Abstract
본 발명은 사이클로아스트라제놀을 유효성분으로 함유하는 퇴행성 뇌질환의 예방, 개선 또는 치료용 조성물에 관한 것으로, 아밀로이드 베타로 유도한 알츠하이머병 동물에게 본 발명의 유효성분인 사이클로아스트라제놀을 투여하여 퇴행성 뇌질환 관련 단백질의 발현량 변화를 조절할 수 있으며, 인지기능을 증진시킬 수 있다.
Description
본 발명은 사이클로아스트라제놀을 유효성분으로 함유하는 퇴행성 뇌질환의 예방, 개선 또는 치료용 조성물에 관한 것이다.
퇴행성뇌질환 환자의 뇌에서 공통적으로 발견되는 병리학적 특징으로는 신경세포의 사멸과 특정단백질의 비정상적 응집이 있다. 아밀로이드 베타(amyloid β), 노인반(senile plaque), 신경섬유농축제(neurofibrillary tangle), 알파 시뉴클린(α-synuclein) 또는 루이소체(lewy body) 등의 단백질의 비정상적 응집이 그 자체로 신경세포의 사멸을 유도하거나, 미세아교세포를 활성화해 염증반응을 일으킴으로써 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 진행성 핵상마비(Progressive supranuclear palsy), 다계통 위축증(Multiple system strophy), 감람핵-뇌교-소뇌 위축증(Olivopontocerebellar atrophy; OPCA), 샤이-드래거 증후군(Shy-Drager syndrome), 선조체-흑질 퇴행증 (Striatonigral degeneration), 헌팅톤병(Huntington's disease), 근위축성 측색 경화증(Amyotrophic lateral sclerosis; ALS), 본태성 진전증(Essential tremor), 피질-기저핵 퇴행증 (Cortico-basal ganlionic degeneration), 미만성 루이 소체 질환(Diffuse Lewy body disease), 파킨스-ALS-치매 복합증(Parkinson-ALS-dementia complex of Guam), 픽병(Pick's disease) 등의 퇴행성 뇌질환의 발병에 중요한 역할을 하는 것으로 알려져 있다.
상기 퇴행성 뇌질환 중에서, 알츠하이머병(AD)은 신경세포(neuron) 상실과, 아밀로이드 전구체 단백질로부터 유래된 39∼43 아미노산 펩티드인 아밀로이드 β 단백질 (Aβ)을 주요 구성성분으로 하는 세포외 노인성반(senile plaque)을 특징으로 한다. 시험관 내 및 생체 내 연구 결과 Aβ 또는 Aβ 펩티드 단편은 독성 효과를 갖는 것으로 보고되어 Aβ가 AD의 발병에 중요한 역할을 하는 것으로 알려져 있다.
한편, 사이클로아스트라제놀(cycloastragenol)은 황기 뿌리에서 추출되는 천연사포닌으로 텔로미어를 연장시키는 텔로머라아제 활성화제로 알려져 있어 노화, 암 또는 심혈관 질환에 도움을 줄 수 있으며, 자외선에 의한 노화 또는 당화형성을 억제하는 것으로 알려져 있다.
사이클로아스트라제놀(cycloastragenol) 관련 기술로는 한국등록특허 제1465515호에 텔로머라제 활성을 증가시키기 위한 조성물 및 방법이 개시되어 있고, 한국공개특허 제2021-0125530호에 노화 및 연령-관련 기관 기능부전과 연관된 질환을 치료하기 위한 텔로머라제-함유 엑소좀에 관한 기술이 개시되어 있으나, 아직까지 본 발명의 사이클로아스트라제놀을 유효성분으로 함유하는 퇴행성 뇌질환의 예방, 개선 또는 치료용 조성물에 대해서는 언급된 바 없다.
본 발명은 상기와 같은 요구에 의해 도출된 것으로서, 본 발명은 사이클로아스트라제놀을 유효성분으로 함유하는 퇴행성 뇌질환의 예방, 개선 또는 치료용 조성물을 제공하고, 아밀로이드 베타로 유도한 알츠하이머병 동물에게 본 발명의 유효성분인 사이클로아스트라제놀을 투여하여 퇴행성 뇌질환 관련 단백질의 발현량 변화를 조절할 수 있다는 것을 확인하였을 뿐만 아니라, MWM(Morris water maze) 분석에서 기억력 및 인지기능이 증진된 것을 확인함으로써, 본 발명을 완성하였다.
