본 발명은 들뜬 에너지 준위에서 발광 특성을 나타내는 형광 물질에 대한 소광 효과를 나타내는 소광자와 이의 다양한 용도에 관한 것이다.
Description
본 발명은 들뜬 에너지 준위에서 발광 특성을 나타내는 형광 물질에 대한 소광 효과를 나타내는 소광자와 이의 다양한 용도에 관한 것이다.
소광자(quencher)란 형광 분자의 형광을 소광(quenching)시킬 수 있는 분자를 의미하며, 일반적으로 광을 흡수할 수 있는 특성을 갖는 염료가 사용된다.
소광 현상의 메커니즘으로는 형광 공명 에너지 전이(fluorescence resonance energy transfer, FRET), 광-유도 전자 전달(photo-induced electron transfer) 및 H-이량체 형성과 같은 염료의 응집을 통해 일어나는 것으로 알려져 있다.
형광 염료의 형광을 제어 또는 소멸시키기 위해서 소광자를 사용할 경우, 소광 염료의 흡수 파장의 범위가 형광 염료가 나타내는 형광의 파장 영역의 상당 부분 또는 전체 영역을 포괄(중첩)하는지가 가장 중요하다.
또한, 소광 효과를 얻는데 있어 형광 염료와 소광자 사이의 길이도 중요한데, 예를 들어 DNA의 경우는 염기의 개수, 펩타이드/단백질의 경우 아미노산의 개수 등이 고려되며 보다 높은 소광 효과를 얻기 위해 형광 염료 및 소광자가 표지되는 링커의 길이를 조절하기도 한다.
바이오 분야에서 상업적으로 주로 사용되는 소광자의 경우 일반적으로 빛을 내지 못하고 흡수만 가능한 염료 구조가 선택되지만, FRET 현상을 이용한 형광-형광 염료 조합도 많이 활용되고 있다. 이렇게 조합된 형광-소광, 형광-형광 염료들은 서로 간의 거리가 멀어지거나 생체분자 내에서 이탈될 경우 원래의 형광이 복원되거나 강해지게 되므로 일종의 형광의 on/off 기능을 부여할 수 있게 되며, 이러한 특성을 고려하여 특정 단백질/효소 등의 바이오 마커에 감응할 수 있는 바이오센서나 활성화 프로브 등을 설계할 때 많이 사용되고 있다.
바이오 분야에서 사용되는 형광 또는 소광 염료들의 경우 단독으로 사용되는 경우는 Indocyanine green이나 Methylene blue와 같이 FDA 승인을 받은 한정된 염료에 한하며, 일반적으로는 생체분자가 갖는 치환기에 결합할 수 있는 반응성기가 도입된다. 상기 반응성기들로는 여러 가지가 알려져 있지만, 치환기 선택성, 반응 속도, 수율, 재현성, 안정성 등이 많은 연구자들로부터 장시간 검증되어 왔으며, 최근에는 실제 연구나 상업적인 목적으로 염료에 도입되는 반응성기는 몇 가지로 한정되어 있다.
예를 들면, 단백질 분자의 아민기와 결합을 목적으로 하는 반응성기로 가장 많이 이용되는 것은 숙신이미딜 에스터와 이소티오시아네이트(isothiocyanate)이고, 단백질 분자의 티올기와 결합을 목적으로 하는 반응성기로서 가장 많이 이용되는 것은 말레이미드이며, 단백질 분자의 히드록시기와 결합을 목적으로 하는 반응성기로는 디클로로트리아진(dichlorotriazine)이 주로 선택된다.
다만, 상기 반응성기들은 치환반응으로 결합이 되거나 수용성 조건에서 장시간 반응 및 보관 안정성을 유지하기 어려운 경우가 대부분이다.
본 발명은 광학 영상 분야에서 생체분자의 동정을 관찰하기 위하여 광범위하게 사용될 수 있는 화합물로서 신규한 소광자를 제공하는 것을 목적으로 한다.
또한, 본 발명은 상기 신규한 소광자를 포함하는 핵산 검출용 올리고뉴클레오타이드, 조성물, 지지체 및 핵산 검출 방법을 제공하는 것을 목적으로 한다.
