本発明は、医薬品等に関する。

で表されるペマフィブラート(化学名:(2R)-2-[3-({1,3-ベンゾオキサゾール-2-イル[3-(4-メトキシフェノキシ)プロピル]アミノ}メチル)フェノキシ]ブタン酸((2R)-2-[3-([1,3-Benzoxazol-2-yl[3-(4-methoxyphenoxy)propyl]amino]methyl)phenoxy]butanoic acid)、国際一般名:Pemafibrate)若しくはその塩又はそれらの溶媒和物は、優れたPPARαアゴニスト活性を有し、血漿トリグリセライド濃度の低下やHDLコレステロールの増加等の作用を示し、脂質異常症(高脂血症)の予防や治療に有用であること(特許文献1、非特許文献1、2)や、NAFLD(非アルコール性脂肪性肝疾患)の予防や治療に有用であること(特許文献2)が知られている。
ところで、医薬品の有効成分として有用な化合物は通常何らかの医薬組成物として製剤化されて投与されることとなるが、医薬組成物が製造されてから投与されるまでに長期間経過することも珍しいことではない。そのため、期待する薬効の発揮の観点や予期せぬ副作用の回避の観点から、医薬組成物中での有効成分の保存安定性の確保が極めて重要となる。

しかしながら、有効成分の保存安定性はその物理的・化学的特性に大きく左右されるところ、当該特性はその化学構造等から予め予測出来ないことが多く、医薬組成物を実際に製造して初めて問題が判明することも少なくない。
そして、ペマフィブラート若しくはその塩又はそれらの溶媒和物については、上記のような薬理効果を示すということが報告されているのみであり、医薬組成物とすることについてはこれまでに具体的に検討がされておらず、医薬組成物中での保存安定性についてはこれまでに全く報告されていなかった。なお、医薬組成物には通常、有効成分のほかに、種々の製剤添加物が配合される。
このような背景の下、本発明者が、ペマフィブラート若しくはその塩又はそれらの溶媒和物を含有する医薬組成物を開発するため、種々の製剤添加物を用いた場合の保存安定性について検討した。しかるところ、ペマフィブラートそのもの自体は極めて安定であるにも拘らず、アミノアルキルメタクリレートコポリマーEに代表される(メタ)アクリル酸系重合体類を共存させた場合に配合変化が生じ、時間の経過に伴い混合物が融解するといった保存安定性の問題が生じることが判明した。
したがって、本発明の課題は、ペマフィブラート若しくはその塩又はそれらの溶媒和物と(メタ)アクリル酸系重合体類との間の配合変化を抑制する技術を提供することにある。
そして、ペマフィブラート若しくはその塩又はそれらの溶媒和物については、上記のような薬理効果を示すということが報告されているのみであり、医薬組成物とすることについてはこれまでに具体的に検討がされておらず、医薬組成物中での保存安定性についてはこれまでに全く報告されていなかった。なお、医薬組成物には通常、有効成分のほかに、種々の製剤添加物が配合される。
このような背景の下、本発明者が、ペマフィブラート若しくはその塩又はそれらの溶媒和物を含有する医薬組成物を開発するため、種々の製剤添加物を用いた場合の保存安定性について検討した。しかるところ、ペマフィブラートそのもの自体は極めて安定であるにも拘らず、アミノアルキルメタクリレートコポリマーEに代表される(メタ)アクリル酸系重合体類を共存させた場合に配合変化が生じ、時間の経過に伴い混合物が融解するといった保存安定性の問題が生じることが判明した。
したがって、本発明の課題は、ペマフィブラート若しくはその塩又はそれらの溶媒和物と(メタ)アクリル酸系重合体類との間の配合変化を抑制する技術を提供することにある。
そこで、本発明者は、上記の状況に鑑み更に検討した結果、ペマフィブラート若しくはその塩又はそれらの溶媒和物と(メタ)アクリル酸系重合体類とを含有する医薬組成物を、ビン包装やPTP包装等の気密包装体に収容することによって、配合変化を抑制できることを見出し、本発明を完成した。
すなわち、本発明は、次の成分(A)及び(B):
(A)ペマフィブラート若しくはその塩又はそれらの溶媒和物;
(B)(メタ)アクリル酸系重合体類;
を含有する医薬組成物が、気密包装体に収容されてなる医薬品を提供するものである。

本明細書において「ペマフィブラート若しくはその塩又はそれらの溶媒和物」には、ペマフィブラート(化学名:(2R)-2-[3-({1,3-ベンゾオキサゾール-2-イル[3-(4-メトキシフェノキシ)プロピル]アミノ}メチル)フェノキシ]ブタン酸((2R)-2-[3-([1,3-Benzoxazol-2-yl[3-(4-methoxyphenoxy)propyl]amino]methyl)phenoxy]butanoic acid)、国際一般名:Pemafibrate)そのもののほか、ペマフィブラートの薬学上許容される塩、さらにはペマフィブラートやその薬学上許容される塩と、水やアルコール(例えば、エタノール)等との溶媒和物も含まれる。薬学上許容される塩としては特に限定されないが、例えば、酸付加塩や塩基付加塩等が挙げられる。酸付加塩としては、具体的には、塩酸塩、臭化水素酸塩、ヨウ化水素酸塩、硫酸塩、硝酸塩、リン酸塩のような無機酸との酸付加塩;安息香酸塩、メタンスルホン酸塩、エタンスルホン酸塩、ベンゼンスルホン酸塩、p-トルエンスルホン酸塩、マレイン酸塩、フマル酸塩、酒石酸塩、クエン酸塩、酢酸塩等の有機酸との酸付加塩が挙げられる。また、塩基付加塩としては、具体的には、ナトリウム塩、カリウム塩、リチウム塩、カルシウム塩、マグネシウム塩等の金属塩;アンモニア、トリメチルアミン、トリエチルアミン、ピリジン、コリジン、ルチジン等のアミンとの塩;リシン、アルギニン、シンコニン、シンコニジン等の有機塩基との塩基付加塩等が挙げられる。

