Abstract
본 발명은 우유 엑소좀을 포함하는 피부 탄력 증진 및/또는 주름 개선용 조성물, 우유 엑소좀을 투여하여 주름을 치료하는 방법에 관한 것이다.

본 발명은 우유 엑소좀을 포함하는 피부 탄력 증진 및/또는 주름 개선용 조성물, 우유 엑소좀을 투여하여 주름을 치료하는 방법에 관한 것이다.
피부는 신체의 외부를 덮고 있는 하나의 막으로 여러 가지 외부의 자극, 장해, 건조 등의 환경요소로부터 신체를 보호해 주는 다양한 생리적 기능을 수행하여 내부 장기와 그 밖의 체내 기관을 보호 조절하는 역할을 한다.
그러나, 피부는 연령이 증가함에 따라 생체의 생리적 기능이 저하되어 신진 대사를 조절하는 각종 호르몬의 분비가 감소하고, 면역 세포의 기능과 세포들의 활성이 저하되게 된다. 또한 광 또는 열에 반복적인 노출로 인해 피부의 외양 또는 기능이 변하게 되는데, 이로 인해 피부에서 콜라겐 합성이 감소하고 수분이 손실되고, 피부 탄력이 감소하면서 피부 노화현상을 촉진하여 피부를 손상시키게 된다. 피부 노화의 대표적인 현상 중 하나가 주름(wrinkles) 생성이다.
이러한 피부노화 및 손상을 방지하여 보다 건강하고 아름다운 피부를 유지하기 위하여 각종 동물, 식물, 미생물 등으로부터 얻어진 생리 활성 물질들을 화장품에 부가하여 사용함으로써 피부의 고유 기능을 유지시키고 피부 세포를 활성화시켜 피부 신진대사를 촉진하여 피부 건강을 유지하고자 하는 노력이 계속되고 있으며, 이에 따른 화장품, 피부 외용제 등이 개발되고 있다.
그러나, 이들 물질은 피부 적용시 자극, 홍반, 발적 등의 안전성 문제로 사용량의 제한이 있거나, 효과가 미미하여 실질적으로 피부 기능 개선 효과를 기대기 어려웠다.
이에 따라, 피부 탄력 증진 및/또는 주름 개선 효과가 우수한 물질에 대한 연구 및 개발이 필요한 실정이다.
본 발명의 목적은 우유 엑소좀을 포함하는, 피부 탄력 증진 및/또는 주름 개선용 조성물을 제공하는 것이다.
본 발명의 다른 목적은 우유 엑소좀을 포함하는, 피부 탄력 증진 및/또는 주름 개선용 화장료 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 우유 엑소좀을 포함하는, 피부 탄력 증진 및/또는 주름 개선용 식품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 우유 엑소좀을 포함하는, 피부 탄력 및/또는 주름 예방 또는 치료용 약학 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 우유 엑소좀을 개체에 투여하는 단계를 포함하는, 주름의 치료방법을 제공하는 것이다.
상기와 같은 목적을 달성하기 위한 본 발명의 일 측면은 우유 엑소좀을 포함하는, 피부 탄력 증진 및 주름 개선용 조성물에 관한 것이다.
본 발명에서, “우유 엑소좀”은 우유 유래의 지질 이중충의 소낭을 말한다. 상기 엑소좀은 우유 유래의 고유의 단백질 및 핵산 등을 포함하고 있으며 30~200 nm 크기의 생체 나노입자로, 내부에 DNA, RNA, 펩타이드 등으로 이루어진 정보가 담겨있다. 엑소좀은 내부의 물질을 체액 내 분해 효소 등으로부터 안전하게 운반하여 인접 세포 또는 원거리 세포에 정보 전달을 하여 세포 주변의 미세 환경에 영향을 미친다. 특히, 우유 유래의 우유 엑소좀은 식품으로도 접할 수 있을 뿐만 아니라 많은 양의 엑소좀이 추출되며, 긴 시간 동안 안정한 상태로 보관할 수 있다는 장점이 있다.
한편 종래 엑소좀은 주로 바이오마커 또는 운반체로 종종 활용되어왔으나, 최근 모유 뿐 아니라 다양한 낙농 유제품 제조에 광범위하게 사용되고 있는 우유 엑소좀 안에 다양한 small RNA가 존재하며, 상기 유전물질이 면역기능 관여 또는 파골세포 생성과정 관여하여 골다공증 치료 및 개선에 유용하다는 것이 밝혀졌다(Mizoguchi et al., 2010). 이에 본 발명자들은 우유 엑소좀에 관하여 연구를 진행하였으며, 상기 우유 엑소좀이 피부 탄력 증진 및 주름 개선과 관련된 유전자 발현 조절, 콜라겐 합성 및 콜라게네아제와 엘라스테아제 활성 억제 등에 관여함을 처음으로 규명하였다.
