本発明は、α-ヒドロキシイソ酪酸エステル化合物、香料組成物、及び香料としての使用に関する。
イソ酪酸エステルには香料として有用な化合物があることが知られている。例えば、非特許文献1には各種のイソ酪酸エステルが主としてフレーバーとして用いられており、具体的にはイソ酪酸メチルが甘いアプリコット様、イソ酪酸プロピルが重いパイナップル様、イソ酪酸ブチルが新鮮なリンゴ及びバナナ様、イソ酪酸イソアミルが甘いアプリコット及びパイナップル様といった、いずれもフルーツ香のフレーバー素材であることの記載がある。
また、特許文献1にはα位に酸素との結合を持つイソ酪酸エステルとして、α-アルコキシイソ酪酸の直鎖又は分岐した炭素数4~12のアルキルエステルが香料として有用であることが開示されており、α-エトキシイソ酪酸ノルマルヘキシルがラベンダー様の香気を持つと記載がある。
また、特許文献1にはα位に酸素との結合を持つイソ酪酸エステルとして、α-アルコキシイソ酪酸の直鎖又は分岐した炭素数4~12のアルキルエステルが香料として有用であることが開示されており、α-エトキシイソ酪酸ノルマルヘキシルがラベンダー様の香気を持つと記載がある。
一方、α-ヒドロキシイソ酪酸エステルの多くは公知な物質である。
例えば特許文献2では、α-ヒドロキシイソ酪酸のノルマルプロピル、イソプロピル、ノルマルブチル、イソブチル及びアミルエステルがニトロセルロースの溶解性に優れた溶媒であることが開示されている。
また、特許文献3では、α-ヒドロキシイソ酪酸のエチル、イソプロピル、ブチルエステルが低毒性で安全性の高い溶媒として脱脂用洗浄剤、フラックス洗浄剤、レジスト剥離剤等に有用であることが開示されている。
例えば特許文献2では、α-ヒドロキシイソ酪酸のノルマルプロピル、イソプロピル、ノルマルブチル、イソブチル及びアミルエステルがニトロセルロースの溶解性に優れた溶媒であることが開示されている。
また、特許文献3では、α-ヒドロキシイソ酪酸のエチル、イソプロピル、ブチルエステルが低毒性で安全性の高い溶媒として脱脂用洗浄剤、フラックス洗浄剤、レジスト剥離剤等に有用であることが開示されている。

本発明が解決しようとする課題は、香料及び調合香料素材として有用なα-ヒドロキシイソ酪酸エステル化合物を提供することである。更に本発明が解決しようとする別の課題は、α-ヒドロキシイソ酪酸エステル化合物を有効成分として含有する香料組成物、及び該化合物の香料としての使用を提供することである。
本発明者らは、種々の化合物を合成し、その香気について鋭意検討したところ、α-ヒドロキシイソ酪酸の特定のエステル化合物が香料及び調合香料素材として有用であることを見出した。

本発明によれば、香料及び調合香料素材として有用なα-ヒドロキシイソ酪酸エステル化合物を提供することができる。更に本発明によれば、α-ヒドロキシイソ酪酸エステル化合物を有効成分として含有する香料組成物、及び該化合物の香料としての使用を提供することができる。
[香料組成物及び使用]
本発明の香料組成物は、下記式(1)で表される化合物を有効成分として含む。また、本発明の使用は、下記式(1)で表される化合物の香料としての使用である。従来、α-ヒドロキシイソ酪酸エステル化合物については、報告があるが、α-ヒドロキシイソ酪酸エステル固有の香りについて、先行文献に記載はなかった。
以下、本発明について、詳細に説明する。
<式(1)で表される化合物>
本発明の香料組成物及び本発明の使用に用いられる化合物は、下記式(1)で表される。

本発明の香料組成物は、下記式(1)で表される化合物を有効成分として含む。また、本発明の使用は、下記式(1)で表される化合物の香料としての使用である。従来、α-ヒドロキシイソ酪酸エステル化合物については、報告があるが、α-ヒドロキシイソ酪酸エステル固有の香りについて、先行文献に記載はなかった。
以下、本発明について、詳細に説明する。
<式(1)で表される化合物>
本発明の香料組成物及び本発明の使用に用いられる化合物は、下記式(1)で表される。

R基が不斉炭素を持つ場合には、式(1)で表される化合物は、それによって生じる光学異性体のいずれか1つ又は任意の割合での混合物を含む。

本発明において、式(1)で表される化合物として、以下の式(1-1)~(1-35)のいずれかで表される化合物が例示される。好ましい化合物は、以下の式(1-1)~(1-10)、(1-13)、(1-16)~(1-18)、(1-21)、(1-24)のいずれかで表される化合物であり、また、好ましい化合物は、以下の式(1-1)~(1-10)のいずれかで表される化合物である。

