Abstract
本発明は、新規な運動ニューロン疾患の予防剤及び/又は治療剤、ならびに該予防剤及び/又は治療剤を含有する医薬組成物を提供することを目的とする。 本発明は、一般式(I): (式中、Rは水素、置換基を有していてもよい鎖状若しくは環状の炭化水素基又は置換基を有していてもよい複素環基を表し、Ar1及びAr2は同一又は異なって、置換基を有してもよい同素環基又は複素環基を表す) で表される化合物を有効成分とする、運動ニューロン疾患の予防剤及び/又は治療剤を包含する。
Description
本発明は、運動ニューロン疾患の予防剤及び/又は治療剤に関する。
筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)をはじめとする運動ニューロン疾患(MND:motor neuron disease)は、運動ニューロンの機能障害や細胞死に伴って、全身の筋肉が萎縮する神経変性疾患である。その一方で、知覚神経系は障害されず、感覚器官や視力・聴力、認知機能などの機能低下はほとんど起こらない。そのため患者の多くは自身の身体機能が次第に衰えていくのを日々実感することとなり、その精神的苦痛は計り知れない。
また、MNDは症状が進行すると寝たきりになるため、介護者の身体的・精神的な負担も大きく、また国が負担する医療費も高額になる。現在、例えば、ALSの患者数は国内で約1万人に上り、65歳以上の高齢者の割合が最も高いが、まだまだ働き盛りで子育て世代でもある30~50代も3割以上を占めている。
ALSの病因については、グルタミン酸による神経過剰興奮毒性や酸化ストレスによる神経細胞障害などが考えられている(非特許文献1参照)。また近年は家族性ALS患者の遺伝子解析等から関連因子がいくつか同定されている(非特許文献2参照)。
しかしながら、ALSの発症機序についてはよく分かっていない点も多く、残念ながらALSには未だ有効な治療法が存在しない。現在承認されているのはグルタミン酸放出抑制剤であるリルゾール(商品名リルテック)と抗酸化剤であるエダラボン(商品名ラジカット)の2剤である。いずれも症状の進行を若干遅らせる程度の対症療法薬である。
また、薬剤投与以外でも対症療法しか存在せず、例えば、呼吸障害が起これば人工呼吸器を装着し、嚥下障害が起これば胃ろうや点滴を使用するといった方法しかないのが現状である。
一方、本発明者らは、特定のクルクミン誘導体がタウ凝集抑制作用やセクレターゼ阻害作用、アミロイドβ蛋白質凝集抑制作用を有することを見出し、アルツハイマー病の治療手段として有用であることを報告している(特許文献1、2参照)。

本発明は、新規な運動ニューロン疾患の予防剤及び/又は治療剤、ならびに当該予防剤及び/又は治療剤を含有する医薬組成物に関するものである。
本発明者らは、上記課題を解決するため鋭意検討した結果、特定のクルクミン誘導体が、ALSをはじめとする運動ニューロン疾患の予防及び/又は治療に優れた効果を示すことを見出し、本発明を完成するに至った。
即ち、本発明は、下記〔1〕~〔8〕に関する。

〔7〕 一日一回経口投与されるように用いられる、前記〔1〕~〔6〕いずれかに記載の予防剤及び/又は治療剤。

〔9〕 ヒト又は動物に対して、一般式(I):

本発明の運動ニューロン疾患の予防剤及び/又は治療剤は、抗酸化・抗炎症作用(例えば、酸化ストレスに対する神経保護作用)、グルタミン酸誘発毒性に対する保護作用の両作用を発揮することができる。
また、本発明の運動ニューロン疾患の予防剤及び/又は治療剤は、運動ニューロン疾患に関連する異常凝集性タンパク質(過酸化水素分解酵素:SOD1等)の凝集も抑制することができる。
さらに、本発明の運動ニューロン疾患の予防剤及び/又は治療剤は、前記作用に加えて、運動ニューロンの細胞死を抑制することができ、ひいては運動機能障害の発症及び/又は進行を抑制するという優れた効果を奏する。
本発明の運動ニューロン疾患(MND:motor neuron disease)の予防剤及び/又は治療剤は、一般式(I):
(式中、Rは水素、置換基を有していてもよい鎖状若しくは環状の炭化水素基又は置換基を有していてもよい複素環基を表し、Ar1及びAr2は同一又は異なって、置換基を有してもよい同素環基又は複素環基を表す)
で表される化合物又はその塩を有効成分とすることに特徴を有する。以降、本発明の運動ニューロン疾患の予防剤及び/又は治療剤のことを、単に、本発明の予防剤及び/又は治療剤と記載することもある。

