Abstract
본 발명은 진세노사이드 Rd 함량이 증가되고 단도가 높아진 흑삼 및 이의 제조방법에 관한 것이다. 본 발명에 따른 흑삼을 제조하는 과정에서 먼저, 인삼 중의 전분을 가수분해하고, 다음으로 인삼 중의 프로토파낙사디올 진세노사이드를 진세노사이드 Rd로 가수분해하는 단계를 포함한다. 본 발명은 흑삼의 진세노사이드 함량을 1.00 내지 7.00 mg/g까지 증가시킬 수 있고 당도를 5.00 내지 7.00Brix 까지 증가시킬 수 있다.
Description
본 발명은 당도가 높고 진세노사이드 Rd 함량이 높은 흑삼 및 이의 제조방법에 관한 것으로 증숙 공정에서 당도를 높이고 진세노사이드 Rd를 강화시킬 수 있어서 기존 농축액을 이용한 제조방법보다 효율성, 경제성이 높다.
인삼의 지표성분인 진세노사이드는 아글리콘(aglycone)의 화학적 구조에 따라 크게 dammarane type과 oleanane type으로 분류된다. Dammarane type은 아글리콘의 3, 6, 12번 탄소에 결합된 hydroxy group에 따라 protopanaxadiol, protopanaxatriol로 분류된다. Protopanaxadiol ginsenoside는 ginsenoside Rb1, Rd, Rc, Rg3 등이 있고 protopanaxatriol ginsenoside는 ginsenoside Rg1, Re, F1 등이 알려져 있다. Protopanaxadiol ginsenosides의 인체대사과정을 살펴보면, 진세노사이드 Rb1은 위, 장에서 위산 또는 장내미생물이 생산하는 효소에 의해서 진세노사이드 Rg3로 가수분해되거나 진세노사이드 Rd, 진세노사이드 F2를 거쳐 Compound K로 가수분해된다. 이 과정에서 여러가지 진세노사이드 대사체가 생선되나 주된 대사경로는 진세노사이드 Rb1에서 진세노사이드 Rg3로 또는 진세노사이드 Rb1에서 진세노사이드 Rd, 진세노사이드 F2를 거쳐 Compound K로 전환되는 경로이다. 다양한 진세노사이드의 대사경로를 도 9에 나타내었다.
한국공개특허 제10-2015-0088844호는 진세노사이드 Rg2, Rg3, Rh1, Rh2 및 Rf를 포함하는 화장료 또는 항산화용 건강기능식품 조성물에 관한 것으로 흑삼을 물로 추출한 흑삼 추출물을 사카로마이세스 세레비지애로 효모발효시키는 것이고, 한국공개특허 제10-2015-0160977호는 진세노사이드 성분 증가를 위한 인삼열매 가공방법에 관한 것으로 엿기름을 이용한 당화발효공정을 실시하였으나 진세노사이드 Rd의 증가가 당화발효공정에 의한 것인지 명확히 규명하지 못했으며, 한국등록특허 제10-1120996호는 진세노사이드 Rg3 및 Rg2를 포함하는 운동능력증진 및 피로회복 증진용 조성물에 관한 것으로 분쇄한 흑삼에 펙티네이즈, 헤미셀룰레이즈, 셀룰레이즈와 같은 섬유소 분해효소 및 알파-아밀레이즈와 같은 다당류 분해효소 중 선택된 1종 이상의 효소로 처리한 것이나 진세노사이드 Rd는 강화시키지 못하였고, 한국등록특허 제10-1423100호는 진세노사이드 Rg3 및 Rb1의 함량을 증폭시키는 발효홍삼의 제조방법에 관한 것이며, 한국등록특허 제10-1308880호는 복합 미생물의 효율적 발효에 의한 발효홍삼농축액의 제조방법에 관한 것으로 원료홍삼농축액에 Lactis균, L.

본 발명은 흑삼을 제조하는 과정에서 인삼 중의 전분을 이당류, 단당류로 가수분해하고 프로토파낙사디올 진세노사이드를 진세노사이드 Rd로 가수분해하는 흑삼 및 이의 제조방법에 관한 것으로 당도가 높고 진세노사이드 Rd 함량이 증가된 흑삼을 제공한다.

