본 발명은 하기 화학식 1의 화합물, 그의 약제학적으로 허용되는 염 또는 이성체, 및 이를 유효성분으로 포함하는 대사성 질환의 치료 또는 예방용 약제학적 조성물에 관한 것이다. Description
본 발명은 케토헥소카이네이즈(Ketohexokinase, KHK)의 저해 활성을 갖는 신규한 3-아자바이사이클로알킬 유도체 화합물 및 이를 유효성분으로서 함유하는 약제학적 조성물에 관한 것이다.
대사성 질환 중 하나인 비알콜성 지방간염의 발병 원인은 크게 지방간 생성, 염증 증가, 세포 사멸 등으로 알려져 있다. 만성질환으로서 비알콜성 지방간염은 점차 간섬유화, 간경화, 간암으로 진행될 수 있다.
한편, 케토헥소카이네이즈(Ketohexokinase, KHK)는 과당(fructose) 대사에 관여하는 효소로서, 과당 대사 중 과당의 인산화를 담당하는 카이네이즈(kinase)의 한 종류이다. 과당 대사는 포도당 대사와 달리 에너지 의존적인 저해를 받지 않아 간의 빠른 지방 축적을 유도하여 지방간 생성에 영향을 준다. 따라서, 케토헥소카이네이즈(KHK)를 저해할 경우 비알콜성 지방간염의 발병 원인 중의 하나인 지방간 생성을 억제하는 효과를 기대할 수 있다.
동물 실험 모델에서도 케토헥소카이네이즈(KHK) 활성을 억제하였을 경우 대사 질환에 대한 전반적인 지표들이 개선되는 것을 관찰할 수 있었고, 케토헥소카이네이즈(KHK) 녹아웃(knock out) 마우스의 표현형이 정상 마우스와의 차이를 보이지 않아 케토헥소카이네이즈(KHK) 저해로 인한 큰 부작용은 없을 것으로 예상된다.
다만, 케토헥소카이네이즈(KHK)가 ATP를 이용하여 인산화를 담당한다는 점에서 케토헥소카이네이즈(KHK)를 저해할 경우 다른 종류의 카이네이즈를 저해할 가능성이 있어 만성 질환 치료제로서의 안전성 측면에서 많은 우려를 가져온 것이 사실이다. 따라서, 케토헥소카이네이즈(KHK) 저해제는 다른 카이네이즈와 선택성을 가지는 것이 중요하다.
케토헥소카이네이즈(KHK)의 저해제에 관한 연구는 현재 활발히 진행되고 있는데, 당뇨병, 당뇨병의 합병증, 비만, 비알콜성 지방간, 지방성 간염 등의 대사성 질환의 예방 또는 치료에 유용하게 사용될 수 있는 치료제의 개발이 여전히 필요한 실정이다.

또한, 약제학적으로 허용되는 카복실산 염에는, 예를 들어 리튬, 나트륨, 칼륨, 칼슘, 마그네슘 등에 의해 형성된 알칼리 금속 또는 알칼리 토금속 염, 라이신, 아르기닌, 구아니딘 등의 아미노산 염, 디사이클로헥실아민, N-메틸-D-글루카민, 트라이스(하이드록시메틸) 메틸아민, 디에탄올아민, 콜린, 트라이에틸아민 등과 같은 유기염 등이 포함된다. 본 발명에 따른 화학식 1의 화합물은 통상적인 방법에 의해 그의 염으로 전환될 수 있다.

본 명세서에서 용어 "치료" 란 발병 증상을 보이는 대상자(subject)에 사용될 때 질병의 진행을 중단, 지연 또는 완화시키는 것을 의미하며, "예방"이란 발병 증상을 보이지는 않지만 그러한 위험성이 높은 대상자에 사용될 때 발병 징후를 중단, 지연 또는 완화시키는 것을 의미한다.
본 발명의 화학식 1의 화합물, 이의 약제학적으로 허용되는 염 또는 이성체의 투여량은 환자의 체중, 나이 및 질병의 특수한 성질과 심각성과 같은 요인에 따라 의사의 처방에 따른다. 그러나, 성인 치료에 필요한 투여량은 투여의 빈도와 강도에 따라 하루에 약 1 내지 500mg 범위가 보통이다. 성인에게 근육 내 또는 정맥 내 투여 시 일회 투여량으로 분리하여 하루에 보통 약 1 내지 300mg의 전체 투여량이면 충분할 것이나, 일부 환자의 경우 더 높은 일일 투여량이 바람직할 수 있다.

