Description
本発明は、夜間頻尿の予防若しくは改善剤、並びに夜間頻尿の予防若しくは改善用飲食品組成物に関する。
頻尿とは、様々な原因により1日あたりの排尿回数が増える症状である。頻尿の原因としては、尿崩症、高血圧症、水分、カフェインないしアルコールの過剰摂取、加齢などの尿量の増加に因るものと、過活動膀胱、尿路感染、間質性膀胱炎など尿量の増加に因らないものに大別される。さらに、膀胱や前立腺など泌尿器系の臓器における機能異常の存在や、神経的な緊張も、頻尿の原因の1つと言われている。
頻尿などの排尿障害の1つとして、夜間頻尿が注目されている。夜間頻尿とは睡眠時に排尿のために起きなければならない症状であり、排尿障害のなかでも夜間頻尿を抱える有症者が特に多いことが知られている。夜間頻尿の有症者は質の高い睡眠をとることができないため、日常生活に支障をきたし生活の質を低下させる。また、夜間頻尿は加齢とともに発症することが多く、40歳以上の日本人の有症者は4500万人を超えると推定されている。そのため、近年の高齢化社会に伴って夜間頻尿は注目を集めている。
夜間頻尿は、夜間の尿量が多いこと、膀胱容量の減少など、発症原因が様々であり、多くの要因が関与して発症すると考えられている。夜間頻尿が発症すると、睡眠中の尿意や下腹部の緊張感により、排尿に関する不安感が生じ、健康な睡眠が妨げられる場合がある。しかし、夜間頻尿は、睡眠時以外の頻尿(「昼間頻尿」とも言われる)とは発症原因が異なると考えられており、その発症機構には未だ不明な点が多い。

これまでに、過活動膀胱の予防又は改善に有効な漢方が、西日本泌尿器科, 2006年, 第68巻, p. 51-54などで提案されている。しかし、西日本泌尿器科, 2006年, 第68巻, p. 51-54に記載されている漢方は、尿意切迫感や昼間頻尿の予防若しくは改善には有効であっても、睡眠時の排尿回数は減少しないと報告されている。
現在のところ、薬物による排尿障害の治療には、膀胱収縮を促すアセチルコリンの作用を抑制する抗コリン薬が有効であることも報告されている。EUROPEAN UROLOGY, 2012年, 第62巻, p. 1040-1060には、抗コリン薬の服用により、過活動膀胱の症状の1つである昼間排尿が改善される一方で、夜間頻尿には満足な効果を示すものがないことが記載されている。また、抗コリン薬の服用は、口の渇き、便秘、排尿の困難性等の副作用を起こすことが知られている。さらに抗コリン薬は、放出されたアセチルコリンが膀胱平滑筋の受容体へ結合するのを阻害するが、これはいわゆる対症療法である。これらの理由から、抗コリン薬の服用は、効果の点から夜間頻尿の十分な治療法とは言い難い。
このような実情から、より安全で効果的な夜間頻尿の予防又は改善剤が望まれている。

一方、ロズマリン酸が、膀胱尿管逆流症、尿失禁、腎障害などの予防又は改善に有効であることが報告されている(特開2017-52747号公報、国際公開第2006/020994号、Tissue Cell, 2011年, 第43巻, p. 392-397及びBasic & Clinical Pharmacology & Toxicology, 2012年, 第110巻,p. 390-395参照)。
しかし、ロズマリン酸の摂取が夜間頻尿の予防又は改善に有効であることについて、全く知られていない。

本発明者らは、夜間頻尿を予防若しくは改善しうる物質について鋭意検討を行った。その結果、ロズマリン酸に夜間頻尿を予防若しくは改善する作用があり、夜間頻尿の予防若しくは改善に有用であることを見出した。
本発明はこれらの知見に基づいて完成するに至ったものである。