상기 목적을 달성하기 위하여, 본 발명은 사이클로아스트라제놀(cycloastragenol) 또는 이의 약리학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물을 제공한다.
또한, 본 발명은 사이클로아스트라제놀(cycloastragenol) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 조성물을 인간을 제외한 동물에게 투여하는 단계를 포함하는 퇴행성 뇌질환의 예방 또는 치료방법을 제공한다.
또한, 본 발명은 사이클로아스트라제놀(cycloastragenol) 또는 이의 식품학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 개선용 건강기능식품 조성물을 제공한다.
또한, 본 발명은 사이클로아스트라제놀(cycloastragenol) 또는 이의 약리학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 치료용 수의학적 조성물을 제공한다.
또한, 본 발명은 사이클로아스트라제놀(cycloastragenol) 또는 이의 식품학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 개선용 사료 첨가제를 제공한다.
본 발명은 사이클로아스트라제놀을 유효성분으로 함유하는 퇴행성 뇌질환의 예방, 개선 또는 치료용 조성물에 관한 것으로, 아밀로이드 베타로 유도한 알츠하이머병 동물에게 본 발명의 유효성분인 사이클로아스트라제놀을 투여하여 퇴행성 뇌질환 관련 단백질의 발현량 변화를 조절할 수 있으며, 기억력 및 인지기능을 증진시킬 수 있다.

본 발명은 하기 화학식 1의 사이클로아스트라제놀(cycloastragenol) 또는 이의 약리학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물에 관한 것이다.
상기 퇴행성 뇌질환은 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 헌팅턴병(Huntington disease), 경도인지장애(mild cognitive impairment), 대뇌 아밀로이드 맥관병증, 아밀로이드성 뇌졸중(stroke), 전신성 아밀로이드병, 노인성 치매, 근위축성 축삭경화증(amyotrophic lateral sclerosis) 및 전두촉두엽 치매(frontotemporal dementia) 중에서 선택된 어느 하나인 것이 바람직하지만 이에 한정하지 않는다.
상기 사이클로아스트라제놀은 황기 뿌리(Astragalus radix)로부터 분리된 것일 수 있지만 이에 한정하지 않는다.
상기 퇴행성 뇌질환은 산화스트레스(Oxidative Stress), 신경영양 기능장애(Neurotrophic Dysfunctions), 신경염증(Neuroinflammation) 또는 세포사멸(Apoptotic cell death)에 의한 것이 특징이다.
상기 퇴행성 뇌질환은 아밀로이드-베타의 축적에 의해 유발된 것일 수 있으며, 상기 사이클로아스트라제놀은 아밀로이드 베타(Aβ)의 축적에 의해 감소된 Nrf2(Nuclear factor erythroid-2-related factor 2), HO-1(Heme oxygenase 1), BDNF(Brain-derived neurotrophic factor), p-TrkB(p-ropomyosin receptor kinase B), p-CREB(cAMP-response element-binding protein), NeuN(neuronal nuclear protein), p-ERK(phospho-extracellular signal-related kinase) 및 Bcl-2(B cell lymphoma/leukemia-2) 중에서 선택된 하나 이상의 단백질 발현량을 증가시킬 수 있고, 아밀로이드 베타(Aβ)의 축적에 의해 증가된 p-JNK(phospho c-Jun-N-terminal kinase), p-P38, Iba-1(ionized calcium-binding adapter molecule-1), GFAP(Glial Fibrillary Acidic Protein), TNF-α(tumour necrosis factor-alpha), IL-1β(interleukin 1-beta), Bax, Cleaved Caspase-3 및 Bim 중에서 선택된 하나 이상의 단백질 발현량을 감소시킬 수 있다.
상기 약학 조성물의 유효성분 이외에 약학적으로 허용가능한 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로즈 용액, 말토덱스트린 용액, 글리세롤, 에탄올, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 중에서 선택된 1종 이상의 담체를 더 함유할 수 있으며, 상기 유효성분 이외에 약학적으로 허용 가능한 항산화제, 완충액, 정균제, 희석제, 계면활성제, 결합제, 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제 및 보존제 중에서 선택된 1종 이상의 보조제를 더 함유할 수 있다.
상기 약학 조성물은 통상의 방법에 따라 경구 또는 비경구의 제형으로 투여될 수 있으며, 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 상기 조성물에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트 (calcium carbonate), 수크로오스, 락토오스, 젤라틴 등을 섞어 조제된다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 또한, 비수성용제, 현탁제로는 프로필렌 글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
또한, 본 발명은 하기 화학식 1의 사이클로아스트라제놀(cycloastragenol) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 조성물을 인간을 제외한 동물에게 투여하는 단계를 포함하는 퇴행성 뇌질환의 예방 또는 치료방법에 관한 것이다.