本発明において、式(1)で表される化合物として、以下の式(1-1)~(1-35)のいずれかで表される化合物が例示される。好ましい化合物は、以下の式(1-1)~(1-10)、(1-13)、(1-16)~(1-18)、(1-21)、(1-24)のいずれかで表される化合物であり、また、好ましい化合物は、以下の式(1-1)~(1-10)のいずれかで表される化合物である。
近年、化学物質の毒性や環境への影響が極めて重視される傾向にあり、それは香料や香料組成物についても例外ではない。人体への感作性や環境への蓄積性などを理由に従来用いられてきた香料の使用条件が厳しく制限されたり、使用禁止になるケースが増える傾向にある。そのために環境負荷の少ない香料及び香料組成物が今まで以上に強く求められる状況にある。従って、調合香料素材としても、生分解性に優れ、生物蓄積性が小さいことが好ましい。
式(1)で表される化合物は、生分解性に優れ、かつ、生物蓄積性が小さい化合物を含み、この観点からは、Rは、エチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、セカンダリーブチル基、ターシャリーブチル基、3-メチルブタン-2-イル基、2-メチルブチル基、3-メチルブチル基、ネオペンチル基、2-メチルペンチル基、4-メチルペンタン-2-イル基、ノルマルヘキシル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択された基であることが好ましい。また、同様にの観点から、Rは、エチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、2-メチルブチル基、3-メチルブチル基、4-メチルペンタン-2-イル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択された基であることも好ましい。
式(1)で表される化合物は、生分解性に優れ、かつ、生物蓄積性が小さい化合物を含み、この観点からは、Rは、エチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、セカンダリーブチル基、ターシャリーブチル基、3-メチルブタン-2-イル基、2-メチルブチル基、3-メチルブチル基、ネオペンチル基、2-メチルペンチル基、4-メチルペンタン-2-イル基、ノルマルヘキシル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択された基であることが好ましい。また、同様にの観点から、Rは、エチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、2-メチルブチル基、3-メチルブチル基、4-メチルペンタン-2-イル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択された基であることも好ましい。
式(1)で表される化合物は、それ自体が後述するように優れた香気を有することから、香料として有用である。また、香料は、一般に単品で使用されることは少なく、複数の香料を目的に合わせて配合した調合香料(香料組成物)として使用することが多い。式(1)で表される化合物は、調合香料(香料組成物)に配合される香料(「調合香料素材」ともいう。)として有用であり、本発明の香料組成物は、式(1)で表される化合物を有効成分として含有するものである。香料として、上記式(1)で表される化合物を1種単独で使用してもよく、2種以上を併用してもよい。
また、式(1)で表される化合物が、本発明の効果を損なわない範囲で、少量の不純物、副生成物、夾雑物などを含むことを排除するものではない。

式(1)で表される化合物は、ミント様の香気を持つと共にウッディ調、スパイシー調、フローラル調、グリーン調などの香気を有し、かつ拡散性にも優れる。また、上記式(1)で表される化合物の中には、グリーンノートの香気又はフルーティノートの香気を持つものもあり、かつ拡散性にも優れる。式(1)で表される化合物を単独で香料として各種香粧品類、保健衛生材料をはじめとして医薬品、日用雑貨品、食品などに添加使用することにより香気を賦与してもよく、また、式(1)で表される化合物を他の調合香料素材等と混合して、後述する香料組成物(調合香料)を調製し、これを各種の製品に配合して香気を付与してもよい。これらの中でも、目的とする香気を得る観点から、式(1)で表される化合物を調合香料素材として香料組成物に配合して、式(1)で表される化合物を有効成分として含有する香料組成物を調製し、該香料組成物を製品に配合することで賦香することが好ましい。