[제조예 1] 1,3-디클로로-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴의 제조
하기 단계 A 및 B의 과정을 거쳐 제조예 1의 화합물을 제조하였다.
(단계 A) 1,3-디하이드록시-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴의 제조
에틸 2-옥소사이클로펜탄-1-카르복실레이트(4.0g, 25.6mmol), 2-사이아노아세타마이드(2.15g, 25.6mmol)와 포타슘 하이드록사이드(1.58g, 28.2mmol)를 메탄올 (100ml)에 녹여서 12시간 동안 환류교반하였다. 반응용액을 감압 농축한 후, 1N 염산 용액을 첨가하였다. 생성된 고체를 여과하고 물로 씻어준 후, 질소를 이용하여 건조시켜 1,3-디하이드록시-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴(1.5g, 8.51mmol, 33%)을 얻었다.
(단계 B) 1,3-디클로로-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴의 제조
단계 A에서 얻은 1,3-디하이드록시-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴(1.5g, 8.51mmol, 벤질트라이에틸암모늄클로라이드(3.6g, 15.9mmol)와 포스포릴 트라이클로라이드(3.7ml, 39.7mmol)를 100℃ 반응 온도에서 5시간 동안 반응하였다. 반응용액을 감압 농축한 후, 물을 첨가하고 에틸 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/5)로 정제하여 1,3-디클로로-6,7-디하이드로-5H-사이클로펜타[c]피리딘-4-카보나이트릴(1.0g, 4.69mmole, 55%)을 얻었다.

제조예 1에 기재된 방법으로, 에틸 2-옥소사이클로펜탄-1-카르복실레이트 대신에 에틸 4-옥소테트라하이드로-2H-피란-3-카르복실레이트(2.0g, 11.6mmol)를 이용하여 6,8-디클로로-3,4-디하이드로-1H-피라노[3,4-c]피리딘-5-카보나이트릴(200mg, 0.87mmol, 7.5%)을 얻었다.

하기 단계 A, B, C, D의 과정을 거쳐 제조예 5의 화합물을 제조하였다.
(단계 A) 메틸 (5S)-5-메틸-4-옥소테트라하이드로퓨란-3-카르복실레이트의 제조
메틸 (S)-2-하이드록시프로피오네이트(42.0g, 38.4mmol), 소듐 하이드라이드(1.54g, 60%, 38.4mmol)를 테트라하이드로퓨란(100ml)에 녹여 0℃에서 30분간 교반한 후, 감압 농축하였다.
농축된 물질에 메틸 아크릴레이트(3.46ml, 38.4mmol)와 디메틸설폭사이드(30ml)를 넣고, 상온에서 12시간 동안 교반하여 반응을 완결하였다. 완결된 반응 용액에 1N 염산 용액을 넣고 에틸아세테이트로 추출하였다. 포화 소듐클로라이드로 세척하고 무수 마그네슘설페이트로 건조한 다음 필터한 여액을 감압증류하였다. 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/5)로 정제하여 메틸 (5S)-5-메틸-4-옥소테트라하이드로퓨란-3-카르복실레이트(2.0g, 12.7mmol, 33%)를 얻었다.

