Abstract
본 발명의 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효 성분으로 이용 시, 동물모델의 행동학적 척도를 개선하고, 미세아교세포의 활성화 및 염증인자(COX-2 및 iNOS)의 생성을 억제하면서, 세포-신경 자극에 대한 반응성에 대한 시경 흥분 정도를 낮추는 효과가 존재하는 바, 스트레스 조절능이 우수하고 이에 따른 스트레스성 질환, 특히 우울증의 예방 및 치료에 유용함을 확인하여, 관련 산업에 유용하게 이용될 수 있다.
Description
본 발명은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 치료용 조성물에 관한 것이다.
인간이 내·외적 환경의 자극에 의해 신체적·심리적인 스트레스를 받게 되면 인체는 스트레스에 적응하기 위해 뇌에서 청반 노르에피네프린(LC-NE: locus ceruleus norepinephrine)과 부신피질 자극 호르몬 유리 호르몬(CRH: corticotropin releasing hormone)을 분비한다. LC-NE는 교감 신경계를 활성화시키고 부교감 신경계의 작용을 억제하여 혈압, 심박동, 호흡을 증가시키며, 기관지를 확장시킬 뿐 아니라 피부에 소름이 끼치면서 털이 일어나는 발한 작용을 증가시킨다. 시상하부에서 분비된 CRH는 뇌하수체 전엽으로부터 부신피질 자극 호르몬(Adrenocorticotropic hormone: ACTH)을 일반 체순환으로 방출한다. 이 ACTH가 혈액을 따라 옮겨가서 부신피질의 ACTH 수용체에 결합하게 되면, cAMP라고 불리는 2차 신호전달자를 합성하게 된다. cAMP에 의해 활성화된 단백질 인산화 효소PKA(protein kinase A)는 미토콘드리아에 위치한 당질코르티코이드(Glucocorticoid) 합성 단백질을 활성화시킨다. 그러면 당질코르티코이드 합성 단백질이 콜레스테롤을 이용하여 스트레스 호르몬들을 만들어 낸다. 코르티졸(cortisol), 코르티코스테론(corticosterone), 코르티손(cortisone)등이 대표적인 당질 코르티코이드의 예이다. CRH-ACTH-당질 코르티코이드까지 연결되는 스트레스 경로를 일반적으로 시상하부-뇌하수체-부신(Hypothalamus-Pituitary gland-Adrenal gland: HPA) 축이라고 한다. 당질코르티코이드는 간에서 포도당 신합성(gluconeogenesis)과 글리코겐의 저장, 혈당치 상승 등의 탄수화물대사조절을 한다. 그밖에 당질코르티코이드는 면역을 담당하는 면역세포의 각종 면역물질 생성 유전자의 발현을 억제함으로써 항염증 작용을 한다. 또한 혈액 내 이 호르몬 함량이 높아지면 음성 피드백(negative feedback)으로 뇌하수체에 작용하여 ACTH 분비를 억제한다.
신체가 허용할 수 있는 수준의 스트레스는 우리 몸의 적절한 자극제로 작용을 하게 되지만, 허용 범위를 벗어난 강하고도 지속적으로 가해지는 심리적 육체적 스트레스는 당질 코르티코이드를 장시간 동안 과량 분비하게 유도한다. 이렇게 장기간 과량 분비되는 당질 코르티코이드는 몸의 면역력을 떨어뜨리게 되므로, 외부 감염에 의한 저항성을 떨어뜨리게 되고, 우리 몸에 다양한 질병을 유발하게 된다. 면역력 감퇴뿐만 아니라 특별히 뇌세포 손상 및 재생을 억제하여 불면증, 무기력증, 기억력 감퇴, 우울증 유발은 물론 자살 충동까지 일으킨다.
우울증을 치료하기 위한 치료제로서 1960년대 개발된 삼환계 약물을 필두로하여 현재는 세로토닌 재흡수 억제제로 알려진 프로작, 졸로푸트, 세로자트, 이팩사, 레메론 등 다양한 종류의 항우울증 치료제가 개발되어 왔다. 또한 이와 같은 항 우울증 치료제와 더불어 스트레스 반응의 1차 물질인 CRH가 결합하는 CRH 수용체 억제제 개발 연구 또한 많이 이루어지고 있으나, 이에 따른 부작용이 보고되고 있다.
