Abstract
本発明は、一般式(I)の化合物(式中、R1、R2及びR3は、互いに独立して水素又は低級アルキルであり、Xは、アルカリ金属カチオン、アンモニウムカチオン又は第四級アンモニウムカチオンである)に関する。本発明の化合物は、水に対する優れた溶解性を示し、またその溶液は生体への投与の際にも中和処理が不要であり、医薬等として有用である。
Description
本発明は、新規の構造を有する有機ゲルマニウム化合物に関するものである。
ゲルマニウム(Ge)は、半導体として古くから研究の対象になっていた元素であるが、その有機化合物に関する研究も盛んに行われ、種々の有機ゲルマニウム化合物が合成されてきた。
例えば、Ge-132(ポリ-トランス-〔(2-カルボキシエチル)ゲルマセスキオキサン]、レパゲルマニウム、アサイゲルマニウムとも呼ばれる)は、免疫賦活作用、抗腫瘍作用、抗炎症作用、鎮痛作用、モルヒネとの協調作用といった様々な生理作用を持つ有機ゲルマニウム化合物である。Ge-132は、ゲルマニウムと酸素の12原子で構成される環状構造を有する常温で固体の結晶性化合物であり、水中では加水分解して単量体である3-(トリヒドロキシゲルミル)プロパン酸(3-(trihydroxygermyl)propanoic acid、THGP)となる。生体内は水分に富んでいるため、Ge-132は生体内では加水分解を受けてTHGP又はその塩を生成していると考えられる。
THGPは、シス-ジオール含有化合物との脱水縮合によりラクトン型のTHGP-シス-ジオール錯体を形成し、当該シス-ジオール含有化合物の生理的機能を制御するものと考えられている(非特許文献1等)。相互作用による生理的機能制御の例として、THGPがアデノシンやATPと錯体を形成することでこれらとP1あるいはP2受容体との結合を阻害し、疼痛抑制に関与し得ることが報告されている(非特許文献2)。このような知見から、Ge-132が示す種々の生理作用は、生体内で加水分解されることで生成するTHGPと生体成分との相互作用によって発揮されているものと考えられている。
THGPの溶液を乾固させると、複数のTHGPが脱水縮合したポリマー構造の固体有機ゲルマニウム化合物が生成される。Ge-132はこうして生成される固体有機ゲルマニウム化合物の一種であるが、その他にも、直鎖状リニアポリマー構造の固体有機ゲルマニウム化合物Poly-[(2-carboxyethyl-hydroxygermanium)oxide]、ラダー状構造(ゲルマニウムと酸素の8原子で構成される環状構造)の固体有機ゲルマニウム化合物Propagermanium(3-oxygermylpropionic acid polymer)等が知られている(特許文献1、非特許文献3、4)。いずれの固体有機ゲルマニウム化合物も水を含む溶媒中で加水分解されることでTHGPを生成し、これによって先に述べた様々な生理作用を示すものと推測される。したがって、生理作用の発揮が期待される固体有機ゲルマニウム化合物は、THGPをより生じやすいもの、すなわち溶解性に優れたもの、具体的には高い溶解度及び溶解速度を有するものが好ましい。
しかしながら、従来の固体有機ゲルマニウム化合物の水に対する溶解度は十分なものではなく、また加水分解して水に溶解するまでに時間を要する、つまり溶解速度が低かった。加えて、従来の固体有機ゲルマニウム化合物を溶解した溶液のpHは酸性を呈するため、当該溶液を生体に投与する際には、溶液の中和処理が必要となる場合があった。

本発明は、溶解性の優れた新規有機ゲルマニウム化合物を提供することを目的とする。
本発明者らは、THGPの結晶化について鋭意研究を重ねた結果、高濃度のTHGPを含む水溶液をアルカリで中和し、アルコール等の水と混和が可能な有機溶媒を加えることで均一な単結晶が生成されること、及びこの結晶はTHGPが8分子会合したかご状オクタマーであることを見いだし、下記の発明を完成させた。
本発明によると、溶解性に優れ、かつ生体への投与の際にも中和処理が不要な有機ゲルマニウム化合物を提供することができる。
以下に記載する本発明の説明は、代表的な実施形態又は具体例に基づくことがあるが、本発明はそのような実施形態又は具体例に限定されるものではない。なお、本明細書において、各数値範囲の上限値及び下限値は任意に組み合わせることができる。また、本明細書において「~」又は「-」を用いて表される数値範囲は、特に断りがない場合、その両端の数値を上限値及び下限値として含む範囲を意味する。
