Abstract
解決しようとする課題は、対症療法ではなく、原因療法となりうる、実際に効果のあるアルツハイマー病の認知症の治療法とその治療物質である。 解決手段 微小管結合タンパク質1B遺伝子を脳内の神経細胞で過剰発現させ、アルツハイマー病の認知症などの症状を改善させる物質。 また、これを達成する方法として、微小管結合タンパク質1B遺伝子に遺伝子治療用ベクターを結合させたものを脳内に注入する方法と、微小管結合タンパク質1B遺伝子 をBBB(血液脳関門)通過型ナノマシンに結合させて血管内投与する大きく分けて2つの方法がある。
Description
本発明は、アルツハイマー病の認知症の治療薬に関するものである。より詳しくは、脳の神経細胞に、遺伝子を導入、発現させるものであり、遺伝子治療に関する。
現状では、アルツハイマー病の原因がアミロイドの脳内蓄積によるということは解っているが、そのアミロイドにより脳の神経細胞がどのようにしてダメージをうけるかが諸説あり決定していない。そのため、原因療法はアミロイド抗体(非特許文献1)などアミロイドを標的としたものにほとんど限られるだけだが、うまくいっていない。そのため、対症療法が主に行われている。
ドネペジルという薬剤により、脳内のアセチルコリンという神経伝達物質を増やすことができる(特許文献1、非特許文献2)。しかしながら記憶をつかさどるシナプスの神経伝達物質はグルタミン酸なので、アセチルコリンは対症療法に過ぎず、アルツハイマー病の初期から中期の症状が進行するのを僅かに抑制できるのみであり、その効果は限定的である。
すなわち、これらの、従来の対症療法では、認知症に対する効果が非常にわずかで、臨床的にほとんど効果が見られないものがほとんどであり、アルツハイマー病の認知症を有効に治療できる薬剤、方法は存在しないと言っても過言ではない。
この問題の解決方法として、原因療法と考えられるアミロイド抗体による治療法があるが、効果が少なく、脳浮腫などの副作用が強いという欠点があり、その問題の解消が求められていた。

本発明によれば、以下の発明が提供される。
(1)i)微小管結合タンパク質1B(MBP1B)をコードするDNA
ii)MBP1BをコードするDNAと70%以上の相同性を有し、同等の機能を有するタンパク質をコードする遺伝子
iii)MBP1BをコードするDNAとストリンジェントな条件でハイブリダイズし、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA
iv)MBP1Bタンパク質のアミノ酸配列と60%以上の同一性を有し、該MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA
のいずれかを含む、認知症治療薬。
本発明においては、微小管結合タンパク質1B遺伝子またはそれと相同で同一又は類似の機能を有する遺伝子を脳内の神経細胞で過剰発現させ、アルツハイマー病の認知症などの症状を改善させる方法と物質を提供する。また、コードする遺伝子とは、オープンリーディングフレーム(ORF)または、コーディング領域(coding region)(好ましくは全長)を意味し、プロモーターに機能するように連結して細胞に導入した場合に、機能するタンパク質が発現するDNAをいう。なお、プロモーターには、必要に応じて、internal ribosomal entry site (IRES)やエンハンサー、インシュレーター等が含まれていてもよい。本発明においては原則として、(cDNAの)ORF全長を導入するが、タンパク質として同等の機能を有する限り、DNAの置換、付加、欠失等を含んでいてもよい。DNAの置換、付加、欠失等は、同等の機能を有する限り制限はないが、全塩基数の30%以下、より好ましくは、20%以下、さらに好ましくは、10%以下、特に好ましくは、1~数個である。
(2)機能するように、ベクターに連結された、(1)の認知症治療薬。
(1)を達成する方法として、微小管結合タンパク質1B遺伝子に遺伝子治療用ベクターを結合させたものを脳内に直接注入する方法と、微小管結合タンパク質1B遺伝子 をBBB(血液脳関門)通過型ナノマシンに結合させて血管内投与する大きく分けて2つの方法があるが、ここではベクターに連結されたものを提供する。