단계 B에서 얻은 메틸 (S)-5-메틸-4-우레이도-2,5-디하이드로퓨란-3-카르보실레이트(2.0g, 9.99mmol)를 28% 암모니아수(20ml)에 녹여, 60℃에서 12시간 동안 교반하였다. 반응이 종결된 후, 감압 농축하였다. 농축된 물질에 1N 염산 용액을 넣고 에틸아세테이트로 추출하였다. 포화 소듐클로라이드로 세척하고 무수 마그네슘설페이트로 건조한 다음 필터한 여액을 감압증류하였다. 컬럼 크로마토그래피(메탄올/디클로로메탄= 1/9)로 정제하여 (S)-7-메틸-5,7-디하이드로푸로[3,4-d]피리미딘-2,4-디올(1.0g, 5.95mmol, 60%)을 얻었다

에틸 2-옥소사이클로펜탄-1-카르복실레이트(4.0g, 25.6mmol), 소듐 하이드라이드(1.0g, 60%, 25.6mmol)를 테트라하이드로퓨란(100ml)에 녹여 0℃에서 30분간 교반한 후, 감압 농축하였다. 반응용액에 니트로소벤젠(2.88g, 26.9mmol)을 첨가하고, 0℃에서 30분 동안 교반하여 반응을 완료시켰다. 완료된 반응 용액에 1N 염산 용액을 넣고 에틸아세테이트로 추출하였다. 포화 소듐 클로라이드 수용액으로 세척하고 무수 마그네슘설페이트로 건조한 다음 필터한 여액을 감압증류하여 6-에톡시-5,6-디옥소헥사노익산(3.6g, 19.1mmol)을 얻었다. 얻어진 물질은 정제없이 다음 반응에 사용하였다.

단계 A에서 얻은 6-에톡시-5,6-디옥소헥사노익산(3.6g, 19.1mmol)을 에테르(100ml)에 녹인 후, 0℃에서 0.15M 디아조메탄 에테르 용액(127ml, 19.1mmol)을 서서히 적가하였다. 반응이 완결된 용액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산=1/2)로 정제하여 1-에틸-6-메틸 2-옥소헥산디오에이트(2.7g, 13.4mmol, 52%, 2단계 수율)를 얻었다.

단계 B에서 얻은 1-에틸-6-메틸 2-옥소헥산디오에이트(2.8g, 13.4mmol)와 디에틸아미노설퍼 트리플루오라이드(6.7g, 41.5mmol)를 클로로포름(150ml)에 녹여 0℃에서 3시간 동안 교반하였다. 완결된 반응용액에 과량의 물을 첨가하고 에틸 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산=1/10)로 정제하여 1-에틸-6-메틸 2,2-디플루오로헥산디오에이트(0.9g, 4.01mmol, 30%)를 얻었다.

단계 C에서 얻은 1-에틸-6-메틸 2,2-디플루오로헥산디오에이트(0.9g, 4.01mmol)와 포타슘 tert-부톡사이드(473mg, 4.21mmol)를 테트라하이드로퓨란(100ml)에 녹여 상온에서 8시간 동안 교반하였다. 완결된 반응용액에 1N 염산 용액을 넣고 에틸 아세테이트로 추출하였다. 유기층을 포화 소듐 클로라이드 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하여 메틸 2-에톡시-3,3-디플루오로-2-하이드록시사이클로펜탄-1-카르복실레이트를 얻었다. 얻어진 물질은 정제없이 다음 반응에 사용하였다.
(단계 E) 7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-올의 제조
단계 D에서 얻은 메틸 2-에톡시-3,3-디플루오로-2-하이드록시사이클로펜탄-1-카르복실레이트(540mg, 2.41mmol), 메틸이소티오우레아 반황산염(672mg, 4.82mmol)과 소듐 카보네이트(767mg, 7.24mmol)를 물(50ml)에 녹여, 상온에서 12시간 동안 교반하였다. 완결된 반응용액에 과량의 물을 첨가하고 에틸 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피 (에틸아세테이트/헥산=1/1)로 정제하여 7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-올(250mg, 1.15mmol, 48%)을 얻었다.