本発明の第1の態様は、一般式(I)
の化合物であって、式中、R1、R2及びR3は、互いに独立して水素又は低級アルキルであり、Xは、アルカリ金属カチオン、アンモニウムカチオン又は第四級アンモニウムカチオンである、前記化合物に関する。
一般式(I)におけるR1、R2及びR3は、互いに独立して水素又は低級アルキルである。低級アルキルとは、炭素数が1~6個、好ましくは1~4個、より好ましくは1~3個の直鎖又は分岐鎖状のアルキルをいう。好ましい実施形態において、R1、R2及びR3はいずれも水素である。
一般式(I)におけるXは、Na+、K+等のアルカリ金属カチオン、アンモニウムカチオン、又は第四級アンモニウムカチオンである。第四級アンモニウムカチオンは、窒素にアルキル基及び/又はアリール基が4個結合したカチオンであり、本発明において好ましく用いられる第四級アンモニウムカチオンとしては、炭素数が1~6個、好ましくは1~4個、より好ましくは1~3個の直鎖又は分岐鎖状である4個のアルキル基が窒素に結合したカチオン、例えば、テトラメチルアンモニウムカチオン、テトラエチルアンモニウムカチオン、テトラプロピルアンモニウムカチオン、エチルトリメチルアンモニウム、トリエチルメチルアンモニウム等を挙げることができる。好ましい実施形態において、XはNa+である。
一般式(I)の化合物の好適な例としては、R1~R3がいずれも水素であり、XがNa+である、1, 7, 9, 15-tetra(2’-sodium carboxyethylgermanium)-3, 5, 11, 13-tetra-[sodium propanato(2-)-C3’, O’]-germanium-2, 4, 6, 8, 10, 12, 14, 16, 17, 18, 19, 20-dodecaoxa-pentacyclo [8.11, 5. 17, 11. 19, 13. 13, 15.] icosane(以下、THGPオクタマーと表記する)を挙げることができる。
一般式(I)の化合物は、一般式(III)の化合物(式中、R1~R3は一般式(I)の説明において記載されているとおりである)を、水及び水と混和可能な有機溶媒の混合溶媒中で結晶化させることによって製造することができる。
一般式(IV)のアクリル酸誘導体とトリクロロゲルマンとのハイドロゲルミレーションは、例えば、濃塩酸、ジエチルエーテル又はクロロホルム等を溶媒として用いて25~40℃程度の温度で行うことができる。次いで、ハイドロゲルミレーションにより得られた化合物を、水の存在下で、典型的には水溶液中で、アルカリ金属の水酸化物又はアミンと反応させて加水分解及び中和することによって、一般式(III)の化合物を調製することができる。
あるいは、Ge-132等の公知のTHGP重合体を、水の存在下でアルカリ金属の水酸化物又はアミンと反応させることによって、式中、R1~R3が水素である一般式(III)の化合物を調製することもできる。
得られた一般式(III)の化合物は、次いで、水及び水と混和可能な有機溶媒との混合溶媒中で結晶化される。混合溶媒中の水と有機溶媒との混合比は、1:3~1:20(v/v)であればよく、好ましくは1:3~1:10(v/v)、より好ましくは1:3~1:5(v/v)である。用いられる有機溶媒としては、例えば、アセトン又は低級アルコール、例としてメタノール、エタノール、1-プロパノール又は2-プロパノール等であり、特にエタノールが好ましい。
一般式(III)の化合物の水溶液に、混合比率が上記の範囲になる量の上記有機溶媒を加えることにより、結晶化のための混合液が調製される。この混合液中の一般式(III)の化合物の濃度は、2%~12%(w/v)であればよく、例えば4%~12%(w/v)、6%~12%(w/v)、8%~12%(w/v)であり得て、好ましくは8%~12%(w/v)である。
上記の混合液を十分に撹拌した後、室温下で12時間以上、好ましくは24時間以上静置することで、一般式(I)の化合物の結晶を析出させることができる。また、結晶化を促すため、一般式(I)の化合物の結晶を種晶として加えてもよい。得られた結晶は、例えば、ろ過、低級アルコール等の有機溶媒による洗浄及び減圧乾燥といった公知の手段によって、混合液から単離精製することができる。