本発明の予防剤及び/又は治療剤は、式(I)で表される化合物を運動ニューロン疾患の代表的疾患であるALSのモデルマウスに投与することにより、運動機能障害が抑制されるという予想外の新知見を見出したことに基づくものである。当該化合物は、本発明者らが既に、アミロイドβ蛋白、タウ蛋白に対し凝集抑制作用を示すことを見出しているが、MNDについては、例えば、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)が典型例として挙げられるが、アミロイドβ、タウ等の蛋白質の異常凝集は認められていない。一方、MND、例えば、ALSにおいては、superoxide dismutase(SOD1)やTAR DNA-binding protein(TDP-43)といった、アミロイドβ、タウとは全く異なる蛋白質の異常凝集によって運動ニューロン機能障害が起こることが知られているが、本発明者らが鋭意検討した結果、式(I)で表される化合物が、意外にも、ALS治療薬として公知のエダラボン(抗酸化剤)やリルゾール(グルタミン酸放出抑制剤)と同じ作用を示しながらも、SOD1をはじめとするタンパク質の異常凝集抑制作用に依る新たなメカニズムに基づいて運動ニューロンの細胞死を抑制するなどしてALSに有効であることが示された。また、公知のALS治療薬の投与経路が点滴静注であるのに対し、本発明の化合物は経口投与時の血中への吸収性および血中から中枢神経組織への移行性を高めるために、前記基本骨格を有する構造において、分子量を小さくし、かつ生体内で抱合体形成や代謝分解を受けにくいよう構造を検討したところ、かかる作用を奏することを見出した。このことから本発明の化合物は経口投与も可能であるため、従来のALS治療薬に比べてより有用であると推察される。
前記一般式(I)におけるRは、水素、置換基を有していてもよい鎖状若しくは環状の炭化水素基又は置換基を有していてもよい複素環基を表し、好ましくは水素を表す。前記鎖状若しくは環状の炭化水素基としては、本発明の効果を妨げない限り特に限定されず、例えば、メチル基、エチル基、プロピル基、イソプロピル基等のアルキル基(好ましくはC1-6)、ビニル基等のアルケニル基(好ましくはC1-6)、フェニル基、ナフチル基等のアリール基(好ましくはC6-12)等を例示できる。前記複素環基としては、本発明の効果を妨げない限り特に限定されず、例えば、1~3個の酸素原子、硫黄原子、窒素原子を含む複素環基等が挙げられ、具体的には、例えば、ピロリル基、ピロリニル基、イミダゾリル基、ピラゾリル基、ピリジル基、フリル基、チエニル基、オキサゾリル基等が挙げられる。また、前記置換基としては、本発明の効果を妨げない限り特に限定されず、通常医薬化合物に適用される、例えば、メチル基、エチル基等のアルキル基(好ましくはC1-6)、ベンジル基等のアラルキル基(好ましくはC7-10)、アセチル基、ベンゾイル基等のアシル基(好ましくはC2-7)、tert-ブトキシカルボニル基等のアルキルカルボニル基(好ましくはC2-7)、メチルスルホニル基、p-トルエンスルホニル基等のアルキルスルホニル基(好ましくはC1-6)、ヒドロキシ基、ニトロ基等を例示できる。
前記一般式(I)においてAr1及びAr2は同一又は異なって、置換基を有してもよい同素環基又は複素環基を表す。「同素環基」としては、本発明の効果を妨げない限り特に限定されず、単環性又は二環性であってもよい。具体的には、ビニル基、フェニル基、ナフチル基等を例示できる。「複素環基」としては、本発明の効果を妨げない限り特に限定されず、単環性又は二環性であってもよい。具体的には、単環性の複素環基としては、ピロリル基、ピロリニル基、イミダゾリル基、ピラゾリル基、ピリジル基、フリル基、チエニル基、オキサゾリル基、二環性の複素環基としては、インドール、イソインドール、ベンズイミダゾール、キノリン、イソキノリン、キノキサリン、クロメン、イソクロメン等を例示できる。また、前記置換基としては、本発明の効果を妨げない限り特に限定されず、例えば、ハロゲン原子、水酸基、ニトロ基、アミノ基、シアノ基、1以上の置換基を有してもよいC1-6アルキル基、1以上の置換基を有してもよいC1-6アルキルオキシ基、1以上の置換基を有してもよいモノ-若しくはジ-C1-6アルキルアミノ基、1以上の置換基を有してもよいC1-6アルキルチオ基、1以上の置換基を有してもよいC1-6アルキルスルホニル基、1以上の置換基を有してもよいC1-6アシル基、1以上の置換基を有してもよいC1-6アシルアミノ基、1以上の置換基を有してもよいC2-6アルケニル基、1以上の置換基を有してもよいC2-6アルケニルオキシ基、1以上の置換基を有してもよいモノ-若しくはジ-C2-6アルケニルアミノ基、1以上の置換基を有してもよいC2-6アルケニルチオ基、又は1以上の置換基を有してもよいカルバモイル基等を例示できる。

(7)ニトロ基
「ニトロ基を有するフェニル基」としては、2-ニトロフェニル基、4-ヒドロキシ-3-ニトロフェニル基、3-メトキシ-4-ニトロフェニル基、5-メトキシ-2-ニトロフェニル基、5-メトキシメトキシ-2-ニトロフェニル基、5-フルオロ-2-ニトロフェニル基、5-ヒドロキシ-2-ニトロフェニル基、5-クロロ-2-ニトロフェニル基、2-クロロ-5-ニトロフェニル基、4,5-ジメトキシ-2-ニトロフェニル基、等を例示できる。
「ニトロ基を有するフェニル基」としては、2-ニトロフェニル基、4-ヒドロキシ-3-ニトロフェニル基、3-メトキシ-4-ニトロフェニル基、5-メトキシ-2-ニトロフェニル基、5-メトキシメトキシ-2-ニトロフェニル基、5-フルオロ-2-ニトロフェニル基、5-ヒドロキシ-2-ニトロフェニル基、5-クロロ-2-ニトロフェニル基、2-クロロ-5-ニトロフェニル基、4,5-ジメトキシ-2-ニトロフェニル基、等を例示できる。
(8)アシル基