本発明の夜間頻尿の予防若しくは改善剤、並びに夜間頻尿の予防若しくは改善用飲食品組成物は、夜間頻尿を予防又は改善し、睡眠時に排尿のために起きる回数を減らすことができる。さらに、本発明の夜間頻尿の予防若しくは改善剤、並びに夜間頻尿の予防若しくは改善用飲食品組成物は、睡眠中の尿意を低減し若しくは下腹部の緊張感を和らげ、健康な睡眠をサポートすることができる。また、本発明の夜間頻尿の予防若しくは改善剤、又は夜間頻尿の予防若しくは改善用飲食品組成物を投与又は摂取させることで、睡眠中の排尿に関する不安感を軽減することができる。
本明細書において「予防」とは、個体における疾患若しくは症状の発症の防止若しくは遅延、又は個体の疾患若しくは症状の発症の危険性を低下させることをいう。
また、本明細書において「改善」とは、疾患、症状若しくは状態の好転、疾患、症状若しくは状態の悪化の防止若しくは遅延、又は疾患、症状若しくは状態の進行の逆転、防止若しくは遅延をいう。

本明細書における「夜間頻尿」とは、睡眠時に排尿のために起きなければならない症状を指す。排尿のために睡眠時に1回以上起きることを通常、夜間頻尿と定義される。
なお、「夜間頻尿」は、「昼間頻尿」とは明確に区別される。その理由としては、例えば、通常夜間であれば分泌が増加する抗利尿ホルモンが加齢に伴い減少するため、夜間頻尿が起こることが挙げられる。さらに、通常夜間であれば昼間よりも膀胱容量が増加するが、シフトワークなどのサーカディアンリズムの乱れに伴い夜間の膀胱容量が低下することも、原因の1つとして挙げられる。

本発明の夜間頻尿の予防又は改善剤(以下「本発明の予防又は改善剤」とも表記する)は、ロズマリン酸又はその塩を有効成分とする。また本発明の夜間頻尿の予防又は改善用飲食品組成物(以下単に「飲食品組成物」とも表記する)は、ロズマリン酸又はその塩を有効成分として含有する。
前記の塩としては、薬学的に許容できる塩であれば特に制限されないが、例えば、ナトリウム塩及びカリウム塩等のアルカリ金属塩、カルシウム塩及びマグネシウム塩等のアルカリ土類金属塩、アンモニウム塩及びトリエチルアミン塩等のアミン塩などが挙げられる。本発明において、前記の塩としては、アルカリ金属塩及びアルカリ土類金属塩からなる群より選ばれる少なくとも1種の塩が好ましく、ナトリウム塩及びカルシウム塩からなる群より選ばれる少なくとも1種の塩がより好ましく、ナトリウム塩がさらに好ましい。
前記の塩としては、薬学的に許容できる塩であれば特に制限されないが、例えば、ナトリウム塩及びカリウム塩等のアルカリ金属塩、カルシウム塩及びマグネシウム塩等のアルカリ土類金属塩、アンモニウム塩及びトリエチルアミン塩等のアミン塩などが挙げられる。本発明において、前記の塩としては、アルカリ金属塩及びアルカリ土類金属塩からなる群より選ばれる少なくとも1種の塩が好ましく、ナトリウム塩及びカルシウム塩からなる群より選ばれる少なくとも1種の塩がより好ましく、ナトリウム塩がさらに好ましい。
ロズマリン酸又はその塩の製造方法に特に制限はなく、通常の有機化学的合成により得ることができる。あるいは、天然物由来の材料から抽出や精製等により得ることもできる。また、試薬として市販されているものを用いることもできる。
ロズマリン酸又はその塩は、例えば、特公平1-29558号公報に記載の方法を参考に、シソ科(Lamiaceae)植物細胞の懸濁培養により製造することができる。