このように、本発明は、一般式(III)の化合物を溶解した水溶液と、水及び水と混和可能な有機溶媒を1:3~1:20(v/v)の比で混合した混合溶媒とを、一般式(III)の化合物の濃度が2%~12%(w/v)となるような比率で混合する工程;並びに混合後の液を静置して一般式(I)の化合物の結晶を析出させる工程を含む、一般式(I)の化合物の製造方法をも提供する。
一般式(I)の化合物は、上記の製造方法によって結晶として製造することができ、かかる結晶は本発明の別の態様である。THGPオクタマーの結晶は、サイズが8 μm×2 μmであり、形状が板状である。またこの結晶を有機溶媒に溶解した飽和溶液から有機溶媒を蒸発させることで再結晶化も可能である。有機溶媒としてメタノールを用いて再結晶化したTHGPオクタマーの単結晶は、X線回折測定に供した場合、以下の結晶データを示す。

X線回折測定においては、測定条件に応じて測定誤差が生じ得る。したがって、一般式(I)の化合物の結晶は、上記結晶データと完全に同一の結晶データを与える結晶に限られるものではなく、上記結晶データと実質的に同一の結晶データを与える結晶もまた本発明の範囲内にある。当業者は、測定条件を適宜変更することにより、結晶データの実質的同一性を確認することができる。
また、一般式(I)の化合物の結晶は、その全てが上記の結晶データを与えるものではない。当業者は、上記の結晶データを与える結晶は一般式(I)の化合物の結晶の一形態に過ぎず、一般式(I)の化合物の結晶には他の形態が存在し得ることを理解する。
一般式(I)の化合物は、水に対して極めて溶けやすく、また速やかに溶解する。例えば、20℃の水に対するGe-132の溶解度(飽和溶液100 g中の溶質の質量(g))は約1%であり(すなわち20℃の飽和水溶液100 g中のGe-132の量は約1 g)、また同温度の水に対して1%のGe-132が完全溶解するまでの時間は30分以上であるのに対して、同条件下におけるTHGPオクタマーの溶解度は58.8%、溶解時間は10秒程度である。また、THGPオクタマーが溶解した水溶液のpHは、生体のpHと同様に中性を示す。さらに、一般式(I)の化合物は、既存THGP重合体が溶解することができない一部の有機溶媒、例えばメタノールにも溶解することができる。
一般式(I)の化合物は、水中で加水分解を受け、構成単位である水溶性の有機ゲルマニウム化合物を生成する。かかる水溶性有機ゲルマニウム化合物は、水溶液中でシスジオール構造を有する化合物(例えばアデノシンやフルクトース)やチオール構造を有する化合物(例えばH2Sやグルタチオン)と相互作用することができる。また、相互作用可能な物質に起因する疼痛及び機械的アロディニアを抑制することができる。さらに、相互作用可能な物質と共に水に溶解することで、相互作用可能な物質の官能基の保護又は揮発性を抑えることができる。したがって、一般式(I)の化合物は、疼痛及び機械的アロディニアの抑制、シスジオール基含有化合物の保護、チオール基含有化合物の保護、硫化水素に起因する疼痛又は掻痒の抑制、消臭又は防臭、又はI型インターフェロン産生抑制等の、Ge-132と同様の用途に利用することができる。
一般式(I)の化合物は、薬学的に許容される緩衝剤、安定剤、保存剤、賦形剤その他の成分及び/又は他の有効成分をさらに含む医薬組成物、食品組成物又は化粧品組成物の形態で生体に対して用いることができる。かかる組成物は、本発明のさらなる態様である。薬学的に許容される成分は当業者において周知であり、当業者が通常の実施能力の範囲内で、例えば第十七改正日本薬局方その他の規格書に記載された成分から製剤の形態に応じて適宜選択して使用することができる。
一般式(I)の化合物を含有する医薬組成物は、有機ゲルマニウム化合物、特にTHGPが有効な疾患の治療又は症状の緩和、例えばがんや感染症の治療、炎症や疼痛の緩和等に用いることができる。したがって本発明は、一般式(I)の化合物を含有する医薬組成物の有効量を対象に投与することを含む、有機ゲルマニウム化合物、特にTHGPが有効な疾患を治療する又は症状を緩和する方法を提供する。疾患の例としては腫瘍、感染症等を、また症状の例としては炎症、疼痛等をそれぞれ挙げることができる。