또한, 본 발명은 하기 화학식 1의 사이클로아스트라제놀(cycloastragenol) 또는 이의 식품학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 개선용 건강기능식품 조성물에 관한 것이다.

본 발명의 건강기능식품 조성물에서, 상기 퇴행성 뇌질환은 전술한 바와 같다.
상기 건강기능식품 조성물은 음료, 환, 정제(tablet), 캡슐제(capsule), 산제 중에서 선택된 어느 하나의 식품으로 제조하거나, 다른 식품 또는 식품의 성분에 첨가하여 제조될 수 있으며, 통상적인 방법에 따라 적절하게 제조될 수 있다.
본 발명의 조성물을 첨가할 수 있는 식품의 일례로는 육류, 소시지, 빵, 초콜릿, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합제 중에서 선택된 어느 하나의 형태일 수 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
상기 건강기능식품 조성물은 여러 가지 영양제, 비타민, 광물(전해질), 합성 및 천연 풍미제, 착색제 및 증진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알킨산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그 밖에 천연 과일 주스 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있고, 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상기 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드; 말토스, 수크로스와 같은 디사카라이드; 및 덱스트린, 사이클로덱스트린과 같은 폴리사카라이드; 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다.
또한, 본 발명은 하기 화학식 1의 사이클로아스트라제놀(cycloastragenol) 또는 이의 약리학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 치료용 수의학적 조성물에 관한 것이다.

본 발명의 수의학적 조성물은 통상의 방법에 따른 적절한 부형제 및 희석제를 더 포함할 수 있다. 본 발명의 수의학적 조성물에 포함될 수 있는 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시 벤조에이트, 탈크, 마그네슘 스테아레이트, 세탄올, 스테아릴알콜, 유동파라핀, 솔비탄모노스테아레이트, 폴리소르베이트 60, 메칠파라벤, 프로필파라벤 및 광물유를 들 수 있다. 본 발명에 따른 수의학적 조성물은 충진제, 항응집제, 윤활제, 습윤제, 향신료, 유화제, 방부제 등을 추가로 포함할 수 있는데, 본 발명에 따른 수의학적 조성물은 동물에 투여된 후 활성 성분의 신속, 지속 또는 지연된 방출을 제공할 수 있도록 당업계에 잘 알려진 방법을 사용하여 제형화될 수 있고, 제형은 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 용액, 시럽, 에어로졸, 연질 또는 경질 젤라틴 캅셀, 좌제, 멸균 주사용액, 멸균 외용제 등의 형태일 수 있다.
본 발명에 따른 수의학적 조성물의 유효한 양은 동물의 개체에 따라 적절하게 선택할 수 있다. 질환 내지 상태의 중증도, 개체의 연령, 체중, 건강상태 또는 성별에 따른 본 발명의 유효성분에 대한 민감도, 투여 경로, 투여 기간, 상기 조성물과 배합 또는 동시 사용되는 다른 조성물을 포함한 요소 및 기타 생리 내지 수의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
또한, 본 발명은 하기 화학식 1의 사이클로아스트라제놀(cycloastragenol) 또는 이의 식품학적으로 허용 가능한 염을 유효성분으로 함유하는 퇴행성 뇌질환의 예방 또는 개선용 사료 첨가제에 관한 것이다.

본 발명의 사료 첨가제는 사료관리법상의 보조사료에 해당한다. 본 발명에서 용어 '사료'는 동물이 먹고, 섭취하며, 소화시키기 위한 또는 이에 적당한 임의의 천연 또는 인공 규정식, 한끼식 등 또는 상기 한끼식의 성분을 의미할 수 있다. 상기 사료의 종류는 특별히 제한되지 아니하며, 당해 기술 분야에서 통상적으로 사용되는 사료를 사용할 수 있다. 상기 사료의 비제한적인 예로는, 곡물류, 근과류, 식품 가공 부산물류, 조류, 섬유질류, 제약 부산물류, 유지류, 전분류, 박류 또는 곡물 부산물류 등과 같은 식물성 사료; 단백질류, 무기물류, 유지류, 광물성류, 유지류, 단세포 단백질류, 동물성 플랑크톤류 또는 음식물 등과 같은 동물성 사료를 들 수 있다. 이들은 단독으로 사용되거나 2종 이상을 혼합하여 사용될 수 있다.