단계 E에서 얻은 7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-올(200mg, 1.15mmol), 포스포릴 트라이클로라이드(1.1ml, 11.5mmol)와 트라이에틸아민(0.3ml, 2.3mmol)을 디클로로에탄(30ml)에 녹인 후, 120℃ 반응 온도에서 2시간 동안 반응시켰다. 반응용액을 감압 농축한 후, 물을 첨가하고 에틸 아세테이트로 추출하였다. 유기층을 포화 염화 소듐 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피 (메탄올/디클로로메탄=1/19)로 정제하여 4-클로로-7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘(130mg, 0.55mmol, 48%)을 얻었다.

[제조예 7] 4-클로로-8,8-디플루오로-2-(메틸티오)-5,6,7,8-테트라하이드로퀴나졸린의 제조
하기 과정을 거쳐 제조예 7 화합물을 제조하였다.
제조예 6의 단계 A 내지 단계 E에 기재된 방법으로, 에틸 2-옥소사이클로펜탄-1-카르복실레이트 대신에 에틸 2-옥소사이클로헥산-1-카르복실레이트(20.0g, 117.5mmol)를 이용하여 8,8-디플루오로-2-(메틸티오)-5,6,7,8-테트라하이드로퀴나졸린-4-올(6.25g, 26.9mmol, 23%, 4단계)을 얻었다.
8,8-디플루오로-2-(메틸티오)-5,6,7,8-테트라하이드로퀴나졸린-4-올(3.0g, 12.9mmol), 포스포릴 트라이클로라이드(12.8ml, 129.2mmol)와 트라이에틸아민(0.89ml, 6.45mmol)을 디클로로에탄(200ml)에 녹인 후, 100℃ 반응 온도에서 1시간 동안 반응시켰다. 반응용액을 감압 농축한 후, 물을 첨가하고 에틸 아세테이트로 추출하였다. 유기층을 포화 염화 소듐 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(헥산/에틸아세테이트=2/1)로 정제하여 4-클로로-8,8-디플루오로-2-(메틸티오)-5,6,7,8-테트라하이드로퀴나졸린(2.43g, 9.72mmol, 75%)을 얻었다.

(단계 A) 에틸 2-(3-(3-클로로-4-시아노-6,7-디하이드로-5H-사이클로펜타[c]피리딘-1-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트의 제조
제조예 1의 단계에서 얻은 1,3-디클로로-6,7-디하이록시-5H-사이클로펜타[c]피리딘-4-카보나이트릴(100mg, 0.47mmol), 에틸 2-(3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트 하이드로클로라이드(97mg, 0.47mmol)와 디이소프로필에틸아민(0.33ml, 1.88mmol)을 다이옥산(10ml)에 녹인 후, 50℃에서 8시간 동안 교반하였다. 반응이 완결된 후, 1N 염산 수용액을 첨가하고 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/5)로 정제하여 에틸 2-(3-(3-클로로-4-시아노-6,7-디하이드로-5H-사이클로펜타[c]피리딘-1-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트 (60mg, 0.17mmol, 37%)를 얻었다.

하기 단계 A, B, C, D의 과정을 거쳐 실시예 8의 화합물을 제조하였다.
(단계 A) 에틸 2-(3-(7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트의 제조
제조예 6에서 얻은 4-클로로-7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘(130mg, 0.55mmol), 에틸 2-(3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트 하이드로클로라이드(121mg, 0.55mmol) 및 디이소프로필에틸아민(0.29ml, 1.65mmol)을 다이옥산(10ml)에 녹인 후, 50℃에서 8시간 동안 교반하였다. 반응이 완결된 후, 1N 염산 수용액을 첨가하고 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/3)로 정제하여 에틸 2-(3-(7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트(170mg, 0.44mmol, 81%)를 얻었다.