(3)血液脳関門(BBB, blood-brain barrier)通過型ナノマシンに結合されることを特徴とする、(1)の認知症治療薬。
血液脳関門(BBB, blood-brain barrier)通過型ナノマシンに微小管結合タンパク質1B遺伝子を結合させて、血管内に投与する方法と物質。血液脳関門は、脳血管内皮細胞を主体として、循環血液と脳神経実質の物質輸送を制御するもので、脳の活動に必須な栄養素を取り込む反面、薬剤の脳への送達を著しく制限している。片岡一則らは、血糖値の変化という簡単な刺激に応答して、既存技術と比較して著しく高い効率で血液脳関門を通過し、さらに脳内の神経細胞へと集積する血液脳関門通過型ナノマシンの開発に成功した。この血液脳関門通過型ナノマシンに微小管結合タンパク質1B遺伝子を結合させ、血管内に投与すれば、脳の手術の必要なく、血管内に投与するだけで脳内の神経細胞に微小管結合タンパク質1B遺伝子を発現させることが可能になる。
(4)微小管結合タンパク質1Bを脳細胞内で発現させることにより、認知症を治療する方法。
(5)微小管結合タンパク質1B遺伝子の認知症治療薬を製造するための使用。
(6)認知症治療に使用する、微小管結合タンパク質1B遺伝子。
(7)(1)~(3)のいずれか1に記載の認知症治療薬を投与することにより、認知症を治療する方法。
(8)以下のいずれかのDNAを導入し発現させることにより、認知症を治療する方法:
i)微小管結合タンパク質1B(MBP1B)をコードするDNA
ii)MBP1BをコードするDNAと70%以上の相同性を有し、同等の機能を有するタンパク質をコードする遺伝子
iii)MBP1BをコードするDNAとストリンジェントな条件でハイブリダイズし、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA
または、
iv)MBP1Bタンパク質のアミノ酸配列と60%以上の同一性を有し、該MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA。導入する部位は、脳内が好ましいが、血液脳関門を通過できる場合は、腕等に注射して血液中に導入し、脳に移動して発現させるようにしてもよい。
(9)認知症治療薬を製造するための
i)微小管結合タンパク質1B(MBP1B)をコードするDNA
ii)MBP1BをコードするDNAと70%以上の相同性を有し、同等の機能を有するタンパク質をコードする遺伝子

(7)(1)~(3)のいずれか1に記載の認知症治療薬を投与することにより、認知症を治療する方法。
(8)以下のいずれかのDNAを導入し発現させることにより、認知症を治療する方法:
i)微小管結合タンパク質1B(MBP1B)をコードするDNA
ii)MBP1BをコードするDNAと70%以上の相同性を有し、同等の機能を有するタンパク質をコードする遺伝子
iii)MBP1BをコードするDNAとストリンジェントな条件でハイブリダイズし、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA
または、
iv)MBP1Bタンパク質のアミノ酸配列と60%以上の同一性を有し、該MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA。導入する部位は、脳内が好ましいが、血液脳関門を通過できる場合は、腕等に注射して血液中に導入し、脳に移動して発現させるようにしてもよい。
(9)認知症治療薬を製造するための

本発明では、アルツハイマー病の認知症を、改善できるという効果を有する。
本明細書において、認知症とは、認知障害の一種であり、後天的な脳の器質的障害により、いったん正常に発達した知能が不可逆的に低下した状態をいう。認知症は犬や猫などヒト以外でも発症する。狭義では「知能が後天的に低下した状態」の事を指すが、本明細書では「知能」の他に「記憶」「見当識」を含む認知障害や「人格変化」などを伴った症候群を意味する。