이하, 실시예를 이용하여 본 발명을 더욱 상세하게 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로 본 발명의 범위가 이들에 의해 제한되지 않는다는 것은 당해 기술분야에서 통상의 지식을 가진 자에게 있어 자명한 것이다.
[재료 및 방법]
1. 실험용 마우스
알츠하이머 질병이 있는 실험용 마우스 모델을 만들기 위하여, Aβ1-42 펩타이드를 마우스(C57BL/6N)의 뇌실내에 헤밀톤 시린지를 이용하여 정위 과정의 방법으로 단일 주입하였다. 수컷 마우스들은 평균적으로 10주차의 나이였으며 한국 울산에 있는 Samtako Bio Labs로부터 구입하여 실온(23±2℃)에서 12시간 밤/낮의 주기로 하며, 사료와 물을 자유롭게 섭취할 수 있는 환경에 두었다.
본 발명의 실시예에서 사용된 실험용 마우스들은 경상국립대학교 Applied life science 부서의 승인된 가이드라인(Approval ID: 125)에 따라 다루어졌다.
2. Aβ1-42의 뇌실내(i.c.v) 주입
생리식염수에 녹여 1mg/㎖의 인간 Aβ1-42 펩티드를 제조하였고, 37℃에서 4시간 동안 보관되었다. 수술과 Aβ1-42의 뇌실내 주입을 위하여, 100g의 체중당 0.05㎖의 럼픈(Rompun)과 0.1㎖의 틸라타민 염산염(tiletamine hydrochloride)이용하여 마취하였고, 0.2mm의 anteroposterior(AP), 1mm의 mediolateral(ML) 및 -2.4mm의 dorsoventral(DV)의 브레그마(Bregma)를 고정하였다. 헤밀톤 마이크로-시린지 2.4를 이용하여 천천히 5㎍의 Aβ1-42 펩티드를 포함하는 용액 5㎕을 투여하고 피부를 봉합하였다.
3. 실험용 마우스의 그룹화 및 약물적 처치
Aβ1-42 펩티드를 투여하고 24시간 후, 실험용 마우스를 4개의 그룹으로 나누었다(각 그룹에는 16마리의 실험용 마우스로 구성하였다). Aβ1-42 펩티드 대신에 생리식염수를 투여한 정상대조군, Aβ1-42 펩티드 투여군, Aβ1-42 펩티드 + 20mg/kg의 사이클로아스트라제놀(cycloastragenol) 투여군 및 20mg/kg의 사이클로아스트라제놀(cycloastragenol) 단독 투여군으로 분류하였다.
98%이상의 순도를 가지고 있는 CAG(Cycloastragenol)(Sigma-Aldrich, MA USA)를 5% DMSO에 녹여 준비하였고, PBS에서 2%의 Tween-20과 함께 희석시켰다. 20~25gauge×1.5 인치의 휘어진 dosing cannula를 이용하여 20mg/kg/day의 복용양으로 1일 1회, 6주 동안 사이클로아스트라제놀을 경구투여하였다.
4. 단백질 채취 및 정량화
단백질을 채취하기 위하여, 조직 용해물들을 PRO-PREPTM 추출 용액(iNtRON Biotechonology, Inc.,성남, 한국)에서 균질화시켰고 4℃에서 25분 동안 14,000 rpm의 속도로 원심분리하였다. 그 상등액을 채취하였고 웨스턴 블로팅에 사용되었다.
5. 웨스턴 블롯 분석
단백질 농축물을 Bio-Rad 어세이 키트(Bio-Rad Laboratories, CA, USA)를 이용하여 종래의 방법에 따라 분석하였다. SDS PAGE Gel(sodium dodecyl polyacrylamide gel)에서 미리 염색해 놓은 단백질 마커(GangNam-STAIN, iNTRON Biotechonology, 서울, 한국)와 함께 시료들을 로딩하였고, 전기영동 후에 단백질은 PVDF(polyvinylidene fluoride membrane)로 옮겨졌고, 이어 5% 탈지유로 블로킹하였다.
분석하고자 하는 각각의 단백질에 따른 1차 및 2차 항체를 처리하고, ECL(enhanced chemiluminescent) 시약을 이용하여 시각화 하였다. β-액틴은 로딩 컨트롤을 위한 용도로 사용되었고, 그 결과는 발현 변화값(fold change)의 용어로 표시되었다.