본 발명에 따른 진세노사이드 Rd가 강화된 흑삼의 제조방법은, 세척한 수삼을 박피해 수삼의 왁스층이 제거된 박피인삼을 제조하는 단계; 상기 박피인삼을 60 내지 70℃에서 수분함량 55 내지 65%까지 건조해 건조인삼을 제조하는 1차 건조단계; 상기 건조인삼을 효소희석액 A에 침지하는 단계; 상기 효소희석액 A에 침지한 건조인삼에 효소희석액 A을 분무하면서 60 내지 80℃에서 5 내지 8시간 동안 증숙해 건조인삼 중의 전분을 이당류, 단당류로 가수분해하는 1차 증숙단계; 상기 1차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 건조해 당화인삼을 제조하는 2차 건조단계; 상기 2차 건조단계를 거친 인삼에 효소희석액 B을 분무하면서 70 내지 80℃에서 6 내지 8시간 동안 증숙해 프로토파낙사디올 계열 진세노사이드를 진세노사이드 Rd로 전환하는 2차 증숙단계; 상기 2차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 건조하는 3차 건조단계 및 3차 건조한 인삼을 60 내지 70℃에서 4 내지 6시간 동안 증숙하는 3차 증숙단계; 상기 3차 증숙한 인삼을 수분함량 20%까지 건조하는 열풍건조단계; 상기 열풍건조단계를 거친 인삼을 수분함량 15%까지 냉풍건조하면서 제습하는 단계를 거쳐 진세노사이드 Rd가 강화된 흑삼이 제조된다.
이하 각 단계를 상세히 설명한다.
먼저, 세척한 수삼을 박피해 수삼의 왁스층을 제거한 박피인삼을 제조한다. 일반적으로 박피하지 않고 수삼을 증숙하고 건조하는 과정을 3회이상 반복해 흑삼을 제조하나, 본 발명에서 왁스층을 제거하는 것은 인삼 중의 전분을 가수분해하는 효소희석액 A, 인삼 중의 프로토파낙사디올 진세노사이드를 가수분해하는 효소희석액 B가 인삼 조직으로 용이하게 침투할 수 있도록 한다.
상기 박피인삼을 60 내지 70℃에서 수분함량 55 내지 65%까지 1차 건조해 건조인삼을 제조한다.
상기 건조인삼을 효소희석액 A에 침지한다. 바람직한 상기 효소희석액 A는 Bacillus
licheniformis에서 얻어진 알파-아밀라제, Bacillus
amyloliquefaciens에서 얻어진 알파-아밀라제 또는 이들의 혼합물을 선택적으로 사용할 수 있다. 더욱 바란직한 효소희석액 A는 Bacillus
licheniformis에서 얻어진 알파-아밀라제를 사용할 수 있다. 건조인삼을 효소희석액 A에 침지하는 것은 인삼 조직 중앙부에 주로 분포하는 전분을 가수분해할 수 있도록 한다.
상기 효소희석액 A에 침지한 건조인삼에 효소희석액 A을 분무하면서 60 내지 80℃에서 5 내지 8시간 동안 1차 증숙한다. 상기 1차 증숙단계에서 인삼 중의 전분이 이당류, 단당류로 전환될 수 있다.
상기 1차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 2차 건조해 당화인삼을 제조한다. 상기 2차 건조단계를 거쳐 제조한 당화인삼 추출액의 당도는 5 내지 7Brix까지 증가할 수 있다.
상기 2차 건조단계를 거친 인삼에 효소희석액 B을 분무하면서 70 내지 80℃에서 6 내지 8시간 동안 2차 증숙한다. 바람직한 효소희석액 B는 Aspergillus
niger에서 얻어진 글루코아밀라제, Trichoderma
reesei에서 얻어진 셀루라제, 베타-글루카나제, 헤미샐룰라제의 복합효소 또는 이들의 혼합물을 선택적으로 사용할 수 있다. 더욱 바람직한 효소희석액 B는 Aspergillus
niger에서 얻어진 글루코아밀라제를 사용할 수 있다. 상기 2차 증숙하는 단계에서 인삼 중의 프로토파낙사디올 진세노사이드가 진세노사이드 Rd로 전환된다.
상기 2차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 3차 건조하고, 상기 3차 건조한 인삼을 60 내지 70℃에서 4 내지 6시간 동안 3차 증숙한다.