이에 본 발명에서는 소에서 유래한 우유로부터 원심분리하여 엑소좀을 수득하였으며, 상기 우유 엑소좀을 피부 탄력 증진 및 주름 개선 용도에 활용하고자 한다. 구체적으로, 상기 엑소좀은 소 유래인 것일 수 있다.
본 발명에서, “피부 탄력 증진”이란 피부 노화에 의해서 피부 탄력이 떨어지는 생기는 주름을 개선시키는 것을 일컫는다. 즉, 피부 탄력이 떨어지는 경우 피부 내 콜라겐 섬유 감소 및 변성으로 인하여 표피의 아래 근육이 접히고 펴지는 운동이 반복되게 되며, 이로인하여 피부 주름이 생기게 된다. 이에 저하된 피부 탄력을 개선시킴으로써 주름 생성을 방지 및 억제하는 것을 모두 포함한다.
본 발명에서 "피부 주름"이란, 피부가 쇠하여 생긴 잔줄을 의미하는데, 유전자에 의한 원인, 피부 진피에 존재하는 콜라겐과 엘라스틴의 감소, 외부환경 등에 의해 유발될 수 있다. 이에 상기 "피부 주름 개선"이란, 피부에 주름이 생성되는 것을 억제 또는 저해하거나, 이미 생성된 주름을 완화시키는 것을 의미한다.
본 발명의 일 실시예에서는 피부 탄력 및/또는 주름 관련 유전자의 발현 양상을 확인한 결과, MMP-1 유전자의 발현량은 감소하는 한편, TIMP1, 프로콜라겐(procollagen), CD34, 엘라스틴(Elastin)의 유전자 발현량은 모두 증가함을 확인하였다(도 4). 상기와 같은 결과는 우유 엑소좀이 피부 탄력 증진 및 주름관련 유전자 발현을 조절함으로써, 피부 탄력 증진 및 주름 개선용 조성물로 활용할 수 있음을 시사한다.
상기 우유 엑소좀은 엘라스테아제 활성 및 콜라게네아제 활성을 저해시킬 수 있다.
상기 엘라스테아제는 진피 내 피부 탄력을 유지하는 기질 단백질인 엘라스틴(elastin) 분해에 관여하여 체내의 엘라스틴을 분해하는 백혈구 과립 효소 중의 하나이다. 상기 효소의 활성이 높아질 경우 피부 조직 파괴에 직접적인 원인이 되므로, 엘라스테아제는 피부의 주름 및 탄력성 저하의 원인이 되는 것으로 알려져 있다. 본 발명의 일 실시예에서 상기 우유 엑소좀을 처리할 경우 엘라스테아제의 활성을 현저히 억제시킴을 확인하였다(표 6).
상기 콜레게네아제는 콜라겐을 분해하는 효소 중 하나로서, 콜라겐 type I을 분해하는 matrix metalloproteinase I(MMP-1)로도 알려져 있다. 본 발명의 일 실시예에서는 상기 우유 엑소좀을 처리하여 콜라게네아제의 저해활성을 측정한 결과, 우유 엑소좀 미처리군에 비해 콜라게네아제 활성이 현저히 저해됨을 확인하였다(표 7).
이에, 우유 엑소좀이 피부 탄력 저하 및 주름 생성에 직접적 원인이되는 것으로 알려져 있는 엘라스테아제 및 콜라게네아제 효소의 활성을 저해 또는 억제시킴으로써 피부 탄력 증진 및 주름 개선 용도로 활용할 수 있음을 확인하였다.
또한, 구체적으로 상기 조성물은 노화 개선 용도를 추가로 더 포함할 수 있다.
본 발명에서 “노화”란 내인성 노화와 외인성 노화로 나누어지는데, 내인성 노화는 시간 흐름에 따라 자연스럽게 신체기능이 저하되어 나타나는 현상이며, 외인성 노화는 햇빛에 의한 광노화, 그 외에 흡연, 음주, 스트레스 등의 요인에 의해 생기는 노화를 모두 포함한다.
상기 노화는 구체적으로 광노화인 것일 수 있다.
상기 광노화는 자외선이 피부 세포 내 신호 전달체계에 영향을 미쳐 DNA 손상과 유전자 발현을 변화시켜 세포 노화 및 세포사멸에 의해 유발된다. 자외선이 피부세포에 지속적으로 노출될 경우, 진피층의 섬유아세포에서는 활성산소가 증가하고 사이토카인 transforming growth factor-beta(TGF-β)의 발현은 감소하고, activator protein 1(AP-1)의 발현이 활성화되면서 노화를 일으킨다.