本明細書において、認知症とは、アルツハイマー型認知症、前頭側頭認知症、パーキンソン病、レビー小体型認知症、ハンチントン病、クロイツフェルドヤコブ病、認知症を伴った筋萎縮性側索硬化症、プリオン病などの疾病において症状として認められることが多いが、これらの疾病に限られない。最近の研究では、アルツハイマー病以外のこれらの認知症を示す疾病でも、棘および/またはシナプスのロスやアミロイドの沈着がみられることが多いという報告が多い。つまり、認知症を示す疾患には共通な点が多くみられ、本明細書で主に述べるアルツハイマー病の認知症に対する治療法と治療物質が、アルツハイマー病の認知症以外の認知症にも有効である可能性がある。
本明細書において、アルツハイマー病とは、一般に高齢で発症し、病理学的に脳内へのアミロイドの沈着を病理学的特徴とする、記憶障害を主症状とする疾患である。
本発明において、微小管とは、神経細胞のなかの軸索や樹状突起や棘に見られる、繊維状の細胞内器官であり、これにより軸索や樹状突起や棘などの細胞突起を形成したり、これに沿って物質を移送したりするものを意味する。
微小管結合タンパク質(microtubule-associated protein:MBP1B、微小管関連タンパク質も同じ意味である)とは、繊維状の微小管の表面に付着して存在し、微小管の安定化、伸長を促すタンパク質である。微小管結合タンパク質として、微小管結合タンパク質1A/1B, 微小管結合タンパク質2、タウの4種類が知られている。本明細書では、特に断らない限り、MBP1BをコードするDNAはオープンリーディングフレーム(ORF)全長を含むDNAを意味し、ORFそのものであってもよく、ORFの前後にリンカー等を有していてもよい。
本発明において、遺伝子の過剰発現とは、目的の遺伝子を大量に試験管内で製造し、そのタンパク質をコードする領域に、目的の組織、器官等でその発現の程度を強くするプロモーターを機能するように連結し、またベクターなどの細胞内への導入を容易にする運搬体を結合した発現カセットを作成し、該発現カセットを目的の細胞内に挿入し(遺伝子導入)、既存細胞のタンパク質合成の仕組みを借りて、該細胞内で最終的に該当タンパク質を過剰に作らせることである。本発明においては、過剰発現には、血中に発現カセットを導入し、血液―脳関門を通過して、脳神経細胞内で過剰発現させることも含まれる。
本明細書において、ベクターとは、遺伝子を細胞内で発現させることが目的で、遺伝子に結合させ、遺伝子を細胞内にいれる運搬体である。
本明細書においてプラスミドとは、大腸菌などの細菌で染色体DNAとは独立して複製する環状の2本鎖DNAを言う。 この性質を利用して目的遺伝子を組み込んだものが、種々の細胞で遺伝子発現を起こさせるために利用される(プラスミドベクターともいう)。
本発明で、認知症(dementiaの訳であるが、患者を侮辱する言葉だということで、厚生労働省が認知症に統一した)の治療に用いるDNAとしては、i)微小管結合タンパク質1B(MBP1B)をコードするDNA、ii)MBP1BをコードするDNAと70%以上の相同性を有し、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA、iii)MBP1BをコードするDNAとストリンジェントな条件でハイブリダイズし、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA、iv)MBP1Bタンパク質のアミノ酸配列と60%以上の同一性を有し、該MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA、があげられる。これらのDNAのいずれかを脳神経細胞中に導入し、MBP1Bタンパク質またはそれと同等の機能を有するタンパク質を脳神経細胞中で発現させることにより、認知症を治療することができる。本発明の主な目的は、認知症の治療薬の提供であるが、本発明の原理から認知症の予防にも効果を発揮する場合もあり得ると考えられる。
ここで、導入、発現させるDNAは、MBP1BをコードするDNAと70%以上の相同性を有し、MBP1Bタンパク質と同等の機能を有することが好ましいが、より好ましくは、75%以上、80%以上、85%以上、90%以上、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、99%以上の相同性を有していてもよい。
また、ストリンジェントな条件とは、例えば、サザンハイブリダイゼーションの後、68℃、0.1×SSC、0.1%SSCで洗浄する条件が挙げられる。