「アシル基を有するフェニル基」としては、2-ヒドロキシカルボニルフェニル基、3-ヒドロキシカルボニルフェニル基、2-メトキシカルボニルフェニル基、3-メトキシカルボニルフェニル基、4-メトキシカルボニルフェニル基、2-ジメチルアミノカルボニルフェニル基、3-(ジ-n-プロピルアミノカルボニル)フェニル基等を例示できる。
「アシル基を有するフェニル基」としては、2-ヒドロキシカルボニルフェニル基、3-ヒドロキシカルボニルフェニル基、2-メトキシカルボニルフェニル基、3-メトキシカルボニルフェニル基、4-メトキシカルボニルフェニル基、2-ジメチルアミノカルボニルフェニル基、3-(ジ-n-プロピルアミノカルボニル)フェニル基等を例示できる。
(9)アルキルカルボニルアミノ基等
アルキルカルボニルアミノ基、スルホニル基、スルフィニル基、又はスルホニルオキシ基を有するフェニル基も、上記「置換基を有してもよいフェニル基」に含まれる。
アルキルカルボニルアミノ基、スルホニル基、スルフィニル基、又はスルホニルオキシ基を有するフェニル基も、上記「置換基を有してもよいフェニル基」に含まれる。
上記(1)~(9)の置換基の個数は特に限定されず、同種又は異種の置換基が1種単独で又は複数導入されていてもよい。また、上記(1)~(9)の置換基のフェニル基上の位置も特に限定されず、オルト位、メタ位、パラ位のいずれに導入されていてもよいが、オルト位、パラ位に導入されていることが好ましい。Ar1で表されるフェニル基は、上記(1)~(9)の置換基以外にも、ヒドロキシ基、臭素原子以外のハロゲン原子等の置換基を有してもよい。
前記一般式(I)におけるAr2は、好ましくは置換基を有してもよい同素環基又は複素環基を表し、より好ましくは置換基を有してもよい単環性又は二環性の同素環基又は複素環基を表し、更に好ましくは置換基を有してもよい二環性の複素環基を表す。
「二環性の複素環基」としては、本発明の効果を妨げない限り特に限定されないが、好ましくはインドール-2-イル基、インドール-3-イル基、インドール-4-イル基、インドール-5-イル基、インドール-6-イル基、インドール-7-イル基、ベンゾトリアゾール-5-イル基、ベンゾイミダゾール-5-イル基、キノキサリン-6-イル基、ベンゾフラン-2-イル基、ベンゾチオフェン-2-イル基、1H-インダゾール-5-イル基、7-アザインドール-3-イル基、キノリン-2-イル基、キノリン-5-イル基、キノリン-8-イル基、1,4-ベンゾジオキサン-6-イル基、1,3-ベンゾジオキソール-5-イル基、クロモン-3-イル基、クマリン-6-イル基、7-メトキシクマリン-4-イル基、4-メトキシクマリン-6-イル基、等を例示できる。二環性の複素環基の置換基としては、メチル基、エチル基、ベンジル基、アセチル基、ベンゾイル基、tert-ブトキシカルボニル基、メチルスルホニル基、p-トルエンスルホニル基、ヒドロキシ基、ニトロ基等を例示できる。
「置換基を有してもよい二環性の複素環基」としては、本発明の効果を妨げない限り特に限定されないが、好ましくは無置換の二環性の複素環基のほか、例えば、1-メチル-インドール-6-イル基、1-メチルインドール-2-イル基、1-メチルインドール-3-イル基、1-エチルインドール-6-イル基、1-ベンジルインドール-3-イル基、1-ベンジルインドール-6-イル基、1-アセチルインドール-3-イル基、1-アセチルインドール-6-イル基、1-ベンゾイルインドール-3-イル基、1-tert-ブトキシカルボニルインドール-5-イル基、1-メチルスルホニルインドール-3-イル基、1-メチルスルホニルインドール-6-イル基、1-p-トルエンスルホニルインドール-3-イル基、1-p-トルエンスルホニルインドール-6-イル基、4-ヒドロキシインドール-3-イル基、4-ニトロインドール-3-イル基等、アルキル基(例えば、C1-6等)、アラルキル基(例えばC7-10等)、アシル基(例えば、C2-10等)、ヒドロキシル基、ニトロ基等で置換された二環性の複素環基を例示できる。
前記一般式(I)で表される化合物は、抗酸化・抗炎症作用、グルタミン酸誘発毒性に対する保護作用、SOD1タンパク質異常凝集に対する抑制作用、運動ニューロンの細胞死抑制作用等の点から、以下のような構造をとることが好ましい。
前記一般式(I)においてAr1は、好適には、上記置換基(1)又は(3)を有するフェニル基であり、より好適には、上記置換基(1a)又は(1c)を有するフェニル基、二つの置換基が一緒になって環を形成した二置換アミノ基を有するフェニル基である。具体的には、例えば、2-メトキシ-4-(ピリジン-2-イルメトキシ)フェニル基、2-メトキシ-4-(2-モルホリノエトキシ)フェニル基、4-(ピリジン-2-イルメトキシ)フェニル基、2-フルオロ-4-(ピリジン-2-イルメトキシ)フェニル基、4-メチルピペラジン-1-イル基である。