6. 면역형광분석을 위한 시료 준비
실험용 마우스들을 Rompun 및 Zolitil을 이용하여 마취한 실험용 마우스들은 10㎖/분의 유속으로 3분 동안 생리식염수로 transcardially perfused(초월적 관류)하였고, 이후 연동 펌프(peristaltic pump)를 사용하여 8분 동안 4% 파라포름알데히드(paraformaldehyde)를 주입하였다. 실험용 마우스의 뇌를 차가운 4% 파라포름알데히드 중성 완충용액에서 48시간 동안 고정하였다. 이어 30% 자당 용액으로 48시간 동안 세척된 다음 OCT(optimal cutting temperature compound) 복합체 (Sakura, Torrance, CA, USA)에 고정하였다. 글리신(glycine)으로 코팅된 슬라이드들(Fisher, Pittsburgh, PA, USA) 위에서 마이크로톰(Leica cryostat CM 3050S, Nussloch, Germany)을 이용하여 14μm의 두께의 조각들로 잘라 준비하였다.
7. 항체 및 시약
본 연구에 사용된 주시약과 항체들은 다음과 같다 : 사이클로아스트라제놀(CAS Number-78574-94-4)은 Sigma Aldrich으로부터 구입하였고, Nrf-2(sc-722), HO-1(sc-136,961), p-TrkB(sc-365842), TrkB(sc-376776), BDNF(sc-546), p-anti-p-P38(9212S), anti-p-p-P38(9211S), JNK 179(sc-6254), JNK(sc-7345), Caspase-3(sc-7272), Bax(sc-7480), Bcl2(sc-7382), Iba-1(sc-32725), GFAP(sc-33673), TNF-α(sc-52746), IL-1β(sc-32294), Cleaved Caspase-3(#9664), Bim(sc-181 374358) 및 β-actin(sc-47,778)은 싼타크루즈 바이오테크놀로지(Santa Cruz Biotechnology)로부터 구입하였다. p-CREB(#87G3), CREB(#48H2), NeuN(sc-33684), p-ERK(#9101) 및 ERK(#9102) 항체들은 셀 시그날링 테크놀로지(Cell Signaling Technology)로부터 구입하였다.
2차-항체인 항-마우스 HRP(Horseradish peroxidase) 및 항-토끼 HRP는 1× TBS-T에서 1:10,000의 비율로 희석하여 사용하였으며, 면역형광분석을 위하여, 상기 2차 항체가 결합된 FITC 또는 TRITC를 사용하였다.
8. 면역형광분석
공초점 현미경 분석을 위하여, 슬라이드들을 세척하고, proteinase-k와 함께 5분 동안 보관하였다. 이후, 0.02% Triton 및 0.01g/㎖ 소혈청 알부민을 포함하는 정상 염소 혈청에서 45분 동안 보관하였다. 블록을 제작한 후, 슬라이드들을 밤새 4℃에서 1차 항체들과 반응시키고, 1차 항체 처리 후, 슬라이드들은 적절한 rabbit/anti-mouse IgG(형광 FITC/TRITC 표지) 2차 항체(0.01M PBS에서 1:100으로 희석된 2차 항체)(Invitrogen Korea Inc)로 2시간 동안 상온에서 처리되었다.
핵염색을 위하여, 4',6-diamidino-2-phenylindole(DAPI, 1;1000으로 PBS에서 희석)을 사용하였으며, DAPI의 처리 이후에, 슬라이드들을 형광 mounting medium DAKO(S3023)을 사용하여 커버슬립으로 덮었다. 역치 아래의 상황과 비교하여 평가한 전 영역의 면역형광 반응성은 이미지 J(v. 1.50, NIH, Bethesda, MD, USA)를 사용하여 분석하였다.
9. 생체 내 ROS 및 LPO 분석
ROS(reactive oxygen species) 분석은 종래의 방법으로 분석하였다. ROS 분석은 2',7'-dichlorodihydrofluorescein diacetate(DCFH-204 DA)가 fluorescent 2',7'-dichlorofluorescein(DCF)로 산화되는 것에 근거하였다. ROS의 정도를 분석하기 위하여, 뇌피질과 해마가 균질화된 실험용 마우스의 뇌는 Loke's buffer (1:20 비율)에서 5mg tissue/㎖ 농도로 희석되었다. 이후, 1㎖의 Loke's 완충용액, 0.2㎖의 뇌 분쇄물 및 10㎖의 DCFH-DA(5mM)를 반응하여 형광 DCF를 얻기 위하여 상온에서 15분 동안 보관하였다.