상술한 기술적 과제를 해결하기 위한 본 발명의 일 측면에 따르면, 하기의 화학식 1로 표시되는 소광자가 제공된다.

본 발명은 들뜬 에너지 준위에서 발광 특성을 나타내는 형광 물질에 대한 소광 효과를 나타내는 소광자와 이의 다양한 용도에 관한 것으로서, 본 발명에 따른 소광자는 기존 소광자 대비 소광 효율이 높음에 따라 우수한 소광 특성을 나타낼 수 있다.

또한, 본 발명의 다른 측면에 따르면, 상기 올리고뉴클레오티드를 포함하는 핵산 검출용 조성물이 제공된다.
본 발명의 일 실시예에 따른 핵산 검출용 조성물은 화학식 1로 표시되는 소광자와 마이너 그루브 바인더 및 형광단을 동시에 포함하는 올리고뉴클레오타이드와 함께 표적 생체분자와의 반응을 위한 효소, 용매(완충액 등) 및 기타 시약 등을 더 포함할 수 있다.
여기서 용매로는 포스페이트 완충액, 카보네이트 완충액 및 트리스 완충액으로 구성된 군에서 선택되는 완충액, 다이메틸설폭사이드, 다이메틸포름아미드, 다이클로로메탄, 메탄올, 에탄올 및 아세토나이트릴로부터 선택되는 유기 용매 또는 물 등이 사용될 수 있으며, 용매의 종류에 따라 소광자에 다양한 작용기를 도입함으로써 용해도를 조절하는 것이 가능하다.
또한, 본 발명의 또 다른 측면에 따르면, 화학식 1로 표시되는 소광자, 지지체 및 상기 소광자와 상기 지지체를 연결하는 링커를 포함하는 핵산 검출용 지지체가 제공된다.
이에 따라, 샘플 내 생체분자는 지지체 상에 고착화된 소광자와의 상호작용을 통해 지지 매트릭스 상에 고정될 수 있다.
상기 지지 매트릭스는 유리, 셀룰로오스, 나일론, 아크릴아마이드 젤, 덱스트란, 폴리스티렌, 알지네이트, 콜라겐, 펩타이드, 피브린, 히알루론산, 아가로즈, 폴리하이드록시에틸메타크릴레이트, 폴리바이닐알코올, 폴리에틸렌글라이콜, 폴리에틸렌옥사이드, 폴리에틸렌글라이콜다이아크릴레이트, 젤라틴, 매트리젤(matrigel), 폴리락트산, 카르복시메틸셀룰로오스, 덱스트란, 키토산, 라텍스 및 세파로즈로부터 선택되는 적어도 하나로 제조될 수 있으며, 비드 또는 멤브레인 형태일 수 있다.
여기서, 링커는 소광자와 지지체를 연결하는 부분으로서, 소광자와 지지체를 연결할 수 있는 임의의 물질은 모두 본원에서 의도한 링커로서 사용될 수 있다.

핵산 검출 방법
본 발명의 일 실시예에 따르면, 표적 핵산에 소광자로 표지된 프로브를 반응시켜 표지하는 방법이 구현될 수 있다. 또한, 표적 생체분자의 종류에 따라 소광자에 적절한 반응성기를 도입함으로써 표적-특이적 상호작용을 이용한 생체분자의 표지 방법이 구현될 수도 있다. 아울러, 소광자로 표지한 생체분자를 전기영동을 통해 동정하는 방법이 구현될 수도 있다.
DNA 마이크로어레이법
DNA 마이크로 어레이법은 표지해야 할 표적 핵산에 염료를 반응시켜 표지하는 한편, 표적 핵산에 대하여 상보적인 염기서열을 가지는 단일사슬의 프로브 핵산을 준비하고, 단일사슬으로 변성시킨 표적 핵산과 프로브 핵산을 기판 위에서 혼성화하여, 표적 핵산의 형광을 측정한다.
본 표지 방법에서 기판에 고정하는 프로브 핵산으로는, 유전자의 발현을 조사하는 경우, cDNA 등의 cDNA의 라이브러리, 게놈의 라이브러리 또는 모든 게놈을 주형(鑄型)으로 하여 PCR법에 의해 증폭하여 조제한 것을 사용할 수 있다.