また、式(1)で表される化合物は、香料として使用することが好ましく、ミントの香り、グリーンノートの香り又はフルーティノートの香りを付与するために使用されることがより好ましい。

<香料組成物>
本発明の香料組成物(調合香料)は、式(1)で表される化合物を有効成分として含有する。なお、式(1)で表される化合物を少なくとも1種以上含有すれば特に限定されず、2種以上の式(1)で表される化合物を含有してもよい。
本発明の香料組成物は、式(1)で表される化合物を有効成分として含有していればよく、その他の成分については特に限定されないが、他の調合香料素材(以下、「従来香料」ともいう。)を更に含有することが好ましい。
なお、「香料組成物(調合香料)」とは、該香料組成物を各種香粧品類、医薬品、食品、飲料等に添加することで、香気を付与する組成物、又はそれ自体として香水等に使用される組成物であり、従来香料に加え、必要に応じて、溶媒等の添加剤を含有してもよい。
式(1)で表される化合物の配合量は、化合物の種類、目的とする香気の種類及び香気の強さ等により異なるが、式(1)で表される化合物の量として香料組成物中に、好ましくは0.001質量%以上、より好ましくは0.01質量%以上、更に好ましくは0.1質量%以上であり、好ましくは90質量%以下、より好ましくは70質量%以下、更に好ましくは50質量%以下である。
本発明の香料組成物(調合香料)は、式(1)で表される化合物を有効成分として含有する。なお、式(1)で表される化合物を少なくとも1種以上含有すれば特に限定されず、2種以上の式(1)で表される化合物を含有してもよい。
本発明の香料組成物は、式(1)で表される化合物を有効成分として含有していればよく、その他の成分については特に限定されないが、他の調合香料素材(以下、「従来香料」ともいう。)を更に含有することが好ましい。

従来香料は、従来公知な香料成分であれば特に制限はなく、広い範囲の香料が使用でき、例えば下記のようなものから単独で又は2種以上を任意の混合比率で選択し、使用することができる。

また、香料組成物は、調合香料素材以外の構成成分として、ポリオキシエチレンラウリル硫酸エーテル等の界面活性剤;ジプロピレングリコール、ジエチルフタレート、エチレングリコール、プロピレングリコール、メチルミリステート、トリエチルシトレート等の溶媒;酸化防止剤;着色剤等も含んでいてもよい。
式(1)で表される化合物は、ミント様の香気を有すると共に、ウッディ調、スパイシー調、フローラル調、グリーン調などの香気を有することから、従来香料と組み合わせることによりミント調と共に自然なウッディ調、スパイシー調、フローラル調、グリーン調を賦与できるため、各種香粧品類、保健衛生材料をはじめとして医薬品、日用雑貨品、食品などへの添加し、香気を賦与するに有用である。
式(1)で表される化合物を含有する香料組成物を、香気付与のため、及び配合対象物の香気の改良を行うために添加できるものとしては香粧品類、健康衛生材料、雑貨、飲料、食品、医薬部外品、医薬品等の各種製品を挙げることができ、例えば、香水、コロン類等のフレグランス製品;シャンプー、リンス類、ヘアートニック、ヘアークリーム類、ムース、ジェル、ポマード、スプレーその他毛髪用化粧料;化粧水、美容液、クリーム、乳液、パック、ファンデーション、おしろい、口紅、各種メークアップ類等の肌用化粧料;皿洗い洗剤、洗濯用洗剤、ソフトナー類、消毒用洗剤類、消臭洗剤類、室内芳香剤、ファニチャーケア、ガラスクリーナー、家具クリーナー、床クリーナー、消毒剤、殺虫剤、漂白剤、殺菌剤、忌避剤、その他の各種健康衛生用洗剤類;歯磨、マウスウォッシュ、入浴剤、制汗製品、パーマ液等の医薬部外品;トイレットペーパー、ティッシュペーパー等の雑貨;医薬品等;食品等の香気成分として使用することができる。
上記製品中の香料組成物の配合量は特に限定されず、賦香すべき製品の種類、性質及び官能的効果などに応じて、香料組成物の配合量は広い範囲に渡って選択することができる。例えば、0.00001質量%以上、好ましくは0.0001質量%以上、更に好ましくは0.001質量%以上であり、例えば香水等のフレグランスの場合には100質量%であってもよく、好ましくは80質量%以下、更に好ましくは60質量%以下、より更に好ましくは40質量%以下である。
式(2)で表される2-ヒドロキシ-2-メチルプロピオン酸4-メチルペンタン-2-イルは新規物質である。この化合物は、1つの不斉炭素を持つため、2つの光学異性体を持ち、いずれか1つ又は任意の割合での混合物を含む。