상기 3차 증숙한 인삼을 수분함량 20%까지 건조하는 열풍건조단계; 상기 열풍건조단계를 거친 인삼을 수분함량 15%까지 냉풍건조하면서 제습하는 단계를 거쳐 진세노사이드 Rd가 강화된 흑삼이 제조된다. 상기한 제조방법으로 제조한 흑삼 중의 진세노사이드 함량은 1.00 내지 7.00 mg/g까지 증가할 수 한다.
이하에서는 실시예를 들어서 본 발명을 상세하게 설명하지만, 이들 실시예에 의하여 본 발명의 권리범위가 제한되는 것은 아니다.
**실시예 1**
진세노사이드 Rb1이 함유된 인삼 전분 희석액의 조제
인삼에서 추출한 전분을 증류수에 희석해 전분희석액을 조제하고 진세노사이드 Rb1의 농도가 2.00 mg/mL가 되도록 표준품을 희석하였다.
**실시예 2**
스페자임 FRED(Spezyme FRED)의 진세노사이드 Rb1 가수분해 특성 파악
상기 실시예 1의 희석액에 스페자임 FRED(이하 FRED) 2%(v/v)를 가하고 70℃에서 24시간 동안 중탕하면서 진세노사이드 Rb1, Rd, F2 및 Compound K를 분석한 결과를 도1에 나타내었다. 24시간 중탕 후 진세노사이드 Rb1 함량은 1.95 mg/mL로 이는 효소 FRED가 진세노사이드 Rb1을 진세노사이드 Rd, F2 또는 Compound K로 가수분해할 수 없음을 증명한다. 그리고 당도계로 측정한 당도가 0.01Brix에서 0.5Brix로 증가했음을 확인하였고 이는 인삼 전분이 이당류, 단당류로 가수분해되었음을 증명한다.
**실시예 3**
스페자임 LT300(Spezyme LT300)의 진세노사이드 Rb1 가수분해 특성 파악
상기 실시예 2과 같이 실시하되 FRED 대신에 LT300을 사용한 결과를 도2에 나타내었다. 24시간 중탕 후 진세노사이드 Rb1 함량은 1.37 mg/mL이고 진세노사이드 Rd, F2 또는 Compound K는 거의 불검출되었으며 당도는 0.4Brix까지 증가하였다. 이는 LT300은 진세노사이드 Rb1을 가수분해할 수 있지만 진세노사이드 Rd, F2 또는 Compound K로 가수분해할 수 없으며 인삼 전분은 가수분해할 수 있음을 증명한다.
**실시예 4**
바이오윈 AG(BioWin AG)의 진세노사이드 Rb1 가수분해 특성 파악
상기 실시예 2과 같이 실시하되 FRED 대신에 BAG를 사용한 결과를 도3에 나타내었다. 24시간 중탕 후 진세노사이드 Rb1, F2 및 Compound K는 검출되지 않았고 진세노사이드 Rd는 1.66 mg/mL까지 증가하였으며 당도는 0.01Brix로 증가하지 않았다. 이는 BAG가 진세노사이드 Rb1을 진세노사이드 Rd로 가수분해하고 이를 다시 진세노사이드 F2, Compound K로는 가수분해할 수 없음을 증명하고 전분은 가수분해할 수 없음을 증명한다.
**실시예 5**
로하멘트 CL(Rohament CL)의 진세노사이드 Rb1 가수분해 특성 파악
상기 실시예 2과 같이 실시하되 FRED 대신에 RCL을 사용한 결과를 도4에 나타내었다. 24시간 중탕 후 진세노사이드 Rb1, F2 및 Compound K는 거의 검출되지 않았고 진세노사이드 Rd는 1.17 mg/mL까지 증가하였으며 당도는 0.01Brix로 증가하지 않았다. 이는 RCL은 진세노사이드 Rb1을 진세노사이드 Rd로 가수분해하고 이를 다시 진세노사이드 F2, Compound K로는 가수분해할 수 없음을 증명하고 전분은 가수분해할 수 없음을 증명한다.
