에서의 TH-면역반응성이 대조군에 비해 현저히 증가되어 뇌의 흑색질에서 도파민 감소 억제능이 있음을 확인하였다 (도 6).
특히, 본 발명의 일실시예에서는 파킨슨이 유발된 동물모델에서 2'-FL의 투여가 마우스의 움직임을 증가시켜 운동활성증진 효과가 있으며 (도 4), 2-FL의 투여는 6-OHDA 투여 후 메스암페타민(Meth)에 의해 유발되는 로테이션 행동을 전체적으로 완화 및 억제시켰음을 확인하였다 (도 5).
본 발명의 식품 조성물에 있어, 2'-FL의 함량은 특별히 제한되지 않으며, 투여 대상의 상태, 구체적인 병증의 종류, 진행 정도 등에 따라 다양하게 변경될 수 있다. 필요한 경우, 식품의 전체 함량으로도 포함될 수 있다.
본 발명의 식품 조성물은 일 예로, 면류, 껌류, 유제품류, 아이스크림류, 육류, 곡류, 카페인 음료, 일반음료, 초콜릿, 빵류, 스낵류, 과자류, 사탕, 피자, 젤리, 알코올성 음료, 술, 비타민 복합제 및 그 밖의 건강보조식품류 중 선택되는 어느 하나일 수 있으나, 반드시 이에 한정되는 것은 아니다.
본 발명의 식품 조성물이 식품 첨가물의 형태로 사용될 경우, 이를 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다.
또한, 본 발명의 "식품 조성물"은 건강기능식품을 포함하는 의미를 포괄하는데, 상기 "건강기능식품"이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
본 발명의 식품 조성물은 통상 사용되어 냄새, 맛, 시각 등을 향상시킬 수 있는 추가 성분을 포함할 수 있다. 예들 들어, 비오틴(biotin), 폴레이트(folate), 판토텐산(pantothenic acid), 비타민 A, C, D, E, B1, B2, B6, B12, 나이아신(niacin) 등을 포함할 수 있다. 또한, 크롬(Cr), 마그네슘(Mg), 망간(Mn), 구리(Cu), 아연(Zn), 철(Fe), 칼슘(Ca) 등의 미네랄을 포함할 수 있다. 또한, 시스테인, 발린, 라이신, 트립토판 등의 아미노산을 포함할 수 있다. 또한, 방부제(소르빈산 칼륨, 벤조산나트륨, 살리실산, 디히드로초산나트륨 등), 착색제(타르색소 등), 발색제(아질산나트륨, 아초산나트륨 등), 표백제(아황산나트륨), 살균제(표백분과 고도 표백분, 차아염소산나트륨 등), 팽창제(명반, D-주석산수소칼륨 등), 강화제, 유화제, 증점제(호료), 피막제, 산화방지제(부틸히드록시아니졸(BHA), 부틸히드록시톨루엔(BHT) 등), 조미료(MSG 글루타민산나트륨 등), 감미료(둘신, 사이클레메이트, 사카린, 나트륨 등), 향료(바닐린, 락톤류 등), 검기초제, 거품억제제, 용제, 개량제 등의 식품 첨가물(food additives)을 첨가할 수 있다. 상기 첨가물은 식품의 종류에 따라 선별되고 적절한 양으로 사용될 수 있다.
이하, 본 발명의 내용에 대해서 하기 실시예를 통해 더욱 상세히 설명하고자 한다. 다만, 본 발명의 권리범위가 하기 실시예만 한정되는 것은 아니고 그와 등가의 기술적 사상의 변형까지를 포함한다.

실시예
1: 도파민 신경세포 배양에서
신경독성물질에
대한 2'-FL (2'-fucosyllactose)의 신경보호 효과 검증]
도파민성 신경세포배양은 임신된 스프라그-돌리(Sprague-Dawley) 랫드에서 얻은 E14-E15 배아의 복뇌(ventral mesencephalon) 조직으로부터 준비하였다. 구체적으로, 해부된 복뇌 조직을 풀링한 다음 37℃에서 15분간 교반하여 미리 워밍업된 트립신 EDTA(025%)에서 세포들을 분리한 후, DMEM (Dulbecco's modified Eagle medium)/F12 배양액으로 세척하였다. 세척 후, 성장필수인자들을 첨가 후 배양을 시작하였다. 배양 3일과 5일에 각각 새로운 배양액을 50% 교체하여 배양을 유지하였고, 배양 7일째부터 항산화인자를 배제하였다.
또한, 배양 10일째에 신경독성물질인 6-OHDA 및 '6-OHDA+2'-FL'이 세포 내로 첨가되었고, 48시간 후 세포들은 4% PFA (paraformaldehyde; Sigma-Aldrich)을 이용하여 고정하였다(도 1). 도 1은 시험관 내(in vitro) 실험의 타임 라인을 나타낸 도이다.
고정된 세포들은 티로신 수산화효소(tyrosine hydroxylase)를 이용한 면역세포화학법을 이용하여 도파민 세포들의 생존비율을 확인하였다. 각 그룹별로 6개의 샘플이 사용되었다. 각각의 그룹의 조성은 하기 표 1에 표시하였고, 세포의 수는 1×106로 동일한 조건을 사용하였다.