또한, 유전자의 변이 등을 조사하는 경우, 표준이 되는 이미 알려진 서열을 근거로 하여, 변이 등에 대응하는 여러가지 올리고뉴클레오티드를 합성한 것을 사용할 수 있다.
프로브 핵산을 기판 위에 고정하는 것은, 핵산의 종류나 기판의 종류에 따라 적당한 방법을 선택할 수 있다. 예를 들어, DNA의 전하를 이용하여, 폴리리신 등의 양이온으로 표면처리한 기판에 정전결합시키는 방법을 이용할 수도 있다.
단일사슬로 변성시킨 표적 핵산을 기판 위에 고정하고, 올리고뉴클레오타이드와 혼성화한다. 여기서, 올리고뉴클레오타이드의 5' 말단에는 형광단이 표지되고, 3' 말단에는 화학식 1로 표시되는 소광자로부터 선택되는 적어도 하나가 표지된다. 5' 말단과 3' 말단 사이에는 표적이 되는 핵산과 상보적으로 결합할 수 있는 프로브가 위치할 수 있다.
혼성화는 실온~70℃, 그리고 2~48시간의 범위에서 하는 것이 바람직하다. 혼성화에 의해 프로브 핵산과 상보적인 염기서열을 가지는 표적 핵산이 선택적으로 프로브 핵산과 결합한다. 그 후, 기판을 세정하고 실온에서 건조한다.
이 때, 올리고뉴클레오타이드는 프로브에 의해 표적 핵산에 혼성화되지만 5' 말단의 형광단은 3' 말단의 소광자에 의해 소광된 상태로 존재한다.
이어서, 표적 핵산에 혼성화된 올리고뉴클레오타이드는 중합 효소에 의해 신장되는데, 올리고뉴클레오타이드는 중합 효소의 엑소뉴클레아제 활성에 의해 표적 핵산으로부터 분리 및 분해되며, 올리고뉴클레오타이드의 5' 말단의 형광단과 3' 말단의 소광자는 서로 분리되며, 이에 따라 형광단은 형광을 발할 수 있게 된다.
이 때, 발생하는 형광 강도를 측정하여 표적 핵산의 증폭량을 측정할 수 있게 된다.
이하에서는 본 발명의 구체적인 실시예들을 제시한다. 다만, 하기에 기재된 실시예들은 본 발명을 구체적으로 예시하거나 설명하기 위한 것에 불과하며, 이로서 본 발명이 제한되어서는 아니된다.
제조예 1. 화합물 1의 합성
중간체 2의 합성
500mL 1구 반응기에 2,3,3-트라이메틸인돌레닌(30g, 0.188mol)과 황산 120ml을 넣고 0℃에서 교반하였다. KNO
3 (19.0g, 0.188mmol)과 황산(120mL)을 섞은 용액을 0℃에서 적가하였다. 1시간 후 얼음을 넣고 NaOH(aq) 중화하였다. 이어서, 생성된 고체를 필터 후 건조하였다.
중간체 3의 합성
2L 반응기에 중간체 2 (36.3g, 0.178mol)과 6M HCl 950ml를 넣고 교반하였다. Tin(II) chloride dehydrate (240.6g, 1.066mol)를 넣고 100℃에서 2시간 교반하였다. 반응이 종료되면 실온으로 식힌 후 NaOH로 중화하였다. 이어서, MC와 물로 추출 후 농축하여 건조하였다.
중간체 4의 합성
반응기에 중간체 3 (5.0g, 0.029mol) DIPEA (14.8g, 0.115mol), CH3I (12.2g, 0.086mol)과 아세톤 50.0ml을 넣고 3일간 교반하였다. 이어서, MC와 물로 추출한 후 농축하여 컬럼(HEP:EA=4:1) 정제하였다.
중간체 6의 합성
반응기에 중간체 4 (1.4g, 0.007mol)와 중간체 5 (1.5g, 0.008mol)를 다이옥산(14ml)에 녹여서 14시간 환류 교반하였다. 이어서, 용매를 농축 후 컬럽 정제하였다.