本発明に使用できるMBP1Bタンパク質と同等な機能を有するタンパク質のMBP1Bタンパク質のアミノ酸配列との同一性については、60%以上が好ましく、より好ましくは、65%以上、70%以上、75%以上、80%以上、85%以上、90%以上、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、99%以上であってもよい。
これらのMAP1BをコードするDNAと相同なDNAから産生されるタンパク質がMAP1Bと同等の機能を有するかどうかは、脳神経細胞に導入して、Y迷路テスト(Neuroscience. 2006 Dec 1;143(2):395-405. Epub 2006 Sep 14. PMID:16973297)を行うことにより確認することができる。
本明細書において、血液脳関門とは、脳血管内皮細胞を主体として、循環血液と脳神経系の物質輸送を制御する機能を担っており、脳の活動に必須な栄養素を選択的に取り込む反面、薬剤の脳への送達を著しく制限する生体バリアともいえる組織をいう。
本明細書において、プロモーターとは、遺伝子からmRNAを転写させる働きを有する配列を言う。目的に応じてDNAであってもRNAであってもよいが、通常はDNAが用いられる。プロモーターにはエンハンサー、サイレンサー等のシスエレメントを有する場合もあるが、本明細書では特に区別せず、エンハンサーなどを有するプロモーターもプロモーターと呼ぶ。また、コアプロモーターの上流や下流にシスエレメントを結合させたプロモーターも使用できる。
本発明においては、脳神経細胞内(好ましくは海馬神経細胞内)で微小管結合タンパク質1Bを発現させることに特徴がある。脳神経細胞内で遺伝子を発現できるプロモーターとしては、例えば、CMV(サイトメガロウイルス)プロモーター、EF1αプロモーター等が挙げられるがこれらに限られず、脳神経細胞内で外来遺伝子を発現できるプロモーター、エンハンサーの組み合わせであればよい。

微小管結合タンパク質1B(MAP1B)をコードする領域を含むMap1b遺伝子は、公知の方法でクローニングできる。例えば、Map1b遺伝子を発現する組織(例えば海馬を含む脳組織など)からpolyA鎖を有するmRNAを調製し、好ましくはリンカー付きのポリT配列(ポリTは18~30程度が好ましい)をアニールさせて逆転写酵素によりcDNAを合成する。このcDNAを用いて、適当なプライマー、例えば、MAP1Bタンパク質をコードする領域の開始コドンを含む領域の20~30塩基程度の配列のDNAを5’-側プライマーに用いて、前記リンカー付きポリT配列を3’-側プライマーとしてPCRによりMAP1Bタンパク質をコードする領域を含むDNAを増幅することができる。PCRの条件は、プライマーの長さ、性質により当業者の周知の方法で決定できる。用いる増幅酵素(ポリメラーゼ)は正確性(fidelity)の高い(エラーの少ない)酵素を用いることが好ましい。正確性の高い増幅酵素としては、pfu,KODポリメラーゼ等が挙げられるがこれらに限られない。必要に応じてPCR産物のDNAをプラスミドにクローン化して塩基配列を決定することにより、増幅による変異がないか確認することができる。
あるいは、Map1b遺伝子の全長または一部の増幅DNA断片またはクローン化したMap1b遺伝子DNA断片などをプローブとして用いて、cDNAライブラリまたはジェノミックライブラリをプラークハイブリダイゼーション等によりスクリーニングすることにより、Map1b遺伝子をクローニングすることができる。この際、ストリンジェンシーを下げてスクリーニングすることで、Map1b遺伝子と相同性を有する遺伝子を見出し、クローニングすることができる。
PCR法またはライブラリーからスクリーニングして得たMap1b遺伝子の開始コドンから終始コドン又はその下流までを適当なプライマーを設計してPCRすることで、リンカーを挿入できる。好ましくは、cDNA合成時にリンカー付きのプライマーを用い、次の相補鎖合成後にリンカーを付けるか、またはPCR時にもリンカー付きの5’-側プライマーを用いることで2度手間を避けることができる。