ペマフィブラート若しくはその塩又はそれらの溶媒和物は公知の化合物であり、例えば、特許文献1、非特許文献1、米国特許第7,109,226号明細書に開示の方法により製造することができる。本発明においては、非特許文献1に記載の方法で製造できる、ペマフィブラートの結晶(好適には、第十七改正日本薬局方 融点測定法第1法に従って測定したとき、95~101℃、特に好適には97~100℃の融点を示す結晶)を用いるのが好ましい。なお、これらの文献の内容は、参照として本明細書に組み込まれる。
医薬組成物におけるペマフィブラート若しくはその塩又はそれらの溶媒和物の含有量は特に限定されず、製剤の種類、服用者の性別、年齢、症状等に応じて適宜検討して決定することができる。例えば、1日あたり、ペマフィブラート若しくはその塩又はそれらの溶媒和物を、ペマフィブラートのフリー体に換算して0.05~0.8mg、より好適には0.075~0.6mg、特に好適には0.1~0.4mg服用できる量を含有せしめることができる。
医薬組成物におけるペマフィブラート若しくはその塩又はそれらの溶媒和物の含有量としては、医薬組成物全質量に対して、ペマフィブラートのフリー体換算で0.001~60質量%であるのが好ましく、0.0025~25質量%であるのがより好ましく、0.005~10質量%であるのが更に好ましく、0.0075~5質量%であるのが更に好ましく、0.01~1質量%であるのが更に好ましく、0.05~0.5質量%であるのが特に好ましい。
医薬組成物におけるペマフィブラート若しくはその塩又はそれらの溶媒和物の含有量としては、医薬組成物全質量に対して、ペマフィブラートのフリー体換算で0.001~60質量%であるのが好ましく、0.0025~25質量%であるのがより好ましく、0.005~10質量%であるのが更に好ましく、0.0075~5質量%であるのが更に好ましく、0.01~1質量%であるのが更に好ましく、0.05~0.5質量%であるのが特に好ましい。
<(メタ)アクリル酸系重合体類>
本明細書において、「(メタ)アクリル酸系重合体類」とは、アクリル酸由来の構造単位、メタクリル酸由来の構造単位、アクリル酸エステル類由来の構造単位及びメタクリル酸エステル類由来の構造単位よりなる群から選ばれる1種以上の構造単位(以下、構造単位Xともいう)を含む重合体を意味する。ここで、アクリル酸エステル類、メタクリル酸エステル類としては、例えば、メタノール、エタノール、プロパノール、ブタノール等の脂肪族アルコール(好適には、炭素数1~12(より好ましくは炭素数1~6)の直鎖又は分岐鎖の脂肪族アルコール)と(メタ)アクリル酸とのエステルが挙げられる。なお、当該脂肪族アルコールには、さらに、ジメチルアミノ基、トリメチルアンモニウム基などの、脂肪族アミノ基(第1~3級アミノ基、又は第4級アンモニウム基。なお、脂肪族アミノ基に含まれる脂肪族基としては、炭素数1~12(より好ましくは炭素数1~6)の直鎖又は分岐鎖の脂肪族基が好ましい。)や水酸基、ホスホリルコリン基が置換していてもよい。
また、構造単位Xを誘導する単量体としては、具体的には例えば、アクリル酸、メタクリル酸、アクリル酸メチル、メタクリル酸メチル、アクリル酸エチル、メタクリル酸エチル、アクリル酸ブチル、メタクリル酸ブチル、アクリル酸オクチル、メタクリル酸オクチル、アクリル酸2-エチルヘキシル、メタクリル酸2-エチルヘキシル、アクリル酸ドデシル、メタクリル酸ドデシル、アクリル酸2-ヒドロキシエチル、メタクリル酸2-ヒドロキシエチル、アクリル酸ジメチルアミノエチル、メタクリル酸ジメチルアミノエチル、アクリル酸塩化トリメチルアンモニウムエチル、メタクリル酸塩化トリメチルアンモニウムエチル、2-アクリロイルオキシエチルホスホリルコリン、2-メタクリロイルオキシエチルホスホリルコリン等が挙げられる。これらのうち1種を単独で用いても2種以上を組み合わせて用いてもよい。