예컨대, TGF-β는 콜라겐의 합성을 촉진하는 사이토카인으로서, 자외선에 의해 TGF-β의 발현이 감소할 경우 콜라겐 합성이 저해되고, 반면 AP-1은 MMP를 조절하여 콜라겐 분해를 촉진시킴으로써 콜라겐 분해를 촉진시킨다.
본 발명의 일 실시예에서는 우유 엑소좀이 노화에 의해 저해된 콜라겐 생합성량을 증가시키고(도 5), 콜라게네아제 및 엘라스테아제 활성 억제를 통해 콜라겐 분화를 막을 수 있음을 확인하였다(도 7).
본 발명의 일 실시예에서 자외선 UVB를 처리하여 광노화를 유발한 후 우유 엑소좀을 처리한 결과, 우유 엑소좀을 처리하지 않은 대조군에 비해 콜라겐 섬유양 감소가 거의 없으며, 섬유 밀도가 조밀하고 배열이 규칙적임을 확인하였다(도 8).
상기와 같은 결과는 우유 엑소좀은 신체기능 저하에 따라 피부 탄력 저하로 유발되는 노화 뿐 아니라, 자외선에 의한 외인성 노화 방지 및 개선에도 우수한 효과가 있음을 시사한다.
본 발명의 다른 목적은 우유 엑소좀을 포함하는, 피부 탄력 증진 및 주름 개선용 화장료 조성물에 관한 것이다.
상기 '우유 엑소좀', '피부 탄력 증진' 및 '주름 개선'에 관한 설명은 전술한 바와 동일하다.
구체적으로 상기 우유 엑소좀은 소 유래인 것일 수 있다.
본 발명의 일 실시예에서 소 유래 우유 엑소좀을 인간진피섬유아세포에 처리한 결과 세포독성이 거의 없음을 확인하였는 바(도 3), 우유 엑소좀을 화장료 조성물에 활용할 수 있음을 확인하였다.
본 발명의 일 실시예에서는 우유 엑소좀이 피부 탄력 및 주름 관련 유전자의 발현을 조절하고(도 4), 콜라겐 합성량을 증가시키며(도 5), 특히 엘라스테아제 및 콜라게네아제 효소 활성 저해에 우수한 효과가 있음을 확인하였는 바(도 6 및 도 7), 피부 탄력 증진 및 주름 개선용 화장료 조성물로 활용할 수 있음을 확인하였다.
또한 구체적으로, 상기 화장료 조성물은 노화 개선 용도를 추가로 더 포함할 수 있다. 구체적으로 상기 노화는 광노화일 수 있다.
본 발명의 일 실시예에서 우유 엑소좀이 자외선 UVB 처리시에도 콜라겐 감소가 거의 없으며, 섬유 밀도가 조밀하고 배열이 규칙적임을 확인하였는 바(도 8), 상기 결과를 통해 우유 엑소좀이 자외선에 의한 노화 개선용 화장료 조성물로 활용할 수 있음을 확인하였다.
본 발명의 화장료 조성물은 용액, 외용 연고, 크림, 폼, 영양 화장수, 유연 화장수, 팩, 유연수, 유액, 메이크업 베이스, 에센스, 비누, 액체 세정료, 입욕제, 선 스크린 크림, 선 오일, 현탁액, 유탁액, 페이스트, 겔, 로션, 파우더, 비누, 계면 활성제-함유 클렌징, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 패취 및 스프레이로 구성된 군으로부터 선택되는 제형으로 제조할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 화장료 조성물은 일반 피부 화장료에 배합되는 화장품학적으로 허용 가능한 담체를 1 종 이상 추가로 포함할 수 있으며, 통상의 성분으로 예를 들면 유분, 물, 계면 활성제, 보습제, 저급 알코올, 증점제, 킬레이트제, 색소, 방부제, 향료 등을 적절히 배합할 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 화장료 조성물에 포함되는 화장품학적으로 허용 가능한 담체는 화장료 조성물의 제형에 따라 다양하다.
본 발명의 제형이 연고, 페이스트, 크림 또는 젤인 경우에는, 담체 성분으로서 동물성 유, 식물성 유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크, 산화 아연 등이 이용될 수 있으나, 이에 제한되는 것은 아니다. 이들은 단독으로 사용되거나 2 종 이상 혼합되어 사용될 수 있다.