前記一般式(I)においてAr2は、好適には、単環性又は二環性の複素環基である。具体的には、例えば、置換基を有してもよいピロリル基、置換基を有してもよいピロリニル基、置換基を有してもよいイミダゾリル基、置換基を有してもよいピラゾリル基、置換基を有してもよいインドール-2-イル基、置換基を有してもよいインドール-3-イル基、置換基を有してもよいインドール-4-イル基、置換基を有してもよいインドール-5-イル基、置換基を有してもよいインドール-6-イル基、置換基を有してもよいインドール-7-イル基、置換基を有してもよいベンゾトリアゾール-5-イル基、置換基を有してもよいベンゾイミダゾール-5-イル基、置換基を有してもよいキノキサリン-6-イル基、置換基を有してもよいベンゾフラン-2-イル基、置換基を有してもよいベンゾチオフェン-2-イル基、又は置換基を有してもよいピロール基であり、より好適には、インドール-2-イル基、インドール-3-イル基、インドール-6-イル基、1-メチル-インドール-6-イル基、ベンゾトリアゾール-5-イル基、ベンゾイミダゾール-5-イル基、キノキサリン-6-イル基、ベンゾフラン-2-イル基、ベンゾチオフェン-2-イル基、1-メチル-ピロール-2-イル基であり、更に好適には、1-メチル-ピロリル-2-イル基、インドール-6-イル基、1-メチル-インドール-6-イル基、である。
一般式(I)で表される化合物の具体例としては、例えば、下記から選択される化合物が挙げられる。
また、前記一般式(I)で表される化合物の塩も、本発明に包含される。このような塩としては、薬理学的に許容される塩が好ましく、例えば、フッ化水素酸塩、塩酸塩、臭化水素酸塩、ヨウ化水素酸塩等のハロゲン化水素酸塩;硫酸塩、硝酸塩、過塩素酸塩、リン酸塩、炭酸塩、重炭酸塩等の無機酸塩;酢酸塩、シュウ酸塩、マレイン酸塩、酒石酸塩、フマル酸塩等の有機カルボン酸塩;メタンスルホン酸塩、トリフルオロメタンスルホン酸塩、エタンスルホン酸塩、ベンゼンスルホン酸塩、トルエンスルホン酸塩、カンファースルホン酸塩等の有機スルホン酸塩;アスパラギン酸塩、グルタミン酸塩等のアミノ酸塩;トリメチルアミン塩、トリエチルアミン塩、プロカイン塩、ピリジン塩、フェネチルベンジルアミン塩等のアミンとの塩;ナトリウム塩、カリウム塩等のアルカリ金属塩;マグネシウム塩、カルシウム塩等のアルカリ土類金属塩等があげられ、好ましくは塩酸塩、シュウ酸塩である。
前記一般式(I)で表される化合物は、例えば、Rajeshwar Narlawar et al., ChemMedChem 2008, 3, 165-172、国際公開2008/066151、国際公開2009/145219等に記載されている方法、又はそれらを組み合わせた方法等を参考にして製造することができる。具体的には、以下の工程1及び工程2によって製造することができる。なお、下記式中、Ar1及びAr2は前記と同意義を示す。
工程1は、溶媒及び触媒存在下、一般式(A)で表されるアルデヒドと、一般式(B)で表される化合物を反応させ、一般式(C)で表されるジケトンを得る工程である。一般式(C)で表されるジケトン又はその塩も本発明に包含される。なお、上記式中では、一般式(A)で表されるアルデヒドがAr1を有し、一般式(B)で表される化合物がAr2を有しているが、Ar2を有する一般式(A)で表されるアルデヒドとAr1を有する一般式(B)で表される化合物とを反応させて、一般式(C)で表されるジケトンを得てもよい。
工程1で使用される一般式(A)で表されるアルデヒドや一般式(B)で表される化合物は、市販品や公知の方法(例えば、国際公開2008/066151、国際公開2009/145219に記載されている方法)によって合成されたものを使用することができる。なお、工程1での原料化合物の使用量やその反応条件は、公知技術に従って適宜設定することができる。また、溶媒及び触媒、必要により助剤も特に限定されず、公知のものから適宜選択して用いることができる。
工程2は、溶媒存在下、一般式(C)で表されるジケトンとヒドラジン又はその誘導体を反応させ、前記一般式(I)で表される化合物を得る工程である。工程2は、目的とする化合物に応じて、前記一般式(C)で表されるジケトンとヒドラジン又はその誘導体(H2N-NHRで示される化合物又はその塩等)とを反応させて得られた化合物に、置換基を付加する工程を有していてもよい。
使用されるヒドラジンは特に限定されず、例えば、ヒドラジン一水和物、ヒドラジン水溶液、無水ヒドラジン、ヒドラジン酢酸塩、ヒドラジン一塩酸塩又はこれらの誘導体等が挙げられる。なお、工程2での原料化合物の使用量やその反応条件は、公知技術に従って適宜設定することができる。また、溶媒、必要により触媒及び助剤も特に限定されず、公知のものから適宜選択して用いることができる。
本発明の予防剤及び/又は治療剤は、一般式(I)で表される化合物又はその塩を含有するのであれば、その他の成分は特に限定されない。一般式(I)で表される化合物又はその塩は単独で又は2種以上組み合わせて用いることができ、本発明の予防剤及び/又は治療剤における一般式(I)で表される化合物又はその塩の含有量としては通常0.00001~100重量%程度である。
本発明の一般式(I)で表される化合物は、抗酸化・抗炎症作用(例えば、活性酸素に基づく酸化・炎症を抑制する作用)、グルタミン酸誘発毒性に対する保護作用、SOD1タンパク質異常凝集に対する抑制作用、運動ニューロン細胞死の抑制作用、などを有することにより、MNDやそれに基づく症状の予防剤、改善剤、及び/又は治療剤として好適に用いることができる。具体的には、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)をはじめとするMNDやそれに基づく症状の予防剤、改善剤、及び/又は治療剤として好適に用いることができる。また、運動障害を発症する疾患、例えば、進行性核上性麻痺(PSP:progressive supranuclear palsy)にも好適に用いることができる。