DCF 정도는 마이크로플레이트 리더기(발광파장: 484/530nm)로 측정하였다. 평행하게 있는 공백들은 DCF가 존재하지 않는 백그라운드 형광을 확인하기 위해 이용되었다.
획득된 결과들은 분(min) 당 및 단백질 밀리그램 당 생성되는 pmol DCF으로 표시되었다. LPO(Lipid peroxidation) 분석은 종래 방법에 따라 수행하였으며, free Malondialdehyde(MDA)는 LPO의 마커로 사용하였으며, 뇌피질과 해마 내에서 MDA colorimetric/fluorometric kit(Cat #K739-100)을 이용하여 측정하였다.
10. Morris Water Maze 시험
기억력 관련 인자들은 비디오 추적 소프트웨어(SMART Panlab, Harvard Apparatus, Holliston, MA, USA)을 사용하는 Morris Water Maze(MWM) 시험을 이용하여 분석하였다. 5일 동안의 훈련을 받은 후에, 플랫폼까지 도달하는 지연시간(초)이 기록하였다. 훈련을 마친 후에, 프로브 테스트가 실시하였고, 플랫폼을 제거하고 실험용 마우스가 1분 동안 자유롭게 수영하여 이전에 위치한 플랫폼 위치에 도달하도록 함으로써 기억 형성을 평가하는 것이다.
6일 차에 해당 플랫폼 위치를 지나치는 횟수, 특정 사분면에서 시간을 소비하는 것 및 해당 위치까지 도달하는데 걸린 지연시간을 확인하였다.
11. 데이터 분석 및 통계
이미지 J 소프트웨어를 사용하여, 웨스턴 블롯 밴드들의 상대적인 밀도들을 계산하였고 각 그룹의 8마리의 실험용 마우스들에 대하여 웨스턴 블롯 및 면역형광을 위한 3번의 반복 실험으로 "평균±표준편차(SD)"를 표시하였다. 각 그룹들 간의 비교를 위하여, one-way ANOVA에 이은 Bonferroni's multiple comparisons test가 사용되었다. 계산은 Prism 6 소프트웨어(GraphPad Software, San Diego, CA, USA)를 이용하여 수행되었다. p<0.05를 기준으로 하여 유의한 차이(significant difference) 여부를 결정하였다.
실시예 1. Aβ1-42 펩티드를 투여한 실험용 마우스에서 사이클로아스트라제놀의 산화 스트레스 및 신경영양학적 인자에 대한 효과 확인
증가된 산화 스트레스에 대한 사이클로아스트라제놀(Cycloastragenol)의 효과를 분석하기 위하여, 실험용 마우스의 뇌 분쇄물에서의 LPO 및 ROS의 함량 변화를 확인하였다.
그 결과, 대뇌 피질(Cortex) 및 해마(Hippocampus)에서 대조군 대비 Aβ1-42 펩티드 투여군의 LPO(Lipid peroxidation) 및 ROS(Reactive oxygen species)의 발현량이 증가하였으며, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 LPO 및 ROS의 함량이 통계적으로 유의미하게 감소하였다(도 1).
또한, 주요 항산화 조절자인 Nrf2(Nuclear factor erythroid-2-related factor 2)의 발현량과 이의 하위 타겟(downstream target)인 HO-1(Heme oxygenase 1)의 발현량, BDNF(Brain-derived neurotrophic factor) 및 이와 관련된 수용체 p-TrkB(p-Tropomyosin receptor kinase B)의 발현량을 확인한 결과, 대조군 대비 Aβ1-42 펩티드 투여군의 HO-1(Heme oxygenase 1)의 발현량, BDNF(Brain-derived neurotrophic factor) 및 이와 관련된 수용체 p-TrkB(p-Tropomyosin receptor kinase B)의 발현량이 통계적으로 유의미하게 감소하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 HO-1(Heme oxygenase 1)의 발현량, BDNF(Brain-derived neurotrophic factor) 및 이와 관련된 수용체 p-TrkB(p-Tropomyosin receptor kinase B)의 발현량이 통계적으로 유의미하게 증가하였다(도 2).
또한, 면역형광 분석에서도, 대조군 대비 Aβ1-42 펩티드 투여군의 BDNF 및 p-TrkB 발현량이 통계적으로 유의미하게 감소하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 BDNF 및 p-TrkB 발현량이 통계적으로 유의미하게 증가하였다(도 3).