본 발명의 제형이 파우더 또는 스프레이인 경우에는, 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록사이드, 칼슘 실케이트, 폴리아미드 파우더 등이 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로하드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진제를 포함할 수 있으나, 이에 제한되는 것은 아니다. 이들은 단독으로 사용되거나 2 종 이상 혼합되어 사용될 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는, 담체 성분으로서 용매, 용해화제 또는 유탁화제 등이 이용될 수 있으며, 예컨대 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌 글리콜, 1,3-부틸글리콜 오일 등이 이용될 수 있고, 특히, 목화씨 오일, 땅콩 오일, 옥수수 배종 오일, 올리브 오일, 피마자 오일 및 참깨 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 이용될 수 있으나, 이에 제한되는 것은 아니다. 이들은 단독으로 사용되거나 2 종 이상 혼합되어 사용될 수 있다.
본 발명의 제형이 현탁액인 경우에는, 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타하이드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있으나, 이에 제한되는 것은 아니다. 이들은 단독으로 사용되거나 2 종 이상 혼합되어 사용될 수 있다.
또한, 본 발명의 조성물은 피부에 직접 도포하거나 살포하는 등의 경피 투여 방법으로 사용될 수 있으며, 본 발명 조성물의 투여 경로는 목적조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다.
본 발명의 조성물의 사용량은 연령, 병변의 정도 등의 개인 차이나 제형에 따라 적절하게 조절될 수 있으며, 1일 1회 내지 수회 적댱량을 피부에 도포하여 1 주일 내지 수개월 사용될 수 있다.
본 발명의 또 다른 측면은 우유 엑소좀을 포함하는, 피부 탄력 증진 및 주름 개선용 식품 조성물에 관한 것이다.
상기 '우유 엑소좀', '피부 탄력 증진' 및 '주름 개선'에 관한 설명은 전술한 바와 동일하다.
상기 식품의 종류에는 특별한 제한은 없다. 본 발명의 우유 엑소좀을 첨가할 수 있는 식품은 소세지, 육류, 빵, 초콜릿류, 스넥류, 캔디류, 과자류, 라면, 피자, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합제 등이 있다. 음료수로 제형화할 경우에 본 발명의 우유 엑소좀 외에 첨가되는 액체 성분으로는 이에 한정되지는 않으나, 통상의 음료와 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다. 상술한 천연 탄수화물은 모노사카라이드(예, 포도당, 과당 등), 디사카라이드(예, 말토오스, 수크로오스 등) 및 폴리사카라이드(예, 덱스트린, 시클로덱스트린 등과 같은 통상적인 당), 및 자일리톨, 소르비톨, 에리스리톨 등의 당 알코올일 수 있다.
상기 식품의 종류는 구체적으로 건강기능식품일 수 있다. 보다 구체적으로 피부탄력 증진 및 주름 개선용 건강기능식품일 수 있다.
상기 건강기능 식품은 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 증진제 (치즈, 초콜릿 등), 펙트산 및 그의 염, 유기산, 보호성 콜로이드 점증제, pH 조절제, 안정화제, 보존제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 또한, 본 발명의 건강기능 식품은 과일 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 단독으로 또는 조합으로 사용될 수 있으며, 이러한 첨가제의 비율은 조성물 전체 중량당 0.001 내지 50 중량부의 범위에서 선택되는 것이 일반적이다.
상기 건강기능식품은 식품의 생체 조절 기능을 강조한 식품으로 물리적, 생화학적, 생물공학적인 방법을 이용하여 특정 목적에 작용 및 발현하도록 부가가치를 부여한 식품이다. 이러한 건강기능 식품의 성분은 생체 방어와 신체 리듬의 조절, 질환의 방지 및 회복에 관계하는 신체 조절 기능을 생체에 대하여 충분히 발휘하도록 설계하여 가공하게 되며, 식품으로 허용 가능한 식품 보조 첨가제, 감미료 또는 기능성 원료를 함유할 수 있다.
본 발명의 우유 엑소좀을 건강기능식품(또는 건강기능 음료 첨가물)으로 사용할 경우, 상기 우유 엑소좀을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용하고, 통상적인 방법에 따라 적절하게 사용할 수 있다. 상기 우유 엑소좀의 혼합량은 그의 사용 목적(예방, 건강 또는 개선, 치료적 처치)에 따라 적합하게 결정될 수 있다.
본 발명의 또 다른 일 측면은 우유 엑소좀을 포함하는, 피부 탄력 증진 및 주름 예방 또는 치료용 약학적 조성물에 관한 것이다.