이에 본 발명자들은 보닐 아세테이트 또는 이의 약학적으로 허용 가능한 염을 유효성분으로 이용 시, 동물모델의 행동학적 척도를 개선하고, 미세아교세포의 활성화 및 염증인자(COX-2 및 iNOS)의 생성을 억제하면서, 세포-신경 자극에 대한 반응성에 대한 시경 흥분 정도를 낮추는 효과가 존재하는 바, 스트레스 조절능이 우수하고 이에 따른 스트레스성 질환, 특히 우울증의 예방 및 치료에 유용하다는 것을 확인하여, 본 발명을 완성하였다.
본 발명의 목적은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 치료용 약학적 조성물을 제공할 수 있다.
또한 본 발명의 목적은 보닐 아세테이트 또는 이의 약학적으로 허용가능한염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 개선용 식품 조성물을 제공할 수 있다.
또한 본 발명의 목적은 약학적으로 유효한 양의 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 스트레스성 질환의 예방 및 치료방법을 제공할 수 있다.
상기 목적의 달성을 위해, 본 발명은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
또한 본 발명은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 개선용 식품 조성물을 제공한다.
또한 본 발명은 약학적으로 유효한 양의 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 스트레스성 질환의 예방 및 치료방법을 제공한다.

본 발명은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
본 명세서에서 상기 "예방"이란 상기 약학적 조성물의 투여로 스트레스성 질환을 억제 또는 지연시키는 모든 행위를 의미한다. 또한, 상기 "치료"란 상기 약학적 조성물의 투여로 스트레스성 질환의 증세가 호전되거나 이롭게 변경하는 모든 행위를 의미한다.
상기 보닐 아세테이트는 하기의 화학식 1로 표시될 수 있고, 상기 보닐 아세테이트는 유도체를 포함할 수 있으며, 약학적으로 허용 가능한 염을 더 포함할 수 있다.

상기 스트레스성 질환은 우울증, 불안, 피로증후군, 신경변성 질환, 섭식장애, 신경성 식욕부진, 약물중독, 약물 및 알코올 금단 증후군 및 스트레스성 정신병 에피소드로 이루어진 군이나, 바람직하게는 우울증이고, 이에 제한되지 않는다.
상기 본 발명의 보닐 아세테이트를 유효성분으로 이용 시, 만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델에서 체중 변화, 꼬리매달기 검사, 강제 수영검사 및 혈청 스트레스 호르몬(코르티코스테론) 수치 변화를 통한 행동학적 척도를 개선함을 확인하였다. 또한, 미세아교세포의 활성화를 억제하고, 염증인자(COX-2 및 iNOS)의 생성을 억제하면서, 세포-신경 자극에 대한 반응성에 대한 시경 흥분 정도를 낮추는 효과가 있어, 스트레스 조절능이 우수하고 이에 따른 스트레스성 질환, 특히 우울증의 예방 및 치료에 효과적이다.