실험 결과, 신경독성 물질인 6-OHDA의 투여는 정상세포군에 비해 심각한 도파민 세포의 감소를 유발하였으나, 2'-FL의 투여로 인해 도파민 세포의 감소가 억제되었다(도 2). 도 2는 1차 복뇌신경세포(ventral mesencephalon neuron) 배양에서 2'-FL의 6-OHDA 매개 도파민성 변성에 대한 작용을 확인한 결과이다.
즉, 2'-FL이 신경독성물질인 6-OHDA가 유발하는 도파민 세포의 감소를 억제한 것으로 확인되었다.
[실시예 2: 파킨슨이 유발된 동물모델에서의 2'-FL의 신경보호 효과 검증]
2-1. 2'-FL 투여에 의한 행동개선 여부 평가 및 조직학적 분석을 통한 도파민 세포의 분포 변화 검증
마우스는 클로로랄 하이드레이트(400 ㎎/㎏, ip)로 마취하여 스테레오탁식 프레임(stereotaxic frame)에 고정시켰다. 6-OHDA HBr (276 ㎍/㎕×5 ㎕)을 해밀턴 미세시린을 통해 4분 동안 좌측 선조체 2개 부위(1 ㎜ AP, 32 ㎜ ML relative to bregma and 51 ㎜ below skull and 1 ㎜ AP, 26 ㎜ ML relative to bregma and 42 ㎜ below skull)에 주입하였다(Day 0). 이후, 마취에서 회복된 마우스를 37℃ 인큐베이터에서 2시간 동안 체온을 유지하였다.
2'-FL (500 μM×20 ㎕) 및 식염수(대조군; 20 ㎕)는 0일에서 28일까지 코로 주입하였다. 파킨슨병의 유발 유무와 행동변화 관찰을 위해 메스암페타민(methamphetamine)을 쥐에게 투여한 후 행동변화를 각각 7일, 21일 및 28일에 실시하였다(도 3). 도 3은 생체 내(in vivo) 실험의 타임 라인을 나타낸 도이다.
첫 번째로, 20일에 전위(locomotor) 행동 테스트를 시행하였다. 구체적으로, 자동화된 아쿠스캔(Accuscan) 활동 모니터(Columbus, OH)를 사용하여 개방된 장소에서의 활동을 측정하였다. 각 동물은 42×42×31 ㎝ 크기의 오픈박스에 2시간 동안 물병과 음식이 제공된 장소에서 자유로운 활동을 하며, 오픈박스에 설치된 적외선 측정기를 이용하여 움직임을 계산하였다.
실험 결과, 5가지의 서로 다른 측정방법(HACTV, TOTDIST, VTIME, VACTV 및 MOVTIME)에서 2'-FL(n=10)의 투여가 대조군(n=11)에 비해 마우스의 움직임을 증가시켰음을 확인하였다(도 4). 도 4는 6-OHDA 투여 후 20일째 2'-FL의 운동 활성 증진 테스트 결과이다.
두 번째로, 7일, 21일 및 28일에 메스암페타민 투여에 의해 유발된 로테이션 행동의 완화 테스트를 2'-FL 투여군(500 μM) 및 대조군에서 시행하였다.
실험 결과, 2'-FL 투여는 6-OHDA 투여 후 메스암페타민(Meth)에 의해 유발되는 로테이션 행동을 전체적으로 완화시켰고, 특히 21일에서 억제하는 것으로 나타났다(도 5). 도 5는 메스암페타민 투여에 의해 유발된 로테이션 행동을 2'-FL이 완화시켜 주는지에 대한 테스트 결과이다.
2-2. 2'-FL 투여에 의한 흑색질에서의 도파민 감소 억제 효과 검증
로테이션 테스트 후 동물들은 도살되어 뇌조직을 적출하였다(Veh, n=5; 2'-FL, n=4). 도파민 신경세포의 분포 정도는 티로신 수산화효소(tyrosine hydroxylase; TH)를 이용한 면역조직화학에 의해 측정되었다.
구체적으로, 냉동절편 기계를 이용하여 25μm 두께로 뇌 전체를 절개하였다. 그런 다음 4℃에서 17-19시간 동안 1차 항체(TH, 농도 1:100; Chemicon, Temecula, CA)에서 뇌조직을 배양했다. 이후, 2차 항체(농도 1:500)와의 결합을 마친 후, 뇌 절편들은 슬라이드 글라스에 붙인 후 형광현미경을 이용하여 발현을 확인하였다.
실험 결과, 6-OHDA에 의한 흑색질에서의 TH(+) 세포들의 숫자는 현저히 줄어들었으나(도 6의 윗면), 2'-FL의 장기간 투여는 이러한 세포들의 사멸을 억제하는 효과를 보였다(도 6의 아랫면).
또한, 2'-FL의 투여로 인해 흑색질(중간의 white box)에서의 TH-면역반응성(immunoreactivity)이 대조군에 비해 현저히 증가되었음을 확인하였다(p=0007, t-test). 도 6은 2'-FL 투여에 의한 흑색질에서의 도파민 감소 억제능을 확인한 결과이다.