본 발명에서, “피부 탄력 개선”이란 피부 노화에 의해서 피부 탄력이 떨어지는 생기는 주름을 개선시키는 것을 일컫는다. 즉, 피부 탄력이 떨어지는 경우 피부 내 콜라겐 섬유 감소 및 변성으로 인하여 표피의 아래 근육이 접히고 펴지는 운동이 반복되게 되며, 이로인하여 피부 주름이 생기게 된다. 이에 저하된 피부 탄력을 개선시킴으로써 주름 생성을 방지 및 억제하는 것을 모두 포함한다.

구체적으로, 상기 우유 엑소좀은 소 유래인 것일 수 있다.
본 발명의 일 실시예에서는 우유 엑소좀이 진피섬유아세포에 대하여 세포독성이 거의 없음을 확인하였다(도 3).
본 발명 일 실시예에서는 우유 엑소좀이 탄력 저하 및 주름 생성 및/또는 억제 관련 유전자 발현을 조절하고(도 4), 콜라겐 합성을 촉진시키며(도 5), 특히 엘라스테아제 및 콜라게네아제 활성을 저해시키는 것을 직접적으로 확인하였다(도 6 및 도 7).
또한 구체적으로, 상기 조성물은 노화 예방 또는 치료용도를 추가로 더 포함할 수 있다.
본 발명의 일 실시예에서 자외선 UVB를 처리하여 광노화를 유발한 후 우유 엑소좀을 처리한 결과, 우유 엑소좀 미처리 군에 비해 콜라겐 섬유양 감소가 거의 없으며, 섬유 밀도가 조밀하고 배열이 규칙적임을 확인하였다(도 8).
상기와 같은 결과로부터 본 발명의 우유 엑소좀을 포함하는 약학적 조성물이 콜라겐 합성을 방해하는 유전자 발현 및 효소 활성을 저해하는 한편, 콜라겐 합성을 유도함으로써 피부 탄력 증진 및 주름 개선을 위한 예방 또는 치료 용도로 활용할 수 있으며, 나아가 자외선 등으로 인한 노화 예방 또는 치료 용도로도 활용할 수 있음을 알 수 있다.
본 발명의 약학적 조성물은 투여를 위하여, 상기 본 발명의 우유 엑소좀 외에 약학적으로 허용 가능한 담체, 부형제 또는 희석제를 포함할 수 있다. 상기 담체, 부형제 및 희석제로는 락토즈, 덱스트로즈, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
또한, 본 발명의 약학적 조성물은 어떠한 제형으로도 적용가능하며, 보다 구체적으로 비경구용 제형일 수 있다. 비경구용 제형으로는 주사용, 도포용, 에어로졸 등의 스프레이 형일 수 있다. 더욱 구체적으로는 주사제 형태일 수 있다.
비경구 투여를 위한 제제에는 멸균된 수용액, 비수성 용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성 용제, 현탁제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다.
본 발명의 또 다른 측면은 우유 엑소좀을 포함하는 약학적 조성물을 치료를 필요로 하는 개체에 약학적으로 유효한 양으로 투여하는 단계를 포함하는 피부 탄력 증진 및 주름 치료방법에 관한 것이다. '피부 탄력' 및 '주름'에 관한 설명은 전술한 바와 동일하다. 구체적으로, 우유 엑소좀을 포함하는 약학적 조성물을 투여하여 주름을 치료하는 방법에 관한 것일 수 있다.
또한, 상기 치료방법은 노화 치료를 추가로 포함하는 것일 수 있다. '노화'는 전술한 바와 동일하다.
본 발명에서 "약학적으로 유효한 양"은 의학적 치료에 적용가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효용량 수준은 환자의 성병, 연령, 질병의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
본 발명의 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있다. 또한 본 발명의 약학적 조성물은 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 용어 "개체"는 본 발명에 따른 약학적 조성물의 투여에 의해 증상이 호전될 수 있는 피부 색소 침착 질환을 가진 동물 또는 인간을 포함한다. 본 발명에 따른 치료용 조성물을 개체에게 투여함으로써, 피부 색소 침착 질환을 효과적으로 예방 및 치료할 수 있다.
본 발명의 용어 "투여"는 어떠한 적절한 방법으로 인간 또는 동물에게 소정의 물질을 도입하는 것을 의미하며, 본 발명에 따른 치료용 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 경구 또는 비경구 투여될 수 있다. 또한, 본 발명에 따른 치료용 조성물은 유효성분이 표적 세포로 이동할 수 있는 임의의 장치에 의해 투여될 수 있다.
본 발명에 따른 약학적 조성물의 바람직한 투여량은 환자의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다.
본 발명의 우유 엑소좀을 포함하는 조성물은 독성이 없어 생체 적합성이 우수하며, 엘라스테아제 및 콜라게네아제 효소 활성 저해 및 콜라겐 합성 효과가 우수하여, 피부 탄력 및 주름 개선용 조성물로서 다양한 분야에 활용될 수 있다.