本発明において、上記組成物の形態は任意であるが、経口剤(錠剤、カプセル剤、散剤、顆粒剤、細粒剤、丸剤、懸濁剤、乳剤、液剤、シロップ剤等)及び外用剤(軟膏剤、貼付剤など)を好ましい例として挙げることができる。
上記組成物の投与方法は特に限定されず、剤形に応じて適宜決定される。好ましい実施形態の一つにおいて、上記組成物は、経口的又は経皮的に生体に投与される。
上記組成物の投与量は、用法、対象の年齢、疾患の種類及び部位その他の条件などに応じて適宜選択されるが、通常成人に対して体重1kgあたり10 μg~2000 μg、好ましくは50 μg~1000 μg、より好ましくは100 μg~500 μgであり、これを1日に1回若しくは複数回に分けて、又は間歇的に投与することができる。
以下の実施例によって本発明をさらに詳細に説明するが、本発明はこれらの例に限定されるものではない。
実施例1 THGPオクタマーの合成と構造解析
1)合成
2 LビーカーにGe-132(500 g, モノマー換算で2.95 mol)を秤量し、蒸留水1 Lを加えて懸濁した。テフロン(登録商標)容器に95%水酸化ナトリウム(124 g, 2.95 mol)を秤量し、蒸留水500 mLを加えて溶解した。先のGe-132水懸濁液に水酸化ナトリウム水溶液を撹拌下で数回に分けて添加して中和し、Ge-132を溶解した。2 Lナスフラスコに中和溶液を移し、減圧下で加熱して約1 Lになるまで濃縮した。濃縮液を5 L容器に移し替え、3 Lのエタノールを添加し、十分に撹拌した後に室温下で24時間静置した。析出した結晶を吸引ろ過及びろ集して風乾し、さらにグラスチューブオーブンで105℃、8時間以上減圧下で加熱して乾燥させ、白色結晶として目的の化合物を収率95%で得た。得られた結晶のサイズは8 μm×2 μm、結晶形状は板状であった。
1)合成

2)単結晶X線構造解析
1)で得られた結晶をメタノールに溶解して飽和メタノール溶液を調製した。これを密栓せずに室温下で静置してメタノールを徐々に蒸発させることで、1 mm以上に成長した板状の結晶を得た。結晶を回収し、単結晶X線構造解析装置(SMART APEX II ULTRA、Bruker社)を用いて、以下の条件でX線回折強度の測定を行った。

1)で得られた結晶をメタノールに溶解して飽和メタノール溶液を調製した。これを密栓せずに室温下で静置してメタノールを徐々に蒸発させることで、1 mm以上に成長した板状の結晶を得た。結晶を回収し、単結晶X線構造解析装置(SMART APEX II ULTRA、Bruker社)を用いて、以下の条件でX線回折強度の測定を行った。

決定した結晶構造のパラメータを下に、原子座標を表1に、これらから構築される分子構造を図1に示す。

1)で得られた結晶は、8分子のTHGPが脱水縮合した構造を有する有機ゲルマニウム化合物((1, 7, 9, 15-tetra(2’-sodium carboxyethylgermanium)-3, 5, 11, 13-tetra-[sodium propanato(2-)-C3’, O’]-germanium-2, 4, 6, 8, 10, 12, 14, 16, 17, 18, 19, 20-dodecaoxa-pentacyclo [8.11, 5. 17, 11. 19, 13. 13, 15.] icosane)、THGPオクタマー)であることが確認された(図1及び表1)。特徴的な点として、オクタマーを構成する8つのTHGP中のプロピオン鎖のうち4つは直鎖構造であるのに対し残る4つはラクトン構造であること、及びラクトン構造中のゲルマニウム原子が5配位であることが挙げられる。
3)溶解度
1)で得られたTHGPオクタマー100 mgに精製水又はメタノールを少量ずつ加えて室温下で30分間撹拌する作業を繰り返し、目視によりTHGPオクタマーが完全に溶解したと認められる液量を測定した。測定した液量からTHGPオクタマーの溶解度(溶質量(g)/総質量(g)×100%)を算出した。また、完全溶解したときの溶液のpHを測定した。
1)で得られたTHGPオクタマー100 mgに精製水又はメタノールを少量ずつ加えて室温下で30分間撹拌する作業を繰り返し、目視によりTHGPオクタマーが完全に溶解したと認められる液量を測定した。測定した液量からTHGPオクタマーの溶解度(溶質量(g)/総質量(g)×100%)を算出した。また、完全溶解したときの溶液のpHを測定した。