상기 염으로는 약학적으로 허용 가능한 유리산(free acid)에 의해 형성된 산부가염이 유용하다. 산 부가염은 염산, 질산, 인산, 황산, 브롬화수소산, 요오드화수소산, 아질산 또는 아인산과 같은 무기산류와 지방족 모노 및 디카르복실레이트, 페닐-치환된 알카노에이트, 하이드록시 알카노에이트 및 알칸디오에이트, 방향족 산류, 지방족 및 방향족 술폰산류와 같은 무독성 유기산, 아세트산, 안식향산, 구연산, 젖산, 말레인산, 글루콘산, 메탄설폰산, 4-톨루엔술폰산, 주석산, 푸마르산과 같은 유기산으로부터 얻는다. 이러한 약학적으로 무독한 염류로는 설페이트, 피로설페이트, 바이설페이트, 설파이트, 바이설파이트, 니트레이트, 포스페이트, 모노하이드로겐 포스페이트, 디하이드로겐 포스페이트, 메타포스페이트, 피로포스페이트 클로라이드, 브로마이드, 아이오다이드, 플루오라이드, 아세테이트, 프로피오네이트, 데카노에이트, 카프릴레이트, 아크릴레이트, 포메이트, 이소부티레이트, 카프레이트, 헵타노에이트, 프로피올레이트, 옥살레이트, 말로네이트, 석시네이트, 수베레이트, 세바케이트, 푸마레이트, 말리에이트, 부틴-1,4-디오에이트, 헥산-1,6-디오에이트, 벤조에이트, 클로로벤조에이트, 메틸벤조에이트, 디니트로 벤조에이트, 하이드록시벤조에이트, 메톡시벤조에이트, 프탈레이트, 테레프탈레이트, 벤젠설포네이트, 톨루엔설포네이트, 클로로벤젠설포네이트, 크실렌설포네이트, 페닐아세테이트, 페닐프로피오네이트, 페닐부티레이트, 시트레이트, 락테이트, 하이드록시부티레이트, 글리콜레이트, 말레이트, 타트레이트, 메탄설포네이트, 프로판설포네이트, 나프탈렌-1-설포네이트, 나프탈렌-2-설포네이트 또는 만델레이트를 포함한다.
본 발명에 따른 약학 조성물은 유효성분을 약학적으로 허용된 담체에 혼입시킨 형태로 제조될 수 있다. 여기서, 약학적으로 허용된 담체는 제약 분야에서 통상 사용되는 담체, 부형제 및 희석제를 포함한다. 본 발명의 약학 조성물에 이용할 수 있는 약학적으로 허용된 담체는 이들로 제한되는 것은 아니지만, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로스, 메틸 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
본 발명의 약학 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀전, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 또는 멸균주사용액의 형태로 제형화하여 사용될 수 있다.
제제화할 경우에는 통상 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제될 수 있다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 그러한 고형 제제는 유효성분에 적어도 하나 이상의 부형제, 예를 들면 전분, 칼슘 카르보네이트, 수크로스, 락토오스, 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용될 수 있다. 경구투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데, 일반적으로 사용되는 희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수용성용제, 현탁제, 유제, 동결건조 제제 및 좌제가 포함된다. 비수용성용제, 현탁제로는 프로필렌 글리콜, 폴리에틸렌 글리콜, 올리브유와 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 트윈(tween) 61, 카카오지, 라우린지, 글리세로 젤라틴 등이 사용될 수 있다.
본 발명에 따른 약학 조성물은 개체에 다양한 경로로 투여될 수 있다. 투여의 모든 방식이 예상될 수 있는데, 예를 들면 경구, 정맥, 근육, 피하, 복강내 주사에 의해 투여될 수 있다.
본 발명에 따른 약학 조성물의 투여량은 개체의 연령, 체중, 성별, 신체 상태 등을 고려하여 선택된다. 상기 약학 조성물 중 포함되는 유효성분의 농도는 대상에 따라 다양하게 선택할 수 있음은 자명하며, 바람직하게는 약학 조성물에 0.01 ~ 5,000 ㎍/ml의 농도로 포함되는 것이다. 그 농도가 0.01 ㎍/ml 미만일 경우에는 약학 활성이 나타나지 않을 수 있고, 5,000 ㎍/ml를 초과할 경우에는 인체에 독성을 나타낼 수 있다.
또한 본 발명은 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 개선용 식품 조성물을 제공한다.
본 발명의 식품 조성물은 유효성분인 보닐 아세테이트를 함유하는 것 외에 통상의 식품 조성물과 같이 여러 가지 향미제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있다.
상술한 천연 탄수화물의 예는 모노사카라이드, 예를 들어, 포도당, 과당 등; 디사카라이드, 예를 들어 말토스, 슈크로스 등; 및 폴리사카라이드, 예를 들어 덱스트린, 시클로덱스트린 등과 같은 통상적인 당, 및 자일리톨,소르비톨, 에리트리톨 등의 당알콜이다. 상술한 향미제는 천연 향미제(타우마틴), 스테비아 추출물(예를 들어 레바우디오시드 A, 글리시르히진 등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다. 본 발명의 식품 조성물은 상기 약학적 조성물과 동일한 방식으로 제제화되어 기능성 식품으로 이용하거나, 각종 식품에 첨가할 수 있다. 본 발명의 조성물을 첨가할 수 있는 식품으로는 예를 들어, 음료류, 육류, 초코렛, 식품류, 과자류, 피자, 라면, 기타 면류, 껌류, 사탕류, 아이스크림류, 알코올 음료류, 비타민 복합제 및 건강보조식품류 등이 있다.