また、(メタ)アクリル酸系重合体類としては、第1~3級アミノ基、第4級アンモニウム基、水酸基及びホスホリルコリン基から選ばれる1種以上の置換基を有する(メタ)アクリル酸エステル類由来の構造単位、並びに(メタ)アクリル酸由来の構造単位から選ばれる1種以上の構造単位(以下、構造単位Yともいう)を少なくとも含むものが好ましい。構造単位Yとしては、(メタ)アクリル酸ジC1~12アルキルアミノC1~12アルキル由来の構造単位、(メタ)アクリル酸トリC1~12アルキルアンモニウムC1~12アルキル由来の構造単位、(メタ)アクリル酸由来の構造単位が好ましい。構造単位Yを含む重合体の中では、構造単位Yと、(メタ)アクリル酸C1~12アルキル由来の構造単位(以下、構造単位Zともいう)とを含有する重合体(以下、重合体Pともいう)が好ましい。
また、本明細書において、(メタ)アクリル酸系重合体類は、構造単位X以外にも、これと共重合可能な反応性不飽和基を有する単量体(酢酸ビニル、ビニルピロリドン、2-メチル-5-ビニルピリジン等)由来の構造単位を含んでいてもよい。
本明細書において、(メタ)アクリル酸系重合体類の重量平均分子量は特に限定されないが、1000~1000000が好ましく、10000~500000がより好ましく、50000~300000がさらに好ましく、100000~200000が特に好ましい。
なお、上記重量平均分子量は、ポリアクリル酸ナトリウムに換算した値をいい、ポリマー・ラボラトリー社製のポリアクリル酸ナトリウム標準物質で作製した検量線を用い、サイズ排除クロマトグラフ法で測定できる。
本明細書において、(メタ)アクリル酸系重合体類の重量平均分子量は特に限定されないが、1000~1000000が好ましく、10000~500000がより好ましく、50000~300000がさらに好ましく、100000~200000が特に好ましい。
なお、上記重量平均分子量は、ポリアクリル酸ナトリウムに換算した値をいい、ポリマー・ラボラトリー社製のポリアクリル酸ナトリウム標準物質で作製した検量線を用い、サイズ排除クロマトグラフ法で測定できる。

(メタ)アクリル酸系重合体類としては、含量均一性の観点から、アクリル酸、メタクリル酸、メタクリル酸メチル、アクリル酸エチル、メタクリル酸ブチル、メタクリル酸ジメチルアミノエチル及びメタクリル酸塩化トリメチルアンモニウムエチルよりなる群から選ばれる1種以上の単量体由来の重合体であるのが好ましく、アクリル酸エチル・メタクリル酸メチルコポリマー、アミノアルキルメタクリレートコポリマーE、アンモニオアルキルメタクリレートコポリマー、カルボキシビニルポリマー、メタクリル酸コポリマーS、メタクリル酸コポリマーL及びメタクリル酸コポリマーLD(乾燥メタクリル酸コポリマーLDを含む。)よりなる群から選ばれる1種以上が特に好ましい。メタクリル酸コポリマーLDとしては、乾燥メタクリル酸コポリマーLDが好ましい。

医薬組成物における(メタ)アクリル酸系重合体類の含有量は特に限定されず、製剤の種類、服用者の性別、年齢、症状等に応じて適宜検討して決定することができるが、含量均一性及び保存安定性の観点から、医薬組成物全質量に対して(メタ)アクリル酸系重合体類の合計量として、0.001~50質量%であるのが好ましく、0.005~30質量%であるのがより好ましく、0.01~20質量%であるのがさらに好ましく、0.1~10質量%であるのがさらに好ましく、0.5~5質量%であるのが特に好ましい。
医薬組成物における、ペマフィブラート若しくはその塩又はそれらの溶媒和物と(メタ)アクリル酸系重合体類との含有質量比率は特に限定されないが、含量均一性及び保存安定性の観点から、ペマフィブラートのフリー体換算で1質量部に対し、(メタ)アクリル酸系重合体類を合計で0.001~500質量部含有するのが好ましく、0.01~100質量部含有するのがより好ましく、0.5~50質量部含有するのがさらに好ましく、1~20質量部含有するのが特に好ましい。
本明細書において「医薬組成物」の剤形は特に限定されず、固形状、半固形状、又は液状製剤のいずれであってもよく、その利用目的等に応じて選択することができる。医薬組成物の剤形としては、例えば、第十七改正日本薬局方 製剤総則等に記載の剤形が挙げられる。具体的には例えば、経口投与用の剤形としては、錠剤(例えば、通常錠、口腔内崩壊錠、チュアブル錠、発泡錠、分散錠、溶解錠、CR錠などを含む)、カプセル剤、顆粒剤(例えば、発泡顆粒剤などを含む)、散剤、丸剤等の固形製剤;経口ゼリー剤等の半固形状製剤;経口液剤(例えば、エリキシル剤、懸濁剤、乳剤、リモナーデ剤などを含む)等の液状製剤等が挙げられる。また、非経口投与用の剤形としては、注射剤、吸入剤、点眼剤、点耳剤、点鼻剤、坐剤、外用固形剤、外用液剤、スプレー剤、軟膏剤、クリーム剤、ゲル剤、貼付剤等が挙げられる。
医薬組成物としては、服用のし易さ及び製造のし易さの観点から、固形製剤であるのが好ましい。
固形製剤としては、経口投与用固形製剤が好ましく、錠剤、カプセル剤、顆粒剤、散剤、丸剤がより好ましく、錠剤が特に好ましい。