2-ヒドロキシ-2-メチルプロピオン酸4-メチルペンタン-2-イルは、単独で香料として有用であり、また、香料組成物の有効成分として有用である。
また、2-ヒドロキシ-2-メチルプロピオン酸4-メチルペンタン-2-イルは、香料として使用することが好ましく、ミント様の香りを付与するために使用されることがより好ましい。
また、2-ヒドロキシ-2-メチルプロピオン酸4-メチルペンタン-2-イルは、香料として使用することが好ましく、ミント様の香りを付与するために使用されることがより好ましい。
[式(1)で表される化合物の製造方法]
式(1)で表される化合物の製造方法に特に制限はなく、従来公知の方法から適宜選択して用いればよい。
例えば、ピルビン酸エステルとメチルハロゲン化マグネシウムをグリニャール反応させることによってα-ヒドロキシイソ酪酸エステルを製造することができる。この反応の反応式を下記式(3)に示した。
式(1)で表される化合物の製造方法に特に制限はなく、従来公知の方法から適宜選択して用いればよい。
例えば、ピルビン酸エステルとメチルハロゲン化マグネシウムをグリニャール反応させることによってα-ヒドロキシイソ酪酸エステルを製造することができる。この反応の反応式を下記式(3)に示した。
また、α-ヒドロキシイソ酪酸とアルコールを触媒の存在下にエステル化反応させることによって、α-ヒドロキシイソ酪酸エステルを製造することができる。この反応の反応式を下記式(4)に示した。
また、別種のα-ヒドロキシイソ酪酸エステルとアルコールを触媒の存在下にエステル交換反応させることによって、目的のα-ヒドロキシイソ酪酸エステルを製造することができる。この反応の反応式を下記式(5)に示した。
これらの反応に用いられる触媒や反応方式、反応条件、及び反応装置などについても、従来公知な触媒、反応方法、反応条件、及び反応装置を用いることができ、特に制限はない。また、得られた式(1)の化合物を精製する方法についても、従来公知な精製方法を採用することができ、何ら制限はない。
以下に、実施例を以って本発明を更に詳細に説明するが、本発明は、これらの実施例に限定されるものではない。

<実施例1:α-ヒドロキシイソ酪酸エチルの合成>
蒸留管を備えた300mlガラス製フラスコにα-ヒドロキシイソ酪酸メチル(三菱ガス化学(株)製)56.7g、エタノール(和光純薬工業(株)製)33.2g、チタンテトラエトキシド(和光純薬工業(株)製)0.92gを充填した。常圧下で加熱還流しながらエステル交換反応を行い、生成するメタノールを系外に抜き出しながら96時間反応を行った。その結果、下記式(6)の反応により反応収率97%でα-ヒドロキシイソ酪酸エチルが得られた。反応系に加水して触媒を失活させた後に減圧蒸留を行い、71mmHg、77℃の留分としてα-ヒドロキシイソ酪酸エチル46.9g(GC分析による純度(以下、GC純度ともいう。):99.6%)を得た。
蒸留管を備えた300mlガラス製フラスコにα-ヒドロキシイソ酪酸メチル(三菱ガス化学(株)製)56.7g、エタノール(和光純薬工業(株)製)33.2g、チタンテトラエトキシド(和光純薬工業(株)製)0.92gを充填した。常圧下で加熱還流しながらエステル交換反応を行い、生成するメタノールを系外に抜き出しながら96時間反応を行った。その結果、下記式(6)の反応により反応収率97%でα-ヒドロキシイソ酪酸エチルが得られた。反応系に加水して触媒を失活させた後に減圧蒸留を行い、71mmHg、77℃の留分としてα-ヒドロキシイソ酪酸エチル46.9g(GC分析による純度(以下、GC純度ともいう。):99.6%)を得た。
<実施例2~13:各種α-ヒドロキシイソ酪酸エステルの合成>
参考例1と同様の反応装置を用い、適量のα-ヒドロキシイソ酪酸メチル(三菱ガス化学(株)製)と各種アルコール(ノルマルプロパノール、イソプロパノール、ノルマルブタノール、イソブタノール、セカンダリーブタノール、2-メチルブタノール、ネオペンチルアルコール、2-メチルペンタノール、ノルマルヘキサノール、シクロペンタノール、シクロヘキサノール、4-メチルペンタン-2-オール)をチタンテトラアルコキシド及び/又はナトリウムアルコキシドのような適当な触媒の存在下、場合によってはヘキサン、トルエンのような溶媒共存下で、加熱しながら適当な反応条件下でエステル交換反応させた。反応によって生成するメタノールを反応条件下で蒸留又は反応溶媒との共沸によって系外へ抜出しながらエステル交換反応を完結し、参考例1と同様の分離操作を行い、以下のα-ヒドロキシイソ酪酸エステルをそれぞれ得た。得られたイソ酪酸エステルのGC純度を併記した。