또한 상기 식품 조성물은 유효성분인 추출물 외에 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제(치즈, 초콜릿 등), 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알콜, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다. 그밖에 본 발명의 식품 조성물은 천연 과일 쥬스 및 과일 쥬스 음료 및 야채 음료의 제조를 위한 과육을 함유할 수 있다.
본 발명의 기능성 식품 조성물은 정제, 캅셀, 분말, 과립, 액상, 환 등의 형태로 제조 및 가공될 수 있다. 본 발명에서 '건강기능성 식품 조성물'이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 말하며, 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다. 본 발명의 건강기능식품은 통상의 식품 첨가물을 포함할 수 있으며, 식품 첨가물로서의 적합 여부는 다른 규정이 없는 한, 식품의약품안전청에 승인된 식품 첨가물 공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다. 상기 '식품 첨가물 공전'에 수재된 품목으로는 예를 들어, 케톤류, 글리신, 구연산칼슘, 니코틴산, 계피산 등의 화학적 합성물; 감색소, 감초추출물, 결정셀룰로오스, 고량색소, 구아검 등의 천연첨가물; L-글루타민산나트륨 제제, 면류첨가알칼리제, 보존료 제제, 타르색소제제 등의 혼합 제제류 등을 들 수 있다. 예를 들어, 정제 형태의 건강기능식품은 본 발명의 유효성분을 부형제, 결합제, 붕해제 및 다른 첨가제와 혼합한 혼합물을 통상의 방법으로 과립화한 다음, 활택제 등을 넣어 압축 성형하거나, 상기 혼합물을 직접 압축 성형할 수 있다. 또한 상기 정제 형태의 건강기능식품은 필요에 따라 교미제 등을 함유할 수도 있다. 캅셀 형태의 건강기능식품 중 경질 캅셀제는 통상의 경질 캅셀에 본 발명의 유효성분을 부형제 등의 첨가제와 혼합한 혼합물을 충진하여 제조할 수 있으며, 연질 캅셀제는 본 발명의 유효성분을 부형제 등의 첨가제와 혼합한 혼합물을 젤라틴과 같은 캅셀기제에 충진하여 제조할 수 있다. 상기 연질 캅셀제는 필요에 따라 글리세린 또는 소르비톨 등의 가소제, 착색제, 보존제 등을 함유할 수 있다. 환 형태의 건강기능식품은 본 발명의 유효성분과 부형제, 결합제, 붕해제 등을 혼합한 혼합물을 기존에 공지된 방법으로 성형하여 조제할 수 있으며, 필요에 따라 백당이나 다른 제피제로 제피할 수 있으며, 또는 전분, 탈크와 같은 물질로 표면을 코팅할 수도 있다. 과립 형태의 건강기능식품은 본 발명의 유효성분의 부형제, 결합제, 붕해제 등을 혼합한 혼합물을 기존에 공지된 방법으로 입상으로 제조할 수 있으며, 필요에 따라 착향제, 교미제 등을 함유할 수 있다.
또한 본 발명은 약학적으로 유효한 양의 상기 약학적 조성물을 개체에 투여하는 단계를 포함하는 스트레스성 질환의 예방 및 치료방법을 제공한다.
본 발명의 약학 조성물은 치료적 유효량 또는 약학으로 유효한 양으로 투여한다. 용어 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다.
하기의 실시예를 통하여 본 발명을 보다 상세하게 설명한다. 그러나 하기 실시예는 본 발명의 내용을 구체화하기 위한 것일 뿐 이에 의해 본 발명이 한정되는 것은 아니다.
<준비예 1>
1-1. 실험군 분류
본 발명의 보닐아세테이트(BA)의 우울증 동물모델에 대한 항우울증 효과를 확인하기 위해, 다음과 같은 실험군으로 구분하였다.