さらに本発明では、有効成分であるロズマリン酸又はその塩として、ロズマリン酸又はその塩を含有する天然物から抽出され、ロズマリン酸又はその塩を含有する抽出物を用いることもできる。ロズマリン酸又はその塩を含有する天然物としてはシソ(Lamiaceae)科の植物が挙げられ、具体的にはアカジソ(Perilla frutescens Britton var.acuta Kudo)、アオジソ(Perilla frutescens var.crispa f.viridis.Makino)、ローズマリー(Rosmarinus officinalis)、レモンバーム(Melissa officinalis)、セージ(Salvia officinalis)及びクミスクチン(Orthosiphon aristatus)からなる群より選ばれる少なくとも1種が好ましく用いられる。このうち本発明では、ロズマリン酸又はその塩を含有するアカジソ抽出物を用いることがより好ましい。
ロズマリン酸又はその塩を含有する、シソ科の植物の抽出物を得るには、前記シソ科植物の全草、葉、茎、根、及び種子からなる群より選ばれる1種、好ましくはシソ科植物の葉から常法に従って抽出すればよい。
抽出方法としては、水溶性溶媒を用いた溶媒抽出法が好ましい。水溶性溶媒としては、水、水溶性有機溶媒又はこれらの混合溶媒が挙げられる。水溶性有機溶媒としては、エタノール、ブタノール等の炭素数1~5の1価アルコール類、エチレングリコール、プロピレングリコール、1,3-ブチレングリコール、1,4-ブチレングリコール等の多価アルコールが挙げられる。これらのうち、水、エタノール又はエタノール水溶液が好ましく用いられる。また抽出条件は、室温下でも高温下でもいずれでもよい。また、常圧で抽出を行ってもよいし、加圧条件下で抽出を行ってもよい。さらには、静置抽出を行ってもよく、撹拌抽出を行ってもよい。
また、得られた抽出物は、ロズマリン酸又はその塩の含有率をより高めるために、公知方法により適宜分画ないし精製を行ってもよい。具体的には、European Journal of Nutrition, 2005年, 第44巻, p. 1-9やPlanta medica, 2016年, 第82巻, p. 388-406に記載された方法を参照することができる。
また、ロズマリン酸又はその塩を含有する抽出物として市販品を用いてもよい。例えば、アカジソ抽出物としては、製品名:ニチノー 赤シソエキスRA15P(日農化学工業社製)、アオジソ抽出物としては、製品名:シソエキスOGI(小川香料社製)、ローズマリー抽出物としては、製品名:ローズマリーエキスMF(丸善製薬社製)、レモンバーム抽出物としては、製品名:レモンバームエキスパウダーMF(丸善製薬社製)、セージ抽出物としては、製品名:サルビア・セージ(Finzelberg社製)、クミスクチン抽出物としては、製品名:クミスクチンエキス末(金秀バイオ社製)などが挙げられる。
アカジソ、アオジソ、ローズマリー、レモンバーム、セージ及びクミスクチンなどのシソ科植物は、従来より食用として用いられている。よって、本発明の有効成分であるロズマリン酸又はその塩としてシソ科植物由来の抽出物を用いると、安全性の高い夜間頻尿の予防若しくは改善剤並びに飲食品組成物を提供することができる。
シソ科植物の抽出物を用いる場合、抽出物中のロズマリン酸及びその塩が占める割合は、抽出物固形分に対してロズマリン酸量換算で0.1質量%以上が好ましく、より好ましくは1質量%以上、さらに好ましくは10質量%以上である。上限値は100質量%であるが、95質量%以下が好ましく、90質量%以下がより好ましい。あるいは、0.1質量%以上100質量%以下が好ましく、1質量%以上95質量%以下がより好ましく、10質量%以上90質量%以下がさらに好ましい。なお、本明細書において「抽出物固形分」とは、抽出物を105℃の乾燥機で3時間乾燥して揮発物質を除いた残分をいう。
抽出方法としては、水溶性溶媒を用いた溶媒抽出法が好ましい。水溶性溶媒としては、水、水溶性有機溶媒又はこれらの混合溶媒が挙げられる。水溶性有機溶媒としては、エタノール、ブタノール等の炭素数1~5の1価アルコール類、エチレングリコール、プロピレングリコール、1,3-ブチレングリコール、1,4-ブチレングリコール等の多価アルコールが挙げられる。これらのうち、水、エタノール又はエタノール水溶液が好ましく用いられる。また抽出条件は、室温下でも高温下でもいずれでもよい。また、常圧で抽出を行ってもよいし、加圧条件下で抽出を行ってもよい。さらには、静置抽出を行ってもよく、撹拌抽出を行ってもよい。