Ge-132を始めとする従来のTHGP重合体は、水に対して約1%の溶解度を持ち、有機溶媒、例えばメタノールには不溶である。これに対し、THGPオクタマーの水及びメタノールに対する溶解度はそれぞれ58.8%及び1.4%であった。また、水溶液のpHは7.5前後と中性付近を示した。
4)溶解速度
精製水10 mLにGe-132及びTHGPオクタマー100 mgをそれぞれ加えて、室温下、300 rpmの速度で撹拌し、目視で完全に溶解したと認められるまでに要する時間を測定した。

Ge-132を始めとする従来のTHGP重合体を室温下で水に溶解する場合、結晶が完全に溶解するまでにある程度の時間を要する。Ge-132 1%水溶液調製時における、結晶が完全に溶解するまでの時間を測定した結果、30分以上を要することを確認した。一方、THGPオクタマーは10秒程度で完全に溶解した。
5)レーザー回折粒度分布
レーザー回折式粒度分布測定装置(HRA、マイクロトラック社製)を用い、エタノールを分散媒として、1分間の超音波分散後、THGPオクタマーの粒度分布を測定した。測定条件は以下のとおりである。
測定範囲:0.06~1400 μm
使用光源:炭酸ガスレーザー×3個
検出器:散乱光検出器
レーザー回折式粒度分布測定装置(HRA、マイクロトラック社製)を用い、エタノールを分散媒として、1分間の超音波分散後、THGPオクタマーの粒度分布を測定した。測定条件は以下のとおりである。
測定範囲:0.06~1400 μm
使用光源:炭酸ガスレーザー×3個
検出器:散乱光検出器
その結果、THGPオクタマーは、Dp10 = 3.4 μm、Dp50(Median径)= 8.0 μm、Dp90 = 16.9 μmと、極めて微細な粒子であることが確認された(図2)。
6)赤外分光光度分析
臭化カリウム(KBr)とTHGPオクタマー又はGe-132との混合物を錠剤化したものについて、赤外分光光度計(FTIR-8100M、島津製作所社製)を用いて赤外分光分析を行った。分析条件は以下のとおりである。

測定範囲:400~4600 cm-1
分解能:4 cm-1
ミラースピード:2.8 mm/sec
積算回数:40回
臭化カリウム(KBr)とTHGPオクタマー又はGe-132との混合物を錠剤化したものについて、赤外分光光度計(FTIR-8100M、島津製作所社製)を用いて赤外分光分析を行った。分析条件は以下のとおりである。
測定手法:KBr錠剤法
測定範囲:400~4600 cm-1
分解能:4 cm-1
ミラースピード:2.8 mm/sec
積算回数:40回
その結果、Ge-132では1700 cm-1に見られるC=Oによる吸収がTHGPオクタマーでは1650~1550 cm-1と低波数側にシフトしたこと、Ge-132では3000 cm-1に見られるカルボン酸による吸収がTHGPオクタマーでは消失したこと、さらに1500 cm-1以下の指紋領域において互いに異なるスペクトルを示すことが確認された(図3)。
7)質量分析
THGPオクタマーのメタノール溶液を質量分析計(TOF-MS、QToF Ultima、Waters社)に供し、質量分析を行った。分析条件は以下のとおりである。

TOF-MSによって測定されたTHGPオクタマーの質量スペクトルと天然同位体の量から計算された理論スペクトルを図4に示す。実測値は理論値とほぼ一致していることが確認された。
8) 1 H NMR測定
THGPオクタマーを重水及び重メタノールにそれぞれ溶解した溶液について、核磁気共鳴装置(Gemini 2000、アジレント・テクノロジー社)を用いて1H NMRスペクトルを測定した。比較対照としてGe-132を重水に溶解後、重水酸化ナトリウム溶液で中和した溶液を調製し、同様に測定した。測定条件は25℃、積算回数16回、300 MHzとし、残存するHODのシグナルを4.80 ppmにオフセットした。
THGPオクタマーを重水及び重メタノールにそれぞれ溶解した溶液について、核磁気共鳴装置(Gemini 2000、アジレント・テクノロジー社)を用いて1H NMRスペクトルを測定した。比較対照としてGe-132を重水に溶解後、重水酸化ナトリウム溶液で中和した溶液を調製し、同様に測定した。測定条件は25℃、積算回数16回、300 MHzとし、残存するHODのシグナルを4.80 ppmにオフセットした。