- Sham군: CRS(chronic restrained stress)를 유도하지 않고, 보닐아세테이트를 투여하지 않은 정상 대조군
- CRS군: CRS를 유도하고, PBS를 구강으로 투여한 군
- CRS + BA군: CRS를 유도하고 보닐아세테이트(25, 50 및 100 mg/kg)를 구강으로 투여한 군
- BA군: CRS를 유도하지 않고, 보닐아세테이트(100 mg/kg)를 구강으로 투여한 군
1-2. 스트레스성 우울증 동물 모델 유도
스트레스성 우울증 동물 모델에 대한 보닐아세테이트의 항우울증 효능을 확인하기 위하여, 만성구속스트레스(chronic restrained stress, CRS)로 유도한 우울증 동물 모델을 제조하였다. 구체적으로, 동물 모델은 출생 후 8주령의 수컷 C57BL/6NTac 마우스(Narabiotechnology, 대한민국; 체중 22-24 g)를 준비하여 1주일 동안 안정시켰다. 50 ml의 conical tube (SPL Life sciences, Pocheon, Korea)에 숨구멍을 뚫어서 구속틀을 만들고, 그 안에 마우스를 넣어 매일 2시간씩 14일 동안 구속하고, 구속이 끝난 뒤에는 홈케이지에서 물과 먹이를 자유롭게 섭취할 수 있게 하였다.
1-3. 행동실험법
1) 꼬리매달기검사(tail suspension test, TST)
만성구속스트레스를 유발한 지 14일째 되는 날에 바닥으로부터 약 50 cm 높이에 설치한 가로 막대기(직경 1 cm)에 마우스의 꼬리 끝을 고정하여 거꾸로 매단 다음에 부동의 시간을 측정하였다. 총 6분간의 테스트 기간 중에서 후반 4분 동안 측정하였다.
2) 강제수영검사(forced swimming test, FST)
꼬리메달기검사가 끝난 뒤 24시간 후에, 원형수조(직경 15 x 높이 25 cm)에 약 14 cm 높이까지 증류수(23-24℃)를 채워 넣고 마우스를 빠뜨려 강제로 수영시켰다. 총 6분간의 테스트 기간 중에서 후반 4분 동안의 부동(immobility; 최소한의 움직임을 보이며 세발이 움직이지 않거나, 뒷발 중 하나를 이용하여 부유하는 경우)의 시간을 측정하였다.
1-4. 보닐아세테이트의 투여
본 발명의 보닐아세테이트(25, 50 및 100 mg/kg)를 0.1 M PBS에 녹여 매일 구속 스트레스를 유도하기 1시간 전에 구강으로 투여하였다. 보닐아세테이트는 개체 당 투여량이 100 ㎕이 되도록 하였다.
1-5. 효소면역흡착법
보닐아세테이트(25, 50 및 100 mg/kg)를 매일 구속스트레스를 유도하기 1시간 전에 구강으로 투여하고, 14일 후에 마우스를 마취하고, 혈액을 채취하였다. 혈액을 5,000 rpm에서 15분간 원심 분리하여 혈청을 분리하였다. 혈청 내에서 스트레스호르몬인 코르티코스테론(corticosterone)의 수치를 확인하기 위하여 효소면역흡착법(enzyme linked immunosorbent assay, ELISA)의 키트(Enzo life sciences, Farmingdale, NY, USA)를 사용하여 분석하였다.