また、得られた抽出物は、ロズマリン酸又はその塩の含有率をより高めるために、公知方法により適宜分画ないし精製を行ってもよい。具体的には、European Journal of Nutrition, 2005年, 第44巻, p. 1-9やPlanta medica, 2016年, 第82巻, p. 388-406に記載された方法を参照することができる。
また、ロズマリン酸又はその塩を含有する抽出物として市販品を用いてもよい。例えば、アカジソ抽出物としては、製品名:ニチノー 赤シソエキスRA15P(日農化学工業社製)、アオジソ抽出物としては、製品名:シソエキスOGI(小川香料社製)、ローズマリー抽出物としては、製品名:ローズマリーエキスMF(丸善製薬社製)、レモンバーム抽出物としては、製品名:レモンバームエキスパウダーMF(丸善製薬社製)、セージ抽出物としては、製品名:サルビア・セージ(Finzelberg社製)、クミスクチン抽出物としては、製品名:クミスクチンエキス末(金秀バイオ社製)などが挙げられる。
アカジソ、アオジソ、ローズマリー、レモンバーム、セージ及びクミスクチンなどのシソ科植物は、従来より食用として用いられている。よって、本発明の有効成分であるロズマリン酸又はその塩としてシソ科植物由来の抽出物を用いると、安全性の高い夜間頻尿の予防若しくは改善剤並びに飲食品組成物を提供することができる。
シソ科植物の抽出物を用いる場合、抽出物中のロズマリン酸及びその塩が占める割合は、抽出物固形分に対してロズマリン酸量換算で0.1質量%以上が好ましく、より好ましくは1質量%以上、さらに好ましくは10質量%以上である。上限値は100質量%であるが、95質量%以下が好ましく、90質量%以下がより好ましい。あるいは、0.1質量%以上100質量%以下が好ましく、1質量%以上95質量%以下がより好ましく、10質量%以上90質量%以下がさらに好ましい。なお、本明細書において「抽出物固形分」とは、抽出物を105℃の乾燥機で3時間乾燥して揮発物質を除いた残分をいう。
後記実施例でも示すように、ロズマリン酸を投与又は摂取させても、昼間の頻尿に対する改善は確認できない。これに対し、ロズマリン酸を投与又は摂取させることで、睡眠時(夜間)の排尿回数が有意に減少する。そのため、ロズマリン酸又はその塩を、夜間頻尿を予防又は改善するために使用することができる。さらに夜間頻尿を予防又は改善することで、睡眠中の尿意を低減し若しくは下腹部の緊張感を和らげ、健康な睡眠をサポートすることができる。ここで、下腹部の緊張感とは、日常生活で感じるトイレに行きたいというソワソワ感(Restless feeling)や落ち着かない感じを意味する。また睡眠中の排尿に関する不安感を軽減することができる。
上記使用は、治療的使用(即ち医療行為)であっても非治療的使用(非医療的な行為)であってもよい。また、上記使用の対象は、ヒト、非ヒト動物、又はそれらに由来する検体であり得る。なお、前記「非治療的」とは、医療行為、すなわち治療による人体への処理行為を含まない概念である。
上記使用は、治療的使用(即ち医療行為)であっても非治療的使用(非医療的な行為)であってもよい。また、上記使用の対象は、ヒト、非ヒト動物、又はそれらに由来する検体であり得る。なお、前記「非治療的」とは、医療行為、すなわち治療による人体への処理行為を含まない概念である。
本発明の予防又は改善剤は、上記使用の具体的態様の1つであり、治療的用途(医療用途)、非治療用途(非医療用途)のいずれにも適用することができる。具体的には、医薬品、医薬部外品等としての使用することができる他、各種の飲食品、飼料、ペットフード等に本発明の予防又は改善剤を配合することもできる。
本発明の予防又は改善剤は、液状、固形状、乳液状、ペースト状、ゲル状、パウダー状(粉末状)、顆粒状、ペレット状、スティック状等、ヒトや非ヒト動物(好ましくは哺乳動物)に適用されうる各種剤型をとることができる。
また、本発明の予防又は改善剤は、有効成分である上記化合物のみからなるものであってもよいし、効果に影響を与えない範囲で他の成分を含有するものであってもよい。他の成分とは、例えば下記の添加剤が挙げられる。
本発明の予防又は改善剤は、液状、固形状、乳液状、ペースト状、ゲル状、パウダー状(粉末状)、顆粒状、ペレット状、スティック状等、ヒトや非ヒト動物(好ましくは哺乳動物)に適用されうる各種剤型をとることができる。