중간체 8의 합성
100 mL 1구 반응기에 중간체 6 (1g, 0.003mol), 말론알데하이드 다이아닐라이드 염산염(0.7g, 0.003mol), 초산(2mL), 무수초산(10mL)를 넣고 100℃에서 1시간 교반하였다. 이어서, 에틸 아세테이트로 고체를 석출시킨 뒤 컬럽 정제하였다(MC:MeOH=50:1).
중간체 11의 합성
반응기에 중간체 9 (100g, 0.366mol)과 중간체 10 (86.2g, 0.439mol), Pd(OAc)
2 (8.2g, 0.037mol), BINAP (22.8g, 0.037mol), STB (49.3g, 0.513mol), 톨루엔 1000ml를 넣고 환류교반하였다. TLC 확인 후 상온으로 냉각하였다. 이어서, 농축 후 MC/H
2O로 추출하여 컬럼 정제하였다.
중간체 13의 합성
반응기에 중간체 11 (100g, 0.257mol)과 중간체 12 (44.3, 0.514mol, Con. HCl 250ml와 에탄올 800ml를 넣고 하루 동안 환류 교반하였다. TLC 확인 후 상온으로 냉각하였다. 이어서, 에틸아세테이트로 고체를 석출하여 필터 후 건조하였다.
중간체 15의 합성
반응기에 중간체 13 (10.0g 0.036mol)과 중간체 14 (8.5g, 0.044mol)과 DCB 20ml를 넣고 하루 동안 환류 교반하였다. 에틸아세테이트를 사용하여 고체 석출 후 필터 하여 건조하였다.
중간체 16의 합성
반응기에 중간체 8 (2.0g, 0.005mol), 중간체 15 (3.3g, 0.005mmol), 무수초산(0.9ml, 9.71mmol), 피리딘(40mL)을 넣고 환류 하에 2시간 교반하였다. 이어서, 농축 후 컬럼정제하였다.
화합물 1 (18)의 합성
반응기에 중간체 16 (500mg, 0.726mmol)과 중간체 17 (110.1mg, 1.089mmol), HATU (331.1mg, 0.871mmol), methylmophorine (220.2mg, 2.177mmol), DMF (10ml)을 넣고 상온에서 교반하였다. 이어서, MC와 물을 사용하여 추출 후 컬럼정제하였다.
제조예 2. 화합물 2의 합성
중간체 20의 합성
반응기에 중간체 6 (10.0g, 0.026mole)과 중간체 19 (5.6g, 0.029mole), 초산(5mL), 무수초산(15mL)를 넣고 100℃에서 1시간 교반하였다. 이어서, 에틸 아세테이트로 고체를 석출시킨 뒤 컬럼 정제하였다(MC:MeOH=50:1).
중간체 21의 합성
반응기에 중간체 8 (2.0g, 0.004mol), 중간체 15 (2.3g, 0.004mmol), 무수초산(0.9ml, 9.71mmol), 피리딘(40mL)을 넣고 환류 하에 2시간 교반하였다. 이어서, 농축 후 컬럼정제하였다.
화합물 2 (22)의 합성
반응기에 중간체 16 (500mg, 0.726mmol)과 중간체 17 (107.9mg, 1.067mmol), HATU (324.5mg, 0.854mmol), methylmophorine (215.8mg, 2.134mmol), DMF (10ml)을 넣고 상온에서 교반하였다. 이어서, MC와 물을 사용하여 추출 후 컬럼정제하였다.
제조예 3. 화합물 6의 합성

반응기에 중간체 23 (25.0g, 0.152mol), 중간체 24 (30.1g, 0.183 mol), K

중간체 26의 합성
반응기에 중간체 25 (10.0g, 0.043mol), 아이오도 프로판올(9.7g, 0.052mol), 1,4-다이옥산(20.0ml)를 넣고 하루 동안 환류 교반하였다. 이어서, TLC 확인 후 컬럼 정제하였다.