2-3. 2'-FL 투여에 따른 염증관련 유전자의 발현 변화 검증
흑색질 조직은 6-OHDA 주입 후 28일에 수집하였다(Veh, n=4; 2'-FL, n=3). 염증성 마커의 발현은 qRTPCR을 통하여 확인하였다.
실험 결과, 2'-FL의 투여는 M1 마커 CD86 (pro-inflammatory)의 발현을 감소시키고(p=0087, t-test), 항염증 M2 표지인자인 CD206의 발현을 증가시키는 패턴을 보였다(p=0053, t-test)(도 7). 도 7은 2'-FL 투여에 의한 염증관련 유전자의 발현 변화를 확인한 결과이다.
[실시예
3: 파킨슨병에서 2'-FL이 장내 미생물 환경에 미치는 영향 검증]
장기능 장애로 파킨슨병 환자의 80%가 변비를 앓고 있는 것으로 알려졌다. 이에, 2'-FL이 파킨슨이 유도된 쥐의 장내 미세환경을 변화시킬 수 있는지에 대하여 확인하였다.
이를 위하여, "누출성 장(leaky gut)"에서 증가하는 장 투과성 표지인자들의 발현을 확인하였다. 최종 로테이션 테스트 후, 도살 전에 꼬리 정맥을 통하여 채혈하고 ELISA 방식을 통해 표지인자들의 발현량을 조사하였다.
3-1. 조눌린(zonulin) 발현 변화 검증
병원체(pathogen)나 상주세균(resident bacteria)이 소장의 벽에 직접 부착되면 장 점막세포로부터 조눌린 분비가 촉진되고 장내 투과성이 높아진다.
조눌린 신호 경로의 조절 장애는 정상적인 장관 점막세포의 장벽 기능을 방해하고 면역 반응을 변화시킨다. 또한, 조눌린 혈청 수치 상승과 장내 투과성 증가는, 증상이 나타나기 훨씬 전에 질병에 걸릴 위험이 있는 환자에게서 흔히 관찰된다고 보고되었다.
본 실험에서는, 총 8마리(Veh, n=4; 2'-FL, n=4)의 쥐로부터 6-OHDA 주입 28일 후 혈액을 채취하여 조눌린의 발현을 조사하였다.
실험 결과, 2'-FL을 장기간 투여한 쥐의 조눌린 발현이 대조군에 비해 감소하는 경향을 보였다(p=0076, t-test)(도 8). 도 8은 2'-FL 투여에 의한 혈액에서 조눌린의 발현 변화를 확인한 결과이다.
3-2. 오클루딘(Occludin, Ocln) 발현 변화 검증
장 장벽 무결성(Gut barrier integrity)은, 상피세포 장벽 기능에 중요한 역할을 하는 클라우딘(Cladin, Cldn), 조눌라 오클루덴스-1(ZO-1), 오클루딘(Occludin, Ocln)과 같은 '막관통 장벽 단백질'에 의해 유지된다고 보고되었다. 또한, 식이조절 또는 고지방식단의 경우 활성화된 오클루딘의 신체에 미치는 유익한 보고들이 있다.
본 실험에서는, 총 23마리(Veh, n=12; 2'-FL, n=11)의 쥐로부터 6-OHDA 주입 28일 후 혈액을 채취하여 오클루딘의 발현을 조사하였다.
실험 결과, 2'-FL을 장기간 투여한 쥐의 오클루딘 발현이 대조군에 비해 증가하는 경향을 보였다(p=0069, t-test)(도 9). 도 9는 2'-FL 투여에 의한 혈액에서 오클루딘의 발현 변화를 확인한 결과이다.
3-3. 밀착 연접(tight junction) 단백질 발현 변화 검증
밀착 연접(tight junction; TJ)은 세포 간 통로를 제어하고 개별 세포 사이에 조절 가능한 반투과성 확산 장벽을 만드는 기작으로, 막관통 단백질(transmembrane protein)에 의해 매개된다. 상피세포에서 밀착 연접은 오클루딘(occluding), 클라우딘(claudins), 연접접착분자(junctional adhesion molecules; JAM), 조눌라 오클루덴스(ZO)-1, 2, 3에 의해 형성된다. ZO-1은 접합 형성에 매우 중요하다. ZO-1이 없으면 세포들이 밀착 연접을 형성하지 못한다고 알려져 있다.
본 실험에서는, 총 9마리(Veh, n=4; 2'-FL, n=5)의 쥐로부터 6-OHDA 주입 28일 후 소장을 채취하여 PCR 방법을 이용하여 ZO-1의 발현을 측정하였다.
실험 결과, 2'-FL 투여가 파킨슨이 유발된 흰쥐의 소장에서, 밀착 연접 단백질에 연관된 ZO-1의 발현을 증가시키는 것으로 확인되었다(p=0025, t-test)(도 10). 도 10은 2'-FL 투여에 의한 소장에서 ZO-1의 발현 변화를 확인한 결과이다.