**실시예 6**
FRED 효소로 가수분해한 당화인삼의 제조
효소액이 잘 침투할 수 있도록 세척한 수삼(10 kg)의 왁스층을 제거한 박피인삼을 준비하고, 상기 박피인삼을 65±5℃에서 수분함량 60±5%까지 건조해 건조인삼을 제조하였다. 상기 건조인삼을 30%(v/v) FRED 효소희석액(효소희석액 A)에 1시간 동안 침지하고, 상기 효소희석액 A에 침지한 건조인삼에 2시간 마다 효소희석액 A를 분무하면서 70±10℃에서 8시간 동안 증숙하고, 상기 증숙한 인삼을 수분함량 60±5%까지 건조하고, 상기 건조한 인삼을 수분함량 20%까지 열풍건조하고, 상기 열풍건조한 인삼을 냉풍건조하면서 제습해 수분함량 15%까지 건조하였다.
**실시예 7**
LT300 효소로 가수분해한 당화인삼의 제조
상기 실시예 6과 같이 실시하되 FRED 효소 대신에 LT300 효소로 제조한 효소희석액 A를 사용해 당화인삼을 제조하였다.
**실시예 8**
효소를 사용하지 않은 당화인삼의 제조
상기 실시예 6과 같이 실시하되 효소희석액 A 대신에 증류수를 사용해 당화인삼을 제조하였다.
**실시예 9**
당화인삼의 진세노사이드 분석
건강기능식품공전의 진세노사이드분석법에 따라 실시예 6, 실시예 7 및 실시예 8에서 제조한 당화인삼 중의 진세노사이드를 분석한 결과를 도 5에 나타내었다. FRED, LT300 효소는 진세노사이드 Rb1를 가수분해할 수 없음을 확인하였다.
**실시예 10**
당화인삼 추출액의 당도측정
상기 실시예 6, 실시예 7 및 실시예 8에서 제조한 당화인삼을 10배수 정제수로 추출하고, 상기 추출액의 당도를 당도계로 측정한 결과를 도 6에 나타내었다. 효소를 사용하지 않은 실시 예 8의 당도는 2.45Brix이나 효소를 사용한 실시예 6, 실시예 7의 당도는 각각 6,56Brix, 5.38Brix로 당도가 증가하엿음을 확인하였다. 이는 인삼 중의 전분이 이당류, 단당류로 가수분해되었음을 증명한다.
**실시예 11**
BAG를 이용해 진세노사이드 Rd를 강화한 흑삼의 제조
효소액이 잘 침투할 수 있도록 세척한 수삼(10 kg)의 왁스층을 제거한 박피인삼을 준비하고, 상기 박피인삼을 65±5℃에서 수분함량 60±5%까지 1차 건조해 건조인삼을 제조하였다. 상기 건조인삼을 30%(v/v) FRED 효소희석액(효소희석액 A)에 1시간 동안 침지하고, 상기 효소희석액 A에 침지한 건조인삼에 2시간 마다 효소희석액 A를 분무하면서 70±10℃에서 8시간 동안 1차 증숙하고, 상기 1차 증숙한 인삼을 수분함량 60±5%까지 2차 건조하고였다. 상기 2차 건조한 인삼에 2시간 마다 30%(v/v) BAG 효소희석액(효소희석액 B)를 분무하면서 70±10℃에서 8시간 동안 2차 증숙하고, 상기 2차 증숙한 인삼을 수분함량 60±5%까지 3차 건조하고였다. 상기 3차 건조한 인삼을 65±5℃에서 6시간 동안 3차 증숙하고, 상기 3차 증숙한 인삼을 수분함량 20%까지 열풍건조하고, 상기 열풍건조한 인삼을 냉풍건조하면서 제습해 수분함량 15%까지 건조하였다.
**실시예 12**
RCL을 이용해 진세노사이드 Rd를 강화한 흑삼의 제조
상기 실시예 11과 같이 실시하되, BAG 효소 대신에 RCL 효소로 제조한 효소희석액 B를 이용해 흑삼을 제조하였다.
**실시예 13**
효소를 사용하지 않고 제조한 흑삼의 제조
상기 실시예 11과 같이 실시하되, 효소희석액 B 대신에 증류수를 이용해 흑삼을 제조하였다.
**실시예 14**
흑삼 중의 진세노사이드 분석
상기 실시예 9에서 사용한 진세노사이드 분석법을 이용해 분석한 진세노사이드 함량을 도 7에 나타내었다. 효소를 사용하지 않은 실시예 13의 진세노사이드 함량은 증가하지 않았고 진세노사이드 Rg3만 1.37 mg/g까지 증가하였다. 이는 진세노사이드 Rb1의 일부가 열역학적인 가수분해를 통해 진세노사이드 Rg3로 전화된 것이다. BAG 효소를 사용한 실시예 11은 진세노사이드 Rb1이 진세노사이드 Rd로 전환되어 6.52 mg/g까지 증가하였고 RCL을 사용한 실시예 12는 진세노사이드 Rb1이 진세노사이드 Rd로 가수분해되어 5.89 mg/g까지 증가하였다.