これらの操作により得られたMAP1Bタンパク質をコードする領域全長を含むDNAは、機能するようにベクターに連結することができる。例えば、クロンテク社の、Lenti-X Vector(Lenti-XTM HTX Packaging System)などのプロモーター(例えば、サイトメガロウイルスプロモーター、EF1αプロモーター等)に機能するように連結させ、細胞や血液中に導入することで、タンパク質を望む細胞中で発現させることができる。
図3は、微小管結合タンパク1B遺伝子にレンチウイルスベクターを結合させたものを、Alzheimer病モデルマウスの頭蓋骨に穴を開け、定位的に右の海馬に注射した時の、Y迷路テストで測ったAlzheimerモデルマウスの痴呆の程度の変化である。ウイルスベクターを結合することにより、海馬内神経細胞での微小管結合タンパク質1B遺伝子の発現の程度が向上した。注射後二日目で、注射群の痴呆の程度が明らかに改善した(図3)。
アルツハイマー病は、脳内にアミロイドが蓄積して、これが神経細胞に悪影響をして起こると考えられているが、その仕組みは解明されていない。アミロイドは微小管結合タンパク質1Bタンパクに結合するので、それにより微小管結合タンパク質1Bタンパクが不活化され、シナプスの後部を形成する棘内の微小管が脱重合(溶ける)して棘が短縮する結果シナプスの機能が落ちると考えられる。局注後1週で、だんだん効果が薄れているが、これは、結果を早く出すために、アミロイドを産生する遺伝子内に5つも遺伝子変異のあるモデルマウスを使ったためと考えられる(Oakley, H. et.al. J. Neurosci. 40, p10129-10140 (2009))。変異が多いほど症状が早く現れる。人間のアルツハイマー病では2割が1つの遺伝子変異で、あとは遺伝子変異がない。微小管結合タンパク質1Bの効果を、非常に速いアミロイドの沈着が打ち消してしまうものと考えられる。
海馬由来の初代(プライマリー)培養神経細胞(培養後約2週間経ってシナプスが形成されたもの)に、レンチウイルスベクターに結合した微小管結合タンパク質1B遺伝子をリン酸カルシウム法により導入し過剰発現させた。レンチウイルスベクターに結合したMAP1B遺伝子を加えると、加えないもの(図4のDE)より10倍ほどにシナプスの後部を形成する樹状突起棘が拡大した(図4のAB)。D, Eは、正常の樹状突起で、微小管結合タンパク質1Bを使用していない。GFP(グリーンフルオレセインプロテイン)だけを導入しており、神経細胞全体が緑に染まっていて、ここでは細い樹状突起が染まっていた。小さな膨らみが正常な棘で、ここに、他からの神経細胞が入力し、シナプスを形成する。A,Bは、GFPに加えて、微小管結合タンパク質1Bにレンチウイルスベクターを結合させたもの+GFPを導入した。樹状突起の突起が巨大化した。Cでは、棘を代表するタンパク質の遺伝子PSD95に、黄色に光る色素の遺伝子YFP95を結合した遺伝子を導入した。大きくなった樹状突起上の突起の中に棘であることを示す遺伝子のスポットが出てきて、この大きくなった突起が棘であると分かる(図4のC, 矢印)。
上記の海馬由来培養神経細胞でレンチウイルスベクターに結合した微小管結合タンパク質1B遺伝子により大きくなった棘を、同時に微小管染色した。左で樹状突起の一部が大きくなっていることが分かる(図5)。赤が微小管染色であるが、棘として拡大していない部分の樹状突起の軸では、微小管が濃く染まっている(赤、矢頭)。棘として拡大している部分では、軸の微小管が薄くなっていた。これから、棘として拡大している部分では、軸の微小管が側方に伸びてこの側方への微小管の圧力が棘の拡大をおこしていると推察される(右はシェ-マ)。
アルツハイマー病では、棘の数が減少していることが解っているが、これは、形で判定している。同じようなことが、電気泳動→ウェスタンブロッティングという標的タンパク質を測定する生化学的な方法でも検出されることが解った。今回使用したモデルマウスの右だけの海馬にレンチウイルスベクター―微小管結合タンパク質1B遺伝子を注入し、1週後に左右の海馬を取り出し電気泳動(SDS-PAGE)した。常法により、PVDF膜等に転写後、棘、つまりシナプス後部にあるタンパク質を認識する抗体で反応させると、左Aの上のように、Rightの方が太くなり、微小管結合タンパク質1B遺伝子注入群の、棘にある減少していたタンパク質が回復していることを示した(図6)。図6Bは、Aをデンシトメーターにより定量化したものであるが、N=4で、5%有意でRightの方が多いことがわかる。