중간체 28의 합성
반응기에 중간체 26 (5.0g, 0.012mol), 중간체 27 (2.6g, 0.013mol), Ac
2O (15ml)를 넣고 3시간 동안 환류 교반하였다. 이어서, 상온으로 냉각 후 용매 제거한 뒤 컬럼정제 하였다.
중간체 30의 합성
반응기에 중간체 28 (1.3g, 0.003mol), 피리딘(13ml)를 넣고 상온에서 교반하였다. 중간체 29 (1.0g, 0.003mol)을 MC(13ml)에 첨가한 후 상온에서 하루 동안 교반하였다. 이어서, 용매를 농축한 후 컬럼 정제하였다.

500mL 1구 반응기에 N,N-다이에틸-1,4-페닐렌디아민(25g, 0.152mol), 3-브로모-3-메틸-2-뷰탄온(30.1g, 0.183mol), 포타슘카보네이트(25.2g, 0.183mol), 다이메틸포름아미드(250mL)를 넣고 50℃에서 12시간 교반하였다. 반응기에 물(100mL)을 넣고 강교반한 뒤 에틸아세테이트(100mL x 2)로 추출하였다. 유기층에 무수황산마그네슘을 넣고 5분간 교반한 뒤 고체를 여과하였다. 여액은 농축하고 농축액에

10mL vial에 화합물 2 (75mg, 0.11mmol), 석신산무수물(9.9mg, 0.099mmol), 4-다이메틸아미노피리딘(12.1mg, 0.099mmol), 다이클로로메탄(5ml)를 넣고 상온에서 1.5시간 롤링하였다. 완전히 농축하고 1-에틸-3-(3-다이메틸아미노프로필)카보디이미드(52.7mg, 0.275mmol), 트라이에틸아민(28μl), 피리딘(5ml), CPG-NH
2 (1g)을 넣고 상온에서 2시간 롤링하였다. powder를 여과하고 아세토니트릴, 메탄올, 다이클로로메탄으로 각각 3회씩 세척하였다. 건조 후 CapA/CapB=1ml/1ml를 넣고 상온에서 2시간 롤링하고 아세토니트릴, 다이클로로메탄으로 각각 3회씩 세척한 후 건조하였다. 또한, 상기와 동일한 방법으로 화합물 6-CPG, 화합물 7-CPG 및 화합물 9-CPG를 합성하였다.
실험예 1. 화합물 2의 소광 특성 측정
이중 표지 올리고뉴클레오타이드의 합성
제조예 6에서 합성한 화합물 2-CPG와 CalfluorRed
® 610을 이용하여 10-Column Polygen DNA Synthesizer으로 이중 표지 올리고뉴클레오타이드를 합성하였다. 합성한 이중 표지 올리고뉴클레오타이드를 일반적인 방법으로 Cleavage 및 Deprotection한 후 RP HPLC로 정제하였다. 이중 표지 합성한 올리고뉴클레오타이드의 서열은 하기의 표 1과 같다.

이중 표지 올리고뉴클레오타이드의 합성
제조예 6에서 합성한 화합물 6-CPG와 CalfluorRed
® 610을 이용하여 10-Column Polygen DNA Synthesizer으로 이중 표지 올리고뉴클레오타이드를 합성하였다. 합성한 이중 표지 올리고뉴클레오타이드를 일반적인 방법으로 Cleavage 및 Deprotection한 후 RP HPLC로 정제하였다. 이중 표지 합성한 올리고뉴클레오타이드의 서열은 하기의 표 3과 같다.

이상, 본 발명의 일 실시예에 대하여 설명하였으나, 해당 기술 분야에서 통상의 지식을 가진 자라면 특허청구범위에 기재된 본 발명의 사상으로부터 벗어나지 않는 범위 내에서, 구성 요소의 부가, 변경, 삭제 또는 추가 등에 의해 본 발명을 다양하게 수정 및 변경시킬 수 있을 것이며, 이 또한 본 발명의 권리범위 내에 포함된다고 할 것이다.