本発明で用いる医薬組成物には、その剤形に応じて、上記成分以外に製薬上許容される担体(製剤添加物)を加えてもよい。こうした製剤添加物としては、例えば、賦形剤、崩壊剤、結合剤、滑沢剤、可塑剤、フィルム形成剤、粉体、抗酸化剤、矯味剤、甘味剤等が挙げられるが、これらに限定されるものではない。なお、これらの製剤添加物としては、具体的には例えば、医薬品添加物辞典2016(株式会社薬事日報社発行)、Handbook of Pharmaceutical Excipients, Seventh Edition(Pharmaceutical Press社発行)等に収載されたものを用いればよい。
賦形剤としては、具体的には例えば、ケイ酸アルミニウム、無水硫酸ナトリウム、無水リン酸水素カルシウム、塩化ナトリウム、ケイ酸カルシウム、軽質無水ケイ酸、重質無水ケイ酸、硫酸カルシウム、リン酸一水素カルシウム、リン酸水素カルシウム、リン酸水素ナトリウム、リン酸二水素カリウム、リン酸二水素カルシウム、リン酸二水素ナトリウム等の無機系賦形剤;アメ粉、カラメル、カンテン、パラフィン、ショ糖、果糖、麦芽糖、乳糖、乳糖水和物、白糖、ブドウ糖、プルラン、ポリオキシエチレン硬化ヒマシ油、還元麦芽糖水アメ、粉末還元麦芽糖水アメ、トレハロース、還元パラチノース、マルトース、ポリビニルアセタールジエチルアミノアセテート、クエン酸カルシウム等の有機系賦形剤等が挙げられる。これらは、1種又は2種以上を組み合わせて使用することができる。

結合剤としては、具体的には例えば、牛脂硬化油、硬化油、水素添加植物油、ダイズ硬化油、カルナウバロウ、サラシミツロウ、ミツロウ、モクロウ等の油脂類の他、デキストリン、プルラン、アラビアゴム、カンテン、ゼラチン、トラガント、アルギン酸ナトリウム、ポリビニルアルコール、ポリビニルアセタールジエチルアミノアセテート等が挙げられる。これらは、1種又は2種以上を組み合わせて使用することができる。
結合剤の合計含有量は、医薬組成物全質量に対して、好ましくは0.001~30質量%、より好ましくは1~25質量%、特に好ましくは2~20質量%である。