본 발명의 효과는 상기한 효과로 한정되는 것은 아니며, 본 발명의 상세한 설명 또는 청구범위에 기재된 발명의 구성으로부터 추론 가능한 모든 효과를 포함하는 것으로 이해되어야 한다.

이하, 본 발명을 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명이 하기 실시예에 의해 한정되는 것은 아니다.
실시예 1. 우유 엑소좀(milk exosome) 제조방법
원심분리기를 이용하여 소 유래 우유에서 엑소좀을 추출하였다. 보다 구체적으로는, 우유를 튜브에 분주하여, 2000 g와 10,000 g에서 각각 10분 동안 원심분리하여 상층액을 수거하였다. 수거한 상층액을 0.45 μm, 0.2 μm 필터로 여과한 다음, phosphate buffered saline (PBS)를 혼합하였다. 이후, Exoquick exosome precipitation solution (System Biosciences)을 PBS와 혼합하여 넣어 준 후, 30분 동안 휴지시키고, 1,500 g에서 30분 또는 10,000 g에서 10분간 다시 원심분리하였다. 상층액을 제거하고 엑소좀 펠렛을 PBS에 용해시켜 이후 실험에 이용하였다.
실험예 1. 우유 엑소좀의 특성 확인
1-1. 우유 엑소좀의 크기 측정
우유 엑소좀의 크기를 확인하기 위해 동적 광산란 측정(dynamic light scattering, DLS)을 통해 엑소좀의 크기를 측정하였으며, 80~190 nm 크기를 나타냄을 확인하였다(도 1).
1-2. CD9 단백질 발현 확인
엑소좀 마커인 CD9 발현 여부를 웨스턴블롯을 이용하여 확인하였으며, 도 1에 나타난 바와 같이 엑소좀 마커인 CD9 단백질이 엑소좀에서는 뚜렷이 관찰되었으며, 엑소좀 외에 상층액에서는 CD9 발현이 되지 않음을 확인하였다.
우유 엑소좀에서 10% SDS 폴리아크릴마이드 젤을 이용하여 단백질을 분리하였다. 상기 분리한 단백질은 니트로셀룰로오스막(GE Health care)으로 옮긴 후, 항체의 비특이적 결합을 방지하기 위하여 1시간 동안 5% 탈지분유에 반응하였다. CD9(abcam) 1차 항체를 1:1000의 비율로 희석하여 4 ℃에서 12 ~ 18시간 동안 막에 결합시켰다. 이후, HRP-tagged anti-rabbit 항체와 결합시켜 상온에서 30분 동안 반응시켰다. ECL kit(Santa Cruz Inc)를 사용하여 단백질 밴드를 관찰하였다.
1-3. 엑소좀의 형태 및 크기 확인
상기 실시예 1에서 추출한 우유 엑소좀의 형태 및 크기를 전자현미경을 이용하여 관찰하였다. 그 결과, 도 2에 나타난 바와 같이 엑소좀이 200 nm 크기 내에서 구형으로 존재하는 것을 확인하였다.
실험예 2. 우유 엑소좀의 세포 독성 확인
2-1. 인간진피섬유아세포 배양
인간진피섬유아세포인 normal human dermal fibroblast(NHDF; Lonza, New Jersey, USA)에서 분양받아 사용하였으며 2% fetal bovine serum (FBS), 0.1% insulin, 0.1% recombinant human fibroblast growth factor-β, 0.1% GA-1000이 첨가된 FGM-2배지(Lonza, New Jersey, USA)를 사용하여 37℃, 5% CO
2 배양기에서 배양하였다.
2-2. 세포 독성 확인
본 발명의 우유 엑소좀에 독성이 있는지 확인하기 위하여, NHDF세포에 우유 엑소좀을 각각 20 μg/ml 및50 μg/ml을 24시간 동안 처리하여 세포 생존율을 측정하였다. 우유 엑소좀을 처리하지 않은 군을 대조군으로 하였다(Mock).
NHDF 세포를 96 웰 플레이트에 5x10
4 cells/ml이 되도록 분주하고 37℃, 5%, CO
2 조건의 배양기에서 24시간 배양하였다. 상기 배양된 세포에 우유 엑소좀을 처리하고, 24시간 동안 배양한 후, EZ-Cytox 세포생존율 분석키트(Daeil Lab Service)를 사용하여 제조사의 방법에 따라 세포 생존율에 대한 효과를 측정하였다.