本発明の予防剤及び/又は治療剤は、一般式(I)で表される化合物又はその塩を体内に摂取できるのであれば、その形態は限定されない。例えば、錠剤、散剤、顆粒剤、カプセル剤、液剤、乳剤、エリキシル剤、懸濁剤、シロップ剤、トローチ剤、吸入剤、坐剤、注射剤、軟膏剤、眼軟膏剤、点眼剤、点鼻剤、点耳剤、パップ剤、ローション剤等の経口製剤又は非経口製剤等の剤として製剤化することができる。血中への吸収性や血中からの中枢神経組織への移行性が高いことから、経口製剤として製剤化してもよい。
現在上市されているMND、特にALSの治療薬の副作用としては、肝機能障害や腎障害等がよく知られている。そのため、副作用の程度を確認しながら治療を行うが、対症療法薬であるために症状の進行を止めることはできず長期間の投薬が必要となる。その間に上記副作用や症状の進行によって、最終的にはALS治療剤を使用し続けることが難しくなり、治療を中止せざるを得なくなることもある。そのため、副作用を予防又は軽減する対策が重要となってくる。本発明の化合物は、詳細なメカニズムは不明なるも、血中滞留時間が長く、また、安全性が高く、毒性や変異原性も低いことから、経口製剤として開発することが可能であり、MNDの初期症状に対する予防剤及び/治療剤として好適であると示唆される。
本発明の予防剤及び/又は治療剤の投与量(有効量ともいう)は、その形態や投与目的、当該剤の投与対象の種類、年齢、体重、症状によって適宜設定され一定ではない。例えば、本発明における一般式(I)で表される化合物又はその塩の有効ヒト投与量としては、一日当たり、0.01~100mg/kg程度が好ましく、1~50mg/kg程度がより好ましい。投与は、所望の投与量範囲内において、1日内において単回で又は数回に分けて行ってもよい。投与期間も任意である。例えば、一日一回経口投与される態様が例示される。
本発明の予防剤及び/又は治療剤の投与対象とは、好ましくは筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)をはじめとする運動ニューロン疾患や運動障害を発症する疾患、例えば、進行性核上性麻痺(PSP:progressive supranuclear palsy)の治療を必要とするヒトであるが、ウシ、ウマ、ヤギ等の家畜動物、イヌ、ネコ、ウサギ等のペット動物、又は、マウス、ラット、モルモット、サル等の実験動物等の動物であってもよい。また投与対象は、経口摂取可能な観点から、在宅での治療を主とする初期症状を呈する患者から入院・全身介助を必要とする中期・末期の患者に至るまで幅広く挙げられる。
また、本発明は、本発明の予防剤及び/又は治療剤を含有する医薬組成物を提供する。該組成物の形態としては、一般式(I)で表される化合物又はその塩を体内に取り込むことができる形態であれば特に限定はなく、例えば、外用剤組成物や内服剤組成物としての形態が例示される。
具体的には、錠剤、散剤、細粒剤、顆粒剤、被覆錠剤、カプセル剤、液剤、乳剤、エリキシル剤、懸濁剤、シロップ剤、トローチ剤、吸入剤、坐剤、注射剤、軟膏剤、眼軟膏剤、点眼剤、点鼻剤、点耳剤、パップ剤、ローション剤等の剤に製剤化することができる。
上記形態を有する本発明の医薬組成物は、一般式(I)で表される化合物又はその塩を含有するものであれば、常法に従って、製剤分野等において通常使用される担体、基剤、及び/又は添加剤等を本発明の目的を達成する範囲内で適宜配合して調製することができる。具体的には、例えば、賦形剤、結合剤、滑沢剤、着色剤、矯味矯臭剤や、及び必要により安定化剤、乳化剤、吸収促進剤、界面活性剤、pH調製剤、防腐剤、抗酸化剤等を使用することができ、一般に医薬品製剤の原料として用いられる成分を、本発明の効果を損なわない範囲内で適量配合して常法により製剤化される。一般式(I)で表される化合物又はその塩の含有量は、剤型、投与方法、担体等により異なるが、本発明の医薬組成物中、通常0.01~100重量%、好ましくは0.1~95重量%である。
本発明の一般式(I)で表される化合物又はその塩は、運動ニューロン疾患全般に亘って適用できるが、その一態様として、例えば、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)の治療方法に利用できる。具体的な方法としては、以下の(a)~(d)を例示できる。また、下記(e)の態様も、本発明の一態様として挙げることができる。
(a)前記一般式(I)で表される化合物又はその塩を、筋萎縮性側索硬化症の患者に投与する工程を含む、筋萎縮性側索硬化症の治療方法。
(b)前記一般式(I)で表される化合物又はその塩を、原発性側索硬化症の患者に投与する工程を含む、原発性側索硬化症の治療方法。
(c)前記一般式(I)で表される化合物又はその塩を、脊髄性進行性筋萎縮症の患者に投与する工程を含む、脊髄性進行性筋萎縮症の治療方法。