粉体としては、タルク、酸化チタン、黄色三二酸化鉄、三二酸化鉄、法定色素等の有機粉体又は無機粉体が挙げられる。これらは、1種又は2種以上を組み合わせて使用することができる。
粉体の合計含有量は、医薬組成物全質量に対して、好ましくは0.005~3質量%、より好ましくは0.01~2質量%である。
粉体の合計含有量は、医薬組成物全質量に対して、好ましくは0.005~3質量%、より好ましくは0.01~2質量%である。
抗酸化剤としては、具体的には例えば、アスコルビン酸、亜硫酸水素ナトリウム、亜硫酸ナトリウム、エデト酸ナトリウム、エリソルビン酸、酢酸トコフェロール、ジブチルヒドロキシトルエン、天然ビタミンE、トコフェロール、ブチルヒドロキシアニソール等が挙げられる。これらは、1種又は2種以上を組み合わせて使用することができる。
矯味剤としては、具体的には例えば、リモネン、ピネン、カンフェン、サイメン、シネオール、シトロネロール、ゲラニオール、ネロール、リナロール、メントール、テルピネオール、ロジノール、ボルネオール、イソボルネオール、メントン、カンフル、オイゲノール、シンゼイラノール等のテルペン;トウヒ油、オレンジ油、ハッカ油、樟脳白油、ユーカリ油、テレピン油、レモン油、ショウキョウ油、チョウジ油、ケイヒ油、ラベンダー油、ウイキョウ油、カミツレ油、シソ油、スペアミント油等のテルペンを含有する精油;アスコルビン酸、酒石酸、クエン酸、リンゴ酸及びこれらの塩等の酸味剤等が挙げられる。これらは、1種又は2種以上を組み合わせて使用することができる。
甘味剤としては、例えば、アスパルテーム、ステビア、スクラロース、グリチルリチン酸、ソーマチン、アセスルファムカリウム、サッカリン、サッカリンナトリウム等が挙げられ、これらの1種又は2種以上を組み合わせて使用できる。
本発明で用いる医薬組成物は、その剤形に応じて公知の方法により製造することができる。
例えば、医薬組成物が固形製剤である場合には、粉砕、混合、造粒、乾燥、整粒、分級、充填、打錠、コーティング等の単位操作を適宜組み合わせることにより製造することができる。
より具体的には例えば、医薬組成物の剤形が顆粒剤、散剤、丸剤等の粒状の製剤の場合、ペマフィブラート若しくはその塩又はそれらの溶媒和物、(メタ)アクリル酸系重合体類に加えて、必要に応じ賦形剤や結合剤、崩壊剤、滑沢剤等の製剤添加物を用い、これらの成分を混合した後、押出造粒、転動造粒、攪拌造粒、流動層造粒、噴霧造粒、溶融造粒、破砕造粒等の公知の造粒方法により造粒して造粒物を得、さらに必要に応じて分級、整粒等することで製造することができる。なお、得られた造粒物は、公知の方法によりコーティング剤等で被覆することもできる。
また、医薬組成物の剤形が錠剤の場合、ペマフィブラート若しくはその塩又はそれらの溶媒和物、(メタ)アクリル酸系重合体類に加えて、必要に応じ賦形剤や結合剤、崩壊剤、滑沢剤等の適当な製剤添加物を用い、これらの成分を混合して混合物を得、これを直接圧縮(打錠)すること(直接粉末圧縮法)や、上記の造粒物を必要に応じて分級、整粒等したあと圧縮(打錠)すること(半乾式顆粒圧縮法、乾式顆粒圧縮法、湿式顆粒圧縮法など)により製造することができる。なお、得られた圧縮物(錠剤)は、公知の方法によりコーティング剤等で被覆することもできる。
さらに、医薬組成物の剤形がカプセル剤の場合、上記の造粒物や圧縮物を、カプセルに充填すればよい。
例えば、医薬組成物が固形製剤である場合には、粉砕、混合、造粒、乾燥、整粒、分級、充填、打錠、コーティング等の単位操作を適宜組み合わせることにより製造することができる。
より具体的には例えば、医薬組成物の剤形が顆粒剤、散剤、丸剤等の粒状の製剤の場合、ペマフィブラート若しくはその塩又はそれらの溶媒和物、(メタ)アクリル酸系重合体類に加えて、必要に応じ賦形剤や結合剤、崩壊剤、滑沢剤等の製剤添加物を用い、これらの成分を混合した後、押出造粒、転動造粒、攪拌造粒、流動層造粒、噴霧造粒、溶融造粒、破砕造粒等の公知の造粒方法により造粒して造粒物を得、さらに必要に応じて分級、整粒等することで製造することができる。なお、得られた造粒物は、公知の方法によりコーティング剤等で被覆することもできる。
また、医薬組成物の剤形が錠剤の場合、ペマフィブラート若しくはその塩又はそれらの溶媒和物、(メタ)アクリル酸系重合体類に加えて、必要に応じ賦形剤や結合剤、崩壊剤、滑沢剤等の適当な製剤添加物を用い、これらの成分を混合して混合物を得、これを直接圧縮(打錠)すること(直接粉末圧縮法)や、上記の造粒物を必要に応じて分級、整粒等したあと圧縮(打錠)すること(半乾式顆粒圧縮法、乾式顆粒圧縮法、湿式顆粒圧縮法など)により製造することができる。なお、得られた圧縮物(錠剤)は、公知の方法によりコーティング剤等で被覆することもできる。
さらに、医薬組成物の剤形がカプセル剤の場合、上記の造粒物や圧縮物を、カプセルに充填すればよい。
本明細書において「気密包装体」とは、通常の取扱い、運搬又は保存等の状態において、固形状又は液状の異物の侵入を抑制し得る包装を意味し、第十七改正日本薬局方 通則に定義される「気密容器」及び「密封容器」を包含する概念である。気密包装体としては、定形、不定形のいずれのものも用いることができ、具体的には例えば、ビン包装、SP(Strip Package)包装、PTP(Press Through Package)包装、ピロー包装、スティック包装等が挙げられる。気密包装体としては、これらを複数組み合わせたものであってもよく、具体的には例えば、医薬組成物をまずPTP包装にて包装し、これをさらにピロー包装にて包装する態様等が挙げられる。
気密包装体の包装材料(素材)としては、固形状又は液状の異物の侵入を抑制し得るものであれば特に限定されず、例えば、医薬品や食品等の分野で、水分に弱い内容物の防湿等を目的として用いられる材料を適宜用いることができる。
ビン包装に用いられるビン本体の材料としては例えば、ガラス、プラスチック(ポリエステル、ポリエチレン(低密度(LDPE)、中密度(MDPE)、高密度(HDPE)を含む)、ポリカーボネート、ポリスチレン、ポリプロピレン等)、金属(アルミニウム等)などが挙げられる。ビン包装するに際しては、例えば、医薬組成物を、市販のビン内に適当な数量格納し、次いで、適当な栓や蓋で封をすればよい。なお、ビンは、格納する医薬組成物の数量等に応じた大きさのものを適宜選択すればよく、ビンの容量としては例えば、10~500mL程度であり、14~400mLが好ましく、24~350mLがより好ましい。ビン包装の材料としては、ガラス、ポリエチレン、ポリプロピレンが好ましく、ガラス、低密度ポリエチレン(LDPE)、高密度ポリエチレン(HDPE)がより好ましく、ガラス、高密度ポリエチレン(HDPE)が特に好ましい。
ビン包装に用いられるビン本体の材料としては例えば、ガラス、プラスチック(ポリエステル、ポリエチレン(低密度(LDPE)、中密度(MDPE)、高密度(HDPE)を含む)、ポリカーボネート、ポリスチレン、ポリプロピレン等)、金属(アルミニウム等)などが挙げられる。ビン包装するに際しては、例えば、医薬組成物を、市販のビン内に適当な数量格納し、次いで、適当な栓や蓋で封をすればよい。なお、ビンは、格納する医薬組成物の数量等に応じた大きさのものを適宜選択すればよく、ビンの容量としては例えば、10~500mL程度であり、14~400mLが好ましく、24~350mLがより好ましい。ビン包装の材料としては、ガラス、ポリエチレン、ポリプロピレンが好ましく、ガラス、低密度ポリエチレン(LDPE)、高密度ポリエチレン(HDPE)がより好ましく、ガラス、高密度ポリエチレン(HDPE)が特に好ましい。