Claims (23)
- 하기의 화학식 1 로 표시되는 소광자:[화학식 1]여기서,R 1 및 R 2는 각각 하기 화학식 2의 a 및 b, b 및 c 또는 c 및 d와 접합되며,[화학식 2]Ar 1은 치환 또는 비치환된 아릴, 치환 또는 비치환된 헤테로아릴 및 화학식 2로부터 선택되며,n은 1 내지 3의 정수이며,R 3 내지 R 19는 각각 독립적으로 수소, 중수소, 치환 또는 비치환된 C1-C10 알킬, 적어도 하나의 헤테로 원자를 포함하는 치환 또는 비치환된 C1-C10 헤테로알킬, 치환 또는 비치환된 C2-C10 알케닐, 치환 또는 비치환된 C2-C10 알키닐, 치환 또는 비치환된 C 3-C 20 사이클로알킬, 치환 또는 비치환된 C1-C10 알콕시, 치환 또는 비치환된 아릴옥시, 치환 또는 비치환된 C1-C10 할로알킬, 할로겐, 사이아노, 하이드록시, 치환 또는 비치환된 아미노, 치환 또는 비치환된 아마이드, 카바메이트, 설프하이드릴, 나이트로, 카르복실, 카르복실산염, 치환 또는 비치환된 아릴, 치환 또는 비치환된 헤테로아릴, 치환 또는 비치환된 아르알킬, 4차 암모늄, 인산, 인산염, 치환된 케톤, 알데하이드, 치환된 에스터, 치환된 설포닐, 치환 또는 비치환된 설폰아마이드, 아실클로라이드, 설폰산, 설폰산염, 히드라진, 티올, 아세탈, 케탈, 포스포네이트(아인산염), 하이포아인산염, 술포히드록시, 설페이트, 아지도, 구아니디움, 케텐, 티오카르보닐, 아미노티오카르보닐, 폴리알킬렌옥사이드, 카르복실 유도체, 친다이엔체, 설포닐 할라이드, 에폭사이드 및 포스포아미디트로부터 선택되는 작용기이거나, 카르복실, 카르복실 유도체, 하이드록실, 할로알킬, 친다이엔체, 알데하이드, 케톤, 설포닐 할라이드, 티올, 아민, 설프하이드릴, 알켄, 에폭사이드 및 포스포아미디트로부터 선택되는 작용기와 공유 결합 가능한 반응성기로부터 선택되며,R 5 와 R 6는 각각 독립적으로 존재하거나 서로 연결되어 고리를 형성하며,R 7 와 R 8은 각각 독립적으로 존재하거나 서로 연결되어 고리를 형성하며,R 18 와 R 19은 각각 독립적으로 존재하거나 서로 연결되어 고리를 형성하며,L 1 및 L 3은 단결합 또는 1 내지 40개의 비수소 원자를 포함하는 링커이며, L 2 및 L 4는 1 내지 40개의 비수소 원자를 포함하는 링커이다.
- 제1항에 있어서,상기 화학식 1의 R 1 및 R 2는 상기 화학식 2의 a 및 b와 접합되는,소광자.
- 제1항에 있어서,상기 화학식 1의 R 1 및 R 2는 상기 화학식 2의 b 및 c와 접합되는,소광자.
- 제1항에 있어서,상기 화학식 1의 R 1 및 R 2는 상기 화학식 2의 c 및 d와 접합되는,소광자.
- 제1항에 있어서,R 3 내지 R 19 중 적어도 하나는 카르복실, 카르복실 유도체, 하이드록실, 할로알킬, 친다이엔체, 알데하이드, 케톤, 설포닐 할라이드, 티올, 아민, 설프하이드릴, 알켄, 에폭사이드 및 포스포아미디트로부터 선택되는 작용기이거나 상기 작용기와 공유 결합 가능한 반응성기인,소광자.
- 제1항에 있어서,R 3 내지 R 19 및 Ar 1의 치환기 중 적어도 하나는 카르복실, 카르복실 유도체, 하이드록실, 할로알킬, 친다이엔체, 알데하이드, 케톤, 설포닐 할라이드, 티올, 아민, 설프하이드릴, 알켄, 에폭사이드 및 포스포아미디트로부터 선택되는 작용기이거나 상기 작용기와 공유 결합 가능한 반응성기인,소광자.