그 결과, 도 3에 나타난 바와 같이, 대조군과 우유 엑소좀을 처리한 군 모두에서 생존율 변화가 나타나지 않은 바, 본 발명의 우유 엑소좀이 독성이 없음을 확인하였다.
실험예 3. 우유 엑소좀의 피부 탄력 및 주름 개선 효과 확인
3-1. 피부 탄력 및 주름 관련 유전자의 발현양상 확인
우유 엑소좀 처리에 따른 피부 탄력 및 주름 관련 유전자의 발현양상을 알아보기 위해, 우유 엑소좀을 NHDF 세포에 처리하고 Timp1, 프로콜라겐(procollagen), CD34, 엘라스틴(Elastin) 및 MMP-1의 발현 양상을 qRT-PCR로 확인하였다.
NHDF 세포에 우유 엑소좀을 처리하고 24시간 후에 세포를 수거하였다. 상기 수거한 세포에Tri 시약(Bioline)을 반응시켜 total RNA를 분리하였다. 역전사 반응을 실시하기 위하여 1 μg RNA에 dNTP, M-MLV reverse-transcriptase (Promega) 등을 첨가한 다음 37℃에서 1시간 반응시켜 cDNA를 합성하였다. Timp1, 프로콜라겐(procollagen), CD34, 엘라스틴(Elastin) 및 MMP-1 mRNA 발현을 측정하기 위해 SYBR Green PCR Master mix(Bioline)를 이용하여 실시간 중합효소 연쇄반응을 수행하였다. 특정 유전자를 증폭하기 위한 PCR 조건은 다음과 같이 시행하였다. 95℃에서 10분간 반응시킨 다음, 15초 95℃, 15초 60℃, 72℃ 15초를 한 주기로 하여 40 cycles 동안 증폭하였다. 표적 유전자 mRNA 발현량은 actin 발현량에 대한 상대적인 발현량으로 보정하였다. 사용된 프라이머는 하기 표 1에 나타난 바와 같다.

그 결과, 도 4에 나타난 바와 같이 우유 엑소좀을 처리한 NHDF 세포에서 MMP-1 유전자의 mRNA 발현이 농도 의존적으로 억제되는 것을 확인하였다. 한편 콜라겐, 엘라스틴 보호에 관여하는 유전자인 TIMP1의 경우, 우유 엑소좀 처리에 따라 발현량이 증가함을 확인하였으며, 프로콜라겐(procollagen), CD34, 엘라스틴(Elastin)의 발현량 또한 증가함을 확인하였다. 상기 결과는 우유 엑소좀은 주름 개선에 관여하는 유전자 발현을 촉진시키는 반면, 콜라겐 분해 효소 관련 유전자의 발현량을 조절함으로써 피부 주름 개선에 우수한 효과가 있음을 시사한다.
3-2. 타입 I콜라겐(Type I collagen) 생성량 확인
우유 엑소좀의 콜라겐 합성 촉진 효과를 확인하기 위하여, NHDF 세포에 우유 엑소좀을 처리한 후 콜라겐 합성량을 확인하였다. NHDF 세포에 우유 엑소좀을 각각 20 μg/ml 및50 μg/ml을 각각 처리하여 콜라겐 생성량을 측정하였으며, 우유 엑소좀을 처리하지 않은 군을 대조군으로 하였다(Mock).
구체적으로, NHDF 세포를 1.5 X 10
5 cells/well에 분주하여 24시간 전배양 후 우유 엑소좀 20 μg/ml 및50 μg/ml을 처리하여 48시간 배양하였다. 이후, 배양 배지를 3000 rpm에서 10분간 원심분리 후, Sircol collagen assay 키트(Bioclolor, UK)를 이용하였다. 상기 획득된 상층액은 키트에 제공된 isolation & concentration reagent를 처리하여 4℃에서 16시간 이상 유지시켰으며, 이후 12,000 rpm에서 10분간 원심분리하여 콜라겐을 농축시켰다. 상층액을 제거한 후 제공된 sircol dye reagent를 펠렛에 1 ml 추가하고 30분동안 유지시켰다. 12,000 rpm에서 10분간 원심분리하고 펠렛을 acid-salt wash reagent로 세척한 후 다시 위 조건에서 원심분리하였다. 원심분리 후, alkali reagent를 처리하여 콜라겐에 흡착된 염료를 녹여내고 555 nm의 파장에서 흡광도를 측정하였다.
그 결과, 도 5에 나타난 바와 같이, 우유 엑소좀을 처리한 경우 콜라겐 합성량이 증가함을 확인하였으며, 특히 우유 엑소좀의 농도 의존적으로 콜라겐 합성 촉진 효과가 현저하게 상승함을 확인하였다.