遺伝子治療用ベクターの種類としては、ウイルスベクター(ウイルスが細胞に感染する機構を利用した遺伝子導入法)の中に、非増殖性ウイルスベクターとして、レトロ、アデノ、レンチ、アデノ随伴ウイルス(AAV)、センダイウイルス等があり、増殖性ウイルスベクターとして、腫瘍溶解性ウイルス等 があり、 非ウイルスベクターとして、プラスミド(Naked DNA(裸のDNA)、リポソームやポリマーとの複合体)や、バクテリアベクター(遺伝子改変した細菌)などがあげられるがこれらに限られない。
本発明に使用するウイルスベクターとしては、レトロ、アデノ、レンチ、アデノ随伴ウイルス(AAV)、センダイウイルス等であってもよく、非ウイルスベクターとしては、プラスミド、バクテリアベクター等でもよい。また、従来のいわゆるベクターの代わりに、血液脳関門(BBB, blood-brain barrier)通過型ナノマシンを用いても良い。要は、ベクターは、遺伝子を脳神経細胞中で発現できるものであればよく、この目的を達成できれば特に制限されない。
本発明によれば、アルツハイマー病の認知症を、対症療法ではなく、原因療法として、劇的に改善するという効果を有する。ベクターを用いる場合では、小さな手術で脳内に注入しなければならないが、血液脳関門通過型ナノマシンを用いる場合には、血管内への注射だけで、脳内に作用させることができる。
(実施例1)
マウス微小管結合タンパク質1B遺伝子にレンチウイルスベクターを結合させたものを、アルツハイマー病モデルマウスの頭蓋骨に穴を開け、定位的に右の海馬に注入し、アルツハイマー病の認知症を改善させる方法

マウス微小管結合タンパク質1B遺伝子は、以下のようにしてクローニングした。BL6マウスの脳の全RNAを、RNeasy Mini Kit (Quiagen)を用いて調製した。次に、全RNAは、ポリTプライマーを用い、SuperScript III(Invitrogen)により逆転写された。常法によりcDNAライブラリーを作成し、Map1b遺伝子断片をプローブとしてスクリーニングすることで、Map1b遺伝子をクローニングし、塩基配列を確認した。マウス微小管結合タンパク質1B遺伝子(Map1b遺伝子のタンパク質コーディング領域)は、Lenti-XTM 293T Cell Line と Lenti-X HTX Packaging System (クロンテクを用いて、レンチウイルスベクターと結合させた。このキメラ遺伝子溶液1マイクロリッターを、モデルマウス B6SJLTg(APPSwFlLon,PSEN1*M146L*L286V)6799Vas/Mmjax(The Jackson Laboratory、https://www.mmrrc.org/about/mmrrcContacts.phpより購入)、034840-JAX-HEMI-F - Hemizygous female、(Oakley, et al. J Neurosci. 2006 26:10129-40)の冠状矢状縫合交差より右に2mm、後ろへ2mm、脳表から1mmのところへ頭蓋固定定位脳内局所注入装置にて注入した。
アミロイド前駆体タンパク質遺伝子、プレセニリン遺伝子(トータル5変異)の2重トランスジェニックマウスの頭蓋骨に、マウス頭蓋固定、マイクロインジェクションシステム(ナリシゲ、東京、日本)を用いて穴を開け、定位的に右の海馬に注射した(ブレグマより右へ2mm、後ろへ2mm、脳表より1mm)。この時の、Y迷路テストで測ったAlzheimer病モデルマウスの痴呆の程度の変化を示す。ウイルスベクターが付いたマウス微小管結合タンパク質1B遺伝子を海馬内に注入することにより、海馬内神経細胞で、微小管結合タンパク質1Bが過発現した。二日で、注射群の痴呆の程度が明らかに改善したことを確認できた(図3)。
(実施例2)