また、本発明の予防又は改善剤は、有効成分である上記化合物のみからなるものであってもよいし、効果に影響を与えない範囲で他の成分を含有するものであってもよい。他の成分とは、例えば下記の添加剤が挙げられる。
本発明の予防又は改善剤を医薬品、医薬部外品に適用する場合、前記化合物を有効量含有させ、必要により添加剤を配合して各種剤形に調製することができる。例えば、錠剤、被覆錠剤、カプセル剤、顆粒剤、散剤、シロップ剤、腸溶剤、トローチ剤、ドリンク剤等の経口医薬として、又は、注射剤、坐剤、経皮吸収剤、外用剤等といった非経口医薬として、医薬品や医薬部外品を調製することができる。これらの形態のうち、好ましい形態は経口医薬である。
医薬品や医薬部外品を種々の剤型に調製するには、添加剤を用いて常法に従って製造すればよい。添加剤は、通常用いられているものを使用することができる。添加剤の例としては、薬学的に許容される賦形剤、液体担体、油性担体、安定化剤、湿潤剤、乳化剤、結合剤、等張化剤、崩壊剤、滑沢剤、増量剤、界面活性剤、分散剤、懸濁剤、希釈剤、浸透圧調整剤、pH調整剤、防腐剤、抗酸化剤、着色剤、紫外線吸収剤、保湿剤、増粘剤、光沢剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、矯臭剤、細菌抑制剤等が挙げられる。
医薬品や医薬部外品を種々の剤型に調製するには、添加剤を用いて常法に従って製造すればよい。添加剤は、通常用いられているものを使用することができる。添加剤の例としては、薬学的に許容される賦形剤、液体担体、油性担体、安定化剤、湿潤剤、乳化剤、結合剤、等張化剤、崩壊剤、滑沢剤、増量剤、界面活性剤、分散剤、懸濁剤、希釈剤、浸透圧調整剤、pH調整剤、防腐剤、抗酸化剤、着色剤、紫外線吸収剤、保湿剤、増粘剤、光沢剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、矯臭剤、細菌抑制剤等が挙げられる。
本発明の予防若しくは改善剤、又は前述の有効成分を飲食品、飼料、ペットフード等に配合適用し、飲食品組成物、飼料組成物、ペットフード組成物等とする場合、食用又は飲料用に適した形態、例えば、顆粒状、粒状、液状、ゼリー状、錠剤、カプセル、ペーストなどに成形して提供することができる。さらに、前記飲食品組成物は、一般飲食品の他、夜間頻尿の予防若しくは改善をコンセプトとし、必要に応じてその旨を表示した美容食品、病者用食品、栄養機能食品、特定保健用食品又は機能性表示食品等の機能性飲食品の形態とすることができる。
飲食品への配合の例としては、小麦粉加工食品、米加工食品、菓子類、飲料類、乳製品、調味料、蓄肉加工食品、水産加工食品、調理油等が挙げられる。また、前記飲食品組成物は、錠剤(タブレット)、カプセル等の錠剤食、濃厚流動食、自然流動食、半消化態栄養剤、成分栄養剤、ドリンク栄養剤等の経口経腸栄養食品等の形態としてもよい。
飼料組成物及びペットフード組成物としては、ウサギ、ラット、マウス等に用いる小動物用飼料、犬、猫、リス等に用いるペットフード等が挙げられる。
これらの飲食品組成物、飼料組成物及びペットフード組成物等は、本発明の予防若しくは改善剤、又は前述の有効成分を含有し、これに食品原料、例えば、甘味剤、着色剤、抗酸化剤、ビタミン類、香料、ミネラル等の添加剤、タンパク質、脂質、糖質、炭水化物、食物繊維等を適宜組み合わせて、常法に従って調製することができる。
飲食品への配合の例としては、小麦粉加工食品、米加工食品、菓子類、飲料類、乳製品、調味料、蓄肉加工食品、水産加工食品、調理油等が挙げられる。また、前記飲食品組成物は、錠剤(タブレット)、カプセル等の錠剤食、濃厚流動食、自然流動食、半消化態栄養剤、成分栄養剤、ドリンク栄養剤等の経口経腸栄養食品等の形態としてもよい。
飼料組成物及びペットフード組成物としては、ウサギ、ラット、マウス等に用いる小動物用飼料、犬、猫、リス等に用いるペットフード等が挙げられる。
これらの飲食品組成物、飼料組成物及びペットフード組成物等は、本発明の予防若しくは改善剤、又は前述の有効成分を含有し、これに食品原料、例えば、甘味剤、着色剤、抗酸化剤、ビタミン類、香料、ミネラル等の添加剤、タンパク質、脂質、糖質、炭水化物、食物繊維等を適宜組み合わせて、常法に従って調製することができる。
本発明の予防若しくは改善剤、並びに飲食品組成物における前記有効成分の配合量又は含有量は、その使用形態により適宜決定することができる。