参考例1と同様の反応装置を用い、適量のα-ヒドロキシイソ酪酸メチル(三菱ガス化学(株)製)と各種アルコール(ノルマルプロパノール、イソプロパノール、ノルマルブタノール、イソブタノール、セカンダリーブタノール、2-メチルブタノール、ネオペンチルアルコール、2-メチルペンタノール、ノルマルヘキサノール、シクロペンタノール、シクロヘキサノール、4-メチルペンタン-2-オール)をチタンテトラアルコキシド及び/又はナトリウムアルコキシドのような適当な触媒の存在下、場合によってはヘキサン、トルエンのような溶媒共存下で、加熱しながら適当な反応条件下でエステル交換反応させた。反応によって生成するメタノールを反応条件下で蒸留又は反応溶媒との共沸によって系外へ抜出しながらエステル交換反応を完結し、参考例1と同様の分離操作を行い、以下のα-ヒドロキシイソ酪酸エステルをそれぞれ得た。得られたイソ酪酸エステルのGC純度を併記した。

<実施例14:α-ヒドロキシイソ酪酸イソアミルの合成>
冷却管、撹拌装置、ディーンスターク装置を備えた300mlガラス製丸底フラスコにα-ヒドロキシイソ酪酸25.0g(三菱ガス化学(株)製)、イソアミルアルコール105.0g(東京化成工業(株)製、2-メチルブタノール17%、3-メチルブタノール83%の異性体混合物)、ヘキサン(和光純薬工業(株)製)25.0g、パラトルエンスルホン酸1.3g(和光純薬工業(株)製)を入れ、常圧下で還流しながらエステル化反応を行い、生成する水をヘキサンと共沸させ、ディーンスターク装置で分離しながら4時間反応させた。10%水酸化ナトリウム水溶液で触媒を中和し、10%炭酸水素ナトリウム水溶液で2回、飽和塩化ナトリウム水溶液で2回洗浄した後に減圧蒸留して19hPa、84℃の留分としてα-ヒドロキシイソ酪酸イソアミル19.3g(α-ヒドロキシイソ酪酸2-メチルブチルエステル17%、α-ヒドロキシイソ酪酸3-メチルブチルエステル83%の異性体混合物としてGC純度:99.8%)を得た。この反応の反応式を下記式(7)に示した。
冷却管、撹拌装置、ディーンスターク装置を備えた300mlガラス製丸底フラスコにα-ヒドロキシイソ酪酸25.0g(三菱ガス化学(株)製)、イソアミルアルコール105.0g(東京化成工業(株)製、2-メチルブタノール17%、3-メチルブタノール83%の異性体混合物)、ヘキサン(和光純薬工業(株)製)25.0g、パラトルエンスルホン酸1.3g(和光純薬工業(株)製)を入れ、常圧下で還流しながらエステル化反応を行い、生成する水をヘキサンと共沸させ、ディーンスターク装置で分離しながら4時間反応させた。10%水酸化ナトリウム水溶液で触媒を中和し、10%炭酸水素ナトリウム水溶液で2回、飽和塩化ナトリウム水溶液で2回洗浄した後に減圧蒸留して19hPa、84℃の留分としてα-ヒドロキシイソ酪酸イソアミル19.3g(α-ヒドロキシイソ酪酸2-メチルブチルエステル17%、α-ヒドロキシイソ酪酸3-メチルブチルエステル83%の異性体混合物としてGC純度:99.8%)を得た。この反応の反応式を下記式(7)に示した。
上記の方法によって得た各種α-ヒドロキシイソ酪酸エステルにつき、調香師により香気評価を行った結果を表1に示した。
<香料材料の生分解性及び生物濃縮性の評価>
化合物の生分解性の評価方法の一つにOECDテストガイドライン301Cがあり、化合物と好気性微生物の共存する水溶液中における生化学的酸素要求量と実際の酸素消費速度から化合物の生分解性の良否を判断することができる。
この試験方法に準じた化合物の生分解する確率を、被験物質の化学構造から容易、かつ、精度よく推算する方法として「Biowin5」、「Biowin6」という計算ソフトウェアが知られている。
化合物の生分解性の評価方法の一つにOECDテストガイドライン301Cがあり、化合物と好気性微生物の共存する水溶液中における生化学的酸素要求量と実際の酸素消費速度から化合物の生分解性の良否を判断することができる。
この試験方法に準じた化合物の生分解する確率を、被験物質の化学構造から容易、かつ、精度よく推算する方法として「Biowin5」、「Biowin6」という計算ソフトウェアが知られている。
また、化合物の生物濃縮性の評価方法の一つにOECDテストガイドライン305があり、魚へ暴露した場合に、化合物が魚体に取り込まれる量によって濃縮度を判断することができる。この試験方法に準じた化合物の生物濃縮度を、被験物質の化学構造から容易、かつ、精度よく推算する方法として「BCFWIN」という計算ソフトウェアが知られている。