1-6. 면역조직화학적 염색법
보닐아세테이트(100 mg/kg)를 매일 구속스트레스를 유도하기 1시간 전에 구강으로 투여하고, 14일 후에 마우스를 마취하고, 4% 파라포름알데하이드(paraformaldehyde)를 심장으로 관류하여 고정하고, 뇌를 적출하였다. 적출한 뇌는 4% 파라포름알데하이드에 담가 4℃에서 하루 정도 더 고정시킨 다음, 30% 수크로스(sucrose)로 바꾸어 4℃에서 3일 이상 기간 동안 보관하여 동결손상을 방지하였다. 동결절편기를 이용하여 뇌를 30 ㎛ 두께로 잘라 동결 절편을 제작하였다. 면역조직화학적 염색을 위하여 절편을 블로킹 솔루션[200 ㎍ 소혈청알부민(bovin serum albumin; BSA, sigma, 미국), 500 ㎕ 소태아혈청(fetal bovine serum; FBS, Gibco, 독일), 500 ㎕ 정상염소혈청(normal goat serum; NGS, Vector Laboratories, 미국) 및 10 ㎕ Triton X-100 (Sigma, 미국)을 인산완충액(phosphate buffed saline; PBS)에 넣어 10 ml을 만듦]으로 1시간 동안 반응시켰다. 그 다음에 PBS로 수세하고, 1차 항체[Iba-1(미세아교세포의 마커) 및 c-Fos(세포흥분의 마커)를 블로킹 솔루션에 희석한 용액으로 교체하고, 12시간 이상 동안 반응시켰다. 그 다음에 PBS로 수세한 후, 2차 항체를 PBS에 1:200으로 희석한 용액으로 교체하고, 1시간 동안 반응시킨 후에, PBS로 수세하였다. 그리고 아비딘-바이오틴-과산화효소복합체(avidin-biotin-peroxidase complex; 1:200; Vector Laboratories, 미국)와 3,3'-diamino-benzidine를 이용하여 면역염색을 시행하였다.
1-7. 웨스턴블롯 분석
뇌의 해마(hippocampus)와 편도체(amygdala)를 적출하였다. 이를 단백질 용해 완충액(protein lysis buffer; 10 mM 트리스, 0.5 mM EDTA, 0.25 M 수크로스) 200 ㎕를 넣고 분쇄기를 이용하여 단백질을 분리하였다. 분리된 단백질은 소혈청알부민(bovine serum albumin; BSA, sigma, 미국)을 사용한 Bradford 방법(Bio-rad, 미국)을 이용하여 각 개체의 단백질의 양을 정량한 뒤, 단백질 25 ㎍은 10% SDS-PAGE(Sodium Dodecyl Sulfate-polyacrylamide gel electrophoresis)에 의해 분리하여 PVDF(polyvinylidene difluoride) 멤브레인(membrane, Gendepot, 영국)으로 이동시켰다. PVDF 멤브레인은 5% 탈지유(skimmed milk, BD, 미국)에서 1시간 동안 블로킹시켰다. TBST(Tris-Buffered Saline and Tween 20)로 씻은 후 1차 항체[CD11b(단핵구의 마커), c-Fos(세포흥분의 마커), COX-2(염증매개인자) 및 GAPDH(glyceraldehyde 3-phosphate dehydrogenase)]를 3% 탈지유에 1:1,000으로 희석시켜 4℃에서 하루 동안 반응시킨 후 TBST로 10분씩 3회 씻고, 2차 항체와 함께 상온에서 1시간 동안 반응시켰다. 2차 항체 반응 후에 TBST에 씻은 뒤 ECL system(Santa Cruz, 미국)로 밴드를 확인하였다. 단백질의 확인 및 정량 분석은 이미지 장비 ChemiDoc XRS+(Bio-Rad)를 이용하여 분석하였다.
1-8. 역전사 중합효소 연쇄반응(RT-PCR, reverse transcription-polymerase chain reaction)
만성구속스트레스로 유도한 우울증 동물모델에서 염증매개인자 발현에 대한 보닐아세테이트의 억제 효과를 확인하기 위하여 역전사 중합효소 연쇄반응(RT-PCR, reverse transcription-polymerase chain reaction)을 이용하여 mRNA 발현을 확인하였다. 매일 구속스트레스를 유도하기 1시간 전에 보닐아세테이트(100 mg/kg)를 구강으로 투여하고, 14일 후에 마우스를 마취하고, 뇌의 해마와 편도체를 분리하였다. 분리한 조직은 트리졸(Trizol, Invitrogen, 미국) 1 ml을 넣고, 분쇄기로 조직을 갈고, 원심 분리하여 RNA를 분리하였다. cDNA를 합성하기 위해 0.5 μg의 Oligo dT, 0.5 mM dNTP mix, 5x first-strand buffer, RNase free, 5 mM dithiothreitol (DTT) 및 M-MLV 역전사효소를 포함하는 반응 혼합물(reaction mixture) 중에서 1 μg의 총 RNA를 37℃에서 1시간 동안 반응시켰다. RT-PCR 분석은 제조사의 메뉴얼에 따라 수행되었다(RT-PCR kit; Roche, Germany). PCR 프라이머 서열은 하기 표 1과 같다. PCR 증폭을 위해, 특정 올리고뉴클레오티드 프라이머 쌍은 6 μl of PCR Master Mix (Lucigen, WI, USA) 중에서 1 μl의 cDNA 및 0.6 U의 Econo TaqDNA 중합효소를 반응시켰다. 결과물의 5-7 μl를 2% 아가로스 젤에서 전기영동하고 트랜드일루미네이터에서 브롬화 에티듐(ethidium bromide)으로 염색한 후 확인하였다. 각 유전자의 발현 수준은 GAPDH(glyceraldehyde 3-phosphate dehydrogenase)으로 표준화하였다.