例えば、錠剤、被覆錠剤、顆粒剤、散剤、カプセル剤等の経口用固形製剤、内服液剤、シロップ剤等の経口用液体製剤の場合は、本発明の予防若しくは改善剤、又は飲食品組成物の総量に対する、前記有効成分の配合量又は含有量はロズマリン酸量換算で0.01質量%以上が好ましく、0.1質量%以上がより好ましく、1質量%以上がさらに好ましく、100質量%以下が好ましく、90質量%以下がより好ましく、70質量%以下がさらに好ましい。あるいは、0.01~100質量%が好ましく、0.1~90質量%がより好ましく、1~70質量%がさらに好ましい。
本発明の予防若しくは改善剤を飲食品やペットフード等に配合する場合は、本発明の予防若しくは改善剤、又は飲食品組成物の総量に対する、前記有効成分の配合量又は含有量はロズマリン酸量換算で0.001質量%以上が好ましく、0.01質量%以上がより好ましく、0.1質量%以上がさらに好ましく、50質量%以下が好ましく、10質量%以下がより好ましく、3質量%以下がさらに好ましい。あるいは、0.001~50質量%が好ましく、0.01~10質量%がより好ましく、0.1~3質量%がさらに好ましい。
例えば、錠剤、被覆錠剤、顆粒剤、散剤、カプセル剤等の経口用固形製剤、内服液剤、シロップ剤等の経口用液体製剤の場合は、本発明の予防若しくは改善剤、又は飲食品組成物の総量に対する、前記有効成分の配合量又は含有量はロズマリン酸量換算で0.01質量%以上が好ましく、0.1質量%以上がより好ましく、1質量%以上がさらに好ましく、100質量%以下が好ましく、90質量%以下がより好ましく、70質量%以下がさらに好ましい。あるいは、0.01~100質量%が好ましく、0.1~90質量%がより好ましく、1~70質量%がさらに好ましい。
本発明の予防若しくは改善剤を飲食品やペットフード等に配合する場合は、本発明の予防若しくは改善剤、又は飲食品組成物の総量に対する、前記有効成分の配合量又は含有量はロズマリン酸量換算で0.001質量%以上が好ましく、0.01質量%以上がより好ましく、0.1質量%以上がさらに好ましく、50質量%以下が好ましく、10質量%以下がより好ましく、3質量%以下がさらに好ましい。あるいは、0.001~50質量%が好ましく、0.01~10質量%がより好ましく、0.1~3質量%がさらに好ましい。
本発明の予防若しくは改善剤、並びに飲食品組成物の投与又は摂取量は、個体の状態、体重、性別、年齢又はその他の要因に従って適宜選択、決定できる。例えば、成人(体重60kg)の1日の投与又は摂取量としては、ロズマリン酸量換算で0.1mg以上が好ましく、10mg以上がより好ましく、50mg以上がさらに好ましく、40000mg以下が好ましく、15000mg以下がより好ましく、5000mg以下がさらに好ましい。あるいは、0.1~40000mgが好ましく、10~15000mgがより好ましく、50~5000mgがさらに好ましい。
また、本発明の予防若しくは改善剤、並びに飲食品組成物は、1日1回~数回に分け、又は任意の期間及び間隔で摂取又は投与され得る。摂取又は投与期間は適宜決定することができ、例えば、1日以上が好ましく、7日以上がより好ましく、30日以上がさらに好ましい。
上記医薬品、医薬部外品又は飲食品組成物の摂取又は投与対象としては、特に限定されないが、夜間頻尿の予防、改善、治療を所望又は必要とするヒトやヒト以外の哺乳動物が好ましい。なお、摂取又は投与対象には、夜間頻尿の症状が認められるヒトやヒト以外の哺乳動物、及びそのおそれがあるヒトやヒト以外の哺乳動物、その疾患又は症状の予防を期待するヒトやヒト以外の哺乳動物も含まれる。ヒトにおける夜間頻尿の具体的な症状としては、排尿のために睡眠時に1回以上起きる日が、週当たりの平均で1日以上、好ましくは2日以上、より好ましくは3日以上、さらに好ましくは4日以上、であることが挙げられる。
また、本発明の予防若しくは改善剤、並びに飲食品組成物の摂取若しくは投与対象の年齢は、20歳以上が好ましく、40歳以上がより好ましく、60歳以上がさらに好ましい。
また、本発明の予防若しくは改善剤、並びに飲食品組成物の摂取若しくは投与対象の年齢は、20歳以上が好ましく、40歳以上がより好ましく、60歳以上がさらに好ましい。
上述した実施形態に関し、本発明はさらに以下の予防若しくは改善剤、飲食品組成物、並びに方法を開示する。
<1>ロズマリン酸又はその塩を有効成分とする、夜間頻尿の予防又は改善剤。