실시예 2. Aβ1-42 펩티드를 투여한 실험용 마우스에서 사이클로아스트라제놀의 단백질 CREB(cAMP Response Element-Binding Protein) 및 NeuN(neuronal nuclei)의 발현량 변화 확인
사이클로아스트라제놀의 신경학적 생존 효과를 확인하기 위하여, 실험용 마우스들의 뇌 중 전두엽 피질 및 해마에서 p-CREB 및 NeuN의 발현량을 면역형광분석법을 이용하여 확인하였다.
그 결과, 대조군 대비 Aβ1-42 펩티드 투여군의 p-CREB 및 NeuN 발현량이 통계적으로 유의미하게 감소하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 p-CREB 및 NeuN의발현량이 통계적으로 유의미하게 증가하였다(도 4).
또한, 웨스턴 블랏 결과에서도 p-CREB, 및 NeuN의 발현량을 확인하였으며, 그 결과 대조군 대비 Aβ1-42 펩티드 투여군의 p-CREB 및 NeuN 발현량이 통계적으로 유의미하게 감소하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 p-CREB 및 NeuN 발현량이 통계적으로 유의미하게 증가하였다(도 5).
실시예 3. Aβ1-42 펩티드를 투여한 실험용 마우스의 뇌의 활성화된 성상세포(astrocytes) 및 미세아교세포(microglia)에서 MAPK(Mitogen-activated Protein kinases) 및 염증성 사이토카인에 대한 사이클로아스트라제놀(cycloastragenol)의 효과
사이클로아스트라제놀의 효과를 확인하기 위하여, 실험용 마우스들의 뇌에서신경염증과 관련된 세포내 시그널 마커인 p-ERK, p-JNK 및 p-P38을 포함하는 MAP Kinases의 발현량을 확인하였다.
그 결과, 대조군 대비 Aβ1-42 펩티드 투여군의 p-ERK 발현량이 통계적으로 유의미하게 감소하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 p-ERK 발현량이 통계적으로 유의미하게 증가하였으며,
p-JNK 및 p-P38 발현량의 경우, 대조군 대비 Aβ1-42 펩티드 투여군의 p-JNK 및 p-P38 발현량이 통계적으로 유의미하게 증가하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 p-JNK 및 p-p38 발현량이 통계적으로 유의미하게 감소하였다(도 6).
또한, 실험용 마우스들의 뇌에서 활성화된 염증성 사이토카인 Iba-1, GFAP(glial fibrillary acidic protein), TNF-α 및 IL-1β의 발현량을 확인하였는데, 그 결과 도 7에 개시한 바와 같이 대조군 대비 Aβ1-42 펩티드 투여군의 염증성 사이토카인 Iba-1, GFAP, TNF-α 및 IL-1β의 발현량이 통계적으로 유의미하게 증가하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 염증성 사이토카인 Iba-1, GFAP, TNF-α 및 IL-1β의 발현량이 통계적으로 유의미하게 감소하였다
한편, p-JNK 및 p-P38에 대한 면역형광 분석 결과에서도, p-JNK 및 p-P38 발현량의 경우, 대조군 대비 Aβ1-42 펩티드 투여군의 p-JNK 및 p-P38 발현량이 통계적으로 유의미하게 증가하였고, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 p-JNK 및 p-p38 발현량이 통계적으로 유의미하게 감소하였다(도 8).
실시예 4. Aβ1-42 펩티드를 투여하여 유도된 세포사멸에 대한 사이클로아스트라제놀의 효과
세포 사멸은 신경퇴행의 주요한 원인으로, 전구-세포사멸(pro-apoptotic) 인자 Bax 및 항-세포사멸(anti-apoptotic) Bcl-2에 의해 조절된다. Aβ1-42 펩티드로 유도된 세포 사멸에 대한 사이클로아스트라제놀의 효과를 확인하기 위하여, Bcl-2, Bax, Casp-3, 및 Bim에 대한 웨스턴 블롯을 실시하였다.
그 결과, 대조군 대비 Aβ1-42 펩티드 투여군의 대뇌 피질 및 해마에서 전구-세포사멸 단백질 Bcl-2 발현양이 감소하였으며, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 Bcl-2의 발현양이 통계적으로 유의미하게 증가하였다. 대조군 대비 Aβ1-42 펩티드 투여군의 대뇌 피질 및 해마에서 전구-세포사멸 단백질 Bax, Casp-3, 및 Bim 발현양이 증가하였으며, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 Bax, Casp-3, 및 Bim 발현양이 통계적으로 유의미하게 감소하였다(도 9).