- 제1항에 있어서,상기 반응성기는 카르복실, 카르복실 유도체, 하이드록실, 할로알킬, 친다이엔체, 알데하이드, 케톤, 설포닐 할라이드, 티올, 아민, 설프하이드릴, 알켄, 에폭사이드 및 포스포아미디트로부터 선택되며, 상기 반응성기는 보호기로 보호되는,소광자.
- 제1항에 있어서,R 14 내지 R 17 중 인접한 치환기는 서로 연결되어 고리를 형성하는,소광자.
- 제1항에 있어서,Ar 1의 치환기 중 적어도 하나는 치환된 아미노기인,소광자.
- 제9항에 있어서,상기 아미노기의 질소에 결합된 치환기는 상기 질소에 결합된 다른 치환기와 서로 연결되어 고리를 형성하거나, 상기 아미노기 외 다른 치환기와 서로 연결되어 고리를 형성하는,소광자.
- 제11항에 있어서,R 20 내지 R 23 중 적어도 하나는 치환된 아미노기인,소광자.
- 제11항에 있어서,R 21은 치환된 아미노기인,소광자.
- 제12항 또는 제13항에 있어서,상기 아미노기의 질소에 결합된 치환기는 상기 질소에 결합된 다른 치환기와 서로 연결되어 고리를 형성하거나, 상기 아미노기 외 다른 치환기와 서로 연결되어 고리를 형성하는,소광자.
- 제1항 내지 제14항 중 어느 한 항에 따른 소광자;마이너 그루브 바인더(MGB; minor groove binder); 및형광단;을 포함하는 올리고뉴클레오타이드.
- 제15항에 있어서,상기 형광단은 쿠마린, 시아닌, 보디피, 플로세인, 로다민, 피렌, 카르보피로닌, 옥사진, 잔텐, 티오잔텐, 아크리딘 및 또는 이의 유도체로부터 선택되는 적어도 하나인,올리고뉴클레오타이드.
- 제15항에 따른 올리고뉴클레오타이드를 포함하는 핵산 검출용 조성물.
- 제1항 내지 제14항 중 어느 한 항에 따른 소광자;지지체; 및상기 소광자와 상기 지지체를 연결하는 링커;를 포함하는,핵산 검출용 지지체.
- 제18항에 있어서,상기 지지체는 유리, 셀룰로오스, 나일론, 아크릴아마이드 젤, 덱스트란, 폴리스티렌 또는 레진인,핵산 검출용 지지체.
- 제18항에 있어서,상기 링커는 치환 또는 비치환된 C 1-C 30 알킬, 적어도 하나의 헤테로 원자를 포함하는 치환 또는 비치환된 C 2-C 30 헤테로알킬, 치환 또는 비치환된 C 6-C 30 아릴 및 치환 또는 비치환된 C 3-C 30 헤테로아릴로부터 선택되는,핵산 검출용 지지체.
- (a) 표적 핵산, 상기 표적 핵산을 증폭시키기 위해 필요한 시약 및 제15항에 따른 올리고뉴클레오타이드를 포함하는 반응 혼합물을 준비하는 단계;(b) 상기 반응 혼합물 중 표적 핵산을 중합효소 연쇄 반응에 의해 증폭하는 단계; 및(c) 상기 반응 혼합물의 형광 강도를 측정하는 단계;를 포함하는,핵산 검출 방법.
- 제21항에 있어서,상기 단계 (b)는,(b-1) 상기 표적 핵산에 혼성화된 올리고뉴클레오타이드가 중합 효소에 의해 신장되는 단계;(b-2) 상기 중합 효소의 엑소뉴클레아제 활성에 의해 상기 표적 핵산으로부터 상기 올리고뉴클레오타이드의 소광자와 형광단이 분리되는 단계; 및(b-3) 상기 소광자로부터 떨어진 상기 형광단이 형광을 발하는 단계;를 포함하는,핵산 검출 방법.
- 제21항에 있어서,상기 단계 (c)에서 측정된 형광 강도로부터 표적 핵산의 증폭량을 측정하는 단계 (d)를 더 포함하는,핵산 검출 방법.