以下、本発明を実施例に基づきさらに詳細に説明するが、本発明はこれに限定されるものではない。
試験例1 ロズマリン酸の経口摂取がラットの排尿機能に及ぼす影響
試験には、高血圧自然発症ラット(Spontaneously Hypertensive Rats:SHR、American Journal of Physiology,1998年,第275巻,p.R1279-1286、Neurourology and Urodynamics,2005年,第24巻,p.295-300参照)(42週齢、雌、日本SLC社より入手)を使用した。
処理群(n=12)のSHRにはロズマリン酸含有飼料(組成:0.5質量%ロズマリン酸(Carbosynth社より入手)、66質量%スターチ、20質量%カゼイン、5質量%コーン油、4質量%セルロース、3.5質量%ミネラル、1質量%ビタミン)を、対照群(n=12)のSHRにはロズマリン酸不含飼料(組成:66.5質量%スターチ、20質量%カゼイン、5質量%コーン油、4質量%セルロース、3.5質量%ミネラル、1質量%ビタミン)をそれぞれ8週間摂取させた後、代謝ケージにて排尿行動を測定した。なお、代謝ケージの測定前には4日間の順化期間を設定し、個別飼育を行った。

排尿行動測定(排尿回数と排尿量の測定)は、自由摂餌及び自由飲水条件下、12時間毎の明暗周期条件下で実施した。排尿量は、1回の排尿毎に尿を回収して電子天秤で秤量することにより測定した。尚、既報(Neurourology and Urodynamics,2017年,第36巻,p.1034-1038参照)を参考に、ラットの休止期に相当する明期の開始時刻をZT0とし、明期をZT0-6とZT6-12に2分し、それぞれの排尿回数を夜間頻尿の指標とした。また、活動期に相当する暗期をZT12-18とZT18-24に2分し、それぞれの排尿回数を昼間頻尿の指標とした。
試験食摂取後の排尿回数を表1に、排尿量を表2に、休止期と活動期それぞれ6時間毎に集計して示す。結果は平均値±標準誤差で示し、群間の平均値の比較にはStudent-t検定を用いた。