各サンプルの1H NMRスペクトルを図5及び6に示す。THGPオクタマーを重水に溶解して得られた1H NMRスペクトルでは1.56 ppm及び2.47ppmにそれぞれトリプレットを観測した。この結果はGe-132水溶液の1H NMRスペクトルと一致しており、どちらもTHGPを生成していることが示唆される(図5)。一方で、THGPオクタマーを重メタノールに溶解して得られた1H NMRスペクトルでは1.40 ppm、1.51 ppm、2.30 ppm及び2.38 ppmにそれぞれトリプレットを観測した(図6)。重水を用いた場合、THGPオクタマーは加水分解を受けてTHGPを生成するため、プロピオン鎖上にある2つのCH2に起因するシグナルが2箇所に観測されたのに対し、重メタノールを用いた場合は加水分解を受けないため、オクタマーに存在する直鎖状のプロピオン鎖と環状のプロピオン鎖がそれぞれ持つ2つのCH2が区別され、合計4箇所に観測されたものと推測される。
実施例2 アデノシンとの相互作用性の確認
重水にTHGPオクタマーとアデノシンとを溶解して得た混合溶液(混合モル比1:1、pH7.0~7.5、100 mM)について、30℃、積算回数16回、300 MHzの測定条件のもと、1H NMRスペクトルを測定した。残存するHODのシグナルを4.80 ppmにオフセットした。比較対照としてGe-132とアデノシンの混合溶液についても同様に測定した。
重水にTHGPオクタマーとアデノシンとを溶解して得た混合溶液(混合モル比1:1、pH7.0~7.5、100 mM)について、30℃、積算回数16回、300 MHzの測定条件のもと、1H NMRスペクトルを測定した。残存するHODのシグナルを4.80 ppmにオフセットした。比較対照としてGe-132とアデノシンの混合溶液についても同様に測定した。
各サンプルの1H NMRスペクトルを図7に示す。THGPオクタマーとアデノシンとの混合溶液の1H NMRスペクトルでは、THGPに由来する1.58 ppm(Ge-CH2起因)及び2.50 ppm (CH2-COO-起因)のトリプレットシグナル以外に、1.76 ppm及び2.57 ppmに新たなシグナルが観測された。アデノシン単独では上記の化学シフト付近にスペクトルを持たないことから、新たに観測されたシグナルはアデノシンとTHGPの複合体由来であることが示唆された。この結果はTHGPオクタマーに代えてGe-132を用いた場合の1H NMRスペクトルと一致しており、THGPオクタマーはGe-132と同様にTHGPを生成してアデノシンとの反応性を示すことが明らかとなった。
実施例3 H 2 Sとの相互作用性の確認
重水で調製した10 mMリン酸緩衝液(pH7.4)にTHGPオクタマーとNaHS(H2Sのナトリウム塩)とを溶解して得た混合溶液(混合モル比1:1、pH7.0~7.5、20 mM)について、25℃、積算回数16回、300 MHzの測定条件のもと、1H NMRスペクトルを測定した。残存するHODのシグナルを4.80 ppmにオフセットした。比較対照としてGe-132とNaHSの混合溶液についても同様に測定した。
重水で調製した10 mMリン酸緩衝液(pH7.4)にTHGPオクタマーとNaHS(H2Sのナトリウム塩)とを溶解して得た混合溶液(混合モル比1:1、pH7.0~7.5、20 mM)について、25℃、積算回数16回、300 MHzの測定条件のもと、1H NMRスペクトルを測定した。残存するHODのシグナルを4.80 ppmにオフセットした。比較対照としてGe-132とNaHSの混合溶液についても同様に測定した。
各サンプルの1H NMRスペクトルを図8に示す。THGPオクタマーとNaHSとの混合溶液の1H NMRスペクトルでは、THGPに由来する1.56 ppm(Ge-CH2起因)及び2.47 ppm (CH2-COO-起因)のトリプレットシグナル以外に、1.40 ppm及び2.39 ppmに新たなシグナルが観測された。NaHS単独では上記の化学シフト付近にスペクトルを持たないことから、新たに観測されたシグナルはTHGPとNaHSとの複合体由来であることが示唆された。この結果はTHGPオクタマーに代えてGe-132を用いた場合の1H NMRスペクトルと一致しており、THGPオクタマーはGe-132と同様にTHGPを生成してNaHSとの反応性を示すことが明らかとなった。