**실시예 15**
흑삼추출액의 당도 측정
상기 실시예 11과 같은 방법으로 흑삼추출액을 제조해 측정한 당도를 도 8에 나타내었다.
**부호의 설명**
본 발명에서 사용한 효소는 다음과 같다.
BioWin AG는 Aspergillus
niger에서 얻어진 글루코아밀라제가 함유된 효소로 본 발명에서는 "BAG"로 표시해 사용하였고, Rohament CL은 Trichoderma reesei에서 얻어진 셀룰라제, 베타-글루카나제 및 헤미셀룰라제가 함유된 효소로 "RCL"로 표시해 사용하였으며, Spezyme FRED는 Bacillus
licheniformis에서 얻어진 알파-아밀라제가 함유된 효소로 "FRED"로 표시해 사용하였고, Spezyme LT300은 Bacillus amyloliquefaciens에서 얻어진 알파-아밀라제가 함유된 효소로 "LT300"으로 표시해 사용하였다.
Claims (6)
- 세척한 수삼을 박피해 수삼의 왁스층이 제거된 박피인삼을 제조하는 단계;상기 박피인삼을 60 내지 70℃에서 수분함량 55 내지 65%까지 건조해 건조인삼을 제조하는 1차 건조단계;상기 건조인삼을 효소희석액 A에 침지하는 단계;상기 효소희석액 A에 침지한 건조인삼에 효소희석액 A을 분무하면서 60 내지 80℃에서 5 내지 8시간 동안 증숙해 건조인삼 중의 전분을 이당류, 단당류로 가수분해하는 1차 증숙단계;상기 1차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 건조해 당화인삼을 제조하는 2차 건조단계;상기 2차 건조단계를 거친 인삼에 효소희석액 B을 분무하면서 70 내지 80℃에서 6 내지 8시간 동안 증숙해 프로토파낙사디올 계열 진세노사이드를 진세노사이드 Rd로 전환하는 2차 증숙단계;상기 2차 증숙단계를 거친 인삼을 수분함량 55 내지 65%까지 건조하는 3차 건조단계 및 3차 건조한 인삼을 60 내지 70℃에서 4 내지 6시간 동안 증숙하는 3차 증숙단계;상기 3차 증숙한 인삼을 수분함량 20%까지 건조하는 열풍건조단계;상기 열풍건조단계를 거친 인삼을 수분함량 15%까지 냉풍건조하면서 제습하는 단계를 포함하는 진세노사이드 Rd가 강화된 흑삼 및 이의 제조방법.
- 제1항에 있어서,효소희석액 A는 Bacillus licheniformis에서 얻어진 α-amylase, Bacillus amyloliquefaciens에서 얻어진 α-amylase 또는 이들의 혼합물을 선택적으로 사용한 것으로 전분의 글루코사이드 결합을 가수분해하는 것을 특징으로 하는 흑삼 및 이의 제조방법.
- 제1항에 있어서,효소희석액 B는 Aspergillus niger에서 얻어진 글루코아밀라제, Trichoderma reesei에서 얻어진 셀루라제, 베타-글루카나제, 헤미샐룰라제의 복합효소 또는 이들의 혼합물을 선택적으로 사용한 것으로 진세노사이드 Rb1을 진세노사이드 Rd로 가수분해하는 것을 특징으로 하는 흑삼 및 이의 제조방법.
- 제1항에 있어서,2차 증숙단계의 프로토파낙사디올 계열 진세노사이드는 진세노사이드 Rb1, Rb2, Rb3 및 Rc인 것을 특징으로 하는 흑삼 및 이의 제조방법.
- 제1항에 있어서,당도는 5.00 내지 7.00Brix인 것을 특징으로 하는 흑삼 및 이의 제조방법.
- 제1항에 있어서,흑삼 중의 진세노사이드 Rd 함량은 1.00 내지 7.00 mg/g인 것을 특징으로 하는 흑삼 및 이의 제조방법.