表1に示すように、対照群と比較して処理群で、ラットの睡眠時に相当する休止期(ZT6-12)における平均排尿回数が有意に減少していた。

試験例2 ロズマリン酸含有シソエキスの経口摂取がヒトの排尿機能に及ぼす影響
(1)被験者
20才代~50才代の健常男性14名。
試験開始時における被験者の平均年齢は38.6±2.8歳、平均BMIは21.8±0.6であった。また、試験開始時の昼間(睡眠時以外)の平均排尿回数は7.6±0.4回、夜間(睡眠時)の平均排尿回数は0.24±0.11回であった。
(1)被験者
20才代~50才代の健常男性14名。
試験開始時における被験者の平均年齢は38.6±2.8歳、平均BMIは21.8±0.6であった。また、試験開始時の昼間(睡眠時以外)の平均排尿回数は7.6±0.4回、夜間(睡眠時)の平均排尿回数は0.24±0.11回であった。
(2)被験品の作製
市販の赤シソの水抽出物(製品名:ニチノー 赤シソエキスRA15P、日農化学工業社製、ロズマリン酸を15,400mg/100g含有)を2号カプセル(製品名:ゼラチンカプセル、クリオカプス社製)に、カプセル1粒当たりのロズマリン酸量が25mgになるように充填し、ロズマリン酸含有シソ抽出物入りカプセルを作製した。ロズマリン酸不含プラセボカプセルとして、ロズマリン酸含有シソ抽出物の代わりに同量の澱粉を充填したものを作製した。
市販の赤シソの水抽出物(製品名:ニチノー 赤シソエキスRA15P、日農化学工業社製、ロズマリン酸を15,400mg/100g含有)を2号カプセル(製品名:ゼラチンカプセル、クリオカプス社製)に、カプセル1粒当たりのロズマリン酸量が25mgになるように充填し、ロズマリン酸含有シソ抽出物入りカプセルを作製した。ロズマリン酸不含プラセボカプセルとして、ロズマリン酸含有シソ抽出物の代わりに同量の澱粉を充填したものを作製した。
(3)試験形態
2群クロスオーバー試験(ウォッシュアウト期間:28日以上)
(i)処理群(ロズマリン酸含有シソ抽出物入りカプセル、4粒/日、ロズマリン酸100mg含有)
(ii)対照群(ロズマリン酸不含プラセボカプセル、4粒/日)

各カプセル(2粒)を100mLのミネラルウォーターと一緒に、1日2回(朝食後及び夕食後それぞれ、30分以内)で28日間摂取した。

カプセルの摂取前後の排尿状況を3日間記録し、排尿回数(昼間、夜間)と、1日あたりの排尿量などを記録した。

(i)ノンカフェイン茶飲料、水以外の飲料を摂取せず、カフェインの摂取を禁止した。
(ii)シソを含有する食品の摂取を禁止した。

(v)普段と同じ睡眠環境(照明、温湿度設定、窓の開閉)で睡眠した。

(6)統計学的検定
排尿回数や1日あたりの排尿量などについて得られた結果は、平均値±標準誤差で示し、群間の平均値の比較にはPaired-t検定を用いた。
被験品摂取前後における昼間及び夜間の排尿回数の変化量について解析した。その結果を表3に示す。また、被験品摂取前後における1日あたりの排尿量の変化量について、結果を表4に示す。
排尿回数や1日あたりの排尿量などについて得られた結果は、平均値±標準誤差で示し、群間の平均値の比較にはPaired-t検定を用いた。
被験品摂取前後における昼間及び夜間の排尿回数の変化量について解析した。その結果を表3に示す。また、被験品摂取前後における1日あたりの排尿量の変化量について、結果を表4に示す。
表3に示すように、対照群と比較して、処理群において昼間の平均排尿回数について有意な変化は認められなかったが(0.52±0.53回減少)、処理群において夜間の平均排尿回数は有意に減少した(0.24±0.11回減少、P<0.05)。
一方、表4に示すように、対照群と比較しても、処理群において1日あたりの平均排尿量に有意な変化は認められなかった。
尚、被験品摂取前における昼間の平均排尿回数と夜間の平均排尿回数、1日あたりの平均排尿量には、対照群と処理群の群間に有意差は認められなかった。
一方、表4に示すように、対照群と比較しても、処理群において1日あたりの平均排尿量に有意な変化は認められなかった。
尚、被験品摂取前における昼間の平均排尿回数と夜間の平均排尿回数、1日あたりの平均排尿量には、対照群と処理群の群間に有意差は認められなかった。
以上の結果から、ロズマリン酸は夜間頻尿の予防又は改善効果を有し、これを摂取することにより夜間の頻尿を予防若しくは改善できることが明らかとなった。
本発明をその実施態様とともに説明したが、我々は特に指定しない限り我々の発明を説明のどの細部においても限定しようとするものではなく、添付の請求の範囲に示した発明の精神と範囲に反することなく幅広く解釈されるべきであると考える。
本願は、2018年5月28日に日本国で特許出願された特願2018-101411に基づく優先権を主張するものであり、これはここに参照してその内容を本明細書の記載の一部として取り込む。