단계 A에서 얻은 에틸 2-(3-(7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트(170mg, 0.44mmol)와 3-클로로벤조퍼옥시산(248mg, 1.11mmol)을 디클로로메탄(50ml)에 녹인 후, 상온에서 3시간 동안 교반하였다. 반응이 완결된 후, 탄산 수소 소듐 수용액을 넣고 디클로로메탄으로 추출하였다. 유기층을 염화 소듐 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/2)로 정제하여 에틸 2-(3-(7,7-디플루오로-2-(메틸설포닐)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트 (180mg, 0.43mmol, 98%)를 얻었다.
(단계 C) 에틸 2-(3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트의 제조
단계 B에서 얻은 에틸 2-(3-(7,7-디플루오로-2-(메틸설포닐)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트(180mg, 0.43mmol), (S)-2-메틸아제티딘 하이드로클로라이드(70mg, 0.65mmol)와 포타슘 카보네이트(180mg, 1.30mmol)를 N-메틸프롤리딘(15ml)에 녹인 후, 120℃에서 4시간 동안 교반하였다. 반응이 완결된 후, 1N 염산 수용액을 첨가하고 아세테이트로 추출하였다. 유기층을 염화 소듐 수용액, 탄산 수소 소듐 수용액, 그리고 물로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/2)로 정제하여 에틸 2-(3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세테이트(60mg, 0.15mmol, 34%)를 얻었다.

하기 단계 A, B, C, D의 과정을 거쳐 실시예 9의 화합물을 제조하였다.
(단계 A) 4-클로로-7,7-디플루오로-2-(메틸설포닐)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘의 제조
제조예 6 에서 얻은 4-클로로-7,7-디플루오로-2-(메틸티오)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘(1.67g, 6.97mmol)을 디클로로메탄에 녹인 후 70wt% mCPBA(4.30g, 17.43mmol)을 첨가하여 상온에서 2시간 교반하였다. 반응이 완결된 후 포화 소듐 메타바이설파이트 용액을 넣고 30분간 교반하고 포화 소듐 바이카보네이트를 넣어주고 디클로로메탄으로 추출하였다. 유기층을 염화 소듐 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하여 농축하였다. 농축 후 얻은 화합물(1.74g, 6.48mmol)을 별도의 정제 과정 없이 다음 반응에 사용하였다.

단계 B 에서 얻은 화합물(1.32g, 3.29mmol)을 NMP에 녹인 후 포타슘 카보네이트(2.272g, 16.44mmol)를 넣고, (S)-메틸아제티딘 하이드로클로라이드를 NMP에 녹여서 넣어주었다. 반응물을 120℃에서 4시간 동안 교반하였다. 반응이 완결되면 상온으로 낮춰주고, 암모늄클로라이드 수용액과 에틸아세테이트를 넣어주고 몇 분간 교반하고 추출하였다. 유기층을 암모늄 클로라이드 수용액과 소듐 클로라이드 수용액으로 씻어주고, 무수 황산 마그네슘을 넣어 건조 후에 여과하였다. 여과액을 감압 농축하여 농축된 화합물을 컬럼 크로마토그래피(헵탄/에틸아세테이트=1/1)로 정제하여 에틸 2-((1R,5S,6S)-3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세테이트(770mg, 59%)를 얻었다.
(단계 D) 2-((1R,5S,6S)-3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산의 제조
단계 C에서 얻은 화합물(305mg, 0.77mmol)을 테트라하이드로퓨란(8ml)에 녹이고, 리튬 하이드록사이드 모노하이드레이트(65mg, 1.55mmol)를 넣어주었다. 물(4ml)과 메탄올(2ml)을 순서대로 넣어주고 반응물을 18시간 동안 교반하였다. 반응이 완결되면 1N HCl을 1.5ml를 넣어주고 암모늄 클로라이드 수용액과 에틸 아세테이트를 넣어 추출하였다. 유기층을 소듐 클로라이드로 씻어주고 무수 황산 마그네슘으로 건조하고 여과하였다. 여과액을 감압 농축하고 농축된 화합물을 컬럼 크로마토그래피(MC/MeOH=10/1)로 정제하여 2-((1R,5S,6S)-3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산(262mg, 93%)을 얻었다.