該ソフトウェアはアメリカ合衆国環境保護庁(United States Environmental Protection Agency, EPA)が化学物質の環境への影響を評価する目的で作成した「The Estimations Programs Interface for Windows version 4.1」という計算ソフトウェアのモジュールの1つとして公共に配布されており、Globally Harmonized System of Classification and Labelling of Chemicals(GHS)の化合物分類やアメリカ合衆国環境保護庁の新規化学物質審査において利用されている。このソフトウェアを用いて、既存の香料材料と本発明の化合物の生分解性及び生物濃縮性の違いを評価した。
該ソフトウェアはアメリカ合衆国環境保護庁(United States Environmental Protection Agency, EPA)が化学物質の環境への影響を評価する目的で作成した「The Estimations Programs Interface for Windows version 4.1」という計算ソフトウェアのモジュールの1つとして公共に配布されており、Globally Harmonized System of Classification and Labelling of Chemicals(GHS)の化合物分類やアメリカ合衆国環境保護庁の新規化学物質審査において利用されている。このソフトウェアを用いて、既存の香料材料と本発明の化合物の生分解性及び生物濃縮性の違いを評価した。
ミントの香調を持つ本発明の化合物に類似する既存の香料材料の代表例としてメントール、メントン、カルボンを選択し、本発明の化合物と共に評価を行った。ソフトウェアへの入力に用いたSMILES式と「Biowin5(線形予測モデル)」、「Biowin6(非線形予測モデル)」による良分解性の確率の出力結果を表2~表3に示した。結果の数字は大きい程、良分解性を示し、0.5以上で良分解性(表中、記号“A”)、0.5未満で難分解性(表中、記号“B”)と判定される。
また、「BCFWIN version3.01」による生体濃縮性の評価として「regression-based method」及び「Arnot-Gobas method」両方法による出力結果を表2~表3に示した。両方法のどちらも数字が大きい程、環境から魚体へ濃縮することを意味し、食物連鎖によって環境へ悪影響を及ぼす指標となる。
表2~表3から類似する既存の香料材料であるメントール、メントン、カルボンに対して、本発明の化合物は良好な生分解性及び低い生物濃縮性が期待できる結果が得られた。本発明の化合物は香料として環境に放出された後に容易に生分解し、かつ生体濃縮し難いことにより、より環境への負荷が少ない傾向を示した。
また、「BCFWIN version3.01」による生体濃縮性の評価として「regression-based method」及び「Arnot-Gobas method」両方法による出力結果を表2~表3に示した。両方法のどちらも数字が大きい程、環境から魚体へ濃縮することを意味し、食物連鎖によって環境へ悪影響を及ぼす指標となる。
表2~表3から類似する既存の香料材料であるメントール、メントン、カルボンに対して、本発明の化合物は良好な生分解性及び低い生物濃縮性が期待できる結果が得られた。本発明の化合物は香料として環境に放出された後に容易に生分解し、かつ生体濃縮し難いことにより、より環境への負荷が少ない傾向を示した。
<実施例15:ホワイトフローラルタイプの香料組成物>
表4に示す組成を持つ香料組成物78.7質量部に、実施例3で得られたα-ヒドロキシイソ酪酸イソプロピル21.3質量部を加えた香料組成物を調合した。
調香師による香気評価により、表4に記載した組成を持つ香料組成物に実施例3のα-ヒドロキシイソ酪酸イソプロピルを添加することにより、爽やかでフレッシュな軽い感じのフローラルグリーンを付与することかできた。その結果、ミント様、及びライラック様フローラルグリーンの香気が付与された新規なホワイトフローラルタイプの香料組成物が得られた。この香料組成物の香気は化粧水、デオドラントシート、ボディーパウダーなどへの賦香に適すると思われる。
表4に示す組成を持つ香料組成物78.7質量部に、実施例3で得られたα-ヒドロキシイソ酪酸イソプロピル21.3質量部を加えた香料組成物を調合した。