また、SP包装、PTP包装、ピロー包装やスティック包装等に用いられる包装材料としては例えば、二軸延伸ポリプロピレン(OPP)、二軸延伸ポリエステル(PET)、グリコール変性PET(PET-G)、二軸延伸ナイロン(ONy、PA)、セロハン、紙、低密度ポリエチレン(LDPE)、直鎖状低密度ポリエチレン(L-LDPE)、エチレン-酢酸ビニル共重合体(EVA)、無延伸ポリプロピレン(CPP、IPP)、アイオノマー樹脂(IO)、エチレン-メタクリル酸共重合体(EMAA)、ポリアクリロニトリル(PAN)、二軸延伸ポリ塩化ビニリデン(PVDC)、エチレン-ビニルアルコール共重合樹脂(EVOH)、ポリ塩化ビニル(PVC)、環状ポリオレフィン(COC)、無延伸ナイロン(CNy)、ポリカーボネート(PC)、ポリスチレン(PS)、硬質塩化ビニル(VSC)等の樹脂や、アルミニウム箔(AL)のような金属箔等が挙げられ、これらの1種又は2種以上を適宜組み合わせた多層構造としてもよい。斯かる多層構造としては例えば、PVCとPVDCを積層したもの(PVC/PVDC。以下、同様に省略して表記する。)、PVC/PVDC/PE/PVC、PVC/PVDC/PE/PVDC/PVC、CPP/COC/CPP、PVC/AL、CPP/AL、CPP/CPP/CPP等が挙げられる。斯かる多層構造を形成する方法としては、押出しラミネート、ドライラミネート、共押出しラミネート、サーマルラミネート、ウェットラミネート、ノンソルベントラミネート、ヒートラミネート等の公知のラミネート方法が挙げられる。SP包装、PTP包装、ピロー包装やスティック包装等に用いられる包装材料としては、ポリ塩化ビニル、アルミニウム箔が好ましい。
PTP包装の形態としては、公知の方法で樹脂シート等に所望数形成したポケットに、医薬組成物を1個又は1投与単位ずつ格納し、次いでアルミニウム箔等の金属箔を構成材料とするシートをフタ材として用いて蓋をすることが挙げられる。なお、ポケットを形成するシートとしてもアルミニウム箔を構成材料とするシートを用いた、いわゆる両面アルミPTP包装としてもよい。本発明においては、保存安定性の観点から、PTP包装をさらにピロー包装(例えば、アルミピロー包装など)により包装するのが好ましい。
SP包装やピロー包装、スティック包装の形態としては、公知の方法で樹脂シートやアルミニウム箔を構成材料とするシート等を用いて、医薬組成物を1個又は1投与単位ずつ包装することが挙げられる。本発明においては、保存安定性の観点から、アルミニウム箔を構成材料とするシートを用いるのが好ましい。
SP包装やピロー包装、スティック包装の形態としては、公知の方法で樹脂シートやアルミニウム箔を構成材料とするシート等を用いて、医薬組成物を1個又は1投与単位ずつ包装することが挙げられる。本発明においては、保存安定性の観点から、アルミニウム箔を構成材料とするシートを用いるのが好ましい。
なお、本明細書において、医薬品における医薬組成物の包装体内部での占有率(容積率)は、包装体がビン包装の場合、通常、25~90%であり、28~75%が好ましく、30~50%がより好ましい。また、包装体がSP包装、PTP包装、ピロー包装、スティック包装の場合、通常、30~98%であり、40~95%が好ましく、45~93%がより好ましく、50~90%が特に好ましい。なお、この場合において、占有率とは、包装体内部の全容積に対する医薬組成物の占有率を意味するものであり、包装体内部に格納した医薬組成物の破損防止のための詰め物や中栓等は、空間占有率を算出するに際して考慮されるものではない。
気密包装体としては、市販の包装体をそのまま用いてもよく、また市販の包装材料を加工して用いてもよい。市販品のビン包装の包装体としては、例えば、Z-シリーズ(阪神化成工業社製)、A-シリーズ(磯谷硝子工業製)等が挙げられる。また、SP包装、PTP包装、ピロー包装やスティック包装用の包装材料としては、スミライトVSS、スミライトVSL、スミライトNS、スミライトFCL(以上、住友ベークライト社製)、TASシリーズ(大成化工社製)、PTP用ビニホイル、PTP用スーパーホイル(以上、三菱樹脂社製)、ニッパクアルミ箔(日本製箔社製)、アルミ箔銀無地(大和化学工業社製)等が挙げられる。
医薬組成物を気密包装体に収容する方法は特に限定されるものではなく、包装体内への医薬組成物の投入等の適当な手段により、医薬組成物を包装体内に配置することで達成できる。この場合において、包装体内に医薬組成物とともに乾燥剤(例えば、円柱状(錠剤型)のものやシート状のもの)を投入する手段を用いてもよい。
本発明の医薬品の適用疾患は何ら限定されず、現時点で公知の、又は将来的に見出される、ペマフィブラートの投与が有効であるとされる疾患の予防又は治療に広く用いることができる。
例えば、ペマフィブラート若しくはその塩又はそれらの溶媒和物は優れたPPARαアゴニスト活性を有し、血漿トリグリセライド濃度の低下やHDLコレステロールの増加等の作用を有する。従って、本発明の医薬品は、好適には、脂質異常症(高脂血症、より詳細には例えば、原発性高脂血症、続発性高脂血症等)の予防及び/又は治療剤として、さらに好適には高トリグリセライド血症の予防及び/又は治療剤等として使用できる。
また、ペマフィブラート若しくはその塩又はそれらの溶媒和物は、NAFLD(非アルコール性脂肪性肝疾患)の予防又は治療に有用である。従って、本発明の医薬品は、NAFLD(より好適には、NASH(非アルコール性脂肪性肝炎))の予防及び/又は治療剤等としても使用できる。
さらに、ペマフィブラート若しくはその塩又はそれらの溶媒和物は、原発性胆汁性肝硬変の治療剤等として使用してもよい。
例えば、ペマフィブラート若しくはその塩又はそれらの溶媒和物は優れたPPARαアゴニスト活性を有し、血漿トリグリセライド濃度の低下やHDLコレステロールの増加等の作用を有する。従って、本発明の医薬品は、好適には、脂質異常症(高脂血症、より詳細には例えば、原発性高脂血症、続発性高脂血症等)の予防及び/又は治療剤として、さらに好適には高トリグリセライド血症の予防及び/又は治療剤等として使用できる。
また、ペマフィブラート若しくはその塩又はそれらの溶媒和物は、NAFLD(非アルコール性脂肪性肝疾患)の予防又は治療に有用である。従って、本発明の医薬品は、NAFLD(より好適には、NASH(非アルコール性脂肪性肝炎))の予防及び/又は治療剤等としても使用できる。
さらに、ペマフィブラート若しくはその塩又はそれらの溶媒和物は、原発性胆汁性肝硬変の治療剤等として使用してもよい。
本発明で用いる医薬組成物の服用経路は特に限定されず、適用する疾患、製剤の種類、服用者の性別、年齢、症状等に応じて適宜検討して決定することができるが、服用の容易性の観点から、経口投与が好ましい。また、医薬組成物は、1日につき1~4回(好ましくは1回)程度に分けて、食前、食間、食後、就寝前等に服用することができる。