3-3. 엘라스테아제(Elastase) 활성저해 효과 확인
엘라스테아제(Elastase) 저해활성 측정은 Cannell 방법(R. J. Cannell, S. J. Kellam, A. M. Owsianka, & J. M. Walker, 1988)에 따라 측정하였다. 기질로서 N-succinyl-(L-Ala)3-
p-nitroanilide(Sigma Aldrich CO., USA)를 사용하여 37℃에서 20분 동안 기질로부터 생성되는
p-nitroanilide의 생성량을 415 nm에서 측정하였다.
구체적으로, 우유 엑소좀 20 μl에 50 mM Tris-HCl buffer (pH 8.0) 120 μl를 처리한 후 엘라스테아제 (1 unit/ml) 10 μl와 elastase 기질인 N-succinyl-(L-Ala)3-
p-nitroanilide 50 μl를 첨가하여 37℃에서 20분간 반응시켰다. 반응 후 415 nm에서 흡광도를 측정하였다.

그 결과, 도 6에 나타난 바와 같이, 우유 엑소좀을 처리한 경우 엘라스테아제(Elastase) 활성 억제 효과가 우유 엑소좀을 처리하지 않은 대조군에 비해 현저히 상승함을 확인하였는 바, 엘라스테아제 활성저해 효과가 우수함을 확인하였다.
3-4. 콜라게네아제(Collagenase) 활성저해 효과 확인
콜라게네아제 저해활성 측정은 Wunsch와 Heindrich(E. Wunsch, & H. G. Heidrich, 1963)의 방법에 따라 측정하였다.
구체적으로 0.1 M tris-HCL buffer (pH 7.5)에 4 mM CaCl
2를 첨가하여 0.3 mg/mL의 4-phenyl azobenzyloxycarbonyl-Pro-Leu-Gly-Pro-D-Arg(Sigma Aldrich CO., USA)를 녹인 기질액 0.25 ml 및 우유 엑소좀 0.1 ml의 혼합액에 0.2 mg/mL의 콜라게네아제(Sigma Aldrich CO., USA) 0.15 ml를 첨가하여 실온에서 20 분 동안 방치한 후 6% 시트릭산(citric acid) 500 μL를 넣어 반응을 정지시킨 후, 에틸 아세테이트(ethyl acetate) 2 mL를 첨가하여 Vmax microplate spectrophotometer (Molecular Devices, Sunnyvale, CA, USA) 320 nm에서 흡광도를 측정하였다. 콜라게네아제 저해활성은 시료 용액의 첨가군과 무첨가군의 흡광도 저해율로 나타내었다.

그 결과, 도 7에 나타난 바와 같이, 우유 엑소좀을 처리한 경우 콜라게네아제(Collagenase) 활성 억제 효과가 현저히 상승함을 확인하였는 바, 우유 엑소좀의 콜라게네아제 효소의 활성 억제에 우수한 효과가 있음을 확인하였다.
실험예 4. 우유 엑소좀의 노화 개선 효과 확인
자외선 UVB로부터 발생한 광노화 현상에 대한 우유 엑소좀의 효과를 조사하기 위해 Masson's trichrome staining 방법으로 콜라겐 섬유의 양과 형태를 관찰하였다.
먼저, 표피와 진피로 구성된 인공피부인 Neoderm-ED (테고사이언스)를 구입하여 8일동안 하루에 1번씩 0.05 J/cm
2 자외선을 조사하였고, 우유 엑소좀 50 μg/ml을 8일동안 이틀에 1번씩 처리하였다.
그 결과, 도 8에 나타난 바와 같이 우유 엑소좀을 처리한 경우 콜라겐 섬유의 밀도가 조밀하고 배열이 규칙적이게 나타나는 반면, 우유 엑소좀을 처리하지 않은 대조군에서는 콜라겐 섬유가 파괴되어 밀도가 엉성해고 배열이 불규칙 적이며, 콜라겐 섬유양이 감소됨을 확인하였다.
상기 결과를 통해 우유 엑소좀이 자외선에 의한 피부 노화 개선에도 우수한 효과가 있음을 확인하였다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다. 예를 들어, 단일형으로 설명되어 있는 각 구성 요소는 분산되어 실시될 수도 있으며, 마찬가지로 분산된 것으로 설명되어 있는 구성 요소들도 결합된 형태로 실시될 수 있다.
본 발명의 범위는 후술하는 청구범위에 의하여 나타내어지며, 청구범위의 의미 및 범위 그리고 그 균등 개념으로부터 도출되는 모든 변경 또는 변형된 형태가 본 발명의 범위에 포함되는 것으로 해석되어야 한다.