調香師による香気評価により、表4に記載した組成を持つ香料組成物に実施例3のα-ヒドロキシイソ酪酸イソプロピルを添加することにより、爽やかでフレッシュな軽い感じのフローラルグリーンを付与することかできた。その結果、ミント様、及びライラック様フローラルグリーンの香気が付与された新規なホワイトフローラルタイプの香料組成物が得られた。この香料組成物の香気は化粧水、デオドラントシート、ボディーパウダーなどへの賦香に適すると思われる。
<実施例16:ミントタイプの香料組成物>
L-メントール10質量部に、実施例3で得られたα-ヒドロキシイソ酪酸イソプロピル90質量部を加えた香料組成物を調合した。また、比較対象としてL-メントール10質量部に、99.5%エタノール90質量部を加えた香料組成物を調合した。調香師による香気評価によって、両者の香気の違いを比較評価した。同様の方法でDL-メントン、L-カルボンについても、各々行った。その香気の比較評価の結果を表5にまとめた。
L-メントール、DL-メントン、L-カルボンなどの代表的なミント調の香料材料を、ミント調のα-ヒドロキシイソ酪酸イソプロピルで希釈すると、L-メントール、DL-メントン、L-カルボンのミント調を邪魔することなく、強くリフトアップして拡散性が増し、ミントの清涼感がより強く感じられるようになり、同時にフローラル、グリーンな感じも付加されて、よりナチュラルなミント様の香気が得られた。実施例3で得られたα-ヒドロキシイソ酪酸イソプロピルはミント調香料素材のリフトアップ材料として有用であった。
L-メントール10質量部に、実施例3で得られたα-ヒドロキシイソ酪酸イソプロピル90質量部を加えた香料組成物を調合した。また、比較対象としてL-メントール10質量部に、99.5%エタノール90質量部を加えた香料組成物を調合した。調香師による香気評価によって、両者の香気の違いを比較評価した。同様の方法でDL-メントン、L-カルボンについても、各々行った。その香気の比較評価の結果を表5にまとめた。
L-メントール、DL-メントン、L-カルボンなどの代表的なミント調の香料材料を、ミント調のα-ヒドロキシイソ酪酸イソプロピルで希釈すると、L-メントール、DL-メントン、L-カルボンのミント調を邪魔することなく、強くリフトアップして拡散性が増し、ミントの清涼感がより強く感じられるようになり、同時にフローラル、グリーンな感じも付加されて、よりナチュラルなミント様の香気が得られた。実施例3で得られたα-ヒドロキシイソ酪酸イソプロピルはミント調香料素材のリフトアップ材料として有用であった。
<実施例17:ガーデニアタイプの香料組成物>
表6に示す組成を持つ香料組成物84.3質量部に、実施例4で得られたα-ヒドロキシイソ酪酸ノルマルブチル15.7質量部を加えた香料組成物を調合した。
調香師による香気評価により、表6に記載した組成を持つ香料組成物に実施例4のα-ヒドロキシイソ酪酸ノルマルブチルを添加することにより、ガーデニア特有の甘さを強調することができ、スッキリとしたグリーンを付与することができた。その結果、グリーン調が付与され、ミルク様の甘さが強調された新規なガーデニアタイプの香料組成物が得られた。この香料組成物の香気はヘアートリートメント、乳液、スキンクリーム、ボディーローションなどへの賦香に適すると思われる。
表6に示す組成を持つ香料組成物84.3質量部に、実施例4で得られたα-ヒドロキシイソ酪酸ノルマルブチル15.7質量部を加えた香料組成物を調合した。
調香師による香気評価により、表6に記載した組成を持つ香料組成物に実施例4のα-ヒドロキシイソ酪酸ノルマルブチルを添加することにより、ガーデニア特有の甘さを強調することができ、スッキリとしたグリーンを付与することができた。その結果、グリーン調が付与され、ミルク様の甘さが強調された新規なガーデニアタイプの香料組成物が得られた。この香料組成物の香気はヘアートリートメント、乳液、スキンクリーム、ボディーローションなどへの賦香に適すると思われる。
<実施例18:ミューゲタイプの香料組成物>
表7に示す組成を持つ香料組成物83.6質量部に、実施例5で得られたα-ヒドロキシイソ酪酸イソブチル16.4質量部を加えた香料組成物を調合した。
調香師による香気評価により、表7に記載した組成を持つ香料組成物に実施例5のα-ヒドロキシイソ酪酸イソブチルを添加することにより、スッキリとしたグリーン調が付与され、ウッディ調が強調された甘さのある上品で新規なミューゲタイプの香料組成物が得られた。この香料組成物の香気はシャンプー、ボディーソープ、洗顔フォームなどへの賦香に適すると思われる。
表7に示す組成を持つ香料組成物83.6質量部に、実施例5で得られたα-ヒドロキシイソ酪酸イソブチル16.4質量部を加えた香料組成物を調合した。