1-9. 통계처리
모든 결과는 SPSS 21.0 package (SPSS Inc, Chicago, USA)을 이용하여 분석하였으며, 실험결과는 mean±standard deviation(평균±표준편차) 값으로 표시하였다. 통계적 유의성 검증은 ANOVA(일원배치 분산분석)를 이용하여 분석 후 Tukey post hoc로 사후분석 하였고, 유의확률(p-value) 값은 p<0.05 및 p<0.01 미만인 경우에만 인정하였다.
<실시예 1> 행동학적인 검사를 통한 보닐아세테이트의 항우울 효능
만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델에서 행동학적인 검사를 통한 보닐아세테이트의 항우울증 효과를 확인하기 위하여, 체중 변화, 꼬리매달기 검사, 강제 수영검사, 혈청 스트레스 호르몬(코르티코스테론) 수치 변화를 확인하였다.
구체적으로, 상기 마우스를 매일 2시간씩 14일간 구속스트레스를 가하는 동안 또는 가한 다음에 체중을 측정하고, 꼬리매달기검사와 강제수영검사를 시행하고, 혈청 스트레스호르몬(코르티코스테론; corticosterone)의 수치를 측정하였다(도 1A와 1B). 그 결과, 만성구속스트레스를 가하면, CRS군에서 체중의 감소는 지속적으로 나타나서 14일째에 Sham군에 비하여 유의미하게 감소하였으나, CRS+BA군에서는 유의미한 증감이 나타나지는 않았다.
또한, 꼬리매달기검사와 강제수영검사의 경우에, CRS군에서의 부동자세의 시간(160초와 133.6초)은 Sham군에서의 부동자세의 시간(64.7초와 35.7초)에 비하여 유의미하게 증가하였으나, 이러한 증가는 CRS+BA군에서는 유의미하게 감소하였다(91.4초와85.2초)(도 1C와 1D). 혈청 내의 스트레스호르몬(코르티코스테론)의 수치는 Sham군에 비해 CRS군에서 유의하게 증가하였고(4822.3 pg/ml), CRS+BA군(50 mg/kg에서 2869.8 pg/ml; 100 mg/kg에서 2202.5 pg/ml)에서 유의하게 감소하였다(도 1E).
<실시예 2> 보닐아세테이트의 미세아교세포 활성화의 억제 효능
만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델의 뇌(해마와 편도체)에서 웨스턴블롯 분석과 면역조직화학적 염색을 통한 미세아교세포의 활성화 정도를 확인하여, 만성스트레스성 우울증 동물모델에서의 미세아교세포(microglia; 신경계 면역세포)의 활성화에 대한 보닐아세테이트의 억제 효과를 검증하였다.
구체적으로, 마우스 뇌의 해마(Hippocampus)와 편도체(Amygdala)를 채취하여 단핵구의 마커인 CD11b에 대한 웨스턴 블롯과, 미세아교세포의 마커인 Iba-1에 대한 면역 염색을 실시하였다.