[3-4] 成分(B)が、アミノアルキルメタクリレートコポリマーE、アンモニオアルキルメタクリレートコポリマー、メタクリル酸コポリマーS、メタクリル酸コポリマーL及びメタクリル酸コポリマーLDよりなる群から選ばれる1種以上である、[3-1]記載の医薬組成物。
[3-5] 固形製剤である、[3-1]~[3-4]のいずれか記載の医薬組成物。
[3-6] 錠剤、カプセル剤、顆粒剤、散剤又は丸剤である、[3-1]~[3-5]のいずれか記載の医薬組成物。
[3-7] 気密包装体が、ビン包装、SP包装、PTP包装、ピロー包装及びスティック包装よりなる群から選ばれる1種以上である、[3-1]~[3-6]のいずれか記載の医薬組成物。
[3-8] 脂質異常症(高脂血症、より詳細には例えば、原発性高脂血症、続発性高脂血症等)、NAFLD(より好適には、NASH(非アルコール性脂肪性肝炎))及び原発性胆汁性肝硬変から選ばれる疾患の予防及び/又は治療剤である、[3-1]~[3-7]のいずれか記載の医薬組成物。
(A)ペマフィブラート若しくはその塩又はそれらの溶媒和物;
(B)(メタ)アクリル酸系重合体類;
を含有する医薬組成物。
[3-2] 成分(B)が、アクリル酸、メタクリル酸、メタクリル酸メチル、アクリル酸エチル、メタクリル酸ブチル、メタクリル酸ジメチルアミノエチル及びメタクリル酸塩化トリメチルアンモニウムエチルよりなる群から選ばれる1種以上の単量体由来の重合体である、[3-1]記載の医薬組成物。
[3-3] 成分(B)が、アクリル酸エチル・メタクリル酸メチルコポリマー、アミノアルキルメタクリレートコポリマーE、アンモニオアルキルメタクリレートコポリマー、カルボキシビニルポリマー、メタクリル酸コポリマーS、メタクリル酸コポリマーL及びメタクリル酸コポリマーLDよりなる群から選ばれる1種以上である、[3-1]記載の医薬組成物。
[3-4] 成分(B)が、アミノアルキルメタクリレートコポリマーE、アンモニオアルキルメタクリレートコポリマー、メタクリル酸コポリマーS、メタクリル酸コポリマーL及びメタクリル酸コポリマーLDよりなる群から選ばれる1種以上である、[3-1]記載の医薬組成物。
[3-5] 固形製剤である、[3-1]~[3-4]のいずれか記載の医薬組成物。