(d)前記一般式(I)で表される化合物又はその塩を、進行性球麻痺の患者に投与する工程を含む、進行性球麻痺の治療方法。
(e)前記一般式(I)で表される化合物又はその塩を、進行性核上性麻痺の患者に投与する工程を含む、進行性核上性麻痺の治療方法。
(a)前記一般式(I)で表される化合物又はその塩を、筋萎縮性側索硬化症の患者に投与する工程を含む、筋萎縮性側索硬化症の治療方法。
(b)前記一般式(I)で表される化合物又はその塩を、原発性側索硬化症の患者に投与する工程を含む、原発性側索硬化症の治療方法。
(c)前記一般式(I)で表される化合物又はその塩を、脊髄性進行性筋萎縮症の患者に投与する工程を含む、脊髄性進行性筋萎縮症の治療方法。
(d)前記一般式(I)で表される化合物又はその塩を、進行性球麻痺の患者に投与する工程を含む、進行性球麻痺の治療方法。
(e)前記一般式(I)で表される化合物又はその塩を、進行性核上性麻痺の患者に投与する工程を含む、進行性核上性麻痺の治療方法。
また、本発明の一態様として、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)、進行性核上性麻痺(PSP:progressive supranuclear palsy)の予防及び/又は治療するための、一般式(I)で表される化合物又はその塩の使用を提供する。また、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)、進行性核上性麻痺(PSP:progressive supranuclear palsy)の予防剤及び/又は治療剤を製造するための、一般式(I)で表される化合物又はその塩の使用を提供する。
以下、実施例により本発明を詳細に説明するが、本発明はこれらに限定されるものではない。なお、以降の試験で用いた化合物(実施例1:化合物A)は、下記に示す構造を有するものであり、WO2012/141228号に記載の方法に従って調製した。
試験例1
酸化ストレスに対する保護作用を検証するため、神経細胞様に分化することが知られているラット副腎由来褐色細胞腫PC12細胞を、過酸化水素(300μM)と化合物A(3、10、30μM)で同時処置し、24時間後に培地中の乳酸脱水素酵素(LDH)を定量した。結果を図1に示す。なお、過酸化水素と化合物Aの処理がないものをControl(CTL)、過酸化水素のみで処理したものを陰性Control(-)として同様に評価を行い、CTLに対する相対LDH量(%)として表した。

図1より、化合物Aは過酸化水素による神経細胞死を濃度依存的に抑制することが分かった(図中、平均±標準偏差で示す;##:P<0.01 vs CTL by Mann-Whitney U test;*:P<0.05, **:P<0.01 vs 陰性Control(-) by Dunnett test)。
試験例2
炎症や小胞体ストレス応答の経路であるNrf2(NFE2 related factor 2)経路の活性化を評価するため、化合物Aで抗酸化剤応答配列(ARE、Nrf2の結合配列)-ルシフェラーゼ発現PC12細胞(Izumi et al., Isolation, identification, and biological evaluation of Nrf2-ARE activator from the leaves of green perilla (Perilla frutescens var. crispa f. viridis). Free Radic Biol Med. 2012, 53(4):669-679.参照)を処置した。結果を図2に示す。なお、化合物Aの処理がないものをControl(CTL)として同様に評価を行い、CTLに対する相対ルシフェラーゼ発現量(%)として表した。
炎症や小胞体ストレス応答の経路であるNrf2(NFE2 related factor 2)経路の活性化を評価するため、化合物Aで抗酸化剤応答配列(ARE、Nrf2の結合配列)-ルシフェラーゼ発現PC12細胞(Izumi et al., Isolation, identification, and biological evaluation of Nrf2-ARE activator from the leaves of green perilla (Perilla frutescens var. crispa f. viridis). Free Radic Biol Med. 2012, 53(4):669-679.参照)を処置した。結果を図2に示す。なお、化合物Aの処理がないものをControl(CTL)として同様に評価を行い、CTLに対する相対ルシフェラーゼ発現量(%)として表した。
図2より、化合物Aは濃度依存的にNrf2経路を活性化しており、酸化ストレス・炎症への耐性を高めることが示唆された(図中、平均±標準偏差で示す;*:P<0.05, ***:P<0.001 vs CTL by Dunnett test)。
試験例3