그 결과, CRS군의 해마와 편도체에서 CD11b의 단백질 발현은 Sham군에 비하여 유의미하게 증가하였고, 이러한 증가는 CRS+BA군에서 유의미하게 억제되었다(도 2A와 2B). 이러한 결과는 Iba-1에 의해 면역 염색된 미세아교세포의 활성화의 정도와 일치하였다(도 2C).
<실시예 3> 보닐 아세테이트의 세포/신경 자극에 대한 반응성 확인
만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델의 뇌(해마 와 편도체)에서 웨스턴블롯 분석과 면역조직화학적 염색을 통한 c-Fos의 활성화 정도를 확인하여, 만성스트레스성 우울증 동물모델에서 신경흥분의 정도에 대한 보닐아세테이트의 억제 효과를 검증하였다.
구체적으로, 해마와 편도체를 채취하여 세포흥분의 마커 c-Fos에 대한 웨스턴블롯 및 c-Fos에 대한 면역염색을 실시하였다.
그 결과, 해마와 편도체에서 c-Fos의 단백질 발현은 Sham군에 비하여 유의미하게 증가하였고, 이러한 증가는 CRS+BA군에서 유의미하게 억제되었다(도 3A-3B). 이러한 결과는 c-Fos에 의해 면역 염색된 세포의 분포정도와 일치하였다(도 3C).
<실시예 4> 보닐 아세테이트의 염증매개인자의 억제 확인
만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델의 뇌(해마와 편도체)에서 웨스턴블롯과 RT-PCR 분석을 통한 염증매개인자의 활성화 정도를 검사하고 정량화하여, 뇌에서 염증반응에 대한 보닐아세테이트의 조절 효과를 검증하였다.
구체적으로, 해마와 편도체를 채취하여 염증매개인자인 COX-2와 iNOS에 대한 웨스턴블롯 또는 RT-PCR 분석을 실시하였다.
그 결과, 해마와 편도체에서 COX-2의 단백질 및 mRNA 발현 그리고 iNOS의 mRNA 발현은 Sham군에 비하여 유의미하게 증가하였고, 이러한 증가는 CRS+BA군에서 유의미하게 억제됨을 확인하였다(도 4A-4F).
상기 일련의 결과를 통하여, 본 발명의 보닐 아세테이트를 유효성분으로 이용 시, 만성구속스트레스(CRS)로 유도된 스트레스성 우울증 동물모델에서 체중 변화, 꼬리매달기 검사, 강제 수영검사 및 혈청 스트레스 호르몬(코르티코스테론) 수치 변화를 통한 행동학적 척도를 개선함을 확인하였다. 또한, 미세아교세포의 활성화를 억제하고, 염증인자(COX-2 및 iNOS)의 생성을 억제하면서, 세포-신경 자극에 대한 반응성에 대한 시경 흥분 정도를 낮추는 효과가 있어, 스트레스 조절능이 우수하고 이에 따른 스트레스성 질환, 특히 우울증의 예방 및 치료에 유용하다는 것을 확인할 수 있다.
Claims (10)
- 보닐 아세테이트(bornyl acetate) 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 스트레스성 질환은 우울증, 불안, 피로증후군, 신경변성 질환, 섭식장애, 신경성 식욕부진, 약물중독, 약물 및 알코올 금단 증후군 및 스트레스성 정신병 에피소드로 이루어진 군에서 선택된, 조성물.
- 제1항에 있어서, 상기 조성물은 염증인자인 COX-2 및 iNOS의 발현을 억제하는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 미세아교세포(microglia)의 활성화를 억제하는 것인, 조성물.
- 제1항에 있어서, 상기 조성물은 세포-신경 자극에 대한 반응성을 억제하는 것인, 조성물.
- 보닐 아세테이트 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 스트레스성 질환의 예방 또는 개선용 식품 조성물.
- 제7항에 있어서, 상기 스트레스성 질환은 우울증, 불안, 피로증후군, 신경변성 질환, 섭식장애, 신경성 식욕부진, 약물중독, 약물 및 알코올 금단 증후군 및 스트레스성 정신병 에피소드로 이루어진 군에서 선택된, 조성물.
- 약학적으로 유효한 양의 제1항의 약학적 조성물을 개체에 투여하는 단계를 포함하는 스트레스성 질환의 예방 및 치료방법.