또한, 면역형광분석에서도, 대조군 대비 Aβ1-42 펩티드 투여군의 대뇌 피질 및 해마에서 전구-세포사멸 단백질 Caspase-3, 및 Bim 발현양이 증가하였으며, Aβ1-42 펩티드 투여군 대비 본 발명의 Aβ1-42 + 20mg의 사이클로아스트라제놀 투여군의 Caspase-3, 및 Bim 발현양이 통계적으로 유의미하게 감소하였다(도 10).
실시예 5. Aβ1-42 펩티드를 투여하여 유도된 기억력 손상에 대한 사이클로아스트라제놀의 효과
인지기능 이상에 대한 사이클로아스트라제놀의 효과를 확인하기 위하여 MWM(Morris water maze) 분석을 실시하였다.
그 결과, Aβ1-42 펩티드 투여군은 해당 플랫폼에 도달하는데 걸리는 시간이 증가하였고, 이에 대비하여 본 발명의 사이클로아스트라제놀 투여군은 해당 플랫폼에 도달하는게 걸리는 시간이 감소하였고, 프로브 테스트에서 목표 사분면에서 소비하는 시간과 해당 플랫폼을 지나치는 횟수를 확인하였으며, Aβ1-42 펩티드 투여군 대비 본 발명의 사이클로아스트라제놀 투여군은 목표 사분면 및 플랫폼을 지나치는 횟수 및 머무는 시간이 통계적으로 유의미하게 증가하였다(도 11).
Claims (12)
- 제1항에 있어서, 상기 퇴행성 뇌질환은 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 헌팅턴병(Huntington disease), 경도인지장애(mild cognitive impairment), 대뇌 아밀로이드 맥관병증, 아밀로이드성 뇌졸중(stroke), 전신성 아밀로이드병, 노인성 치매, 근위축성 축삭경화증(amyotrophic lateral sclerosis) 및 전두촉두엽 치매(frontotemporal dementia) 중에서 선택된 어느 하나인 것을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 사이클로아스트라제놀은 황기 뿌리(Astragalus radix)로부터 분리된 것을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 퇴행성 뇌질환은 산화스트레스(Oxidative Stress), 신경영양 기능장애(Neurotrophic Dysfunctions), 신경염증(Neuroinflammation) 또는 세포사멸(Apoptotic cell death)에 의한 것임을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 퇴행성 뇌질환은 아밀로이드-베타의 축적에 의한 것임을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 사이클로아스트라제놀은 아밀로이드 베타(Aβ)의 축적에 의해 감소된 Nrf2(Nuclear factor erythroid-2-related factor 2), HO-1(Heme oxygenase 1), BDNF(Brain-derived neurotrophic factor), p-TrkB(p-ropomyosin receptor kinase B), p-CREB(cAMP-response element-binding protein), NeuN(neuronal nuclear protein), p-ERK(phospho-extracellular signal-related kinase) 및 Bcl-2(B cell lymphoma/leukemia-2) 중에서 선택된 하나 이상의 단백질 발현량을 증가시키는 것을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제1항에 있어서, 상기 사이클로아스트라제놀은 아밀로이드 베타(Aβ)의 축적에 의해 증가된 p-JNK(phospho c-Jun-N-terminal kinase), p-P38, Iba-1(ionized calcium-binding adapter molecule-1), GFAP(Glial Fibrillary Acidic Protein), TNF-α(tumour necrosis factor-alpha), IL-1β(interleukin 1-beta), Bax, Cleaved Caspase-3 및 Bim 중에서 선택된 하나 이상의 단백질 발현량을 감소시키는 것을 특징으로 하는 퇴행성 뇌질환의 예방 또는 치료용 약학 조성물.
- 제9항에 있어서, 상기 퇴행성 뇌질환은 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 헌팅턴병(Huntington disease), 경도인지장애(mild cognitive impairment), 대뇌 아밀로이드 맥관병증, 아밀로이드성 뇌졸중(stroke), 전신성 아밀로이드병, 노인성 치매, 근위축성 축삭경화증(amyotrophic lateral sclerosis) 및 전두촉두엽 치매(frontotemporal dementia) 중에서 선택된 어느 하나인 것을 특징으로 하는 퇴행성 뇌질환의 예방 또는 개선용 건강기능식품 조성물.