ラット脳初代培養細胞を30μMグルタミン酸で処置し、同時に化合物Aを0.3~10μMで処置した後、24時間後に培地中のLDHを定量した。結果を図3に示す。なお、グルタミン酸と化合物Aの処理がないものをControl(CTL)、グルタミン酸のみで処理したものを陰性Control(-)として同様に評価を行い、CTLに対する相対LDH量(%)として表した。

図3より、化合物Aはグルタミン酸による神経細胞死を濃度依存的に抑制することが分かった(図中、平均±標準偏差で示す;##:P<0.01 vs CTL by Mann-Whitney U test;*:P<0.05, **:P<0.01 vs 陰性Control(-) by Dunnett test)。
試験例4
ALSモデル動物のSOD1 H46R Tgマウス(オス、n=20~25、東北大学医学部 神経内科学講座より提供;Chang-Hong et al.,, Exp Neurol. 2005 194(1):203-11参照)の10週齢に、化合物Aを10mg/kg/day又は40mg/kg/dayで1日1回経口投与し、ロータロッド試験(条件:20回転/分、420秒)でローターから落下するまでの秒数を計測し、運動機能障害の発症及び進行を評価した。結果を図4及び図5に示す。
ALSモデル動物のSOD1 H46R Tgマウス(オス、n=20~25、東北大学医学部 神経内科学講座より提供;Chang-Hong et al.,, Exp Neurol. 2005 194(1):203-11参照)の10週齢に、化合物Aを10mg/kg/day又は40mg/kg/dayで1日1回経口投与し、ロータロッド試験(条件:20回転/分、420秒)でローターから落下するまでの秒数を計測し、運動機能障害の発症及び進行を評価した。結果を図4及び図5に示す。
図4及び図5より、10mg/kg/day、40mg/kg/dayの化合物Aを投与した両群とも、Control群と比較して運動機能障害の発症及び進行が有意に抑制された。
試験例5
SOD1 H46R Tgマウス(オス、10週齢)に化合物Aを40mg/kg/dayで1日1回経口投与し続け、20週齢または24週齢になったマウスの脊髄を摘出し(n=3~4)、厚さ30μmの凍結切片(連続切片)を作製した。各切片をニッスル染色し、その中から第3~第5腰髄の12切片毎(360μm毎)の計10枚について、Stereo Investigatorを用いて、脊髄前角大型ニューロン(下位運動ニューロン)をカウントした。脊髄前角大型ニューロンは、長径20μm以上で核小体を確認できる細胞と定義し、10切片の合計値を1個体の細胞数として算出し、化合物A投与群とControl群の細胞数を比較検討した。その結果を図6に示す。
SOD1 H46R Tgマウス(オス、10週齢)に化合物Aを40mg/kg/dayで1日1回経口投与し続け、20週齢または24週齢になったマウスの脊髄を摘出し(n=3~4)、厚さ30μmの凍結切片(連続切片)を作製した。各切片をニッスル染色し、その中から第3~第5腰髄の12切片毎(360μm毎)の計10枚について、Stereo Investigatorを用いて、脊髄前角大型ニューロン(下位運動ニューロン)をカウントした。脊髄前角大型ニューロンは、長径20μm以上で核小体を確認できる細胞と定義し、10切片の合計値を1個体の細胞数として算出し、化合物A投与群とControl群の細胞数を比較検討した。その結果を図6に示す。
図6より、化合物Aを投与した群ではControl群と比較して脊髄中の運動神経ニューロンの細胞数減少が有意に抑制された(図中、平均±標準誤差で示す;*:P<0.05, **:P<0.01 by Student’s t-test)。
試験例6
SOD1 H46R Tgマウス(オス、10週齢)に化合物Aを40mg/kg/dayで1日1回経口投与し続け、20週齢または24週齢になったマウスの脊髄を摘出し(n=9)、湿重量の10倍量のリン酸緩衝生理食塩水(PBS)を加えて超音波破砕した。破砕後、以降の抽出を行なってタンパク質を抽出し、凝集度の低いものを含む画分から高いものを含む画分へと順に5段階の抽出画分を得た。具体的には、100,000g・4℃で20分間遠心し、その遠心上清をPBS可溶性画分(PBS-Soluble)として回収した。遠心後の沈殿に1% Triton X-100を含有するPBSを最初のPBSと等量加えて超音波破砕し、100,000g・4℃で20分間遠心し、その遠心上清をTriton可溶性画分(Triton-Soluble)として回収した。遠心後の沈殿に5% SDSを含有するPBSを最初のPBSと等量加えて超音波破砕し、100,000g・20℃で20分間遠心し、その遠心上清をSDS可溶性画分(SDS-Soluble)として回収した。遠心後の沈殿に8M 尿素を含むPBSを最初のPBSと等量加えて超音波破砕し、100,000g・20℃で20分間遠心し、その遠心上清を尿素可溶性画分(Urea-Soluble)として回収した。遠心後の沈殿に88% ギ酸を最初のPBSと等量加えて超音波破砕した後、1M トリス・HCl(pH=10)を加えて中和し、不溶性画分(Insoluble)として回収した。