以下、実施例により本発明をより具体的に説明するが、本発明はこれらにより何ら限定されるものではない。
[試験例1]安定性試験
以下に示すサンプル1-1~1-4をそれぞれ調製後、40℃、75%相対湿度(RH)の条件下で1ヶ月間保存し、保存開始直後及び1ヶ月間後におけるサンプルの状態変化の有無を評価し、配合変化の有無を確認した。
結果を表1に示す。

〔サンプル1-1〕
ペマフィブラートを、蓋を開けたガラス製の容器にとり、そのままサンプル1-1とした。
〔サンプル1-2〕
アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を、蓋を開けたガラス製の容器にとり、そのままサンプル1-2とした。
〔サンプル1-3〕
ペマフィブラート1質量部に対し、アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を1質量部の割合で混合して混合物を得、これを、蓋を開けたガラス製の容器にとり、サンプル1-3とした。
〔サンプル1-4〕
ペマフィブラート1質量部に対し、アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を1質量部の割合で混合して混合物を得た。得られた混合物のうち0.25gをガラス瓶(2K規格ビン:磯谷硝子工業)に入れて蓋をし、これをサンプル1-4とした。
ペマフィブラートを、蓋を開けたガラス製の容器にとり、そのままサンプル1-1とした。
〔サンプル1-2〕
アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を、蓋を開けたガラス製の容器にとり、そのままサンプル1-2とした。
〔サンプル1-3〕
ペマフィブラート1質量部に対し、アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を1質量部の割合で混合して混合物を得、これを、蓋を開けたガラス製の容器にとり、サンプル1-3とした。
〔サンプル1-4〕
ペマフィブラート1質量部に対し、アミノアルキルメタクリレートコポリマーE(オイドラギットEPO:Evonik Rohm GmbH)を1質量部の割合で混合して混合物を得た。得られた混合物のうち0.25gをガラス瓶(2K規格ビン:磯谷硝子工業)に入れて蓋をし、これをサンプル1-4とした。
表1に示す試験結果から、ペマフィブラートのみのサンプル1-1においては1ヶ月間保存後も保存開始直後と同様に白色粉体の状態が保たれ、また、アミノアルキルメタクリレートコポリマーEのみのサンプル1-2においては白色から微黄色へと若干の変色が認められたのみであり粉体として殆ど変わらない状態が保たれるのに対し、ペマフィブラートとアミノアルキルメタクリレートコポリマーEとを混合したサンプル1-3においては配合変化が生じ、混合物が融解し、粉状から飴状に大きく変化した。
一方、ペマフィブラートとアミノアルキルメタクリレートコポリマーEとを混合し、これを気密包装体に収容したサンプル1-4においては、1ヶ月間保存後も保存開始直後と同様に白色粉体の状態が保たれることが判明した。

以上の試験結果から、ペマフィブラートと、アミノアルキルメタクリレートコポリマーEに代表される(メタ)アクリル酸系重合体類との間の配合変化は、気密包装体に収容することによって、抑制されることが明らかとなった。
[製造例1~9]
表2~4に記載の成分及び分量(mg)を1錠当りに含有する錠剤(例1~9)を常法により製造し、高密度ポリエチレン製のビン(ボトル)に収容して、それぞれ製造例1~9の医薬品を得る。

[製造例10~18]
表2~4に記載の成分及び分量(mg)を1錠当りに含有する錠剤(例1~9)を常法により製造し、予めポケット部分を成形した樹脂シート(住友ベークライト社製:商品名 スミライトVSS-1202)のポケット部分に入れ、次いでPTP用アルミ箔(大和化学工業社製:商品名 アルミ箔銀無地)で蓋をしてPTP包装する。得られたPTP包装体3シート(1シート当たり錠剤を10錠収容する)をさらにアルミピロー包装し、それぞれ製造例10~18の医薬品を得ることができる。

表2~4に記載の成分及び分量(mg)を1錠当りに含有する錠剤(例1~9)を常法により製造し、予めポケット部分を成形した樹脂シート(住友ベークライト社製:商品名 スミライトVSS-1104)のポケット部分に入れ、次いでPTP用アルミ箔(大和化学工業社製:商品名 アルミ箔銀無地)で蓋をしてPTP包装する。得られたPTP包装体2シート(1シート当たり錠剤を12錠収容する)をアルミピロー包装し、それぞれ製造例19~27の医薬品を得ることができる。

表2~4に記載の成分及び分量(mg)を1錠当りに含有する錠剤(例1~9)を常法により製造し、予めポケット部分を成形した樹脂シート(住友ベークライト社製:商品名 スミライトVSL-4501)のポケット部分に入れ、次いでPTP用アルミ箔(大和化学工業社製:商品名 アルミ箔銀無地)で蓋をしてPTP包装する。得られたPTP包装体3シート(1シート当たり錠剤を10錠収容する)をさらにアルミピロー包装し、それぞれ製造例28~36の医薬品を得ることができる。

表2~4に記載の成分及び分量(mg)を1錠当りに含有する錠剤(例1~9)を常法により製造し、ガラスビンに収容し、それぞれ製造例37~45の医薬品を得ることができる。

本発明によれば、血漿トリグリセライド濃度の低下やHDLコレステロールの増加等の作用を示すペマフィブラートを含有し、保存安定性に優れる医薬組成物を提供できるため、例えば医薬品産業等において利用できる。
Claims (6)
- 次の成分(A)及び(B):
(A)ペマフィブラート若しくはその塩又はそれらの溶媒和物;
(B)(メタ)アクリル酸系重合体類;
を含有する医薬組成物が、気密包装体に収容されてなる医薬品。 - 成分(B)が、アクリル酸、メタクリル酸、メタクリル酸メチル、アクリル酸エチル、メタクリル酸ブチル、メタクリル酸ジメチルアミノエチル及びメタクリル酸塩化トリメチルアンモニウムエチルよりなる群から選ばれる1種以上の単量体由来の重合体である、請求項1記載の医薬品。
- 成分(B)が、アクリル酸エチル・メタクリル酸メチルコポリマー、アミノアルキルメタクリレートコポリマーE、アンモニオアルキルメタクリレートコポリマー、カルボキシビニルポリマー、メタクリル酸コポリマーS、メタクリル酸コポリマーL及びメタクリル酸コポリマーLDよりなる群から選ばれる1種以上である、請求項1又は2記載の医薬品。
- 医薬組成物が、固形製剤である、請求項1~3のいずれか1項記載の医薬品。
- 医薬組成物が、錠剤、カプセル剤、顆粒剤、散剤又は丸剤である、請求項1~4のいずれか1項記載の医薬品。
- 気密包装体が、ビン包装、SP包装、PTP包装、ピロー包装及びスティック包装よりなる群から選ばれる1種以上である、請求項1~5のいずれか1項記載の医薬品。