調香師による香気評価により、表7に記載した組成を持つ香料組成物に実施例5のα-ヒドロキシイソ酪酸イソブチルを添加することにより、スッキリとしたグリーン調が付与され、ウッディ調が強調された甘さのある上品で新規なミューゲタイプの香料組成物が得られた。この香料組成物の香気はシャンプー、ボディーソープ、洗顔フォームなどへの賦香に適すると思われる。
<実施例19:金木犀タイプの香料組成物>
表8に示す組成を持つ香料組成物84.8質量部に、実施例5で得られたα-ヒドロキシイソ酪酸イソブチル15.2質量部を加えた香料組成物を調合した。
調香師による香気評価により、表8に記載した組成を持つ香料組成物に実施例5のα-ヒドロキシイソ酪酸イソブチルを添加することにより、スッキリとしたグリーン調が付与され、ミルク様の甘さ及びウッディ調が強調されたシャープで温かな甘さのある新規な金木犀タイプの香料組成物が得られた。この香料組成物の香気はボディーローション、ハンドクリームなどへの賦香に適すると思われる。
表8に示す組成を持つ香料組成物84.8質量部に、実施例5で得られたα-ヒドロキシイソ酪酸イソブチル15.2質量部を加えた香料組成物を調合した。
調香師による香気評価により、表8に記載した組成を持つ香料組成物に実施例5のα-ヒドロキシイソ酪酸イソブチルを添加することにより、スッキリとしたグリーン調が付与され、ミルク様の甘さ及びウッディ調が強調されたシャープで温かな甘さのある新規な金木犀タイプの香料組成物が得られた。この香料組成物の香気はボディーローション、ハンドクリームなどへの賦香に適すると思われる。
<実施例20:ジャスミンタイプの香料組成物>
表9に示す組成を持つ香料組成物80.5質量部に、実施例13で得られたα-ヒドロキシイソ酪酸4-メチルペンタン-2-イル19.5質量部を加えた香料組成物を調合した。
調香師による香気評価により、表9に記載した組成を持つ香料組成物に実施例13のα-ヒドロキシイソ酪酸4-メチルペンタン-2-イルを添加することにより、全体的にまとまりがでて、バランスが良くなった。その結果、スパイシーな温かさ、フルーティな甘さが付与されたナチュラルで華やかさのある新規なジャスミンタイプの香料組成物が得られた。この香料組成物の香気はスキンクリーム、石鹸、ヘアムースなどへの賦香に適すると思われる。
表9に示す組成を持つ香料組成物80.5質量部に、実施例13で得られたα-ヒドロキシイソ酪酸4-メチルペンタン-2-イル19.5質量部を加えた香料組成物を調合した。
調香師による香気評価により、表9に記載した組成を持つ香料組成物に実施例13のα-ヒドロキシイソ酪酸4-メチルペンタン-2-イルを添加することにより、全体的にまとまりがでて、バランスが良くなった。その結果、スパイシーな温かさ、フルーティな甘さが付与されたナチュラルで華やかさのある新規なジャスミンタイプの香料組成物が得られた。この香料組成物の香気はスキンクリーム、石鹸、ヘアムースなどへの賦香に適すると思われる。
本発明のα-ヒドロキシイソ酪酸エステル化合物は、優れた香気を有し、それ自体を香料として使用することが期待されると共に、該化合物を調合香料素材として使用することにより、香気性に優れた香料組成物が得られ、各種製品に配合することにより、所望の賦香性を発揮するものである。
更に、実施例で得られた化合物は、いずれも優れた生分解性及び低い生物濃縮性を有し、環境への負荷が低いものであり、使用に適するものであることが示された。

Claims (8)
- 式(1)中、Rがエチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、セカンダリーブチル基、ターシャリーブチル基、3-メチルブタン-2-イル基、2-メチルブチル基、3-メチルブチル基、ネオペンチル基、2-メチルペンチル基、4-メチルペンタン-2-イル基、ノルマルヘキシル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択される、請求項1に記載の香料組成物。
- 式(1)中、Rがエチル基、ノルマルプロピル基、イソプロピル基、ノルマルブチル基、イソブチル基、セカンダリーブチル基、ターシャリーブチル基、3-メチルブタン-2-イル基、2-メチルブチル基、3-メチルブチル基、ネオペンチル基、2-メチルペンチル基、4-メチルペンタン-2-イル基、ノルマルヘキシル基、シクロペンチル基、及びシクロヘキシル基よりなる群から選択される、請求項3に記載の使用。
- 式(1)で表される化合物が、ミント様の香りを付与する、請求項3又は4に記載の使用。
- 式(1)で表される化合物が、グリーンノートの香りを付与する、請求項3又は4に記載の使用。