すなわち、海馬は、1mlエッペンドルフ中で、プロテアーゼインヒビターを含む2倍容量の4倍レムリサンプルバッファー(Laemmli sample buffer, Bio-Rad、東京)中でホモジェナイズされ、その10~20μLが12%ミニプロテアンTGXゲル(Bio-Rad)で電気泳動された。ポリビニリデン・ジフルオライド膜(PVDF膜)に転写後、棘、つまりシナプス後部にあるタンパク質を認識する1次抗体(HOMER1ウサギポリクローナル抗体、1:500, Proteintec Group Inc., Rosemont, IL, USA)及び内部コントロールとしてanti-tublin(1:10000, DM1A, Sigma, St. Lois, MO)で反応させた。その後、PVDF膜は、西洋ワサビペルオキシダーゼを連結した2次抗体(アマシャム)とインキュベートし、シグナルは、Image Quant 400(GEヘルスケア、Buckinghamshire, 英国)により検出した。定量的バンドパターンはImageJ(NIH)により得た。結果は、左Aの上のように、Rightの方が太くなり、微小管結合タンパク質1B遺伝子注入群の、棘にある減少していたタンパク質が回復していることを示した(図6)。
マウス微小管結合タンパク質1B遺伝子にレンチウイルスベクターを結合させたものの製造方法はすでに述べたので、マウス微小管結合タンパク質1B遺伝子に血液脳関門(BBB, blood-brain barrier)通過型ナノマシンを結合したものの作成方法を述べる。
ミセル化ナノ粒子(高分子ミセル)は、水に溶けやすい性質を示すポリエチレングリコール(PEG)からなる親水性ポリマーと水に溶けにくい性質を示すポリアミノ酸からなる疎水性ポリマーを分子レベルで結合させたブロックコポリマー(共重合体)から構成される。ブロックコポリマーを水中で拡散すると、外側が親水性ポリマーで内側が疎水性ポリマーという明確な二層構造を有する20~100ナノメートルサイズの球状の集合体であるミセルを形成する。このミセルの疎水性内核部分に薬物や生理活性物質を封入することができる。アミノ酸の種類や構造を化学的に変化させることで様々な化合物に対応が可能である。表面をPEG(ポリエチレングルコール)が覆うことで血液中での安定性を確保できる。脳には、血液脳関門という特殊な血液と脳組織とを隔てる組織があり、それは、脳血管内皮細胞を主体として、循環血液と脳神経系の物質輸送を制御する機能をになっており、脳の活動に必須な栄養素を選択的に取り込む反面、薬剤の脳への送達を著しく制限する生体バリアともいえる組織である。脳の活動に必須なブドウ糖などを選択的に取り込む反面、薬剤の送達を著しく制限している。このような背景において、片岡一則らは、血糖値の変化で著しく高い効率で脳血液関門を通過しさらに脳内の神経細胞へと集積する脳血液関門通過型ナノマシン)を開発した(Nature Communications 8、 Article number 1001, 2017).
疎水性ブロックの端にグルコース分子を付けることで、空腹時にナノマシンを注射しその後に食事をするという簡単な方法で、脳内に薬を効率よく運ぶことができる。かかるナノマシンに微小管結合タンパク質1B遺伝子を発現するように結合させて、静脈注射し、その後血糖値を上げることにより血液脳関門を通過させ、脳神経細胞内で、微小管結合タンパク質1Bを発現させることにより、認知症を予防ないし治療できる。
本発明の微小管結合タンパク質1B遺伝子に血液脳関門(BBB, blood-brain barrier)通過型ナノマシンを結合したものや、微小管結合タンパク質1B遺伝子にベクターを結合したものは、人のアルツハイマー病の治療に使用できる可能性が高いことから、医療産業において利用できる。

Claims (3)
- i)微小管結合タンパク質1B(MBP1B)をコードするDNA、
ii)MBP1BをコードするDNAと70%以上の相同性を有し、MBP1Bと同等の機能を有するタンパク質をコードするDNA、
iii)MBP1BをコードするDNAとストリンジェントな条件でハイブリダイズし、MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA、
iv)MBP1Bタンパク質のアミノ酸配列と60%以上の同一性を有し、該MBP1Bタンパク質と同等の機能を有するタンパク質をコードするDNA
のいずれかを含む、認知症治療薬。 - 機能するように、ベクターに連結された、請求項1の認知症治療薬。
- 血液脳関門(BBB, blood-brain barrier)通過型ナノマシンに結合されることを特徴とする、請求項1または2の認知症治療薬。