4-클로로-8,8-디플로로-2(메틸티오)-5,6,7,8-테트라하이드로퀴나졸린(500mg, 2.00mmol)과 3-클로로벤조퍼옥시산(862mg, 5.00mmol)을 디클로로메탄(30ml)에 녹인 후, 상온에서 18시간 동안 교반하였다. 반응이 완결된 후, 메타중아황산 나트륨 수용액을 넣고 디클로로메탄으로 추출하였다. 유기층을 탄산수소 나트륨 수용액으로 씻어준 후, 무수 황산 마그네슘으로 건조하고 여과하였다. 여액을 감압 농축하고 컬럼 크로마토그래피(에틸아세테이트/헥산 = 1/2)로 정제하여 4-클로로-8,8-디플루오로-2-(메틸설포닐)-5,6,7,8-테트라하이드로퀴나졸린(527 mg, 1.86 mmol, 93%)을 얻었다.

2-((1R,5S,6S)-3-(8,8-디플루오로-2-(피롤리딘-1-일)-5,6,7,8-테트라하이드로퀴나졸린-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산
실시예 11에 기재된 방법으로, (S)-2-메틸아제티딘 하이드로클로라이드 대신에 피롤리딘을 이용하여 2-((1R,5S,6S)-3-(8,8-디플루오로-2-(피롤리딘-1-일)-5,6,7,8-테트라하이드로퀴나졸린-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산(17mg, 0.044mmol, 54%, 2단계)을 얻었다.
1H-NMR (400MHz, CDCl3) δ 4.01(d, J=11.0Hz, 2H), 3.54(m, 6H), 3.48(s, 1H), 2.61(m, 2H), 2.33(d, J=7.3Hz, 2H), 2.22(m, 2H), 1.91(m, 4H), 1.80-1.85(m, 2H), 1.48(s, 2H), 0.98-1.03(m, 1H)
[실험예] 케토헥소카이네이즈 저해제 in vitro 활성 측정
대장균에서 케토헥소카이네이즈를 발현하고 His tag를 이용하여 정제하였다. 정제된 케토헥소카이네이즈의 활성을 측정하기 위해 BellBrook Labs의 Transcreener ADP2 TR-FRET Red Assay kit를 이용하였다. 적정 농도의 케토헥소카이네이즈와 ATP, 과당(fructose)이 포함된 용액을 15분 반응시킨 후 TR-FRET을 이용하여, 생성된 ADP의 양을 측정하는 방법이다. 케토헥소카이네이즈 저해제의 활성을 보기 위해서는 케토헥소카이네이즈와 적정 농도의 저해제를 30분간 우선 반응시킨 후, 케토헥소카이네이즈의 기질이 포함된 용액과 15분 반응시켰다. 그리고 TR-FRET을 위한 반응을 1시간 시킨 후 PerkinElmer의 Envision 기기를 이용하여 형광을 측정하였다. BellBrook Labs에서 발행한 TR-FRET의 최적화(optimization) 절차에 따라 Envision 기기의 설정값을 결정하였다.
케토헥소카이네이즈 저해제의 각 농도별 결과 값은 665nm 파장과 615nm 파장의 비율로 결정된다. 이때, 665/615 비율 값은 ADP 표준 곡선(standard curve)을 바탕으로 ADP 농도로 환산하였고, 통계 소프트웨어(Prizm)에서 모리슨 방정식(Morrison equation)을 사용하여 저해 활성 Ki값을 구하였다.
상기 실험을 통해 얻어진 실시예 화합물의 Ki는 하기 표 1에 나타내었다.