SOD1 H46R Tgマウス(オス、10週齢)に化合物Aを40mg/kg/dayで1日1回経口投与し続け、20週齢または24週齢になったマウスの脊髄を摘出し(n=9)、湿重量の10倍量のリン酸緩衝生理食塩水(PBS)を加えて超音波破砕した。破砕後、以降の抽出を行なってタンパク質を抽出し、凝集度の低いものを含む画分から高いものを含む画分へと順に5段階の抽出画分を得た。具体的には、100,000g・4℃で20分間遠心し、その遠心上清をPBS可溶性画分(PBS-Soluble)として回収した。遠心後の沈殿に1% Triton X-100を含有するPBSを最初のPBSと等量加えて超音波破砕し、100,000g・4℃で20分間遠心し、その遠心上清をTriton可溶性画分(Triton-Soluble)として回収した。遠心後の沈殿に5% SDSを含有するPBSを最初のPBSと等量加えて超音波破砕し、100,000g・20℃で20分間遠心し、その遠心上清をSDS可溶性画分(SDS-Soluble)として回収した。遠心後の沈殿に8M 尿素を含むPBSを最初のPBSと等量加えて超音波破砕し、100,000g・20℃で20分間遠心し、その遠心上清を尿素可溶性画分(Urea-Soluble)として回収した。遠心後の沈殿に88% ギ酸を最初のPBSと等量加えて超音波破砕した後、1M トリス・HCl(pH=10)を加えて中和し、不溶性画分(Insoluble)として回収した。
各サンプルに10%β-メルカプトエタノール・4.3%ドデシル硫酸ナトリウムを含有するサンプル処理液を等量加えて100℃・10分間加熱した後、15%ポリアクリルアミドゲルで250V・45分間電気泳動した。泳動後、ポリフッ化ビニリデンメンブレンに15V・30分間転写し、2.5%スキムミルクを溶解した0.1% Tween20含有トリス緩衝生理食塩水(T-TBS)で1時間ブロッキングした。ブロッキング後、GeneTex製抗SOD1抗体(T-TBSで1000倍希釈)で4℃・一晩インキュベートし、T-TBSで10分間×3回洗浄した。さらにHRP標識抗ウサギIgG抗体(T-TBSで3000倍希釈)で室温・1時間インキュベートし、T-TBSで10分間×3回洗浄した。洗浄後、HRP用化学発光検出薬を用いてバンドを検出・定量し、Control群に対する相対SOD1量(%)として表した。その結果を図7に示す。
図7より、化合物Aを投与した群ではControl群と比較して不溶性画分におけるSOD1凝集体の量が有意に低下していた(図中、平均±標準誤差で示す;*:P<0.05, by Student’s t-test)。また、尿素可溶性画分やSDS可溶性画分においても、化合物Aを投与した群ではControl群と比較してSOD1凝集体の減少傾向がみられた。一方、未凝集のSOD1を多く含むPBS可溶性画分やTriton可溶性画分においてはSOD1量に大きな差はなかった。このことから、化合物A投与によるSOD1凝集体の減少は、SOD1産生抑制作用というよりはSOD1凝集抑制作用が大きく関与するものと示唆される。
本発明の予防剤及び/又は治療剤は、抗酸化・抗炎症作用やグルタミン酸誘発毒性に対する保護作用の他、運動ニューロン細胞死の抑制作用、SOD1タンパク質異常凝集に対する抑制作用を有し、運動ニューロン疾患、なかでも筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、進行性球麻痺(PBP:progressive bulbar palsy)の予防剤及び/又は治療剤として好適に用いることができる。
Claims (8)
- 式(I)のRが、水素である、請求項1に記載の予防剤及び/又は治療剤。
- 式(I)のAr1が、置換基を有してもよいフェニル基である、請求項1又は2に記載の予防剤及び/又は治療剤。
- 置換基が、置換基を有してもよいヘテロシクロアルキル基で置換されたC1-3アルキルオキシ基、置換基を有してもよいシクロアルキル基で置換されたC1-3アルキルオキシ基、置換基を有してもよいヘテロアリール基で置換されたC1-3アルキルオキシ基、置換基を有してもよいアリール基で置換されたC1-3アルキルオキシ基、置換基を有してもよいジアルキルアミノ基で置換されたC1-3アルキルオキシ基、置換基を有してもよいアルキルオキシ基で置換されたC1-3アルキルオキシ基、置換基を有してもよいアルキル基で置換されたC1-3アルキルオキシ基、無置換のC1-3アルキルオキシ基、及び二置換アミノ基から選ばれる1種又は2種以上である、請求項3に記載の予防剤及び/又は治療剤。
- 式(I)のAr2が、複素環基である、請求項1~4いずれかに記載の予防剤及び/又は治療剤。
- 運動ニューロン疾患が、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、原発性側索硬化症(PLS:primary lateral sclerosis)、脊髄性進行性筋萎縮症(SPMA:spinal progressive muscular atrophy)、又は進行性球麻痺(PBP:progressive bulbar palsy)である、請求項1~5いずれかに記載の予防剤及び/又は治療剤。
- 一日一回経口投与されるように用いられる、請求項1~6いずれかに記載の予防剤及び/又は治療剤。
- 請求項1~7いずれかに記載の予防剤及び/又は治療剤を含有してなる医薬組成物。