Ki값은 저해제와 효소의 결합 친화성(binding affinity)을 나타내는 값으로, 낮은 값일수록 좋은 저해 활성을 보인다. 따라서, 상기 결과로부터, 본 발명의 실시예의 화합물이 우수한 저해 활성을 나타내는 것을 확인할 수 있다.
본 발명에 따른 화학식 1의 3-아자바이사이클로알킬 유도체 화합물은 케토헥소카이네이즈(KHK)를 저해하는 활성을 가지며, 당뇨병, 당뇨병의 합병증, 비만, 비알콜성 지방간, 지방성 간염 등의 대사성 질환의 예방 또는 치료에 효과적으로 사용할 수 있다.
Claims (9)
- 청구항 1에 있어서,하기 군에서 선택되는 화합물, 그의 약제학적으로 허용되는 염 또는 이성체:2-((1R,5S,6S)-3-(4-시아노-3-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[c]피리딘-1-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-(4-시아노-3-((S)-2-메틸아제티딘-1-일)-5,6,7,8-테트라하이드로아이소퀴놀린-1-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-(3-(4-시아노-3-((S)-2-메틸아제티딘-1-일)-6,7,8,9-테트라하이드로-5H-사이클로헵타[c]피리딘-1-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-(5-시아노-6-((S)-2-메틸아제티딘-1-일)-3,4-디하이드로-1H-피라노[3,4-c]피리딘-8-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-(3-(7,7-디메틸-2-((S)-2-메틸아제티딘-1-일)-5,7-디하이드로푸로[3,4-d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-((S)-7-메틸-2-((S)-2-메틸아제티딘-1-일)-5,7-디하이드로푸로[3,4-d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-((S)-7-메틸-2-((S)-2-메틸아제티딘-1-일)-5,7-디하이드로푸로[3,4-d]피리미딘-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산;2-((1R,5S,6S)-3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-(7,7-디플루오로-2-((S)-2-메틸아제티딘-1-일)-6,7-디하이드로-5H-사이클로펜타[d]피리미딘-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산;2-((1R,5S,6S)-3-(8,8-디플루오로-2-((S)-2-메틸아제티딘-1-일)-5,6,7,8-테트라하이드로퀴나졸린-4-일)-3-아자바이사이클로[3.1.1]헵탄-6-일)아세트산;2-((1R,5S,6S)-3-(8,8-디플루오로-2-((S)-2-메틸아제티딘-1-일)-5,6,7,8-테트라하이드로퀴나졸린-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산;2-((1R,5S,6S)-3-(8,8-디플루오로-2-(피롤리딘-1-일)-5,6,7,8-테트라하이드로퀴나졸린-4-일)-3-아자바이사이클로[3.1.0]헥산-6-일)아세트산.
- 청구항 1 또는 청구항 2에 있어서,케토헥소카이네이즈(ketohexokinase, KHK) 저해제로 사용되는 화합물, 그의 약제학적으로 허용되는 염 또는 이성체.
- 청구항 1 또는 청구항 2에 있어서,대사성 질환의 예방 또는 치료에 사용하기 위한 화합물, 그의 약제학적으로 허용되는 염 또는 이성체.
- 청구항 1 또는 청구항 2에 따른 화합물, 그의 약제학적으로 허용되는 염 또는 이성체; 및 약제학적으로 허용되는 담체를 포함하는 케토헥소카이네이즈(ketohexokinase, KHK) 저해용 약제학적 조성물.
- 청구항 1 또는 청구항 2에 따른 화합물, 그의 약제학적으로 허용되는 염 또는 이성체; 및 약제학적으로 허용되는 담체를 포함하는 케토헥소카이네이즈(ketohexokinase, KHK) 관련 대사성 질환의 예방 또는 치료용 약제학적 조성물.
- 청구항 6에 있어서,상기 대사성 질환은 당뇨병, 당뇨병의 합병증, 비만, 비알콜성 지방간염, 및 지방성 간염으로 이루어지는 군에서 선택되는 것인, 대사성 질환의 예방 또는 치료용 약제학적 조성물.
- 유효성분으로서 청구항 1 또는 청구항 2에 따른 화합물, 그의 약제학적으로 허용되는 염 또는 이성체를, 약제학적으로 허용되는 담체와 혼합하는 단계를 포함하는 케토헥소카이네이즈(KHK) 관련 대사성 질환의 예방 또는 치료용 약제학적 조성물을 제조하는 방법.
- 청구항 8에 있어서,상기 대사성 질환은 당뇨병, 당뇨병의 합병증, 비만, 비알콜성 지방간염, 및 지방성 간염으로 이루어지는 군에서 선택되는 것인, 약제학적 조성물을 제조하는 방법.
